目的 对新型消毒剂邻苯二甲醛(OPA)的消化内镜消毒效果与戊二醛的进行比较研究.方法 用细菌培养的方法随机研究OPA与戊二醛消毒消化内镜的效果,并比较其临床优劣.结果 有效浓度的OPA和戊二醛在按照要求使用的情况下,所有检测的内镜细菌培养都未见细菌生长,但前者消毒所需时间更短,临床使用更安全.结论 OPA可以有效的对消化内镜进行消毒,与戊二醛相比有临床优势.  相似文献

邻苯二甲醛(OPA)法检测铝吸附疫苗中抗原含量   总被引:1,自引:0,他引:1  
目的:建立快速测定铝胶吸附的疫苗中抗原含量的OPA荧光检测法。 方法:利用邻苯二甲醛(OPA)在2-巯基乙醇存在下与氨基酸的N端或L-谷氨酸侧链反应,在460nm处生成荧光衍生物的原理,建立了无需进行抗原蛋白提取的检测方法,并对该方法的特异性、线性、精密度、回收率、重复性进行考察。同时,配合钠十二烷基的硫酸盐-聚丙烯酰胺凝胶电泳法(SDS-PAGE),研究在配制过程中抗原蛋白与佐剂是否分离。结果: OPA荧光法,在抗原蛋白含量为0.02~0.16mg/ml时,线性良好,准确度达90%~115%,批内和批间相对标准偏差≤15%。电泳结果显示,CpG的加入可能会导致少量抗原蛋白从铝胶中解离。结论:该方法快速、准确、灵敏度较高,重复性好,可以应用于含铝胶疫苗制剂的实验室甚至生产质控过程。  相似文献

本实验用邻苯二甲醛-β-巯基乙醇衍生化、反相梯度洗脱、荧光检测分析法测定豚鼠脑的氨基酸。60min内可测20种氨基酸。脑组织样本的预处理简单,平均回收率达95%。氨基酸保留时间的变异系数平均为1.1±0.3%(均值±标准差);峰面积的变异系数平均为2.9+0.8%;在10pmol—2nmol范围内,线性关系的相关系数平均为0.987±0.009。本文并对实验所用的色谱条件作了讨论。  相似文献

OPT修饰GAPDH及gGAPDH的荧光衍生物与Trp残基之间存在非辐射的能量传递。在不同浓度的GuHCl溶液中,糖基化和非糖基化酶OPT衍生物的荧光的变化具有一定的差异。特别是两者的荧光在碘化钾溶液中的淬来有明显的不同。OPT修饰动力学研究表明,gGAPDH的修饰速度快于GAPDH的修饰速度。以上结果提示:糖基化的位点可能在赖氨酸残基上,并且被糖基化的残基可能位于或靠近活性部位。  相似文献

研究复合季铵盐消毒剂含量的测定与滴定误差分析。探讨了四苯硼钠标准液配制及对滴定分析终点颜色的影响,并与单季铵盐定量分析方法进行了对比。结果表明:四苯硼钠两相滴定法适合于复合季铵盐定量分析,但要注意四苯硼钠标准液配制的正确性及滴定终点颜色的判断。  相似文献

A series of model peptides in the form of quaternary ammonium salts at the N‐terminus was efficiently prepared by the solid‐phase synthesis. Tandem mass spectrometric analysis of the peptide quaternary ammonium derivatives was shown to provide sequence confirmation and enhanced detection. We designed the 2‐(1,4‐diazabicyclo[2.2.2] octylammonium)acetyl quaternary ammonium group which does not suffer from neutral losses during MS/MS experiments. The presented quaternization of 1,4‐diazabicyclo[2.2.2]octane (DABCO) by iodoacetylated peptides is relatively easy and compatible with standard solid‐phase peptide synthesis. Twenty-four novel 5-phenyl-1,3,4-oxadiazole-2-thiol (POT) analogues, benzo[d]oxazole-2-thiol, benzo[d]thiazole-2-thiol and 5-methyl-1,3,4-thiadiazole-2-thiol-substituted N,N-bis(2-hydroxyethyl) quaternary ammonium salts (QAS) (5a-d, 6a-d, 7a-d, 10a-d, 13a-d, 16a-d) were prepared and characterised by FTIR, NMR and elemental analysis. Part of target compounds (5d, 6d, 7d, 10d, 13d, 16d) displayed potent antimicrobial effect against ten common pathogens (S. aureus, α-H-tococcus, β-H-tococcus, E. coli, P. aeruginosa, Proteus vulgaris, Canidia Albicans, Cytospora mandshurica, Physalospora piricola, Aspergillus niger) and had relatively low cytotoxity against two human cell lines (HaCat and LO2). TEM and SEM images of E. coli and S. aureus morphologies treated with 7d showed that the antibacterial mechanism might be the QAS fixing on cell wall surfaces and puncturing to result in the release of bacterial cytoplasm. This study provides new information of QAS, which could be used to design novel antimicrobial agents applied in clinic or agriculture.  相似文献

With the view of incorporating quaternary ammonium salts (QAs) in marine paints, nineteen of these were tested against a community of marine bacteria, at a temperature and salinity close to those of seawater. The concentration of QAs and the length of the main substituting chain are the main parameters affecting the growth and adhesion of bacteria, but the nature of (i) the other chains, (ii) the counter‐ion and (iii) the rings when inserted in the QA molecule also influenced the bacteria. Increasing the concentration of the QAs decreased the growth rate of the bacteria, the maximum cell density at the plateau and the rate of adhesion. The effect of increasing the length of the main chain depended on the range of carbon numbers. Below 7 carbon atoms, the growth rate was not significantly modified, but the numbers of cells at the plateau increased in contrast with the adhesion rate which decreased rapidly. Increasing the length of the chain to between 7 and 16 carbon atoms resulted in a decrease in the growth rate, a decrease and then a stabilisation in the numbers of cells at the plateau and no further change in the adhesion rate. Possibly an increase in growth rate, adhesion rate and in the numbers of cells at the plateau may occur above 16 carbon atoms. In contrast, the length of the other chains influenced positively the cell concentration at the plateau, and more generally the efficiency of QAs decreased substantially when these chains had the same numbers of carbon atoms. QAs with iodide as counter‐ion were more effective than those with chloride or bromide and phenyl was more effective than benzyl as rings inserted in QAs. The minimum inhibitory concentrations (MIC) were often very high if compared to standard methods with laboratory strains, and this can be tentatively explained by the dominance of Gram— bacteria in the community assayed, the development of resistant strains in the cultures used with time and the presence of organic matter in the culture medium.  相似文献

The syntheses have been developed for quaternary N-(1,4-anhydro-5-deoxy-2,3-O-isopropylidene-D,L-ribitol-5-yl)ammonium salts derived from five aromatic amines, pyridine, 2-methylpyridine, 3-carbamoylpyridine, 4-(N,N-dimethylamino)pyridine, and quinoline, as well as two tertiary aliphatic amines, trimethylamine and triethylamine. Reactions of 1,4-anhydro-2,3-O-isopropylidene-5-O-tosyl-D,L-ribitol with tri-n-propylamine and tri-n-butylamine were unsuccessful. The products were identified on the basis of their 1H and 13C NMR spectra. The structure of N-(1,4-anhydro-5-deoxy-2,3-O-isopropylidene-D,L-ribitol-5-yl)trimethylammonium tosylate was additionally elucidated by X-ray diffractometry.  相似文献
