Wirkung von Cyanid auf das Membranpotential von Blattzellen von Mnium cuspidatum

NaCN in Konzentrationen von mehr als 5 · 10^-5 M depolarisieren das Membranpotential von Mnium-Blattzellen (˜ —210 mV) rasch und reversibel um etwa 75%; 1 · 10^-5 M NaCN bewirkt eine etwa 50%ige Hemmung. Der Effekt tritt sowohl im Licht als auch im Dunkeln auf. Die Ergebnisse deuten auf die Existenz elektrogener Ionentransportmechanismen.

Modellversuch zur Ableitung von Aktionspotentialen von der Nervenfaser

Zusammenfassung In dieser Arbeit wird an einem Modell im elektrolytischen Trog das Feld um eine kontinuierlich leitende Nervenfaser nachgebildet, wie es bei der Erregungsleitung auftritt. In dieses stationäre Feld werden Modelle von Ableitelektroden gebracht. Es handelt sich um ca. 1000fach vergrößerte, unisolierte und isolierte Wolframelektroden, wie sie aus 0,1 mm Wolframdraht im Institut hergestellt werden.Eine Ableitelektrode, die sich nahe an der Nervenfaser befindet und somit in dem Potentialfeld um die Nervenfaser liegt, nimmt in diesem ein gewisses Potential an. Auf diese Weise kann ein Teil des Aktionspotentials abgegriffen werden. Wie groß dieser Teil im Verhältnis zur Gesamtspannung über der Membran ist und von welchen Größen die Spannung der Ableitelektrode im Modell abhängt, wird geklärt.Die Aktionsfelder werden dargestellt, wie sie sich ungestört und mit den Modellen der Ableitelektroden ergeben. Zusätzlich werden Modelle mit zylindrischem und halbkugelförmigem Ende zum Vergleich getestet. Die Spannung der Ableitelektrode in Abhängigkeit vom Abstand von der Nervenfaser wird gemessen.

Model experiments for the study of tracing action potentials from nerve fibres

Summary The electrical field surrounding a continuosly conducting nerve fibre was artificially produced in a model in an electrolytic trough. Models of insulated and uninsulated microelectrodes about thousand times the diameter of the tungsten electrodes as produced in this institute from 0.1 mm wire were inserted into this stationary field. An electrode located near the nerve fibre in the field asumes an electrical potential. Thus a part of the action potential can be recorded. The magnitude of this voltage-together with the parameters on which it depends — with respect to the voltage across the membrane of the nerve fibre was determined. The action potential fields are shown both undisturbed and with the model electrodes inserted. Electrodes with cylindrical and hemispherical tips were also tested. In addition the influence of the distance from nerve fibre on the voltage of the model electrodes is described.

     

Wirkung von Endoxan auf die Chromosomen von HeLa-Zellen

Mutagenic effects induced by solutions of Cytoxan-Substance and its activated compound (rat-passage) are tested in He-La cells.Cytoxan-substance induces chromatid aberrations as well as mitotic depression 44–48 hs after treatment in proportion to the concentration. 22–24 hs after treatment no effects are seen. Activated Cytoxan induces gaps and breaks in HeLa cells harvested 22 hs after treatment. Reunion-figures are missing.Incubating cultures up to 46 hs after treatment leads to few gaps, more breaks and a high number of reunion-figures.It is not known if activated Cytoxan delays the cell cycle or single phases of the cycle. It is discussed that autoradiographic experiments are nessessary to get more information on sensitive and/or delayed phases of the cycle under influence of activated Cytoxan. Furthermore those studies have proved to be helpful to solve questions about the rejoining mechanism.

Der Jahresgang von Neuston im Golf von Neapel

The annual fluctuations in the occurrence of 91 taxa which were identified, made the recognition of 4 main seasonal populations possible. Trends were observed, as well as analogies to other patterns of distribution. On an average, more than half of the fauna was displaced from month to month, but the variation in the density of subtropical neuston appeared to be less over an annual period than it was during the day, and from region to region. The dominance of certain species and occurrence of neuston throughout the year demonstrated, that regionally at least a biocoenosis exists continously at the surface of the sea.     

Anreicherung von Morphogenetischen Substanzen in Lichtpflanzen von Acetabularia Mediterranea Unter dem Einfluss von Puromycin

Zusammenfassung Unter dem Einfluß von Puromycin, das spezifisch aber reversibel die Proteinsynthese blockiert, erfolgt in belichteten kernhaltigen Pflanzenteilen von Acetabularia mediterranea eine Anreicherung morphogenetischer Substanzen. Wie Transplantationsversuche zwischen A. mediterranea und A. crenulata zeigen, betrifft diese Anreicherung sowohl die artspezifischen als auch die nichtartspezifischen morphogenetischen Substanzen. Die Anreicherung wird durch Actinomycin stark gehemmt.Es wird diskutiert, welche Schlußfolgerungen daraus für die Natur der artspezifischen morphogenetischen Substanzen gezogen werden können.

Accumulation of morphogenetic substances in plants of Acetabularia mediterranea in light under the influence of puromycin

Summary Morphogenetic substances are accumulated in nucleated parts of the alga Acetabularia mediterranea in light, if they are treated with puromycin, which inhibits specifically but reversibel protein synthesis. Transplantation experiments between A. mediterranea and A. crenulata show, that there was an accumulation of both species specific morphogenetic substances and also of nonspecies specific ones. This accumulation is strongly inhibited if the parts are treated with actinomycin together with puromycin.The conclusions, which can be drawn from these results for the nature of the species specific substances, are discussed.

Cytochemischer Nachweis von saurer Desoxyribonuclease im Cytoplasma von Blutzellen

Zusammenfassung Es wird ein cytochemisches Verfahren zum Nachweis der sauren Desoxyribonuclease in Blutzellen beschrieben. Das Prinzip des Nachweises beruht auf der Depolymerisierung von Desoxyribonucleinsäuren der Inkubationslösung zu Mononucleotiden, deren in 3-Stellung stehendes Phosphation durch ebenfalls der Inkubationslösung zugegebene saure Phosphatase freigesetzt und mit dem Verfahren der Schwermetallsalz-Präzipitation in Form des Bleisulfids mikroskopisch erkennbar dargestellt wird.In allen Leukocytenarten, Erythroblasten, Reticulocyten und Thrombocyten ist saure Desoxyribonuclease nachweisbar. Die Spezifität der Methode wird diskutiert.

Cytochemical staining for acid deoxyribonuclease in cytoplasm of blood cellsI. Method

Summary A technique for the cytochemical demonstration of acid deoxyribonuclease in blood cells is presented in this paper. Blood slides were incubated in a medium containing highly polymerized DNA, lead nitrate an acid phosphatase. DNA is depolymerized by the deoxyribonuclease and mononucleotides are formed. These serve as substrate for exogenous acid phosphatase, which release phosphate ions. The latter are locally precipitated by lead ions.Acid deoxyribonuclease occurs in all leucocytes, erythroblasts, reticulocytes and thrombocytes. The specifity of this method is discussed.

Auswahl von GC-Phasen und Optimierung von Trichothecen-Trennungen mittels Computersimulation

Gas chromatography combines high resolution with good sensitivity (ECD) for the simultaneous analysis of trichothecenes. With the aid of computer-supported GC simulation based on thermodynamic retention indices, optimized GC methods for 4 different phase polarities have been developed. The verification with polar and unpolar columns in 2 GCs under optimized separation conditions provides highly reliable results.

Variation von Glycerin,Sorbit und anderen Metabolitgehalten infolge von Temperaturstress bei Larven von Callitroga macellaria

Sorbitol, sorbitol phosphate, glycerol, α-glycerol phosphate, lactic and pyruvic acid were determined after storing of non-feeding Callitroga larvae hot or cold. Compared with controls, the high levels of pyruvic acid and lactic acid indicate the increase of anaerobic metabolism by temperature stress. At the same time glycerol increased about threefold, sorbitol increased about seventeenfold after heating and about thirty-fourfold after cooling. The level of glycerol phosphate is lowered after heating and after cooling. Sorbitol phosphate shows after heating a trend to decrease. Heat causes a great standard error.Based on this and other facts it is supposed that different kinds of stress result in a lack of metabolic energy. Thus the pathway of glucose phosphate to direct oxidation and formation of triose phosphate by the pentose phosphate cycle is energetically favoured because the phosphorylation of hexose phosphate to hexose diphosphate is avoided. Triose phosphate can enter the glycolytic pathway. The reduced coenzyme nicotinamide adenine dinucleotide phosphate (NADPH) is generated by the formation of polyols (or polyolphosphates).     

Wirkungen von PS-II-Herbiziden auf das endogene Hormonsystem von Hordeum-Keimlingen

Bestimmungen endogener Cytokiningehalte in Hordeum-Keimlingen zeigten eine starke Erhöhung in fünf und sieben Tage alten Pflanzen nach Applikation von Simazin. Im Zusammenhang mit früheren Untersuchungen scheint es denkbar, daß Simazin den Stoffwechsel der Prenyllipide (Chlorophylle, Carotinoide, Benzochinone) in Chloroplasten über eine Erhöhung des Cytokininspiegels in Richtung auf eine Sonnentypadaptation umstellt. Mögliche Bedeutungen der Wirkung von Simazin auf die Gehalte endogener Abscisine, Gibberelline und Indol-3-essigsäure in Hordeum-Keimlingen verschiedenen Alters werden erörtert.
