The physiology of Lactobacillus delbrueckii ssp. bulgaricus and Lactobacillus casei, extensively used in the dairy industry, was studied in order to evaluate key parameters in the synthesis of exopolysaccharides and to improve their production through novel fermentation processes. Selected strains were studied in shake flasks and in fermentor experiments using glucose and lactose as main carbon sources and bacto casitone as the only complex component, in a temperature range between 35 and 42°C. The production of exopolysaccharides was monitored and correlated to the growth conditions using both a colorimetric assay and chromatographic methods. Fermentor experiments in batch mode yielded 100 mg l−1 of EPS from L. bulgaricus and 350 mg l−1 from L. casei. Moreover, the use of a microfiltration (MF) bioreactor resulted in exopolysaccharides (EPS) concentrations threefold and sixfold those of batch experiments, respectively. The monosaccharidic composition of the two analyzed polymers differed from those previously reported. The optimization of the production of EPSs using the MF fermentation strategy could permit the use of these molecules produced by generally recognised as safe (GRAS) microorganisms in the place of other polysaccharides in the food industry.  相似文献

The production of fungal spores using on-site, non-sterile, portable fermentation equipment is technically constrained. Very little information is available on the production requirements, such as medium concentration, inoculum stabilization, required fermentation times, and maintenance of axenic growth. In this study, we developed a two-part, liquid concentrate of the production medium that remains stable and soluble at room temperature. We also examined inoculum stability and showed that freeze- or air-dried blastospore preparations were stable for 7 days after rehydration when stored at 4 °C. The use of a low-pH (pH 4), relatively rich complex medium provided a growth environment deleterious to bacterial growth yet conducive to rapid sporulation by Paecilomyces fumosoroseus. High concentrations of blastospores (7.9×108/ml) of P. fumosoroseus were produced in a 40-h fermentation with very low levels of bacterial contamination when the fermentor was charged with a blastospore production medium with a starting pH of 4 and inoculated with blastospore concentrations greater than 1×106 spores/ml. These studies demonstrate that the use of disinfected, portable fermentation equipment has potential for on-site production of high concentrations of blastospores of the bioinsecticidal fungus P. fumosoroseus.  相似文献

利用分批发酵研究了灵芝(Ganoderma lucidum)胞外多糖的合成特性,结果表明Ganoderma lucidum多糖合成和菌体生长呈部分生长关联型。菌体干重、胞外多糖分别达到15.56g·L-1<、3.02g·L-1<,胞外多糖对细胞干重得率系数(Yp/x)为0.19。根据分批发酵试验结果采用Logistic方程、Luedeking-Piret方程和类似Luedeking-Piret方程,得到了描述灵芝生长、胞外多糖以及葡萄糖底物消耗分批发酵动力学模型。同时在初始葡萄糖变化较大范围内,试验数据与模型预测值进行了比较拟合,平均相对误差小于5%,表现出很好的适用性。表明该动力学模型对指导灵芝胞外多糖的发酵生产具有实际意义。  相似文献

A high-performance liquid chromatographic method with electrochemical detection has been developed for the simultaneous determination of epirubicin, 13-S-dihydroepirubicin, doxorubicin and 13-S-dihydrodoxorubicin in human plasma. An aliquot of 200 μl plasma, spiked with internal standard, was extracted by solid-phase extraction using polymeric adsorbent columns. Chromatography was performed using a C18 reversed-phase column with a mobile phase consisting of water–acetonitrile (71:29, v/v) containing 0.05 M Na2HPO4 and 0.05% v/v triethylamine adjusted to pH 4.6 with citric acid. Linearity of the method was obtained in the concentration range of 1–500 ng/ml for all the analytes. Analytical recoveries of the analytes ranged from 89 to 93%. The assay can be used for the simultaneous determination of the four analytes, or for epirubicin and its metabolite or doxorubicin and its metabolite, using the other parent drug as an internal standard. The method was applied to analyze human plasma samples from patients treated with epirubicin using doxorubicin as an internal standard.  相似文献

Long-term cell recycle fermentations of Candida tropicalis were performed over 14 rounds of fermentation. The average xylitol concentrations, fermentation times, volumetric productivities and product yields for 14 rounds were 105 g l–1, 333 h, 4.4 g l–1 h–1 and 78%, respectively, in complex medium; and 110 g l–1, 284 h, 5.4 g l–1 h–1 and 81%, respectively, in a chemically defined medium. These productivities were 1.7 and 2.4 times those with batch fermentation in the complex and chemically defined media, respectively. The xylitol yield from xylose with cell recycle fermentation using the chemically defined medium was 81% (w/w), which was 7% greater than the xylitol yield with batch fermentation (74%); both modes of fermentation gave the same yield using the complex medium. These results suggest that the chemically defined medium is more suitable for production of xylitol than complex medium.  相似文献

Removal of phosphorus (P) by Ceratophyllum demersum L. and associated epiphytic periphyton was quantified by measuring the disappearance of soluble reactive P (SRP) from microcosms during 1-h in situ incubations conducted over a 1-year period. Initial P concentrations in these incubations ranged from 30 to >10,000 μg P L−1. Phosphorus removal was proportional to initial P concentrations and was weakly correlated with solar irradiance and water temperature. Removal rates (0.6–32.8 mg P m−2 d−1) and kv coefficients (0.68–1.93 h−1) from experiments run at low initial P concentrations (up to 200 μg P L−1) were comparable to results reported for other macrophytes. Removal rates from experiments run at the highest (>10,000 μg P L−1) initial P concentrations (5300 and 11,100 mg P m−2 d−1) most likely represented luxury nutrient consumption and were not thought to be sustainable long term. We were unable to determine a Vmax for P removal, suggesting that the nutrient-storage capability of the C. demersum/periphyton complex was not saturated during our short-term incubations. Based on N:P molar ratios, the marsh was P limited, while the C. demersum/periphyton complex was either N limited or in balance for N and P throughout this study. However, despite its tissue stoichiometry, the C. demersum/periphyton complex always exhibited an affinity for P. It appeared that the biochemical mechanisms, which mediate P removal, at least on a short-term basis, were more influenced by increases in ambient P levels than by tissue nutrient stoichiometry.  相似文献

Camalexin is a phytoalexin of Arabidopsis thaliana and an important component of inducible defenses. Accurate quantification of low concentrations suffers from interference by structurally related metabolites. A. thaliana plants were induced with silver nitrate and camalexin was extracted using methanol and identified and quantified by (i) TLC as a blue fluorescent band, (ii) microtiter plate-based fluorescence spectroscopy, (iii) GC on a midpolar column coupled to flame ionization detection, (iv) C18 HPLC coupled to a photodiode detector, and (v) UPLC coupled to a mass spectrometer detector. Standard curves over the range of 0.1–15 μg ml−1 gave R2 values from 0.996 to 0.999. The different methods were compared and evaluated for their ability to detect and quantify increasing concentrations (<0.4–8 μg g−1 FW) of camalexin. Each of the techniques presented advantages and disadvantages with regard to accuracy, precision, interference, analytical sensitivity, and limits of detection. TLC is a good qualitative technique for the identification of camalexin and fluorescence spectroscopy is subject to quenching when performed on crude extracts. Comparable results were obtained with GC–FID, HPLC–PDA, and UPLC–MS, with UPLC–MS having the added advantage of short analysis times and detection based on accurate mass.  相似文献

Larvae of the South African clawed frog, Xenopus laevis (Daudin), are efficient, obligate suspension feeders. We examine the relationship between the ambient particle concentration offered these larvae as food and their filtering, ingestion, and buccal pumping rates. We demonstrate that: (i) the larvae can sense and respond to a broad range of particle concentrations, down to 0.2 mg 1–1 (dry weight); (ii) their metabolic needs theoretically can be met by particle concentrations as low as 5 mg 1–1; and (iii) their patterns of regulation of filtering and ingestion fit predictions from certain models used to describe zooplankton feeding dynamics. Two such models are discussed: the modified Monod (Michaelis-Menten) model, with a lower threshold below which the tadpoles do not feed, and an energy optimization model. Both the models and the observed behavior of the tadpoles allow for stability of populations of food organisms. Tadpole feeding dynamics apparently are compatible with both the predictions and assumptions of these models, suggesting similar regulation of feeding by tadpoles and zooplankton. Food limitation was tested in the laboratory by individual growth and reproduction of two cladoceran species, Ceriodaphnia richardi and Daphnia gessneri, from the shallow tropical Brazilian Lake Monte Alegre. The cladocerans were fed cultivated green alga Scenedesmus spinosus in concentrations of 0.20, 0.10, 0.05, and 0.025 mg C l−1. Higher biomass and growth rates occurred in the two highest-food concentrations; the two lowest ones negatively affected clutch size and first reproduction. The threshold food concentration is lower than 0.025 mg C l−1 and the incipient limiting level is a value between 0.10 and 0.20 mg C l−1. The largest species, D. gessneri, was more sensitive to low food concentrations. The effects of low and high temperatures (19 and 27°C) were evaluated by life table experiments with three cladocerans from the lake—Daphnia ambigua, D. gessneri, and Moina micrura—with no food limitation (1 mg C l−1 of S. spinosus). Higher population growth rates for the three species were found at 27°C; better performance in most life table parameters was observed for the former two species at the highest temperature, D. gessneri being the most sensitive to the lowest temperature. There are indications that temperature is an important abiotic factor that constrains populations of cladocerans for a short period in winter in the lake, when temperature decreases to 18–19°C. However, its influence cannot be separated from a biotic factor such as food, whose effect is stronger in the cool season, when concentrations are lower and contribution of inedible algae is relatively higher.  相似文献

The methylotrophic bacteriumHyphomicrobium VS was enriched and isolated, using activated sewage sludge as inoculum in mineral medium containing dimethylsulfide (DMS) at a low concentration to prevent toxicity. DMS concentrations above 1 mM proved to be growth inhibiting.Hyphomicrobium VS could use DMS, dimethylsulfoxide (DMSO), methanol, formaldehyde, formate, and methylated amines as carbon and energy source. Carbon was assimilated via the serine pathway. DMS-grown cells respired sulfide, thiosulfate, methanethiol, dimethyldisulfide and dimethyltrisulfide.To testHyphomicrobium VS for application in biofiltration of air polluted with volatile sulfur compounds two laboratory scale trickling biofilters with polyurethane and lava stone as carrier material were started up by inoculation with this bacterium. Both methanol- and DMS-grown cells could be used. Only a short adaptation period was needed. Atlantic sea scallops, Placopecten magellanicus, in most areas of the Bay of Fundy, New Brunswick, Canada, have year-round concentrations of paralytic shellfish posioning (PSP) toxins greater than the regulatory concentration of 80 μg STX eq. 100 g−1 wet weight. Scallops (mean shell height of 10.7 cm, age 3–5 years) were collected by SCUBA and individually tagged near Parker Island, Bay of Fundy. Half were hung 2 m below the low tide water level and the remainder were placed on the bottom (11 m depth at low tide) under the scallops held at 2 m. Scallop, water and sediment samples were collected monthly for determination of concentrations of PSP toxins and Alexandrium fundyense.In October, 1993, mean concentrations of PSP toxins in digestive gland, and mantle were 3205 and 1018 μg STX eq. 100 g−1 wet weight, respectively. Eight months later (June 1994), PSP concentrations in digestive glands from the surface and bottom had declined to 504 and 682 μg STX eq. 100 g−1 wet weight, respectively, whereas those in the mantle had declined to 802 and 681 μg STX eq. 100 g−1 wet weight. During July 1994, A. fundyense concentrations observed at Parker Island and offshore were 320 cells l−1 and 14,200 cells l−1, respectively. Subsequently, toxin concentrations in surface and bottom scallop digestive glands increased to 12,720 and 11,408 μg STX eq. 100 g−1 wet weight, whereas concentrations in mantles increased to 2126 and 1748 μg STX eq. 100 g−1 wet weight, respectively. Concentrations of PSP toxins in these tissues in October 1994 were similar to those measured in October 1993. Concentrations of PSP toxin were less than the regulatory concentration in the gonads and non-detectable in adductor muscles of all scallops sampled.There were no statistically significant differences in profiles for uptake and depuration of PSP toxins in scallops held at the surface compared to those from bottom, suggesting that A. fundyense cysts at the concentrations found in the sediment (45 cysts cm−3) did not contribute significantly to the year-round presence of PSP toxins within scallop tissues. The year-round occurrence of PSP toxin is probably due to accumulation during summer blooms followed by a very slow rate of depuration.  相似文献

Spatial distribution patterns of arsenic (As) in the tissues of a lesser-known As hyperaccumulating fern Pityrogramma calomelanos (L.) Link var. austroamericana (Domin) Farw. (Pteridaceae) have been studied. Quantitative micro-proton-induced X-ray emission (micro-PIXE) spectroscopy was employed to examine As localization in pinnule and stipe cross-sections of this species. In addition, As hyperaccumulation status of P. calomelanos var. austroamericana was compared with the well-known As hyperaccumulating fern Pteris vittata L. Both species were grown in pots under controlled conditions and exposed to four levels of As (0–500 mg As kg−1) for 20 weeks. Pityrogramma calomelanos var. austroamericana accumulated up to 16 415 mg As kg−1 dry weight (DW), however, phytotoxicity symptoms such as necrotic pinnule tips and margins, appeared in fronds with concentrations >3,008 mg As kg−1 DW. Arsenic was readily translocated to fronds, with concentrations up to 75 times greater in fronds than in roots. Quantitative elemental maps of As generated using micro-PIXE analysis revealed that As concentrations in pinnule cross-sections were higher than in stipe cross-sections with concentrations of 3.7 × 103 and 1.6 × 103 mg As kg−1 DW, respectively (as determined by region selection analysis; RSA). In pinnules, RSA revealed variable concentrations of As, however did not resolve a clear pattern of compartmentalization across different anatomical regions. In stipe tissues, As concentrations followed the order vascular bundle > cortex > epidermis (as determined by RSA). Our results show that P. calomelanos var. austroamericana is an As hyperaccumulator and has the potential for use in phytoremediation of soils with low levels (up to 50 mg kg−1) of As contamination.  相似文献

1. The effects of inorganic Hg2+ and methylmercuric chloride on the ionic currents of cultured hippocampal neurons were studied and compared. We examined the effects of acute exposure to the two forms of mercury on the properties of voltage-activated Ca2+ and Na+ currents and N-methyl-D-aspartate (NMDA)-induced currents.2. High-voltage activated Ca2+ currents (L type) were inhibited by both compounds at low micromolar concentrations in an irreversible manner. Mercuric chloride was five times as potent as methylmercury in blocking L-channels.3. Both compounds caused a transient increase in the low-voltage activated (T-type) currents at low concentrations (1 M) but blocked at higher concentrations and with longer periods of time.4. Inorganic mercury blockade was partially use dependent, but that by methylmercury was not. There was no effect of exposure of either form of mercury on the I–V characteristics of Ca2+ currents.5. Na+- and NMDA-induced currents were essentially unaffected by either mercury compound, showing only a delayed nonspecific effect at a time of overall damage of the membrane.6. We conclude that both mercury compounds show a relatively selective blockade of Ca2+ currents, but inorganic mercury is more potent than methylmercury.  相似文献

A sensitive, robust gas chromatographic–mass spectrometric assay suitable for use in pharmacokinetic or bioequivalence studies is presented for the selective serotonin reuptake inhibitor, fluoxetine, and its major metabolite, norfluoxetine (N-desmethylfluoxetine). This method employs solid-phase extraction followed by acetylation with trifluoroacetic anhydride and analysis of the derivatives using selected ion monitoring. The lower limit of quantification was 1.0 ng/ml, and the assay was linear for both analytes from 1 to 100 ng/ml. Mean recoveries following solid-phase extraction at concentrations of 5.0, 20 and 100 ng/ml were 91% (fluoxetine) and 87% (norfluoxetine). Assay precision (as mean RSD) and accuracy (as mean relative error) for both analytes were tested at the same three nominal concentrations and were found to be within 10% in all cases. Analysis of fluoxetine concentrations in plasma samples from 18 volunteers following administration of a single 40 mg dose of fluoxetine provided the following pharmacokinetic data (mean±SD): Cmax, 32.73±9.21 ng/ml; AUC0–∞, 1627±1372 ng/ml h; Tmax, 3.08 h (median); ke, 0.022±0.007 h−1; elimination half-life, 37.69±21.70 h.  相似文献

The antagonistic activity of two yeast strains (Pichia anomala (E.C. Hansen) Kurtzman, strain K and Candida oleophila Montrocher, strain O) against the parasitic complex responsible for banana crown rot was evaluated. The strains were applied at three different concentrations (106, 107, 108 cfu/ml) and their efficacy tested in vivo on three separate fungi (Colletotrichum musae (Berk. & Curt.) Arx, Fusarium moniliforme Sheldon, and Cephalosporium sp.) and on a parasitic complex formed by association of these three fungi. At the concentrations used C. musae appeared to be the most pathogenic. The complex showed intermediate aggressiveness between C. musae and both other fungi.Statistically significant antagonistic effects were observed on C. musae, F. moniliforme, and the fungal complex. The highest protection level (54.4%) was observed with strain O added at 108 cfu/ml on crowns previously inoculated with the fungal complex. The level was lower when the fungi were inoculated separately.Furthermore, the antagonistic effect was strongly reinforced when strain O at 108 cfu/ml was applied 24 h before fungal complex inoculation (59.9%), as compared to its application 15 min (24.3%) or 3 h (27.3%) after fungal complex inoculation. Bananas showed increased susceptibility to the fungal complex from March to June, and this influenced the level of protection by yeast, which decreased over the same period. A strict negative correlation (R2 = 0.83) was highlighted between susceptibility of banana to crown rot and protection provided by yeast.  相似文献

Continuous ethanol fermentation of cheese whey powder solution was realized using pure culture of Kluyveromyces marxianus (DSMZ 7239) at hydraulic residence times (HRT) between 12.5 and 60 h. Sugar utilization, ethanol and biomass formation were investigated as functions of HRT. Effluent sugar concentration decreased, but percent sugar utilization, ethanol and biomass concentrations increased with HRT. Ethanol productivity was maximum (0.745 gE l−1h−1) at an HRT of 43.2 h where the biomass productivity was almost minimum (0.18 gX l−1 h−1). The ethanol yield coefficient was almost constant at 0.4 gE g−1S up to HRT of 43.2 h and the growth yield coefficient was minimum at HRT of 43.2 h. Kinetic models were developed and the constants were determined by using the experimental data.  相似文献

Algal food density is known to influence life history variables of cladoceran species. It is not, however, well established whether both littoral and planktonic cladocerans show similar trends when exposed to increasing food concentrations. In the present work, we studied the life table demography of four cladoceran species (Ceriodaphnia cornuta, Moina macrocopa, Pleuroxus aduncus and Simocephalus vetulus) in relation to three algal food concentrations (low: 0.5 × 106, medium: 1.5 × 106 and high: 4.5 × 106 cells ml–1 of Chlorella vulgaris) (in terms of carbon content, these were equivalent to 0.15, 0.45 and 1.35 g ml–1, respectively) at 25 °C. In general, for all the tested cladoceran species, values of average lifespan, gross reproductive rate, net reproductive rate, generation time and the rate of population growth were higher at lower food concentrations. Furthermore, high food concentration resulted in a negative population growth rate (mean ± standard error: –0.091 ± 0.026) for P. aduncus. The highest population growth rate (0.602 ± 0.014) was recorded for M. macrocopa at low food density. S. vetulus had the longest average lifespan (40 ± 1 d) while M. macrocopa had the lowest (5 ± 1 d). C. cornuta showed better performance at medium food concentration. We conclude that among the algal concentrations used here, 0.5 × 106 – 1.5 × 106 was beneficial not only to the planktonic species but also to the littoral P. aduncus and S. vetulus while 4.5 × 106 cells ml–1 was unsuitable for all the cladocerans tested.  相似文献

Biosurfactants have been suggested as a method to control harmful algal blooms (HABs), but warrant further and more in-depth investigation. Here we have investigated the algicidal effect of a biosurfactant produced by the bacterium Pseudomonas aeruginosa on five diverse marine and freshwater HAB species that have not been tested previously. These include Alexandrium minutum (Dinophycaee), Karenia brevis (Dinophyceae), Pseudonitzschia sp. (Bacillariophyceae), in marine ecosystems, and Gonyostomum semen (Raphidophyceae) and Microcystis aeruginosa (Cyanophyecae) in freshwater. We examined not only lethal but also sub-lethal effects of the biosurfactant. In addition, the effect of the biosurfactant on Daphnia was tested. Our conclusions were that very low biosurfactant concentrations (5 μg mL−1) decreased both the photosynthesis efficiency and the cell viability and that higher concentrations (50 μg mL−1) had lethal effects in four of the five HAB species tested. The low concentrations employed in this study and the diversity of HAB genera tested suggest that biosurfactants may be used to either control initial algal blooms without causing negative side effect to the ecosystem, or to provoke lethal effects when necessary.  相似文献

Oilseed rape (Brassica napus L.) needs very high nitrogen fertilizer inputs. Significant amounts of this nitrogen are lost during early leaf shedding and are a source of environmental and economic concern. The objective of this study was to investigate whether the remobilization of leaf amino acids could be limiting for nitrogen use efficiency. Therefore, amino acid concentrations were analyzed in subcellular compartments of leaf mesophyll cells of plants grown under low (0.5 mM NO3) and high (4 mM NO3) nitrogen supply. With high nitrogen supply, young leaves showed an elevated amino acid content, mainly in vacuoles. In old leaves, however, subcellular concentrations were similar under high and low nitrogen conditions, showing that the excess nitrogen had been exported during leaf development. The phloem sap contained up to 650 mM amino acids, more than four times as much than the cytosol of mesophyll cells, indicating a very efficient phloem-loading process. Three amino acid permeases, BnAAP1, BnAAP2, and BnAAP6, were identified and characterized. BnAAP1 and BnAAP6 mediated uptake of neutral and acidic amino acids into Xenopus laevis oocytes at the actual apoplastic substrate concentrations. All three transporters were expressed in leaves and the expression was still detectable during leaf senescence, with BnAAP1 and BnAAP2 mRNA levels increasing from mature to old leaves. We conclude that phloem loading of amino acids is not limiting for nitrogen remobilization from senescing leaves in oilseed rape.  相似文献

When several concentrations of brassinolide (BL) were added to a shoot induction medium (SIM) that contained only BA, redifferentiation of adventitious shoots from tobacco leaf discs was unaffected at low BL levels (10-10~10-8 M), but was inhibited at higher concentrations. In comparison, when BL was applied without BA, only cell expansion occurred and no shoots formed. The determination time for shoot formation was shortened at low BL concentrations, but their formation was postponed (i.e., time was lengthened) at higher concentrations. Elongation of shoots incubated for 30 d was unaffected at low BL concentrations, but was inhibited as that amount increased.NTH1, a tobacco homeobox gene that is expressed in the central zone of the tobacco shoot apex, showed greater expression levels in the SIM over time, and its expression was stronger in media treated with low concentrations of BL compared with the SIM control at the same time point. Expression ofNTH1 was postponed at higher BL concentrations. In conclusion, at low concentrations, brassinolide has no effect on shoot formation. However, it inhibits their formation at high concentrations when cytokinin is included in the media.  相似文献
