At least five of the biotypes described in the Bemisia tabaci (Gennadius) (Homoptera: Aleyrodidae) complex are known to be present in the Mediterranean Basin area. Only two of them, however, are economically relevant, that is, biotypes B and Q. Biological and genetic differences between the two biotypes have been well studied, but less is known about their patterns of genetic variation and population structure. To address these issues, a study was undertaken based on variation at six microsatellite loci among a subset of nine B. tabaci populations (five belonging to the Q and four to the B biotype). The data obtained show that (i) these loci showed considerable polymorphism in the Q and B biotypes populations although the presence of null alleles can obscure the picture; (ii) the Iberian‐Q, Canarian‐Q, and Egyptian‐B populations exhibit heterozygosity excess as a result of bottleneck events; (iii) the low genetic differentiation between the Israeli, Iberian Peninsula, and Italian populations suggest that these populations share a common gene pool; (iv) the genetic distances between the Canarian‐Q population and the geographically close population from Morocco indicates spatial isolation and a limited gene flow; and finally (v) the microsatellite data for the B populations indicate that the whiteflies from Egypt and Israel have a close phylogenetic relationship, but the source of these biotype B invasions into the Mediterranean area remains unknown.  相似文献

In recent years, the dominant biotype of Bemisia tabaci (Gennadius) has shifted from biotype B to Q in many locations of China. Our field monitoring showed that B. tabaci biotype Q could be found on pepper Capsicum frutescens L. while biotype B could not be found on the plant. To study the role of the host plant in the displacement of biotype B by Q, the adult mortality, female fecundity, and adult emergence of both biotypes B and Q on different host combinations were studied using a model system involving pepper and eggplant. The results showed that pepper is not a preferred host for either biotype B or Q adults in comparison with eggplant. The negative impact of pepper to biotype B is, however, much greater than that to biotype Q. Because both the survival rates of the adult whitefly and the average number of emerged adults per female of biotype Q on pepper are higher than that of biotype B, biotype Q showed higher fitness in comparison with biotype B. Our results suggest that the existence and differential impact of a nonpreferred host might affect the population fitness and therefore could play a role in the displacement of biotype B in some regions.  相似文献

烟粉虱Bemisia tabaci(Gennadius)是由多种生物型或物种组成的复合种。Q型烟粉虱在我国部分地区正在取代B型烟粉虱成为优势生物型。烟粉虱Q型与B型遗传多样性比较研究为解析这2种生物型入侵的遗传学基础具有重要的意义。本文利用AFLP技术研究了Q型(包括Q1型、Q2型)、B型烟粉虱种群的遗传多样性。结果表明:Q型烟粉虱遗传多样性高于B型烟粉虱;Q1型烟粉虱各项遗传多样性指数均接近于Q2型。最后探讨了AFLP与SSR分子标记的各自特点。  相似文献

在室内用棉花植株饲养情况下,对采自浙江杭州的烟粉虱Bemisia tabaci(Gennadius)B型和Q型混合种群的生物型组成以及抗药性水平进行了多代的监测。在室内饲养的F0代,两种生物型比例相当;在F10代,Q型已经完全被B型所取代。与F0代相比,F10代烟粉虱种群对吡虫啉、阿维菌素和氟虫腈的抗性水平明显下降,而对α-氯氰聚酯的抗性保持稳定。该研究表明:在没有杀虫剂选择压力的室内条件下,B型比Q型更具生存优势。然而在烟碱类杀虫剂广泛使用的田间条件下,烟粉虱Q型因比B型对烟碱类杀虫剂具有更强的抗性而获得竞争优势。  相似文献

本研究利用mtDNA COI基因片段作标记,通过序列分析对湖北省烟粉虱Bemisa tabaci(Gennadius)生物型进行了鉴定。结果表明当阳、仙桃、天门、监利、宜昌和武汉6个地理种群烟粉虱的720bp COI基因序列的同源性高达100%,且该6个地理种群烟粉虱与USA-Q型烟粉虱、湖北Q型烟粉虱的同源性也高达100%。枣阳地区烟粉虱与Israel-B型烟粉虱、我国浙江B型烟粉虱、我国湖北B型烟粉虱的同源性为100%。由此可见当阳、仙桃、天门、监利、宜昌和武汉6个地区烟粉虱生物型属于Q型,仅枣阳地区烟粉虱属于B型。  相似文献

利用烟粉虱Bemisia tabaci(Gennadius)内共生菌特异性引物,研究了内共生菌在B、Q型烟粉虱种群中的分布和感染率,同时评价了3种不同的抗生素利福平、氨苄青霉素和硫酸卡那霉素分别在3种不同的浓度下(100.0、50.0及25.0μg/mL)对烟粉虱内共生菌的去除效果。结果表明:B、Q型烟粉虱原生内共生细菌Portiera的带菌率均为100.0%;B、Q型烟粉虱次生内共生菌Hamiltonella的带菌率分别为91.7%和100.0%;B型烟粉虱次生内共生菌Rickettsia的带菌率为87.5%,Q型为0;其它次生内共生菌在B、Q型烟粉虱中均未检测到。利福平、氨苄青霉素和硫酸卡那霉素在3种不同的浓度下均不能去除B、Q型烟粉虱Portiera;利福平、氨苄青霉素在3种不同的浓度下均能完全去除B型烟粉虱Rickettsia,硫酸卡那霉素在不同浓度下去除Rickettsia的效果不同;3种抗生素去除Hamiltonella的能力受抗生素种类以及浓度的影响。同一抗生素在不同浓度下去除Hamiltonella的效果均是100.0μg/mL>50.0μg/mL>25.0μg/mL;不同浓度的抗生素去除Hamiltonella的效果均是利福平>氨苄青霉素>硫酸卡那霉素,各浓度与各抗生素之间的去除Hamiltonella的效果均具有显著性差异。  相似文献

Biotypes B and Q of Bemisia tabaci (Gennadius), collected from the islands of Tenerife and Majorca respectively, were exposed to competition conditions on tomato cv. Marmande in the laboratory. Both biotypes were established in single and mixed cultures at different densities. Increased mortality of biotype Q females and immature instars was observed together with a lower rate of fecundity and progeny size compared to biotype B, when reared in single or mixed cultures. The female:male sex ratio of F1 individuals of biotype Q was higher in single than in mixed cultures. However, the sex ratio of F1 individuals of biotype B was the same in single and mixed cultures, suggesting reproductive interference. Whitefly density did not affect interspecific interactions. It had a moderate effect on developmental rate of both biotypes, and on mortality of immature instars and progeny size of biotype B only. The results indicate that under laboratory conditions the studied biotype B should displace biotype Q.  相似文献

The invasive, insecticide-resistant, Q whitefly biotype, has gradually spread to other countries including the US via human-mediated movement of plant materials. We assessed the utility of the VspI -based mtCOI (mitochondrion cytochrome oxidase I) polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) technique as a rapid, cost-effective, and reliable alternative for differentiating the Q from the dominant B biotype in Arizona. Using the standard mtCOI gene sequencing and mtCOI PCR-RFLP techniques, we biotyped eight whitefly strains of five individuals each collected from poinsettia and cotton at different locations in Arizona. Complete concordance was observed between the two methods, with three strains being identified as the Q biotype and five samples as the B biotype. We also scanned the mtCOI gene sequences for VspI polymorphisms in the B and Q biotype whiteflies currently available in the GenBank database. This global screening revealed the existence of three and four VspI polymorphic types for the Q and B biotypes, respectively. Nevertheless, all three VspI polymorphic Q biotype whiteflies shared a common and unique VspI site that can be used to differentiate Q biotype from the four VspI polymorphic B biotype whiteflies identified. These results demonstrate that the VspI -based mtCOI gene PCR-RFLP provides a reliable diagnostic tool for differentiating the Q and B biotype whiteflies in the US and elsewhere.  相似文献

Horizontal transmission of begomoviruses between Bemisia tabaci biotypes   总被引:1,自引:0,他引:1  
We have previously shown that the monopartite Tomato yellow leaf curl virus (TYLCV), a begomovirus (family Geminiviridae, genus Begomovirus) infecting tomato plants can be transmitted in a gender-dependent manner among its insect vector the whitefly Bemisia tabaci type B (Gennaduis) (Aleyrodidae: Hemiptera) during mating. Viruliferous females were able to transmit the virus to non-viruliferous males and vice versa, in the absence of any other virus source. The recipient insects were able to infect tomato plants. In this communication, we present evidence that two bipartite begomoviruses infecting cucurbits, Squash leaf curl virus (SLCV) and Watermelon chlorotic stunt virus (WmCSV) can be transmitted in a gender-dependent manner among whiteflies. In addition we show that TYLCV can be transmitted during mating among individuals from the same biotype (from B-males to B-females and vice versa; and from Q-males to Q-females and vice versa). However, viruliferous males of the B biotype are unable to transmit the virus to females of the Q biotype (and vice versa); similarly, viruliferous males of the Q biotype are unable to transmit the virus to females of the B biotype (and vice versa). These findings support the hypothesis that a pre-zygotic mating barrier between the Q and B biotypes is the cause for the absence of gene flow between the two biotypes, and that virus transmission can be used as a marker for inter-biotype mating. To be transmitted during mating, the virus needs to be present in the haemolymph of the donor insect. Abutilon mosaic virus (AbMV), a bipartite begomovirus that can be ingested but not transmitted by B. tabaci, is absent in the whitefly haemolymph, and cannot be transmitted during mating. Mating was a precondition for horizontal virus transfer from male to female, or female to male. Virus was not transmitted when viruliferous B. tabaci were caged with the non-vector non-viruliferous whitefly Trialeurodes vaporariorum (Westwood) (Aleyrodidae: Hemiptera) and vice versa.  相似文献

烟粉虱B em isia tabaci(G ennad ius)是一种取食植物汁液的重要农业害虫,同时也是许多植物病毒最重要的传播介体之一。烟粉虱被认为是由许多具有明显遗传分化的不同种群即生物型组成的复合种,其中B型烟粉虱是一种入侵性最强的生物型,几乎在世界各地均有分布。由于不同生物型的烟粉虱在寄主范围、传毒能力、地理分布、抗药性等许多生物学方面存在差异,因此对烟粉虱生物型的鉴定及其遗传分化研究对于该害虫的可持续控制具有重要的指导意义。烟粉虱生物型的鉴定方法包括生物学鉴定、酶谱鉴定以及分子标记鉴定的方法,其中使用的分子标记包括RAPD、AFLP、rDNA-ITS1、m tDNA CO I以及SSR等标记。目前,不同生物型的烟粉虱尤其B型烟粉虱的分类地位仍然存在争议。介绍了不同生物型烟粉虱的生物学差异、鉴定方法及其遗传分化研究的最新进展,同时探讨了不同生物型烟粉虱的分类地位和研究方向。  相似文献

The two most widespread biotypes of Bemisia tabaci (Gennadius) in southern Europe and the Middle East are referred to as the B and Q-type, which are morphologically indistinguishable. In this study various DNA markers have been developed, applied and compared for studying genetic diversity and distribution of the two biotypes. For developing sequence characterized amplified regions (SCAR) and cleaved amplified polymorphic sequences (CAPS) techniques, single random amplified polymorphic DNA (RAPD) fragments of B and Q biotypes, respectively, were used. The CAPS were investigated on the basis of nuclear sodium channel and the mitochondrial cytochrome oxidase I genes (mtCOI) sequences. In general, complete agreement was found between the different markers used. Analysis of field samples collected in Israel for several years, using these markers, indicated that the percentage of the Q biotype tends to increase in field populations as time progresses. This may be attributed to the resistance of the Q biotype to neonicotinoids and pyriproxyfen and the susceptibility of the B biotype to these insecticides.  相似文献

The B and Q 'biotypes' of Bemisia tabaci (Gennadius) (Hemiptera: Aleyrodidae) have been invading many parts of the world and causing severe damage to a range of crops. Recent phylogenetic analyses indicate that B and Q are cryptic species within the B. tabaci species complex. Although various attempts have been made to examine the reproductive compatibility between B and Q, few studies have tested the fertility of the F1 females and so the extent of possible gene flow remains unclear. In this study, we conducted a series of crossing experiments and behavioural observations to examine in detail the reproductive compatibility between the B and Q biotypes collected from Zhejiang, China, a region recently invaded by these whiteflies. Crossing experiments between the two biotypes using either single-pairs or small groups demonstrated that proportions of females in the F1 progeny were only 0-2% in the inter-biotype crosses compared to 58-68% in the intra-biotype treatments. Furthermore, all inter-biotype F1 females were sterile. Continuous video observations showed that B and Q adults very rarely copulated, and copulation occurred only when adults of opposite sex from different biotypes were enclosed in dense cohorts for a relatively long period of time. These data show that the B and Q biotypes examined in this study are completely isolated in reproduction. The isolation was due to mainly a copulation barrier, but post-copulation barriers were also involved.  相似文献

Abstract Bemisia tabaci shifted unexpectedly in China from a predominance of B biotype to Q biotype during 2005–2008. This observation stimulated an interest in investigating whether environmental factors, including host, temperature and relative humidity (RH) could possibly explain the observed shift in biotypes distribution. Results indicated that all three parameters examined influenced biotype survivability. The percentage of B biotype, when reared together on pepper plants with the Q biotype, decreased significantly from 66.7% in the founder population, to 13.6% and 3.7% in the first and second generations, respectively. When the B (founder at 66.7%) and Q (founder at 33.3%) biotypes were reared together on eggplant alone, or on pepper‐plus‐eggplant combination, the population size of the B biotype either remained constant, or increased somewhat in the first and second generations. On eggplant, the effects of RH and temperature on the competitiveness between the Q and B biotypes (3 pairs of Q and 6 pairs of B) were not significant.  相似文献

Tomato yellow leaf curl virus (TYLCV) and tomato yellow leaf curl Sardinia virus (TYLCSV) (genus Begomovirus, family Geminiviridae) as well as their whitefly vector Bemisia tabaci were reported from the south‐west and central regions of Morocco. To establish a more comprehensive view of tomato begomoviruses and B. tabaci biotypes throughout Morocco, 32 tomato fields were surveyed for tomato yellow leaf curl disease (TYLCD) symptoms in southern and northern regions, and 54 samples of leaves from cultivated plants or weeds and 35 B. tabaci individuals were collected and analysed by PCR, randomly amplified polymorphic DNA and sequencing. Only TYLCV or TYLCSV were detected. TYLCV was detected in 15 plant samples whereas TYLCSV only in 4. Sequence analyses revealed the presence of the ‘Spanish’ strain of TYLCSV and distinguished two genetically distinct strains of TYLCV. The begomovirus infections were unevenly distributed throughout Morocco. In the north‐west and north‐central regions where tomato plants exhibiting TYLCD symptoms were rarely observed, only 1 sample out of 13 tested positive for the presence of a begomovirus. In the north‐east region, the ratio of begomovirus‐positive samples was higher, 6/13, and in the south‐west region, it was the highest, 13/14. Consistently the frequency of plants exhibiting TYLCD‐like symptoms in the northern regions was lower than that in the south‐west region. B. tabaci biotype Q is present throughout the country and in Algeria, whereas biotype B, identified for the first time in Morocco, was detected only in the north‐east region.  相似文献

烟粉虱Bemisia tabaci (Gennadius) B型与Q型是烟粉虱复合种中入侵性较强、分布较广的2种生物型, 当前在许多地区混合发生。这2种生物型的快速鉴别对其种群动态调查及入侵生态学研究具有重要价值。为了验证微卫星位点BEM06与BEM23鉴别B型与Q型烟粉虱的有效性, 本研究分析了这2个微卫星位点的等位基因在国内外17个B型、4个Q型、3个非B/Q型烟粉虱种群的分布特点。结果表明: 这2个微卫星位点的联合使用可鉴别B型与Q型烟粉虱, 但是无法有效地将B型、Q型与一些非B/Q型烟粉虱某些个体区分开来。结果提示: 利用微卫星位点BEM06与BEM23鉴别B型与Q型烟粉虱具有一定的局限性, 尤其是田间烟粉虱存在其他生物型时需要慎重使用这种鉴别方法。  相似文献

Differential invasion success among biotypes: case of Bemisia tabaci   总被引:2,自引:0,他引:2  
Studies on success or failures of biological invasions by different insect biotypes are scarce and could provide interesting insights into the traits that determine greater or lower ability to invade. Life history traits of invasive whiteflies Bemisia tabaci of the B biotype (known as a worldwide invasive biotype) and of the indigenous biotype Ms (not known as an invader anywhere in the world), both from the island of La Réunion (Indian Ocean), were compared for this purpose. In our study we demonstrated that within a cultivated host plant (tomato), the B biotype differs from the Ms by a combination of several life-history traits. This combination gives the invasive biotype an advantage over the resident both in terms of rapid demographic growth (increased intrinsic rate of increase and associated traits such as short developmental times and high fecundity) and in terms of competition (large adult and offspring sizes), without any recorded trade off. However, in the field the resident biotype remains dominant on non-cultivated hosts (weeds) and in a particular climate (high humidity). This suggests that invasive biotypes are characterized by physiological, morphological and biological adaptations to a disturbed environment created by anthropic activities at different places in the world, while resident biotypes may persist in less altered habitats.  相似文献

After the 2004 discovery of the Bemisia tabaci (Gennadius) (Hemiptera Aleyrodidae) Q biotype in the United States, there was a vital need to determine the geographical and host distribution as well as its interaction with the resident B biotype because of its innate ability to rapidly develop high-level insecticide resistance that persists in the absence of exposure. As part of a coordinated country-wide effort, an extensive survey of B. tabaci biotypes was conducted in North America, with the cooperation of growers, industry, local, state, and federal agencies, to monitor the introduction and distribution of the Q biotype. The biotype status of submitted B. tabaci samples was determined either by polymerase chain reaction amplification and sequencing of a mitochondrial cytochrome oxidase I small subunit gene fragment and characterization of two biotype discriminating nuclear microsatellite markers or esterase zymogram analysis. Two hundred and eighty collections were sampled from the United States, Bermuda, Canada, and Mexico during January 2005 through December 2011. Host plants were split between ornamental plant and culinary herb (67%) and vegetable and field crop (33%) commodities. The New World biotype was detected on field-grown tomatoes (Solanum lycopersicum L.) in Mexico (two) and in commercial greenhouses in Texas (three) and represented 100% of these five collections. To our knowledge, the latter identification represents the first report of the New World biotype in the United States since its rapid displacement in the late 1980s after the introduction of biotype B. Seventy-one percent of all collections contained at least one biotype B individual, and 53% of all collections contained only biotype B whiteflies. Biotype Q was detected in 23 states in the United States, Canada (British Columbia and Ontario territories), Bermuda, and Mexico. Forty-five percent of all collections were found to contain biotype Q in samples from ornamentals, herbs and a single collection from tomato transplants located in protected commercial horticultural greenhouses, but there were no Q detections in outdoor agriculture (vegetable or field crops). Ten of the 15 collections (67%) from Canada and a single collection from Bermuda contained biotype Q, representing the first reports of biotype Q for both countries. Three distinct mitochondrial haplotypes of B. tabaci biotype Q whiteflies were detected in North America Our data are consistent with the inference of independent invasions from at least three different locations. Of the 4,641 individuals analyzed from 517 collections that include data from our previous work, only 16 individuals contained genetic or zymogram evidence of possible hybridization of the Q and B biotypes, and there was no evidence that rare hybrid B-Q marker co-occurrences persisted in any populations.  相似文献

Wolbachia是广泛分布于节肢动物体内一类共生细菌,它能够通过多种机制调控宿主的生殖方式。近年来的研究表明,Wolbachia与许多外来生物的成功入侵相关。本文利用长PCR的方法特异扩增了不同生物型烟粉虱(共24个种群)体内Wolbachiawsp基因,结果发现B型和Q型烟粉虱入侵种群体内均未检测到Wolbachia,而在非B/Q型的浙江种群和肯尼亚种群体内检测到了Wolbachia。对该wsp基因进行测序并和已知序列进行同源性分析发现,浙江烟粉虱种群的Wolbachia属于B组Con/Rug亚种群,而肯尼亚种群属于B组Btab1亚种群。Wolbachia的存在与否可能与烟粉虱的成功入侵有一定的关系。图2表2参23  相似文献

【目的】为了揭示山东省农区烟粉虱 Bemisia tabaci 隐种的分布情况,对2013年7-8月份期间在该省不同农区采集的烟粉虱隐种组成进行了系统调查研究。【方法】在山东省15市22个地点作物上采集了69份烟粉虱样品,利用mtCOI基因PCR-RFLP方法进行了分子鉴定,并分析了烟粉虱Q隐种在不同寄主作物及地理区域内的分布状况。【结果】22个采集地点中,17个地点的采集样品全部为烟粉虱Q隐种;其他5个地点(潍坊、菏泽、泰安、淄博和临沂)的烟粉虱Q隐种所占比例均大于90%,B隐种比例较低(<10%)。烟粉虱Q隐种比例在5种寄主(茄子、辣椒、番茄、黄瓜和棉花)种群间以及在山东省东部丘陵、中部山地和西部平原地区间均没有显著差异。【结论】2013年田间调查发现烟粉虱Q隐种在山东各地已广泛取代B隐种成为该地区优势种群。  相似文献
