Kinetin at physiological concentrations causes significant reduction of GA3-promoted growth in excised Avena stem segments. Kinetin is therefore considered to be a gibberellin-antagonist in this system. A Lineweaver-Burke plot reveals that kinetin acts non-competitively with GA3. The kinetin inhibition of GA3-promoted growth can be seen within 6 hours. It was found that soluble protein is markedly increased by kinetin in the tissue during the first 3 hours, thus preceding the inhibition of GA3-promoted growth by several hours. At the cellular level, kinetin negated the blocking effect of GA3 on cell division in the intercalary meristem portions of these segments. In fact, kinetin promotes both lateral and longitudinal cell divisions in intercalary meristem cells.  相似文献

The purpose of this study was to analyze the nature of the interaction between gibberellic acid (GA3) and abscisic acid (ABA) in the regulation of growth in excised Avena (oat) stem segments. Growth, compared to sucrose controls, was inhibited by ABA in the range of 10?4 to 10?6M. GA3-promoted growth was also inhibited by ABA in the same concentration range. A Lineweaver-Burk analysis of the interaction between GA3 and ABA indicated that ABA acts in a non-competitive fashion with GA3. This same result was obtained previously with GA3-indoleacetic acid (IAA) and GA3-kinetin interactions with Avena stem sections. Our results indicate that ABA can inhibit GA3-promoted growth within physiological concentrations, and that it is probably acting at a different physiological site from that for GA3.  相似文献

The accumulation of silicon was studied in Avena intercalary-meristemstem-segments treated in GA3, sucrose, GA3+sucrose, and water. Electron-probe analysis was used for the detection of silicon.GA3 and sucrose, together and separately, appear to inhibitthe accumulation of silicon in the silica cells which are thespecific sites for silicon deposition. The results suggest thatsucrose and gibberellin may play a role in regulating the silicificationprocess in developing internodes.  相似文献

Ethylene at 5–80 µl l–1 inhibited elongationand induced swelling in internodes of light-grown normal anddwarf pea plants; GA3 did not prevent swelling in response toethylene. GA3 neither inhibited nor enhanced the activity of isoperoxidasesin the internodes, regardless of its effect on their elongation.Ethylene at 80 µl l–1 enhanced peroxidase in GA3-untreatedand treated normal and dwarf plants. At 5 µl l–1,ethylene had only a weak effect on peroxidase activity or none.The enzyme enhancement by ethylene was not related to its effecton cell expansion and seems do be due, at least in part, tochemical injury. Electron microscopy revealed peroxidase activity in the roughER and cell walls, including intercellular spaces. Stainingof walls in ethylene-treated tissues was more pronounced thanin untreated ones. Golgi vesicles did not seem to be involvedin the assembly of the enzyme carbohydrate moiety in ethylene-treatedcells. The peroxidase fraction extracted with 20 mM phosphate buffer,pH 6, and that extracted from wall debris with 1 M NaCl accountedfor 98% of total enzyme activity. Both fractions contained thesame six cathodic isoforms which comprised 85–90% of theiractivity. Electrophoresis did not reveal differences in thequalitative isoenzyme patterns in relation to variety, age,GA3, or ethylene. The only observed quantitative differenceswere age-dependent. Procedural artefacts during separation of protoplast and wallionically bound peroxidase fractions are discussed.  相似文献

Pulvini of excised segments from oats (Avena sativa L. cv Victory) were treated unilaterally with indoleacetic acid (IAA) or gibberellic acid (GA3) with or without gravistimulation to assess the effect of gravistimulation on hormone action. Optimum pulvinus elongation growth (millimeters) and segment curvature (degrees) over 24 hours were produced by 100 micromolar IAA in vertical segments. The curvature response to IAA at levels greater than 100 micromolar, applied to the lower sides of gravistimulated (90°) pulvini, was significantly less than the response to identical levels in vertical segments. Furthermore, the bending response of pulvini to 100 micromolar IAA did not vary significantly over a range of presentation angles between 0 and 90°. In contrast, the response to IAA at levels less than 10 micromolar, with gravistimulation, was approximately the sum of the responses to gravistimulation alone and to IAA without gravistimulation. This was observed over a range of presentation angles. Also, GA3 (0.3-30 micromolar) applied to the lower sides of horizontal segments significantly enhanced pulvinus growth and segment curvature, although exogenous GA3 over a range of concentrations had no effect on pulvinus elongation growth or segment curvature in vertical segments. The response to GA3 (10 micromolar) plus IAA (1.0 or 100 micromolar) was additive for either vertical or horizontal segments. These results indicate that gravistimulation produces changes in pulvinus responsiveness to both IAA and GA3 and that the changes are unique for each growth regulator. It is suggested that the changes in responsiveness may result from processes at the cellular level other than changes in hormonal sensitivity.  相似文献

Different amounts of abscisic acid, 2.7 × 10?9? 5 × 10?8 moles, were chromatographed in isopropanol: ammonia: water (100:14:6), firstly alone and secondly together with 5 × 10?8 moles kinetin. The same amount of kinetin was also chromatographed alone. The chromatograms were tested biologically with the Avena straight-growth test. Whereas a large part of the chromatograms of kinetin gives growth stimulation, the Rf region 0.4–0.6 of abscisic acid chromatograms is strongly growth-inhibiting. The inhibition within this Rf region does not become less if abscisic acid and kinetin are chromatographed together.  相似文献

Kinetin has a stimulating effect in the Avena straight-growth test. The action of different concentrations of kinetin, 2.5 × 10?7, 2.5 × 10?6 and 2.5 × 10?5M, in combination with different concentrations of IAA was studied in this test. It was shown that the effect of low IAA concentrations, 0.25 × 10?7 and 1 × 10?7M, was strongly enhanced by the addition of all the kinetin concentrations investigated. The effect of the highest IAA concentrations, 25 × 10?7 and 100 × 10?7M, on the other hand, was inhibited relatively strongly by the highest employed concentration of kinetin. The results are explained as due to a kinetin-produced increase of auxin in the coleoptile segment, which in combination with low IAA concentrations can lead to a growth stimulation and with high IAA concentrations to a growth inhibition. Since kinetin in purification and chromatography of auxin can partly follow IAA, thereby affecting the quantitative yield, it is emphasized that, prior to the test, auxin extracts containing cytokinins should be freed from the latter by, for example, gel filtration or paper electrophoresis.  相似文献

A micro-growth measuring technique was used to determine the growth response of stem segments of Avena sativa cv. Avon to a variety of gibberellic acid (GA3) concentrations over a range of incubation temperatures. Growth rate varied with GA3 concentration, the temperature at which the rate was measured, and the growth temperature of the plants prior to excision of the segments. The curves relating segment extension rates to temperature were affected by GA3 such that the linear portion of the curve was shifted to higher rates as GA3 concentration was increased. The results seem to be analogous to the GA3-induced shifts of thermally induced phase transitions in glucose leakage from liposomes, observed earlier (Wood, Paleg 1974 Aust J Plant Physiol 1: 31-40).  相似文献

Gibberellic acid (GA) does not delay maturation of pea internodes;on the con-trary, maturation (i.e. cessation of extension) takesplace slightly earlier. Thus the increased length of internodesresulting from GA treatment is due entirely to increased rateof extension. In this experiment, GA treatment of plants accelerated the visibleproduction of the first flower bud by about 4 days: the nodebearing the first flower was not altered. The total number offlower buds produced by the end of the experiment was increasedas a result of GA treatment, but many of those first formedon plants receiving high doses (I-IOµg.) withered beforeopening.  相似文献

The effect of coumarin, IAA, ethylene, kinetin and gibberellic acid on roots of maize and wheat was investigated. Sterile attached and detached roots and isolated elongation zones were used. In some experiments a semi-sterile procedure was followed. The effects of the different regulators separately or in various combinations together with coumarin were studied on the root growth with regard to division, elongation and swelling of the cells. The ethylene production in isolated elongation zones was measured after treatment with coumarin, IAA, PCIB, kinetin, colchicine and dinitrophenol. The results show the following: 1) Each substance produces a specific morphologic pattern. 2) Changes in polarity were demonstrated for auxin-induced swelling in cell divisions and cell expansion and for coumarin-induced swelling in cell divisions. Other cell expansion in swollen parts was due to cylindric cells increasing in width while retaining their cylindric form. 3) Coumarin-induced inhibition could not be counteracted by IAA, PCIB, carbon dioxide, kinetin, gibberellic acid or Cycocel. 4) The ethylene production in isolated elongation zones increases noticeably after kinetin treatment, less strongly after auxin treatment and least after coumarin treatment. The production of ethylene does not seem to be correlated with the morphogenetic effect of the different substances. 5) The isolated elongation zones reacted to a) IAA and kinetin with an increase in length in some cases and b) gibberellic acid with a reduction of their width. 6) The inhibitory effect of coumarin on the growth in length of the elongation zones was diminished by kinetin. The swelling produced by coumarin in these zones was reduced by gibberellic acid. The effects just mentioned of kinetin and gibberellic acid were considered as indirect ones. - From the present findings it was concluded that concomitant effects of auxin, ethylene, cytokinins and gibberellins are not obligatory for coumarin to exert its morphogenetic effects on root growth. In discussing the results some pitfalls in studies of growth reactions after application of hormones to roots containing meristem were emphasized.  相似文献

Movement of [14C]kinetin and [14C]gibberellic acid was examined in cotton (Gossypium hirsutum L.) cotyledonary petiole sections independent of label uptake or exit from the tissue. Sections 20 millimeters in length were taken from well watered, stressed, and poststressed plants. Transport capacity was determined using a pulse-chase technique. Movement of both kinetin and gibberellic acid was found to be nonpolar with a velocity of 1 millimeter per hour or less, suggesting passive diffusion. Neither water stress nor anaerobic conditions during transport of labeled material affected the transport capacity of the petioles.  相似文献

The properties of two strains of carrot (Daucus carota) callus are presented. One has a very low acid invertase activity which is accompanied by differences in morphology and metabolic rate, but not in growth rate. We conclude that one of the main functions of plant acid invertases is in controlling the levels of sugars which, by interaction with hormones, affect differentiation, both morphological and biochemical. The effect of tris on sucrose metabolizing enzymes, and the cause of the “sucrose effect” are considered.  相似文献
