Summary More than 80 different gliadin electrophoretic patterns (spectra) have been found in 109 accessions of the diploid wheat Triticum monococcum. Each pattern consists of 15–20 gliadin bands. Some patterns are clearly related and might arise from one another through single mutations in the gliadin-coding loci. From the analysis of 15 grains of each, only 61 accessions were found to be uniform; others consisted of two or more grain variants differing in their gliadin spectrum. An analysis of F2 grains from three crosses between different accessions showed that groups (blocks) of components are jointly and codominantly inherited. Two independent major Gli loci were established. The close resemblance of the composition of some blocks of T. monococcum to some of those in polyploid wheats indicates that one locus in each T. monococcum genotype is located on chromosome 1A (Gli-A1) and the other on 6A (Gli-A2). However, the blocks of T. monococcum include more bands than corresponding (equivalent) blocks of polyploid wheats. Two out of 275 F2 grains of the cross k-14244 x k-20409 were found to have gliadin spectra which can be explained as a result of intralocus recombination. Also, a second gliadin-coding locus on chromosome 1A was found in the cross k-46140 x k-46753. This locus recombines with the main Gli-A1 locus with a frequency of about 22% and was clearly analogous to the additional Gli locus found earlier on chromosome 1A of certain polyploid wheats.  相似文献

 Chromosome pairing at metaphase-I was analyzed in F1 hybrids among T. turgidum (AABB), T. aestivum (AABBDD), and T. timopheevii (AtAtGG) to study the chromosome structure of T. timopheevii relative to durum (T. turgidum) and bread (T. aestivum) wheats. Individual chromosomes and their arms were identified by means of C-banding. Homologous pairing between the A-genome chromosomes was similar in the three hybrid types AAtBG, AAtBGD, and AABBD. However, associations of B-G were less frequent than B-B. Homoeologous associations were also observed, especially in the AAtBGD hybrids. T. timopheevii chromosomes 1At, 2At, 5At, 7At, 2G, 3G, 5G, and 6G do not differ structurally from their counterpart in the A and B genomes. Thus, these three polyploid species inherited translocation 5AL/4AL from the diploid A-genome donor. Chromosome rearrangements that occurred at the tetraploid level were different in T. turgidum and T. timopheevii. Translocation 4AL/7BS and a pericentric inversion of chromosome 4A originated only in the T. turgidum lineage. The two lines of T. timophevii studied carry four different translocations, 6AtS/1GS, 1GS/4GS, 4GS/4AtL, and 4AtL/3AtL, which most likely arose in that sequence. Quinacrine fluorescent banding patterns of chromosomes 9 and 13 are very similar in mitotic preparations of Mus musculus. Meiotic studies were carried out in male and female mice heterozygous for two translocations involving these chromosomes to determine whether the translocations have a common chromosome. The results indicate that chromosome 9 is involved in the T163H translocation but not in either the T70H or T264Ca translocations. The T70H and T264Ca translocations, but not the T163H, have chromosome 13 in common. These results support the interpretations based on mitotic studies.  相似文献

A hydroponic experiment was conducted to investigate the effects of indole-3-acetic acid (IAA) on arsenic (As) uptake and antioxidative enzymes in fronds of Pteris cretica var. nervosa (As hyperaccumulator) and Pteris ensiformis (non-hyperaccumulator). Plants were exposed to 2 mg L?1 As(III), As(V) or dimethylarsinic acid (DMA) and IAA concentrations for 14 d. The biomass and total As in the plants significantly increased at 30 mg L?1 IAA. Superoxide dismutase (SOD) activities significantly increased with IAA addition. Catalase (CAT) activities showed a significant increase in P. ensiformis exposed to three As species at 30 or 50 mg L?1 IAA but varied in P. cretica var. nervosa. Peroxidase (POD) activities were unchanged in P. ensiformis except for a significant decrease at 50 mg L?1 IAA under As(III) treatment. However, a significant increase was observed in P. cretica var. nervosa at 10 mg L?1 IAA under As(III) or DMA treatment and at 50 mg L?1 IAA under As(V) treatment. Under DMA stress, malondialdehyde contents in fronds of P. cretica var. nervosa showed a significant decrease at 10 mg L?1 IAA but remained unchanged in P. ensiformis. Therefore, IAA enhanced As uptake and frond POD activity in P. cretica var. nervosa under As stress.  相似文献

Analyses of RFLPs, isozymes, morphological markers and chromosome pairing were used to isolate 12 Triticum aestivum cv Chinese Spring (genomes A, B, and D)-T. peregrinum (genomes Sv and Uv) disomic chromosome addition lines. The evidence obtained indicates that each of the 12 lines contains an intact pair of T. peregrinum chromosomes. One monosomic addition line, believed to contain an intact 6Sv chromosome, was also isolated. A CS-7Uv chromosome addition line was not obtained. Syntenic relationships in common with the standard Triticeae arrangement were found for five of the seven Sv genome chromosomes. The exceptions were 4Sv and 7Sv. A reciprocal translocation exists between 4S1 and 7S1 in T. longissimum and evidence was obtained that the same translocation exists in T. peregrinum. In contrast, evidence for syntenic relationships in common with the standard Triticeae arrangements were found for only one Uv chromosome of T. peregrinum.; namely, chromosome 2Uv. All other Uv genome chromosomes are involved in at least one translocation, and the same translocations were found in the U genome of T. umbellulatum. Evidence was also obtained indicating that the centromeric regions of 4U and 4Uv are homoeologous to the centromeric regions of Triticeae homoeologous group-6 chromosomes, that the centromeric regions of 6U and 6Uv are homoeologous to the centromeric regions of group-4 chromosomes, and that 4U and 4Uv are more closely related overall to Triticeae homoeologous group-6 chromosomes than they are to group-4 chromosomes.  相似文献

Trifolium ambiguum M. Bieb and T. repens L. are taxonomically related but very difficult to cross. The rare hybrids so far reported between these two species were obtained only by embryo culture. This difficulty has been overcome in the present research by the creation of a “fertile bridge” between T. ambiguum and T. repens. Characters of interest can now be transferred from T. ambiguum to T. repens by using this “fertile bridge” without the use of sophisticated techniques. An array of backcross progenies was generated from crosses between a T. ambiguum×T. repens F1 hybrid (8x H-435) and its parental species. The 8x hybrid was cross-fertile only with T. repens and resulted in 145 seeds from 1578 reciprocal crosses. Eleven of nineteen initially grown BC1F1 plants were all hexaploid with an average pollen stainability of 41.6%. A high frequency of multivalents at metaphase-I indicated that both autosyndetic and allosyndetic pairing occurred. Backcrosses of 6x BC1F1 plants to T. repens resulted in 5x BC2F1 plants with an average pollen stainability of 59.3%. On the other hand, 6x BC1F1×6x T. ambiguum crosses did not produce any seed and only two pentaploid plants were obtained from 6x BC1F1×4x T. ambiguum crosses. The difficulty encountered in generating 6x backcross progeny with 6x T. ambiguum was overcome by intercrossing the 6x BC1F1 plants and producing 6x BC1F2 plants with an average pollen stainability of 65.8%. One of these 6x BC1F2 plants was cross-compatible as a female with 6x T. ambiguum and resulted in CBC2 plants that were all cross-compatible with 6x T. ambiguum. The 6x BC1F2 plants are likely to be superior to 6x BC1F1 progeny, as they have exhibited better expression of the combined rhizomatous and stoloniferous growth habit, improved fertility, more frequent nodal rooting and heavier nodulation. Consequently, the 6x BC1F2 plants can either be used directly in the selection programme or as a “fertile bridge” between the two parental species. The present work has resulted in the development of a series of fertile hybrids by the manipulation of chromosome numbers, combining the agronomic characteristics of the parent species in varying genome balances and at a range of ploidy levels. In India, 55 species of Berberis have been reported and 22 species are found in Uttarakhand state. Berberis rawatii (Berberidaceae), a new species from Chamoli and Pithoragarh districts of Uttarakhand state of India (western Himalaya) is here described and illustrated. In terms of leaf and inflorescence, Berberis rawatii shows affinities with B. cretica L. Both these species have entire leaves, fascicled inflorescences, conspicuous style and black berries, but B. rawatii differs in the presence of a petiole, the dorsal surface of leaf pruinose, presence of elongated‐ovate glands, presence of prophylls, three ovules and pruinose fruits.  相似文献

The aim of this study was to investigate influence of transgenic and commercial corn hybrids on the behaviour and feeding activity of the pink corn borer (Sesamia cretica Led.). Food consumption was different according to feeding period and hybrids. The feeding ratio of S. cretica on maize hybrids was significantly different between transgenic and commercial hybrids. It appears from the results of this experiment that the antifeeeding activity of transgenic hybrids had greater effect than commercial hybrids one. Thus, it is apparent from these results that transgenic maize was unsuitable because larvae were dead, but the other commercial hybrids were preferable. It can be concluded from the data that the feeding on different maize hybrids had different effects on certain biological aspects of pink corn borer. The impact of untreated Bt. Corn; 0.5% water emulsion of Neem-Azal-T/S and 0.005% water emulsion of Chlorpyrifos insecticides on the behaviour and feeding activity of the pink corn borer S. cretica has been studied. The data showed that larvae stopped feeding from the time when it was fed on untreated Bt. corn or/and non Bt. Corn treated with NeemAzal-T/S 0.5%. The results indicated that Bt. corn, NeemAzal-T/S 0.5% and Chlorphan 48% (0.005%) possessed the toxic effect on pink corn borer S. cretica. According to results, it could be stated that the tested compounds can play an important role in controlling the pink corn borer.  相似文献

The aim of this work was to investigate the enzyme inhibition, antioxidant activity, and phenolic compounds of Lecokia cretica (Lam .) DC. Acetylcholinesterase (AChE), butyrylcholinesterase (BChE), and α‐glycosidase enzymes were strongly inhibited by the L. cretica extracts. IC50 values for the three enzymes were found as 3.21 mg/mL, 2.1 mg/mL, and 2.07 mg/mL, respectively. Antioxidant activities were examined in both aqueous and ethanol (EtOH) extracts using CUPRAC, FRAP, and DPPH method. Also, the phenolic compounds of the endemic plant were identified and quantified by using HPLC/MS/MS. According to the results, the extracts have remarkable antioxidant activities. The most abundant phenolic acids of L. cretica in EtOH extract were determined as quinic acid (12.76 mg/kg of crude extract), chlorogenic acid (3.39 mg/kg), and malic acid (2.38 mg/kg).  相似文献

Plant genome size evolution is a very dynamic process: the ancestral genome of angiosperms was initially most likely small, which led to a tendency towards genome increase during evolution. However, findings in several angiosperm lineages demonstrate mechanisms that also led to genome size contraction. Recent molecular investigations on the Asteraceae genus Crepis suggest that several genomic reduction events have occurred during the evolution of the genus. This study focuses on the Mediterranean Crepis sect. Neglectoides, which includes three species with some of the smallest genomes within the whole genus. Crepis neglecta has the largest genome in sect. Neglectoides, approximately twice the size of the two species Crepis cretica and Crepis hellenica. Whereas C. cretica and C. hellencia are more closely related to each other than to C. neglecta the karyotypes of the latter species and C. cretica are similar, while that of C. hellenica differs considerably. Here, the karyotypic organisation of the three species is investigated with fluorescence in‐situ hybridisation and studied in a molecular phylogenetic framework based on the nuclear markers Actin, CHR12, CPN60B, GPCR1 and XTH23. Our findings further corroborate the occurrence of genome size contraction in Crepis, and suggest that the difference in genome size between C. neglecta and C. cretica is mostly due to elimination of dispersed repetitive elements, whereas chromosomal reorganisation was involved in the karyotype formation of C. hellenica.  相似文献

We have measured the kinetics of proflavin binding to T-even bacteriophages—the 700 S and 1000 S forms of T2L, T4D, and T4D os41—by difference spectroscopy at 430 nm. Measurements were carried out from 22° to 37°C. Binding is very slow to encapsulated DNA compared to free DNA, requiring hours to reach equilibrium. The kinetic data are compatible with the two-step mechanism where P is proflavin, N is nucleotide, and PN1 and PN2 are complexes. Computer integration of the rate equations allows evaluation of the rate constants; previous equilibrium measurements gave thermodynamic parameters. For all phage studied, the bimolecular step is endothermic with high positive entropy; the second, unimolecular step is highly exothermic with small negative entropy change. Both forms of T2L bind proflavin with essentially the same rate, as do T4D and the osmotic shock resistant mutant T4D os41. This suggests that the encapsulated DNA is equally accessible to proflavin in both forms of each phage. However, T4D binds dye appreciably faster than T2L, indicating that capsid permeability or DNA environment (glucosylation or packing) is different in the two species.  相似文献

Summary Translocation of 14C-labelled carbohydrates between the parent stolon and branches, and among branches, of Trifolium repens plants was investigated in two glasshouse experiments to determine patterns of physiological organisation in this clonal species. Differential defoliation treatments were applied to the parent stolon and/or branches to test the sensitivity of translocation to the short-term carbon needs of defoliated sinks. Strong reciprocal exchange of carbohydrate between the parent stolon and branches was observed, with 18 41% of the 14C exported from leaves on the parent stolon moving to branches, while branches simulta-neously exported 25% (for old source branches) to 54% (for young source branches) of the 14C they assimilated to the parent plant, including translocation to other branches. Branch-to-branch translocation occurred both acropetally and basipetally. Parent-to-branch, branch-to-parent and branch-to-branch carbon fluxes all increased in response to defoliation of the sink, at the expense of carbon supply to stolon tissue or roots of the source module. Reduced export to stolon tissue of the parent axis played a major role in facilitating C reallocation from leaves on the parent stolon to defoliated branches. The observed patterns of C allocation and translocation could be adequately explained by accepted source-sink theory, and are consistent with a high degree of intra-plant physiological integration in resource supply and utilisation. This information provides mechanistic explanations for aspects of the growth dynamics and ecological interactions of T. repens in the patchy environment of a grazed pasture.  相似文献

Genetic diversity in the introduced diploids Tragopogon dubius, T. porrifolius, and T. pratensis and their neoallotetraploid derivatives T. mirus and T. miscellus was estimated to assess the numbers of recurrent, independent origins of the two tetraploid species in the Palouse region of eastern Washington and adjacent Idaho. These tetraploid species arose in this region, probably within the past 50–60 yr, and provide one of the best models for the study of polyploidy in plants. The parental species of both T. mirus and T. miscellus have been well documented, and each tetraploid species has apparently formed multiple times. However, a recent survey of the distributions of these allotetraploids revealed that both tetraploid species have expanded their ranges considerably during the past 50 yr, and several new populations of each species were discovered. Therefore, to evaluate the possibility that these recently discovered populations are of recent independent origin, a broad analysis of genetic diversity in T. mirus, T. miscellus, and their diploid progenitors was conducted. Analyses of allozymic and DNA restriction site variation in all known populations of T. mirus and T. miscellus in the Palouse and several populations of each parental diploid species revealed several distinct genotypes in each tetraploid species. Four isozymic multilocus genotypes were observed in T. mirus, and seven were detected in T. miscellus. Tragopogon mirus possesses a single chloroplast genome, that of T. porrifolius, and two distinct repeat types of the 18S-26S ribosomal RNA genes. Populations of T. miscellus from Pullman, Washington, have the chloroplast genome of T. dubius; all other populations of T. miscellus have the chloroplast DNA of T. pratensis. All populations of T. miscellus combine the ribosomal RNA repeat types of T. dubius and T. pratensis, as demonstrated previously. When all current and previously published data are considered, both T. mirus and T. miscellus appear to have formed numerous times even within the small geographic confines of the Palouse, with estimates of five to nine and two to 21 independent origins, respectively. Such recurrent polyploidization appears to characterize most polyploid plant species investigated to date (although this number is small) and may contribute to the genetic diversity and ultimate success of polyploid species.  相似文献

Restriction fragment length polymorphism (RFLP) markers were used to map male fertility restoring gene that was transferred from chromosome 6U of Aegilops umbellulata Zhuk. to wheat. Segments of chromosome 6U bearing the gene that restore fertility to T. timopheevi Zhuk. male sterile cytoplasm were identified in all four translocation lines by two probes, BCD21 and BCD342. Lines 040-5,061-1 and 061-4 are T6BL.6BS6U translocations, while line 2114 is a T6AL.6AS-6U translocation. Line 2114 has a much larger 6U chromosomal segment and lower frequency of transmission of male gametes with the alien segment than the other three lines. The restoring gene carried by the 6U segment in 2114 showed high expressivity and complete penetrance. This restoring gene is designated Rf6.  Resistance to clover cyst nematode (Heterodera trifolii) has been successfully transferred from Trifolium nigrescens to T. repens by interspecific hybridisation. A sterile triploid hybrid (H-6909-5, 2n=3x=24) was initially produced with the aid of embryo culture. The hybrid was chromosome-doubled from axillary meristems by an in vitro colchicine method. Three chromosome-doubled plants were obtained, and these showed a marked increase in pollen stainability from 10% in 3x H-6909-5 to an average of 89% (range 88–91%) in 6x H-6909-5. T. nigrescens was a source of clover cyst nematode resistance. A mean of 23 (range 0–150) cysts per plant was recorded for T. nigrescens in comparison to a mean of 150 (range 50–240) cysts per plant for T. repens. The 3x and 6x interspecific hybrids were shown to be as resistant as the most resistant. T. nigrescens genotype and were significantly lower in cyst number per gram of root dry weight than the susceptible T. nigrescens and T. repens genotypes.  Taxa of the Viola alba complex were investigated using allozymes and morphometry. A taxonomic revision is presented. A wide delimitation of V. alba with only three morphological and geographical subspecies is suggested: (1) ssp. dehnhardtii distributed in the Mediterranean eastwards to Turkey; (2) ssp. alba flanking ssp. dehnhardtii in the north and east; and (3) ssp. cretica endemic to Crete. Ssp. cretica, up to now treated as a separate species, is particularly close to ssp. dehnhardtii. Viola cadevallii (NW Mediterranean) is included in the synonymy of ssp. dehnhardtii. Ssp. scotophylla (S Europe), ssp. thessala (Balkan), V. armena (Turkey), and V. besseri (Caucasus) are reduced to synonyms of V. alba ssp. alba. Viola pentelica (Greece) might represent transitional forms between ssp. alba and ssp. dehnhardtii. Glacial refugia for ssp. alba are suggested from the eastern Mediterranean via Turkey to the Caucasus, for ssp. dehnhardtii in the Mediterranean area in general, and for ssp. cretica in Crete. Meiosis of T70H/+, Ts(113)70H translocation trisomic male mice has been studied using C-banded preparations and 3H-thymidine autoradiography of the first meiotic division. Epididymal sperm counts and sperm morphology scores were also collected. As reported earlier, at the first meiotic division the translocation involved chromosomes 1, 13, 131 and 113 (twice) formed mainly three multivalent configurations: Chain III+II, CIV+I and CV. — The autoradiographic study indicated an abnormal, precocious spiralization pattern for the chromatin in CIV+I primary spermatocytes. These cells, occurring together with the CIII+II and CV configurations in recognizable groups, usually descending from single spermatogonial stem cells, are delayed through meiotic prophase. Both delay and disturbed chromosome spiralization in these cells are attributed to the uniform association of the univalent (I) chromosome 113 with the sex chromosomes during pachytene. Primary spermatocytes of the CIV+I configuration and those carrying a CV take longer to develop from metaphase I into secondary spermatocytes than does the CIII+II type. — In T70H tertiary trisomics with a similar chromosome imbalance, the majority of primary spermatocytes degenerates during the diakinesis-metaphase I stages of meiosis. Fertility is low in contrast to the translocation trisomics. Comparison between the two types leads to the conclusion, that trisomy per se reduces the size of the testes and that the univalent containing CIV+I primary spermatocytes, contrary to the almost uniformly 113 univalent carrying spermatocytes of the T70H tertiary trisomics are rescued by the neighbouring CIII+II and CV carrying cells to form normal secondary spermatocytes and morphologically normal sperm.  相似文献

Accurate assessment of pest potential distributions is needed to identify their establishment risks that play a key role in pest management in agricultural ecosystems. We used a correlative niche modelling method (Maxent) to predict and map the spatial distributions of two important rice stem borers, Chilo suppressalis and Sesamia cretica, in paddy fields of Iran. In total, 195 presence occurrence records (101 records for C. suppressalis and 94 records for Scretica) were compiled. A set of environmental and topographic variables, with the highest effects on the species distributions and the lowest correlations among themselves, were used. The results showed that mainly the northern parts of Iran were the most suitable areas for C. suppressalis, and north, north‐east and south‐west of Iran as the most suitable areas for Scretica. Both models performed well, with an area under the receiver operating characteristic curve (AUC) of 0.983 and 0.786 for C. suppressalis and Scretica, respectively. The Maxent models showed higher accuracy for predicting the distribution of the specialist pest with the small range sizes compared to the generalist species. Assessing the importance of environmental variables, which were derived from the jackknife test, showed the precipitation as the variable with the highest contribution (66%) in explaining the spatial distribution of C. suppressalis compared to the other variables. The distribution of Scretica was influenced by a set of variables derived from both the precipitation and temperature. The Maxent predictions were useful to map the geographical distributions of the risk for both rice stem borers that is needed to develop effective management strategies.  相似文献
