Ultrastructure of male reproductive accessory glands and ejaculatory duct in the Queensland fruit fly (Q-fly), Bactrocera tryoni, were investigated and compared with those of other tephritid flies. Male accessory glands were found to comprise one pair of mesodermic glands and three pairs of ectodermic glands. The mesodermic accessory glands consist of muscle-lined, binucleate epithelial cells, which are highly microvillated and extrude electron-dense secretions by means of macroapocrine transport into a central lumen. The ectodermic accessory glands consist of muscle-lined epithelial cells which have wide subcuticular cavities, lined with microvilli. The electron-transparent secretions from these glands are first extruded into the cavities and then forced out through small pores of the cuticle into the gland lumen. Secretions from the two types of accessory glands then flow into the ejaculatory duct, which is highly muscular, with epithelial cells rich in rough endoplasmic reticulum and lined with a thick, deeply invaginated cuticle. While there are some notable differences, reproductive accessory glands of male Q-flies generally resemble those of the olive fruitfly, Bactrocera oleae, and to a lesser extent the Mediterranean fruit fly, Ceratitis capitata.  相似文献

Amazona aestiva Linnaeus, 1758 is known as the blue-fronted parrot. These animals are considered seed dispersion, contributing to ecological balance. Despite its ecological importance, A. aestiva is listed as an imminent danger of becoming extinct, as it is one of the most illegally traded native species in Brazil, besides losing its habitat due to deforestation and forest fires. The cranial oesophagus mucosa is lined with a non-keratinized stratified squamous epithelium, and the basal layer presents melanocyte accumulation. No mucous glands were observed in the lamina propria/submucosa. The ingluvium two different muscular layer patterns were observed, one internal and circular and one very thick, external and longitudinal. The proventriculum submucosa consists of polygonal proventricular glands and contents into a broad central duct lined with simple columnar epithelium Periodic acid-reactive Schiff positive (PAS+) and Alcian blue positive (AB+). The 5-HT cells are dispersed in the proventricular glands located at base and apex. The ventricle mucous layer contains numerous branched folds lined by a simple columnar epithelium. The epithelium was negative reaction for PAS and positive for AB only on the fold surfaces. The 5-HT cells were scarce and observed only at the base of the ventricular gland, as well as in the adjacent connective tissue.  相似文献

The morphology and the ultrastructure of the male accessory glands and ejaculatory duct of Ceratitis capitata were investigated. There are two types of glands in the reproductive apparatus. The first is a pair of long, mesoderm-derived tubules with binucleate, microvillate secretory cells, which contain smooth endoplasmic reticulum and, in the sexually mature males, enlarged polymorphic mitochondria. The narrow lumen of the gland is filled with dense or sometimes granulated secretion, containing lipids. The second type consists of short ectoderm-derived glands, finger-like or claviform shaped. Despite the different shape of these glands, after a cycle of maturation, their epithelial cells share a large subcuticular cavity filled with electron-transparent secretion. The ejaculatory duct, lined by cuticle, has epithelial cells with a limited involvement in secretory activity. Electrophoretic analysis of accessory gland secretion reveals different protein profiles for long tubular and short glands with bands of 16 and 10 kDa in both types of glands. We demonstrate that a large amount of accessory gland secretion is depleted from the glands after 30 min of copulation.  相似文献

Human deep posterior lingual glands (von Ebner's glands) are located beneath the circumvallate papillae. They are formed by tubuloalveolar adenomeres, intercalated ducts and excretory ducts coming together in the main excretory duct. The tubuloalveolar cells, pyramid-shaped, show large and dense secretory granules (clear cored) throughout the cytoplasm, rare basal folds and packed cisternae of rough endoplasmic reticulum (RER) at the basal pole. The columnar cells of the intercalated ducts are arranged in a monolayer. They are characterized by dense, clear-core secretory granules (mostly in the apical cytoplasm), a basal nucleus, well-developed RER and Golgi apparatus, and thin filaments distributed in supra- and perinuclear cytoplasm. Striated ducts are absent. Excretory ducts, coming together in the main duct, are lined by a bistratified epithelium. The inner layer consists of columnar cells showing bundles of tonofilaments with scarce secretory activity. The outer layer is composed of basal cells lying on the basal lamina. The main excretory duct, which opens at the bottom of the vallum, shows a stratified epithelium. The outer side is composed of 2-3 layers of malpighian cells lying on the basal lamina. The inner side consists of a single layer of cuboidal-columnar cells with dense apical granules and well-developed organelles synthesizing and condensing secretions. These cells interpolate with goblet cells, rare mitochondria-rich cells, ciliated cells and numerous small globous cells showing a clear matrix and lacking secretory granules. The cilia show a 9 + 2 microtubular structure with basal bodies provided with striated rootlets. Myoepithelial cells surround with their processes the basal portions of the secretory cells and the intercalated ducts. The conclusions concern some comparative aspects and some hypothesis on the functional role of goblet cells, ciliated cells and epithelial cells lining the different ducts, also in relation to the final secretory product.  相似文献

The urinary bladder of a euryhaline marine teleost, Gillichthys mirabilis , was studied by light and electron microscopy. An enlargement of the mesonephric ducts forms a sac-like structure lined by an epithelium composed of two major cell types. Tall columnar cells continuous with the duct epithelium are characterized by a large number of mitochondria and well-developed rough and smooth endoplasmic reticulum. Tubular smooth endoplasmic reticulum is more developed in the basal cytoplasm and often opens into the extracellular space. A second cell type, the low cuboidal cells, forming most of the bladder epithelium, has fewer mitochondria. Basal cells are rarely observed and mucous cells are absent.
In seawater Gillichthys , cells of both types are separated by narrow intercellular spaces. In 5% seawater fish, the columnar cells show functional activation, as evidenced by an increased number of mitochondria and more extensive tubular smooth endoplasmic reticulum. No such changes were noted in the cuboidal cells; however, the lateral intercellular spaces are dilated probably owing to hypotonicity of the urine in the hypotonic environment. A functional difference between the two cell types is strongly suggested. The columnar cells may be responsible for active sodium uptake in hypotonic seawater environments.  相似文献

The histological characteristics of the digestive tract and the ultrastructure of mucosal cells of the stomach and intestine of rice field eel, Monopterus albus, are described to provide a basis for future studies on its digestive physiology. The digestive tract of the rice field eel is a long and coiled tube composed of four layers: mucosa, lamina propria‐submucosa, muscularis and serosa. The pharynx and oesophagus mucosa is lined with a stratified epithelium. The stomach includes the cardiac and pyloric portions and the fundus. Many gastric pits are formed by invaginations of the mucosal layer and tubular gastric glands formed by the columnar cells in the fundus. The intestine is separated from the stomach by a loop valve and divided into a proximal portion and a distal portion. The proximal intestinal epithelium consists of columnar cells with microvilli towards the lumen and goblet cells. The enterocytes are joined at the apical surface by the junctional complex, including the evident desmosomas. Numerous lysosomes and some vesicles are evident in the upper cytoplasm of the cells, and a moderate amount of endoplasmic reticulum and lysosomes are scattered in the supranuclear cytoplasm. The epithelium becomes progressively thicker and the folds containing large numbers of goblet cells are fewer and shorter in the distal portion of the intestine. At the ultrastuctural level, the columnar cells of the tubular gastric glands have numerous clear vacuoles and channels. A moderate amount of pepsinogen granules are present in the stomach. The enterocytes of the intestinal mucosa display a moderate amount of endoplasmic reticulum and lysosomes, and long and regular microvilli.  相似文献

The salivary glands of two species of Zoraptera, Zorotypus caudelli and Zorotypus hubbardi, were examined and documented mainly using transmission electron microscopy (TEM). The results obtained for males and females of the two species are compared and functional aspects related to ultrastructural features are discussed. The salivary glands are divided into two regions: the secretory cell region and the long efferent duct, the latter with its distal end opening in the salivarium below the hypopharyngeal base. The secretory region consists of a complex of secretory cells provided with microvillated cavities connected by short ectodermal ducts to large ones, which are connected with the long efferent duct. The secretory cell cytoplasm contains a large system of rough endoplasmic reticulum and Golgi apparatus producing numerous dense secretions. The cells of the efferent duct, characterized by reduced cytoplasm and the presence of long membrane infoldings associated with mitochondria, are possibly involved in fluid uptaking from the duct lumen.  相似文献

Changes in the ultrastructure of epithelial cells from long hyaline glands of male Melanoplus sanguinipes (Fabr.) (Orthoptera : Acrididae) have been examined during sexual maturation and after allatectomy. In newly emerged males, the long hyaline gland epithelium is composed of 1–3 cell layers. The cells contain almost no rough endoplasmic reticulum, inconspicuous Golgi complexes, and large numbers of free ribosomes and polysomes. Within 24 hr, the cells undergo considerable reorganization to form a 1-cell-thick layer. Changes in cytostructure include proliferation of the rough endoplasmic reticulum and the development of several elaborate Golgi complexes. The developing lumen contains a coarse fibrous material. By 3 days postemergence, columnar epithelial cells are clearly capable of considerable synthesis and export of secretory protein. Rough endoplasmic reticulum, and large, elaborate Golgi complexes are the major structural features of the cytoplasm. From day 3 to sexual maturity (day 7), no major ultrastructural changes occur, although massive accumulation of secretion in the lumen causes the epithelium to become cuboidal or flattened. Isoelectric focusing of soluble proteins from long hyaline gland secretions shows that maturing glands contain increasing numbers and quantities of secretory proteins.Allatectomy has minor effects on long hyaline gland ultrastructure. A reduction in the density of rough endoplasmic reticulum and ribosomes suggests that glands from operated males are metabolically less active. This is confirmed by qualitative and quantitative changes in the amount of secretion as revealed by isoelectric focusing. The observations are discussed in terms of the juvenile hormone control of long hyaline gland maturation.  相似文献

Males of a solitary digger wasp, the European beewolf, Philanthus triangulum, possess large mandibular glands that have been reported to produce a scent marking pheromone. We analysed the morphology and ultrastructure of these glands using light microscopy as well as scanning and transmission electron microscopy. The paired glands are located laterally in the head and each side consists of a larger and a smaller part. Both parts possess a collecting duct each with distinct openings at the mandible base. However, the collecting duct of the larger part is additionally connected to the pharynx through a lateral extension. The collecting ducts are bordered by a monolayered epithelium lined with cuticle that exhibits conspicuous ramified protuberances. About 1400 acini consisting of class 3 gland cells surround the ducts and are connected to them through conducting canals. The main components in the cytoplasm of these gland cells are mitochondria, well-developed smooth endoplasmatic reticulum, and electron lucent vesicles suggesting a high secretory activity. The connection between the large gland parts and the pharynx suggests that the secretion of the mandibular glands might not only be delivered directly onto the mandibles but might also be transported to and stored in the postpharyngeal gland.  相似文献

Electron microscopy of the cells of the follicle duct of Triops cancriformis shows that the follicular ducts are lined by a single-layered epithelium which also produces the eggshell material. The cytoplasm is rich in rough endoplasmic reticulum that synthesizes the eggshell material which subsequently aggregates into preformed vacuoles. Newly formed spheres of eggshell material are then excreted into the lumen. At the end of vitellogenesis the oocytes descend toward the longitudinal oviduct and pass through the eggshell material which fills the follicle ducts. The production of the eggshell and its chemical composition in some Phyllopoda are compared. The paper discusses the relationship between the eggshell construction and the reproductive biology of the population.  相似文献

Cytological variations of the median and the 2 lateral accessory glands of Bruchidius atrolineatus Pic (Coleoptera : Bruchidae) were examined as a function of age and the reproduction of the male. In sexually active virgin males, the secretory epithelium is columnar at emergence, but progressively flattens, and the secretions formed and stored by its cells are expelled by exocytosis into the glandular lumen. After 10 days, the male accessory glands exhibit a stage of repletion, characteristic of glands temporarily storing their secretions in their lumen. In diapausing males, the genital tract is relatively undeveloped and the accessory glands are reduced to tubules, whose lumen, surrounded by an epithelium composed of narrow cells, contains little secreted material. The presence of secretion aggregates in the secretory epithelial cells, the abundance of rough endoplasmic reticulum in them, and the release of a part of their secretions into the glandular lumen, indicate that reproductive diapause in B. atrolineatus is characterized by a decrease in the reproductive function. and not its total arrest.  相似文献

Several investigators have described in some crocodilians a row of round or oval organs, called dorsal glands, lying under scutes on each side of the dorsal midline. The function of these glands is unknown, but they are hypothesized to produce skin-conditioning secretions. We investigated the anatomy and histochemistry of the dorsal glands of adult American alligators ( Alligator mississippiensis ). Twenty to 22 pairs of glands containing a viscous, often black, material were observed lying from the mid-cervical to the anterior caudal regions in the axial musculature or on the inner surface of the dermis. The capsule of each gland consists of dense collagenous fibres and numerous short elastic fibres, and is surrounded by skeletal muscle. The single lumen is lined by one to several layers of cuboidal to columnar epithelium in varying stages of degeneration, indicating a holocrine secretory mode. The epithelial cell membranes often interdigitate and tight junctions and desmosomes occasionally are observed between them. The epithelial cells and secretory product contain slight to considerable amounts of lipid; glycoproteins may be present. Crystals exhibiting a dense core and/or layering occur in the epithelial cells and secretory product. Energy-dispersive X-ray analysis demonstrates calcium, copper, iron, lead, potassium, and zinc in the crystals. Mitochondria, vacuoles, and short segments of rough endoplasmic reticulum also occur in the cells.  相似文献

The male reproductive system of the fire ant, Solenopsis invicta Buren (Hymenoptera : Formicidae), consists of the testes, vasa efferentia, vasa deferentia, seminal vesicles, accessory glands, ejaculatory duct, wedge, aedeagal bladder, and external genitalia. The testes in newly eclosed males appear as 4 large white lobes filled with packets of sperm. Each lobe of the testes contains only one follicle. As the testes degenerate, the maturing sperm migrate through the vasa efferentia and vasa deferentia into the seminal vesicles. The seminal vesicles attach to the accessory glands, which are lined with secretory columnar epithelium. The posterior ends of the accessory glands taper and unite to form the ejaculatory duct. A sclerotized wedge is found at the junction of the accessory glands and the ejaculatory duct. An aedeagal bladder, joining the ejaculatory duct posterior to the wedge, is lined with squamous epithelium enveloped by heavy musculature. The ejaculatory duct continues posteriorly to form a distal aedeagus surrounded by 3 pairs of valves, comprising the external genitalia.  相似文献

In squirrel monkey (Saimiri sciureus) the position of submandibular glands in the neck, on either side of the trachea, more closely resembles that of rodents than that of other primates. The glands exhibit seromucous acini and mucous tubules with seromucous demilunes. Electron microscopy shows basal cytoplasmic folds and well-developed intercellular tissue spaces and canaliculi only in relation to seromucous cells. Greatly dilated cisternae of the granular endoplasmic reticulum and prominent Golgi membranes are characteristic of the mucous cells. The secretory granules of seromucous and mucous cells are morphologically distinct and indicate chemically different products for the two cell types. Histochemically, the seromucous cell shows the presence of acid mucosubstance as indicated by the PAS and Alcian blue techniques. Preliminary studies showed no appreciable quantity of amylase in submandibular glands. The intercalated duct cell is juxtaposed with the acinar cell or mucous tubule cell. Short luminal microvilli, prominent Golgi complexes and scant apical granules are notable features of intercalated duct cells. Four cell types compose the striated ducts, viz., granular light cells, agranular dark cells, vesiculated dark cells, and basal cells. Peripheral nerves are found in five different locations: in the connective tissue (interstitial), between adjacent myoepithelial and mucous-secreting cells, in the intercellular space between adjacent secretory cells, and between basal plications of striated ducts and between adjacent myoepithelial and intercalated duct cells.  相似文献

The midgut of the females of Syringophilopsis fringilla (Fritsch) composed of anterior midgut and excretory organ (=posterior midgut) was investigated by means of light and transmission electron microscopy. The anterior midgut includes the ventriculus and two pairs of midgut caeca. These organs are lined by a similar epithelium except for the region adjacent to the coxal glands. Four cell subtypes were distinguished in the epithelium of the anterior midgut. All of them evidently represent physiological states of a single cell type. The digestive cells are most abundant. These cells are rich in rough endoplasmic reticulum and participate both in secretion and intracellular digestion. They form macropinocytotic vesicles in the apical region and a lot of secondary lysosomes in the central cytoplasm. After accumulating various residual bodies and spherites, the digestive cells transform into the excretory cells. The latter can be either extruded into the gut lumen or bud off their apical region and enter a new digestive cycle. The secretory cells were not found in all specimens examined. They are characterized by the presence of dense membrane-bounded granules, 2–4 μm in diameter, as well as by an extensive rough endoplasmic reticulum and Golgi bodies. The ventricular wall adjacent to the coxal glands demonstrates features of transporting epithelia. The cells are characterized by irregularly branched apical processes and a high concentration of mitochondria. The main function of the excretory organ (posterior midgut) is the elimination of nitrogenous waste. Formation of guanine-containing granules in the cytoplasm of the epithelial cells was shown to be associated with Golgi activity. The excretory granules are released into the gut lumen by means of eccrine or apocrine secretion. Evacuation of the fecal masses occurs periodically. Mitotic figures have been observed occasionally in the epithelial cells of the anterior midgut.  相似文献

Harderian glands of the Wistar albino rats normal and adrenalectomized were investigated by light microscopy. In normal, these glands have a tubuloalveolar structure. The gland is located in the medio posterior aspect of the orbit. It is lobulated and appears homogeneous in colour and texture. Harderian gland consist of tubules with wide lumina lined by a single layer of columnar epithelial cells surrounded by myoepithelial cells within their basal lamina. It contains porphyrin pigment which is stored as solid intraluminal deposits. The glandular epithelium possesses two cell types, termed A and B. Type A cells are more numerous. The single excretory duct of the gland is directly continuous with endpieces at the hilus and opens nasally and ventrally to the third eyelid. The excretory duct is accompanied by many acini of small serous glands around it. The tubuloalveoli of the gland is not divided into lobules. There is no branched duct system within the gland. The secretion seems to be associated with porphyrins, is essentially released by exocytosis, but holocrine secretion also occurs. The single excretory duct is lined by a stratified epithelium. The gland is surrounded by a collagenous capsule. The adrenalectomy, caused degenerative changes in the glands. Epithelial height was lower than in normal gland epithelium. Most of the acini were completely disorganised. The acinar lumina were filled with porphyrin debris. The results suggest that rat harderian glands are sensitive to adrenal androgen changes in both male and female rats.  相似文献

ABSTRACT The fine structure of female accessory reproductive gland (FARG) of the adult mealworm beetle, Tenebrio molitor is studied with light and electron microscopes. The FARG is a simple tubular organ that composed of two kinds of cells-secretory epithelial cells and duct forming cells. The lumen of FARG is lined with a thin cuticle and filled with secretory materials. Each secretory epithelial cell has its peculiar end apparatus in addition to well-developed rough endoplasmic reticulum (rER), mitochondria, and secretory vesicles. They are forming basal infolding along the plasma membrane. Along the inner surface of the plasma membrane, numerous secretory vesicles are seen. The glandular secretions of the epithelial secretory cells are synthesized via rER to Golgi apparatus, and are stored in the extracellular cavity in the epithelial cell. These secretions are drained to the lumen through the end apparatus and this type of glandular secretion in the insects is type III. Histochemical reactions reveal the major component of these glandular secretions is an acid mucopolysaccharide.  相似文献

The unusual idiosomal glands of a water mite Teutonia cometes (Koch 1837) were examined by means of transmission and scanning electron microscopy as well as on semi-thin sections. One pair of these glands is situated ventrally in the body cavity of the idiosoma. They run posteriorly from the terminal opening (distal end) on epimeres IV and gradually dilate to their proximal blind end. The terminal opening of each gland is armed with the two fine hair-like mechanoreceptive sensilla (‘pre-anal external’ setae). The proximal part of the glands is formed of columnar secretory epithelium with a voluminous central lumen containing a large single ‘globule’ of electron-dense secretory material. The secretory gland cells contain large nuclei and intensively developed rough endoplasmic reticulum. Secretory granules of Golgi origin are scattered throughout the cell volume in small groups and are discharged from the cells into the lumen between the scarce apical microvilli. The distal part of the glands is formed of another cell type that is not secretory. These cells are composed of narrow strips of the cytoplasm leaving the large intracellular vacuoles. A short excretory cuticular duct formed by special excretory duct cells connects the glands with the external medium. At the base of the terminal opening a cuticular funnel strengthens the gland termination. At the apex of this funnel a valve prevents back-flow of the extruded secretion. These glands, as other dermal glands of water mites, are thought to play a protective role and react to external stimuli with the help of the hair-like sensilla.  相似文献

Electron microscopic examination of the paired coxal glands of the horseshoe crab Limulus polyphemus, focusing on urinary and vascular channels, shows six morphologically distinct regions. Each of four nephridial lobes consists of two cortical layers surrounding a medulla. The outer and inner cortexes contain blood vessels separated by a basement membrane from the urinary space lined by podocytes. Podocyte foot processes are applied to the basement membrane, interdigitate with those from other podocytes, and have a filtration slit diaphragm between them. Cortical morphology demonstrates ultrafiltration of blood, a previously undescribed function of the gland, as well as possible endocytic reabsorption of materials by the podocytes. The medulla drains into the stolon connecting the four lobes. These two areas have urinary tubules of cuboidal epithelium featuring microvillous-like apical projections, cytoplasmic vesicles and vacuoles, elaborate lateral interdigitations with septate junctions, and basal invaginations containing numerous mitochondria. These tubules are closely surrounded by blood channels, lined by a basement membrane containing embedded support cells. The medulla and stolon morphology are suggestive of both ion transport and water movement, in keeping with the gland's role in osmoregulation. The stolon empties into the end sac in the base of the most posterior lobe. It is lined by tall epithelium exhibiting apical overlap, blunt projections into the lumen, apparent endocytic vesicles, and basal plasma membrane infoldings with mitochondria. Cephalaspideans are a group of opisthobranch gastropods that comprises carnivorous and herbivorous species, allowing an investigation of the relationship between these diets and the morphofunctional features of the salivary glands. In this study, the salivary glands of the carnivorous cephalaspidean Philinopsis depicta were observed by light and electron microscopy. The secretory epithelium of these ribbon-shaped glands is formed by ciliated cells, granular cells and cells with apical vacuole. In ciliated cells the nucleus and most cytoplasmic organelles are located in the wider apical region and a very thin stalk reaches the base of the epithelium. These cells possess significant amounts of glycogen. Granular cells are packed with electron-dense secretory granules and also contain several cisternae of rough endoplasmic reticulum and Golgi stacks. The other type of secretory cell is mainly characterized by the presence of a large apical vacuole containing secretion. These cells possess high amounts of rough endoplasmic reticulum cisternae and several Golgi stacks. Vesicles with peripheral electron-dense material are also abundant, and seem to fuse to form the apical vacuole. The available data point out to a significant difference between the salivary glands of carnivorous and herbivorous cephalaspidean opisthobranchs, with an intensification of protein secretion in carnivorous species.  相似文献
