The morphology, distribution and function of nematocysts in the hydroid Halocordyle disticha were studied by light and scanning electron microscopy. Two types of stenoteles, two types of microbasic b-mastigophores (one with a capsular inclusion) and one type of desmoneme were identified. Prey-capturing experiments were performed with Artemia sp. nauplii. Stenoteles and microbasic mastigophores (those with inclusions) penetrated and paralyzed Artemia larvae, whereas desmonemes kept them in a firm grip by winding around setae.  相似文献

Glycerol-isolated basitrichs of Calliactis parasitica responsive to thioglycolate had open apical flaps, while unresponsive capsules isolated with Triton X100 or by freezing had closed apical flaps. Limited treatment with trypsin induced the apical flaps to open without causing discharge, suggesting that nematocysts can maintain the resting condition even with open flaps. Trypsin-treated basitrichs acquire a high responsiveness to thioglycolate. Microbasic mastigophores of Aiptasia mutabilis are more responsive to distilled water after controlled trypsin treatment but the apical flaps are unchanged. Ca2+ is inhibitory regardless of trypsin treatment. It is proposed that the capsule tip may control the penetration of the discharging agents rather than providing mechanical resistance to inner pressure.  相似文献

The cnidom of the sea anemone Sagartiogeton viduatus (Muller, 1776) is described from interference‐contrast light micrographs (LMs) and scanning electron micrographs (SEMs). Special attention is given to nematocyst maturation, including the differentiation of the shaft into proximal and main regions as helical folding of the shaft wall proceeds. Comparisons are made with Metridium senile (Linnaeus, 1761), whose cnidom, with a few exceptions, is closely similar to that of S. viduatus. The two anemones possess b‐ and p‐mastigophores, p‐amastigophores, isorhizas and spirocysts. Although the majority of cnidae in S. viduatus is smaller than corresponding ones in M. senile, they are grouped into the same size classes as those of M. senile, namely small, medium and large. The main differences from M. senile cnidae are the followings: (1) Large acontia p‐amastigophores are the largest nematocysts in S. viduatus. (2) They are noticeably larger than the large acontia b‐mastigophores, and (3) they are separated from the p‐amastigophores of M. senile by the sinusoid pattern of their U‐shaped capsular matrix. (4) The large acontia b‐mastigophores are microbasic and not mesobasic as in M. Senile, and (5) they do not produce darts. (6) Another difference from M. senile is the absence of catch‐tentacle isorhizas.  相似文献

The qualitative and quantitative carotenoid composition is reported for (i) a red and a green strain of Oscillatoria limnetica and a green strain of Spirulina platensis cultivated under identical conditions and (ii) a red and a green strain of Spirulina subsalsa grown under identical conditions. No correlation between colour and carotenoid content was obtained. However, differences in carotenoid composition within the Oscillatoria and Spirulina strains were observed. Both oscillol diglycoside ex Oscillatoria limnetica and myxol glycoside ex Spirulina platensis were mixed α-glycosides with chinovose-fucose present in ca 7:2 ratio. Analytical procedures are given. The chemosystematic implications for the current taxonomy of the genera Oscillatoria and Spirulina are discussed.  相似文献

The equilibrium moisture contents of sun-cured (Kroumougrad), flue-cured (Bright Yellow—4) and air-cured (Burley-21 and Matsukawa) tobaccos were measured over a relative humidity range from 5 to 80% at 20°C. The moisture sorption isotherms of tobaccos were of sigmoid type, and classified into two groups. In a lower humidity range below ca. 40% RH, the A group (Kroumougrad and BY-4) had a smaller moisture sorption capacity than B group (Burley-21 and Matsukawa), while in a higher humidity range above ca. 50% RH the former had a larger moisture sorption capacity than the latter. By extracting with water, the moisture content of BY-4 was increased in the lower humidity range, while it decreased in the higher humidity range. However, the moisture content of Matsukawa was scarecely changed by extracting it with water. These results suggest that the differences in equilibrium moisture content with the type of curing were due to the differences in contents of water soluble com- ponents. To control the hygroscopic properties of a tobacco, therefore, the influences of the addition of sucrose and glycerol on the equilibrium moisture content were quantitatively analysed. The moisture sorption capacity of tobacco was greatly different from its nitrogen sorption capacity. The specific surface area of tobacco calculated from moisture sorption isotherm was ca. 110 times larger than the specific surface area calculated from the nitrogen sorption isotherm. Both the nitrogen and moisture sorption data should be necessary for better understanding of the complicated sorption-desorption phenomena in tobaccos.  相似文献

In culture fluid, Klebsiella pneumoniae type 1 Kasuya strain produces polysaccharide exhibiting a strong adjuvant effect. The active substance responsible for the strong adjuvant effect of the polysaccharide is not its acidic polysaccharide fraction (the type-specific capsular antigen) but the neutral polysaccharide fraction. In the present study, a mutant which did not produce the type-specific capsular polysaccharide was isolated from ultraviolet-irradiated cells of K. pneumoniae type 1 Kasuya strain which had been labeled with leucine-requiring marker by selecting unagglutinable cells with the antiserum to the type-specific capsular polysaccharide. Serological tests showed that the type-specific acidic capsular polysaccharide was present neither on the cells surface nor in the culture fluid of the mutant. Electron microscopically, the mutant did not possess any capsular material. On the other hand, nearly an equal amount of neutral polysaccharide antigen was produced in culture fluids of the noncapsulated mutant and the parent strain. The neutral polysaccharide antigen produced by the noncapsulated mutant exhibited the same degree of strong adjuvant effect on antibody response to bovine gammaglobulin in mice as that produced by the parent strain. The relationship between the neutral polysaccharide antigen in culture fluid and the O antigen of K. pneumoniae was discussed.  相似文献

Palmaria decipiens, one of the most abundant red seaweeds of the chilean Antarctic, was collected in King George Island. The hot water extract (26% yield) showed by acid hydrolysis to contain xylose, galactose and traces of glucose. Fractionation with cetrimide gave a soluble neutral xylan and an insoluble fraction. The insoluble fraction afforded an acidic polysaccharide that contained 4.8% of uronic acids, 2.8% of sulfate and 18.9% of protein. Polyacrylamide gel electrophoresis showed that it was homogeneous. The GLC and HPLC analysis of the total acidic hydrolysis products showed that the acidic polysaccharide was composed of the neutral sugars galactose and xylose in the molar ratio 8.2:1.0 and of galacturonic and glucuronic acid in the ratio 1.5:1.0. The second-derivative FT-IR spectrum showed the characteristic amide I, II and III bands of proteins. Alkaline cleavage with 0.1 M NaOH indicated the presence of a glycoprotein with O-glycosidic linkage.Results found in this work suggest that the acidic polysaccharide extracted from Palmaria decipiens is an acidic xylogalactan-protein complex.  相似文献

Cystocarpic, tetrasporic and vegetative thalli of Gelidium chilense were extracted with water at 95 °C. The contents of 3,6-anhydro-galactose and sulfate group of the hydrocolloids correspond to those of an agar-type polysaccharide. The percentages of 6-O-methyl-galactose and of pyruvic acid ranged between 5.7–6.2% and 0.42–0.54%, respectively. The gel melting and gelation temperatures of Gelidium chilense, G. rex and G. lingulatum agars were determined. A correlation between 6-O-methylgalactose content and gelation temperatures was not observed. It was found by anion-exchange chromatography that 19.8% of tetrasporic and 4.9% of vegetative G. chilense agars are unsulfated polymers. Structural studies on the neutral fraction from tetrasporic G. chilense agar by partial hydrolysis and 1H NMR spectroscopy have shown that it is mainly composed of agarose. Methylation analysis, oxidative hydrolysis and partial hydrolysis, followed by 1H NMR spectroscopy, have shown that the neutral fraction of the agar from tetrasporic Gelidium rex is agarose. The results obtained in this work are compared with previously reported data on studies of agars from Chilean Gelidium species.  相似文献

Remodelling of a smoothly tapering internal tubule to form the enlarged shaft of microbasic mastigophores of the siphonophore Rosacea takes place very late in development. It occurs after deployment of the nematocyst in its final position, and after the pleats of the internal tubule have been completely obliterated in the shaft region by the growth of spine material. The optical and mechanical properties of the internal tubule at the earlier (bedspring) stage give no hint that a shaft will develop. The possible ways in which remodelling might occur inside the capsule and remote from the cytoplasm are discussed.  相似文献

The calcium-binding vesicles from the green alga Mougeotia scalaris were isolated and characterized after staining in vivo by neutral red or rhodamine B. They were found to possess, a protonated group with a pKa-9.9, typifying phenolic hydroxyl groups; upon titration, both, phenolic compound(s) and vital dye were concomitantly released from the vesicular matrix. A shift in peak absorbance from 450 nm to 540 nm of the vitally stained vesicles indicated that the neutral form of neutral red was bound to the vesicular, matrix as an intermediate form, stabilized via intermolecular hydrogen bonds to the phenolic compound(s). Up to 8.5.109 dye molecules were calculated to be adsorbed to a mean-size vesicle. Analysis of Langmuir adsorption isotherms, indicated that there were two binding sites each for both neutral red and rhodamine B. The isolated vesicles were devoid of calcium, probably because vesicular calcium, bound to the vesicle matrix, was displaced upon dye binding. Dye adsorption to the vesicles in vivo results in substantial inhibition of the reorientational movement of the Mougeotia chloroplast and is explained by dye-mediated disorder of the cellular calcium homoeostasis.Abbreviations NR neutral red - RB rhodamine B - SDS sodium dodecyl sulfate This paper is part of the Ph.D. thesis of F. Haemophilus influenzae type b meningitis has been reported predominantly among infants who have not completed the primary immunization series, whereas infection by capsular f has been reported mostly in patients with significant underlying diseases. The present study analyses H. influenzae type b & f infection among 25 unvaccinated patients with respiratory tract infections and symptoms of meningitis by PCR. 5/7 isolates from CSF sample were capsular type b; whereas capsular type f was detected in 2/10 and 5/8 isolates from throat swab and sputum samples, respectively. Rest 13 strains were negative for both the capsular types. To conclude, in India where H. influenzae type b conjugate vaccine is not included in national immunization program, individuals with respiratory tract infections and meningitis are at a greater risk of developing infection with H. influenzae type b strains along with other capsular types and non-capsular types.  相似文献

An intertidal population of Iridaea capensis in the south-western Cape Province of South Africa has been studied in detail for a year. The species is generally restricted to sand-inundated rocky shores, which appears not to be the case for Iridaea species in the Pacific. In early winter (June) biomass, plant density and plant size were at a minimum. A small increase in these parameters occurred in August, but there was a massive increase in plant numbers and biomass in October. The largest plants were recorded in summer (February). The population was over 70% sporophytes in April, but switched to almost 80% gametophytes following heavy recruitment between August and October, and remained significantly gametophyte-dominated through April of the following year. Carrageenan levels were high (ca 42% of dry weight) in late winter-spring, and lowest (ca 30–35%) in summer-autumn. Strong offshore wind conditions in early summer caused a reduction in biomass, and it is hypothesized that the associated upwelling of nutrient rich water may be implicated in the lower carrageenan levels at this time.  相似文献

The rough bark of orchard trees (Malus) around Darmstadt is predominantly covered in red to purple‐brown layers (biofilms) of epiphytic terrestrial alga of Trentepohlia umbrina. The smooth bark of forest trees (Fagus sylvatica L. and Acer sp.) in the same area is covered by bright green biofilms composed of the green algae Desmococcus, Apatococcus and Trebouxia, with a few cells of Coccomyxa and ‘Chlorella’ trebouxioides between them. These algae are desiccation tolerant. After samples of bark with the biofilms were kept in dry air in darkness for various periods of time, potential quantum yield of PSII, Fv/Fm, recovered during rehydration upon rewetting. The kinetics and degree of recovery depended on the length of time that the algae were kept in dry air in the desiccated state. Recovery was better for green biofilm samples, i.e. quite good even after 80 days of desiccation (Fv/Fm = ca. 50% of initial value), than the red samples, where recovery was only adequate up to ca. 30–40 days of desiccation (Fv/Fm = ca. 20–55% of initial value). It is concluded that the different bark types constitute different ecophysiological niches that can be occupied by the algae and that can be distinguished by their capacity to recover from desiccation after different times in the dry state.  相似文献

Isolates belonging to an undescribed Phytophthora species were frequently recovered during an oak forest soil survey of Phytophthora species in eastern and north-central USA in 2004. The species was isolated using an oak leaf baiting method from rhizosphere soil samples collected from Quercus rubra, Q. macrocarpa, and Q. phellos. This species is formally described as P. quercetorum. It is homothallic and has aplerotic oogonia and paragynous antheridia. It produces papillate sporangia (occasionally bipapillate) of ovoid-elongated shapes. Its temperature optimum for growth is ca 22.5 °C with the upper limit of ca 32.5 °C. P. quercetorum differs from the morphologically related P. quercina in producing distinct submerged colony-patterns, different growth-temperature requirements, and oogonial shapes and sizes. Phylogenetic analyses using seven nuclear loci supported P. quercetorum as a novel species within clade 4, closely related to P. arecae, P. palmivora, P. megakarya, and P. quercina.  相似文献

We checked the hypothesis that the transient presence of anthocyanins in young leaves serves a photoprotective function. For this purpose, Rosa sp. and Ricinus communis L., whose young leaves are red to become green upon maturation, were used. Thus, young leaves with high and mature leaves with low anthocyanin contents were analysed concerning their carotenoid (Car) composition and susceptibility to photoinhibition. Cars, including the components of the xanthophyll cycle, had similar contents in young and mature leaves, when expressed on a chlorophyll basis. Yet, when expressed on a leaf area basis or on the assumed photon absorptive capacity of leaves, Cars contents were considerably lower in anthocyanic young leaves. Although this may indicate a low photodissipative potential, red young leaves were considerably less susceptible to photoinhibitory damage. The results are compatible with a photoprotective function of anthocyanins, indicating also that their presence may compensate for a low capacity in the xanthophyll cycle-dependent harmless dissipation of excess excitation energy.  相似文献

The neutral red retention assay was adapted to assess the effects of a matrix of temperature and salinity combinations on haemocytes of the European flat oyster Ostrea edulis (L.). Oysters were collected from a commercially fished site in the western Solent and were acclimated to different temperature and salinity combinations in the laboratory. The results indicate that the haemocytes are most stable at high salinity and intermediate temperature (28‰/15°C). Furthermore it is evident from the data that at low salinities the adaptive capacity of the cellular processes is reduced to such an extent that the haemocytes are unable to respond to favourable temperature regimes. The use of neutral red as a probe to monitor cellular stability is discussed.  相似文献

山羊支原体山羊肺炎亚种(Mycoplasma capricolum subsp. capripneumoniae, Mccp)是山羊传染性胸膜肺炎(contagious caprine pleuropneumonia, CCPP)的病原,可用灭活疫苗和荚膜多糖(capsular polysaccharide, CPS)间接血凝试剂进行预防和血清学检测,但高昂的培养成本和复杂的抗原定量一直困扰着生产人员。为解决生产实际中出现的这些问题,本研究基于Mccp代谢组学的前期理论基础,通过改变初始pH值的方法,初步筛选出初始pH值为7.8的可以同时提高2种抗原产量的糖发酵培养基。利用紫外可吸收光谱可识别酚红,以及十六烷基三甲基溴化铵(cetyltrimethylammonium bromide, CTAB)可与阴离子荚膜多糖结合的理论依据,建立了利用紫外光谱分析Mccp达到的培养阶段,以及利用CTAB沉淀法相对定量发酵液荚膜多糖抗原产量的方法。通过紫外图谱观察的方法可对应Mccp生长曲线进行指导生产,大大节省传统颜色变化单位(color change unit, CCU)法的监测时间,提高了原肉眼观察方法的精确度。建立的CTAB沉淀法可在5 h内完成对CPS含量的监测,与传统的差值法相比大大缩短了时间,并且其准确度得到苯酚-硫酸法的验证。本研究优化的一种培养基和建立的两种相关性比较方法,可有效降低Mccp生产成本,提高生产效率,这些方法已在本实验室的研究阶段得到应用,为进一步改进CCPP灭活疫苗和荚膜多糖的生产工艺以及快速定量提供了实验数据。  相似文献

Recognizing that it takes Acanthoscelides obtectus (Say)(Coleoptera: Bruchidae) larvae over 24 h to bore into a dry red kidney bean (Phaseolus vulgaris L.) and that boring can occur only at particular sites where a bean abuts some other surface, we postulated that this pest might be controlled by periodic tumbling of beans so as to place initiated holes out of register with requisite touching surfaces. Larvae repeatedly forced to initiate holes might die of exhaustion, if not smashed by tumbling beans. Indeed, brief daily tumbling of beans held in half-filled jars, buckets, and gunny sacks reduced A. obtectus populations by 97% relative to stationary controls. We recommend that small lots of beans can be protected indefinitely from the bean weevil when stored in <75% filled cylinders rolled ca 1 circumference every morning and evening. This control method should be immediately useful, particularly among subsistence families, since the only external input is knowledge. Also, this control principle might be broadly applicable to other bean and grain pests having strict spatial and temporal requirements for seed penetration.  相似文献

Till about 1965 a rich characean community occurred in the shallow peat lake Botshol with six species of which the rare Nitellopsis obtusa and Chara major dominated at many sites. In the period 1980–1988 characean biomass strongly decreased and only two species, Chara globularis and C. connivens, remained in small populations at a few localities. Of the macrophyte Najas marina also some small populations remained, while the aquatic moss Fontinalis antipyretica and the filamentous alga Vaucheria dichotoma predominated at many sites. These phenomena may have been due to eutrophication by the input of polluted water. This process of impoverishment was stopped by restoration measures in 1989, resulting in a lower phosphorus concentration (ca 25 µg l-1) and a higher water transparency. Immediately after these measures the Characeae community increased in abundance and number of species. During the summer of 1990, and especially 1991, a spectacular growth occurred of Chara connivens. C. connivens was often accompanied by C. major. Other species with scattered occurrence were C. aculeolata, C. aspera, C. contraria and C. globularis. The reasons for the shift in dominance from Nitellopsis obtusa to Chara connivens are discussed. From growth experiments evidence was obtained that neither the recent higher chloride level, nor the lowered phosphate concentration were the main factors for the domination of Chara connivens.  相似文献
