Nonhost angiosperm volatiles and verbenone were tested for their ability to disrupt the response of western pine beetle, Dendroctonus brevicomis LeConte (Coleoptera: Scolytidae), to attractant-baited multiple funnel traps. Verbenone significantly reduced attraction; however, no difference was observed between 4 and 50 mg/24-h release rates. Combinations of six bark volatiles (benzyl alcohol, benzaldehyde, trans-conophthorin, guaiacol, nonanal, and salicylaldehyde), three green leaf volatiles [(E)-2-hexenal, (E)-2-hexen-1-ol, and (Z)-2-hexen-1-ol], and the nine compounds combined did not significantly reduce D. brevicomis response to attractant-baited traps. However, a significant effect was observed when the bark and green leaf volatiles were combined with verbenone. The nine nonhost angiosperm volatiles (NAVs) significantly augmented the effect of both release rates of verbenone, reducing trap catches to levels significantly below that of either release rate of verbenone alone. trans-Conophthorin, a compound reported to have behavioral activity in a number of other scolytids, was not critical to the efficacy of our NAV blend. Our results suggest that the addition of nonhost angiosperm volatiles to verbenone could be important for developing successful semiochemical-based management techniques for D. brevicomis.

Diamondback moth, Plutella xylostella (L.) (Lepidoptera: Plutellidae), is one of the most serious pests of Brassicaceae crops worldwide. Electrophysiological and behavioral responses of P. xylostella to green leaf volatiles (GLVs) alone or together with its female sex pheromone were investigated in laboratory and field. GLVs 1-hexanol and (Z)-3-hexen-1-ol elicited strong electroantennographic responses from unmated male and female P. xylostella, whereas (Z)-3-hexenyl acetate only produced a relatively weak response. The behavioral responses of unmated moths to GLVs were further tested in Y-tube olfactometer experiments. (E)-2-Hexenal, (Z)-3-hexen-1-ol, and (Z)-3-hexenyl acetate induced attraction of males, reaching up to 50%, significantly higher than the response to the unbaited control arm. In field experiments conducted in 2008 and 2009, significantly more moths were captured in traps baited with synthetic sex pheromone with either (Z)-3-hexenyl acetate alone or a blend of (Z)-3-hexenyl acetate, (Z)-3-hexen-1-ol, and (E)-2-hexenal compared with sex pheromone alone and other blend mixtures. These results demonstrated that GLVs could be used to enhance the attraction of P. xylostella males to sex pheromone-baited traps.

We tested the hypothesis that green leaf volatiles (GLVs) disrupt the response of overwintered pine shoot beetles, Tomicus piniperda (L.) to multiple-funnel traps baited with the attractive host volatile α-pinene. A combination of four GLV alcohols, 1-hexanol ( E )-2-hexen-1-ol ( Z )-2-hexen-1-ol, and ( Z )-3-hexen-1-ol, caused 54 and 36% reduction in the number of pine shoot beetles captured in two separate trapping experiments. Similarly, a combination of the four alcohols plus two GLV aldehydes, hexanal and ( E )-2-hexenal, caused 38% reduction in the number of pine shoot beetles captured compared with α-pinene alone. A blend of the two GLV aldehydes was not disruptive. None of the four GLV alcohols nor the two GLV aldehydes were disruptive when tested individually. The finding that the blend of four GLV alcohols reduced attraction of T. piniperda supports the general hypothesis that GLVs common to nonhost angiosperms are disruptive to conifer-attacking bark beetles (Scolytidae).

Abstract  Effects of maize ( Zea mays L.) volatiles induced by larvae of the Asian corn borer, Ostrinia furnacalis (Guenée), on the orientation behaviors of Asian corn borer larvae and oviposition of the females were investigated. Nineteen volatile chemicals, with terpenes being the major components, were identified from maize plants attacked by third instar Asian corn borer larvae. Coupled gas chromatographic-electroantennographic detection (GC-EAD) analyses revealed some electroantennographic differences between female and male Asian corn borer antennae in response to larvae-induced maize volatiles; female responded to ( E )-2-hexenal, nonanal, ( Z )-3-hexen-1-ol and three unknown compounds while the male only responded to ( E )-2-hexenal, nonanal and one unknown compound. In laboratory orientation bioassays, Asian corn borer neonate larvae were attracted to extracts collected from Asian corn borer-damaged plants as well as to synthetic farnesene, but were repelled by ( Z )-3-hexen-1-ol. In laboratory oviposition bioassays, gravid females laid fewer eggs on plants damaged by larvae than on mechanically damaged plants or undamaged plants. Adult Asian corn borer females deposited fewer eggs on wax paper treated with ( E )-2-hexenal or ( Z )-3-hexen-1-ol than on wax paper treated with hexane (control). The results suggest that Asian corn borer can affect the behaviors of conspecific larvae and adults by changing host plant volatiles.

1 The western pine beetle Dendroctonus brevicomis LeConte (Coleoptera: Scolytidae) is one of the most damaging insect pests of ponderosa pines Pinus ponderosa Douglas ex P. & C. Lawson in Western U.S.A. We compared the effect of verbenone, a well known bark beetle anti‐aggregation pheromone, with that of acetophenone on the attraction of D. brevicomis to its aggregation pheromone in a ponderosa pine forest in northern California. We tested the D. brevicomis aggregation pheromone alone and with three different release ratios of the aggregation pheromone (attractant) to verbenone or acetophenone (1 : 1, 1 : 2 and 1 : 5). 2 All treatments containing acetophenone or verbenone resulted in a significant reduction in the catch of D. brevicomis relative to the aggregation pheromone alone. When beetle responses to the three verbenone or three acetophenone treatments were pooled, the pooled verbenone treatment caught more D. brevicomis than the pooled acetophenone treatment. 3 There was no significant difference in the number of D. brevicomis caught among the three release rates of verbenone. By contrast, the 1 : 2 attractant : acetophenone ratio attracted significantly more D. brevicomis than the traps with the 1 : 5 attractant : acetophenone ratio. 4 Attraction of a major predator, Temnochila chlorodia (Mannerheim) (Coleoptera: Trogositidae), to the aggregation pheromone of D. brevicomis was reduced by verbenone, but not by acetophenone. Moreover, the T. chlorodia : D. brevicomis ratio for the pooled acetophenone treatment was 1.7‐fold greater than that for the attractant alone and two‐fold greater than the ratio for the pooled verbenone treatment, suggesting that acetophenone would not disrupt populations of this natural enemy. The importance of anti‐attractants in the biology of D. brevicomis and other bark beetles is discussed.

A blend of eight nonhost angiosperm volatiles (benzyl alcohol, benzaldehyde, guaiacol, nonanal, salicylaldehyde, (E)‐2‐hexenal, (E)‐2‐hexen‐1‐ol and (Z)‐2‐hexen‐1‐ol) without [NAV] and with [NAVV] (–)‐verbenone (4,6,6‐trimethylbicyclo[3.1.1]hept‐3‐en‐2‐one) were tested at low (L), medium (M) and high (H) release rates for their ability to reduce attraction of western pine beetle, Dendroctonus brevicomis LeConte, to attractant‐baited (exo‐brevicomin [racemic, 3 mg/d], frontalin [racemic, 3 mg/d] and myrcene [18 mg/d]) multiple funnel traps. NAV‐L (40 mg/d) had no significant effect. Verbenone alone (50 mg/d) and NAV‐M (240 mg/d) both significantly reduced attraction, but no significant difference was observed between the two treatment means. NAV‐H (430 mg/d) significantly reduced catches by ~60% and 78% compared to verbenone alone and the baited control, respectively. In a second experiment, combining (–)‐verbenone with NAV (NAVV) increased the effects observed in Experiment 1. NAVV‐M (240 mg/d) resulted in an ~69% and 83% reduction in trap catch compared to verbenone alone and the baited control, respectively. Significantly fewer D. brevicomis were captured in NAVV‐H (430 mg/d) than any other treatment resulting in an ~93% reduction in trap catch compared to the baited control. In a third experiment, NAVV was tested at three release rates for its ability to protect individual ponderosa pines, Pinus ponderosa Dougl. ex Laws., from attack by D. brevicomis. Cumulative release rates varied in direct proportion to tree diameter, but represented quarter, half and full NAVV rates. NAVV significantly reduced the density of D. brevicomis attacks, D. brevicomis successful attacks, and levels of tree mortality on attractant‐baited trees. Only three of 15 NAVV‐treated trees died from D. brevicomis attack while ~93% mortality (14/15) was observed in the untreated, baited control. Quarter and half rates were ineffective for reducing tree mortality.

A number of angiosperm nonhost volatiles (NHVs) and green leaf volatiles (GLVs) were tested alone and as supplements to the antiaggregation pheromone, verbenone, for their ability to disrupt attack by the mountain pine beetle (MPB), Dendroctonus ponderosae Hopkins (Coleoptera: Scolytidae), on lodgepole pine, Pinus contorta Dougl. ex Loud. var. latifolia Engel. Preliminary experiments led to a refined NHV blend [benzyl alcohol, guaiacol, benzaldehyde, nonanal, salicylaldehyde, and conophthorin] and a refined GLV blend [(Z)-3-hexen-1-ol and (E)-2-hexen-1-ol]. In a 20-replicate experiment, NHV and GLV groups both singly, and verbenone alone, significantly reduced MPB mass attack on pheromone-baited trees and on trees within 5 m of the pheromone-baited trees. Both blends in combination with verbenone reduced the number of mass attacked, baited trees to three out of 20 compared to 20 out of 20 of the baited controls. Each binary combination was also effective at reducing mass attack. In these experiments, all tested repellents were released from devices stapled to trees at the same point as the pheromone bait, suggesting that the repellency could have been to a point source, rather than to the whole tree. Therefore, in two further experiments bands of release devices were wrapped around the treated trees and the pheromone bait was removed from the treated trees. In one experiment, when the aggregation pheromone bait was suspended between pairs of trees treated with the NHV blend plus GLV blend plus verbenone, only three out of 25 treated pairs had mass attack on at least one member of the pair. In the other 60-replicate experiment, with no pheromone baits present, attack occurred on 13 untreated and 11 banded trees, all in the path of a large advancing infestation. However, the mean attack density on the banded trees was significantly reduced to a level below the 40 attacks m–2 of bark surface required to kill a healthy lodgepole pine. As a result of these experiments, operational trials are recommended.

Conifer feeding bark beetles (Coleoptera, Curculionidae, Scolytinae) pose a serious economic threat to forest production. Volatiles released by non-host angiosperm plants (so called non-host volatiles, NHV) have been shown to reduce the risk of attack by many bark beetle species, including the European spruce bark beetle, Ips typographus. However, the most active blend for I. typographus, containing three green leaf volatiles (GLVs) in addition to the key compounds trans-conophthorin (tC) and verbenone, has been considered too expensive for use in large-scale management. To lower the cost and improve the applicability of NHV, we aim to simplify the blend without compromising its anti-attractant potency. Since the key compound tC is expensive in pure form, we also tested a crude version: technical grade trans-conophthorin (T-tC). In another attempt to find a more cost effective substitute for tC, we evaluated a more readily synthesized analog: dehydro-conophthorin (DHC). Our results showed that 1-hexanol alone could replace the three-component GLV blend containing 1-hexanol, (3Z)-hexen-1-ol, and (2E)-hexen-1-ol. Furthermore, the release rate of tC could be reduced from 5 mg/day to 0.5 mg/day in a blend with 1-hexanol and (–)-verbenone without compromising the anti-attractant activity. We further show that T-tC was comparable with tC, whereas DHC was a less effective anti-attractant. DHC also elicited weaker physiological responses in the tC-responding olfactory receptor neuron class, providing a likely mechanistic explanation for its weaker anti-attractive effect. Our results suggest a blend consisting of (–)-verbenone, 1-hexanol and technical trans-conophthorin as a cost-efficient anti-attractant for forest protection against I. typographus.

Two candidate attractants, phenol and 1,4-benzoquinone, a synthetic mixture of typical compounds from green-leaf odours [(Z)-3-hexenyl acetate: (Z)-3-hexen-1-ol: benzaldehyde: (E)-2-hexen-1-ol: 1-hexanol; 100:20:10:1:1] and freshly damaged oak leaves were screened for field attractancy in funnel traps in Hungary. Males of two Rhizotrogus spp. (Coleoptera, Scarabaeidae, Melolonthinae), R. aestivus 01. and R. vernus Germ. were caught in larger numbers. While R. aestivus catches were probably chance captures, male R. vernus was significantly attracted to the baits containing 1,4-benzoquinone. This compound represents a promising basis for the development of a monitoring trap for R. vernus.

1 Olfactory responses of the Colorado potato beetle (CPB), Leptinotarsa decemlineata (Say) (Coleoptera: Chrysomelidae), a generalist predator, Podisus maculiventris (Say) (Hemiptera, Heteroptera: Pentatomidae) (Pm), and a specialist predator, Perillus bioculatus (F.) (Hemiptera, Heteroptera: Pentatomidae) (Pb) were investigated. Volatiles tested included 20 compounds emitted by undamaged potato plants (Solanum tuberosum), plants that had been artificially damaged, or plants damaged by feeding by CPB larvae. 2 Coupled gas chromatography/electroantennogram detector (GC/EAD) recordings revealed five compounds for which reliable responses were recorded from CPB antennae: (E)-2-hexen-1-ol, (Z)-3-hexen-1-ol, (±)-linalool, nonanal, methyl salicylate, and indole. Both Pm and Pb responded selectively to the same compounds as the CPB with exceptions: (1) (Z)-3-hexenyl butyrate elicited reliable responses for both Pm and Pb, and (2) (E)-2-hexen-1-ol and (Z)-3-hexen-1-ol were inactive for Pm and Pb under these conditions. Dose–response curves showed that CPB was at least 100 times more sensitive to (E)-2-hexen-1-ol than were the predators. Both predators were more sensitive to each of the other compounds than were CPB. Both CPB and Pm were attracted to a five component blend comprising (E)-2-hexen-1-ol, (Z)-3-hexen-1-ol, (±)-linalool, nonanal and methyl salicylate. However, attraction of CPB to the blend occurred only with lower doses of (E)-2-hexen-1-ol and (Z)-3-hexen-1-ol. 3 These results show that the herbivore (CPB) has olfactory receptors which are more sensitive to constitutive host plant volatiles, e.g. green leaf volatiles, while both generalist (Pm) and specialist (Pb) predators are more sensitive to systemic volatiles produced in response to prey feeding.

In the present study, Y-tube olfactometric assays demonstrated that headspace volatile extracts collected from leaf miner-damaged, or artificially damaged, bean plants were more attractive to naive females of the parasitoid insect Opius dissitus than those collected from healthy plants. Headspace extracts from both Liriomyza huidobrensis and Liriomyza sativae second-instar larvae-damaged beans were analyzed by coupled gas chromatography-electroantennographic detection (GC-EAD). Of nine EAD-active volatiles identified, (3E)-4,8-dimethyl-1,3,7-nonatriene, (3Z)-hexenyl acetate, (syn)-2-methylpropanal oxime, and (syn)-2-methylbutanal oxime were the most abundant compounds that evoked significant electroantennogram (EAG) responses. Compounds (3Z)-hexen-1-ol, (anti)-2-methylbutanal oxime, linalool, beta-caryophyllene, and (3E,7E)-4,8,12-trimethyl-1,3,7,11-tridecatetraene also elicited clear EAG responses but were present in smaller amounts. Choice experiments in a Y-tube olfactometer indicated that synthetic versions of (3Z)-hexen-1-ol, 2-methylpropanal oxime, 2-methylbutanal oxime, 3-methylbutanal oxime, linalool, (E,E)-alpha-farnesene, and (3E,7E)-4,8,12-trimethyl-1,3,7,11-tridecatetraene were attractive individually, while (3Z)-hexenyl acetate and (3E)-4,8-dimethyl-1,3,7-nonatriene were unattractive at concentrations similar to those obtained from the headspace collection. Moreover, a blend of nine EAD-active volatiles was significantly more attractive relative to hexane (solvent). A mixture of oximes, tereponids, and green leaf volatiles may facilitate host location by female O. dissitus.

The response of the two most abundant cockchafer species in central Europe, Melolontha hippocastani and M. melolontha, towards phenol, mixtures of phenol with the leaf alcohol (Z)-3-hexen-1-ol and the known cockchafer pheromones, 1,4-benzoquinone (M. hippocastani) and toluquinone (M. melolontha), was investigated in the field. During the swarming period at dusk, phenol attracted males of both species, and enhanced the known attraction of cockchafer males towards (Z)-3-hexen-1-ol. A mixture of phenol plus (Z)-3-hexen-1-ol was less attractive for M. hippocastani males than a mixture of (Z)-3-hexen-1-ol plus 1,4-benzoquinone, whereas phenol plus (Z)-3-hexen-1-ol attracted as many M. melolontha males as a mixture of (Z)-3-hexen-1-ol plus toluquinone. In both species three component mixtures containing phenol, (Z)-3-hexen-1-ol, and the respective benzoquinone did not capture more males than two component mixtures consisting of only (Z)-3-hexen-1-ol and the benzoquinone. A possible role of phenol as another cockchafer sex pheromone component is discussed.

Headspace volatiles from apple-bearing twigs were collected in the field with a Radiello sampler during three different diurnal periods over the complete fruit growing season. Analyses by thermal desorption-GC-MS identified a total of 62 compounds in changing quantities, including the terpenoids alpha-pinene, camphene, beta-pinene, limonene, beta-caryophyllene and (E,E)-alpha-farnesene, the aldehydes (E)-2-hexenal, benzaldehyde and nonanal, and the alcohol (Z)-3-hexen-1-ol. The variations in emission of these plant odours were statistically related to temperature, humidity and rainfall in the field. Remarkably, rainfall had a significant positive influence on changes in volatile release during all three diurnal periods, and further factors of significance were temperature and relative humidity around noon, relative humidity in the late afternoon, and temperature and relative humidity during the night. Rainfall was associated consistently with an increase in the late afternoon in terpene and aldehyde volatiles with a known repellent effect on the codling moth, one of the key pests of apple fruit. During the summer of 2003, a season characterized by below-average rainfall, some postulated effects of drought on trees were tested by establishing correlations with rainfall. Emissions of the wood terpenes alpha-pinene, beta-pinene and limonene were negatively correlated with rainfall. Another monoterpene, camphene, was only detected in this summer but not in the previous years, and its emissions were negatively correlated with rainfall, further supporting the theory that drought can result in higher formation of secondary metabolites. Finally, the two green leaf volatiles (E)-2-hexenal and (Z)-3-hexen-1-ol were negatively correlated with rainfall, coinciding well with the expectation that water deficit stress increases activity of lipoxygenase. To our knowledge, this work represents the first empirical study concerning the influence of abiotic factors on volatile emissions from apple trees in situ.

Subtle differences in pheromone components of sympatric species should be attractive only to the producing species and unattractive or repellent to the nonproducing species, and thereby maintain reproductive isolation and reduce competition between species. Bark beetles Dendroctonus brevicomis and D. frontalis (Coleoptera: Curculionidae) are known to have common pheromone components, except for exo-brevicomin, which is produced by D. brevicomis. We predicted that D. frontalis would not respond to exo-brevicomin outside of the zone of sympatry with D. brevicomis. We conducted a field experiment to determine the effect of exo-brevicomin on attraction of D. frontalis and associated species in Mississippi. We determined whether D. frontalis pheromone production differed inside and outside the sympatric zone and compared the pheromone profiles with D. brevicomis within the sympatric zone. Trapping studies revealed that D. frontalis can perceive and respond positively to exo-brevicomin, an aggregation pheromone of a sympatric congener (D. brevicomis), at locations hundreds of kilometers outside the sympatric zone. Qualitative pheromone profiles showed that both species emit similar pheromone components: frontalin, endo-brevicomin, exo-brevicomin, trans-verbenol, verbenone, and myrtenol. Although not previously reported, D. frontalis males from Arizona produced exo-brevicomin. The predator Thanasimus dubius did not discriminate traps baited with exo-brevicomin and was most attracted to traps with frontalin. Hylastes beetles were significantly attracted to traps baited with exo-brevicomin in combination with other compounds. Our results raise new practical and evolutionary questions on the role of exo-brevicomin in the behavioral ecology of D. frontalis. The addition of exo-brevicomin to the current lure might increase the efficiency of trapping programs in the southeastern United States.

Optimized trap lure for male Melolontha cockchafers
Abstract:  Melolontha cockchafer males search for mates using green leaf volatiles (GLV), released by host plants after female feeding. Thus, the feeding-induced plant volatiles act as sexual kairomones. Males of both Melontha hippocastani and Melontha melolontha are strongly attracted by the GLV ( Z )-3-hexen-1-ol ( Z -3-ol). Sex pheromones enhance the attractiveness of Z -3-ol and have been identified as toluquinone (TQ) in M. melolontha , and 1,4-benzoquinone (BQ) in M. hippocastani . Additionally, phenol acts as a male attractant in both species. From the perspective of potential application, we investigated by field experiments with volatile-baited traps the ways of enhancing the number of captured males by the use of specific binary or ternary blends of Z -3-ol with phenol, and TQ or BQ respectively. The data show that in both species binary lures containing Z -3-ol combined with TQ or BQ at a ratio of 10 : 1 are the most potent male attractants.

Peach shoot volatiles were attractive to mated female oriental fruit moth, Cydia molesta (Busck), in a dual choice arena. No preference was observed between leaf odours from the principle host plant, peach, and the secondary host plant, apple. Twenty-two compounds were identified in headspace volatiles of peach shoots using gas chromatography-mass spectrometry. Green leaf volatiles accounted for more than 50% of the total emitted volatiles. A bioassay-assisted fractionation using different sorbent polymers indicated an attractant effect of compounds with a chain length of 6-8 carbon atoms. The major compounds of this fraction were tested either singly or in combinations for behavioural response of females. Significant bioactivity was found for a three-component mixture of (Z)-3-hexen-1-yl acetate, (Z)-3-hexen-1-ol and benzaldehyde in a 4:1:1 ratio. This synthetic mixture elicited a similar attractant effect as the full natural blend from peach shoots as well as the bioactive fraction.

Green leaf volatiles (GLV), a series of saturated and monounsaturated six-carbon aldehydes, alcohols, and esters are emitted by plants upon mechanical damage. Evidence is increasing that intact plants respond to GLV by activating their own defense mechanisms, thus suggesting that they function in plant-plant communication. The present paper demonstrates that exposure of maize plants to naturally occurring GLV, including (Z)-3-, (E)-2- and saturated derivatives, induce the emission of volatile blends typically associated with herbivory. Position or configuration of a double bond, but not the functional group of the GLV influenced the strength of the emissions. (Z)-3-Configured compounds elicited stronger responses than (E)-2- and saturated derivatives. The response to (Z)-3-hexen-1-ol increased linearly with the dose between 200 and 1000 nmol per plant. Not only the naturally occurring (E)-2-hexenal, but also (E)-2-pentenal and (E)-2-heptenal induced maize plants, although to a lesser extent. Externally applied terpenoids [(3E)-4,8-dimethyl-1,3,7-nonatriene, beta-caryophyllene, and (E)-beta-farnesene] did not significantly increase the total amount of inducible volatiles in maize. Of three tested maize cultivars Delprim and Pactol responded much stronger than Attribut. Recovery experiments in the presence and absence of maize plants demonstrated that large proportions of externally applied GLV were assimilated by the plants, whereas (3E)-4,8-dimethyl-1,3,7-nonatriene was recovered in much higher amounts. The results furthermore suggested that plants converted a part of the assimilated leaf aldehydes and alcohols to the respective acetates. We propose that GLV not only can alert neighboring plants, but may facilitate intra-plant information transfer and can help mediate the systemic defense response in a plant.
