Non- H. pylori Helicobacter species (NHPHS) are associated with several important human and animal diseases. In the past year research into this group of bacteria has continued to gain attention, and novel species have been described in new niches owing to improvements in detection methods. Polymerase chain reaction and/or sequencing remain the gold standard for the detection of this genus. New insights into the pathogenesis of the NHPHS in hepatobiliary, gastric, and intestinal diseases were gained. In particular, data revealed interaction between hepatic steatosis and infectious hepatitis in the development of hepatocellular carcinoma. Evidence of an association between hepatitis C virus and Helicobacter spp. in hepatocarcinoma development was also provided; and male sex hormone signaling appeared to influence infectious hepatitis induced by Helicobacter hepaticus . More findings support an association between Helicobacter heilmannii and gastric adenocarcinoma; and in mice, mucins MUC4 and MUC5 but not MUC1 influence the colonization and pathogenesis of Helicobacter felis . Data indicated that the roles of the adaptive immune system in H. hepaticus -induced intestinal tumorigenesis are different in the small and large intestines, and environmental factors, such as bile acids may modulate H. hepaticus carcinogenic potential. New reports in the prevention and eradication of NHPHS showed a protective response against Helicobacter suis induced by vaccine administration, and a successful cross-foster rederivation method successfully eradicated Helicobacter spp. from contaminated mice litters. Overall, the studies provided insights into the pathophysiology of Helicobacter species other than Helicobacter pylori.  相似文献

Among the 372 phytophagous Hemiptera examined, 133 insects of 28 species (Coreidae 18, Pentatomidae 7, Pyrrhocoridae 2, Lygaeidae 1) were infected with trypanosomatids. Gut infections only were found in 68.4%, gut and salivary gland infections in 29.3% and salivary infections alone in 2.3%. Fifty-one cultures were isolated from 38 insects. Cultures were characterized by assay of certain ornithinc-arginine metabolism enzymes and by indirect immunofluorescence against monoclonal antibodies specific for Phytomonas spp. Ten cultures were identified as either Crithidia or Leptomonas. Twenty-one promastigote cultures had an enzyme pattern hitherto recorded only for Leishmania and 16 cultures were identified as Phytomonas.  相似文献

The lipopolysaccharides (LPS) of 19 marine bacteria were examined for size heterogeneities by using sodium dodecyl sulfate-polyacrylamide gel electrophoresis in conjunction with an LPS-specific silver staining method. Fifteen marine bacteria had an R-type LPS instead of the ladderlike LPS array characteristic of most bacteria. In addition, three marine bacteria also had a single large LPS molecule. Without constraints (e.g., surface masking), R-type LPS, a more hydrophobic molecule, predominates in Shewanella and Hyphomonas species and in other marine bacteria.  相似文献

ABSTRACT. Plant trypanosomatids cause lethal vascular wilting in palms of the Arecaceae family. Infections, affecting plants in South and Central America, can result in significant economic loss. The study of trypanosomatids that cause these diseases has been complicated due to the inability to culture these organisms for in vitro analyses. To develop a protocol that would facilitate studies of trypanosomatids, continuous in vitro cultures of phloemic trypanosomatids were established from apical stems of diseased coconut trees collected in endemic and non-endemic regions of Brazil (the states of Bahia and Rio de Janeiro, respectively). Although attempts at establishing axenic cultures were unsuccessful, it was found that trypanosomatid co-cultures could be successfully established and maintained. The procedure was to preculture media with 104 Aedes albopictus cells in Grace's medium supplemented with 10% heat-inactivated fetal bovine serum (without antibiotics or fungicides) for 3 d before adding 106 trypanosomatids/ml harvested from either fresh apical stem extracts or with 2 mm3 fragments of coconut apical stems. By day 7 under these conditions the parasites grew exponentially. Using this strategy, two isolates were identified and have been maintained in our laboratory for over 400 passages, demonstrating the efficacy of this culturing procedure. In situ the organisms were observed in vascular bundles and inside sieve elements of the phloem of diseased palms. In vitro parasites retained their mobility. Morphometric analysis revealed differences between Bahia and Rio de Janeiro isolates.  相似文献

The uptake kinetics of sorbitol, sucrose, glucose and fructoseacross the plasma membrane using protoplasts isolated from applefruit flesh (Malus pumila Mill. var. domestica Schneid.) wasinvestigated. When sorbitol was taken up into the cell, PCMBS-sensitivesaturable transport was distinguishable from the diffusive transport.At a low sorbitol concentration, the saturable transport systemaccounted for more than 50% of the total uptake, whereas ata high concentration the diffusive transport system was moredominant. The saturable transport was suggested be a carrier-mediatedtransport system coupled with ATP because the system was inhibitedCCCP or orthovanadate. The Km value for sorbitol was computedto be 3.6mM. A carrier-mediated transport system coupled withATP was also observed for glucose and fructose with correspondingKm values of 5.0 and 2.5 mM. However, no saturable transportfor sucrose was observed over a range of 0.1 to 10 mM sucroseconcentration. ABSTRACT. We report the characterization of cell-associated and extracellular peptidases of Bodo sp., a free-living flagellate of the Bodonidae family, order Kinetoplastida, which is considered ancestral to the trypanosomatids. This bodonid isolate is phylogenetically related to Bodo caudatus and Bodo curvifilus . The proteolytic activity profiles of Bodo sp. were determined by sodium dodecyl sulfate-polyacrylamide gel electrophoresis containing co-polymerized gelatin, casein, hemoglobin, or bovine serum albumin as substrates. The enzymatic complex degraded gelatin better in acidic pH, and under these conditions four proteolytic bands (120, 100, 90, and 75 kDa) were detected in the cellular or extracellular extracts. Two peptidases (250 and 200 kDa) were exclusively detected with the substrate casein. All these enzymes belong to the serine peptidase class, based on inhibition by aprotinin and phenylmethylsulfonyl fluoride. This is the first biochemical characterization of peptidases in a free-living Bodo sp., potentially providing insight into the physiology of these protozoa and the evolutionary importance of peptidases to the order Kinetoplastida as some of these enzymes are important virulence factors in pathogenic trypanosomatids.  相似文献

A total of 139 surface water samples from seven lakes and 15 rivers in southwestern Finland were analyzed during five consecutive seasons from autumn 2000 to autumn 2001 for the presence of various enteropathogens (Campylobacter spp., Giardia spp., Cryptosporidium spp., and noroviruses) and fecal indicators (thermotolerant coliforms, Escherichia coli, Clostridium perfringens, and F-RNA bacteriophages) and for physicochemical parameters (turbidity and temperature); this was the first such systematic study. Altogether, 41.0% (57 of 139) of the samples were positive for at least one of the pathogens; 17.3% were positive for Campylobacter spp. (45.8% of the positive samples contained Campylobacter jejuni, 25.0% contained Campylobacter lari, 4.2% contained Campylobacter coli, and 25.0% contained Campylobacter isolates that were not identified), 13.7% were positive for Giardia spp., 10.1% were positive for Cryptosporidium spp., and 9.4% were positive for noroviruses (23.0% of the positive samples contained genogroup I and 77.0% contained genogroup II). The samples were positive for enteropathogens significantly (P < 0.05) less frequently during the winter season than during the other sampling seasons. No significant differences in the prevalence of enteropathogens were found when rivers and lakes were compared. The presence of thermotolerant coliforms, E. coli, and C. perfringens had significant bivariate nonparametric Spearman's rank order correlation coefficients (P < 0.001) with samples that were positive for one or more of the pathogens analyzed. The absence of these indicators in a logistic regression model was found to have significant predictive value (odds ratios, 1.15 x 10(8), 7.57, and 2.74, respectively; P < 0.05) for a sample that was negative for the pathogens analyzed. There were no significant correlations between counts or count levels for thermotolerant coliforms or E. coli or the presence of F-RNA phages and pathogens in the samples analyzed.  相似文献

The coastal ecosystems of California are highly utilized by humans and animals, but the ecology of fecal bacteria at the land–sea interface is not well understood. This study evaluated the distribution of potentially pathogenic bacteria in invertebrates from linked marine, estuarine, and freshwater ecosystems in central California. A variety of filter-feeding clams, mussels, worms, and crab tissues were selectively cultured for Salmonella spp., Campylobacter spp., Escherichia coli-O157, Clostridium perfringens, Plesiomonas shigelloides, and Vibrio spp. A longitudinal study assessed environmental risk factors for detecting these bacterial species in sentinel mussel batches. Putative risk factors included mussel collection near higher risk areas for livestock or human sewage exposure, adjacent human population density, season, recent precipitation, water temperature, water type, bivalve type, and freshwater outflow exposure. Bacteria detected in invertebrates included Salmonella spp., C. perfringens, P. shigelloides, Vibrio cholerae, Vibrio parahaemolyticus, and Vibrio alginolyticus. Overall, 80% of mussel batches were culture positive for at least one of the bacterial species, although the pathogens Campylobacter, E. coli-O157, and Salmonella were not detected. Many of the same bacterial species were also cultured from upstream estuarine and riverine invertebrates. Exposure to human sewage sources, recent precipitation, and water temperature were significant risk factors for bacterial detection in sentinel mussel batches. These findings are consistent with the hypothesis that filter-feeding invertebrates along the coast concentrate fecal bacteria flowing from land to sea and show that the relationships between anthropogenic effects on coastal ecosystems and the environmental niches of fecal bacteria are complex and dynamic.  相似文献

The specific aim of this study was to assess the faecal shedding of zoonotic enteropathogens by semi-domesticated reindeer (Rangifer tarandus tarandus) to deduce the potential risk to human health through modern reindeer herding. In total, 2,243 faecal samples of reindeer from northern regions of Finland and Norway were examined for potentially enteropathogenic bacteria (Campylobacter species, Enterococcus species, Escherichia coli, Salmonella species and Yersinia species) and parasites (Cryptosporidium species) in accordance with standard procedures. Escherichia coli were isolated in 94.7%, Enterococcus species in 92.9%, Yersinia species in 4.8% of the samples and Campylobacter species in one sample only (0.04%). Analysis for virulence factors in E. coli and Yersinia species revealed no pathogenic strains. Neither Salmonella species nor Cryptosporidium oocysts were detected. The public health risk due to reindeer husbandry concerning zoonotic diseases included in this study has to be considered as very low at present but a putative epidemiological threat may arise when herding conditions are changed with respect to intensification and crowding.  相似文献

ABSTRACT. A large number of trypanosomatids was seen in the latex of the milkweed plant Euphorbia hyssopifolia found in Rio de Janeiro, Brazil. The parasites, which belong to the genus Phytomonas, were isolated and axenically cultivated in a biphasic culture medium. The form and the structural organization of the parasites as found in the intact plant, in the isolated latex, and in the culture medium was studied by scanning and transmission electron microscopy.  相似文献

Thein vitroculture of immature soybean cotyledons (in direct contact with the medium) and immature fruit explants (stem dipping into the medium) on a defined medium containing glutamine and sulphate as sole sources of N and S for 7 d led to rates of growth and reserve protein accumulation close to, or greater than, those occurringin situ. Supplementation of the medium with 8.4 mMmethionine had little effect on growth and protein accumulation of the cotyledons in the explant system, but did result in significant increases in the isolated cotyledon system. Methionine suppressed the synthesis of the 7S β-subunit in both systems. The free amino pool of the cotyledons increased more than three-fold when methionine was present in the explant medium. In the isolated cotyledon system, the basal medium alone caused a large increase (over 30-fold) in the free amino acid fraction, but methionine resulted in an even greater increase (over 50-fold). In both systems the expansion involved a very large increase in the methionine pool, but many other amino acids also showed large increases. Specific effects of methionine on individual amino acids were more clear in the explant system, where its presence resulted in marked increases in serine, alanine and asparagine. The data show that an abnormal situation arises on feeding with methionine, a fact to be considered before attributing effects on growth and protein synthesis directly to methionine.  相似文献

Rapid identification of enteropathogenic bacteria in stool samples is critical for clinical diagnosis and antimicrobial therapy. In this study, we describe the development of an approach that couples multiplex PCR with hybridization to a DNA microarray, to allow the simultaneous detection of the 10 pathogens. The microarray was synthesized with 20-mer oligonucleotide probes that were designed to be specific for virulence-factor genes of each strain. The detection limit for genomic DNA from a single strain was approximately 10 fg. In the presence of heterogeneous non-target DNA, the detection sensitivity of the array decreased to approximately 100 fg. We did not observe any non-specific hybridization. In addition, we successfully used this oligonucleotide-based DNA microarray to identify the causative agents in clinical stool samples from patients with food-borne enteritis.  相似文献

The growth characteristics of five bacteria, Brevibacterium aurantiacum 1-16-58, Corynebacterium casei DPC 5298T, Corynebacterium variabile DPC 5310, Microbacterium gubbeenense DPC 5286T, and Staphylococcus saprophyticus 4E61, all of which were isolated from the surface of smear cheese, were studied in complex and chemically defined media. All of the coryneforms, except M. gubbeenense, grew in 12% salt, while B. aurantiacum and S. saprophyticus grew in 15% salt. All five bacteria assimilated lactate in a semisynthetic medium, and none of the coryneform bacteria assimilated lactose. Glucose assimilation was poor, except by S. saprophyticus and C. casei. Five to seven amino acids were assimilated by the coryneforms and 12 by S. saprophyticus. Glutamate, phenylalanine, and proline were utilized by all five bacteria, whereas utilization of serine, threonine, aspartate, histidine, alanine, arginine, leucine, isoleucine, and glycine depended on the organism. Growth of C. casei restarted after addition of glutamate, proline, serine, and lactate at the end of the exponential phase, indicating that these amino acids and lactate can be used as energy sources. Pantothenic acid was essential for the growth of C. casei and M. gubbeenense. Omission of biotin reduced the growth of B. aurantiacum, C. casei, and M. gubbeenense. All of the bacteria contained lactate dehydrogenase activity (with both pyruvate and lactate as substrates) and glutamate pyruvate transaminase activity but not urease activity.  相似文献

Twenty years ago an Arctic cryptophyte was isolated from Baffin Bay and given strain number CCMP 2045. Here, it was described using morphology, water‐ and non‐water soluble pigments and nuclear‐encoded SSU rDNA . The influence of temperature, salinity, and light intensity on growth rates was also examined. Microscopy revealed typical cryptophyte features but the chloroplast color was either green or red depending on the light intensity provided. Phycoerythrin (Cr‐PE 566) was only produced when cells were grown under low‐light conditions (5 μmol photons · m?2 · s?1). Non‐water‐soluble pigments included chlorophyll a , c 2 and five major carotenoids. Cells measured 8.2 × 5.1 μm and a tail‐like appendage gave them a comma‐shape. The nucleus was located posteriorly and a horseshoe‐shaped chloroplast contained a single pyrenoid. Ejectosomes of two sizes and a nucleomorph anterior to the pyrenoid were discerned in TEM . SEM revealed a slightly elevated vestibular plate in the vestibulum. The inner periplast component consisted of slightly overlapping hexagonal plates arranged in 16–20 oblique rows. Antapical plates were smaller and their shape less profound. Temperature and salinity studies revealed CCMP 2045 as stenothermal and euryhaline and growth was saturated between 5 and 20 μmol photons · m?2 · s?1. The phylogeny based on SSU rDNA showed that CCMP 2045 formed a distinct clade with CCMP 2293 and Falcomonas sp. isolated from Spain. Combining pheno‐ and genotypic data, the Arctic cryptophyte could not be placed in an existing family and genus and therefore Baffinellaceae fam. nov. and Baffinella frigidus gen. et sp. nov. were proposed.  相似文献

本研究分别从台湾槟榔病果中分离出病原菌Ar8904、Ar8905;从海南槟榔病果中分离出病原菌Ar8801、Ar8802、Ar8904以及从云南槟榔病果中分离出病原菌Ar8901、Ar892.将这些病原菌进行对比实验.研究结果表明,这7株病原菌在菌株形态、培养形状、寄主范围、致病性以及症状特征都十分的相似.因此,根据炭疽菌现代分类标准,我们可以初步确认台湾、海南、云南的槟榔病果均为同一种炭疽菌引起的.  相似文献

ABSTRACT. Two new hemimastigophoran flagellates are described using light and electron microscopy, and the family Spironemidae is revised. Spironema terricola n. sp. occurs in soil from the Grand Canyon (southwest USA). It moves in a conspicuously euglenoid manner and differs from S. multiciliatum Klebs by its vermiform shape and shorter kinetics. Spironema terricola is similar to Goodey's Spironema multiciliatum from soil in England. However, Goodey's vermiform species has a very elongate nucleus and is thus neither identical with S. terricola , which has a roundish nucleus, nor with Klebs' lanceolate S. multiciliatum ; we consider it a new species, Spironema goodeyi n. sp, Stereonema geiseri n. g., n. sp. was discovered in the Aufwuchs (periphyton) of a river in Bavaria, Germany. the new genus differs from Spironema by its a contractility, and from Hemimastix by its shorter kinetics and less plicate cortex. the fine structure of Spironema and Stereonema is very similar to that of Hemimastix Foissner et al., viz., the cortex is composed of two plates having diagonal symmetry and the flagellated basal bodies have associated a short and a long microtubular ribbon. All species have unique extrusomes of the same type. the main differences between the three genera and five species recognized are contractility, length of kinetics, body size, shape of cell and nucleus, and particulars of the cortex and extrusomes. the phylogenetic relationships of the Hemimastigophora are still uncertain. However, the diagonal symmetry of the cortical plates and the pronounced euglenoid movement of Spironema spp. suggest a common ancestor with euglenids.  相似文献

A group of 277 yeasts isolated from burned children and 14 reference strains were tested for phosphatase activity by using phenolphthalein phosphate substrates. Phosphatase activity was widely distributed among various species and strains representing seven genera. Candida albicans, which was the most common yeast isolated from clinical material, was notably absent in producing the enzyme, whereas Candida tropicalis was the most consistent, strong, and rapidly active phosphatase-producing organism. The characteristic enzyme activity of a selected isolate of C. tropicalis was demonstrated in the presence of concentrations of inorganic phosphate which inhibited enzyme activity of other species. The greater enzyme activity of C. tropicalis was not related to more rapid or greater cell growth or decrease in the pH of culture media. Extracellular constitutive heat-labile acid phosphatase was found in broth filtrates of C. tropicalis, C. krusei, and a strain of Staphylococcus aureus.  相似文献
