The Brazilian swallowtail Eurytides lysithous is evidently a Batesian mimic of several Parides species. It is polymorphic for mimetic patterns in both sexes. Various populations contain from one to three major forms, and these seem to depend on two unlinked loci or supergenes. Samples from natural populations, and one reared brood, suggest that one locus controls two white-marked forms, with incomplete dominance producing a third heterozygous form. The heterozygotes are everywhere deficient from Hardy-Weinberg expectations. The second putative locus has an allele epistatic to the first locus which converts the white-marked forms to black, but epistasis is apparently incomplete in heterozygotes. The incomplete dominance and epistasis result in extraordinarily variable polymorphic populations and would allow a genetic analysis for comparison with those already done in the classic Batesian polymorphic swallowtails of the Old World.  相似文献

Collections of D. melanogaster from Japanese populations were analyzed for enzyme and chromosomal polymorphisms. Allelic frequencies at the Adh and alphaGpd loci were compared with polymorphic inversion (In(2L)B, In(2R)C) frequencies in the second chromosome. There was a significant positive correlation between the frequencies of AdhS and In(2L)B, caused by linkage. On the other hand, inversion-free cage populations maintained in the laboratory for a long time showed considerably larger variation in the frequencies of these enzyme alleles, which seem very likely to be a consequence of random drift. Two fitness components of these enzyme and chromosomal variants were measured in two different environmental conditions; neither of the two loci showed heterozygote superiority in viability or productivity, while the inversion heterozygotes showed a superior productivity compared to the corresponding homozygotes in the fluctuating environment. These findings are compatible with the hypothesis that polymorphic isozyme genes are maintained by random drift of neutral genes in natural populations, and that association with linked inversions is a historical accident.  相似文献

Three populations of the orchid species Cephalanthera rubra localized on neighbouring mineral islands in the Biebrza National Park (north-east Poland) were examined using 16 allozyme loci. The percentage of polymorphic loci in these C. rubra populations ranged from 25 to 31.3% and the mean number of alleles per polymorphic locus from 1.25 to 1.31. The levels of observed heterozygosity at polymorphic loci were higher than expected for all populations. The overabundance of heterozygotes was noted in every population. The values of fixation indices were highly negative. The genetic differentiation among the three populations was small ( F ST = 0.0173), albeit statistically significant ( P  < 0.001). The level of gene flow varied from 13 to 16. Fourteen multilocus genotypes were found among the 401 ramets sampled from the populations. Only four of them were common for all populations. The frequency of genotypes was different in these three populations of C. rubra . Some of the multilocus genotypes dominated in the given populations, others were found sporadically. Genetic variation within C. rubra populations is mainly the result of small population sizes and reflects the influences of breeding system and type of reproduction. The pink bollworm, Pectinophora gossypiella (Saunders) (Lepidoptera: Gelechiidae), a cotton pest probably native to Indo-Pakistan, invaded China at the beginning of the 20th century. Chinese P. gossypiella have been assumed to be the result of indiscriminate introductions from Pakistan and America by transport of cotton seed. We tested this long-held hypothesis and genotyped a total of 527 individuals from 14 sites at 13 microsatellite loci. We analyzed these data with traditional statistics as well as with Bayesian methods. The loci were, for the most part, highly polymorphic. The allelic richness of Chinese populations at six loci was greater than those of the Pakistani and American populations. Significant deficits of heterozygotes were recorded for all 14 populations, and null alleles were the most probable factor contributing to these deficits. Pairwise FST estimates showed that there was significant differentiation among the pooled Chinese, Pakistani, and American populations, and there was structure within most of the Chinese populations. The Bayesian analysis revealed that the combined Chinese, American, and Pakistani populations formed separate clusters, and the nine Chinese populations were divided into two clusters. Allelic frequency distributions showed that private and shared alleles within Chinese P. gossypiella were derived only partly from the Pakistani and American populations. The microsatellite-based genetic analyses suggested that the Chinese P. gossypiella populations originated from multiple sources.  相似文献

The California population of Composite Cross V of barley was used as the source of three subpopulations that were started from generations 10, 20 and 30, respectively, and were grown in parallel environmental conditions in Cambridge for eight generations. Outcrossing rates (0.2%) were even lower than in the California material, and heterozygotes were correspondingly rare, so that the populations were essentially mixtures of homozygous lines. Four esterase loci that were polymorphic in the base Composite Cross V remained so in all the derived populations, but showed considerable changes in allelic frequency over time, particularly at two of the genes. Multilocus analysis showed that strong directional changes occurred in all three populations, but they were not consistent. One particular genotype became predominant in the population derived from generation 10, whereas in the other two populations it was a genotype with different alleles at the Est1 and Est3 loci that rose to frequencies of more than 50%. Strong directional selection undoubtedly occurred in these populations, but did not cause parallel changes in esterase gene frequencies. These data do not facilitate a discrimination between the alternative explanations of hitchhiking or multilocus selection at these loci.  相似文献

Microsatellite markers were used to assess the genetic diversity and population structure in four populations of Elymus trachycaulus from British Columbia and one population of Elymus alaskanus from Northwest Territories. Fourteen microsatellite loci were used in this study. Our results indicated that E. trachycaulus is highly polymorphic, with an average percentage of polymorphic loci of 96.5% over the four populations. Average expected heterozygosity values (HE or gene diversity) varied from 0.418 to 0.585 with a mean of 0.497. Most of the genetic variation was found within populations (85%) and the differentiation among populations was found to be 15% (Fst = 0.15). Interpopulation genetic distances corresponded well with the geographic distance between the population sites of origin, as well as morphological characteristics. Tests for Hardy–Weinberg equilibrium (HWE) for all loci and all populations revealed that all loci significantly differ from HWE. Subsequent analysis indicated that departure from HWE at some loci was due to an excess of heterozygotes. Possible explanations for heterozygote excess are discussed. The most likely reason for observed heterozygote excess could be due to the polyploidy nature of the species.  相似文献

Allozyme genetic variability in five natural populations of Cyclobalanopsis championii (Fagaceae) in Taiwan was investigated using 12 loci from 9 enzyme systems. The average values of parameters describing within-population variation, expected heterozygosity (He = 0.151), the percentage of polymorphic loci per individual (P = 50%), the average number of alleles per locus (A = 1.7), effective number of alleles per locus (Ae = 1.25), and the average number of alleles per polymorphic loci (AP = 2.2) are comparable to those of other long-lived woody plants. The overall fixation index (Fis = 0.208) indicates a significant deficiency of heterozygotes at the population level. Allelic frequency deviation from Hardy-Weinberg equilibrium was found for different loci in different populations. An exact test for population differentiation using the Tools for Population Genetic Analyses program also indicates that allelic frequencies among populations are significantly different (P < .001). Among-population variation, Gst, accounted for 9.2% of the total heterozygosity. The population at Shouchia and the southernmost population Nanjenshan had higher inbreeding coefficients (0.177 and 0.153, respectively) than did the northern populations. Genetic drift is supported by the observations of the variance components of linkage disequilibrium and a large proportion of loci in Nanjenshan and Shouchia that show pairwise locus disequilibrium. We believe continuous genetic drift in the southern populations will increase genetic divergence among populations of C. championii in Taiwan. Significant correlation was found between elevation and expected heterozygosity. We therefore inferred that temperature is the most important ecological factor to influence the genetic diversity of C. championii.  相似文献

Blood specimens from Jalari and Brahmin caste populations of Andhra Pradesh, India, were examined for blood groups, red cell enzymes, and serum proteins. Of 33 genetic loci studied, 16 were observed to be invariant among both the castes, while common polymorphism or rare variants were observed in one or both populations for the other loci. Three rare heterozygotes at the phosphoglucoisomerase locus, two different peptidase A variants occurring once each and single cases of rare 6-phosphogluconate dehydrogenase and transferrin variants were recorded. Also a few cases of hemoglobin AS and anhaptoglobinemia were observed. The difference in rare variants between the two castes is conspicuous but large differences in their gene frequencies at the polymorphic loci were not observed. It is pointed out that the frequency of rare variants in the tribal and caste populations of Southern India appears to be higher than observed in temperate-dwelling civilized populations.  相似文献

Allozyme variation at 3 polymorphic enzyme loci (GPI, PGM, MDH) was studied in Trichostrongylus colubriformis. By means of a multivariate analysis it was shown that populations of worms harboured by naturally infected goats were genetically different from populations laboratory reared in lambs or rabbits. We have characterized 10 microsatellite loci for the tropical tree Entandrophragma cylindricum (Sprague) Sprague (sapelli) in order to genotype individuals in forest stands for estimation of the genetic diversity of the species. We used the technique of building a (GA)n microsatellite-enriched library by capture with streptavidin-coated magnetic beads. We assessed the polymorphism of seven microsatellites in 186 mature trees in a selectively logged stand (Dimako) and an unlogged stand (Ndama), both in Cameroon. All the loci were polymorphic, and the number of alleles was high, ranging from eight to 36, with a mean of 22.1. Both stands showed the same high level of genetic diversity (mean H(E) = 0.85) and a low genetic differentiation (FST = 0.007), indicating that genetic diversity was within rather than among populations. Five and three out seven loci in Dimako and Ndama, respectively, showed a deficit of heterozygotes. The seven loci enabled more than 97% of the mature trees in each stand to be identified. It was concluded that these markers can be efficiently used for gene flow studies.  相似文献

Allozyme electrophoresis was used to study genetic diversityamong populations of the freshwater, hermaphroditic snail Biomphalariacamerunensis in Cameroon. Five of 19 loci studied in 15 enzymesystems were polymorphic. Intrapopulation variation was evidentin 8 of 12 populations sampled and heterozygotes were presentin 6 of these. Two populations with large sampled sets werepanmictic. Although results for the remaining 4 populationswith heterozygotes present were inconclusive, our findings supportthe hypothesis that Biomphalaria are out-breeders. The allelesfor two loci, asparate aminotransferase-1 (AAT-1) and 6-phosphogluconatedehydrogenase (6-PGD), showed distribution patterns that suggestthey may be limited by climate or habitat. The slow migratingAAT-1 was the only allele found in the mountainous regions ofthe southwest. This region falls under the Cameroon-type climateand is the wettest region in the country. The faster allelewas restricted to the lowland rain forest of the south. For6-PGD the fast allele was found throughout the range of B. camerunensis,but the slower allele was found only in the mountainous regionswithin the Cameroon-type climate. Caenorhabditis elegans is a major laboratory model system yet a newcomer to the field of population genetics, and relatively little is known of its biology in the wild. Recent studies of natural populations at a single time point revealed strong spatial population structure and suggested that these populations may be very dynamic. We have therefore studied several natural C. elegans populations over time and genotyped them at polymorphic microsatellite loci. While some populations appear to be genetically stable over the course of observation, others seem to go extinct, with full replacement of multilocus genotypes upon regrowth. The frequency of heterozygotes indicates that outcrossing occurs at a mean frequency of 1.7% and is variable between populations. However, in genetically stable populations, linkage disequilibrium between different chromosomes can be maintained over several years at a level much higher than expected from the heterozygote frequency. C. elegans seems to follow metapopulation dynamics, and the maintenance of linkage disequilibrium despite a low yet significant level of outcrossing suggests that selection may act against the progeny of outcrossings.  相似文献

For isocitratedehydrogenase and phosphoglucoisomerase, the faster alleleswere found throughout the range of B. forskalii extending fromthe Sahelian regions of the north to rain forest of the south.The slower, rarer alleles for these loci were restricted tothe equatorial rain forest regions. For hydroxybutyrate dehydrogenase,the opposite was true for the rarer allele, which was restrictedto a small region in the arid, tropical climate. Historically documented founder events provide opportunities to assess the effects of population size reductions on genetic variation, but the actual magnitude of genetic change can be measured only when direct comparisons can be made to the source or ancestral population. We assayed variation at nine microsatellite loci in the translocated population of the Laysan finch ( Telespiza cantans ) at Pearl and Hermes reef (PHR), and compared the level of variation to that in the source population on Laysan Island. Heterogeneity in allele frequencies was highly significant at eight of the nine loci, primarily as a result of fluctuations in allele frequencies in the three PHR populations. Intra- and interpopulational measures of genetic diversity generally matched predictions based on the well-documented history of three islet populations at PHR: significantly lower numbers of alleles and polymorphic loci, as well as higher pairwise F ST values and genetic distance, were observed for the two populations that underwent severe size reductions. Changes in heterozygosity at single loci were unpredictable, as both significant increases and decreases were observed in founder populations. A significant excess of heterozygotes was found in two populations and was highly significant over all four finch populations ( P < 0.003). Estimates of effective population size from temporal changes in heterozygosity and allele frequencies were very small ( N e≤ 30) as a result of the founding events and the constraints of islet area on population numbers. We concluded that the PHR population is not adequate as a secondary genetic reserve for T. cantans , and an alternative refuge needs to be established.  相似文献

The Shanyi inbred A and E strains of the Chinese hamster are widely used in biomedical research, but detailed genetic characterization has been lacking. We developed microsatellite markers that could be used for genetic diversity analysis and linkage map construction. We isolated and characterized 16 novel microsatellite loci from a microsatellite-enriched genomic DNA library. These loci were genotyped in 48 animals from the two strains, and the polymorphic information content was determined. In the Shanyi A and E populations, 14 and 15 loci were found to be polymorphic, respectively, with polymorphic information content ranging from 0.1393 to 0.8082 and from 0.1109 to 0.7397, respectively. A total of 115 alleles were found for the 16 microsatellite loci in the two populations; the mean observed heterozygosity (H(O)) was 0.5191 and 0.4333 for the A and E populations, respectively, indicating marked genetic variation within the two populations. Correspondingly, the F(ST) values ranged from 0.002 to 0.9253, with an overall mean of 0.1935, indicating significant genetic difference between the two strains. The population differentiation levels were substantiated by Nei's genetic distance and full Bayesian analyses computed with STRUCTURE. Despite the genetic diversity and differentiation within and between the two inbred populations, the 48 individuals were correctly allocated into their original populations with high statistical confidence based on these 16 microsatellite loci. These novel microsatellite loci should be useful genetic markers for these two Chinese hamster inbred strains.  相似文献

Twelve polymorphic microsatellite DNA loci were isolated from genomic guppy DNA. We assessed the level of genetic diversity for these loci using individuals from five native Trinidadian populations. All the loci were polymorphic although there were considerable differences among loci and populations in allele frequencies. Within populations, the number of alleles ranged from 1 to 22 and observed heterozygosities ranged from 0.000 to 0.990. Allele frequencies differed substantially between populations suggesting divergence.  相似文献

Candida albicans is a diploid yeast that can undergo mating and a parasexual cycle, but is apparently unable to undergo meiosis. Characterization of the population structure of C. albicans has shown that reproduction is largely clonal and that mating, if it occurs, is rare or limited to genetically related isolates. Because molecular typing has delineated distinct clades in C. albicans, we have tested whether recombination was common within clades, but rare between clades. Two hundred and three C. albicans isolates have been subjected to multilocus sequence typing (MLST) and the haplotypes at heterozygous MLST genotypes characterized. The C. albicans isolates were distributed among nine clades, of which five corresponded to those previously identified by Ca3 fingerprinting. In each of these clades with more than 10 isolates, polymorphic nucleotide positions located on between 3 and 4 of the six loci were in Hardy-Weinberg disequilibrium. Moreover, each of these polymorphic sites contained excess heterozygotes. This was confirmed by an expanded analysis performed on a recently published MLST dataset for 1044 isolates. On average, 66% of polymorphic positions in the individual clades were in significant excess of heterozygotes over the five clades. These data indicate that mating within clades as well as self-fertilization are both limited and that C. albicans clades do not represent a collection of cryptic species. The study of haplotypes at heterozygous loci performed on our dataset indicates that loss of heterozygosity events due to mitotic recombination is moderately common in natural populations of C. albicans. The maintenance of substantial heterozygosity despite relatively frequent loss of heterozygosity could result from a selective advantage conferred by heterozygosity.  相似文献

Genetic diversity within and among six subpopulations of Larix decidua Mill. from two altitudinal transects of Swiss Alps was investigated using 6 enzyme systems coding for 8 loci. Globally, the mean proportion of polymorphic loci was 22.9%, the average number of alleles per locus was 1.3, and the mean expected heterozygosity was 0.095. Only 5.8% of the genetic variation resided among populations. The mean genetic distance was 0.006. Several significant differences of gene frequencies were found between different age classes. Positive values of the species mean fixation index observed in this study suggested a considerable deficit of heterozygotes in the populations of L. decidua of Swiss Alps. At one of the sites (Arpette), the highest subpopulation in elevation gave the lowest level of genetic diversity (as evidenced by the lowest proportion of polymorphic loci and the lowest mean expected heterozygosity) and the largest value of genetic distance when compared to other subpopulations. The genetic differences between the highest subpopulation and the other ones suggest that the founder effect may be an important factor influencing genetic differentiation of L. decidua populations at Arpette transect.  相似文献

The central and marginal populations of D. robusta differ greatly in the level of inversion polymorphism; the marginal populations are monomorphic or nearly so and the central populations are highly polymorphic. This paper presents the frequencies of alleles at forty gene loci in various populations of D. robusta, studied by electrophoresis of proteins and enzymes. Population samples were obtained from eight widely separated populations of D. robusta which included the central, the extreme marginal and the intervening populations between the center and the margins. We find that the proportion of polymorphic loci and average heterozygosity per individual is slightly higher in the marginal populations than the central populations. In D. robusta on an average, 39% of the loci are polymorphic and the average proportion of loci heterozygous per individual is 11%. A breakdown of loci in three categories, viz, hydrolytic enzymes and some other enzymes, larval proteins and glycolytic and Kreb's cycle enzymes, shows that in all populations the level of polymorphism is highest in the hydrolytic enzymes, intermediate in larval proteins and least in the glycolytic and Kreb's cycle enzymes. On the average, the proportion of loci heterozygous per individual for three groups of loci is: hydrolytic enzymes and others (.164), larval proteins (.115) and glycolytic and Kreb's cycle enzymes (.037). We also observe that in all populations the level of polymorphism on the X chromosome is far less than the expected 38%; in salivary gland cells the euchromatic length of the X chromosome is 38% of the entire genome. Lower levels of polymorphism for the X chromosome loci are explained due to low probability of balanced polymorphisms for the X-linked loci since the conditions for establishment of balanced polymorphism for X-linked loci are more restrictive than for the autosomal loci.-The polymorphic loci can be grouped according to pattern of allele frequencies in different populations as follows: (1) The allele frequencies are similar in all populations at the XDH, Pep-1 and Hex-1 loci. (2) The alleles at the Est-1, Est-2, Amy loci and the AP-4(1.0) and the LAP-1(.90) alleles show north south clinal change in frequency. (3) There is north south and east west differentiation at the Pt-5, Pt-8 and Pt-9 loci and the allele AP-4(.81). (4) Polymorphism at loci such as Fum, B.Ox, Hex-8, Pep-2 and Pep-3 are restricted to only one or two of the populations. (5) Allele frequencies at the MDH and ODH loci fluctuate between populations. (6) Allele frequencies at many polymorphic loci such as Est-1, Est-2, LAP-1, AP-4, Pt-5, Pt-8, Pt-9, Pt-16, MDH, Fum change clinally within a gene arrangement. The pattern of gene variation in D. robusta is very complex and cannot be easily explained due to migration of neutral alleles between once-isolated populations or to semi-isolation of neutral alleles. The observations of the pattern of allele variation in different populations, high levels of polymorphism in the marginal populations which have small population size and low levels of polymorphism of the X chromosome loci all support the argument in favor of balancing selection as the main mechanism for the maintenance of these polymorphisms. Environmental factors must play a role in the maintenance of a great deal of these polymorphisms, since we observe clinal allele frequency changes even within a given inversion type.  相似文献
