  • 1.1. Adipokinetic responses to injection of synthetic adipokinetic hormone I (sAKH) and relevant physiological parameters were studied in azadirachtin-induced 31 to 34-day-old Vth-instar over-aged male nymphs of Locusta migratoria which did not undergo the metamorphic moult to adult.
  • 2.2. The resting lipid and carbohydrate levels in the haemolymph and sAKH-induced activation of fat body glycogen phosporylase did not differ markedly between over-aged nymphs, normal nymphs and normal adults.
  • 3.3. Total fat body glycogen phosphorylase activity and the effect of sAKH on haemolymph carbohydrate level in over-aged nymphs were similar to those in normal adults and differed from those in normal nymphs.
  • 4.4. sAKH-induced elevation of haemolymph lipid level in the over-aged nymphs was higher than in normal nymphs but lower than in normal adults; thus, the over-agad nymphs attained only a partial adult competence in this respect.

  • 1.1. The yolk proteins of hermaphrodite Dolichorhabditis sp. (Nematode, Rhabditida) are composed of at least three polypeptides: VT1, VT2 and VT3 with molecular masses of 175.2, 107 and 82 kDa respectively.
  • 2.2. All three yolk polypeptides make up at least one native protein complex which can be resolved by PAGE.
  • 3.3. The yolk proteins are glycosylated and can be isolated by chromatography in Con A-Sepharose.
  • 4.4. Partial chymotryptic hydrolysis shows that VT2 in different from its C. elegans homologue, YP115.
  • 5.5. The main polypeptides synthesized by whole animals are the yolk components which are actively secreted in the incubation medium.

  • 1.1. Slow-growing juvenile Nile crocodiles were injected with recombinant bovine growth hormone (rbGH) once a week for 6 weeks and then re-treated after 4 weeks.
  • 2.2. The feed intake of the treated crocodiles was 26 g/kg/meal during the three periods, while the intakes of the controls were 15, 20 and 2 g/kg.
  • 3.3. The treated gained 2.3 and 0.9%/week in weight during the first and second injection period and the feed conversion efficiencies were 28 and 13%. The controls lost weight.
  • 4.4. The treated animals grew at rates of 0.98 and 0.43%/week during the first and the second injection period.
  • 5.5. Bovine GH enhances growth in juvenile crocodiles and seems to have less adverse effects than human GH.

  • 1.1. Adult male and female cockroaches (Blattella germanica) were maintained on a positive nitrogen balance diet (66% protein) containing various levels of allopurinol (0–3%) to determine the effects of allopurinol on urate synthesis and storage.
  • 2.2. Each insect was injected with [14C]hypoxanthine and after 1 week was analyzed for whole-body hypoxanthine, xanthine and urate radiolabel.
  • 3.3. There was a general trend of decreased whole-body radiolabel retention, radiolabeled body urates and total-body urate content in both sexes with increasing amounts of dietary allopurinol.
  • 4.4. Virgin female adults were allowed to feed on diets containing 0, 25 and 66% protein plus 0.1% allopurinol and were injected with [14C]xanthine.
  • 5.5. After 1 week radiolabel content in the whole-body xanthine and urate pools was determined.
  • 6.6. Females on the 0% protein diets contained less radiolabel in the whole-body and body urates than those on either 25 or 66% protein diets.

  • 3.3. Young adults were treated with precocene or ketoconazole in an attempt to influence the steroid contents in gonads.

  • 1.1. A comparison was made of the mechanical performance of heart muscle from mouse, an atricial mammal, with corticosterone as glucocorticoid and spiny mouse (Acomys cahirinus), a precocial mammal, with cortisol as glucocorticoid.
  • 2.2. Force-frequency responses were negative in mouse and positive in spiny mouse.
  • 3.3. During recovery, there was a gradual increase and an overshoot in the mouse, while in the spiny mouse there was an initial enhanced response, diminishing gradually with time.
  • 4.4. High calcium concentration inhibited contractile tension in mouse heart, while it was positively inotropic in spiny mouse heart. Changes in the concentration of calcium did not change the patterns of force-frequency response.
  • 5.5. Lowering the experimental temperature increased the time course and amplitude of the tension curve. However, various parameters exhibited different temperature sensitivity.
  • 6.6. There was a significant difference in the levels of circulating cortisol between male and female spiny mice.
  • 7.7. It is proposed that the differences in the mechanical responses of mouse and spiny mouse hearts may be explained in terms of the effects of the specific glucocorticoid hormone on the development of the sodium-calcium exchanger.

  • 1.1. DNase-I-like activity occurs in the carp (Cyprinus carpio) liver cytosol (supernatant 105,000g).
  • 2.2. The enzyme resembles DNase I from bovine pancreas in respect to the molecular mass (~31 kDa), pH (7.4) and ion requirements (Mg2+, Ca2+) and the ability to degrade native as well as denatured DNA.
  • 3.3. As judged by comparison of DNase zymograms obtained after native- and SDS-PAGE, the enzyme occurs in the three molecular forms of similar molecular weight and different charges.
  • 4.4. All these forms are inhibited by rabbit skeletal muscle actin as well as by endogenous actin isolated from the carp liver cytosol.

  • 1.1. Ejaculation of seminal fluid into the spermatophore formed inside the female bursa copulatrix terminates 20 min after the beginning of copulation of Bombyx mori. The amounts of amino acids transferred are small, but the amounts of amino acids in the spermatophore continue to increase after this time due to protein degradation and amino acid interconversions.
  • 2.2. Arginase, which has high activity in the vesicula seminalis, is transferred to the spermatophore without loss of activity during ejaculation. During mating, ornithine and much urea are formed in the spermatophore, indicating activity of the transferred arginase.
  • 3.3. In the spermatophore, marked increase of alanine with low concentrations of ornithine and glutamate suggests the presence of a pathway for the active formation of 2-oxoglutarate with pyruvate via glutamate from arginine. During mating, proline and glutamine also increase, but at low rates.
  • 4.4. Of the exocrine glands in the male reproductive system, the vesicula seminalis secretes the highest concentration of glutamate (30% of the total amino acids); serine is the amino acid present at highest concentration in secretions of other glands (20–30%). No urea was found in the secretions of any of the glands.

  • 1.1. Blood, liver, heart, testis, skin, eye, muscle and kidney samples were obtained from elephants (Loxodonta africana) in the Kruger National Park during a culling programme in April 1992.

  • 3.3. Eighteen protein coding loci (72%) displayed monomorphic gel banding patterns whereas only seven (28%) displayed polymorphic gel banding patterns.

  • 5.5. Relative gene diversities within and between populations are 84% and 16% respectively.
  • 6.6. Two population simulation programmes were utilized to predict the duration of the current variability present in this species, based on current genetic variation and gene transfer from one generation to the next.

  • 1.1. Fat body from feeding-phase, last instar gypsy moth females incorporates l-[35S]methionine in vitro into two vitellogenins with the same molecular masses (165 and 180 kDa) as the apo-vitellogenins found in teh hemolymph and the apo-vitellins in teh eggs.
  • 2.2. Both apo-vitellogenins are observed in the medium of fat body cultures, but only the 180 kDa apo-vitellogenin is observed in extracts of cultured tissue.
  • 3.3. Synthesis and accumulation of the apo-vitellogenins are suppressed in a dose-dependent manner by topical treatment with the juvenile hormone analog, methoprene, prior to day 4.
  • 4.4. This suppression suggests that a declining juvenile hormone titre is involved in the initiation of vitellogenin synthesis.

  • 1.1. A method is presented for the determination of the polycyclic aromatic hydrocarbon benzo(a)pyrene (BaP) in the isopod Porcellio scaber, using reversed-phase HPLC with fluorescence detection.
  • 2.2. This technique has a detection limit for BaP in P. scaber of approximately 3.2 ng g−1 fresh weight.
  • 3.3. BaP was assimilated from food by P. scaber.
  • 4.4. After four weeks of ad libitum feeding on BaP-contaminated food, concentrations in the isopods were approximately 30–40 times lower than those in the food.
  • 5.5. Male and female isopods did not differ in BaP concentration. Variation among males seemed to be much higher than among females.

  • 1.1. Recombinant salmon growth hormone at doses of 0.8 and 2.1 μg/g significantly enhanced linear growth in hypophysectomized male killifish, Fundulus heteroclitus, over that of controls and a significant regression was found between growth and the logarithm of dose.
  • 2.2. Bovine growth hormone elicited significant growth enhancement at all three dosages tested (1,4 and 10 μg/g) and a significant log/dose relationship was also observed.
  • 3.3. Observations on the relative weight of the gonads indicate that whole salmon pituitary extract (25 μg/g) possesses strong gonadotropic activity and that both bGH and rsGH had smaller but significant effects on the gonads.
  • 4.4. It is suggested that growth hormone may play a subsidiary synergistic role to other pituitary hormones in gonadal development.

  • 1.1. Hormonal regulation of apolipoprotein E (apoE) gene expression by insulin and thyroid hormone was studied in a human hepatoma cell line, HepG2.
  • 2.2. Changes at the mRNA level, mRNA translation, in vivo synthesis and secretion were monitored.
  • 3.3. Both insulin and triiodothyronine were found to have no significant effect on apoE mRNA levels.
  • 4.4. Insulin treatment caused an inhibition of: (a) the in vitro translation of endogenous apoE mRNA in a HepG2 cell-free system (25%), and (b) the incorporation of radioactivity into newly-synthesized apoE in an in vivo pulse-chase labeling experiment (32%).
  • 5.5. Interestingly, apoE secretion rate was found to be significantly reduced with insulin (84%) suggesting that a major portion of newly-synthesized apoE may be shunted into a degradative pathway.
  • 6.6. Using a similar experimental approach, triiodothyronine showed no significant effect on the rate of apoE synthesis or translation (6–15% decrease), however a slight reduction (20%) in secretion rate was shown.
  • 7.7. Overall, apoE gene expression does not appear to be influenced by triiodothyronine significantly but is modulated by insulin at the translational and post-translational level.

  • 1.1. A mechanical tissue chopper was used to obtain liver explants (35–75 mg) from 2- to 3-week-old chickens to determine both tissue sensitivity and metabolic effects of isoproterenol, avian insulin and glucagon.
  • 2.2. Avian insulin had no effect on lipogenesis; however, lipogenesis was decreased by dibutyryl cyclic AMP. Insulin did not overcome a decrease in lipogenesis caused by catecholamines. Therefore, this control mechanisms is not modulated by insulin.
  • 3.3. Preincubation in the presence of glucagon decreased in vitro lipogenesis. Preincubation in the presence of a 19–29 amino acid construct that approximated the radioimmune site for glucagon did not result in a similar effect. Therefore, this site does not relate to the biopotency of the hormone.
  • 4.4. A previously noted catecholamine induced decrease in in vitro lipogenesis was verified, showing that points of in vitro regulation are under phosphorylation-dephosphorylation control.
  • 5.5. Preincubation of slices (1 hr) with propranolol blocked the inhibition of lipogenesis caused by α and β adrenergic agonists (arterenol or isoproterenol) during a subsequent 2-hr incubation.
  • 6.6. Preincubation of slices with either of these agonists decreased lipogenesis even following an extensive washout.
  • 7.7. Inhibition could be overcome with propranolol, a β adrenergic antagonist.

  • 1.1. Soluble proteins extracted from male and female Penaeus vannamei tissues such as eyes, eyestalks, brain, nerve cord, hemolymph, heart, muscle, hepatopancreas, hepatopancreas membrane and cuticular epidermis were analyzed and compared by high-resolution mini-two-dimensional polyacrylamide gel electrophoresis (mini-2D-PAGE).
  • 2.2. In each shrimp tissue a large number of discrete polypeptides was observed.
  • 3.3. The polypeptide patterns from the same tissue of female and male shrimp were mostly similar but both qualitative and quantitative differences were noted, suggesting the presence of sex-specific gene products in various shrimp tissues.
  • 4.4. Future applications of these results are discussed.
