55 accessions of wild peanuts (Arachis spp.) introduced from South America were analyzed for seed storage protein composition using SDS-PAGE electrophoresis. The objectives of the study were to evaluate variability within sect.Arachis and to classify taxa based on protein composition. 25 different band positions were resolved. Individual accessions had 11 to 18 bands which included the conarachin region (MW > 50 kD), two to five bands in the acidic arachin region (MW 38–49.9 kD), three to seven in the intermediate MW region (23 to 37.9 kD), two to five bands in the basic arachin region (18–22.9 kD), and one to three bands in the low MW protein region (14–17.9 kD). These data were utilized in a principal coordinate analysis based on the matrix of genetic distances between all pairs of the 55 accessions. Several groups of accessions conformed to expected species classification includingA. batizocoi, A. stenosperma, andA. monticola; whileA. duranensis, A. cardenasii, A. helodes, andA. correntina did not form good groups. The study showed that great diversity exists for protein profiles and seed storage proteins have potential for aiding species classification and for serving as markers for interspecific hybridization studies.  相似文献

Cactaceae is considered the fifth most endangered taxonomic group. In light of this, the aim of this study was to evaluate the efficiency of different low‐temperature storage techniques in maintaining the viability of seeds of cacti in different threat categories. Seeds of six cacti taxa were stored in a cold chamber (8°C), a freezer (?5°C), in liquid nitrogen (?196°C) and at room temperature (25–27°C) for a period of 0, 1, 3, 6, 9 and 13 months. At each evaluation interval we removed a seed sample for each taxon studied, which was distributed into four repetitions of 25 seeds maintained at room temperature under 12‐h light/dark photoperiods. We evaluated the germinability, mean germination time and synchronization index. Most of the studied taxa presented germinability of above 50%, which was influenced by time and by storage temperatures. Also, most taxa stored at room temperature presented a significant reduction in germinability, whereas almost all taxa showed maintenance of the seed viability when stored in a cold chamber, a freezer or liquid nitrogen. This response can be justified by the reduction of the seed metabolism and the degradation of the reserve compounds of the seeds while at lower temperatures. Our results indicate that storage at low temperatures is an effective method for the conservation of cacti seeds and can be used for the formation of artificial seed banks of threatened cacti species.  相似文献

Cultivated peanut, Arachis hypogaea L., is a tetraploid (2n = 4x = 40) species thought to be of allopolyploid origin. Its closest relatives are the diploid (2n = 2x = 20) annual and perennial species included with it in Arachis sect. Arachis. Species in section Arachis represent an important source of novel alleles for improvement of cultivated peanut. A better understanding of the level of speciation and taxonomic relationships between taxa within section Arachis is a prerequisite to the effective use of this secondary gene pool in peanut breeding programs. The AFLP technique was used to determine intra- and interspecific relationships among and within 108 accessions of 26 species of this section. A total of 1328 fragments were generated with 8 primer combinations. From those, 239 bands ranging in size from 65 to 760 bp were scored as binary data. Genetic distances among accessions ranged from 0 to 0.50. Average distances among diploid species (0.30) were much higher than that detected between tetraploid species (0.05). Cluster analysis using different methods and principal component analysis were performed. The resulting grouping of accessions and species supports previous taxonomic classifications and genome designations. Based on genetic distances and cluster analysis, A-genome accessions KG 30029 (Arachis helodes) and KSSc 36009 (Arachis simpsonii) and B-genome accession KGBSPSc 30076 (A. ipaensis) were the most closely related to both Arachis hypogaea and Arachis monticola. This finding suggests their involvement in the evolution of the tetraploid peanut species.  相似文献

Storage proteins have been found to play a major role in insect metamorphosis and egg production and are accumulated during the actively feeding larval stage. Yet few studies have focused on how nutrition affects storage protein levels. Three storage proteins were identified in male and female Heliothis virescens pupae, one arylphorin and two putative high-methionine hexamers. Storage proteins were quantified in early pupae and in pharate adults. Storage protein levels peaked in 48-h pupae and were more abundant in females across all stages. Both male and female pharate adults retained a portion of total storage protein levels and females retained greater levels overall. In females, post-eclosion protein reserves will likely be used toward egg manufacturing, while the role of protein reserves in males remains speculative. In our previous study of H. virescens larvae, we found that protein-derived growth in females progressively increased as dietary protein levels increased. Our present data show that levels of storage protein also increased progressively along with dietary protein levels. This suggests that females allocated protein, in excess of adult tissue formation needs, toward storage protein. Our study is the first to demonstrate how responsive storage protein levels can be in face of varying levels of dietary protein.  相似文献

Genome size variation within species is a frequently reported, but still a controversial problem. In the present study, we re-evaluated recently published Feulgen densitometric data on genome size and its infraspecific variation in Arachis hypogaea, and also conducted measurements in one accession of its wild relative A. monticola. The methods applied were propidium iodide flow cytometry and Feulgen densitometry using Pisum sativum as an internal standard. The 2C DNA contents previously published cannot be confirmed, but values obtained in this study are about half as large. Additionally, we could not reproduce the previously reported 1.15-fold variation within A. hypogaea; our data indicate genome size stability between respective accessions of this species. Based on 8.84 pg (2C) for Pisum sativum the DNA amounts (2C) were: 5.914 pg in A. hypogaea, and 5.979 pg in A. monticola.  相似文献

The 2C nuclear DNA amounts were determined for 99 accessions, representing 23 Arachis species from 8 of 9 taxonomic sections, and two synthetic amphidiploids. Mean 2C DNA amounts varied by 15.20%, ranging from 10.26 to 11.82 pg, between accessions of Arachis hypogaea (2n = 4x = 40). Nuclear DNA content variation (5.33-5.91 pg) was also detected among Arachis duranensis (2n = 2x = 20) accessions. The intraspecific variation in the two species may have resulted from indirect selection for favourable genome sizes in particular environmental conditions. The accessions belonging to A. hypogaea ssp. hypogaea (mean value 11.27 pg) with longer life cycle had significantly larger mean DNA content than the accessions of A. hypogaea ssp. fastigiata (mean value 10.97 pg). For 20 diploid (2n = 2x = 20) species of the genus, 2C nuclear DNA amounts ranged from approximately 3 to 7 pg. The diploid perennial species of section Arachis have about 12% more DNA than the annual species. Comparisons of DNA amounts show that evolutionary rating is not a reliable guide to DNA amounts in generic sections of the genus; lower DNA values with evolutionary advancement were found in sections Heteranthae and Triseminatae, but the same was not true for sections Arachis and Caulorrhizae. Similarly, there is evidence of significant differences in DNA content between 4 ancient sections (Procumbentes, Erectoides, Rhizomatosae, and Extranervosae) of the genus. The occurrence of genome size plasticity in both A. duranensis and A. hypogaea provides evidence that A. duranensis could be one of the diploid progenitors of A. hypogaea. The DNA content in the two synthetic amphidiploids corresponded to the sum value estimated for parental species. Total seed storage proteins were studied in 50 accessions of A. hypogaea (11 A. hypogaea ssp. hypogaea var hypogaea, 13 A. hypogaea ssp. hypogaea var hirsuta, 11 A. hypogaea ssp. fastigiata var fastigiata and 15 A. hypogaea ssp. fastigiata var. vulgaris accessions) in SDS PAGE. These accessions were also analysed for albumin and globulin seed protein fractions. Among the six seed protein markers presently used, it was found that globulin fraction showed maximum diversity (77.2%) in A. hypogaea accessions followed by albumin (52.3%), denatured total soluble protein fraction in embryo (33.3%) and cotyledon (28.5%). The cluster analysis based on combined data of cotyledons, embryos, albumins and globulins seed protein fractions demarcated the accessions of two subspecies hypogaea and fastigiata into two separate clusters supported by 51% bootstrap value, with few exceptions, suggesting the genotypes to be moderately diverse. Native and denatured total soluble seed storage proteins were also electrophoretically analysed in 27 wild Arachis species belonging to six sections of the genus. Cluster analysis using different methods were performed for different seed proteins data alone and also in combination. Section Caulorrhizae (C genome) and Triseminatae (T genome) formed one, distantly related group to A. hypogaea and other section Arachis species in the dendrogram based on denatured seed storage proteins data. The present analysis has maintained that the section Arachis species belong to primary and secondary genepools and, sections Procumbenetes and Erectoides belong to tertiary gene pools.  相似文献

Summary The morphology of pachytene chromosomes was studied in A. glabrata Benth. and A. pusilla Benth. belonging respectively to the sections Rhizomatosae and Triseminale. These two species can not be crossed with the cultivated groundnut A. hypogaea L. All 20 chromosomes of A. glabrata could be identified individually and further classified into 5 basic types. The features that enabled the identification of chromosomes were: total length, arm ratios, nucleolus attachment and position and extent of heterochromatin. A simple key has been proposed for classifying different chromosomes to facilitate their easy identification. The genomes of A. glabrata did not resemble those of A. hypogaea except for the presence of an A chromosome, 2 euchromosomes and 2 nucleolus organisers. A. glabrata did not appear to be an amphidiploid but rather an allopolyploid hybrid. The genome of A. pusilla contained chromosomes unlike those of any other species of section Arachis. It was concluded that both these species are quite unrelated to other species of the section Arachis.  相似文献

The genus Arachis contains a large number of species and undescribed taxa with patterns of genetic variation that are little understood. The objectives of this investigation were to estimate genetic diversity among species of Arachis by utilizing electrophoretic techniques and to establish the potential for use of isozymes as markers for germplasm introgression. One-hundred-and-thirteen accessions representing six of the seven sections of the genus were analyzed for isozyme variation of 17 enzymes. Section Rhizomatosae species were not included because they produce very few seeds. Seeds were macerated and the crude extract was used for starch-gel electrophoretic analyses. Although the cultivated species has few polymorphic isozymes, the diploid species are highly variable and two-to-six bands were observed for each isozyme among accessions. Because of the large number of isozyme differences between A. hypogaea and A. batizocoi (the presumed donor of the B genome), this species can no longer be considered as a progenitor of the cultivated peanut. Seed-to-seed polymorphisms within many accessions were also observed which indicate that germplasm should be maintained as bulk seed lots, representative of many individuals, or as lines from individual plants from original field collections. The area of greatest interspecific genetic diversity was in Mato Grosso, Brazil; however, the probability of finding unique alleles from those observed in A. hypogaea was greatest in north, north-central, south and southeast Brazil. The large number of polymorphic loci should be useful as genetic markers for interspecific hybridization studies.  相似文献

One hundred and sixteen accessions representing 28 species in the genus Arachis were evaluated for resistance to groundnut rosette disease using an infector row technique during the 1996/97, 1997/98, 1998/99 and 1999/2000 growing seasons at Chitedze, Malawi. Of these, a total of 25 accessions belonging to Arachis diogoi (1 accession), A. hoehnei (2), A. kretschmeri (2), A. cardenasii (2), A. villosa (1), A. pintoi (5), A. kuhlmannii (2), A. appressipila (3), A. stenosperma (5), A. decora (1), and A. triseminata (1) showed resistance to the groundnut rosette disease. No visible disease symptoms were observed in several accessions belonging to A. appressipila, A. cardenasii, A. hoehnei, A. kretschmeri, A. villosa, A. pintoi, A. kuhlmannii, and A. stenosperma. Some accessions in A. appressipila, A. diogoi, A. stenosperma, A. decora, A. triseminata, A. kretschmeri, A. kuhlmannii, and A. pintoi were resistant to all three components of rosette, Groundnut rosette ass is tor virus (GRAV), Groundnut rosette virus (GRV) and its satellite RNA (sat. RNA). Two accessions in A. stenosperma and one accession in A. kuhlmannii showed the presence of all three components of the rosette disease. Several wild Arachis accessions were resistant to GRAV. All the accessions of A. batizocoi (4), A. benensis (2), A. duranensis (46), A. dardani (1), A. ipaensis (1), A. magna (1), A. monticola (3), A. oteroi (1), A. pusilla (4), and A. valida (2) were susceptible to rosette disease. In all these accessions, infected plants were chlorotic and severely stunted. The value of exploitation of the resistance in wild Arachis species in rosette resistance breeding programmes is discussed.  相似文献

Background and Aims

The most plausible explanation for treeline formation so far is provided by the growth limitation hypothesis (GLH), which proposes that carbon sinks are more restricted by low temperatures than by carbon sources. Evidence supporting the GLH has been strong in evergreen, but less and weaker in deciduous treeline species. Here a test is made of the GLH in deciduous–evergreen mixed species forests across elevational gradients, with the hypothesis that deciduous treeline species show a different carbon storage trend from that shown by evergreen species across elevations.

Methods

Tree growth and concentrations of non-structural carbohydrates (NSCs) in foliage, branch sapwood and stem sapwood tissues were measured at four elevations in six deciduous–evergreen treeline ecotones (including treeline) in the southern Andes of Chile (40°S, Nothofagus pumilio and Nothofagus betuloides; 46°S, Nothofagus pumilio and Pinus sylvestris) and in the Swiss Alps (46°N, Larix decidua and Pinus cembra).

Key Results

Tree growth (basal area increment) decreased with elevation for all species. Regardless of foliar habit, NSCs did not deplete across elevations, indicating no shortage of carbon storage in any of the investigated tissues. Rather, NSCs increased significantly with elevation in leaves (P < 0·001) and branch sapwood (P = 0·012) tissues. Deciduous species showed significantly higher NSCs than evergreens for all tissues; on average, the former had 11 % (leaves), 158 % (branch) and 103 % (sapwood) significantly (P < 0·001) higher NSCs than the latter. Finally, deciduous species had higher NSC (particularly starch) increases with elevation than evergreens for stem sapwood, but the opposite was true for leaves and branch sapwood.

Conclusions

Considering the observed decrease in tree growth and increase in NSCs with elevation, it is concluded that both deciduous and evergreen treeline species are sink limited when faced with decreasing temperatures. Despite the overall higher requirements of deciduous tree species for carbon storage, no indication was found of carbon limitation in deciduous species in the alpine treeline ecotone.  相似文献

Summary A batch of peanutArachis hypogaea seed were divided into 4 2-kilogram portions and stored 6 months at 22–28° C. One portion was stored with the moisture content slowly increasing from 4 to 28 %; the other portions were maintained at approximately 4.5, 8.9 and 11.5 %. After 1, 2, 3, 4, and 6 months storage, 100 seed of each treatment were cultured on malt-10 % NaCl agar and the fungi isolated identified. The storage fungi present after intervals of storage were compared with those initially present. In seed stored at 4.4 % moisture,Aspergillus species remained about the same andPenicillium decreased. At 8.9 % moistureA. flavus, A. repens andPenicillium were reduced, andA. amstelodami andA. ruber increased. At 11.5 % moisture, 100% of the seed yieldedA. ruber after 4 months storage and the other storage fungi decreased to 0–2 %. The moisture content of seed stored at 99 % R. H. increased slowly to 28 % and resulted in monthly changes in the dominant fungi. Convicilins, a set of seed storage proteins, differ from vicilins, a related group of seed storage proteins, mainly because of the presence of the N-terminal extension, an additional sequence of amino acids in the sequence corresponding to the first exon. Convicilins have been described only in species of the legume tribe Vicieae. One or two genes for convicilins have been identified in most species of this tribe. The genus Pisum is the main exception, since two genes have been identified in most of its species. Thirty-four new convicilin gene sequences from 29 different species (Lathyrus, Lens, Pisum, and Vicia spp.) have been analyzed here. Convicilin gene sequences are generally organized in 6 exons, but in some instances one of the internal introns (2nd or 4th) is lost. In these 29 species, the N-terminal extension is formed by a stretch of 99 to 196 amino acids particularly rich in polar and charged amino acids (on average, it contains 29.43% glutamic acid and 15.38% arginine residues). This N-terminal extension has the characteristics of an intrinsically unstructured region (IUR), one of the categories of protein "degenerate sequences". A comparative analysis indicates that the N-terminal extension sequence has evolved faster than the surrounding sequence, which is common to all vicilins, and it evolved mainly through a series of duplications of short internal sequences and triplet expansions, the predominant one being GAA. This agrees with the evolution of IURs, which is faster than the evolution of surrounding sequences and is mainly due to replication slippage and unequal crossover recombination. Alternative maximum-likelihood trees of phylogenetic relationships among the 29 Vicieae species based on the convicilin exon sequences are presented and discussed.  相似文献

The genus Arachis encompasses 80 validly described species and the most striking characteristics is the geocarpic fruit, which occurs in every species of the genus. Taking into consideration that much of the current knowledge about this subject is based on the anatomy and morphology of A. hypogaea, a cultivated species with fruits that were modified during domestication, the objective of this study was to learn more about the development of the geocarpic fruits of wild species of Arachis (A. paraguariensis, A. pintoi, A. stenosperma, and A. tuberosa), and to provide characters that can be used in future phylogenetic works about this group. Buds, flowers and fruits at different stages of development were collected and processed according to standard methods used for light field, interferential contrast, and scanning electron microscopy. Although the geocarpic fruits of the wild species share many adaptations, this study identified potentially diagnostic infrageneric characters, such as the type of trichomes on the pericarp, the presence of sclerenchyma caps in the bundles of the pericarp of the seed chamber of the fruit and isthmus, the presence of tracheoid cells in the pith of the isthmus, the location of crystalliferous cells, the presence of tannins in the cells of mesocarp outer layers, and the presence of projections in the sclerenchyma tissue. In addition, this article brings some inferences about functional characteristics of this peculiar geocarpic fruit.  相似文献

采用样方法收集了安徽万佛山自然保护区常见的206种植物的种子, 通过测量种子千粒重, 研究了种子大小(seed mass, SM)在不同分类水平及生活型间的变异规律。结果表明: 1) SM在植物种间变异较大, 从小到大分为A、B、C、D、E、F、G 7个数量级, SM平均为21.487 g; C级种子(0.1 g ≤ SM < 1.0 g)最多, 共计73种, 占35.4%; D级种子(1.0 g ≤ SM < 10.0g)次之, 共计56种, 占27.2%; 野菰(Aeginetia indica)种子最小, 仅为0.006 g; 木鳖子(Momordica cochinchinensis)种子最大, 重达1775.192 g。2) SM在科间(平均种子大小)存在极显著差异(p < 0.001), 且变异沿系统发育方向呈显著变小趋势(r = -0.079, p = 0.048)。其中, SM较大的5个科分别为葫芦科、柿树科、楝科、安息香科和金缕梅科; SM较小的5个科分别为列当科、兰科、秋海棠科、苦苣苔科和灯心草科。3)该研究中植物的生活型分为8种类型。其中, 多年生草本优势明显, 计83种, 占39.9%, SM变异包括A-E 5个数量级; 一年生草本次之, 计60种, 占28.8%, SM变异包括A-F 6个数量级; 灌木第三, 计34种, 占16.3%, SM变异包括B-F 5个数量级。SM在8种生活型间(平均种子大小)也存在极显著差异(p < 0.001); 从大到小分别为藤本、乔木、灌木、一年生草本、多年生草本、半灌木、一二年生草本、二或多年生草本; 从草本、半灌木、灌木、乔木到藤本, SM与生活型具有极显著的正相关性(r = 0.220, p < 0.001)。4)种类较多的菊科、禾本科、莎草科及蔷薇科, 各科内属间SM也存在极显著差异(p < 0.001)。本研究结果有助于加深对植物生活史对策中的种子大小对策的理解, 提高对森林植被的管理水平, 丰富保育生态学的种子恢复理论。  相似文献

Summary Charge and molecular weight heterogeneity of globulin-1 (G1) polypeptides of the bean, Phaseolus vulgaris L., were revealed by sodium dodecyl sulphate-polyacrylamide gel electrophoresis (SDS-PAGE). Different bean cultivars were classified into three groups: Tendergreen, Sanilac, and Contender on the basis of their protein subunit composition. Nine distinct major bands: 51,49, 48.5,48T, 48S, 47, 45.5, 45S, and 45C, and two minor bands: 46T and 46S were found to account for the three profiles seen on one-dimensional SDS-PAGE. Two-dimensional analysis revealed these eleven protein bands to be composed of a minimum of fourteen distinct protein subunits. The Tendergreen and Sanilac types differ in their G1 polypeptide composition. The protein patterns of the Contender types are intermediate, containing many protein subunits found in the patterns of the Tendergreen and Sanilac types suggesting a genetic and evolutionary relationship.  相似文献

In alpine species the classification of the various mechanisms underlying seed dormancy has been rather questionable and controversial. Thus, we investigated 28 alpine species to evaluate the prevailing types of dormancy. Embryo type and water impermeability of seed coats gave an indication of the potential seed dormancy class. To ascertain the actual dormancy class and level, we performed germination experiments comparing the behavior of seeds without storage, after cold-dry storage, after cold-wet storage, and scarification. We also tested the light requirement for germination in some species. Germination behavior was characterized using the final germination percentage and the mean germination time. Considering the effects of the pretreatments, a refined classification of the prevailing dormancy types was constructed based on the results of our pretreatments. Only two out of the 28 species that we evaluated had predominantly non-dormant seeds. Physiological dormancy was prevalent in 20 species, with deep physiological dormancy being the most abundant, followed by non-deep and intermediate physiological dormancy. Seeds of four species with underdeveloped embryos were assigned to the morphophysiologial dormancy class. An impermeable seed coat was identified in two species, with no additional physiological germination block. We defined these species as having physical dormancy. Light promoted the germination of seeds without storage in all but one species with physiological dormancy. In species with physical dormancy, light responses were of minor importance. We discuss our new classification in the context of former germination studies and draw implications for the timing of germination in the field.  相似文献
