Addition of the valproic acid (histone deacetylases inhibitor) to a culture of an endophytic fungus Diaporthe sp. harbored from Datura inoxia significantly altered its secondary metabolic profile and resulted in the isolation of three novel compounds, identified as xylarolide A (1), diportharine A (2) and xylarolide B (3) along with one known compound xylarolide (4). The structures of all the compounds (14) were determined by detailed analysis of 1D and 2D NMR spectroscopic data. The relative configurations of compounds 13 were determined with the help of NOESY data and comparison of optical rotations with similar compounds with established stereochemistry. All the isolated compounds were screened for antibacterial, antioxidant and cytotoxic activities. Xylarolide A (1) and xylarolide (4) displayed significant growth inhibition of MIAPaCa-2 with an IC50 of 20 and 32?µM respectively and against PC-3 with an IC50 of 14 and 18?µM respectively. Moreover, compound 1 displayed significant DPPH scavenging activity with EC50 of 10.3?µM using ascorbic acid as a positive control.  相似文献

A new coumarin, (?)-cis-(3′R,4′R)-4′-O-angeloylkhellactone-3′-O-β-d-glucopyranoside (1) and two new chalcones, 3′-[(2E)-5-carboxy-3-methyl-2-pentenyl]-4,2′,4′-trihydroxychalcone (4) and (±)-4,2′,4′-trihydroxy-3′-{2-hydroxy-2-[tetrahydro-2-methyl-5-(1-methylethenyl)-2-furanyl]ethyl}chalcone (5) were isolated from the aerial parts of Angelica keiskei (Umbelliferae), together with six known compounds: (R)-O-isobutyroyllomatin (2), 3′-O-methylvaginol (3), (?)-jejuchalcone F (6), isoliquiritigenin (7), davidigenin (8), and (±)-liquiritigenin (9). The structures of the new compounds were determined by interpretation of their spectroscopic data including 1D and 2D NMR data. All known compounds (2, 3, and 69) were isolated as constituents of A. keiskei for the first time. To identify novel hepatocyte proliferation inducer for liver regeneration, 19 were evaluated for their cell proliferative effects using a Hep3B human hepatoma cell line. All isolates exhibited cell proliferative effects compared to untreated control (DMSO). Cytoprotective effects against oxidative stress induced by glucose oxidase were also examined on Hep3B cells and mouse fibroblast NIH3T3 cells and all compounds showed significant dose-dependent protection against oxidative stress.  相似文献

Phytochemical study of the ethanol extract of the twigs of Eriosema robustum, a Cameroonian medicinal plant resulted to the isolation of two new flavones, 2′,3′,5′,5,7-pentahydroxy-3,4′-dimethoxyflavone (1) and 2′,3,5′,5,7-pentahydroxy-4′-methoxyflavone (2), along with five known compounds: 6-prenylpinocembrin (3), 1-O-heptatriacontanoyl glycerol (4), β-sitosterol (5), stigmasterol (6) and 3-O-β-d-glucopyranoside of sitosterol (7). The structure of the isolated compounds were elucidated on the basis of their NMR, UV and MS data, and by comparison with those reported in the literature. The ethanol crude extract, fractions and some isolated compounds (14) were evaluated for their radical scavenging capacity using 2,2-diphenyl-1-picryhydrazyl (DPPH). The crude extract, fraction II, the new compounds namely robusflavones A (1) and B (2) exhibited significant antioxidant activity.  相似文献

Phytochemical research on the anti-inflammatory activities of Chaenomeles speciosa (Sweet) Nakai (Rosaceae) to investigate the main components of 10% ethanol fraction of the crude extract of C. speciosa fruit in an attempt to find bioactive compounds or new compounds from this medicinal plant. The phytochemical investigation succeeded in isolating two new phenolic compounds, specpolyphenol A (1) and specphenoside A (2), together with three known phenyl glycosides (35) from the fraction. The structures of the new compounds were deduced from comprehensive spectroscopic analyses including IR, EI-MS, 1H NMR, 13C NMR, DEPT, COSY, HMBC and HMQC. The structures of the three known compounds 3, 4 and 5 were identified by comparison of their spectral data with those reported in the literature.  相似文献

A phytochemical investigation to obtain new NO inhibitors led to the isolation of nine compounds including one new guaiane-type sesquiterpenoid (1) and two new cleistanthane diterpenoids (2 and 3) from the stems of Trigonostemon howii. Their structures were elucidated on the basis of extensive 1D and 2D NMR spectroscopic data analysis, and the absolute configurations of new compounds 13 were established via comparison of experimental and calculated electronic circular dichroism (ECD) spectra. Compounds 2 and 3 possess a rare 3,4-seco-cleistanthane diterpenoid skeleton. All of the compounds showed inhibitory effects on lipopolysaccharide-induced NO production in murine microglial BV-2 cells. The further molecular docking studies indicated the strong interactions between some bioactive compounds with the iNOS protein, which revealed the possible and potential mechanism of NO inhibition of bioactive compounds.  相似文献

A chemical study of the roots of Euphorbia fischeriana resulted in the isolation of seven triterpenes (17), including two new compounds: (24R,S)-3β-24,31-epoxy-24-methylcycloartane (1) and (24R,S)-3β,31-dihydroxy-24-methoxy-24-methylcycloartane (2). Their structures were elucidated through extensive spectroscopic analyses. Cycloartanes 14 showed significant human CYP3A4 promoter activity through a series of luciferase reporter assays. Of these compounds, 3 and 4 activated the pregnane X receptor (PXR) and induced CYP3A4 mRNA expression in human primary hepatocytes. However, despite showing the most potent human CYP3A4 promoter activity via a PXR-independent pathway, 2 did not affect CYP3A4 mRNA expression in human primary hepatocytes. This difference is correlated to substitutions in C-24 and C-25 of the cycloartane structure.  相似文献

Two new cadinane-type sesquiterpenes, hypocreaterpenes A (1) and B (2), along with five known compounds (37) were isolated from a marine-derived fungus Hypocreales sp. strain HLS-104 isolated from a sponge Gelliodes carnosa. Their structures were determined by a combination of spectroscopic methods. All compounds were tested for the inhibitory effects on the nitric oxide (NO) production in lipopolysaccharide (LPS)-treated RAW264.7 cells. Among them, compounds 3 and 6 showed moderate anti-inflammatory activity with average maximum inhibition (Emax) values of 10.22% and 26.46% at 1 μM, respectively.  相似文献

Two sulfur-containing compounds, (S)-2-amino-5-((R)-1-carboxy-2-((E)-3-(4-hydroxy-3-methoxyphenyl)allylthio)ethyl-amino)-5-oxopentanoic acid (1) and (S)-2-amino-5-((R)-1-(carboxymethylamino)-3-((E)-3-(4-hydroxyphenyl)allylthio)-1-oxopropan-2-ylamino)-5-oxopentanoic acid (2), and one 1H-pyrrole-2-carboxylic acid derivative, 6-(3-(1H-pyrrole-2-carbonyloxy)-2-hydroxypropoxy)-3,4,5-trihydroxy-tetrahydro-2H-pyran-2-carboxylic acid (3), together with eighteen known phenolic compounds, were isolated from the fruits of pineapple. Their structures were elucidated by a combination of spectroscopic analyses. Some of these compounds showed inhibitory activities against tyrosinase. The half maximal inhibitory concentration values of compounds 1, 4, 5, 6, 7 are lower than 1 mM. These compounds may contribute to the well-known anti-browning effect of pineapple juice and be potential skin whitening agents in cosmetic applications.  相似文献

Two new protolimonoid compounds, namely, argentinin A (1) and B (2) along with five known triterpenoid compounds, dammar-24-en-3α-ol (3), 3-epi-cabraleahydroxy lactone (4), (E)-25-hydroperoxydammar-23-en-3β,20-diol (5), mixture of eichlerianic acid and shoreic acid (6a and 6b), and dammar-24-en-3α,20-diol (7), were isolated from the stem bark of Aglaia argentea. The structure of new compounds were elucidated by spectroscopic methods including one and two-dimensional NMR as well as high-resolution mass spectrometric analysis. All of the compounds were tested for their cytotoxic effects against P-388 murine leukemia cells in vitro. Among those isolated compounds, argentinin A (1) showed the strongest activity with an IC50 value of 1.27 μg/mL (3.05 μM).  相似文献

The first phytochemical investigation of Caesalpinia furfuracea twigs led to the isolation and identification of four new compounds including two isopimarane diterpenes, caesalfurfuric acids A (1) and B (2), and two flavans, (2R)-caesalflavans A (5) and B (6), together with four known compounds, 4-epi-isopimaric acid (3), methyl (E)-3-(3,4-dihydroxyphenyl)acrylate (4), (E)-resveratrol (7) and oxyresveratrol (8). Their structures were elucidated by intensive spectroscopic analysis. Compound 1 was found to exhibit antibacterial activity against MRSA SK1 with an MIC value of 16 μg/mL.  相似文献

Four phenolic compounds, (2R)-3-(4′-hydroxyphenyl) lactic acid, trans-cinnamic acid (2), protocatechuic acid (3) and rutin (4), and three ecdysteroids, amarasterone A1 (5), makisterone C (6) and ponastrone A (7) were isolated and identified from the young fronds of Diplazium esculentum (Retz.) Sw. (Athyriaceae). The structures of these compounds were elucidated on the basis of NMR spectral data and comparison with literature values. Two new steroidal alkaloids, (20 S)-(bennzamido)-3β-(N,N-dimethyamino)-pregnane (1), and (20 S)-(bennzamido)-pregnane-3-one- (2), and two known steroidal alkaloids, pachysanaximine A (3) and 3β, 20α-diacetamido-5α-pregnane (4) were isolated from the whole plant of Sarcococca saligna. The structures of these compounds were identified with the help of spectroscopic techniques while spectra for known compounds were compared with spectra reported in literature. The immunomodulatory potential of the new compounds were found to be significant and dose dependent. Compound 1 showed inhibition of T cells proliferation at 10 μg/mL (95%), and inhibition of IL-2 production with an IC50 = 1.6 μg/mL.  相似文献

A pair of new sesquiterpene isomers containing a spiro heterocyclic skeleton, dothimes A (1) and B (2), together with six known compounds, quindoline (3), (S)-3-(3-indolyl)lactic acid methyl ester (4), dankasterone B (5), dibutyl phthalate (6), (1S,3R,4R,7S)-3,4-dihydroxy-α-bisabolol (7), and p-hydroxybenzaldehyde (8), were isolated from the plant-derived fungus Botryosphaeria dothidea. The structures of all isolated compounds were determined based on extensive spectroscopic analyses, including 1D/2D nuclear magnetic resonance (NMR), and high resolution electrospray ionization mass spectrometry (HRESIMS) data, as well as by comparison with literature reports. Compounds 1 and 2 exhibited inhibitory effects on lipopolysaccharide (LPS)-induced nitric oxide (NO) production with IC50 values of 63.66 and 58.29 μM, respectively.  相似文献

During the course of our continuing search for biologically active compounds from Korean medicinal sources, we investigated the white flower of Impatiens balsamina. From the MeOH extract, two new phenolic compounds (12) containing a nitrile group and eleven known phenolic compounds (313) were isolated. The chemical structures of new compounds (12) were determined through NMR, HRMS, and CD data. We tested the isolated compounds (113) for their cytotoxic activities by determining their inhibitory effects on human tumor cell lines (A549, SK-OV-3, SK-MEL-2, and HCT15) in vitro using the sulforhodamine B (SRB) assay. We also investigated their neuroprotective activity by determining their effects on nerve growth factor (NGF) secretion in C6 cells, and anti-neuroinflammatory activity by measuring nitric oxide (NO) production in lipopolysaccharide (LPS)-stimulated BV-2 cells.  相似文献

A series of 2-(chloromethyl)-3-(4-methyl-6-oxo-5-[(E)-phenyldiazenyl]-2-thioxo-5,6-dihydropyrimidine-1(2H)-yl)quinazoline-4(3H)-ones 9a-j was synthesized by treating 2-(chloroacetyl)amino benzoic acid with 3-amino-6-methyl-5-[(E)-phenyldiazenyl]-2-thioxo-2,5-dihydropyrimidine-4(3H)-one 8a-j and was screened for in vitro antibacterial activities against a representative panel of Gram-positive and Gram-negative bacteria. The compounds were synthesized in excellent yields and the structures were corroborated on the basis of IR, 1H NMR, Mass and elemental analysis data. All the synthesized compounds elicited the potent inhibitory action against all the tested bacterial stains. Furthermore, in order to explore the antioxidant potential of newly synthesized compounds, the free radical scavenging activity measurement were performed by the 1,1-diphenyl-2-picryl-hydrazyl (DPPH) assay method. It is revealed from the antioxidant screening results that the compounds 9c and f manifested profound antioxidant potential.  相似文献

Three compounds, ficusamide (1), ficusoside (2) and elasticoside (3), were isolated from the bark of aerial roots of Ficus elastica (Moraceae), together with nine known compounds, including four triterpenes, three steroids and two aliphatic linear alcohols. The chemical structures of the three compounds were established by extensive 1D and 2D NMR spectroscopy, mass spectrometry and by comparison with published data. The growth inhibitory effect of the crude extract and isolated compounds was evaluated against several microorganisms and fungi. The cytotoxicity against human cancer cell lines was also assessed. Ficusamide (1) displayed a moderate in vitro growth inhibitory activity against the human A549 lung cancer cell line and a strong activity against Staphylococcus saprophyticus, while elasticoside (3) showed a potent activity on Enterococcus faecalis.  相似文献

A new prenylated xanthone, butyraxanthone F (1) together with 20 known compounds were isolated from the leaves of Pentadesma butyracea. Their identification was achieved by spectroscopic means and comparison of the data with literature or by direct comparison on TLC with authentic samples. To the best of our knowledge, this is the first report of 2-hydroxy-2,3-dihydrosqualene (17) from a natural source. Compounds 26 and 1116 were found for the first time in this genus. The antiproliferative activity of the crude extract, fractions and some isolated compounds was evaluated against three human cancer cell lines, BGC-823, Hela and A549. The crude extract, fractions B and C showed weak activities. Globuxanthone (2) and 30-epi-cambogin (7) were found to be the most active compounds on the three cancer cells. The effect of 2 on Hela cells and that of 7 on BGC-823 cells were noteworthy. The antiproliferative potential of allanxanthone A (3), ergosta-4,6,8(14),22-tetraen-3-one (14) and 2-hydroxy-2,3-dihydrosqualene (17) against BGC-823 and Hela cells could be classified as moderate-to-weak.  相似文献

An investigation of compounds extracted from Ochna schweinfurthiana roots with ethyl acetate led to the isolation of three new compounds 4⿴-methoxylophirone A (1), 4,4⿲,4⿴⿿trimethoxylophirone A (2) and (4E;7Z)-3,8-dicarboxy-1-(O-β-d-glucopyranosyl-(1 ⿿ 6)-O-β-d-glucopyranosyl-2,9-dihydroxyhexeicosa-4,7-diene (3). Six known compounds were also identified, including Calodenone (4), Calodenine B (5), Lophirone A (6), Gerontoisoflavone A(7), 16α,17-dihydroxy-ent-kauran-19-oic acid (8) and 3β-O-d-glucopyranosyl-β-sitosterol (9). This report describes the first time that compounds 4-8 have been isolated from this plant, while 8 has never been identified in the genus Ochna. Some of the isolated compounds were evaluated for their antiplasmodial activity against the chloroquine-sensitive Plasmodium falciparum strain 3D7 and antioxidant activity using DPPH radical scavenging and Ferric reducing-antioxidant power (FRAP) assays. Compound 5 exhibited prominent radical scavenging and FRAP activities, while 7 had weak activity. Compound 1 showed good in vitro anti-plasmodial activity. The structures of the isolated compounds were elucidated by spectroscopic methods and comparisons with prior data in the literature.  相似文献

The flowers of Chrysanthemum morifolium Ramat. (family: Asteraceae) are used in Traditional Chinese Medicine to treat fever, cold and swelling. However the aerial part is considered as agriculture waste and the chemical and pharmacological information is scanty. From the stems of this plant, four new compounds named as morineoliganosides A (1), B (2), C (3) and heterophyllol-1-O-β-d-glucopyranoside A (4) together with 27 known isolates (5–31), including flavonoids and caffeoylquinic acids were isolated. Their structures were elucidated by chemical and spectroscopic methods The inhibitory effects on xanthine oxidase of these compounds have been determined. This research provided a scientific development and utilization of C. morifolium stems.  相似文献

Bioactivity-guided isolation of Uvaria rufa leaves and Nephelium hypoleucum bark resulted in the successful isolation of nine compounds (19) bearing a new polyoxygenated cyclohexene scaffold: zeylenol-6-shikimate (1), (−)-zeylenol (2), microcarpin A (3), uvarigranol F (4), quercetin (5), kaempferol (6), and p-coumaric acid (7) from U. rufa, and epicatechin (8) and cinnamtannin B1 (9) from N. hypoleucum. The structures of the isolated compounds were elucidated using various spectroscopic techniques. All compounds, except for 7, exhibited weak-to-strong TRAIL-resistance-overcoming activity and an increased gastric cancer cell line (AGS) inhibition by 17–32% as compared to the treatment with the compounds alone. Compounds 3 and 9 were studied for their ability to overcome TRAIL resistance using western blot analysis, which indicated that they sensitised AGS cells to apoptosis via both extrinsic and intrinsic pathways by increasing the expression of several proapoptotic proteins (cleaved caspase-3, −8, and −9) and by decreasing the expression of anti-apoptotic protein Bcl-2.  相似文献
