Bacterial β-ketoacyl-ACP reductase (FabG) and the β-ketoacyl reductase domain in mammalian fatty acid synthase (FAS) have the same function and both are rendered as the novel targets for drugs. Herein we developed a convenient method, using an available compound ethyl acetoacetate (EAA) as the substitutive substrate, to measure their activities by monitoring decrease of NADPH absorbance at 340 nm. In addition to the result, ethyl 3-hydroxybutyrate (EHB) was detected by HPLC analysis in the reaction system, indicating that EAA worked effectively as the substrate of FabG and FAS since its β-keto group was reduced. Then, the detailed kinetic characteristics, such as optimal ionic strength, pH value and temperature, and kinetic parameters, for FabG and FAS with this substitutive substrate were determined. The Km and kcat values of FabG obtained for EAA were 127 mM and 0.30 s− 1, while those of this enzyme for NADPH were 10.0 μM and 0.59 s− 1, respectively. The corresponding Km and kcat values of FAS were 126 mM and 4.63 s− 1 for EAA; 8.7 μM and 4.09 s− 1 for NADPH. Additionally, the inhibitory kinetics of FabG and FAS, by a known inhibitor EGCG, was also studied.  相似文献

The present report describes the synthesis and anticancer evaluation of certain 11-substituted 6H-indolo[2,3-b]quinolines and their methylated derivatives. These 6H-indolo[2,3-b]quinoline derivatives 11–13 were prepared from the commercially available 1,4-dihydroxyquinoline through alkylation, chlorination, nucleophilic reaction, and ring cyclization. Depending on the ratio of 11, (MeO)2SO2, and K2CO3, alkylation occurred primarily on N-5 (1:0.8:0.8) or N-6 (1:1.5:1.5) leading to the isolation of 14a or 14b as a major product. Accordingly, major product 15a (2/(MeO)2SO2/K2CO3 = 1:2:2) or 15b (1:1:1), respectively, was obtained by alkylation of 12 while 16a (13/(MeO)2SO2/K2CO3 = 1:2:2) or 16b (1:1:1), respectively, was obtained by alkylation of 13. The in vitro anticancer assay indicated 5-methylated derivatives 14a, 15a, 16a are more cytotoxic than their respective 6-methylated counterparts 14b, 15b, 16b and 6H-indolo[2,3-b]quinoline precursors 11, 12, 13. Among them, 11-(4-methoxyanilino)-6-methyl-6H-indolo[2,3-b]quinoline (16a) was the most cytotoxic with a mean GI50 value of 0.78 μM and also exhibited selective cytotoxicities for HL-60 (TB), K-562, MOLT-4, RPMI-8226, and SR with GI50 values of 0.11, 0.42, 0.09, 0.14, and 0.19 μM, respectively.  相似文献

Anti-allergic substances from the rhizomes of Dioscorea membranacea   总被引:2,自引:0,他引:2  
Extracts of five species of Thai medicinal plants, locally known as Hua-Khao-Yen, were screened for anti-allergic activities using RBL-2H3 cells. Of the five species studied, the ethanolic extract of Dioscorea membranacea exhibited potent inhibitory activity against β-hexosaminidase release as a marker of degranulation in RBL-2H3 cells, with an IC50 value of 37.5 μg/mL. Eight compounds were isolated from this crude ethanolic extract, [two naphthofuranoxepins (1, 2), one phenanthraquinone (3), three steroids (4–6), and two steroidal saponins (7, 8)], and tested for their anti-allergic activities. The results showed that dioscorealide B (2) possessed the highest activity with an IC50 value of 5.7 μM, followed by dioscoreanone (3, IC50 = 7.7 μM), dioscorealide A (1, IC50 = 27.9 μM), and diosgenin (9, IC50 = 29.9 μM). Structure–activity relationship studies of naphthofuranoxepins on anti-allergic activity revealed that the hydroxylation at position 8 conferred higher activity than methoxylation. For diosgenin derivatives, the aglycone was found to possess higher activity than the diglucosylated molecule; whereas substitution with rhamnoglucosides apparently results in loss of activity. Furthermore, effects of dioscorealide A, dioscorealide B, and dioscoreanone on antigen-induced release of TNF- and IL-4 in the late phase reaction were also examined.  相似文献

Dry biomass of Spirulina platensis re-hydrated for 48 h was employed as a biosorbent in tests of cadmium(II) removal from water. Various concentrations of biomass (from 1 to 4 g l−1) and metal (from 100 to 800 mg l−1) were tested. Low biomass levels (Xo  2 g l−1) ensured metal removal up to 98% only at Cd0= 100 and 200 mg l−1, while Xo  2.0 g l−1 were needed at Cd0 = 400 mg l−1 to achieve satisfactory results. Whereas Xo = 4.0 g l−1 was effective to remove up to Cd0 = 500 mg l−1, a further increase in metal concentration (Cd0 = 600 and 800 mg l−1) led to progressive worsening of the system performance. At a given biomass levels, the kinetics of the process was better at low Cd2+ concentrations, while, raising the adsorbent level from 1.0 to 2.0 g l−1 and then to 4.0 g l−1, the rate constant of biosorption increased by about one order of magnitude in both cases and the adsorption capacity of the system progressively decreased from 357 to 149 mg g−1.  相似文献

This paper deals with the chiral separation of triiodothyronine (T3) and thyroxine (T4) by HPLC and micro-HPLC. The separation of T3 and T4 is of great pharmaceutical and clinical interest, since the enantiomers exhibit different pharmacological activities. The HPLC measurements were performed on a chiral stationary ligand-exchange phase using l-4-hydroxyproline bonded via 3-glycidoxypropyltrimethoxysilane to silica gel as a selector. Also a chiral teicoplanin (Chirobiotic ™®) phase was used.

In micro-HPLC the chiral separation behaviour of l-4-hydroxyproline, and of the macrocyclic antibiotics teicoplanin and teicoplanin aglycone was investigated for the enantioseparation of T3 and T4. l-4-Hydroxyproline was bonded to 3 μm and the glycopeptide antibiotics were bonded to 3.5 μm silica gel and separations were accomplished by microbore HPLC columns (10 cm × 1 mm I.D.). With both techniques and all chiral selectors investigated T3 and T4 were baseline resolved. micro-HPLC was found to be superior to analytical HPLC with respect to low consumption of packing material, mobile phase and analyte.  相似文献

In order to elucidate early Aptian marine paleotemperature evolution across the period of enhanced organic carbon (Corg)-burial [Oceanic Anoxic Event (OAE) 1a], stable isotope analyses were performed on pelagic limestones at Deep Sea Drilling Project Site 463, central Pacific Ocean. The δ18O data exhibit a distinct anomaly by ~ − 2‰ spanning the OAE 1a interval (i.e., a ~ 6 m-thick, phytoplanktonic Corg-rich unit constrained by magneto-, bio- and δ13C stratigraphy). Elucidation of paleotemperature significance of the δ18O shift is made by taking account of recent Sr/Ca evidence at the same section, which revealed that geochemical signals in carbonate-poor lithologies are relatively unaltered against burial diagenesis. By discriminating δ18O values from carbonate-poor samples (CaCO3 contents = 5–30 wt.%), it appears that an abrupt rise in sea-surface temperatures (SSTs) by 8 °C (= − 1.7‰ shift in δ18O) occurred immediately before OAE 1a, whereas a cooling mode likely prevailed during the peak Corg-burial. In terms of its stratigraphic relationship as to the Corg-rich interval and to a pronounced negative δ13C excursion, as well as its timescale, the observed SST rise resembles those associated with the Paleocene–Eocene thermal maximum and, more strikingly, Jurassic Toarcian OAE. This observation is consistent with the hypothesis that these paleoenvironmental events were driven by a common causal mechanism, which was likely initiated by the greenhouse effect via massive release of CH4 or CO2 from the isotopically-light carbon reservoir and terminated by a negative productivity feedback.  相似文献

The effects of prolonged oral administration (21 days) of fumonisin B1 (FB1) and aflatoxin B1 (AFB1) were studied in male New Zealand rabbits by clinical, pathological, biochemical and sphingolipid analyses. Twenty-four animals were randomly divided into the following four experimental groups: (A) 0 mg FB1 + 0 μg AFB1/(kg body weight (bw) day) (control); (B) 0 mg FB1 + 30 μg AFB1/(kg bw day); (C) 1.5 mg FB1/(kg bw day) + 30 μg AFB1/(kg bw day); (D) 1.5 mg FB1/(kg bw day) + 0 μg AFB1. Animals from group B and principally from group C presented clinical signs of intoxication. Rabbits from group C presented a lower body weight gain than controls. Differences were observed between intoxicated rabbits and controls with respect to absolute and relative liver and kidney weight, hepatic function, serum urea and creatinine levels and Sa/So ratio. The most frequent hepatic and renal injuries were vacuolar degeneration of the liver and kidney as shown by the histopathological and serum biochemical results. Combined administration of AFB1 and FB1 resulted in synergistic toxic effects both in the liver and in the kidney, but hepatic injuries were more marked.  相似文献

The first examples of binary palladium(II) derivatives of unsaturated carboxylic acids are reported. It was found that the interaction of Pd3(μ-OAc)6 with the ,β-unsaturated 1-methylcrotonic (tiglic) and crotonic acids leads to the corresponding carboxylates of composition Pd3[μ-O2CC(R′) = CHMe]6, where R′ = Me (1) or H (2). The new compounds have been characterized by elemental analysis, solid and solution IR, 1H and 13C NMR, and ESI mass spectrometry. The crystal structure of 1 has been determined. This molecule displays a central Pd3 cyclic core with Pd–Pd distances of 3.093–3.171 Å. Each Pd–Pd bond is bridged by a pair of carboxylate ligands, one above and the other below the Pd3 plane, providing a square planar coordination for each Pd atom in an approximate D3h overall symmetry arrangement. Solution spectroscopic data show that the bridging η112 interaction of the carboxylates of 1 and 2 is readily displaced, with a change of the ligand to the terminal (η1) coordination mode.  相似文献

Two new spin-crossover complexes, [Fe(Medpq)(py)2(NCS)2] · py · 0.5H2O (1) and [Fe(Medpq)(py)2(NCSe)2] · py (2) (Medpq = 2-methyldipyrido[3,2-f:2′,3′-h]-quinoxaline, py = pyridine), have been synthesized. The crystal structures were determined at both room temperature (298 K) and low temperature (110 K). Complexes 1 and 2 crystallize in the orthorhombic space group Pbca and monoclinic space group P21/n, respectively. In both complexes, the distorted [FeN6] octahedron is formed by six nitrogen atoms from Medpq, the trans pyridine molecules and the cis NCX groups. The thermal spin transition is accompanied by the shortening of the mean Fe–N distances by 0.194 Å for 2. The mononuclear [Fe(Medpq)(py)2(NCS)2] and [Fe(Medpq)(py)2(NCSe)2] neutral species interact each other via π-stacking, resulting in a one-dimensional extended structure for both 1 and 2. There exist C–HX (X = S, Se) hydrogen bonds for both complexes. Variable-temperature magnetic susceptibility measurements and Mössbauer spectroscopy reveal the occurrence of a gradual spin transition. The transitions are centered at T1/2 = 120 K for 1 and T1/2 = 180 K for 2, respectively.  相似文献

Evidence is presented suggesting, for the first time, that the protein foldability metric σ = (Tθ − Tf) / Tθ, where Tθ and Tf are, respectively, the collapse and folding transition temperatures, could be used also to measure the foldability of RNA sequences. These results provide further evidence of similarities between the folding energy landscapes of proteins and RNA. The importance of σ is discussed in the context of the in silico design of rapidly foldable RNA sequences.  相似文献

Structure–activity relationship (SAR) studies on the tricyclic isoxazole series of MRP1 modulators have resulted in the identification of potent and selective inhibitors containing cyclohexyl-based linkers. These studies ultimately identified compound 21b, which reverses drug resistance to MRP1 substrates, such as doxorubicin, in HeLa-T5 cells (EC50 = 0.093 μM), while showing no inherent cytotoxicity. Additionally, 21b inhibits ATP-dependent, MRP1-mediated LTC4 uptake into membrane vesicles prepared from the MRP1-overexpressing HeLa-T5 cells (EC50 = 0.064 μM) and shows selectivity (1115-fold) against the related transporter, P-glycoprotein, in HL60/Adr and HL60/Vinc cells. Finally, when dosed in combination with the oncolytic MRP1 substrate vincristine, 21b showed tumor regression and growth delay in MRP1-overexpressing tumors in vivo.  相似文献

Three feeding trials with different dietary treatments were conducted to evaluate predictions of dry matter intake and average daily gain of Chinese local beef cattle by Cornell Net Carbohydrate and Protein System (CNCPS) version 5.0. In Trial 1, 60 growing Jin Nan yellow steers (local yellow cattle breed originated from Shanxi Province) were assigned to two treatments with five pens in each treatment and six animals in each pen. Trial 2 was conducted with 72 Lu Xi yellow growing steers (local yellow cattle breed originated from Shandong Province) in 12 conventional pens. These animals were assigned to four treatments with six animals in each pen. In Trial 3, 60 Lu Xi yellow steers were allocated to four treatments with five animals in each pen. Average daily dry matter intake (DMI) for each steer was measured as a mean of each pen. All the data about animals, environment, management and feeds required by the CNCPS model were collected, and model predictions were generated for animals of each pen. Model predictions were evaluated by mean bias, mean square prediction error (MSPE) and regression between the observed and predicted values. Animals in 9 of 10 treatments showed a very close model-predicted DMI values to the observed. In another treatment, animals had a mean bias of −0.72 kg DM/day (TRT 10 in Trial 3). With overall data included, the regression equation between observed and predicted DMI was: YOBS = 1.10XCNCPS − 0.78 (R2 = 0.82; P<0.0001), with an intercept not different (P=0.32) from zero, but a slope still different (P<0.0001) from 1. Although the model-predicted ADG values were very close to the observed values in all trials, most of the predicted values were always lower than the observed values with absolute mean bias less than 0.2. When all data were pooled, the regression equation between observed and predicted ADG was: YOBS = 1.07XCNCPS + 0.02 (R2 = 0.73; P<0.0001), with an intercept not different from zero (P=0.88), but a slope that differed from unity (P<0.0001). Results suggest that the CNCPS model is an acceptable model to predict dietary DM intake and average daily gain of Chinese local beef cattle breeds. Further studies are warranted to introduce model adjustment for Chinese conditions.  相似文献

The effects of β-glucan (BG) prepared from spent brewer’s yeast on gelatinization and retrogradation of rice starch (RS) were investigated as functions of mixing ratio and of storage time. Results of rapid visco-analysis (RVA) indicated that addition of BG increased the peak, breakdown, setback, and final viscosities, but decreased the pasting temperatures of the rice starch/β-glucan (RS/BG) mixtures. Differential scanning calorimetry (DSC) data demonstrated an increase in onset (To), peak (Tp), and conclusion (Tc) temperatures and a decrease in gelatinization enthalpy (ΔH1) with increasing BG concentration. Storage of the mixed gels at 4 °C resulted in a decrease in To, Tp, Tc, and melting enthalpy (ΔH2). The retrogradation ratio (ΔH2H1) and the phase transition temperature range (Tc − To) of the mixed gels increased with storage time, but this effect was reduced by the addition of BG. BG addition also slowed the syneresis of the mixed gels. Results of dynamic viscoelasticity measurement indicated that the addition of BG promoted RS retrogradation at the beginning and then retarded it during longer storage times. The added BG also retarded the development of gel hardness during refrigerated storage of the RS/BG mixed gels.  相似文献

Cyclic voltammetry at potential range − 1.1 to 0.5 V from aqueous buffer solution (pH 7) containing CoCl2 produced a well defined cobalt oxide (CoOx) nanoparticles deposited on the surface of glassy carbon electrode. The morphology of the modified surface and cobalt oxide formation was examined with SEM and cyclic voltammetry techniques. Hemoglobin (Hb) was successfully immobilized in cobalt-oxide nanoparticles modified glassy carbon electrode. Immobilization of hemoglobin onto cobalt oxide nanoparticles have been investigated by cyclic voltammetry and UV–visible spectroscopy. The entrapped protein can take direct electron transfer in cobalt-oxide film. A pair of well defined, quasi-reversible cyclic voltammetric peaks at about − 0.08 V vs. SCE (pH 7), characteristic of heme redox couple (Fe(III)/Fe(II)) of hemoglobin, and the response showed surface controlled electrode process. The dependence of formal potential (E0′) on the solution pH (56 mV pH− 1) indicated that the direct electron transfer reaction of hemoglobin was a one-electron transfer coupled with a one proton transfer reaction process. The average surface coverage of Hb immobilized on the cobalt oxide nanoparticles was about 5.2536 × 10− 11 mol cm− 2, indicating high loading ability of nanoparticles for hemoglobin entrapment. The heterogeneous electron transfer rate constant (ks) was 1.43 s− 1, indicating great of facilitation of the electron transfer between Hb and electrodeposited cobalt oxide nanoparticles. Modified electrode exhibits a remarkable electrocatalytic activity for the reduction of hydrogen peroxide and oxygen. The Michaels–Menten constant Km of 0.38 mM, indicating that the Hb immobilized onto cobalt oxide film retained its peroxidases activity. The biosensor exhibited a fast amperometric response < 5 s, a linear response over a wide concentration range 5 μM to 700 μM and a low detection limit 0.5 μM. According to the direct electron transfer property and enhanced activity of Hb in cobalt oxide film, a third generation reagentless biosensor without using any electron transfer mediator or specific reagent can be constructed for determination of hydrogen peroxide in anaerobic solutions.  相似文献

The reaction of thiamine with K2PtIICl4 and with PtIVCl4 in the presence of excess NaSCN in aqueous solution gave thiamine salts, (H-thiamine)[Pt(SCN)4] · 3H2O (1) and (H-thiamine)[Pt(SCN)6] · H2O (2), respectively, structures of which have been determined by X-ray diffraction. The thiamine molecule adopts the usual F conformation in each salt. In 1, [Pt(SCN)4]2− ions act as large planar spacers in the crystal lattice and interact scarcely with thiamine, except for a hydrogen bonding with the terminal hydroxy O(5γ). Instead, water molecules form two types of host–guest-like interactions with the pyrimidine and the thiazolium moieties of a thiamine molecule, one being a C(2)–Hwaterpyrimidine bridge and the other being an N(4′)–Hwaterthiazolium bridge. In 2, despite the much larger ion size, octahedral [Pt(SCN)6]2− ions form a C(2)–Hanionpyrimidine bridge and an N(4′)–Hanionthiazolium bridge. An additional hydrogen bonding between the anion and the terminal O(5γ) of thiamine creates a hydrogen-bonded macrocyclic ring {thiaminium–[Pt(SCN)6]2−}2, a supramolecule.  相似文献

We measured eddy covariance fluxes of CO2 and H2O over a flat irrigated olive orchard during growth, in different periods from Leaf Area Index (LAI) of 0.3–1.9; measurements of soil respiration were also collected. The daily net ecosystem exchange flux (FNEE) was practically zero at LAI around 0.4 or when the orchard intercepted 11% of the incoming daily radiation; at the end of the experiment, with LAI of 1.9 (and the fraction of intercepted daily radiation close to 0.5), FNEE was around 10 g CO2 m−2 day−1. The night-time ecosystem respiration (Reco), calculated from eddy fluxes in well-mixed night conditions, show a clear but non-linear dependence with LAI; it ranged from 0.05 to 0.15 mg CO2 m−2 s−1 (in average), being the lower limit ideally close to the heterotrophic soil respiration at the site. The gross primary production flux (FGPP) was linearly related to LAI within the LAI range of this experiment (with 11 g CO2 m−2 day−1 increments per unit of LAI) and to the fraction of intercepted radiation. The maximum rates of FGPP (0.75 mg CO2 m−2 s−1) were obtained in the summer mornings of 2002, at LAI close to 1.9. FGPP was strongly modulated by vapour pressure deficit (VPD) through the canopy conductance, even in absence of water stress. Hence, especially in the summer, the maximum rates of carbon assimilation are reached always before noon. The daily course of FGPP shows a two-phase pattern, first related to irradiance and then to canopy conductance. The water use efficiency (WUE) was, in average, 3.8, 6.3 and 7 g CO2 L−1 in 1999, 2001 and 2002, respectively, with maxima always in the early morning. Hourly WUE was strongly related to VPD (WUE = −10.25 + 22.52 × VPD−0.34). Our results suggest that drip irrigated orchards in general, and olive in particular, deserve specific carbon exchange and carbon budget studies and cannot be easily included in other biomes.  相似文献

Ferritins are ubiquitous iron storage and detoxification proteins distributed throughout the plant and animal kingdoms. Mammalian ferritins oxidize and accumulate iron as a ferrihydrite mineral within a shell-like protein cavity. Iron deposition utilizes both O2 and H2O2 as oxidants for Fe2+ where oxidation can occur either at protein ferroxidase centers or directly on the surface of the growing mineral core. The present study was undertaken to determine whether the nature of the mineral core formed depends on the protein ferroxidase center versus mineral surface mechanism and on H2O2 versus O2 as the oxidant. The data reveal that similar cores are produced in all instances, suggesting that the structure of the core is thermodynamically, not kinetically controlled. Cores averaging 500 Fe/protein shell and diameter  2.6 nm were prepared and exhibited superparamagnetic blocking temperatures of 19 and 22 K for the H2O2 and O2 oxidized samples, respectively. The observed blocking temperatures are consistent with the unexpectedly large effective anisotropy constant Keff = 312 kJ/m3 recently reported for ferrihydrite nanoparticles formed in reverse micelles [E.L. Duarte, R. Itri, E. Lima Jr., M.S. Batista, T.S. Berquó and G.F. Goya, Large Magnetic Anisotropy in ferrihydrite nanoparticles synthesized from reverse micelles, Nanotechnology 17 (2006) 5549–5555.]. All ferritin samples exhibited two magnetic phases present in nearly equal amounts and ascribed to iron spins at the surface and in the interior of the nanoparticle. At 4.2 K, the surface spins exhibit hyperfine fields, Hhf, of 436 and 445 kOe for the H2O2 and O2 samples, respectively. As expected, the spins in the interior of the core exhibit larger Hhf values, i.e. 478 and 486 kOe for the H2O2 and O2 samples, respectively. The slightly smaller hyperfine field distribution DHhf for both surface (78 kOe vs. 92 kOe) and interior spins (45 kOe vs. 54 kOe) of the O2 sample compared to the H2O2 samples implies that the former is somewhat more crystalline.  相似文献

Two series of N6-substituted adenosines with monocyclic and bicyclic N6 substituents containing a heteroatom were synthesized in good yields. These derivatives were assessed for their affinity ([3H]CPX), potency, and intrinsic activity (cAMP accumulation) at the A1 adenosine receptor in DDT1 MF-2 cells. In the monocyclic series, the N6-tetrahydrofuran-3-yl and thiolan-3-yl adenosines (1 and 26, respectively) were found to possess similar activities, whereas the corresponding selenium analogue 27 was found to be more potent. A series of nitrogen containing analogues showed varying properties, N6-((3R)-1-benzyloxycarbonylpyrrolidin-3-yl)adenosine (30) was the most potent at the A1AR; IC50 = 3.2 nM. In the bicyclic series, the effect of a 7-azabicyclo[2.2.1]heptan-2-yl substituent in the N6-position was explored. N6-(7-Azabicyclo[2.2.1]heptan-2-yl)adenosine (38) proved to be a reasonably potent A1 agonist (Ki = 51 nM, IC50 = 35 nM) while further substitution on the 7″-nitrogen with tert-butoxycarbonyl (31, IC50 = 2.5 nM) and 2-bromobenzyloxycarbonyl (34, IC50 = 9.0 nM) gave highly potent A1AR agonists.  相似文献

铝毒是限制酸性土壤中作物产量的主要因素之一。番茄(Solanum lycopersicum)是适合在酸性土壤中种植的主要经济作物, 不同品种番茄对铝胁迫的响应存在差异, 因此, 筛选苗期耐铝毒种质对番茄生产及研究具有重要意义。以10个番茄品种为材料, 采用室内土培盆栽, 设置1 000 µmol∙L-1 AlCl3·6H2O处理, 测定反映植物铝胁迫下生长状况的16个形态、生理生化及光合指标。通过主成分分析, 将铝胁迫下番茄幼苗的16个指标转化为5个独立的综合指标, 累积贡献率达90.779%。基于耐铝性综合评价值(A)的系统聚类分析, 将供试种质划分为5类, 第I类为高度耐铝品种Qianxi, 第V类为高度不耐铝品种Puluowangsi。经多元线性逐步回归分析得出番茄苗期耐铝评价方程: y=0.046+0.405X6+0.515X10-0.207X15+0.028X3 (R2=0.997), 从16个指标中提取出与A值显著相关(P<0.01)的4个指标: 丙二醛含量(X3)、净光合速率(X6)、叶面积(X10)和地下部干重(X15)。利用评价方程可判断不同番茄品种苗期的耐铝性, 使番茄耐铝性鉴定工作快速简便。  相似文献

The present work examines chemical and structural response in B. anthracis spores killed by a mixture of supercritical carbon dioxide (SCCO2) and hydrogen peroxide (H2O2). Deactivation of 6-log of B. anthracis spores by SCCO2 + H2O2 was demonstrated, but changes in structure were observed in only a small portion of spores. Results from phase contrast microscopy proved that this treatment is mild and does not trigger germination-like changes. TEM imaging revealed mild damage in a portion of spores while the majority remained intact. Dipicolinic acid (DPA) analysis showed that < 10% of the DPA was released from the spore core into the external milieu, further demonstrating only modest damage to the spores. Confocal fluorescent microscopy, assessing uptake of DNA-binding dyes, directly demonstrated compromise of the permeability barrier. However, the magnitude of uptake was small compared to spores that had been autoclaved. This work suggests that SCCO2 + H2O2 is quite mild compared to other sterilization methods, which has major implications in its application. These results provide some insight on the possible interactions between spores and the SCCO2 + H2O2 sterilization process.  相似文献
