The application of dye–ligand expanded bed chromatography adsorption (EBA) of glucose-6-phosphate dehydrogenase (G6PDH) from unclarified yeast extract was undertaken by using a commercially available expanded bed column (20 mm i.d.) and UpFront adsorbent (ρ = 1.5 g/mL) from UpFront Chromatography. The influence of biomass concentration on the adsorption capacity was explored by employing yeast extracts containing various biomass concentrations (5–30%, w/v). It was demonstrated that the biomass concentration had little effect on G6PDH adsorption performance. Feedstock containing 15% (w/v) biomass gave a relatively high recovery yield (>90%) of G6PDH compared to feedstock containing 30% (w/v) biomass, which gave a recovery of 75% G6PDH. Nevertheless, the enzyme specific activity of 7 U mg−1 with a purification factor of 6 was achieved in the feedstock containing biomass concentration of 30% (w/v). The generic applicability of dye–ligand as an affinity tool in expanded bed chromatography is discussed.  相似文献

The removal of Zn(II) ions from aqueous solution using pure and chemically pretreated biomass of Moringa oleifera was investigated at 30 ± 1 °C in this study. The experimental results explored that the maximum pH (pHmax) for efficient sorption of Zn(II) was 7 ± 0.1 at which evaluated biosorbent dosage and biosorbent particle size, were 0.5 g/L, <0.255 mm, respectively. The cellular Zn(II) concentration increased with the concentrations of Zn(II) in solution. Pretreatment of M. oleifera biomass affected the sorption process and the uptake capacity (mg/g) of biomass for Zn(II) uptake was in following order: NaOH (45.76) > H2SO4 (45.00) > CTAB (42.80) > Ca(OH)2 (42.60) > Triton X-100 (42.06) > H3PO4 (41.22) > Al(OH)3 (41.06) > SDS (40.41) > HCl (37.00) > non-treated biomass (36.07). There was significant increase in uptake capacity of M. oleifera biomass, which suggested that affinity between metal and sorbent can be increased after some sort of pretreatment. Both Langmuir and Freundlich isotherm model fitted well to data of Zn(II) biosorption as represented by high value of their correlation coefficient (i.e. R2  1). Kinetic studies revealed that Zn(II) uptake was fast with 90% or more of uptake occurring with in 40 min of contact time and the equilibrium was reached in 50 min of contact time. The sorption rates were better described by a second order expression than by a more commonly applied Lagergren equation. Finally it was concluded that pretreatment of M. oleifera biomass can achieve superior Zn(II) uptake capacity in comparison to non-pretreated biomass.  相似文献

Biomass of Phragmites australis growing in four constructed wetlands with horizontal sub-surface flow (HF CWs) designed for treatment of municipal sewage in the Czech Republic have been analyzed for 19 trace elements. The biomass was harvested during the peak standing crop in early September and divided into stems, leaves, flowers, roots and rhizomes. Concentrations of monitored elements in both aboveground and belowground plant tissues were similar to those found in plants growing in natural stands. The highest concentrations were recorded for Al, Fe, Mn, Ba and Zn while the lowest concentrations were those of Hg, U and Cd. Concentrations decreased in the order of roots > rhizomes > leaves > stems. The root/leaf ratio averaged 70 and varied between 1.4 for molybdenum and 392 for cobalt. The belowground/aboveground concentration ratio ranged between 0.9 and 69.5 with an average value of 19. Due to average aboveground/belowground biomass ratio > 1, the belowground/aboveground standing stock ratios were lower with six elements (Ba, Zn, Se, Hg, Mo, and Mn) having this ratio < 1.  相似文献

Plant growth, biomass allocation, root distribution and plant nutrient content were investigated in the submerged macrophyte Potamogeton crispus growing in heterogeneous sediments. Three experimental sediments heterogeneous in nutrient content and phosphorus release capacity were used: sandy loam with low nutrient content (A), clay with intermediate nutrient content (B), and clay with high nutrient content (C). Biomass accumulation was significantly affected by the sediment type, and was highest in clay C (1.23 mg per plant dry weight) but lowest in sandy loam (0.69 mg per plant dry weight). The root:shoot ratios in treatments A, B and C were 0.30, 0.14 and 0.09, respectively. P. crispus allocated more biomass to roots in sandy loam compared with the other sediments. The average root numbers in sediments A, B and C were 16, 19 and 20, respectively, and the total root lengths in sediments A, B and C were 238.84, 200.36 and 187.21 cm, respectively. Almost 90% of the root biomass was distributed in the 0–15 cm depth in sediments B and C, compared with 64.53% in sediment A. The rank order of plant nitrogen and phosphorus concentrations in the sediment types was C > B > A. These results indicate that both sediment structure and nutrient availability influence the growth and distribution of the root system of P. crispus.  相似文献

In this research, micro and nanoparticles of Spirulina platensis dead biomass were obtained, characterized and employed to removal FD&C red no. 40 and acid blue 9 synthetic dyes from aqueous solutions. The effects of particle size (micro and nano) and biosorbent dosage (from 50 to 750 mg) were studied. Pseudo-first order, pseudo-second order and Elovich models were used to evaluate the biosorption kinetics. The biosorption nature was verified using energy dispersive X-ray spectroscopy (EDS). The best results for both dyes were found using 250 mg of nanoparticles, in these conditions, the biosorption capacities were 295 mg g?1 and 1450 mg g?1, and the percentages of dye removal were 15.0 and 72.5% for the FD&C red no. 40 and acid blue 9, respectively. Pseudo-first order model was the more adequate to represent the biosorption of both dyes onto microparticles, and Elovich model was more appropriate to the biosorption onto nanoparticles. The EDS results suggested that the dyes biosorption onto microparticles occurred mainly by physical interactions, and for the nanoparticles, chemisorption was dominant.  相似文献

The adsorption of papain on Reactive Blue 4 dye–ligand affinity membrane was investigated in a batch system. The combined effects of operating parameters such as initial pH, temperature, and initial papain concentration on the adsorption were analyzed using response surface methodology. The optimum adsorption conditions were determined as initial pH 7.05, temperature 39 °C, and initial papain concentration 11.0 mg/ml. At optimum conditions, the adsorption capacity of dye–ligand affinity membrane for papain was found to be 27.85 mg/g after 120 min adsorption. The papain was purified 34.6-fold in a single step determined by fast protein liquid chromatography. More than 85% of the adsorbed papain was desorbed using 1.0 M NaCl at pH 9.0 as the elution agent. The purification process showed that the dye–ligand immobilized composite membrane gave good separation of papain from aqueous solution.  相似文献

Conditions influencing bioluminescence output from Pseudomonas putida TVA8 harboring chromosomal tod-luxCDABE fusion were followed. In complex media, cell growth was not influenced by the presence of toluene at 53 mg/L. Bioluminescence induction was tested in minimal medium. At 15 °C the highest bioluminescence induced with toluene (1.325 mg/L) was reached after 6 h. At 25 °C the bioluminescence maximum was approximately 20% lower but this was reached after 3.5 h, and at temperatures of 7 °C, 28 °C, 30 °C and 34 °C, bioluminescence peaked at ≤60% of the maximum. Time courses of bioluminescence were dependent on cell concentrations. The heights of bioluminescence maxima were proportional to toluene concentration in the range 0–26 mg/L. Twenty-three organic pollutants (103× diluted saturated solutions) were tested as bioluminescent inducers. The bioluminescence maximum decreased in the order: ethylbenzene > toluene > phenol > benzene > 4-ethyltoluene > 4-fluorotoluene > cumene > isobutylbenzene > styrene > trichloroethylene > o-, p-xylene > cresol > m-xylene > 2-methylnaphthalene > benzylchloride > naphthalene > salicylic acid > hexachlorobenzene > 2-chloronaphthalene > biphenyl > 2-bromonaphthalene > 1,3,5-triethylbenzene. Bioluminescence was also induced with ethanol and methanol and the presence of these alcohols in concentrations of ≤1% increased bioluminescence of toluene. The induction of bioluminescence from samples of wastewater and groundwater contaminated with BTEX (benzene, toluene, ethylbenzene and xylene) from 0.5 to 120 mg/L was demonstrated.  相似文献

Effects of pellet morphology on broth rheology are reported for pelleted submerged cultures of the lovastatin producing filamentous fungus Aspergillus terreus, growing in fluidized bed and stirred tank bioreactors. The pellet diameter and compactness were affected by the agitation intensity of the broth; however, the total biomass productivity was not affected. In fluidized beds and stirred tanks with agitation intensity of up to 300 rpm (impeller tip speed of 1.02 m s−1), the fungal pellets were stable at diameters of up to about 2300 μm. In more intensely agitated stirred tanks (≥600 rpm; impeller tip speed of ≥2.03 m s−1), the stable pellet size was only about ≤900 μm. The biomass concentration and the pellet diameter were the main factors that influenced the flow index and the consistency index of the power-law broths. Because the biomass productivity was the same in all experiments in a given type of reactor and the oxygen concentration was kept at ∼400% of air saturation, the pellet size and morphology were not influenced by oxygen mass transfer effects. Pellets were always dense in the core region and no necrosis of the biomass occurred.  相似文献

Biomonitoring of mercury pollution of Dubasari reservoir of the Dniester River has been carried out using macrophytes, epiphytosuspensions and lichens. Macrophytes uptake mercury from water column and bottom sediments in the succession Potamogeton pectinatus L. > Myriophyllum verticillatum L. > Potamogeton perfoliatus L. > Butomus umbellatus L. These species possess the properties of bio-indicators. The mercury content in their biomass is in good positive correlation (r = 0.908–0.990) with the mercury content both in water and bottom sediments. It has been shown that the maximum mercury content in these bio-indicators is found in the area of the Rezina–Rybnitsa industrial complex that consists of two cement plants and a metallurgical one. The use of lichens as bio-indicators for the ecological state of the air in the vicinity of the investigated aquatic ecosystem and of the epiphytosuspensions precipitated on submerged macrophytes, denote the active state of the source for mercury pollution of the Dubasari reservoir.  相似文献

The cel5C gene, coding for an endoglucanase (Cel5C) of Penicillium brasilianum was cloned and heterologously expressed in Aspergillus oryzae. This is only the second GH5 EG from the genus Penicillium reported in the CAZy database. The promoter region of the gene has putative binding sites for both the carbon catabolite repressor CreA and the activator XlnR. The pH optimum of Cel5C was found to be 4.0 and the temperature optimum was 70 °C. At a typical temperature for lignocellulose hydrolysis Cel5C retained full residual activity after 20 h of incubation at pH 5.0 and 6.0. Adsorption to Avicel and steam pretreated spruce, was found to follow the Langmuir isotherm, and the maximum adsorption was similar for both substrates, 40 and 49 mg/g, respectively. The affinity for Avicel was 10 times higher than for steam pretreated spruce, 0.040 and 0.0035 L/mg, respectively. Non-productive binding of cellulolytic enzymes to lignin is an important obstacle to overcome for commercial biomass to ethanol production. Therefore, the adsorption on residual lignin produced from various biomass samples was investigated. Both substrate and pretreatment conditions resulted in different adsorptions of Cel5C to the residual lignin.  相似文献

The effect of addition of autoclaved and filter-sterilized culture filtrate of Piriformospora indica (a root endophytic fungus) to the growing Linum album hairy root cultures on growth and lignan production was investigated. The addition resulted in a significant enhancement in lignan production and growth. The podophyllotoxin and 6-methoxypodophyllotoxin (the lignans) concentrations were maximally improved by 3.8 times (233.8 mg/L) and 4.4 times (131.9 mg/L) in comparison to control cultures, respectively, upon addition of 3.0% (v/v) filter-sterilized culture filtrate of P. indica to the hairy root cultures of L. album for exposure time of 48 h. This increase in the lignan content also coincided with the increase in phenylalanine ammonia lyase activity, which was 3.1-fold (371.4 μkat/kg protein) higher compared to control cultures under the same conditions. The maximal increase in hairy root biomass was, however, obtained under different conditions; it was enhanced by 1.4 times (21.8 g/L) in comparison to control cultures, when 2% (v/v) filter-sterilized culture filtrate was in contact with L. album cultures for 96 h.  相似文献

The hydroquinone glucoside arbutin is a plant derived compound medically applied due to its uroantiseptic activity. It also has skin whitening properties and thus is widely used in dermatology and cosmetology. Origanum majorana L. (Lamiaceae) is known to produce arbutin, however the content of the compound in cultivated plants is very variable and low. Since plant cell and tissue cultures are capable to perform specific biotransformation reactions including glucosylation, this investigation targeted the formation of arbutin from hydroquinone in agitated O. majorana shoot cultures. For this purpose different doses of hydroquinone (96, 144, 192, 288 and 384 mg/L of medium) were added to the culture flasks in one, two or three portions. Arbutin was qualitatively and quantitatively determined in methanol extracts from dry biomass and lyophilized media using HPLC-DAD. Cells of O. majorana shoot cultures efficiently converted hydroquinone into arbutin. The product was accumulated in the biomass and was not observed (or in trace amounts) in the medium samples. Different doses as well as portioning of the precursor had a significant impact on the biotransformation process. Arbutin accumulation increased from 0.23 ± 0.03 mg/g DW up to 52.6 ± 4.8 mg/g DW in the biomass. The highest product content was observed after the addition of 192 mg/L hydroquinone in three portions. The highest efficiency of the biotransformation process, i.e. 67.5 ± 5.2% was calculated for a dose of 96 mg/L precursor divided into three portions. After further optimization of the biotransformation process, O. majorana shoot cultures could serve as a rich source of arbutin.  相似文献

In a shallow multifunction dam reservoir, perennial water blooms formed by several toxin-producing cyanobacteria (Anabaena spp., Aphanizomenon spp., Planktothrix agardhii and Microcystis spp.) were observed. Over a seven-year period, concomitantly with a gradual decrease in phosphate and total phosphorus concentrations in the water and an increase in the DIN to DIP ratio, a reduced biomass of cyanobacteria was noted. Simultaneously, a twofold increase in cyanobacterial species richness was found. The concentration of intracellular anatoxin-a was positively correlated with the total cyanobacterial biomass, but the concentration of intracellular microcystins was significantly negatively correlated with the level of phosphorus in the water. Therefore, in a period with a very low (2.3–3.6) DIN:DIP ratio, intracellular ANTX prevailed in the reservoir, while in the following years (at DIN:DIP = 23–36) much higher MC levels were noted. The highest total concentrations (22.2 μg L−1) of intracellular MCs (MC-LF > -LY > -LR > -LA = -LW) and ANTX (14.4 μg L−1) were found in 2010. In the following year, eight MC iso-forms were detected (MC-LF > -LY > -LA > -LR > -LW > -WR > -YR > -RR). The number of MC variants was positively correlated with the increased contribution of Anabaena planctonica/A. affinis and Microcystis spp. to cyanobacteria biomass. The indigenous bentho-pelagic fish Abramis brama L. accumulated in their tissues relatively high amounts of both ANTX (e.g. 6.2–18.4 μg g−1 FW of liver) and different variants of MCs (up to 4.4 μg g−1 FW of liver). Cyanotoxin tissue contents decreased in the following order: gills > liver > muscles. These observed strong changes in the species structure of cyanobacteria assemblages, even at their considerably smaller biomass, appeared to be an undesirable phenomenon due to the predominance of the efficient MC and ANTX producers, such as Anabaena spp., which is easily digested by fish. The variability of the profile of cyanobacterial blooms that depends on nutrient fluctuations and may account for the diverse toxin accumulation and tissue distribution in freshwater ichthyofauna is noteworthy, especially in water bodies used for fishery.  相似文献

The objective of this study is to assess the environmentaly friendly Ni(II) adsorption from synthetic wastewater using waste pomace of olive oil factory (WPOOF). Batch kinetic studies were performed in order to investigate the adsorbent and adsorbate dose, solution pH, agitating speed and temperature. The maximum Ni(II) adsorption was obtained at pH 4.0. The equilibrium nature of Ni(II) adsorption at different temperature was described by the Freundlich, Langmuir and Temkin isotherms. The equilibrium data fit well the Temkin and Langmuir isotherm. The monolayer adsorption capacities of WPOOF as obtained from Langmuir isotherm at 60 °C was found to be 14.80 mg/g. The adsorption mechanism was examined by the FTIR technique. The results of the thermodynamic investigations indicated that the adsorption reactions were spontaneous (ΔG < 0), slightly endothermic (ΔH > 0) and irreversible (ΔS > 0). The pseudo first-order, pseudo second-order, Elovich and intraparticle diffusion kinetic models were used to describe the kinetic data.  相似文献

Hyphal growth and biocontrol efficacy of Trichoderma harzianum may depend on its interactions with biotic components of the soil environment. Effects of soil microbial biomass on growth and biocontrol efficacy of the green fluorescent protein transformant T. harzianum ThzID1-M3 were investigated using different levels of soil microbial biomass (153, 328, or 517 μg biomass carbon/g of dry soil). Hyphal growth of T. harzianum was significantly inhibited in soil containing 328 or 517 μg biomass carbon/g of dry soil compared with soil containing 153 μg biomass carbon/g. However, when ThzID1-M3 was added to soil as an alginate pellet formulation, recoverable populations of ThzID1-M3 varied, with the highest populations in soil containing 517 μg biomass carbon/g. When sclerotia of Sclerotinia sclerotiorum were added to soils (10 sclerotia per 150 g soil) with ThzID1-M3 (20 pellets per 150 g soil), colonization of sclerotia by ThzID1-M3 was significantly lower in the soil containing the highest level of biomass. Addition of alginate pellets of ThzID1-M3 to soils (10 pellets per 50 g) resulted in increased indigenous microbial populations (total fungi, bacteria, fluorescent Pseudomonas spp., and actinomycetes). Our results suggest that higher levels of microbial soil biomass result in increased interactions between introduced T. harzianum and soil microorganisms, and further that microbial competition in soil favors a shift from hyphal growth to sporulation in T. harzianum, potentially reducing its biocontrol efficacy.  相似文献

Use of suitable plants that can extract and concentrate excess P from contaminated soil serves as an attractive method of phytoremediation. Plants vary in their potential to assimilate different organic and inorganic P-substrates. In this study, the response of Duo grass (Duo festulolium) to variable rates of soil-applied potassium dihydrogen phosphate (KH2PO4) on biomass yield and P uptake were studied. Duo grown for 5 weeks in soil with 2.5, 5 and 7.5 g KH2PO4 kg?1 soil showed a significantly higher biomass and shoot P content of 8.3, 11.4 and 12.3 g P kg?1 dry weight respectively compared to plants that received no soil added P. Also, the ability of Duo to metabolize different forms of P-substrates was determined by growing them in sterile Hoagland's agar media with different organic and inorganic P-substrates, viz. KH2PO4, glucose-1-phosphate (G1P), inositiol hexaphosphate (IHP), adenosine triphosphate (ATP) and adenosine monophosphate (AMP) for 2 weeks. Plants on agar media with different P-substrates also showed enhanced biomass yield and shoot P relative to no P control and the P uptake was in the order of ATP > KH2PO4 > G1P > IHP = AMP > no P control. The activities of both phytase (E.C.3.1.3.26) and acid phosphatases (E.C.3.1.3.2) were higher in all the P received plants than the control. Duo grass is capable of extracting P from the soil and also from the agar media and thus it can serve as possible candidate for phytoextraction of high P-soil.  相似文献

The removal of Remazol Blue and Reactive Black B by the immobilized thermophilic cyanobacterial strain Phormidium sp. was investigated under thermophilic conditions in a batch system, in order to determine the optimal conditions required for the highest dye removal. In the experiments, performed at pH 8.5, with different initial dye concentrations between 9.1 mg l−1 and 82.1 mg l−1 and at 45 °C, calcium alginate immobilized Phormidium sp. showed high dye decolorization, with maximum uptake yields ranging from 50% to 88% at all dye concentrations tested. When the effects of high dye concentrations on dye removal were investigated, the highest uptake yield in the beads was 50.3% for 82.1 mg l−1 Remazol Blue and 60.0% for 79.5 mg l−1 Reactive Black B. The highest color removal was detected at 45 °C and 50 °C incubation temperatures for all dye concentrations. As the temperature decreased, the removal yield of immobilized Phormidium sp. also decreased. At about 75 mg l−1 initial dye concentrations, the highest specific dye uptake measured was 41.29–41.17 mg g−1 for Remazol Blue and 47.69–43.82 mg g−1 for Reactive Black B at 45 °C and 50 °C incubation temperatures, respectively, after 8 days incubation.  相似文献

The present study aimed to evaluate the morphological characteristics and biomass of Desmostachya bipinnata and their relation to the environmental variables in three main habitats (canal banks, railway and roadside shoulders) where it is found in Egypt. In addition, the ability of this plant is evaluated to accumulate nutrients and heavy metals in its aboveground shoots. Twenty five quadrats (1 m × 1 m per quadrat) were selected along five sites representing the different habitats of D. bipinnata for this study. The aboveground shoots displayed considerable morphological variations, differing in the different habitats. The above ground biomass, number of spikes and leaves, rachis length and diameter, leaf length, width and area, leaf sheath length, and spike length and diameter were in the order: road sides > canal banks > railway shoulders. A regression equation: biomass = (67.37 × density) + 108.2, was used to estimate the shoot biomass from the plant growth density. Metal uptake capability from soil to grass is in the order Fe > Zn > Cu > Mn, and all of them are in safe concentration ranges. These heavy metals had a transfer factor more than unity, which indicates that D. bipinnata is a powerful accumulator for heavy metals. In addition, the plant shoots exhibit high accumulation of inorganic and organic nutrients.  相似文献

In this paper, the authors made an attempt to measure appropriation of ecosystems more reasonably and effectively based on examining the advantages and disadvantages of emergy analysis and ecological footprint. By combining emergy analysis with ecological footprint (EF) in terms of spatial equivalent of energy productivity, a new method of solar space (SS) and its calculating framework was proposed, and some necessary parameters for this method of analysis were calculated. A case study was followed under this calculating framework for Yangzhou city, Jiangsu province, Eastern China, using the data of the year 2000. The main results are as follows: (1) The ecological space in Yangzhou city includes earth surface space and solar space, and their demands were bigger than their supply. In the year of 2000, the earth surface space had an absolute deficit of 2.3063E?02 Ga-ha per capita (global hectares), and the solar space had an absolute deficit of 1.4944E?04 Sa-ha per capita (solar hectares). (2) There was a relative ecological deficit of 3.68% in earth surface space, and that of 154.86% in solar space. (3) Only freshwater areas and built-up areas were in a state of surplus, and there was a relative ecological surplus of 82.66% for freshwater areas, being greater than that for built-up areas. (4) The ranking of absolute ecological deficit in a descending order was as follows: solar space > arable land > pasture > forest > marine area > garden land, while the relative ecological deficit was the following: solar space > pasture land > forest land > arable land > garden land, except marine areas.A qualitative comparison of methodology was made between solar space method and ecological footprint according to their capability to describe the temporal-spatial characteristics of ecosystems appropriation. Moreover, a quantitative comparison was also conducted to show their differences in applicability to the assessment of ecosystems appropriation. It was shown that the calculating results from solar space method were bigger than that from ecological footprint because the former supplemented additional items of appropriation omitted by the later and included some new items of indirect appropriation that were excluded by the later. It is found that the new measure of solar space operates well as an indicator of the city's appropriation to ecosystems.  相似文献
