Abstract.  The effect of humidity on the activity of Metarhizium anisopliae IP 46 (Metsch.) Sorokin (Hypocreales: Clavicipitaceae) formulated in sunflower oil against Aedes aegypti (L.) (Diptera: Culicidae) eggs was examined. After exposure of eggs at 75% relative humidity (RH) for ≤ 25 days, ovicidal activity was not increased by oil-in-water formulated conidia, hyphal bodies or pure-oil formulated conidia, compared with conidia or hyphal bodies prepared in water only. At optimal > 98% RH, eclosion was ≤ 13.7% after treatment with oil-in-water formulated propagules in ≤ 10% oil, and it was completely inhibited when conidia were applied in pure oil. At 86–100% RH, new conidia were found on eggs treated with oil-formulated conidia and incubated down to 91% RH. Ovicidal activity was still detected at 93% RH and was augmented with increasing humidity and time of exposure of eggs. Eclosion of larvae was distinctly reduced by IP 46 pure-oil formulated conidia after a minimal initial exposure of 3 days at > 98% RH, followed by: (a) a 12-day exposure at 75% RH before submersion in water; (b) a minimal 5-day exposure at > 98% RH and direct subsequent transfer of treated eggs to water, or (c) a minimal daily 20-h exposure at > 98% RH alternating with 4 h at 75% RH for 10 days. We demonstrate that oil-based formulations of conidia of M. anisopliae enhance ovicidal activity at high humidities and conclude that these formulations have potential in the integrated control of Ae. aegypti .  相似文献

The effect of both moisture and temperature on the infective potential of Beauveria bassiana to the Chagas' disease vector, Rhodnius prolixus, was studied under fluctuating regimes. At constant 25 degrees C, contaminated first-instar nymphs exposed to increasing daily periods of initial exposure to 97% RH, followed by transfer to reduced humidity (43, 53, 75, and 86% RH), showed a significant reduction in mortality when the 97% RH exposure time declined from 12 to 8 h per day. The duration of disease incubation depended on the daily 97% RH exposure time. Under fluctuating regimes of both humidity (97% RH versus 75% RH) and temperature (15/28, 20/25, 25/28, and 25/35 degrees C), first-instar mortality was affected by weather conditions, daily 97% RH exposure time (8, 12, and 16 h per day), and number of temperature and humidity fluctuations before transferring tested insects to constant unfavorable conditions. In most cases, at 12/12 h alternating cycles, high and rapid mortality required five cycles. Under these fluctuating regimes, fungus-induced mortality and mortality time were similarly affected in third- and fifth-instar nymphs by the daily 97% RH exposure time. Despite a lower susceptibility of older larval stages, mortality rates in insects exposed for at least 12 h per day at 97% RH remained very high except at 15 degrees C. Moisture and temperature regimes at 12/12 h cycling significantly affected the dose-mortality response in first-instar nymphs. The most favorable conditions consisted of 97%-20 degrees C combined with either 75%-25 degrees C or 43%-25 degrees C. Under less favorable alternating conditions (lower and higher temperatures) the amounts of inoculum required for killing 50% of first-instar nymphs were 10 or 20 times higher. From a vector control standpoint, daily high humidity appears to be the most crucial climatic constraint. B. bassiana has the potential to control R. prolixus populations with applications made during the rainy seasons when humidity is high.  相似文献

Foraging groups of Formosan subterranean termites, Coptotermes formosanus Shiraki were tested for their relative humidity (RH) preference in a humidity gradient arena in the laboratory at a constant temperature of 26°C. Five RH levels (9%, 33%, 53%, 75%, and 98%) were maintained in the test arena comprising of a series of closed containers by using dry silica gel, saturated salt solutions, or distilled water alone. Termites gradually aggregated to the highest RH chamber in the arena. After 1 h, a significantly greater percentage of termites (≈46%) aggregated to the highest RH chamber (98%) than to the lower RH chambers (≤75%). After 12 h, > 97% of the termites aggregated to the 98% RH chamber. In survival tests, where termites were exposed to 15 combinatorial treatments of five RH levels (9%, 33%, 53%, 75%, and 98%) and three temperatures (20°C, 28°C, and 36°C) for a week, the survival was significantly influenced by RH, temperature, and their interaction. A significantly higher mortality was observed on termites exposed to ≤75% RH chambers than to 98% RH chamber at the three temperatures and significantly lower survival was found at 36°C than at 28°C or 20°C. The combination of temperature and RH plays an important role in the survival of C. formosanus.  相似文献

Experiments were conducted on laboratory populations of Eriworm, Philosamia ricini Boisduval to study the effect of temperature and humidity on development, growth, survival and oviposition. Experiments were performed at four different temperatures (22 +/- 2 degrees C, 26 +/- 2 degrees C, 28 +/- 2 degrees C and 34 +/- 1 degrees C) each with three different humidities (50%, 70% and 90% relative humidity). Shortest developmental period (42 days) was observed in 28 +/- 2 degrees C and 70% RH. Longest developmental period was (63.6 days) observed in 22 +/- 2 degrees C at 50% RH. Highest larval weight (9.1 g) was found in 28 +/- 2 degrees C at 70% RH. Best pupal weight was observed in 26 +/- 2 degrees C at 70% RH. Weight of silk material was found to be maximum in 26 +/- 2 degrees C at 70% RH. Survival was best in 28 +/- 2 degrees C and 26 +/- 2 degrees C at 70% RH. Oviposition was found to be highest in 28 +/- 2 degrees C at 70% RH.  相似文献

从产自美国加利福尼亚州的新鲜脐橙样品中发现多个腐烂病果,通过分离培养得到3个疑似丁香疫霉Phytophthora syringae菌株,对3个菌株进行形态学研究、致病性测定和分子序列比对分析。结果表明病菌在V8A培养基上菌落稀疏、平铺,呈星状,菌丝紧贴培养基生长或埋于基质内生长;在PDA培养基上菌落呈菊花花瓣状,菌丝致密,乳白色;游动孢子囊和菌丝膨大体在无菌水和土壤浸出液中黑暗条件下48h后产生;菌株为同宗配合,卵孢子在带有新鲜脐橙果实组织或杜鹃叶片的V8A培养基中大量产生;创伤接种脐橙果实,7d后接种脐橙出现典型的褐腐症状;通用引物ITS1/ITS4扩增测序,Blastn分析表明序列与GenBank中P. syringae序列相似性为99%。依据上述研究结果,将分离获得的3株菌鉴定为丁香疫霉Phytophthora syringae,系国内首次截获的一种植物检疫性真菌病害。  相似文献

Sporangia of three isolates of Phytophthora ramorum representing three different clonal lineages were subjected to relative humidity (RH) levels between 80 and 100% for exposure periods ranging from 1 to 24 h at 20°C in darkness. Plastic containers (21.5 × 14.5 × 5 cm) were used as humidity chambers with 130 ml of glycerine solution added to each container. Glycerine concentrations corresponded to 100, 95, 90, 85 and 80% RH based on refractive index measurements. Sporangia suspensions were pipeted onto nitrile mesh squares (1.5 × 1.5 cm, 15 micron pore size) which were placed in the humidity chambers and incubated at 20°C in darkness. Following exposure periods of 1, 2, 4, 8, 12 and 24 h, mesh squares were inverted onto Petri dishes of selective medium and sporangia germination assessed after 24 and 48 h. At 100% RH, we observed a mean value of 88% germination after 1 h exposure declining to 18% germination following 24 h incubation. At 95% RH, a steeper decline in germination was noted, with means ranging from 79% at 1 h to less than 1% at 24 h exposure. At 90% RH, no germination was noted after 8 or more h exposure, and values were 57%, 22% and 3% germination for the 1, 2 and 4 h exposures, respectively. Germination was only observed at 1 h exposure for both the 85% RH treatment (52% germination) and the 80% RH treatment (38% germination). The three isolates responded similarly over the range of RH values tested. The germination response of P. ramorum sporangia to RH values between 80% and 100% was comparable to that reported for other Phytophthora species. Knowledge of conditions that affect Pramorum sporangia germination can shed light on pathogenesis and epidemic potential and lead to improved control recommendations.  相似文献

用飞虱虫疠霉(Pandora delphaics)“孢子浴”接种的桃蚜(Myzus persicae)无翅成蚜在离体甘蓝菜叶片(65cm^2)上建立蚜群,在不同温度(10-30℃)和湿度(74%-100%RH)的组合条件下任其繁衍,发病和交互感染,以评价该菌的控蚜效果。在25个温,湿度组合处理(8次重复,每重复含3头接种成蚜)中,蚜群均不同程度的发病死亡,在历时30d的观察中,以高温(20-30℃),高湿(95%RH)组合条件下的蚜群发病快且死亡率高,蚜尸上产生的孢子有效地引起若蚜继发性感染。与相同温度下不带菌的对照蚜群相比,次于30℃下,各湿度除个别例外,第8d的控蚜率达30%以上,第20d达80%以上。在10℃和15℃下,控蚜效果一般不如上述较高温度下,且与湿度的关联程度相对较低,但最大控蚜效果均发生在100%RH处理中。结果表明,飞虱虫疠霉用于蚜虫防治的潜力很大,值得深入研究和开发利用。  相似文献

The effects of temperature on conidial germination and susceptibility of adults of the desert locust , Schistocerca gregaria, to four isolates of Metarhizium flavoviride were determined . In addition , the effects of inoculation method (topical or spray) , spore carrier (oil or water) and ambient relative humidity (RH) on susceptibility of the locust to the most thermotolerant isolate (Mf324) were investigated . There were differences among the isolates in the effects of temperature on germination of conidia after a 24 - h incubation period . Over 90% of conidia of all isolates germinated after 24 h at 30 o C . In contrast , at 40 o C , none of the isolates germinated for up to 72 h . However , there were differences in germination between the isolates at 35 o C . Locust mortality and disease progression were significantly affected by temperature . At both 25 and 30 o C , all isolates induced 98 - 100% mortality within 8 days; however , there were differences between isolates at 35 o C . None of the isolates caused significant mortality at 40 o C . Humidity and inoculation method had no influence on levels of mortaility in fungus treated locusts . In contrast , carrier type significantly affected cumulative mortality . Topical oil treatment resulted in higher overall mycosis than the three other treatments . Control mortality on the other hand was strongly affected by inoculation method and to a lesser extent by humidity and carrier . In these conditions , application of oil by spray was generally toxic at all humidities whereas topical application of water was most toxic at near saturation . The results of these laboratory studies demonstrate the importance of strain selection , formulation and application method in the development of a microbial control agent against the desert locust . Low RH should not impede use of this fungus under dry conditions .  相似文献

Phytophthora ramorum, recently found in the US, is causing concern for hardwood forests and the nursery industry. In an effort to identify some of the environmental limitations to growth and sporulation we undertook a laboratory study of four US and three European (EU) isolates. On V8 media, isolates grew when incubated at 2-28 C and produced chlamydospores at 8-28 C. Sporangia were produced at all temperatures tested: 10-30 C for US isolates and 6-26 C for EU isolates. Optimal temperatures were 16-26 C for growth, 14-26 C for chlamydospore production and 16-22 C for sporangia production. US isolates grew less and produced fewer spores when exposed to increasing doses of near-UV radiation (50-300 microW/cm(2)) and visible radiation (250-1500 microW/cm(2)). EU isolates were exposed to 300 microW/cm(2) near-UV only, which significantly reduced growth of one of three isolates and had no significant effect on spore production. In our studies P. ramorum tolerated a broad range of temperature and light conditions, which suggests that it is capable of establishment in a wide geographic area.  相似文献

Individual blastospores of Candida albicans were deposited on the surface of 50-mm membranes (Millipore Corp.) and placed within sealed glass chambers at various relative humidities (RH). After 48 hr, virtually all cells maintained at 100 and 10% RH had survived, but 84% of the cells maintained at 60% RH failed to develop into colonies when transferred to Sabouraud medium. No morphological abnormalities could be observed in cells surviving low RH values, but their initial rate of multiplication after transfer to Sabouraud medium was greatly reduced, compared to that demonstrated by cells maintained at 100% RH. At 60% RH, the exposure time required to kill 50% of the blastospores was 2 to 3.5 days. The inimical effect of 60% RH was confirmed in a total of 21 isolates of C. albicans. No deleterious effect was noted when 12 other species of yeasts were subjected to 10, 60, and 100% RH. The single isolate of Candida brumptii and 1 out of the 20 isolates of Cryptococcus neoformans tested also failed to grow after blastospores had been exposed to 60% RH for 4 days.  相似文献

Nondiapausing pupae ofBathyplectes curculionis (Thomson) were studied under laboratory conditions. The mortality caused by 8 temperatures between 25–48°C at 20% and 70% relative humidity was measured at 10 exposure times between 15 min-24 h. There was no significant mortality at 25°C. Between 30 and 40°C, mortality occurred from long exposures only, with lethal effects becoming greater at each increase in temperature. At 43°C mortality occurred from relatively short exposures, with 100% at 4 h. Exposure times for 50% mortality averaged 16.58 h at 38°C, 1.08 h at 43°C and 0.31 h at 48°C. A slightly higher mortality occurred at 20% relative humidity than at 70% at temperatures between 35 and 40°C. At temperatures above 43°C no effects of relative humidity were noted. Afternoon soil surface temperatures in recently cut alfalfa fields commonly exceeded 50°C during July in northern Utah.  相似文献

Large populations of Pratylenchus thornei, a winter pest of cereals, legumes, and potatoes in the northern Negev region of Israel, survive 7-8 months of summer drought and return to full activity at the beginning of the rainy season. To demonstrate that it survives the summer in an anhydrobiotic state, all developmental stages of P. thornei were exposed to gradually reduced relative humidity (RH) using glycerin water solutions. At 97.7% RH the nematodes were coiled and able to survive exposure to 0% RH. About 40% of artificially desiccated nematodes could be reactivated by gradually increasing the humidity to the final water environment. Desiccated nematodes could withstand temperatures up to 40 C. Reactivated individuals showed intestines apparently devoid of reserve materials. Only 3% survived three cycles of desiccation and reactivation. P. thornei reactivated after anhydrobiosis multiplied twice as much within Vicia sativa roots as did fresh nematodes.  相似文献

1. 1. The mean durations of development in the pupae of Drosophila melanogaster (Meigen) and their survival were measured at combinations of six constant temperatures (15, 20, 22.5, 25, 27.5 and 30°C) and up to 11 levels of relative humidity. The thermal survival range for the pupae is between 15 and 30°C, and the humidity viable range is between 60 and 100% RH.
2. 2. The percentage water loss of the pupae was measured at six constant temperatures and four levels of relative humidity. There was a rapid increase in the percentage of water lost during the first 24 h exposure at all tested conditions. However, pupae reared at 100% RH at each constant temperature, sustained the lowest water loss. The percentage water loss increased as temperature increased, as humidity decreased and also with time.
3. 3. The duration of larval development studied at six constant temperatures (15, 20, 22.5, 25, 27.5 and 30°C) was inversely related to temperature. A wide range of alternating temperature regimes had a small, though statistically significant, accelerative effect on larval developmental time. Thus, the present results may be used as a basis for modelling development under changing temperatures, with the assumption that the developmental rate is nearly identical to that from a series of constant temperatures.

研究了早稻收获和储藏期间稻水象甲Lissorhoptrus oryzophilus Kuschel一代成虫的数量动态及其存活时间与温湿度的关系。早稻收割时尚未迁离稻田的象甲中,仅3~13%的个体随脱落操作落入稻谷中,且这些个体经太阳曝晒2天后存活率不足1%;收割时残留于稻草中的虫量为稻谷中的5.1~12.3倍。储藏1个月的稻谷中所有成虫均死亡,但储藏相同时间的秕谷和稻草中存活成虫分别达到250头/50 kg秕谷和17头/50 kg稻草。成虫在≥90% RH下的存活时间显著长于35~75%RH下,在玻璃管中的存活时间显著长于稻谷中。在稻谷中,温度对成虫存活时间的影响不明显,但在玻璃管中,20℃(恒温)下的存活时间显著长于27.0~33.2℃(室温)下。利用生存分析的指数模型分析了成虫存活与湿度、温度和存放基质的关系,表明在75~98% RH之间,湿度越低则成虫死亡风险越大,较高温度(27.0~33.2℃)和处于稻谷中其死亡风险亦高。这些结果为开展稻谷中稻水象甲的检疫提供了依据。  相似文献

Many metabolic and developmental processes in fungi are controlled by biological rhythms. Circadian rhythms approximate a daily (24 h) cycle and have been thoroughly studied in the model fungus, Neurospora crassa. However relatively few examples of true circadian rhythms have been documented among other filamentous fungi. In this study we describe a circadian rhythm underlying hyphal melanization in Cercospora kikuchii, an important pathogen of soybean. After growth in light or light : dark cycles, colonies transferred to darkness produced zonate bands of melanized hyphae interspersed with bands of hyaline hyphae. Rhythmic production of bands was remarkably persistent in the absence of external cues, lasting at least 7 d after transfer to darkness, and was compensated over a range of temperatures. As in N. crassa, blue light but not red light was sufficient to entrain the circadian rhythm in C. kikuchii, and a putative ortholog of white collar-1, one of the genes required for light responses in N. crassa, was identified in C. kikuchii. Circadian regulation of melanization is conserved in other members of the genus: Similar rhythms were identified in another field isolate of C. kikuchii as well as field isolates of C. beticola and C. sorghi, but not in wild-type strains of C. zeae-maydis or C. zeina. This report represents the first documented circadian rhythm among Dothideomycete fungi and provides a new opportunity to dissect the molecular basis of circadian rhythms among filamentous fungi.  相似文献

The hygropreference of adult Onychiurus arcticus (Tullberg) was investigated over 2 h at 0, 10 and 20°C, along humidity gradients (12–98% RH) established using different salt solutions. At all temperatures O. arcticus preferred the highest humidity (98% RH). At 0 and 20°C, saturated conditions were preferred to 98% RH. The hygropreference of the mite Lauroppia translamellata (Willmann) was also assessed at 20°C, and no clear RH preference was observed. This species survived the loss of 24.9 ± 2.1% of its initial water content when held for 24 h at 20°C and 12% RH. A range of assays designed to eliminate the influence of thigmotactic behaviour and population clumping permit exclusion of these factors as being responsible for the observed results. The mean initial water content of O. arcticus samples (71.7 ± 10.9, 73.4 ± 4.0 and 73.8 ± 23.5% at 0, 10 and 20°C, respectively) did not differ significantly between temperatures, indicating that the results were not influenced by differences in initial hydrated state. The percentage water loss of individuals within the gradient increased with temperature, and differed significantly between regimes. The ecological significance of the observed humidity preferences are discussed.  相似文献

本文报道细脚拟青霉(paecilomyces tenuipes)不同田间分离菌株单孢子后代间的异核现象。用来自荣园、菜地、水稻田三种生境的四个菌株(803、2801、1401和3101)的单孢子培养后代,在加有酵母膏和麦芽糖的改良萨氏培养基上进行配接实验,仅在803与1401两菌株间能形成异核体,其频率为13.5%。在配接实验中两亲和菌落交界处长出白色致密的菌丝簇组成的实线可推测为异核体。从来自菌丝簇组线的每个单菌丝尖端培养物中分离出30个以上的单孢子,井分别移接到 PDA 平板上。将来源于单孢子的菌落与两亲本菌落进行比较,有三个单菌丝尖端培养物(C_3、B_3、F_3)重现了两亲本类型或出现了新的菌落类型,从而异核现象得到证实。  相似文献

The effects of relative humidity (RH) and temperature on the sporulation of Metarhizium anisopliae var. acridum on mycosed cadavers of desert locust, Schistocerca gregaria, were assessed in the laboratory. Quantitative assessments of conidial production over 10 days under constant conditions showed that sporulation was optimized at RH > 96% and at temperatures between 20 and 30 degrees C. Under both these conditions >10(9) conidia/cadaver were produced. At 25 degrees C, conidial yield was maximized under conditions in which cadavers remained in contact with damp substrate. Relatively little sporulation occurred at 15 degrees C (< 3 x 10(7) conidia/cadaver) and 40 degrees C (< 4 x 10(6) conidia/cadaver) and no sporulation occurred at 10 or 45 degrees C. Following incubation, conidial yield was closely related to the water content of locust cadavers. In separate tests, locust cadavers were incubated for 10 days under diurnally fluctuating temperature and RH that comprised favorable (25 degrees C/100% RH) alternating with unfavorable (40 degrees C/80% RH) conditions for sporulation. In this case, fewer conidia were produced compared with cadavers that were incubated under the favorable conditions for an equal period cumulatively but were not periodically exposed to unfavorable conditions. However, this reduced sporulation observed with the fluctuating condition was not observed when cadavers were similarly incubated under favorable/unfavorable conditions of temperature but were not periodically exposed to the low RH condition. This result implies that sporulation is a dynamic process, dependent not only on periodic exposure to favorable RH but also on the interrelation of this with low RH. Associated tests and the monitoring of changes in cadaver weights imply that the mechanism driving the reduced sporulation under fluctuating RH is the net water balance of cadavers, i.e. the cumulative ability of the fungus/cadaver to adsorb water necessary for sporulation at high RH is restricted by water loss associated with intermittent exposure to a low RH. The duration of daily exposure to high humidity appears to be a crucial constraint to the recycling ability of M. anisopliae var. acridum.  相似文献

The effects of both moisture and temperature on the sporulation of an isolate of Beauveria bassiana , highly virulent to the Chagas' disease vector, Rhodnius prolixus , were studied in the laboratory under fluctuating regimes. Quantitative assays for conidial production from funguskilled cadavers from 10-day-old tests showed that recycling was highly dependent on the duration of exposure to high relative humidity (RH). On first-instar larval cadavers, sporulation reached more than 10 5 conidia/nymph under most regimes with a favourable phase combining high humidity (97% RH) and temperatures of 20 or 25 C for at least 12 h/day. Sporulation on third- and fifth-instar nymphs required a longer period (16 h/day or more) of favourable conditions (97% RH and 25 C) per day. The intensity of the conidial production from Rhodnius cadavers was enhanced when diurnal temperatures were high (28 and 35 C). From a vector control standpoint, daily high humidity requirements appear to be a crucial constraint. The recycling ability of B. bassiana on fungus-killed R. prolixus is most likely to contribute to the regulation of this triatomine vector if applications are made during the rainy seasons and in the most favourable habitats.  相似文献

Epizootiological features of Zoophthora anhuiensis, a fungal pathogen specific to aphids in southern China, were studied in six aptera colonies of Myzus persicae at 16 regimes of temperature (T = 10, 15, 20 and 25 degrees C) and relative humidity (H = 90%, 95%, 98% and 100% RH) with initially infected proportion (Ip) of 0.5 in experiment (Expt) 1 or at a fixed regime of 15 degrees C and 100% RH with a variable Ip of 0.17-1.00 in Expt 2. Mycosis-caused mortalities (Mp) varied with aphid densities (D) over time after colony initiation (t) were well fitted to a Gompertz growth model modified to include the variables T, H, Ip and D in the form of Mp = 91.72exp[-5.282exp[-(0.0095T + 0.0128H/T-0.5407D2/H)t]] for Expt 1 (r2 = 0.94) or Mp = 95.49exp[-4.314exp[-(0.1479Ip + 0.1636/D)t]] for Expt 2 (r2 = 0.97). These variables were effective in determining the rate of epizootic development in the M. persicae colonies. Based on the observed and fitted results, Z. anhuiensis was most adaptive to cool climate or season and epizootiologically was subject to a minimal effect of RH at an optimum of approximately 15 degrees C. However, the humidity became determinant for epizootic progress at higher temperatures. Interactive effects of the variables and application of the fitted models for predicting epizootic development in field situations are discussed.  相似文献
