人参发根的诱导及其适宜培养条件的研究   总被引:23,自引:0,他引:23  
利用发根农杆菌A4菌株在人参根外植体上直接诱导产生发根。在1/2MS固体培养基上建立起发根离体培养系,经连续多代的培养,发根仍保持旺盛生长状态。PCR扩增结果表明,发根农杆菌RI质粒的rolC基因已在人参发根基因组中整合并得到表达。液体培养基中发根生长速度约为固体培养的2倍。经对发根中人参皂苷含量及比生长速率的测定,筛选出高产发根系R9923。利用HPLC法测定了R9923发根系中单体皂苷Rg1、Re、Rf、Rb1、Rc、Rb2和Rd的含量,人参总皂苷含量达15.2mg/g。确定1/2MS培养液(30g/L蔗糖)、摇床转速110r/min、每2周更换一次培养液、继代培养时间4周,为人参发根生长适宜条件。探讨了培养容积、发根初始接种量以及分级放大培养工艺对发根大规模生产过程中生物产量和皂苷含量的影响。  相似文献

美洲商陆毛状根诱导及其离体培养的影响因素   总被引:1,自引:0,他引:1  
为了探讨利用美洲商陆毛状根生产其药用成分的可能性,研究了美洲商陆毛状根诱导及其离体培养的影响因素。结果表明,美洲商陆叶片外植体被发根农杆菌ATCC 15834感染约18 d后,从其叶片外植体形态学下端叶脉切口处产生毛状根,其中以预培养1 d,农杆菌感染20 min,共培养4 d时的毛状根诱导率最高,达到70%。PCR扩增和硅胶薄层层析结果显示,发根农杆菌Ri质粒的rol C基因以及冠瘿碱合成酶基因已在美洲商陆毛状根基因组中整合和表达。所获得的美洲商陆毛状根系都能在无外源激素的MS固体培养基上快速自主生长;其中以毛状根根系2的生长速度最快、分生侧根能力最强和根表面的根毛密度最高;毛状根根表面呈紫红色或呈白色。在供试的MS、1/2MS、B5和6,7-V液体培养基中,以无外源激素的MS培养基最适合美洲商陆毛状根根系生长。与无外源激素的MS培养基相比,6,7-V培养基更有利于毛状根中商陆皂苷甲的合成与积累。本文所建立的美洲商陆毛状根诱导及其离体培养的适宜条件为今后利用其毛状根株系的规模培养来生产其药用有效成分商陆皂苷甲奠定了实验和技术基础。  相似文献

Information concerning the sugar status of plant cells is of greatimportance during all stages of the plant life cycle. The aim of this work wasto study primary carbohydrate metabolism in hairy roots of red beet. Growth ofhairy roots of red beet in vitro and changes in concentration of major nutrientsand sugar in the media were measured over a growth cycle of 16 days. We havealso determined the levels of key enzymes in the pathways of sucrose metabolism.Sucrose concentration decreased as hairy root growth proceeded while no changein glucose and fructose levels in the medium was found during the first 3 daysindicating that external sucrose is preferably taken to the cell before it ishydrolyzed by extracellular invertase. The increase in glucose and fructoselevels in the media after 5 days of culture indicates extracellular hydrolysisof sucrose which was further supported by the activity of acid invertaseobserved during that time in the culture medium. The uptake of mineral nutrientsby hairy root of red beet was monitored continuously during the culture cycle.The preferential use of NH4 + overNO3 at the beginning of the culture andacidification of culture media were the two most notable results concerningnitrogen nutrition during hairy root growth of red beet.  相似文献

The leaves of axenically grown Coleus blumei were inoculated with Agrobacterium rhizogenes strain A4 and hairy root were established. PCR and Southern hybridization analysis confirmed transgenic nature of hairy root clones. Cultures of normal roots, induced by α-naphthaleneacetic acid on leaf explants, and hairy roots were evaluated for growth and rosmarinic acid content. Significantly better growth and up to 2.8 higher amount of rosmarinic acid was detected by HPLC analysis in hairy root clones. Methyl jasmonate stimulated rosmarinic acid accumulation in 6 out of 11 tested clones, while yeast extract induced RA accumulation in two and diminished it in 5 out of 11 tested hairy root clones.  相似文献

To better characterize the development and growth of hairy roots in a mist-fed root bed, a single root aerosol reactor was developed. Growth kinetics studies were conducted on hairy roots of Artemisia annua as a function of the mist cycle, carrier gas, and nutrient compositions. Sustained rapid growth was only observed when conditioned medium was fed to the roots. The presence of 1% CO(2) in the carrier gas did not enhance the growth kinetics but it did prevent necrosis of the tissue at the highest mist cycle.  相似文献

毛状根的构型是影响其生长速度和生物量积累的重要因素,为了规模化培养金铁锁毛状根,进一步解决金铁锁资源短缺问题,该研究以金铁锁毛状根为材料,通过改变培养基类型、碳源及碳源浓度,观察和分析了毛状根的生长状态,找出影响毛状根构型的因素。结果表明:最适合金铁锁毛状根生长的培养基为B5+蔗糖30 g·L~(-1),金铁锁毛状根主根长而粗壮,一级、二级侧根生长量大,根系表面积较大,生长效果最佳。经液体悬浮培养验证,测定毛状根的生长量,与在固体培养基培养的毛状根生长状态基本一致。通过该项研究,优化了培养基中营养成分的配比,实现了金铁锁毛状根的快速生长和生物量的积累。  相似文献

An efficient transformation system for the medicinal plant Pueraria phaseoloides was established by using agropine-type Agrobacterium rhizogenes ATCC15834. Hairy roots could be obtained directly from the cut edges of petioles of leaf explants or via callus 10 days after inoculation with the bacteria. The highest frequency of explant transformation by A. rhizogenes ATCC15834 was about 70% after infection for 30 days. Hairy roots could grow rapidly on solid, growth regulator-free Murashige and Skoog medium and had characteristics of transformed roots such as fast growth and high lateral branching. Paper electrophoresis revealed that bacteria-free hairy roots of P. phaseoloides could synthesize agropine and mannopine. The polymerase chain reaction amplification of rooting locus genes showed that left-hand transferred DNA of the root inducing plasmid of A. rhizogenes was inserted into the genome of transformed P. phaseoloides hairy roots. The content of puerarin in hairy roots reached a level of 1.190 mg/g dry weight and was 1.067 times the content in the roots of untransformed plants.  相似文献

含农杆碱型Ri质粒pRiA4b的发根农杆菌(Agrobacteriumrhizogenes)ATCC15834感染黄瓜子叶外植体5d后产生毛状根,毛状根可直接从叶片外植体叶脉处产生。感染10d后,约90%的子叶外植体产生毛状根。毛状根能在无外源生长调节剂的MS固体和液体培养基上自主生长。PCR扩增结果证实,发根农杆菌Ri质粒的rolB和rolC基因已在黄瓜毛状根基因组中整合并得到表达。挑选一无菌黄瓜毛状根系置于含不同浓度6-BA的MS培养基中悬浮培养来探讨细胞分裂素6-BA对黄瓜毛状根生长及其形态变化的影响。结果表明,细胞分裂素6-BA能促进黄瓜毛状根的生长及改变其生长形态。随着培养基中6-BA浓度的升高,黄瓜毛状根变得越来越短而粗,更少分枝。与对照相比,培养基中添加0·1~3·0mg/L6-BA能推迟毛状根最大生长峰的出现,降低其可溶性蛋白含量、SOD和POD酶活性;而黄瓜毛状根的乙烯释放高峰比其最大生长峰和SOD和POD酶活性高峰提前5天,但6-BA处理能降低毛状根的内源乙烯释放量。该结果表明细胞分裂素6-BA可能通过对乙烯生成和释放的调节来影响黄瓜毛状根的生长和形态并延缓毛状根的衰老。  相似文献

Transgenic hairy roots of Brassica napus (cv. Omega) have been developed, using Agrobacterium rhizogenes strain AR 25, for use as a model system in the investigation of physiological and morphological differences between transgenic and normal roots. The basic parameters of growth and normal or altered gravitropical behaviour of hairy roots are for the first time presented in this paper together with an ultrastructural and morphological analysis of the root statocytes. The results obtained also represented the basis for the TRANSF0RM-experiment on the IML-2 mission performed onboard the Space Shuttle Columbia. Typical hairy root traits such as hormone-autonomous growth high growth rate, lateral branching, and changed/absence of gravitropism were detected. The transformed nature of the roots was confirmed by Southern blot analyses. The gravitropical behaviour of apices from hairy root cultures of this clone has been compared with root tips from normal seedlings. While the wild type roots curved progressively with increasing stimulation angles, the transformed roots showed no curvature when stimulated at 45 degrees, 90 degrees or 135 degrees on the ground. The morphology and ultrastructure of the root tip regions were examined by light microscopy and transmission electron microscopy. At the ultrastructural level no major differences could be detected between the roots studied. There was, however, a slight reduction in the starch content of most of the amyloplasts of the transgenic root tips, and the root cap was more V-shaped in the transgenic roots than in the wild type. Preliminary results from the Shuttle experiment TRANSFORM show a random distribution of amyloplasts in the root cells of both transformed and wild type root caps after 14 h on a 1xg centrifuge followed by 37 h in microgravity.  相似文献

为了探讨利用褐脉少花龙葵毛状根来修复重金属镉(Cd)污染的可能性,采用溶液培养法研究了Cd单独及其与钙(Ca)组合对褐脉少花龙葵毛状根生长、抗氧化酶超氧化物歧化酶(SOD)和过氧化物酶(POD)活性及对Cd吸收的影响。结果表明,Cd≤50μmol/L时能促进毛状根生长,而高于100μmol/LCd则抑制毛状根生长,使其侧根根尖变褐和变短,数目减少。与对照相比,不同浓度Cd培养的毛状根可溶性蛋白含量和SOD活性先升高后逐渐下降;其丙二醛(MDA)含量显著提高;100μmol/LCd使毛状根POD活性逐渐升高,但300μmol/LCd则使毛状根POD活性逐渐降低。与对照(仅添加100μmol/L或300μmol/LCd的毛状根)相比,Cd和10~30mmol/LCaCl2组合培养使毛状根可溶性蛋白含量和MDA含量降低;但提高其SOD活性;而100μmol/LCd和10~30mmol/LCaCl2结合培养的毛状根POD活性均比对照低;而300μmol/LCd和10~30mmol/LCaCl2结合培养的毛状根POD活性则均比对照提高。原子吸收分光光度法测定结果表明,毛状根吸收和吸附的重金属Cd含量随着培养基中Cd浓度的升高而增加。但外源加入10~30mmol/LCaCl2能减少毛状根对Cd的吸收,并调节其抗氧化酶SOD和POD活性,降低其膜脂过氧化水平而解除重金属Cd对毛状根生长的抑制或毒害。  相似文献

Summary Hairy roots of Brassica napus (rape cv. Giant) were produced by cocultivating leaf and cotyledon explants with Agrobacterium rhizogenes strain A4T. The hairy roots grew prolifically on solid and in liquid media. Incorporation of ammonium sulphate or phosphinothricin (PPT) into the media reduced growth. PPT treatment reduced glutamine synthetase (GS) activity and increased the ammonia content of the hairy roots. We have found that PPT treatment also induces a loss of glutamine from the roots and this may influence root growth. To test this we grew hairy roots in a liquid medium containing 10 mM glutamine. This glutamine treatment overcame the PPT induced suppression of growth but also significantly increased GS activity, reduced ammonia accumulation and increased the levels of glutamate and asparagine.  相似文献

Picrorhiza kurroa Royle ex Benth. is an endangered plant producing various compounds of medicinal importance. Hairy roots of P. kurroa were obtained following cocultivation of shoot tip explants with Agrobacterium rhizogenes strains A 4 and PAT 405. Bacterial strain A 4 appeared to be better than the strain PAT 405 in terms of both growth of respective hairy root cultures and secondary metabolite production. The optimal growth of both the hairy root cultures occurred on half-strength semisolid medium with 3% sucrose. Picrotin and picrotoxinin from the roots of wild type field grown plants were compared with 8-week-old hairy root cultures induced by the A 4 and PAT 405 strains of A. rhizogenes. Picrotin and picrotoxinin content were evaluated in hairy root cultures as well as roots of field grown plant of P. kurroa. In terms of the production of picrotin and picrotoxinin, the A 4 induced hairy roots appeared to be a better performer than the PAT 405 induced hairy root cultures. The picrotin and picrotoxinin content was highest in 8-week-old A 4 induced hairy roots (8.8 μg/g DW and 47.1 μg/g DW, respectively). Rapid growth of the hairy roots of P. kurroa with in vitro secondary metabolite production potential may offer an attractive alternative to the exploitation of this endangered plant species.  相似文献

为了探讨外源钙对重金属镉(Cd)缓解南美蟛蜞菊Wedelia trilobata毛状根毒害的生理机理,采用溶液培养法研究了重金属Cd单独及其与Ca组合对南美蟛蜞菊毛状根生长、抗氧化酶超氧化物岐化酶(SOD)和过氧化物酶(POD)活性及对Cd2+吸收的影响。结果表明,Cd≤50μmol/L时促进毛状根生长;高于100μmol/LCd则抑制其生长,使其侧根短小,根尖变褐或变黑。与对照相比,不同浓度Cd培养的毛状根POD活性、SOD活性和MDA含量都比对照明显提高,但高于100μmol/L Cd培养的毛状根可溶性蛋白含量均比对照降低。与仅添加200μmol/L或300μmol/L Cd的毛状根相比,Cd和10~30 mmol/L Ca组合培养可促进毛状根生长,使其主、侧根变粗;提高其可溶性蛋白含量;降低其MDA含量、POD活性及SOD活性。原子吸收分光光度法测定结果表明,南美蟛蜞菊毛状根能吸收和吸附重金属Cd2+,当Cd2+浓度为100μmol/L时毛状根对Cd2+的吸收量最大,而Cd2+浓度为300μmol/L时毛状根对Cd2+的吸附量最大。外源加入10~30 mmol/L Ca2+可显著减少毛状根对Cd2+的吸收和吸附,并可调节其抗氧化酶活性,降低其膜脂过氧化水平而解除重金属镉对毛状根生长的抑制或毒害。  相似文献

Summary The effect of exogenously fed hormones on hairy root cultures of Cichorium intybus L. ev. Lucknow Local was studied. It was seen that auxin in the presence of low levels of kinetin induces rapid disorganization in hairy root cultures of C. intybus, ultimately to form suspension cultures, and this process was associated with the decrease in coumarin content in the cells. Of various treatments, it was observed that with an increase in the auxin: cytokinin ratio, the biomass decreased with the increase in disorganization index during the culture period of 28 d. The disorganization index was less when the inoculum size was enhanced to 10-fold. The total endogenous indole-3-acetic acid titers and indole-3-acetic acid oxidase activity also decreased with an increase in disorganization index, and was independent of initial inoculum size, with only a magnitude difference. The total coumarin content strictly correlated with growth in all the treatments. In contrast, exogenously supplied gibberellic acid at the 0.5 mg l−1 level enhanced growth, coumarin content, and branching patterns over the control and other treatments on day 28. The exogenously fed growth regulators had an effect on growth, auxin and coumarin biosyntheses, wherein transformed roots treated with increasing concentration of auxin to cytokinin ratios lost their ability for coumarin biosynthesis. The behavior of hairy roots from an Indian cultivar of chicory upon growth regulator treatment is discussed in terms of growth, coumarin and auxin biosyntheses.  相似文献

用发根农杆菌(Agrobacterium rhizogenes)转化药用植物青蒿(Artemisia annua L.)并建立了发根体外培养系统。Southern杂交、NPT Ⅱ酶的检测证明Ri质粒的T—DNA转移并整合到植物的核基因组上。在发根培养系统中,检测了青蒿的重要次生代谢物一青蒿素的含量,检测了不同理化因子对发根生长及青蒿素含量的影响。结果表明:光照(日光灯,12h光周期,20001x)有利于次生产物青蒿素的积累。培养基的pH值为5.4。蔗糖浓度为3%不仅促进发根的生长,而且促进青蒿素的积累。低浓度萘乙酸(NAA)对发根生长具有促进作用,但抑制青蒿素的合成。赤霉素GA,对发根的生长及次生产物的合成都具有促进作用,其最适浓度为4.8mg/L。  相似文献

Red beet hairy root cultures, obtained after genetic transformation with Agrobacterium rhizogenes, are completely heterotrophic and synthesize betalaines (BNs). Upon subjecting the hairy roots to treatments containing different sugars (3% w/v) it was found that sucrose was rapidly utilized, followed by maltose, and a very limited use of glucose, but the other hexoses – fructose, lactose, xylose and galactose or glycerol totally suppressed both growth and BN synthesis. No habituation or adaptability to maltose or glucose occurred, evidenced by the lack of growth upon re-culture in respective medium. Glycerol, was not taken up alone, but was utilized to a considerable extent in the presence of low levels of sucrose for growth only but not BN synthesis. Red beet hairy root culture did not exogenously hydrolyse sucrose to hexoses, as there were only traces of reducing sugar present in the medium soon after inoculation, without an increase later, confirmed by HPLC. There was an increase in medium osmolarity in the presence of fructose indicating the exudation of certain compounds from the roots. Red beet hairy roots appear useful as a model system to study sugar metabolism/signalling due to their sensitivity to different sugars that may directly link to morphological changes and BN synthesis.  相似文献
