微生物许多非核糖体肽类次生代谢产物主要是由非核糖体肽合成酶(NRPS)催化合成。参考Gontang发布的非核糖体肽合成酶(NRPS)通用引物设计扩增NRPS腺苷酰化结构域基因序列的特异引物,从海洋链霉菌L1的基因组DNA中扩增获得一个715 bp的NRPS基因序列。测序结果及比对分析表明该片段属于NRPS腺苷酰化结构域部分序列。对其拟翻译的氨基酸序列组成成分、理化性质进行分析,显示其包含AFD class I超基因家族核心结合区,为NRPS腺苷酰化结构域(A结构域)所在区域。对氨基酸序列的二级结构预测和三级结构模拟,发现与数据库中肠菌素合酶F组分的结构相似。为后续研究A结构域的特异性及完整NRPS基因簇克隆提供了参考。  相似文献

根据不同物种间同一基因核苷酸序列的保守性及相似性的特点 ,在人和鼠的Pit 1基因第三外显子上设计上游引物 ,而将下游引物设计在猪Pit 1基因第四外显子上。利用聚合酶链式反应 (PCR)技术 ,扩增出猪Pit 1基因第三内含子序列 ,并经由酶切及序列同源性比对确认该序列即为猪Pit 1基因第三内含子序列。此序列的确定为下一步进行遗传变异分析的研究奠定了基础  相似文献

Sox9基因在人和多种动物中具有进化保守性,在性别决定过程中发挥着重要作用。通过对黄鳝Sox9基因的分子克隆及其限制酶切分析,确定了该克隆的限制酶图谱。从限制酶片段的PCR分析以及HMG盒的DNA序列测定,鉴定该阳性克隆为黄鳝Sox9基因克隆,黄鳝Sox9基因的克隆进一步说明Sox9基因在进行上的高度保守性,对其基因组DNA和cDNA序列结构和转基因黄鳝的深入研究,无疑有助于从进化上阐明Sox9基因功能。  相似文献

热不对称性PCR(thermal asymmetric interlaced PCR,TAIL-PCR)是一种用来分离与已知序列邻近的未知DNA序列的分子生物学技术.该技术利用3个根据已知序列设计的嵌套特异性引物分别和简并引物组合进行PCR反应,选择恰当退火温度对目标片段进行PCR扩增.TAIL-PCR技术作为一种使用技术简单易行,反应高效灵敏,产物特异性高,重复性好,能够在较短的时间内获得目标片段,已经在分子生物学研究领域广泛应用.本文从TAIL-PCR技术原理出发,对该技术特异性引物设计、随机引物组合选择、PCR反应条件等关键性问题进行综述,并介绍TAIL-PCR技术在植物基因克隆上的应用现状及发展前景.  相似文献

本研究通过设计引物进行PCR扩增得到绿色荧光蛋白GFP基因,将PCR产物酶切以后与芜菁花叶病毒表达载体相连,得到的阳性克隆通过多对引物组合进行PCR验证及序列测定,说明GFP基因已经连接到该病毒表达载体中,而且没有发生碱基误配及错配。该GFP表达载体的构建为进一步利用TuMv的全长cDNA侵染性克隆进行外源基因的转化奠定了基础。  相似文献

对来源于假单胞菌sp.130的戊二酰-7-氨基头孢烷酸(GL-7-ACA)酰化酶结构基因的全序列及所编码蛋白质的α,β亚基的N末端和C末端的氨基酸序列进行了测定。将蛋白质序列与其他同类的GL-7-ACA酰化酶进行了同源性比较,结果显示该酶与来源于假单胞菌GK16和C427的酰化酶的序列有较高同源性,而与其它同类酰化酶的同源性较低。这些酶的α亚基N-末端差别较大,但是β-亚基的N-末端有较高的保守性。  相似文献

Dm rt基因家族是一个与性别决定相关的基因家族。该家族成员都含有一个具有DNA结合能力的保守基序———DM结构域,在性别决定和分化发育的调控中担负着重要的功能。本文采用简并PCR技术,扩增和克隆了山地麻蜥(Erem ias breuchleyi)基因组中的DM结构域,通过SSCP技术筛选和测序得到了7个具有不同DM序列的克隆。结果显示,在山地麻蜥基因组中存在着Dm rt基因家族的多个成员,与其他动物相关的Dm rt基因进行聚类分析,显示该基因家族在动物系统进化上具有高度的保守性。  相似文献

从基因家族成员的视角开展基因表达研究是阐明基因功能的重要组成内容,实时定量PCR(QPCR)技术是分析基因表达的有效手段.以猕猴桃脂氧合酶(LOX)基因家族6个成员为对象,分析了引物特异性的检测方法.该方法整合了熔点曲线分析、琼脂糖电泳、交叉PCR扩增和PCR产物测序等分析手段,有效消除其他成员的交叉扩增干扰,为利用QPCR检测基因家族成员表达提供了特异、准确与可行的途径.  相似文献

多发性骨髓瘤细胞中一个表达上调基因的克隆与分析   总被引:1,自引:0,他引:1  
根据GenBank收录的多发性骨髓瘤细胞株 (ARH 77)表达上调ESTAF4 2 5 30 0设计引物 ,运用RT PCR检测了 5例多发性骨髓瘤患者及 4例正常人骨髓细胞中该EST的表达水平 .Northern印迹杂交分析该EST在多种组织中的表达 .进一步利用该EST作探针 ,筛选ARH 77cDNA文库 ,获得全长cDNA克隆 ,对该序列进行了分析 .结果显示 ,该EST在多发性骨髓瘤患者骨髓细胞中亦有较高的表达 ,而在正常人骨髓细胞中低表达 .经测序证实 ,该cDNA全长为 4 5 2bp(GenBank收录号 :AF4 87338) .预测其编码一个 5 7个氨基酸的小分子量蛋白质 ,属于与DNA复制有关的解旋酶 引物酶基因家族的新成员 .该基因在多发性骨髓瘤细胞中表达上调 ,其表达水平的改变可能与多发性骨髓瘤的发生与发展有关  相似文献

介绍从简单序列重复间区(ISSR)开发微卫星(SSR)引物的方法。该方法分两步:(1)正向引物的分离,即试材ISSR扩增,产物纯化回收并克隆至T载体,测序后设计正向引物(引物1)和巢式引物(引物2);(2)反向引物的分离,即基医l组DNA酶切并连接接头,结合抑制PCR技术,对产物克隆测序后设计反向引物。应用该方法前人已在多个物种中开发出多对SSR引物.  相似文献

It has now been over twenty years since a novel herpesviral genome was identified in Kaposi's sarcoma biopsies. Since then, the cumulative research effort by molecular biologists, virologists, clinicians, and epidemiologists alike has led to the extensive characterization of this tumor virus, Kaposi's sarcoma-associated herpesvirus(KSHV; also known as human herpesvirus 8(HHV-8)), and its associated diseases. Here we review the current knowledge of KSHV biology and pathogenesis, with a particular emphasis on new and exciting advances in the field of epigenetics. Curcumin is the yellow pigment of turmeric that interacts irreversibly forming an adduct with thioredoxin reductase (TrxR), an enzyme responsible for redox control of cell and defence against oxidative stress. Docking at both the active sites of TrxR was performed to compare the potency of three naturally occurring curcuminoids, namely curcumin, demethoxy curcumin and bis-demethoxy curcumin. Results show that active sites of TrxR occur at the junction of E and F chains. Volume and area of both cavities is predicted. It has been concluded by distance mapping of the most active conformations that Se atom of catalytic residue SeCYS498, is at a distance of 3.56 from C13 of demethoxy curcumin at the E chain active site, whereas C13 carbon atom forms adduct with Se atom of SeCys 498. We report that at least one methoxy group in curcuminoids is necessary for interation with catalytic residues of thioredoxin. Pharmacophore of both active sites of the TrxR receptor for curcumin and demethoxy curcumin molecules has been drawn and proposed for design and synthesis of most probable potent antiproliferative synthetic drugs.  相似文献

Comprises species occurring mostly in subtidal habitats in tropical, subtropical and warm-temperate areas of the world. An analysis of the type species, V. spiralis (Sonder) Lamouroux ex J. Agardh, a species from Australia, establishes basic characters for distinguishing species in the genus. These characters are (1) branching patterns of thalli, (2) flat blades that may be spiralled on their axis, (3) width of the blade, (4) primary or secondary derivation of sterile and fertile branchlets and (5) position of sterile and fertile branchlets on the thalli. Application of the latter two characters provides an important basic method for separation of species into three major groups. Osmundaria , a genus known only in southern Australia, was studied in relation to Vidalia , and its separation from the Vidalia assemblage is not accepted. Species of Vidalia therefore are transferred to the older genus name, Osmundaria. Two new species, Osmundaria papenfussii and Osmundaria oliveae are described from Natal. Confusion in the usage of the epithet, Vidalia fimbriala Brown ex Turner has been clarified, and Vidalia gregaria Falkenberg, described as an epiphyte on Osmundaria pro/ifera Lamouroux, is revealed to be young branches of the host, Osmundaria prolifera.  相似文献

Fifteen chromosome counts of six Artemisia taxa and one species of each of the genera Brachanthemum, Hippolytia, Kaschgaria, Lepidolopsis and Turaniphytum are reported from Kazakhstan. Three of them are new reports, two are not consistent with previous counts and the remainder are confirmations of very scarce (one to four) earlier records. All the populations studied have the same basic chromosome number, x = 9, with ploidy levels ranging from 2x to 6x. Some correlations between ploidy level, morphological characters and distribution are noted.  相似文献
