For the first time the scanning electron microscope was used to compare developmental changes in scorpion embryos and the first and second stadia. In the buthid species of this study, Centruroides vittatus, and all other scorpions, the newborn climb up on their mother's back and remain there without feeding for several days. At this location, they undergo their first molt and in a few days they disperse, fully capable of foraging in the terrestrial environment. The results here support earlier suggestions that the first stadium (pronymph) is a continuation and extension of embryological development. The first molt results in a nymph with exoskeletal features much like those in the adult. In the first molt the metasoma becomes relatively longer, and the sting (aculeus) becomes sharp and functional. The metasomal segments are modified for dorsal flexion and sting use. The embryos and the pronymphs have spiracles that open into an invagination near the posterior margin of flap-like abdominal plates in segments 4-7 of the ventral mesosoma. The second instars have spiracles that lead to book lungs farther anterior in sternites. Tubular legs with cylindrical segments in embryos and pronymphs become more sculptured and oval in the transverse plane. Each leg in the pronymph has a blunt, cup-shaped tip while distal claws (ungues, dactyl) are present in the second instar and subsequent stages. There are some sharp bristles and primordial sensilla in the pronymphs, but the second stadium has adult-like surface features: rows of knobs or granulations (carinae), serrations on the inner surfaces of cheliceral and pedipalpal claws, filtering hairs at the mouthparts, peg sensilla on the pectines, and mechano- and chemoreceptor sensilla on the body and appendages. Scorpion embryos and pronymphs have some structures like fossil scorpions thought to have been aquatic. There is a gradual development of features that appear to be terrestrial adaptations. Evidence is provided for the formation of the sternum from third and fourth leg coxal primordia and possibly from the first abdominal segment. The mature spermatozoa of Buthus occitanus are threadlike in shape and divided into sperm head, middle piece, and end piece. The sperm head is corkscrew shaped anteriorly and in this region bears an unusual acrosomal complex consisting of a ring-shaped acrosomal vacuole associated with a subacrosomal filament and a perinuclear amorphous component. The subacrosomal filament extends posteriorly into a tube-like invagination of the elongated nucleus. The middle piece is characterized by elongated mitochondria which spiral around the anterior part of the flagellum in an extended collar separated from the flagellum by an extracellular cleft, termed the central flagellar tunnel. In addition to the usual 9 × 2 + 2 axonemal pattern in flagella, 9 × 2 + 1 and 9 × 2 + 3 patterns also were observed. The end piece is represented by the free flagellum. Similarities and diversities of scorpionid spermatozoa are discussed with respect to systematic relationships. 相似文献

The venom apparatus of the scorpion, C. sculpturatus (Ewing) was studied with light and electron microscopy. Each of the paired glands is lined by secretory epithelium made up of a single layer of columnar cells. Extensive folding in the epithelial layer creates a primitive acinar gland. The secretory products are either membrane-bound or unbound vesicles with discrete morphologies and are observed in the extruded venom, within the lumen of the gland, and within single secretory cells. The venom apparatus, including connective tissues, nerve cells, and muscle tunic is described and correlations are made with observations in other Athropods. 相似文献

The mitochondrial genome of the scorpion Centruroides limpidus (Chelicerata; Arachnida) has been completely sequenced and is 14519 bp long. The genome contains 13 protein-encoding genes, two ribosomal RNA genes, 21 transfer RNA genes and a large non-coding region related to the control region. The overall A + T composition is the lowest among the complete mitochondrial sequences published within the Chelicerata subphylum. Gene order and gene content differ slightly from that of Limulus polyphemus (Chelicerata: Xiphosura): i.e., the lack of the trnD gene, and the translocation–inversion of the trnI gene. Preliminary phylogenetic analysis of some Chelicerata shows that scorpions (C. limpidus and Mesobuthus gibbosus) make a tight cluster with the spiders (Arachnida; Araneae). Our analysis does not support that Scorpiones order is the sister group to all Arachnida Class, since it is closer to Araneae than to Acari orders. 相似文献

Abstract. Water permeability of cuticular membranes (CM) from the inner bulb scales of Allium cepa has been investigated. CM have a thickness ranging from 0.6 to 1.3 μm. They are composed of a thin (120–200 nm) lamellated cuticle proper and a thicker (300–900 nm) cuticular layer. Permeability coefficients for diffusion of water across these thin membranes are very low (4 × lO−10 ms−10 ). There was no difference in permeability of CM from successive scales of the same onion. Extraction of soluble cuticular lipids (SCL) with chloroform increased permeability by a factor of 1350 to 2050. Preliminary data indicate that only 1 μg cm−10 of SCL are removed by this treatment, hydrocarbons being the main (75%) consistuent. Permeability coefficients of cuticular transpiration were little affected by relative humidity, showing that transport is limited by a hydrophobic barrier that lacks dipoles. However, following extraction, permeability of the membranes depended strongly on humidity due to the presence of polar functional groups in the polymer matrix. Soluble cuticular lipids undergo a phase transition around 47°C. Temperatures higher than that irreversibly increased water permeability. 相似文献

- 1.1. The relationship between nitrogen metabolism and osmoregulation has been studied in the prawn Palaemon elegans (Rathke) following sudden exposure to hyper- and hyposaline conditions.
- 2.2. Animals acclimated to a salinity of 30‰ showed a pronounced increase in the rates of ammonia excretion during the first 2 hr after transfer to lower salinities. These gradually declined during the next 6 hr to rates that were significantly higher than that of control animals (30‰) and were maintained throughout the rest of the experiment.
- 3.3. Rates of ammonia excretion in animals transferred to hypersaline conditions (40‰) fluctuated considerably during the experiment. It was consistently observed, however, that there were two periods during the experiments when ammonia excretion rates had negative values indicating that NH+4 ions were being taken up by the prawns.
- 4.4. Experiments in which small quantities of (NH4)2SO4 containing the stable isotope 15N were added to the sea-water confirmed that P. elegans was able to take NH+4 ions from the sea-water.
- 5.5. Changes in the Na+ ion concentration in the blood and the changes in free amino acid concentration in the blood and in the muscle after exposure to differing salinities were also determined. Their significance and relationship to the observed changes in the rates of ammonia excretion are discussed.

The SEM was used to study the development of respiratory structures in successive stages in relation to the overall changes occurring in the scorpions. Book lung development is a slow process, starting with spiracles and a sac‐like atrium in the early embryo and continuing lamellar formation to 150 or more in the adult. In the embryo, the primordial epithelial cells become aligned in a planar pattern as they secrete granules of material that aggregate spontaneously to form the cuticular walls of the lamellae. A blade‐like structure is formed consisting of cells sandwiched within the two cuticle walls they secreted. These cells are in the primordial air channel. The adjacent hemolymph channel is nearly devoid of cells, but cross‐bridges develop and help stabilize the cuticle walls and maintain the width of the channel. The cells in the primordial air channel undergo cytolysis, leaving it open for air except for cuticular cross‐bridges. Development continues in the newborn (first instars); the air channels of some lamellae still contain cells and are not yet functional for gas exchange. The first instars are weak and relatively inactive. They climb up on the mother's dorsum until the first molt (about 8 days). With the cuticular walls of the lamellae in place, cells adhering to the wall in the hemolymph channel produce a thin, new tissue layer (epithelium) on the lamellar wall facing the hemolymph channel. This layer has many discontinuities as though it is slowly developing. Formation of the tissue layer and cytolysis of the cells in the air channels continue through the first molt in which little book lung cuticle is shed as exuvium. The air channels of the second instars (foraging nymphs) are now cell free and open for air passage except for the cross‐bridges. The tissue layer is still incomplete and continues to be formed. It may provide the hypodermal primordium for cuticle replacement in later molts, but development was not studied beyond the second instar except for comparison with book lungs in the adult. The blade‐like lamellae in the adult are larger and more numerous than in the second instar, but in the anterior book lung the shape of the cuticle wall and cross‐bridges and the widths of the air and hemolymph channels are about the same as in the second instar. The air channels in the posterior part of the lamellae have distinctive, vein‐like space‐holders. The similarity of the adult anterior lamellae with those in the second instar suggests retention of this part through the 4–5 molts to maturation, and/or cell processes like those in the embryo are repeated, but this needs to be examined in further studies of cell and cuticle changes before and during the molts. The freshwater copepod Skistodiaptomus oregonensis was exposed,in laboratory bioassays, to a 3 x 3 factorial array of pH andaluminum treatments (pH 4.5, 6.0 and 7.5, and 0, 100 or 200µg l1 Al). After 12 and 24 h, whole-body Na contentand survivals were determined for groups of 10 animals per treatment.Declines in bodily Na were taken as evidence of osmoregulatorydysfunction. Both Na content and survival were reduced at pH6.0 and especially at pH 4.5. The magnitude of responses dependedon the Al concentration. Exposure to the highest Al level tendedto reduce Na content and survival at pH 6.0, but tended to enhanceNa content and survival at pH 4.5. - 1.1. Since glucose is one of the main energetic substrates for general metabolic processes in crustaceans, analysis of carbohydrate levels can furnish information on the energy metabolism of intact animals during osmoregulation.
- 2.2. Different groups of Chasmagnathus granulata were transferred to different salinities (0 and 40%), and the glucose and glycogen concentrations in blood, gills, muscle and hepatopancreas were determined at the beginning of the experiment and 24, 72, 168 and 360 hr after the salinity changes.
- 3.3. Differences in tissues carbohydrate levels were observed between summer and winter, that reflected differences in reserve mobilization.
- 4.4. In the summer, hypo- and hyperosmotic shocks induced an increase in carbohydrate levels in almost all tissues studied, indicating gluconeogenesis.
- 5.5. In the winter, a carbohydrate mobilization occurred only in the gills and hepatopancreas after both osmotic shocks.
- 6.6. Thus, the substrate reserve used for energy production required for osmoregulation seems to be dependent on the season and tissues.

The effect of rainfall on the metabolism of birds is poorly understood. We measured the metabolism as rate of oxygen consumption (VO2) of four male and four female American kestrels (Falco sparverius) using open-circuit respirometry. We measured VO2 during the spring at ambient temperatures (Ta) of 5, 10, 15, and 25 degrees C in air without rainfall and with simulated rainfall of 2.5 (low rainfall) and 6.1 cm h(-1) (high rainfall). Kestrel metabolism was significantly higher when exposed to the two rainfall levels compared to no rainfall. However, kestrel metabolism was not significantly different at the two rainfall levels. Body temperature (Tb) was significantly lower under high rainfall compared to low rainfall. In addition, under both rainfall levels Tb decreased with decreasing Ta. Calculated thermal conductance was significantly higher in kestrels exposed to rain compared to no rainfall. Kestrels may use sleeking behavior at high rainfall levels to decrease water penetration of the plumage. Daily energy expenditure (DEE) of kestrels exposed to rain may increase markedly, and kestrel energetics may be further exacerbated by wind that often accompanies natural rainstorms. 相似文献

Global warming is having an impact on the temperature and salinity of Baltic Sea waters. Therefore, it is important to determine the conditions in which animals can exist and how these changes may influence their functioning. Hence, the purpose of this research was to determine the broad tolerance limits of temperature and salinity of the glacial relict Saduria entomon by studying its behaviour, osmoregulatory ability and haemocyanin concentration. This effect of temperature was confirmed in the laboratory for individuals acclimated to different salinity and temperature regimes. Changes in the physiological parameters of S. entomon at various temperatures (5.5–21.5°C) and salinity levels (1–15 PSU) were recorded. There were statistically significant differences in haemolymph osmotic pressure under the influence of salinity and temperature. The mean haemolymph osmotic pressures were the lowest at 1 PSU at all the temperatures examined and the highest at 15 PSU and high temperatures 16.5 and 21.5°C. The haemocyanin concentration decreased significantly with increasing temperature at 1 PSU. There was a significant difference in haemocyanin concentration due to salinity at temperatures of 5.5 and 10.0°C (the haemocyanin concentration decreased with increasing salinity). The results showed that, although S. entomon is classified as a cold-water animal, it can survive at high temperatures above 16.5°C at least for a short time, as it is capable of osmoregulation. Summary Third larval instar hemolymph of the fruitflyDrosophila hydei did not metabolize juvenile hormone (JH) at all developmental stages. In contrast, prepupal and pupal body fluid showed JH-esterase activity with a maximum at 4 h after puparium formation. In body wall and fat body of all developmental stages investigated, JH-metabolic activity was found. In both tissues JH catabolism was most active in the 120,000g supernatant and pellet. The 800g and 15,000g pellet showed a lower activity. In all subcellular fractions the JH-acid was identified as the predominant metabolite. There is evidence that JH-specific esterases are responsible for ester cleavage in the 120,000g supernatant. During mid and late third larval instar development in both body wall and fat body JH-esterase activity remains relatively constant. 相似文献

Seven polypeptides highly toxic to mice were isolated from the venom of the scorpion, Centruroides suffusus suffusus (Css), and their chemical and toxic properties were characterized. It was shown that the most active toxins by intracerebroventricular injection are less active when injected subcutaneously. The complete amino acid sequence (66 residues) of toxin II (Css II) has been determined. The C-terminal end is amidated as found for most other scorpion toxins. Css II is a beta-type toxin, previously used to define the binding site for activation of the sodium channel. Using rat brain synaptosomes, we demonstrated that all Css toxins compete with 125I-Css II to bind to site 4 and should be considered as beta-scorpion toxins. Specific binding parameters for Css VI, one of the most active toxins, were determined: KD = 100 pM; capacity in binding sites, 2.2 pmol of toxin/mg of synaptosomal protein. Css VI was shown to inhibit gamma-aminobutyric acid uptake by synaptosomes: K 0.5 = 100 pM, which agrees with its KD. Competition experiments between the seven Css toxins and 125I-Css II for antiserum raised against Css II demonstrated that all these toxins have common antigenic properties. 相似文献

The results of laboratory tests indicated the average survival rates for Psorophora columbiae eggs remained quite high for all of the egg populations exposed to a temperature of 27°C (range 83.0–100.0% survival) after 96 days of exposure, except for the non‐diapausing eggs on dry soil (66.3%). In regard to the exposure of egg populations to moderately cold temperatures (i.e. 8°C, 4°C and ?2°C) for periods of up to 16 days, survival rates for egg populations exposed to 8°C continued to remain relatively high (average >85%) for the remainder of the experimental exposure period (i.e. 96 days). Diapausing Ps. columbiae eggs were more tolerant (82.0% survival) to low temperatures (?2°C) than non‐diapausing eggs (2.4% survival) for 64 days, particularly at temperatures of and below 4°C. Diapausing and non‐diapausing eggs were similar in their ability to survive under high temperatures (34°C and 38°C). High soil moisture (30–40%) or substrate moisture (95% relative humidity) content appeared to enhance the ability of the mosquito eggs to survive both low and high temperature extremes. 相似文献

Chortoicetes terminifera lays a mixture of diapause and non-diapause eggs during the autumn months of March and April. Diapause eggs cease development in late anatrepsis when kept in moist soil at 32.5°C. These conditions favour rapid embryogenesis and hatching in non-diapause eggs. Significant reductions in the incidence of diapause were obtained when newly-laid eggs were kept for 40 days at either 15.5°C or 32.5°C in conditions that prevented the uptake of water. In eggs held for a similar period in moist soil at 15.5°C, diapause was virtually eliminated. These eggs did not develop to the stage at which water uptake occurs. It is suggested that the inception of diapause is dependent upon the occurrence of warm, moist conditions at the time of laying and that these requirements are an integral part of the overall inductive process, which also involves the temperature and photoperiod at which the parent insects are reared. A flow-diagram illustrating the development of diapause eggs in relation to temperature and moisture is discussed in relation to the survival of the species. 相似文献

The three-dimensional structures of the long-chain mammalian scorpion β-toxin CssII from Centruroides suffusus suffusus and of its recombinant form, HisrCssII, were determined by NMR. The neurotoxin CssII (nCssII) is a 66 amino acid long peptide with four disulfide bridges; it is the most abundant and deadly toxin from the venom of this scorpion. Both native and recombinant CssII structures were determined by nuclear magnetic resonance using a total of 828 sequential distance constraints derived from the volume integration of the cross peaks observed in 2D NOESY spectra. Both nCssII and HisrCssII structures display a mixed α/β fold stabilized by four disulfide bridges formed between pairs of cysteines: C1-C8, C2-C5, C3-C6, and C4-C7 (the numbers indicate the relative positions of the cysteine residues in the primary structure), with a distortion induced by two cis-prolines in its C-terminal part. The native CssII electrostatic surface was compared to both the recombinant one and to the Cn2 toxin, from the scorpion Centruroides noxius, which is also toxic to mammals. Structural features such N- and C-terminal differences could influence toxin specificity and affinity towards isoforms of different sub-types of Na(v) channels. 相似文献

In the present study, the combined effects of temperature and diet on development and survival of a crab spider, Misumenops tricuspidatus (Fabricius) (Araneae: Thomisidae) in laboratory conditions were investigated. The experiments were carried out at five constant temperatures ranging from 15°C to 35°C on two kinds of diets, fruit flies (Drosophila melanogaster) and a mixed diet of fruit flies and dung flies. It was found that development rate of eggs increased with successive temperature increments, reaching a maximum at 30°C, then declined at 32°C and that eggs survived well between 20°C and 30°C (>70%), but no eggs survived to hatching at 35°C regardless of whether the spiders were fed on single or mixed diet. Juveniles completed development on both diets at all constant temperatures tested, but survival was low at the extreme temperatures. Juvenile development times decreased over successive temperature increments up to 30°C, then increased at 32°C. Females developed faster than males. Diet also influenced development time, survival and number of moults to reach maturity. Juveniles raised on the mixed diet composed of fruit flies and dung flies developed faster, survived better, and required fewer moults to reach maturity than on a diet composed of only fruit flies. Plots of development rates (reciprocal of mean development times) and survival rates (expressed as percentages) against constant temperatures indicated that M. tricuspidatus is well adapted to low temperatures, but detrimentally affected by high temperatures. Using linear regression, the lower development threshold (LDT) and the sum of effective temperatures (SET) for all life stages of M. tricuspidatus on each diet were estimated. An erythroagglutinin from the hemolymph of the scorpion, Heterometrus bengalensis, has been purified by gel filtration and ion-exchange chromatography. Its homogeneity has been demonstrated by polyacrylamide gel electrophoresis. The purified agglutinin appears to be a monomeric protein having a possible molecular weight between 146,000 and 148,000. It has no divalent cation requirement for erythroagglutination. The erythroagglutination is not inhibited by saccharides, glycoproteins and mucin. Identical erythroagglutination pattern is obtained with normal as well as neuraminidase treated erythrocytes. 相似文献
