Background

Hybridization events are relatively common in vascular plants. However, the frequency of these events is unevenly distributed across the plant phylogeny. Plant families in which individual species are pollinated by specific pollinator species are predicted to be less prone to hybridization than other families. However, exceptions may occur within these families, when pollinators shift host-plant species. Indeed, host shifts are expected to increase the rate of hybridization events. Pollinators of Ficus section Galoglychia are suspected to have changed host repeatedly, based on several cases of incongruence between plant phylogeny and taxonomy, and insect phylogeny and taxonomy. We tracked cyto-nuclear discordance across section Galoglychia as evidence for hybridization. To achieve a proper global view, we first clarified the monophyly of section Galoglychia as it had been questioned by recent phylogenetic studies. Moreover, we investigated if fig size could be a factor facilitating host shifts.  相似文献

Background

Yersinia enterocolitica, an important food- and water-borne enteric pathogen is represented by six biovars viz. 1A, 1B, 2, 3, 4 and 5. Despite the lack of recognized virulence determinants, some biovar 1A strains have been reported to produce disease symptoms resembling that produced by known pathogenic biovars (1B, 2-5). It is therefore imperative to identify determinants that might contribute to the pathogeniCity of Y. enterocolitica biovar 1A strains. Y. enterocolitica invariably produces urease and the role of this enzyme in the virulence of biovar 1B and biovar 4 strains has been reported recently. The objective of this work was to study genetic organization of the urease (ure) gene complex of Y. enterocolitica biovar 1A, biochemical characterization of the urease, and the survival of these strains under acidic conditions in vitro.  相似文献

Background

Escherichia coli strains are commonly found in the gut microflora of warm-blooded animals. These strains can be assigned to one of the four main phylogenetic groups, A, B1, B2 and D, which can be divided into seven subgroups (A0, A1, B1, B22, B23, D1 and D2), according to the combination of the three genetic markers chuA, yjaA and DNA fragment TspE4.C2. Distinct studies have demonstrated that these phylo-groups differ in the presence of virulence factors, ecological niches and life-history. Therefore, the aim of this work was to analyze the distribution of these E. coli phylo-groups in 94 human strains, 13 chicken strains, 50 cow strains, 16 goat strains, 39 pig strains and 29 sheep strains and to verify the potential of this analysis to investigate the source of fecal contamination.  相似文献

Background

Hydrogenosomes are organelles that produce molecular hydrogen and ATP. The broad phylogenetic distribution of their hosts suggests that the hydrogenosomes of these organisms evolved several times independently from the mitochondria of aerobic progenitors. Morphology and 18S rRNA phylogeny suggest that the microaerophilic amoeboflagellate Psalteriomonas lanterna, which possesses hydrogenosomes and elusive "modified mitochondria", belongs to the Heterolobosea, a taxon that consists predominantly of aerobic, mitochondriate organisms. This taxon is rather unrelated to taxa with hitherto studied hydrogenosomes.  相似文献

Background

The lipopolysaccharide is a major antigen and virulence factor of Brucella, an important bacterial pathogen. In smooth brucellae, lipopolysaccharide is made of lipid A-core oligosaccharide and N-formylperosamine O-polysaccharide. B. ovis and B. canis (rough species) lack the O-polysaccharide.  相似文献

Background

Bacillus anthracis is an animal and human pathogen whose virulence is characterized by lethal and edema toxin, as well as a poly-glutamic acid capsule. In addition to these well characterized toxins, B. anthracis secretes several proteases and phospholipases, and a newly described toxin of the cholesterol-dependent cytolysin (CDC) family, Anthrolysin O (ALO).  相似文献

Population dynamics and zonation of the amphipods Bathyporeia pilosa and B. sarsi, co-occurring on some beaches, were studied through monthly sampling of eight cross-shore transects along the Belgian coast (October 2003–October 2004). Their biomass and production were assessed for the first time. Abundance and biomass of B. pilosa were ten times higher along western ultra-dissipative transects than along slightly more reflective, eastern transects. For B. sarsi (less prominent), differences between the two westernmost transects (2–5× higher) and all others were observed, whereas P/B ratio was comparable for all. B. pilosa could reach two times higher abundance and biomass and higher levels of production (max B. sarsi = 7,580 g m−2 y−1; max B. pilosa = 16,040 g m−2 y−1), while the species was nearly absent from the eastern transects. Continuous reproduction and recruitment with three relative peaks of the latter (February, July, October) were observed. Fecundity showed parallel temporal variation for both species, peaking in February and September–October. Interestingly, the July relative “recruitment” peak could not be explained by relative abundance of gravid females or fecundity, but was probably caused by adult mortality. Both species displayed comparable gonad production (B. pilosa: P g  = 0.73 mg/ind year; B. sarsi: P g  = 0.71 mg/ind year), but B. pilosa produced fewer yet larger embryos. Peak abundances were found at 436 ± 25 SD cm (B. pilosa) and 357 ± 40 SD cm (B. sarsi) above MLLWS, corresponding to a 40–62 m cross-shore distance between the peaks of both species. The occupied cross-shore range was larger for B. sarsi than for B. pilosa (35–54 m), for females than for males (15–23 m), and for adults than for juveniles of B. pilosa (5–8 m). Both species displayed many comparable life history features. Differences in abundance and biomass may be related to beach morphodynamics and zonation.  相似文献

Background

Costs of adaptation play an important role in host-parasite coevolution. For parasites, evolving the ability to circumvent host resistance may trade off with subsequent growth or transmission. Such costs of virulence (sensu plant pathology) limit the spread of all-infectious genotypes and thus facilitate the maintenance of genetic polymorphism in both host and parasite. We investigated costs of three virulence factors in Puccinia striiformis f.sp. tritici, a fungal pathogen of wheat (Triticum aestivum).  相似文献

Background

Metabolically versatile soil bacteria Burkholderia cepacia complex (Bcc) have emerged as opportunistic pathogens, especially of cystic fibrosis (CF). Previously, we initiated the characterization of the phenylacetic acid (PA) degradation pathway in B. cenocepacia, a member of the Bcc, and demonstrated the necessity of a functional PA catabolic pathway for full virulence in Caenorhabditis elegans. In this study, we aimed to characterize regulatory elements and nutritional requirements that control the PA catabolic genes in B. cenocepacia K56-2.  相似文献

As part of a 3-fold approach to select potential mycoinsecticides for whitefly control, we evaluated infectivity, thermal requirements, and toxicogenic activity of the entomopathogenic fungus Beauveria bassiana (Ascomycota: Clavicipitaceae) under laboratory conditions. Twenty-five native B. bassiana isolates and a commercially available mycoinsecticide (based on B. bassiana) were evaluated for virulence to fourth instar nymphs of sweetpotato whitefly, Bemisia tabaci, and greenhouse whitefly, Trialeurodes vaporariorum, at a concentration of 1 × 107 conidia/ml. All isolates were pathogenic for both whitefly species, whereas mortality rates varied from 3 to 85%. A second series of bioassays was conducted on 10 selected isolates using four 10-fold concentrations ranging from 1 × 105 to 1 × 108 conidia/ml. Median lethal concentrations (LC50) of the four most virulent isolates varied from 1.1 × 105 to 6.2 × 106 conidia/ml and average survival time (AST) of treated nymphs from 5.9 to 7.4 days. T. vaporariorum were significantly more susceptible to all B. bassiana isolates than B. tabaci. The thermal biology of the eight most virulent isolates to both whitefly species was investigated at six temperatures (10–35 °C). The colony radial growth rate was estimated from the slope of the linear regression of colony radius on time and data were then fitted to a modified generalized β function that accounted for 90.5–99.3% of the data variance. Optimum temperatures for extension rate ranged from 23.1 to 27.1 °C, whereas maximum temperatures for fungal growth varied from 31.8 to 36.6 °C. On the basis of their virulence and thermal requirements, three isolates showed promise as candidates for whitefly management in Mediterranean greenhouses. Whilst in vitro production of macromolecular compounds toxic to Galleria mellonella larvae was not a requisite for virulence, ASTs of larvae injected with Sephadex G-25 fractions from candidate isolates ranged from 1.4 to 3.7 days compared with 5–6 days for non-toxic G-25 fractions. In addition, proteinase K treatment significantly reduced their toxic activity suggesting that they were proteins and revealing the potential of these isolates to be further improved through biotechnology to kill the pest more quickly.  相似文献

The effect of the entomopathogenic fungus Beauveria bassiana on the biological characteristics and life table of Aphidius matricariae, a parasitoid of the green peach aphid, Myzus persicae, was studied under laboratory conditions. Aphids were first infected with twice the LC95 of B. bassiana for third-instar M. persicae (2 × 108 conidia/ml). Subsequently, at different intervals they were exposed to 1-day-old mated parasitoid females for 24 h. The number of mummies produced per female and the percentage emergence of the F1 generation differed significantly as a function of the time interval between application of the fungus and exposure to the parasitoid. The interference of B. bassiana on parasitoid development was also studied by first exposing the aphid hosts to the parasitoid for 24 h and subsequently applying B. bassiana. The number of mummies produced by a female A. matricariae varied from 11.8 to 24.8 and was significantly different when the aphids were first exposed to the parasitoids and then treated with B. bassiana 24, 48, 72, and 96 h after exposure. There were no significantly different effects of B. bassiana on net reproductive rate (R0), mean generation time (T), intrinsic rate (rm) and the finite rate of increase (λ) of A. matricariae as a result of development in hosts exposed to low or high conidial concentrations (1 × 102, 2 × 108 conidia/ml). The parasitoids developed in infected hosts had lower rm, λ, T and DT (doubling time) values compared with those that developed in uninfected hosts but no differences were observed in R0 values. With proper timing, A. matricariae and B. bassiana can be used in combination in the successful biological control of M. persicae.  相似文献

Background

Staphylococcus aureus immunodominant surface antigen B (IsaB) elicits an immune response during septicemia and is generally classified as a virulence factor, but its biological function remains completely undefined. In an attempt to identify staphylococcal RNA-binding proteins, we designed an RNA Affinity Chromatography assay and subsequently isolated IsaB.  相似文献

Background

Salmonella enterica serovar Typhimurium (Typhimurium) is an important pathogen that infects a broad range of hosts. In humans, Typhimurium causes a gastroenteritis characterized by vomiting, diarrhea, and abdominal pains. Typhimurium infection occurs mainly through the ingestion of contaminated food including poultry, pork, eggs, and milk. Chickens that are asymptomatic carriers of Typhimurium constitute a potential reservoir for infection. The type three secretion systems encoded by Salmonella pathogeniCity islands (SPI) 1 and 2 are major virulence factors of Salmonella. However, only a few studies have investigated their role during the infection of chickens.  相似文献

Background

Sinorhizobium meliloti is a symbiotic nitrogen-fixing bacterium that elicits nodules on roots of host plants Medicago sativa. During nodule formation bacteria have to withstand oxygen radicals produced by the plant. Resistance to H2O2 and superoxides has been extensively studied in S. meliloti. In contrast resistance to organic peroxides has not been investigated while S. meliloti genome encodes putative organic peroxidases. Organic peroxides are produced by plants and are highly toxic. The resistance to these oxygen radicals has been studied in various bacteria but never in plant nodulating bacteria.  相似文献

Background

Endothelin-1 (ET-1) is a potent vasoactive peptide, which induces vasoconstriction and proliferation in vascular smooth muscle cells (VSMCs) through activation of endothelin type A (ETA) and type B (ETB) receptors. The extracellular signal-regulated kinase 1 and 2 (ERK1/2) mitogen-activated protein kinases (MAPK) are involved in ET-1-induced VSMC contraction and proliferation. This study was designed to investigate the ETA and ETB receptor intracellular signaling in human VSMCs and used phosphorylation (activation) of ERK1/2 as a functional signal molecule for endothelin receptor activity.  相似文献

Abstract Three populations of Culex pipiens pallens Coquillett, from Hangzhou, Linhai and Jinhua, were determined for their probability of resistant esterase phenotypes. Further, each population was selected with dipterex, chlorpyrifos, temephos and malathion at a given dose of 60%‐70% mortality and was determined for the variation of resistant esterase phenotypes in its offspring. The results indicated that esterase B1 phenotype was dominant in three populations although different natural populations have a different probability distribution of esterase B phenotypes. Insecticide selection showed that various esterase loci seemed to have different selective advantages under different insecticide pressures. The esterase B1 phenotype was selected under temephos, and the esterase B2 phenotype was selected under dipterex and chlorpyrifos. Except for the esterase B1 phenotype, the heterozygote of B1/B2 phenotypes was detected under malathion pressure. These results are discussed in relation to the selective advantages of esterase genes to OP pressure and mosquito control strategies.  相似文献

Background

Relaxed molecular clock models allow divergence time dating and "relaxed phylogenetic" inference, in which a time tree is estimated in the face of unequal rates across lineages. We present a new method for relaxing the assumption of a strict molecular clock using Markov chain Monte Carlo to implement Bayesian modeling averaging over random local molecular clocks. The new method approaches the problem of rate variation among lineages by proposing a series of local molecular clocks, each extending over a subregion of the full phylogeny. Each branch in a phylogeny (subtending a clade) is a possible location for a change of rate from one local clock to a new one. Thus, including both the global molecular clock and the unconstrained model results, there are a total of 22n-2 possible rate models available for averaging with 1, 2, ..., 2n - 2 different rate categories.  相似文献

Background

Rickettsia are intracellular symbionts of eukaryotes that are best known for infecting and causing serious diseases in humans and other mammals. All known vertebrate-associated Rickettsia are vectored by arthropods as part of their life-cycle, and many other Rickettsia are found exclusively in arthropods with no known secondary host. However, little is known about the biology of these latter strains. Here, we have identified 20 new strains of Rickettsia from arthropods, and constructed a multi-gene phylogeny of the entire genus which includes these new strains.  相似文献

Background

Non-coding satellite DNA (satDNA) usually has a high turn-over rate frequently leading to species specific patterns. However, some satDNA families evolve more slowly and can be found in several related species. Here, we analyzed the mode of evolution of the pDo500 satDNA family of Dolichopoda cave crickets. In addition, we discuss the potential of slowly evolving satDNAs as phylogenetic markers.  相似文献

Summary The falcifolia (fal) syndrome is a malformation characterized by shoot sectors with sickle-shaped leaves, which appears in hybrids between Oenothera suaveolens and O. lamarckiana and shows a non-chromosomal inheritance of a previously undescribed type. The determinants, their location in the cell, and the mechanism of their expression are unknown. The defect is the result of a cross in which mixing of two different cytoplasms occurs, without the usual predominantly maternal inheritance. F1 progeny of reciprocal crosses show a quantitative difference in the frequency and degree of expression of the fal character. When the F1 progeny are backcrossed to the parents, the percentage of fal is high in crosses to O. suaveolens and low in those to O. lamarckiana. This manner of transmission is observed regardless of whether the hybrid is used as seed or pollen parent or shows a normal or fal phenotype. F2 generations from F1 plants having either a normal or a fal phenotype always include a certain percentage of fal plants, although the latter generally produce a higher percentage of fal progeny. If a second backcross is carried out, plants that produce normal progeny on self-pollination behave differently from those that produce some fal off-spring when selfed. The latter are similar to the F1 with regard to the transmission of the fal trait. Plants of the F1B1 yielding normal progeny upon selfing produce normal progeny in the F1B2 if the parent to which they are backcrossed is the same as in the first backcross; if the parents of the first and second backcross differ, a high percentage of fal offspring is obtained. Again, whether the hybrid is used as seed or pollen parent is not relevant. Exceptions to this behaviour have been observed only rarely in that the character of the penultimate cross is retained. There is some evidence of somatic segregation of the fal determinants, since sister plants may react differently; this suggestion is supported by comparing the progenies of different branches of a self-pollinated fal plant of the F1 generation.Abbreviations F1, F2, F3, F4 First through fourth filial generation, obtained by self-pollination - F1B1 First backcross generation, i.e. the F1 was backcrossed to one of the original parents - F1B2 Second backcross generation, i.e. the F1B1 was backcrossed to one of the original parents - F1B3 Third backcross generation, i.e. the F1B2 was backcrossed to one of the original parents - (F1B1)D1 Descendants obtained by self-pollination of a F1B1 plant; further generations obtained by self-pollination are designated as D2, D3, D4 - (F1B1)D1B1 Descendant or generation obtained by a backcross of the D1 of an F1B1. Backcrosses of the D2 and D3 are designated mutatis mutandis - (F1B1)D1B2 Generation obtained by a backcross of the (F1B1)D1B1  相似文献
