Net photosynthesis and stomatal conductance were measured in attached leaves of Paphiopedilum insigne. At 20°C and a vapor-pressure deficit of 0.5 kilopascal, both net photosynthesis and stomatal conductance were light-saturated below 0.2 millimole per square meter per second, a response typical of shade plants. The absolute values of photosynthetic rate and conductance however were remarkably low, presumably reflecting an adaptation to the low-light, limited-nutrient habitat characteristic of these orchids. The leaves also showed a vapor-pressure deficit response, with net photosynthesis and conductance varying over a 2-fold range between 0.3 and 1.6 kilopascals.

These results confirm that Paphiopedilum stomata are functional. The correlation between achlorophyllous guard cells and low conductance rates, however, singles them out as an exceptional biological system, exhibiting basic differences from typical stomata in higher plants. Available evidence showing that guard-cell chloroplasts are needed to sustain high conductance rates at moderate to high irradiances indicates that the genetic changes leading to the loss of chloroplast differentiation in Paphiopedilum guard cells were not deleterious because of the low conductance rates characteristic of this genus.

Transgenic antisense tobacco plants with a range of reductions in sedoheptulose-1,7-bisphosphatase (SBPase) activity were used to investigate the role of photosynthesis in stomatal opening responses. High resolution chlorophyll a fluorescence imaging showed that the quantum efficiency of photosystem II electron transport (F(q)(')/F(m)(')) was decreased similarly in both guard and mesophyll cells of the SBPase antisense plants compared to the wild-type plants. This demonstrated for the first time that photosynthetic operating efficiency in the guard cells responds to changes in the regeneration capacity of the Calvin cycle. The rate of stomatal opening in response to a 30 min, 10-fold step increase in red photon flux density in the leaves from the SBPase antisense plants was significantly greater than wild-type plants. Final stomatal conductance under red and mixed blue/red irradiance was greater in the antisense plants than in the wild-type control plants despite lower CO(2) assimilation rates and higher internal CO(2) concentrations. Increasing CO(2) concentration resulted in a similar stomatal closing response in wild-type and antisense plants when measured in red light. However, in the antisense plants with small reductions in SBPase activity greater stomatal conductances were observed at all C(i) levels. Together, these data suggest that the primary light-induced opening or CO(2)-dependent closing response of stomata is not dependent upon guard or mesophyll cell photosynthetic capacity, but that photosynthetic electron transport, or its end-products, regulate the control of stomatal responses to light and CO(2).  相似文献

Controversies regarding the function of guard cell chloroplasts and the contribution of mesophyll in stomatal movements have persisted for several decades. Here, by comparing the stomatal opening of guard cells with (crl‐ch) or without chloroplasts (crl‐no ch) in one epidermis of crl (crumpled leaf) mutant in Arabidopsis, we showed that stomatal apertures of crl‐no ch were approximately 65–70% those of crl‐ch and approximately 50–60% those of wild type. The weakened stomatal opening in crl‐no ch could be partially restored by imposing lower extracellular pH. Correspondingly, the external pH changes and K+ accumulations following fusicoccin (FC) treatment were greatly reduced in the guard cells of crl‐no ch compared with crl‐ch and wild type. Determination of the relative ATP levels in individual cells showed that crl‐no ch guard cells contained considerably lower levels of ATP than did crl‐ch and wild type after 2 h of white light illumination. In addition, guard cell ATP levels were lower in the epidermis than in leaves, which is consistent with the observed weaker stomatal opening response to white light in the epidermis than in leaves. These results provide evidence that both guard cell chloroplasts and mesophyll contribute to the ATP source for H+ extrusion by guard cells.  相似文献

Element content and expression of genes of interest on single cell types, such as stomata, provide valuable insights into their specific physiology, improving our understanding of leaf gas exchange regulation. We investigated how far differences in stomatal conductance (gs) can be ascribed to changes in guard cells functioning in amphistomateous leaves. gs was measured during the day on both leaf sides, on well-watered and drought-stressed trees (two Populus euramericana Moench and two Populus nigra L. genotypes). In parallel, guard cells were dissected for element content and gene expressions analyses. Both were strongly arranged according to genotype, and drought had the lowest impact overall. Normalizing the data by genotype highlighted a structure on the basis of leaf sides and time of day both for element content and gene expression. Guard cells magnesium, phosphorus, and chlorine were the most abundant on the abaxial side in the morning, where gs was at the highest. In contrast, genes encoding H+-ATPase and aquaporins were usually more abundant in the afternoon, whereas genes encoding Ca2+-vacuolar antiporters, K+ channels, and ABA-related genes were in general more abundant on the adaxial side. Our work highlights the unique physiology of each leaf side and their analogous rhythmicity through the day.  相似文献

The relationship between cytokinin- and auxin-induced stomatal opening and nitric oxide (NO) levels in guard cells in broad bean was studied. Results indicate that cytokinins and auxins reduced the levels of NO in guard cells and induced stomatal opening in darkness. In addition, cytokinins not only reduced NO levels in guard cells caused by sodium nitroprusside (SNP) in light but also abolished NO that had been generated by dark, and then promoted the closed stomata reopening, as did NO scavenger 2-(4-carboxyphenyl)-4,4,5,5-tetramethylimidazoline-1-oxyl-3-oxide. However, unlike cytokinins, auxins not only had incapability to reduce NO levels by SNP but also could not abolish NO having been generated by dark, so auxins could not promote the closed stomata to reopen. The above-mentioned effects of auxins were similar to that of nitric oxide synthase (enzyme commission 1.14.13.39) inhibitor N G-nitro- l -Arg-methyl ester. Hence, it is concluded that cytokinins reduced probably the levels of NO in guard cells via scavenging, and auxins reduced NO levels through restraining NO generation in all probability, and then induced stomatal opening in darkness.  相似文献

Responses of plant leaf stomatal conductance and photosynthesis to water deficit have been extensively reported; however, little is known concerning the relationships of stomatal density with regard to water status and gas exchange. The responses of stomatal density to leaf water status were determined, and correlation with specific leaf area (SLA) in a photosynthetic study of a perennial grass, Leymus chinensis, subjected to different soil moisture contents. Moderate water deficits had positive effects on stomatal number, but more severe deficits led to a reduction, described in a quadratic parabolic curve. The stomatal size obviously decreased with water deficit, and stomatal density was positively correlated with stomatal conductance (g(s)), net CO(2) assimilation rate (A(n)), and water use efficiency (WUE). A significantly negative correlation of SLA with stomatal density was also observed, suggesting that the balance between leaf area and its matter may be associated with the guard cell number. The present results indicate that high flexibilities in stomatal density and guard cell size will change in response to water status, and this process may be closely associated with photosynthesis and water use efficiency.  相似文献

Initiation of stomatal closure by various stimuli requires activation of guard cell plasma membrane anion channels, which are defined as rapid (R)- and slow (S)-type. The single-gene loss-of-function mutants of these proteins are well characterized. However, the impact of suppressing both the S- and R-type channels has not been studied. Here, by generating and studying double and triple Arabidopsis thaliana mutants of SLOW ANION CHANNEL1 (SLAC1), SLAC1 HOMOLOG3 (SLAH3), and ALUMINUM-ACTIVATED MALATE TRANSPORTER 12/QUICK-ACTIVATING ANION CHANNEL 1 (QUAC1), we show that impairment of R- and S-type channels gradually increased whole-plant steady-state stomatal conductance. Ozone-induced cell death also increased gradually in higher-order mutants with the highest levels observed in the quac1 slac1 slah3 triple mutant. Strikingly, while single mutants retained stomatal responsiveness to abscisic acid, darkness, reduced air humidity, and elevated CO2, the double mutant lacking SLAC1 and QUAC1 was nearly insensitive to these stimuli, indicating the need for coordinated activation of both R- and S-type anion channels in stomatal closure.

Combined impairment of guard cell slow and rapid anion channels results in increased stomatal conductance and complete stomatal insensitivity to abscisic acid, darkness, and elevated CO2.  相似文献

Field studies were conducted to investigate ontogenic changes in leaf photosynthesis and chloroplast ultrastructure of a single cotton (Gossypium hirsutum L.) leaf subtending the fruit. A 20-d old leaf was the most physiologically active with net photosynthetic rate (PN) of 16.5 mol m-2 s-1 and nitrogen (N) concentration of 168 mmol m-2. These values declined with leaf age and a close relationship existed between them. Concurrent with declines in PN, ultrastructural alterations occurred in the chloroplast: the 20-d old leaf had increased grana number and thylakoids per granum and a few plastoglobuli. Afterwards, the grana number and thylakoids per granum declined with leaf age indicating disintegrated grana and stroma lamellae. Concomitant with disintegrated membrane system was the presence of numerous large plastoglobuli. The PN was closely related to grana number and thylakoids per granum suggesting that the decline in PN with leaf age was associated with ultrastructural changes in the chloroplast.  相似文献

Stomatal movement is an energetic oxygen-requiring process. In the present study, the effect of oxygen concentration on mitochondrial respiratory activity and red-light-dependent photosynthetic oxygen evolution by Vicia faba and Brassica napus guard cell protoplasts was examined. Comparative measurements were made with mesophyll cell protoplasts isolated from the same species. At air saturated levels of dissolved oxygen in the protoplast suspension media, respiration rates by mesophyll protoplasts ranged from 6 to 10μmoles O2 mg?1 chl h?1, while guard cell protoplasts respired at rates of 200–300 μmoles O2 mg chl?1 h?1, depending on the species. Lowering the oxygen concentration below 50–60 mmol m?3 resulted in a decrease in guard cell respiration rates, while rates by mesophyll cell protoplasts were reduced only at much lower concentrations of dissolved oxygen. Rates of photosynthesis in mesophyll cell protoplasts isolated from both species showed only a minor reduction in activity at low oxygen concentrations. In contrast, photosynthesis by guard cell protoplasts isolated from V. faba and B. napus decreased concomitantly with respiration. Oligomycin, an inhibitor of oxidative phos-phorylation, reduced photosynthesis in mesophyll cell protoplasts by 27–46% and in guard cell protoplasts by 51–58%. The reduction in both guard cell photosynthesis and respiration following exposure to low oxygen concentrations suggest close metabolic coupling between the two activities, possibly mediated by the availability of substrate for respiration associated with photosynthetic electron transport activity and subsequent export of redox equivalents.  相似文献

Photosynthetic carbon gain in rapidly fluctuating light is controlled by stomatal conductance, activation of ribulose-1,5-bisphosphate carboxylase-oxygenase, a fast induction step in the regeneration of ribulose-1,5-bisphosphate, and the build-up of pools of photosynthetic intermediates that allow post-illumination CO2 fixation. Experimental work over recent years has identified and characterised these factors. A physiologically-based dynamic model is described here that incorporates these factors and allows the simulation of carbon gain in response to any arbitrary sequence of light levels. The model output is found to conform well to previously reported plant responses of Alocasia macrorrhiza (L.) G. Don. observed under widely differing conditions. The model shows (i) responses of net assimilation rate and stomatal conductance to constant light levels and different CO2 concentrations that are consistent with experimental observations and predictions of a steady-state model; (ii) carbon gain to continue after the end of lightflecks, especially in uninduced leaves; (iii) carbon gain to be only marginally reduced during low-light periods of up to 2 s; (iv) a fast-inducing component in the regeneration of ribulose-1,5-bisphosphate to be limiting for up to 60 s after an increase in light in uninduced leaves: the duration of this limitation lengthens with increasing CO2 concentration and is absent at low CO2 concentration; (v) oxygen evolution to exceed CO2 fixation during the first few seconds of a lightfleck, but CO2 fixation to continue after the end of the lightfleck whereas oxygen evolution decreases to low-light rates immediately. The model is thus able to reproduce published responses of leaves to a variety of perturbations. This provides good evidence that the present formulation of the model includes the essential rate-determining factors of photosynthesis under fluctuating light conditions. Summary Ramets from stem cuttings of three populations of Populus deltoides Bartr. from Wisconsin, Illinois, and Louisiana representing a latitudinal gradient were grown in pots outdoors at Urbana, Illinois and brought indoors for growth chamber studies. Leaf resistance and photosynthetic response to low night temperatures of 4° and 10° C were determined relative to 20° C controls for plants measured over one growing season. Plants from Louisiana, where nights are warm, reacted to cool nights of 4° and 10° C by opening their stomata slower upon illumination the following day than those from farther north where nights are cooler. The optimum night temperature for rate of opening was lower in the Wisconsin population than in populations from farther south. The Wisconsin population showed more ideal homeostasis of photosynthesis at different temperatures than the southern population which exhibited greater plasticity. No seasonal differences in these relationships were apparent other than at the time of leaf senescence.As plants approached senescence, which occurred earliest in the Wisconsin population, leaf resistance increased and photosynthesis declined, but stomata still retained their functional ability to respond to changes in night temperature. The change in leaf resistance, noted in the Wisconsin population, was related more to closure of lower-leaf surface stomata than upper. Only the Louisiana population had significantly more stomata on the lower than upper leaf surface.  相似文献

Recent studies have demonstrated that hydrogen sulfide (H2S) produced through the activity of l -cysteine desulfhydrase (DES1) is an important gaseous signaling molecule in plants that could participate in abscisic acid (ABA)-induced stomatal closure. However, the coupling of the DES1/H2S signaling pathways to guard cell movement has not been thoroughly elucidated. The results presented here provide genetic evidence for a physiologically relevant signaling pathway that governs guard cell in situ DES1/H2S function in stomatal closure. We discovered that ABA-activated DES1 produces H2S in guard cells. The impaired guard cell ABA phenotype of the des1 mutant can be fully complemented when DES1/H2S function has been specifically rescued in guard cells and epidermal cells, but not mesophyll cells. This research further characterized DES1/H2S function in the regulation of LONG HYPOCOTYL1 (HY1, a member of the heme oxygenase family) signaling. ABA-induced DES1 expression and H2S production are hyper-activated in the hy1 mutant, both of which can be fully abolished by the addition of H2S scavenger. Impaired guard cell ABA phenotype of des1/hy1 can be restored by H2S donors. Taken together, this research indicated that guard cell in situ DES1 function is involved in ABA-induced stomatal closure, which also acts as a pivotal hub in regulating HY1 signaling.  相似文献

High concentrations of orthophosphate (Pi) inhibited CO2-dependent O2 evolution and prevented the inactivation of glucose-6-P dehydrogenase by light in intact spinach and barley chloroplasts. Addition of glycerate-3-P to chloroplasts inhibited by Pi in the light, induced O2 evolution and caused rapid inactivation of glucose-6-P dehydrogenase. High resolution chlorophyll a fluorescence imaging was used to compare the photosynthetic efficiency of PSII electron transport (estimated by Fq'/Fm') in guard cell chloroplasts and the underlying mesophyll in intact leaves of six different species: Commelina communis, Vicia faba, Amaranthus caudatus, Polypodium vulgare, Nicotiana tabacum, and Tradescantia albifora. While photosynthetic efficiency varied between the species, the efficiencies of guard cells and mesophyll cells were always closely matched. As measurement light intensity was increased, guard cells from the lower leaf surfaces of C. communis and V. faba showed larger reductions in photosynthetic efficiency than those from the upper surfaces. In these two species, guard cell photosynthetic efficiency responded similarly to that of the mesophyll when either light intensity or CO2 concentration during either measurement or growth was changed. In all six species, reducing the O2 concentration from 21% to 2% reduced guard cell photosynthetic efficiency, even for the C4 species A. caudatus, although the mesophyll of the C4 species did not show any O2 modulation of photosynthetic efficiency. This suggests that Rubisco activity is significant in the guard cells of these six species. When C. communis plants were water-stressed, the guard cell photosynthetic efficiency declined in parallel with that of the mesophyll. It was concluded that the photosynthetic efficiency in guard cells is determined by the same factors that determine it in the mesophyll.  相似文献

Abscisic acid (ABA) induces a transient stimulation of 86Rb+ efflux from isolated guard cells of Commelina communis L. The form of the efflux transients produced in suboptimal conditions (low concentrations of ABA and/or high external pH at which ABA will penetrate poorly) has been compared with the full transient. In suboptimal conditions the stimulation of efflux is both delayed and reduced. The pH-dependence of the delay before initiation of the efflux transient suggests that a threshold internal concentration of ABA is required. However in suboptimal conditions even when the threshold internal concentration is reached and a transient is triggered, the degree of stimulation is reduced, an effect which also appears to depend on internal ABA. It is suggested that the differences reflect activation of different numbers of tonoplast ion channels for release of vacuolar ions. By contrast, the same end-state seems to be reached in optimal and suboptimal conditions, but after different times. The relative efflux stimulation during the efflux transient tracks the declining ion content; both the peak and the end of the transient are reached at the same ion content, but at different times. It is suggested that this reflects an ABA-induced change in the set-point of a regulated ion channel which is sensitive to ion content, perhaps a stretch-activated channel. This effect is independent of external concentration in the range 0.1–10 µM, and pH 6 and pH 8 are equally effective, suggesting an external site of action. Thus the results suggest multiple actions of ABA, involving both internal and external receptors. Regulation of both tonoplast ion channels by internal ABA, and of a regulated channel responsive to ion content by external ABA are suggested.  相似文献

Rubisco activity decreases under water stress, for reasons as yet unclear. Here, the covariation of stomatal conductance (gs) and relative water content (RWC), often observed during water stress, was impaired to assess the separate effects of these factors on Rubisco activity. Three different treatments were applied to soybean (Glycine max) and tobacco (Nicotiana tabacum): leaf desiccation (LD), in which stomatal closure was accompanied by large decreases of RWC; water stress (WS), in which minor decreases of RWC were observed along with stomatal closure; and exogenous application of abscisic acid (ABA), which triggered stomatal closure without changing RWC. Decreased RWC did not induce decreased initial Rubisco activity, which was impaired only in soybean by 40% when the gs dropped below 50 mmol m(-2) s(-1), regardless of the treatment. The mechanism for decreased activity differed among treatments, owing to decreased activation in LD and to total activity and protein content in WS and ABA. Despite the occurrence of Rubisco regulation, CO2 availability in the chloroplast, not impairment of Rubisco activity, limits photosynthesis during WS.  相似文献

Actin filaments in guard cells and their dynamics function in regulating stomatal movement. In this study, the array and distribution of actin filaments in guard cells during stomatal movement were studied with two vital labeling, microinjection of alexa-phalloidin in Vicia faba and expression of GFP-mTn in tobacco. We found that the random array of actin filaments in the most of the closed stomata changed to a ring-like array after stomatal open. And actin filaments, which were throughout the cytoplasm of guard cells of closed stomata (even distribution), were mainly found in the cortical cytoplasm in the case of open stomata (cortical distribution). These results revealed that the random array and even distribution of actin filaments in guard cells may be required for keeping the closed stomata; similarly, the ring-like array and cortical distribution of actin filaments function in sustaining open stomata. Furthermore, we found that actin depolymerization, the trait of moving stomata, facilitates the transformation of actin array and distribution with stomatal movement. So, the depolymerization of actin filaments was favorable for the changes of actin array and distribution in guard cells and thus facilitated stomatal movement.  相似文献
