Isolation of bacteria and 16S rDNAs from Lake Vostok accretion ice   总被引:6,自引:0,他引:6  
Lake Vostok, the largest subglacial lake in Antarctica, is separated from the surface by ≈ 4 km of glacial ice. It has been isolated from direct surface input for at least 420 000 years, and the possibility of a novel environment and ecosystem therefore exists. Lake Vostok water has not been sampled, but an ice core has been recovered that extends into the ice accreted below glacial ice by freezing of Lake Vostok water. Here, we report the recovery of bacterial isolates belonging to the Brachybacteria , Methylobacterium , Paenibacillus and Sphingomonas lineages from a sample of melt water from this accretion ice that originated 3593 m below the surface. We have also amplified small-subunit ribosomal RNA-encoding DNA molecules (16S rDNAs) directly from this melt water that originated from α- and β-proteobacteria, low- and high-G+C Gram-positive bacteria and a member of the Cytophaga / Flavobacterium / Bacteroides lineage.  相似文献

The 3561 m Vostok ice core sample originating from the subglacial Lake Vostok accretion (frozen lake water) ice with sediment inclusions was thoroughly studied by various means to confirm the presence of the thermophile bacterium Hydrogenophilus thermoluteolus reported earlier in the 3607 m accretion ice sample. PCR and molecular-phylogenetic analyses performed in two independent laboratories were made using different 16S rRNA gene (rrs) targeted primers. As a result, rrs-targeted PCR permitted to recover several very closely related clones with a small genetic distance to Hydrogenophilus thermoluteolus (< 1%). In addition, RubisCO (cbbL or rbcL) and NiFe-Hydrogenase (hoxV or hupL) targeted PCR have also allowed to recover sequences highly related to Hydrogenophilus thermoluteolus. All these results point to the presence of thermophilic chemoautotrophic microorganisms in Lake Vostok accretion ice. They presumably originate from deep faults in the bedrock cavity containing the lake in which episodes of seismotectonic activity would release debris along with microbial cells.  相似文献

Two ~4 m vertical sequences of basal ice were collected from tunnels dug into the northern lateral margin of Taylor Glacier, McMurdo Dry Valleys, Antarctica. In both cases the basal sequences exhibit two contrasting ice facies groups; clean (debris-free) and banded dispersed (debris-rich). Debris-rich ices exhibit elevated CO2 and depleted O2 concentrations compared to the clean facies. Bacterial cell numbers, respiration rates, and nutrient concentrations are highest in debris-rich layers. Together, our geochemical and biological data indicate that microbial heterotrophic respiration is likely occurring in situ within the basal ice matrix at ambient temperatures near ?15°C. This implies that the basal ice zone of polar glaciers and larger ice sheets is a viable subglacial microbial habitat and active biome of significant volume that has not previously been considered.  相似文献

Solute yields, laboratory dissolution data and both chemical and isotopic markers of rock weathering reactions are used to characterise the biogeochemistry of glacial meltwaters draining a maritime Antarctic glacier. We find that delayed flowpaths through ice-marginal talus and moraine sediments are critical for the acquisition of solute from rock minerals because delayed flowpaths through subglacial sediments are absent beneath this small, cold-based glacier. Here the mechanisms of weathering are similar to those reported in subglacial environments, and include sub-oxic conditions in the early summer and increasingly oxic conditions thereafter. Up to 85% of the NO3 ? and 65% of the SO4 2? are most likely produced by bacterially mediated reactions in these ice marginal sediments. However, reactive pyrite phases are sparse in the host rocks, limiting the export of Fe, SO4 2? and cations that may be removed by weathering once pyrite oxidation has taken place. This means that dissolution of Ca2+ and Na+ from carbonate and silicate minerals dominate, producing moderate cationic denudation yields from Tuva Glacier (163 Σ*meq+ m?2 a?1) compared to a global range of values (94–4,200 Σ*meq+ km?2 a?1). Overall, crustally derived cations represent 42% of the total cationic flux, the rest being accounted for by snowpack sources.  相似文献

Lake Pääjärvi, a boreal Finnish lake, was investigated in winter for weather conditions, structure and thickness of ice and snow, solar radiation, and under-ice current and temperature. Heat budget of Lake Pääjärvi in January–March was governed by terrestrial radiation losses of 20–35 W m?2 recompensed by ice growth of 0.5–1.0 cm day?1. In April, snow melted, albedo decreased from 0.8 to <0.1, and the mean ice melt rate was 1.5 cm day?1. Internal melting and surface melting were about equal. The mean turbulent heat loss was small. The heat flux from the water to ice was about 5 W m?2 in winter, increasing to 12 W m?2 in the melting season. The light attenuation coefficient was 1.1 m?1 for the congelation ice (black ice) in winter, compared with 1.5 m?1 for the lake water, and it was up to 3 m?1 for candled congelation ice in spring, and about 10 m?1 for superimposed ice (white ice) and snow. Gas bubbles were the main factor that reduced the transparency of ice. The radiation penetrating the ice heated the water body causing convective currents and horizontal heat transfer. This increased the temperature of the water body to about 3°C before the ice break-up. After the snow had melted, the euphotic depth (the depth of 1% surface irradiance) was estimated as 2.0 m, only two-thirds that in summer.  相似文献

Decontamination of ice cores is a critical issue in phylogenetic studies of glacial ice and subglacial lakes. At the Vostok drill site, a total of 3650 m of ice core have now been obtained from the East Antarctic ice sheet. The ice core surface is coated with a hard-to-remove film of impure drilling fluid comprising a mixture of aliphatic and aromatic hydrocarbons and foranes. In the present study we used 16S rRNA gene sequencing to analyze the bacterial content of the Vostok drilling fluid sampled from four depths in the borehole. Six phylotypes were identified in three of four samples studied. The two dominant phylotypes recovered from the deepest (3400 and 3600 m) and comparatively warm (-10 degrees C and -6 degrees C, respectively) borehole horizons were from within the genus Sphingomonas, a well-known degrader of polyaromatic hydrocarbons. The remaining phylotypes encountered in all samples proved to be human- or soil-associated bacteria and were presumed to be drilling fluid contaminants of rare occurrence. The results obtained indicate the persistence of bacteria in extremely cold, hydrocarbon-rich environments. They show the potential for contamination of ice and subglacial water samples during lake exploration, and the need to develop a microbiological database of drilling fluid findings.  相似文献

Biogeochemical dynamics under seasonal ice cover were investigated in the shallow (<10 m) water column of highly productive Georgetown Lake, western Montana, USA. This high altitude (1,800 m) reservoir is well-mixed in summer, but becomes strongly stratified under ice cover (mid-November–mid-May). A rapid drop in dissolved oxygen (DO) concentration and rise in dissolved inorganic carbon (DIC) concentration was observed after the onset of ice, with a corresponding increase in δ18O-DO and decrease in δ13C-DIC, likely caused by respiration (R) of organic carbon. Photosynthesis/respiration ratios (P/R) estimated from simultaneous measurement of DO and δ18O-DO were near unity prior to ice formation but then systematically decreased with time and depth in the lake under ice cover. P/R in the water column was higher at a shallower monitoring site compared to a deeper site near the dam outlet, which may have been important for over-winter survival of salmonids. By March, the bottom 3 m of the water column at both sites was anoxic, with the bottom 1 m being euxinic. Elevated concentrations of dissolved sulfide, ammonium, phosphate, Fe2+, and Mn2+ in deep water suggest coupling of organic carbon degradation with reduction of a number of electron acceptors (e.g., Fe3+, \({\text{NO}}_{3}^{ - } ,\;{\text{SO}}_{4}^{2 - }\) ). The concentrations and δ34S values of H2S in the deep water and \({\text{SO}}_{4}^{2 - }\) in the shallow water were similar, indicating near-complete reduction of sulfate in the euxinic zone. Late in the winter, an influx of isotopically heavy DIC was noted in the deep water coincident with a buildup of dissolved CH4 to concentrations >1 mM. These trends are attributed to acetoclastic methanogenesis in the benthic sediments. This pool of dissolved CH4 was likely released from the lake to the atmosphere during spring ice-off and lake turnover.  相似文献

Knowledge on the relative effects of biological activity and precipitation/dissolution of calcium carbonate (CaCO3) in influencing the air-ice CO2 exchange in sea-ice-covered season is currently lacking. Furthermore, the spatial and temporal occurrence of CaCO3 and other biogeochemical parameters in sea ice are still not well described. Here we investigated autotrophic and heterotrophic activity as well as the precipitation/dissolution of CaCO3 in subarctic sea ice in South West Greenland. Integrated over the entire ice season (71 days), the sea ice was net autotrophic with a net carbon fixation of 56 mg C m?2, derived from a sea-ice-related gross primary production of 153 mg C m?2 and a bacterial carbon demand of 97 mg C m?2. Primary production contributed only marginally to the TCO2 depletion of the sea ice (7–25 %), which was mainly controlled by physical export by brine drainage and CaCO3 precipitation. The net biological production could only explain 4 % of this sea-ice-driven CO2 uptake. Abiotic processes contributed to an air-sea CO2 uptake of 1.5 mmol m?2 sea ice day?1, and dissolution of CaCO3 increased the air-sea CO2 uptake by 36 % compared to a theoretical estimate of melting CaCO3-free sea ice. There was a considerable spatial and temporal variability of CaCO3 and the other biogeochemical parameters measured (dissolved organic and inorganic nutrients).  相似文献

We describe the climatology, hydrology and biogeochemistry of an extreme nitrogen deposition event that occurred in the highly glacierised environment of the European High Arctic during June 1999. Meteorological analysis, three-dimensional air mass trajectories and a 3D transport model show that blocking high pressures over Scandinavia and the rapid advection of western European pollution toward Svalbard were sufficient to cause the most concentrated (1.15 ppm NO3–N and 1.20 ppm NH4–N), high magnitude (total 26 mm and up to 2.4 mm h?1 at 30 m above sea level) nitrogen deposition event on record in this sensitive, high Arctic environment (78.91° N, 11.93° E). Since the event occurred when much of the catchment remained frozen or under snow cover, microbial utilisation of nitrogen within snowpacks and perennially unfrozen subglacial sediments, rather than soils, were mostly responsible for reducing N export. The rainfall event occurred long before the annual subglacial outburst flood and so prolonged (ca. 10 day) water storage at the glacier bed further enhanced the microbial assimilation. When the subglacial outburst eventually occurred, high runoff and concentrations of NO3 ? (but not NH4 +) returned in the downstream rivers. Assimilation accounted for between 53 and 72% of the total inorganic nitrogen deposited during the event, but the annual NO3 ? and NH4 + runoff yields were still enhanced by up to 5 and 40 times respectively. Episodic atmospheric inputs of reactive nitrogen can therefore directly influence the biogeochemical functioning of High Arctic catchments, even when microbial activity takes place beneath a glacier at a time when terrestrial soil ecosystems remain frozen and unresponsive.  相似文献

Antarctic subglacial lakes have, over the past few years, been hypothesised to house unique forms of life and hold detailed sedimentary records of past climate change. Testing this hypothesis requires in situ examinations. The direct measurement of subglacial lakes has been considered ever since the largest and best-known lake, named Lake Vostok, was identified as having a deep water-column. The Subglacial Antarctic Lake Environments (SALE) programme, set up by the Scientific Committee on Antarctic Research (SCAR) to oversee subglacial lakes research, state that prior exploration of smaller lakes would be a “prudent way forward”. Over 145 subglacial lakes are known to exist in Antarctica, but one lake in West Antarctica, officially named Ellsworth Subglacial Lake (referred to hereafter as Lake Ellsworth), stands out as a candidate for early exploration. A consortium of over 20 scientists from seven countries and 14 institutions has been assembled to plan the exploration of Lake Ellsworth. An eight-year programme is envisaged: 3 years for a geophysical survey, 2 years for equipment development and testing, 1 year for field planning and operation, and 2 years for sample analysis and data interpretation. The science experiment is simple in concept but complex in execution. Lake Ellsworth will be accessed using hot water drilling. Once lake access is achieved, a probe will be lowered down the borehole and into the lake. The probe will contain a series of instruments to measure biological, chemical and physical characteristics of the lake water and sediments, and will utilise a tether to the ice surface through which power, communication and data will be transmitted. The probe will pass through the water column to the lake floor. The probe will then be pulled up and out of the lake, measuring its environment continually as this is done. Once at the ice surface, any water samples collected will be taken from the probe for laboratory analysis (to take place over subsequent years). The duration of the science mission, from deployment of the probe to its retrieval, is likely to take between 24 and 36 h. Measurements to be taken by the probe will provide data about the following: depth, pressure, conductivity and temperature; pH levels; biomolecules (using life marker chips); anions (using a chemical analyzer); visualisation of the environment (using cameras and light sources); dissolved gases (using chromatography); and morphology of the lake floor and sediment structures (using sonar). After the probe has been retrieved, a sediment corer may be dropped into the lake to recover material from the lake floor. Finally, if time permits, a thermistor string may be left in the lake water to take time-dependent measurements of the lake’s water column over subsequent years. Given that the comprehensive geophysical survey of the lake will take place in two seasons during 2007–2009, a two-year instrument and logistic development phase from 2008 (after the lake’s bathymetry has been assessed) makes it possible that the exploration of Lake Ellsworth could take place at the beginning of the next decade.  相似文献

Nitrate dual stable isotopes (δ15N and δ18O of NO3 ?) have proven to be a powerful technique to elucidate nitrogen (N) cycling pathways in aquatic systems. We applied this technique for the first time in the pelagic zone of a small temperate meso-eutrophic lake to identify the dominant N cycling pathways, and their spatial and temporal variability. We measured the lake NO3 ? δ15N and δ18O signatures over an annual cycle and compared them to that of the watershed. Both δ15N and δ18O of NO3 ? in the lake increased during summer relative to the inputs. Relationships between lake NO3 ? isotopic composition and concentrations were different across thermal strata with an apparent isotope effect in the epilimnion of 15εepi = 4.6‰ and 18εepi = 10.9‰. We found a strong deviation of the lake NO3 ? δ18O and δ15N from the expected 1:1 line for assimilation (slope = 1.73) suggesting that nitrification was co-occurring. We estimated that nitrification could support between 5 and 30% of nitrate-based production during the growing season, but was negligible in early spring and fall, and probably more dominant under ice. We showed that the technique is promising to study N processes at the ecosystem scale in shallow lakes, particularly during winter. Our results suggest that recycled NO3 ? could support primary productivity and influence phytoplankton composition in the surface waters of small lakes.  相似文献

In the last 40 years, the shallow steppe lake, Neusiedler See, was ice covered between 0 and 97 days. The North Atlantic Oscillation (NAO) as well as the Mediterranean Oscillation affected the lake and its conditions during winter. Both climate indices correlated negatively with the duration of ice cover and the timing of ice-out. Average winter phytoplankton biomass increased from less than 0.2 (0.05–0.84) mg FM l?1 in the late 1960s/beginning of 1970s to 3.1 (1.72–5.61) mg FM l?1 in the years 2001–2004. The increase in annual winter biomass of phytoplankton was associated with a significant shift in the composition of the algal assemblage. In the winter 1997/1998, diatoms contributed between 40 and 80% to the phytoplankton biomass while in 2006/2007 cyanoprokaryotes contributed 46%. Mean chlorophyll-a concentrations during winter were significantly correlated with those of total phosphorus (Ptot). Together with cold-water species (rotifer Rhinoglena fertöensis), perennial, eurythermal ones (copepod Arctodiaptomus spinosus) contributed to the zooplankton community. High zooplankton numbers were encountered when rotifers, particularly when densities of Rhinoglena fertöensis were high (r 2 = 0.928). Zooplankton abundance and biomass varied from year to year but correlated positively with Chl-a (biomass ? r 2 = 0.69; numbers ? r 2 = 0.536). Winter zooplankton populations were primarily influenced by winter conditions, but in early winter also by survival of autumn populations, i.e., the more adults of Arctodiaptomus spinosus survived into winter, the higher was the zooplankton biomass in early winter. Phyto- and zooplankton dynamics in shallow lakes of the temperate region seem to critically depend on the biomass in autumn and on winter conditions, specifically on ice conditions and thus are related to climate signals such as the NAO.  相似文献

Subglacial environments are largely anoxic, contain organic carbon (OC) overridden by glacier ice during periods of advance, and harbour active microbial communities. This creates favourable conditions for OC degradation via methanogenesis. It has been hypothesized that OC beneath ice sheets is converted to methane (CH4) and may be released to the atmosphere during retreat. However, there are limited data available to support this contention. Here, we present new data on the abundance, diversity and activity of methanogenic archaea and the amount and character of OC in subglacial sediments from Arctic and Antarctic glacial systems based on different substrate types. We employed long‐term laboratory incubations to quantify the CH4 production potential in different subglacial settings. Significant numbers of methanogens (up to 7 × 104 cells g?1) were detected in the samples and clones of Methanomicrobiales and Methanosarcinales were identified in clone libraries. Long lag periods (up to >200 days) were observed before significant CH4 concentrations were measured. We report order of magnitude differences in rates of CH4 production (101–105 fmol g?1 d?1) in different subglacial sediments, reflecting contrasts in the origin of the sediment and the OC character. Hence, we predict that contrasting rates of CH4 production are likely to occur beneath glaciers and ice sheets that overran different types of substrate. We subsequently estimated the potential for CH4 production beneath the Laurentide/Inuitian/Cordilleran and Fennoscandian Ice Sheets during a typical 85 ka Quaternary glacial/interglacial cycle. CH4 production from lacustrine‐derived OC is likely to be an order of magnitude higher (~6.3–27 Pg C) than that from overridden soils (~0.55–0.68 Pg C), possibly due to a difference in lability between lacustrine and soil OC. While representing a fraction of the entire OC pool (~418–610 Pg C), this finding highlights the importance of considering the character of different OC pools when calculating subglacial CH4 production.  相似文献

The debris-rich basal ice layers of a high Arctic glacier were shown to contain metabolically diverse microbes that could be cultured oligotrophically at low temperatures (0.3 to 4°C). These organisms included aerobic chemoheterotrophs and anaerobic nitrate reducers, sulfate reducers, and methanogens. Colonies purified from subglacial samples at 4°C appeared to be predominantly psychrophilic. Aerobic chemoheterotrophs were metabolically active in unfrozen basal sediments when they were cultured at 0.3°C in the dark (to simulate nearly in situ conditions), producing 14CO2 from radiolabeled sodium acetate with minimal organic amendment (≥38 μM C). In contrast, no activity was observed when samples were cultured at subfreezing temperatures (≤−1.8°C) for 66 days. Electron microscopy of thawed basal ice samples revealed various cell morphologies, including dividing cells. This suggests that the subglacial environment beneath a polythermal glacier provides a viable habitat for life and that microbes may be widespread where the basal ice is temperate and water is present at the base of the glacier and where organic carbon from glacially overridden soils is present. Our observations raise the possibility that in situ microbial production of CO2 and CH4 beneath ice masses (e.g., the Northern Hemisphere ice sheets) is an important factor in carbon cycling during glacial periods. Moreover, this terrestrial environment may provide a model for viable habitats for life on Mars, since similar conditions may exist or may have existed in the basal sediments beneath the Martian north polar ice cap.  相似文献

Recent measurements have demonstrated unprecedented increase in atmospheric deposition of nutrients in many parts of India. To determine whether atmospheric nutrient inputs would increase phytoplankton growth and catchment dissolved organic carbon (DOC) flushing to constrain benthic algae, we analyzed NO3 ? and PO 4 ?3 in atmospheric deposits; nutrients and DOC in runoff and lake water and standing crop biomass of phytoplankton and periphyton at Jaisamand Lake of Rajasthan, India. Atmospheric deposition of NO3 ? (7.18–29.95 kg ha?1 year?1) and PO 4 ?3 (0.56–2.15 kg ha?1 year?1) showed a consistently rising trend across the year. Microbial biomass and activity in catchment increased in response to atmospheric deposition. Lake DOC and nutrients showed strong coherence with their terrestrial and atmospheric fluxes. Phytoplankton development showed significant linearity with atmospheric input of nutrients. Air-driven input appeared to have compensated the nutrient constraints to phytoplankton during drought. The N:P stoichiometry of deposition and that of lake water indicated that, although there was a seasonal switchover to N- or P-limitation, phytoplankton were mainly co-limited by N and P due probably to the synergistic effects of combined N + P enrichment in the pelagic zone of the lake. Periphyton standing crop showed inverse relationship with phytoplankton and lake DOC. The study indicated that enhanced phytoplankton development and terrestrial DOC flushing in response to atmospheric nutrient input attenuated light penetration to constrain algal periphyton. We suggests that data on these issues may be considered in developing aquatic ecosystem models to establish future links between changing air–water–land interactions and associated shifts in lake ecosystem functioning for more accurately predicting climate change drivers and designing integrated lake basin management strategies.  相似文献

We determined fatty acid (FA) profiles and carbon stable isotopic composition of individual FAs (δ13CFA values) from sea ice particulate organic matter (i-POM) and pelagic POM (p-POM) in the Bering Sea during maximum ice extent, ice melt, and ice-free conditions in 2010. Based on FA biomarkers, differences in relative composition of diatoms, dinoflagellates, and bacteria were inferred for i-POM versus p-POM and for seasonal succession stages in p-POM. Proportions of diatom markers were higher in i-POM (16:4n-1, 6.6–8.7 %; 20:5n-3, 19.6–25.9 %) than in p-POM (16:4n-1, 1.2–4.0 %; 20:5n-3, 5.5–14.0 %). The dinoflagellate marker 22:6n-3/20:5n-3 was highest in p-POM. Bacterial FA concentration was higher in the bottom 1 cm of sea ice (14–245 μg L?1) than in the water column (0.6–1.7 μg L?1). Many i-POM δ13CFA values were higher (up to ~10 ‰) than those of p-POM, and i-POM δ13CFA values increased with day length. The higher i-POM δ13CFA values are most likely related to the reduced dissolved inorganic carbon (DIC) availability within the semi-closed sea ice brine channel system. Based on a modified Rayleigh equation, the fraction of sea ice DIC fixed in i-POM ranged from 12 to 73 %, implying that carbon was not limiting for primary productivity in the sympagic habitat. These differences in FA composition and δ13CFA values between i-POM and p-POM will aid efforts to track the proportional contribution of sea ice algal carbon to higher trophic levels in the Bering Sea and likely other Arctic seas.  相似文献

Background: Subsurface ice preserved as ice lenses and within rock glaciers as well as glacial and lake ice provides sensitive indicators of climate change and serve as a late-season source of meltwater.

Aims: We synthesise the results of geomorphological, geophysical and geochemical studies during the period of 1995–2014, building on a long history of earlier work focused on ice and permafrost studies on Niwot Ridge and the adjacent Green Lakes Valley (GLV), which is part of the Niwot Ridge Long Term Ecological Research Site.

Methods: These studies are discussed in the context of how bodies of ice and rock glaciers reflect changing local climate. We review recent results from geophysical investigations (resistivity, seismic refraction and ground-penetrating radar) of the shallow subsurface, ongoing monitoring of the Arikaree Glacier, three rock glaciers and lake ice in the GLV, and interpretations of how subsurface ice melt regulates the flow and chemistry of alpine surface water after seasonal snowfields melt.

Results and conclusions: Permafrost conditions reported from Niwot Ridge in the 1970s are generally absent today, but ice lenses form and melt seasonally. Ice is present permanently within the Green Lakes 5 rock glacier and at nearby favourable sites. The Arikaree Glacier has shown a marked decline in cumulative mass balance during the past 12 years after a 30-year period when net mass balance was ca. 0. Duration of seasonal lake ice increases with elevation in GLV, but duration has decreased at all seven lakes that have been monitored during the last three decades. This decrease has been most marked at the lowest elevation where it amounted to a reduction of about 1 d year?1 and least at Green Lake 5 where the loss has been at a rate of 0.5 d year?1. Surface temperature measurements from rock glaciers have not shown strong trends during the past 15 years. It has been suggested that almost all of the 2.5-mm year?1 increase in stream discharge from the upper GLV in September and October has been derived from melting of subsurface ice.  相似文献
