During tethered flight in Drosophila melanogaster, spike activity of the second basalar flight-control muscle (M.b2) is correlated with an increase in both the ipsilateral wing beat amplitude and the ipsilateral flight force. The frequency of muscle spikes within a burst is about 100 Hz, or 1 spike for every two wing beat cycles. When M.b2 is active, its spikes tend to occur within a comparatively narrow phase band of the wing beat cycle. To understand the functional role of this phase-lock of firing in the control of flight forces, we stimulated M.b2 in selected phases of the wing beat cycle and recorded the effect on the ipsilateral wing beat amplitude. Varying the phase timing of the stimulus had a significant effect on the wing beat amplitude. A maximum increase of wing beat amplitude was obtained by stimulating M.b2 at the beginning of the upstroke or about 1 ms prior to the narrow phase band in which the muscle spikes typically occur during flight. Assuming a delay of 1 ms between the stimulation of the motor nerve and muscle activation, these results indicate that M.b2 is activated at an instant of the stroke cycle that produces the greatest effect on wing beat amplitude.  相似文献

Spinosad was evaluated in Hawaii as a replacement for organophosphate insecticides (naled, dichlorvos [DDVP], and malathion) in methyl eugenol and cue-lure bucket traps to attract and kill oriental fruit fly, Bactrocera dorsalis Hendel, and melon fly, B. cucurbitae Coquillett, respectively. In the first and second methyl eugenol trials with B. dorsalis, naled was in the highest rated group for all evaluation periods (at 5, 10, 15, and 20 wk). Spinosad was equal to naled at 5 and 10 wk during both trials 1 and 2, and compared favorably with malathion during trial 2. During the first cue-lure trial with B. cucurbitae, naled and malathion were in the top rated group at 5, 10, 15, and 20 wk. Spinosad was equal to naled at 5 wk. During the second cue-lure trial, spinosad and naled were both in the top rated group at 10, 15, and 20 wk. Use of male lure traps with methyl eugenol or cue-lure had no effect on attraction of females into test areas. Our results suggest that spinosad, although not as persistent as naled or malathion, is safer to handle and a more environmentally friendly substitute for organophosphate insecticides in methyl eugenol and cue-lure traps for use in B. dorsalis and B. cucurbitae areawide integrated pest management programs in Hawaii.  相似文献

The activity and resting behaviour of resistant and susceptible Anopheles stephensi Liston were recorded in acoustic actograph chambers lined with residual deposits of malathion, dieldrin or gamma HCH. In gamma HCH-treated flight chambers, SS and RS mosquitoes became active only after picking up lethal doses of insecticide, which explains why few SS and RS mosquitoes survive release into gamma HCH-treated experimental huts. Similar results were obtained in flight chambers treated with dieldrin; however, more mosquitoes would be expected to survive dieldrin under field conditions because resistance to this insecticide is greater than to gamma HCH. Mosquitoes in contact with malathion showed a three-phase activity pattern: an initial active phase, an inactive phase, and hyper-activity/convulsions. Initial activity or irritability was especially pronounced in SS and RS but absent in RR mosquitoes. Whether or not irritability would protect RS mosquitoes from malathion would probably depend on the ratio of sprayed to unsprayed surfaces in treated huts.  相似文献

Acute toxicities of three naturally occurring monoterpenoid essential oil constituents and the essential oil of rosemary were tested against late instars of Agriotes obscurus (L.) (Coleoptera: Elateridae). Both contact and volatile toxicities of thymol, citronellal, eugenol, and rosemary oil were determined. Also, phytotoxicity of these compounds was evaluated on corn germination and seedling development. Thymol had the greatest contact toxicity (LD50 = 196.0 microg/larva), whereas citronellal and eugenol were less toxic (LD50 = 404.9 and 516.5 microg/larva, respectively). Rosemary oil did not show any significant contact toxicity, even at 1,600 microg/larva. In terms of volatile toxicity, citronellal was the most toxic to wireworm larvae (LC50 = 6.3 microg/cm3) followed by rosemary oil (LC50 = 15.9 microg/cm3), thymol (LC50 = 17.1 microg/cm3), and eugenol (LC50 = 20.9 microg/cm3). Thymol, eugenol, and citronellal significantly inhibited corn seed germination and development, whereas rosemary oil had only minimal phytotoxic effects.  相似文献

The pheromone-mediated flight behavior of male Oriental fruit moths in a sustained-flight tunnel was observed after males were treated topically with sublethal concentrations of permethrin, carbaryl, chlordimeform, dieldrin, octopamine, serotonin, yohimbine, and cyproheptadine. With the exception of serotonin all compounds were found to disrupt one or more specific elements of the male precopulatory flight sequence. Among the insecticides, dieldrin was least active, whereas permethrin, carbaryl, and chlordimeform induced unique effects at specific phases of the sequence. Octopamine induced a hypersensitivity to the olfactory signal and mimicked one of the effects observed with chlordimeform. Yohimbine and cyproheptadine significantly decreased moth activation to the chemical signal but did not alter flight performance in responding moths. Yohimbine and cyproheptadine also reversed the effects induced by octopamine. The results of our study show that the complex precopulatory sequence of behaviors exhibited by males is very sensitive to sublethal concentrations of a range of neuroactive compounds.  相似文献

The effects of the monoterpenoids d-limonene, linalool, beta-myrcene, and alpha-terpineol on the growth and development of the German cockroach Blattella germanica (L.) were examined. We evaluated the chemicals' attractiveness in cockroach diet, quantified growth effects induced by monoterpenoids, examined embryotoxic properties of the compounds, and examined their effects on reproduction when administered by oral, topical, and vapor routes of entry. Untreated diet was significantly preferred compared with diet treated with high levels of d-limonene, linalool, and alpha-terpineol. The threshold for acceptance was between 1 and 10% (AI). All four monoterpenoids significantly influenced the days required by nymphs to reach the adult stage; in general, higher monoterpenoid concentrations in the diet were associated with a reduction in the days required by nymphs to mature. Application of high doses of d-limonene or linalool to oothecae of gravid female cockroaches significantly decreased the probability of young emerging from them but did not affect female mortality. Feeding on diet treated with monoterpenoids during nymphal development and through the early, premating period of the adult stage did not significantly influence numbers of broods produced per pair, numbers of offspring per brood, days required to produce a brood, total offspring produced by a pair, or female life span. Topical applications of linalool, beta-myrcene, or alpha-terpineol at near lethal rates to adult cockroaches before they mated had no significant influence on the reproductive parameters examined. A single exposure to sublethal levels of monoterpenoid vapors before mating had no significant influence on any reproductive parameters examined.  相似文献

Methyl eugenol (4-allyl-1,2-dimethoxybenzene-carboxylate) and cue-lure [4-(p-acetoxyphenyl)-2-butanone] are highly attractive kairomone lures to oriental fruit fly, Bactrocera dorsalis (Hendel), and melon fly, B. cucurbitae (Coquillett), respectively. Plastic bucket traps were evaluated as dispensers for methyl eugenol and cue-lure for suppression of the 2 fruit flies in Hawaii. Methyl eugenol and cue-lure mixtures were compared with pure methyl eugenol or cue-lure over 4 seasons. B. dorsalis captures differed significantly with treatment and season. B. dorsalis captures with 100% methyl-eugenol were significantly greater than all other treatments (25, 50, and 75%). B. cucurbitae captures also differed significantly with treatment but not with season. Captures with 100, 75, and 50% cue-lure were not significantly different. Bucket traps baited with cue-lure (+ malathion) and weathered under Hawaiian climatic conditions were attractive to B. cucurbitae up to 8 wk. Two methyl eugenol dispensers (canec disks and Min-U-Gel) were compared with bucket traps. Dispensers (methyl eugenol + malathion) were weathered for 2-16 wk under Hawaiian climatic conditions and bioassayed during summer and winter. Initially, captures of B. dorsalis were not significantly different for the 3 dispensers. Bucket traps and canec disks were most resistant to weather, remaining attractive to B. dorsalis flies up to 16 wk. Min-U-Gel was least resistant, losing attractiveness to B. dorsalis flies within 2 wk. On the basis of performance, bucket traps and canec disks were equally long-lived up to 14 wk; thereafter, bucket traps were slightly more attractive during winter. Canec disks were cheapest, but on the basis of possible environmental concerns, bucket traps may be the best all-around choice for areawide suppression of fruit flies.  相似文献

The differential palatability of monoterpenoid compositional types of Satureja douglasii to a native slug herbivore, Ariolimax dolichophallus, was determined. Types containing a high proportion of bicyclic monoterpenoids (camphene, camphor) were more palatable than those containing high proportions of p-menthane monoterpenoids (isomenthone, pulegone or carvone). In the case of individual monoterpenoids characteristic of these types, camphor proved to be most palatable and pulegone the least; carvone and isomenthone were intermediate. Differential herbivory on the compositional types was observed under field conditions. It is suggested that differential herbivory could have been important in determining the patterns of monoterpenoid variation and their distribution in S. douglasii.  相似文献

Many evolutionary ecological studies have documented sexual dimorphism in morphology or behaviour. However, to what extent a sex-specific morphology is used differently to realize a certain level of behavioural performance is only rarely tested. We experimentally quantified flight performance and wing kinematics (wing beat frequency and wing stroke amplitude) and flight morphology (thorax mass, body mass, forewing aspect ratio, and distance to centre of forewing area) in the butterfly Pararge aegeria (L.) using a tethered tarsal reflex induced flight set-up under laboratory conditions. On average, females showed higher flight performance than males, but frequency and amplitude did not differ. In both sexes, higher flight performance was partly determined by wing beat frequency but not by wing stroke amplitude. Dry body mass, thorax mass, and distance to centre of forewing area were negatively related to wing beat frequency. The relationship between aspect ratio and wing stroke amplitude was sex-specific: females with narrower wings produced higher amplitude whereas males show the opposite pattern. The essential oil of the leaves of P. racemosa var. terebinthina and P. racemosa var. grisea were examined by GC and GC/MS. The major constituents were alpha-terpineol acetate (27%), alpha-terpineol (20%) and 4-methoxy eugenol (12.6%) for Pracemosa var. terebinthina and 4-methoxy-isoeugenol (75.2%) and 4-methoxy-eugenol (4.5%) for P. racemosa var. grisea.  相似文献

Low-angle X-ray diffraction patterns from relaxed fruitfly (Drosophila) flight muscle recorded on the BioCat beamline at the Argonne Advanced Photon Source (APS) show many features similar to such patterns from the "classic" insect flight muscle in Lethocerus, the giant water bug, but there is a characteristically different pattern of sampling of the myosin filament layer-lines, which indicates the presence of a superlattice of myosin filaments in the Drosophila A-band. We show from analysis of the structure factor for this lattice that the sampling pattern is exactly as expected if adjacent four-stranded myosin filaments, of repeat 116 nm, are axially shifted in the hexagonal A-band lattice by one-third of the 14.5 nm axial spacing between crowns of myosin heads. In addition, electron micrographs of Drosophila and other flies (e.g. the house fly (Musca) and the flesh fly (Sarcophaga)) combined with image processing confirm that the same A-band superlattice occurs in all of these flies; it may be a general property of the Diptera. The different A-band organisation in flies compared with Lethocerus, which operates at a much lower wing beat frequency (approximately 30 Hz) and requires a warm-up period, may be a way of optimising the myosin and actin filament geometry needed both for stretch activation at the higher wing beat frequencies (50 Hz to 1000 Hz) of flies and their need for a rapid escape response.  相似文献

Plant essential oils are widely used as fragrances and flavours in the cosmetic, perfume, drug and food industries. Oxygenated monoterpenes are widespread components of the essential oils, usually occurring in high amount. In this paper, the antibacterial activities of twenty-one oxygenated monoterpenes (borneol, borneol acetate, camphor, carvone, 1,8-cineole, citronellal, beta-citronellol, dihydrocarvone, fenchol, fenchone, geraniol acetate, isomenthol, limonene oxide, linalool, linalool acetate, nerol, nerol acetate, terpinen-4-ol, alpha-terpineol, menthol and menthone) and penicillin (standard antibiotic) were determined using a disc diffusion method (in vitro) against 63 bacterial strains, belonging to 37 different genera and 54 species (plant, food and clinic origins). The results showed that the oxygenated monoterpenes exhibited a variable degree of antibacterial activities. These compounds also inhibited the growth of bacterial strains by producing a weak zone of inhibition from 7 to 11 mm in diameter, depending on the susceptibility of the tested bacteria. Among the tested compounds, nerol, linalool alpha-terpineol, fenchol and terpinen-4-ol showed antibacterial activity at a broad spectrum. However, their antibacterial activities were lower than those of penicillin. In contrast to these compounds, camphor and 1,8-cineole exhibited no inhibition effects on the growth of all tested bacteria.  相似文献

Toxicological studies of pesticides on cytoplasmic streaming in Nitella   总被引:2,自引:0,他引:2  
In the present study, changes in velocity of cytoplasmic streaming in the giant internodal cells of Nitella for varying concentration of the pesticides, 2,4-D, dieldrin, malathion, methyl parathion and endosulfan, were measured. Marked decrease in the velocity of cytoplasmic streaming was found at the concentrations of 0.01, 0.1, 1, 10 and 100mM. Dieldrin was the most toxic to all the pesticides investigated, followed by methyl parathion, endosulfan, malathion and 2,4-D. Threshold values for dieldrin, methylparathion, endosulfan, malathion and 2,4-D as indicated by the onset of decrease in the normal cytoplasmic streaming velocity were less than 6.25 x 10(-6), 2.5 x 10(-5), 5 x 10(-5), 5 x 10(-5) and 1.25 x 10(-5)M respectively. Cessation of streaming was noticed above 1mM in dieldrin and above 10mM when exposed to methylparathion and endosulfan. Cessation of streaming was not seen up to 100mM concentration of 2,4-D and malathion.  相似文献

Wheat seeds, when exposed to essential oils, are able to metabolise certain monoterpenes. The actual amounts of the compounds and their derivatives in the endosperm and embryo of wheat seeds, after exposure to the monoterpenes were determined. Neral and geranial, which are the constituents of citral, are reduced and oxidised to the corresponding alcohols and acids. Similarly citronellal, pulegone and carvacrol are converted partly to the corresponding reduction and oxidation products. The aromatic compound vanillin is partly reduced to vanillyl alcohol or oxidised to vanillic acid. In all cases it seems that part of the compounds applied are degraded, as indicated by the inability to account for all the compounds, which were supplied to the germinated seeds. In most cases the derivatives of the essential oil applied were less toxic than the parent compound. The possible role of non-specific enzymes by which the compounds are oxidised or reduced is discussed.  相似文献

A method for determining and analyzing the wing beat frequency in Diptera is presented. This method uses an optical tachometer to measure Diptera wing movement during flight. The resulting signal from the optical measurement is analyzed using a Fast Fourier Transform (FFT) technique, and the dominant frequency peak in the Fourier spectrum is selected as the wing beat frequency. Also described is a method for determining quantitatively the degree of variability of the wing beat frequency about the dominant frequency. This method is based on determination of a quantity called the Hindex, which is derived using data from the FFT analysis. Calculation of the H index allows computer-based selection of the most suitable segment of recorded data for determination of the representative wing beat frequency. Experimental data suggest that the H index can also prove useful in examining wing beat frequency variability in Diptera whose flight muscle structure has been genetically altered. Examples from Drosophila indirect flight muscle studies as well as examples of artificial data are presented to illustrate the method. This method fulfills a need for a standardized method for determining wing beat frequencies and examining wing beat frequency variability in insects whose flight muscles have been altered by protein engineering methods.  相似文献

Volatiles from mouse carcasses in decay stages ranging from fresh to 33 days old were used to investigate oriented flight and landings in male and female blow flies of Calliphora vicina Robineau‐Desvoidy (Diptera: Calliphoridae). Oriented flight increased significantly from 36% towards fresh carcasses to 68%, 61% and 65% towards carcasses aged 3 days, 6 days and 9 days, respectively. Carcasses aged 20 days and 33 days were significantly less attractive, achieving 51% and 41% attraction, respectively. No differences emerged between the sexes in oriented flight, but a significant increase in female landings at the most attractive carcasses was observed. Headspace collections from the different stages of decay showed a succession in the volatile profile emitted from the carcasses and identified nine chemicals which peak in quantity in concurrence with the most attractive stages of decay. Three of these chemicals also showed dose–response effects as indicated by a significant correlation between the amount present and the proportion of flies responding. Blow flies are important pests and efficient traps are needed. The significant interaction between fly sex and carcass age highlights behavioural differences between male and female blow flies which can be exploited in blow fly trapping. Three new volatile chemicals, butylated hydroxyl toluene, 3‐hydroxy‐2‐butanone and nonanal, emitted from dead mice are suggested as potential attractants.  相似文献

Anti-Candida potential of six terpenoids were evaluated in this study against various isolates of Candida albicans (n = 39) and non-C. albicans (n = 9) that are differentially susceptible to fluconazole. All the six terpenoids tested, showed excellent activity and were equally effective against isolates of Candida sps., tested in this study. Linalool and citral were the most effective ones, inhibiting all the isolates at ≤0.064% (v/v). Five among the six terpenoids tested were fungicidal. Time dependent kill curve assay showed that MFCs of linalool and eugenol were highly toxic to C. albicans, killing 99.9% inoculum within seven min of exposure, while that of citronellal, linalyl acetate and citral required 15 min, 1 h and 2 h, respectively. FIC index values (Linalool - 0.140, benzyl benzoate - 0.156, eugenol - 0.265, citral - 0.281 and 0.312 for linalyl acetate and citronellal) and isobologram obtained by checker board assay showed that all the six terpenoids tested exhibit excellent synergistic activity with fluconazole against a fluconazole resistant strain of C. albicans. Terpenoids tested arrested C. albicans cells at different phases of the cell cycle i.e. linalool and LA at G1, citral and citronellal at S phase and benzyl benzoate at G2-M phase and induced apoptosis. Linalool, citral, citronellal and benzyl benzoate caused more than 50% inhibition of germ tube induction at 0.008%, while eugenol and LA required 0.032 and 0.016% (v/v) concentrations, respectively. MICs of all the terpenoids for the C. albicans growth were non toxic to HeLa cells. Terpenoids tested exhibited excellent activity against C. albicans yeast and hyphal form growth at the concentrations that are non toxic to HeLa cells. Terpenoids tested in this study may find use in antifungal chemotherapy, not only as antifungal agents but also as synergistic agents along with conventional drugs like fluconazole.  相似文献

Males of the midge Chironomus plumosus fly solely to mate. They maintain station for long periods in moving air. Females patrol in search of males and, after receiving a spermatophore, fly to oviposition sites. The requirements of flight in males and females are therefore fundamentally different. Females are larger than males, on average, so these differences could stem from scaling rules governing the geometry of space. The same explanation might apply to flight differences within the sexes and even to peculiarities of flight architecture in C. plumosus compared to other flying animals. In other words, might flight design be accounted for entirely by mechanical constraints without recourse to natural selection?
To test this hypothesis, the power output of C. plumosu was measured as size-specific muscle mass. Contrary to expectation, little evidence was found of scaling effects in this measure of power. Despite its being among the smallest of animals to fly, C. plumosus turns out to have the largest mass of flight muscle, relative to body size, yet found among animals. Differences both between C. plumosus and other species and within C. plumosus are, in general, more readily accounted for by the requirements of the mating system within the viscous universe encountered by small flying animals. We conclude that it is natural selection rather than mechanical constraint that is the primary influence determining the architecture of flight in this small animal.  相似文献
