Sensitive two-color fluorescence staining and cell-sorting techniques were used to isolate a T4+T8+ thymocyte subpopulation with high T3 density from human thymocyte cultures. Previously, this population was shown to give rise to both T4+T8- and T8+T4- thymocytes. In the present study, this T3+T4+T8 population was shown to be functionally as well as phenotypically distinct from either T8+T4- cells or T4+T8- cells present in the same culture. The T3+T4+T8+ cell had intermediate cytotoxic capacities relative to T8+T4- and T4+T8- thymocyte fractions. The proliferative capacity of the T4+T8+ population although less than that of the T8+T4- subset exceeded the proliferative response of the T4+T8- population. The time of appearance of large numbers of T3+T4+T8+ cells in culture as well as functional properties exhibited by T3+T4+T8+ cells are consistent with the notion that the T3+T4+T8+ cell represents an activated intermediate in thymocyte differentiation. The T3+T4+T8+ thymocyte may be an important intermediate in in vivo as well as in in vitro thymic differentiation. Moreover, the analysis of its functional properties may contribute to an understanding of functional responses exhibited by the most mature (T3+) population isolated from human thymus.  相似文献

Biosynthetic labeling with 35S-methionine and 35S-cysteine of isolated T4+ cells from Con A-activated T cells demonstrated that the T8 antigen was synthesized by activated T4+ cells. Two-color fluorescence analysis of the activated T cell population from which the T4+ fraction was obtained showed that both T4+T8- and T4+T8+ cells were present. The T8 antigen that was immunoprecipitated by monoclonal anti-T8 from activated T4+ cells migrated with an electrophoretic mobility corresponding to an m.w. of approximately 33,000, a previously reported m.w. value for T8 antigen. Con A activation of highly purified peripheral T4+T8- and T8+T4- subsets indicated that both T4+T8- and T8+T4- cells can give rise to T4+T8+ cells. However, substantial T4, T8 coexpression by T4+T8- cells required a signal from T8+T4- cells which could be supplied by incubating T4+T8- cells with irradiated T8+ cells or the supernatant from Con A-activated T8+T4- cells. The generation of T4+T8+ cells from a subset of T4+T8- T cells may be an important mechanism in immune activation and/or the further differentiation of peripheral T4+ cells.  相似文献

Pronunciation variation is ubiquitous in the speech signal. Different models of lexical representation have been put forward to deal with speech variability, which differ in the level as well as the nature of mental representation. We present the first mismatch negativity (MMN) study investigating the effect of allophonic variation on the mental representation and neural processing of lexical tones. Native speakers of Standard Chinese (SC) participated in an oddball electroencephalography (EEG) experiment. All stimuli have the same segments (ma) but different lexical tones: level [T1], rising [T2], and dipping [T3]. In connected speech with a T3T3 sequence, the first T3 may undergo allophonic change and is produced with a rising pitch contour (T3V), similar to the lexical T2 pitch contour. Four oddball conditions were constructed (T1/T3, T3/T1, T2/T3, T3/T2; standard/deviant). All four conditions elicited MMN effects, with the T1–T3 pair eliciting comparable MMNs, but the T2–T3 pair asymmetrical MMN effects. There were significantly greater and earlier MMN effects in the T2/T3 condition than that in the reversed T3/T2 condition. Furthermore, the T3/T2 condition showed more rightward MMN effects than the T2/T3 condition and the T1–T3 pair. Such asymmetries suggest co-activation of long-term memory representations of both T3 and T3V when T3 serves as the standard. The acoustic similarity between the activated T3V (by the standard T3) and the incoming deviant stimulus T2 induces acoustic processing of the tonal contrast in the T3/T2 condition, similar to that of within-category lexical tone processing, which is in contrast to the processing of between-category lexical tones observed in the T2/T3, T1/T3, and T3/T1 conditions.  相似文献

By using sensitive three-color fluorescence flow cytometric techniques, we were able to identify a T4+T8+ thymocyte with high T3 surface density (T3H) representing 4 to 9% of thymocytes. To characterize the T3HT4+T8+ cell, thymic subpopulations with high T3 surface density (T3H) and lower T3 density (T3L/T3-) were compared with regard to T6 expression. The T3H subpopulation was characterized by lower numbers of T6+ cells and reduced levels of T6 antigen density, whereas the T3L/T3- population was greater than 90% T6+ and expressed this antigen at high cell surface density. In addition, T3H fractions appeared to possess higher levels of nuclear activation with respect to the T3L/T3- population as indicated by increased log 90 degrees scatter profiles. These results suggest that thymocytes with high T3 surface expression are not only more differentiated, but also more activated than the majority of the thymic population. The T3HT4+T8+ fraction could be distinguished from T4+T8+ thymocytes with lower T3 density not only by an increased log 90 degrees scatter profile, but also by the presence of T4+T8+ cells with reduced levels of T8 surface antigen. Our results indicate that T4+T8+ thymocytes with high T3 surface density are a distinct subpopulation and may represent the immediate precursors of the phenotypically more mature T3HT4+T8- and T3HT8+T4- subpopulations found in human thymus.  相似文献

The effect of induced erythrocythemia on hemoglobin concentration ([Hb]) and aerobic work capacity was determined for nine women. Cycle tests were performed at prereinfusion (T1), 2 days after a placebo infusion (T2), 2 days postreinfusion of 334 ml of red blood cells (T3), 8 days postreinfusion (T4), and 14 days postreinfusion (T5). T1 and T2 responses did not differ, negating a placebo effect. [Hb] increased from 12.7 g X dl at T1 to 14.7 g X dl at T3 and then remained constant at T4 and T5. Hematocrit increased from 38.1% at T1 to 44.9% at T3 and then remained constant at T4 and T5. Submaximal O2 uptake (VO2) and stroke volume (SV) did not change from T1 through T5. Submaximal cardiac output (Q) and heart rate (HR) decreased from T1 to T3 and then remained constant at T4 and T5. Arteriovenous O2 difference increased from T1 to T3 and then remained constant at T4 and T5. Maximal VO2 was greater at T3 (2.65 l X min-1), T4 (2.66 l X min-1), and T5 (2.60 l X min-1) than at T1 (2.41 l X min-1). Physical work capacity was greater at T3 (10,740 kg X m), T4 (10,980 kg X m), and T5 (10,380 kg X m) than at T1 (8,747 kg X m). Maximal values for Q, HR, and SV were unchanged from T1 through T5. At maximum, arteriovenous O2 difference and Hb flow rate increased from T1 to T3 and then remained constant at T4 and T5. The greater postreinfusion [Hb] improved O2 transport capacity and appeared to regulate circulatory responses.  相似文献

Effects of endosulfan, malathion and carbaryl on circulating T4, T3 and T3/T4 ratio in the freshwater catfish, C. batrachus have been studied during reproductively active phases, i.e. vitellogenic and post-vitellogenic phases of its annual reproductive cycle. 2. Endosulfan provoked a significant increase in T4 and a decrease in T3 as well as the T3/T4 ratio. 3. Malathion decreased T3 and T3/T4 ratio without altering the level of T4 in circulation. 4. Carbaryl caused a decline in the level of T4 and an elevation in T3 and T3/T4 ratio.  相似文献

Maturing thymocytes express a series of cell surface glycoproteins which can be identified by monoclonal antibodies. The stage II or common thymocyte expresses the phenotype T4+T8+T6+T3-. In response to unknown signals, but presumably involving interactions with products of the major histocompatibility complex, the thymocyte suppresses either the T8 or T4 gene, becoming committed to the T4+T8- or T4-T8+ phenotype. With maturation, the thymocyte also becomes T6-T3+. To study whether DNA methylation may be involved in regulating expression of these determinants in mature T cells, we treated cloned interleukin 2-dependent T8- and T4-bearing T cells with 5-azacytidine (5-azaC), a nucleoside analog which inhibits methylation of newly synthesized DNA. In this report, we show that T8+ T cells treated with 5-azaC express the phenotype T8+T4+T6-T3+. Treatment of the same cells with hydroxyurea, an inhibitor of DNA synthesis, failed to induce T4 on T8+ cells. These results suggest that expression of the T4 gene may be suppressed by DNA methylation in mature T8+ cells.  相似文献

Aspects of the morphology of II species of Triviidae are described, including the living animal, shell, radula, mantle complex, central nervous system and reproductive system. Interspecific differences in radular morphology are important in distinguishing closely allied species. Trivia aperta, T. costata and T. verhoeft are protandric hermaphrodites while T. caluariola, T. neglccta, T. pellucidula, T. rubra, T. solandri and T. suavis are gonochoric. The elaboration of the receptaculum seminis varies between species. In T. aperta, T. calvariola, T. neglecta and T. who& the receptaculum is undivided while in T. costafa, T. pellucidula, T. solandri and T. suavis there are two or more lobes of the receptaculum.  相似文献

Activation of protein kinase C-zeta (PKC-zeta) by insulin requires phosphatidylinositol (PI) 3-kinase-dependent increases in phosphatidylinositol-3,4,5-(PO(4))(3) (PIP(3)) and phosphorylation of activation loop and autophosphorylation sites, but actual mechanisms are uncertain. Presently, we examined: (a) acute effects of insulin on threonine (T)-410 loop phosphorylation and (b) effects of (i) alanine (A) and glutamate (E) mutations at T410 loop and T560 autophosphorylation sites and (ii) N-terminal truncation on insulin-induced activation of PKC-zeta. Insulin acutely increased T410 loop phosphorylation, suggesting enhanced action of 3-phosphoinositide-dependent protein kinase-1 (PDK-1). Despite increasing in vitro autophosphorylation of wild-type PKC-zeta and T410E-PKC-zeta, insulin and PIP(3) did not stimulate autophosphorylation of T560A, T560E, T410A/T560E, T410E/T560A, or T410E/T560E mutant forms of PKC-zeta; thus, T560 appeared to be the sole autophosphorylation site. Activating effects of insulin and/or PIP(3) on enzyme activity were completely abolished in T410A-PKC-zeta, partially compromised in T560A-PKC-zeta, T410E/T560A-PKC-zeta, and T410A/T560E-PKC-zeta, and largely intact in T410E-PKC-zeta, T560E-PKC-zeta, and T410E/T560E-PKC-zeta. Activation of the T410E/T560E mutant suggested a phosphorylation-independent mechanism. As functional correlates, insulin effects on epitope-tagged GLUT4 translocation were compromised by expression of T410A-PKC-zeta, T560A-PKC-zeta, T410E/T560A, and T410A/T560E-PKC-zeta but not T410E-PKC-zeta, T560E-PKC-zeta, or T410E/T560E-PKC-zeta. Insulin, but not PIP(3), activated truncated, pseudosubstrate-lacking forms of PKC-zeta and PKC-lambda by a wortmannin-sensitive mechanism, apparently involving PI 3-kinase/PDK-1-dependent phosphorylations but independent of PIP(3)-dependent conformational activation. Our findings suggest that insulin, via PIP(3), provokes increases in PKC-zeta enzyme activity through (a) PDK-1-dependent T410 loop phosphorylation, (b) T560 autophosphorylation, and (c) phosphorylation-independent/conformational-dependent relief of pseudosubstrate autoinhibition.  相似文献

在扫描电镜下观察了豆科黄华属Thermopsis 18种植物种子的表面纹饰,发现 T.alpina,T.bar- bata,T.inflata,T.lupinoides,T. licentiana,T.smithiana和T.turkestanica的种子表面为粗网状,T californica,T.divaricarpa,T. macrophylla,T.mollis的种子表面为细网状,T.gracilis,T.montana,T. fabacea的种子表面为相对平滑型纹饰,T.alterniflora的种子表面为不规则条形,T.chinensis的种子表 面为粘膜状,T.rhombifolia的种子表面为条形及 T.viuosa的种子表面为碎屑状纹饰。结果表明黄华属的种子表面特征对属下类群的划分有一定意义,对澄清某些混乱的种有一定价值。  相似文献

Sequences of the internal transcribed spacer region 1 (ITS1) of the ribosomal DNA were used to determine the phylogenetic relationships of species of Trichoderma sect. Pachybasium. To this end, 85 strains-including all the available ex-type strains-were analyzed. Parsimony analysis demonstrated that the section is nonmonophyletic, distributing the 85 strains among three main groups that were supported by bootstrap values. Group A comprises two clades (A1 and A2), with A1 including T. polysporum, T. piluliferum, and T. minutisporum, while A2 included T. hamatum, T. pubescens, and T. strigosum in addition to species previously included in sect. Trichoderma (i.e., T. viride, T. atroviride, and T. koningii). The ex-type strain of T. fasciculatum formed a separate branch basal to clade A. Clade B contained the sect. Pachybasium members T. harzianum, T. fertile, T. croceum, T. longipile, T. strictipile, T. tomentosum, T. oblongisporum, T. flavofuscum, T. spirale, and the anamorphs of Hypocrea semiorbis and H. cf. gelatinosa. Sequence differences among clades A1, A2, and B were in the same order of magnitude as between each of them and T. longibrachiatum, which was used as an outgroup in these analyses. Sequence differences within clades A1, A2, and B were considerably smaller: in some cases (i.e., T. virens and T. flavofuscum; T. strictipile and H. cf. gelatinosa), the ITS1-sequences were identical, suggesting conspecifity. In other cases (e.g., T. crassum and T. longipile; T. harzianum, T. inhamatum, T. croceum, T. fertile, and H. semiorbis; T. hamatum and T. pubescens; and T. viride, T. atroviride, and T. koningii) differences were in the range of 1-3 nt only, suggesting a very close phylogenetic relationship. The sequence of a previously described aggressive mushroom competitor group of T. harzianum strains (Th2) was strikingly different from that of the ex-type strain of T. harzianum and closely related species and is likely to be a separate species. 利用光学显微镜和扫描电镜对小麦印度腥黑粉菌及其近似种的形态学特征进行了系统研究。T. indica和T. horrida不同菌株冬孢子大小变化范围均较大。在所研究的种中,T. indica与T. walkeri最近似,前者冬孢子大小平均值比后者略大,分别为:38.35?5.92祄和32.86?1.53祄。T. indica与其它具有疣状或刺状突起的腥黑粉菌:T. horrida, T. barclayana, T. setariae, T. opaca, T. sumatii和T. savilei等区别明显,T. indica冬孢子大小平均值明显大于这些腥黑粉菌,前者大于30祄,后者则小于30祄, T. indica孢壁纹饰与这些腥黑粉菌也有一定区别。应用光学显微镜和扫描电镜,能将T. indica与除T. walkeri之外的其它近似种区别开,但在区别T. indica与T. walkeri方面则有一定的局限性。  相似文献

Using two-color fluorescence flow cytometry, we were able to detect the presence of small numbers of T4+T8+ cells (about 3%) in freshly isolated peripheral T cell populations derived from normal healthy donors. Coexpression of T4 and T8 was predominantly found on large blastlike cells and appeared to be related to activation. Stimulation of peripheral T cells with concanavalin A (Con A) for 5 days resulted in the generation of up to 60% of T4+T8+ cells. Coexpression was accompanied by a twofold increase in the number of T8 antigenic sites per cell. The T4+T8+ cells in lectin-stimulated cultures expressed high levels of the activation antigens T9, T10, and the IL-2 receptor but lacked T6, an antigen found on a majority of stage II thymocytes. Coexpression of T4 and T8 appeared to be a transitory process, because prolonged culture of T cells in the absence of lectin resulted in the loss of the T4+T8+ phenotype. Our data suggest that T cell activation in peripheral blood results in the generation of a T4+T8+ cell population which is distinct from previously described thymic and peripheral blood cells. Because T4 and T8 molecules may interact directly with MHC antigens, coexpression of these molecules may have an important role in immune function.  相似文献

Chromosome banding with nucleotide base-specific fluorochromes chromomycin A3 (CMA) and Hoechst 33258 (H33258) was used to study the karyotypes and to construct cytological maps for diploid Trillium camschatcense (2n = 10), tetraploid T. tschonoskii (2n = 20), hexaploid T. rhombifolium (2n = 30), and a triploid T. camschatcense x T. tschonoskii hybrid (T. x hagae, 2n = 15). With H33258, species- and genome-specific patterns with numerous AT-rich heterochromatin bands were obtained for each of the four forms; CMA revealed a few small, mostly telomeric GC-rich bands. In T. tschonoskii, the two subgenomes were similar to each other and differed from the T. camschatcense genome; on this evidence, the species was considered to be a segmental allotetraploid. In T. x hagae, one T. camschatcense and both T. tschonoskii subgenomes were identified. The subgenomes of T. rhombifolium only partly corresponded to the T. camschatcense and T. tschonoskii genomes, in contrast to the morphologically identical Japanese species T. hagae. This was assumed to indicate that allohexaploids T. rhombifolium and T. hagae originated independently at different times; i.e., their origin is polyphyletic. Based on the chromosome maps, a new nomenclature was proposed for the Trillium genomes examined: K1K1 for T. camschatcense, T1T1T2T2 for T. tschonoskii, T1T1T2T2 for T. x hagae, and K1RK1RT1RT1RT2RT2R for T. rhombifolium.  相似文献

Nel contempo si ristudia la sistematica del genere e delle specie ivi incluse, redigendo, a titolo provvisorio, una chiave analitica delle specie fondamentali : Trichosporon giganteum, T. proteolyticum, T. infestans, T. Luchettii, T. Balzeri, T. granulosum, T. Beigeli, T. cutaneum, T. Uffreduzzii, tutte del sottogenere Proteomyces.
Summary Three strains of Trichosporon isolated from man, one from wood pulp, and two from cheese, are described. The strains are found to be T. infestans (2 strains), T. cutaneum, T. Beigeli and the new species T. Luchettii (2 strains). The sistematic position of the genus and the subdivision of species are discussed. A provisional analytical key of the master species is appended, including: T. giganteum, T. proteolyticum, T. infestans, T. Luchettii, T. Balzeri, T. granulosum, T. Beigeli, T. cutaneum, T. Uffreduzzii, all of the subgenus Proteomyces,

Sumario Tres cultivos de Trichosporon aislados del hombre, uno de pasta de madera para papel, y dos del queso están descritos. Los cultivos fueron referidos al T. infestans (dos), T. cutaneum, T. Beigeli, y a la nueva especie T. Luchettii (dos). La posición sistematica del género y la subdivisión en especies fueron examinado criticamente, y una clave analítica de las especies más conocidas o clasicas ha sido instituida. Las especies consideradas (T. giganteum, T. proteolyticum, T. infestans, T. Luchettii, T. Balzeri, T. granulosum, T. Beigeli, T. cutaneum, T. Uffreduzzii) pertenecen todas al subgénero Proteomyces.

Milk was collected for the first 21 days of lactation twice daily from dairy cows and once daily from goats, sheep, and guinea pigs. Thyroxine (T4) and triiodothyronine (T3) were extracted from 100 microliter of milk using acidified ethanol. T4 and T3 were reconstituted in 100 microliter buffer and measured by radioimmunoassay. Concentrations (ng/ml) of T4 and T3 for milk of cows, goats, sheep, and guinea pigs, respectively, were: 0.97 and 0.94, 1.24 and 0.52, 0.99 and 0.79, and 1.41 and 0.53. T4 concentration for guinea pig milk was significantly higher than for cow and sheep milk, but not for goat milk (P less than 0.05). T3 was found in higher concentration in milk of cows and sheep than in milk of goats and guinea pigs (P less than 0.05). Species differences in conversion of T4 to T3 in mammary gland cells are suggested. Summations of T4 and T3 concentrations in milk indicated no differences among the four species. Regression analyses of changes in milk production, T4 and T3 concentrations, total T4 and T3 in milk per day, and ratios of T4 to T3 revealed variations in patterns. Concentrations of T4 or T3 tended to decrease as lactation progressed over 21 days. Total T3 tended to increase, and the ratio of T4 to T3 tended to decrease. Amounts of T4 and T3 available to offspring from milk were calculated to be minor sources (4 to 7%) of total requirements for maintenance of metabolic functions.  相似文献
