Biogenic isoprene substantially affects atmospheric chemistry, but it is not known how or why many plants, especially trees, make isoprene. We fed 13CO2 to leaves of Quercus rubra and monitored the incorporation of 13C into isoprene by mass spectrometry. After feeding 13CO2 for 9 min we found all possible labelling patterns from completely unlabelled to fully labelled isoprene. By 18 min, 84% of the carbon atoms in isoprene were 13C. Labelling of the last 20% of the carbon atoms was much slower than labelling of the first 80%. The rate of labelling of isoprene was similar to that reported for phosphoglyceric acid indicating that there is a close linkage between the carbon source for isoprene synthesis and the photosynthetic carbon reduction pathway. 相似文献

A method based on simultaneous short-term exposure to 14 CO2 and 15 N2 is described for studying nitrogen fixation and distribution in legumes relative to carbon assimilation and use. Equipment designed to accomodate experiments under natural conditions with very little disturbance of the N2 fixing association is used. It permits continuous measurement and regulation of variables such as air temperature, humidity and CO2 concentration as well as soil aeration. Measurements of distribution and use of assimilates, respiration of nodulated roots, quantitative N2 fixation and the distribution and fate of fixed N as a function of time lead to a precise estimation of C and N budgets for each labelling period. When experiments are done at several phenological stages they give a new insight into the complex C and N interrelations in legume symbiosis.

Reduced soil N availability under elevated CO2 may limit the plant's capacity to increase photosynthesis and thus the potential for increased soil C input. Plant productivity and soil C input should be less constrained by available soil N in an N2‐fixing system. We studied the effects of Trifolium repens (an N2‐fixing legume) and Lolium perenne on soil N and C sequestration in response to 9 years of elevated CO2 under FACE conditions. 15N‐labeled fertilizer was applied at a rate of 140 and 560 kg N ha?1 yr?1 and the CO2 concentration was increased to 60 Pa pCO2 using 13C‐depleted CO2. The total soil C content was unaffected by elevated CO2, species and rate of 15N fertilization. However, under elevated CO2, the total amount of newly sequestered soil C was significantly higher under T. repens than under L. perenne. The fraction of fertilizer‐N (fN) of the total soil N pool was significantly lower under T. repens than under L. perenne. The rate of N fertilization, but not elevated CO2, had a significant effect on fN values of the total soil N pool. The fractions of newly sequestered C (fC) differed strongly among intra‐aggregate soil organic matter fractions, but were unaffected by plant species and the rate of N fertilization. Under elevated CO2, the ratio of fertilizer‐N per unit of new C decreased under T. repens compared with L. perenne. The L. perenne system sequestered more 15N fertilizer than T. repens: 179 vs. 101 kg N ha?1 for the low rate of N fertilization and 393 vs. 319 kg N ha?1 for the high N‐fertilization rate. As the loss of fertilizer‐15N contributed to the 15N‐isotope dilution under T. repens, the input of fixed N into the soil could not be estimated. Although N2 fixation was an important source of N in the T. repens system, there was no significant increase in total soil C compared with a non‐N2‐fixing L. perenne system. This suggests that N2 fixation and the availability of N are not the main factors controlling soil C sequestration in a T. repens system. 相似文献

While this suggests a contribution of reserves to grain filling, the actual efficiency of mobilized assimilate conversion into grain mass (ME) is unknown. In the present study the contribution of pre-anthesis C (C fixed prior to anthesis) to grain filling in main stem ears of two spring wheat (Triticum aestivum L.) cultivars was determined by 13C/12C steady-state labelling. Mobilization of pre-anthesis C in vegetative plant parts between anthesis and maturity, and the contributions of water-soluble carbohydrates (WSC) and protein to pre-anthesis C mobilization were also assessed. Experiments were performed with two levels of N fertilizer supply in each of 2 years. Pre-anthesis reserves contributed 11–29% to the total mass of C in grains at maturity. Pre-anthesis C accumulation in grains was dependent on both the mass of pre-anthesis C mobilized in above-ground vegetative plant parts (r2 = 0·87) and ME (defined as g pre-anthesis C deposited in grains per g pre-anthesis C mobilized in above-ground vegetative plant parts; r2 = 0·40). ME varied between 0·48 and 0·75. The effects of years, N fertilizer treatments and cultivars on ME were all related to differences in the fractional contribution of WSC to pre-anthesis C mobilization. Multiple regression analysis indicated that C from mobilized pre-anthesis WSC may be used more efficiently in grain filling than C present in proteins at anthesis and mobilized during grain filling. Measurements of rapid changes in concentrations and fluxes of gaseous compounds relating to photosynthetic gas exchange are commonly performed using flow-through cuvettes in connection with infrared gas analysers. The accuracy and repeatability of these measurements relies ultimately upon the design of the system as a whole, rather than upon each of its components, and therefore the calibration and testing of the system should be performed keeping this in mind. We present here a simple and efficient method for the calibration of such a measurement system using a precisely determined CO2 flow. This method gives us the opportunity to take into account any disturbing effects caused by undesired properties of the chamber or tubing materials. With the proposed calibration method, the accuracy of the CO2 flux measurement is improved from 8% up to the level of 2%, determined mainly by the accuracy of the control gas used for calibration of the CO2 analyser. 相似文献

When whole plants were exposed to 14 CO2 , almost the same amount of radioactivity was taken up initially by each leaf regardless of its position on the stem and of the presence of beans at that node. Thus, although developing beans are a powerful sink for assimilated carbon, they do not increase the CO2 uptake by adjoining leaves.
The distribution of labelled assimilates 6 hours after feeding14 CO2 to a single leaf for 1 hour varied with both the position of the treated leaf and the stage of development of the plant. Before any flowers were set most of the radioactivity from all expanded leaves moved downwards to the roots and the stem below the treated leaf (lower stem). Later, during pod-fill, the upper leaves maintained this supply to the roots and lower stem, whilst most of the carbon translocated from the lower and mid-stem leaves went to the beans. However, we found no exclusive relationship between a leaf and the supply to beans developing on the same node.

The influence of N availability on C sequestration under prolonged elevated CO2 in terrestrial ecosystems remains unclear. We studied the relationships between C and N dynamics in a pasture seeded to Lolium perenne after 8 years of elevated atmospheric CO2 concentration (FACE) conditions. Fertilizer‐15N was applied at a rate of 140 and 560 kg N ha2?1 y2?1 and depleted 13C‐CO2 was used to increase the CO2 concentration to 60 Pa pCO2. The 13C–15N dual isotopic tracer enabled us to study the dynamics of newly sequestered C and N in the soil by aggregate size and fractions of particulate organic matter (POM), made up by intra‐aggregate POM (iPOM) and free light fraction (LF). Eight years of elevated CO2 did not increase total C content in any of the aggregate classes or POM fractions at both rates of N application. The fraction of new C in the POM fractions also remained largely unaffected by N fertilization. Changes in the fractions of new C and new N (fertilizer‐N) under elevated CO2 were more pronounced between POM classes than between aggregate size classes. Hence, changes in the dynamics of soil C and N cycling are easier to detect in the POM fractions than in the whole aggregates. Within N treatments, fractions of new C and N in POM classes were highly correlated with more new C and N in large POM fractions and less in the smaller POM fractions. Isotopic data show that the microaggregates were derived from the macro‐aggregates and that the C and N associated with the microaggregates turned over slower than the C and N associated with the macroaggregates. There was also isotopic evidence that N immobilized by soil microorganisms was an important source of N in the iPOM fractions. Under low N availability, 3.04 units of new C per unit of fertilizer N were sequestered in the POM fractions. Under high N availability, the ratio of new C sequestered per unit of fertilizer N was reduced to 1.47. Elevated and ambient CO2 concentrations lead to similar 15N enrichments in the iPOM fractions under both low and high N additions, clearly showing that the SOM‐N dynamics were unaffected by prolonged elevated CO2 concentrations. 相似文献

Results were compared with estimates obtained by indirect approaches to mobilization of pre-anthesis C: ‘classical’ growth analysis and balance sheets of water soluble carbohydrates (WSC) and protein. Experiments were performed with two spring wheat cultivars grown with differential nitrogen fertilizer supply in 1991 and 1992. The fraction of pre-anthesis C mobilized in above-ground vegetative biomass ranged between 24 and 34% of total C present at anthesis. Treatment effects on mobilization of pre-anthesis C in total above-ground vegetative biomass were closely related (r2 = 0·89) to effects on mobilization of WSC-C plus protein-C (estimated as N mobilized × 3·15). On average, 81% of pre-anthesis C mobilization was attributable to the balance of pre-anthesis WSC (48%) and protein (33%) between anthesis and maturity. In roots, WSC and protein mobilization accounted for only 29% of the loss of pre-anthesis C. Notably, mobilization of pre-anthesis C was 1·4–2·6 times larger than the net loss of C from above-ground vegetative biomass between anthesis and maturity. This discrepancy was mainly due to post-anthesis C accumulation in glumes and stem. Post-anthesis C accumulation was related to continued synthesis of structural biomass after anthesis and accounted for a mean 15% of total C contained in above-ground vegetative plant parts at maturity. A close correspondence between net loss of C and mobilization of pre-anthesis C was only apparent in leaf blades and leaf sheaths. Although balance sheets of WSC and protein also underrated the mobilization of pre-anthesis C by ≈ 19%, they gave a much better estimate of pre-anthesis C mobilization than growth analysis. 相似文献

Fruiting structures of a number of legumes including chickpea are known to carry out photosynthetic CO2 assimilation, but the pathway of CO2 fixation and particularly the role of phosphoenolpyruvate carboxylase (EC 4.1.1.31) in these tissues is not clear. Activities of some key enzymes of the Calvin cycle and C4 metabolism, rates of 14CO2 fixation in light and dark, and initial products of photosynthetic 14CO2 fixation were determined in podwall and seedcoat (fruiting structures) and their subtending leaf in chickpea (Cicer arietinum L.). Compared to activities of ribulose-1,5-bisphosphate carboxylase (EC 4.1.1.39) and other Calvin cycle enzyme, viz. NADP+-glyceraldehyde-3-phosphate dehydrogenase (EC 1.2.1.13), NAD+-glyceraldehyde-3-phosphate dehydrogenase (EC 1.2.1.12) and ribulose-5-phosphate kinase (EC 2.7.1.19), the levels of phosphoenolpyruvate carboxylase and other enzymes of C4 metabolism viz. NADP+-malate dehydrogenase (EC 1.1.1.82), NAD+-malate dehydrogenase (EC 1.1.1.37), NADP+ malic enzyme (EC 1.1.1.40), NAD+-malic enzyme (EC 1.1.1.39), glutamate oxaloacetate transaminase (EC 2.6.1.1) and glutamate pyruvate transaminase (EC 2.6.1.2), were generally much higher in podwall and seedcoat than in the leaf. Podwall and seedcoat fixed 14CO2 in light and dark at much higher rates than the leaf. Short-term assimilation of 14CO2 by illuminated fruiting structures produced malate as the major labelled product with less labelling in 3-phosphoglycerate, whereas the leaf showed a major incorporation into 3-phosphoglycerate. It seems likely that the fruiting structures of chickpea utilize phosphoenolpyruvate carboxylase for recapturing the respired carbon dioxide. 相似文献

Concurrent measurements of gas exchange, instantaneous isotope discrimination (Δ) against 13CO2 and C18O16O, and extent of 18O enrichment in H2O at the evaporative sites, were followed in a tropical forest pioneer, Piper aduncum, on two different days in Trinidad during February 1995. Δ13CO2 differed from that predicted from measurements of internal:external CO2 concentration (Ci/Ca) and showed a wide range of values which decreased throughout the course of the day. Derivation of Cc (the CO2 concentration at the carboxylation site) was not possible using carbon isotope discrimination under field conditions in situ and was derived assuming a constant value of internal transfer conductance (gw). Under low rates of assimilation the derived Cc/Ca, like Ci/Ca, remained relatively stable over the course of both days and ΔC18O16O followed evaporative demand. Lower values of ΔC18O16O on day 2 occurred in response to the indirect effect of increased leaf-to-air vapour pressure deficits (VPD) and reduced stomatal conductance. For the first time, direct determination of the δH218O of transpired water vapour (δt) allowed derivation of evaporative site enrichment without the prerequisite of isotopic steady state (ISS) defined in the Craig and Gordon model. Generally, δt was less enriched than the source water (δs) in the morning and more enriched in the afternoon, which would be predicted from an increase and decrease in ambient VPD, respectively. On both days, leaves of P. aduncum approached ISS (indicated where δt≈δs) between 1300 and 1500 h. Evaporative site enrichment was maintained into the late afternoon, despite a decrease in ambient VPD. The data presented provide a greater insight into the natural variation in isotopic discrimination under field conditions, which may help to refine models of terrestrial biome discrimination. 相似文献

Abstract. An apparatus is described to carry out pulse and pulse-chase experiments with 14 CO2 on intact, attached leaves with known, steady-state rates of photosynthesis under defined conditions of temperature, vapour pressure deficit and photon flux density. Data are presented which show that the pattern of distribution of 14 C between compounds in extracts of such leaves is a true reflection of the pathways of photosynthetic carbon metabolism in the leaf during steady-state photosynthesis. 相似文献

It is a matter of debate if there is a direct (short‐term) effect of elevated atmospheric CO2 concentration (Ca) on plant respiration in the dark. When Ca doubles, some authors found no (or only minor) changes in dark respiration, whereas most studies suggest a respiratory inhibition of 15–20%. The present study shows that the measurement artefacts – particularly leaks between leaf chamber gaskets and leaf surface, CO2 memory and leakage effects of gas exchange systems as well as the water vapour (‘water dilution’) effect on DCO2 measurement caused by transpiration – may result in larger errors than generally discussed. A gas exchange system that was used in three different ways – as a closed system in which Ca increased continuously from 200 to 4200 mmol (CO2) mol‐1 (air) due to respiration of the enclosed leaf; as an intermittently closed system that was repeatedly closed and opened during Ca periods of either 350 or 2000 mmol mol‐1, and as an open system in which Ca varied between 350 and 2000 mmol mol‐1– is described. In control experiments (with an empty leaf chamber), the respective system characteristics were evaluated carefully. When all relevant system parameters were taken into account, no effects of short‐term changes in CO2 on dark CO2 efflux of bean and poplar leaves were found, even when Ca increased to 4200 mmol mol‐1. It is concluded that the leaf respiration of bean and poplar is not directly inhibited by elevated atmospheric CO2. 相似文献

Δ13C were determined for herbarium specimens of 12 C3 plants (trees, shrubs and herbs) collected during the last 240 years in Catalonia, an area with a Mediterranean climate. Values were 19.91 (S.E. = 0.32, n= 21) for 1750–1760, 19–86 (S.E. = 0.21, n= 49) for 1850–1890 and 19.95 (S.E. = 0.29, n= 25) for 1925–1950, and decreased significantly to 18.87 (S.E. = 0.31, n= 29) for 1982–1988. More irregular temporal changes were found in Δ13C of two C4 species, but they also suggest a decrease in discrimination in recent decades. These results suggest that either carbon assimilation rates have increased or stomatal conductance has decreased, and therefore, that there has been an increase in water use efficiency over the last few decades. 相似文献

Influences of soil volume and an elevated CO2 level on growth and CO2 exchange for the Crassulacean acid metabolism plant Opuntia ficus-indica 总被引:1,自引:0,他引:1
Effects of the current (38 Pa) and an elevated (74 Pa) CO2 partial pressure on root and shoot areas, biomass accumulation and daily net CO2 exchange were determined for Opuntia ficus-indica (L.) Miller, a highly productive Crassulacean acid metabolism species cultivated worldwide. Plants were grown in environmentally controlled rooms for 18 weeks in pots of three soil volumes (2 600, 6 500 and 26 000 cm3 ), the smallest of which was intended to restrict root growth. For plants in the medium-sized soil volume, basal cladodes tended to be thicker and areas of main and lateral roots tended to be greater as the CO2 level was doubled. Daughter cladodes tended to be initiated sooner at the current compared with the elevated CO2 level but total areas were similar by 10 weeks. At 10 weeks, daily net CO2 uptake for the three soil volumes averaged 24% higher for plants growing under elevated compared with current CO2 levels, but at 18 weeks only 3% enhancement in uptake occurred. Dry weight gain was enhanced 24% by elevated CO2 during the first 10 weeks but only 8% over 18 weeks. Increasing the soil volume 10-fold led to a greater stimulation of daily net CO2 uptake and biomass production than did doubling the CO2 level. At 18 weeks, root biomass doubled and shoot biomass nearly doubled as the soil volume was increased 10-fold; the effects of soil volume tended to be greater for elevated CO2 . The amount of cladode nitrogen per unit dry weight decreased as the CO2 level was raised and increased as soil volume increased, the latter suggesting that the effects of soil volume could be due to nitrogen limitations. 相似文献
