Biopharmaceutical and biotechnology companies and regulatory agencies require novel methods to determine the structural stabilities of proteins and the integrity of protein-protein, protein-ligand, and protein-membrane interactions that can be applied to a variety of sample states and environments. Infrared spectroscopy is a favorable method for a number of reasons: it is adequately sensitive to minimal sample amounts and is not limited by the molecular weight of the sample; yields spectra that are simple to evaluate; does not require protein modifications, a special supporting matrix, or internal standard; and is applicable to soluble and membrane proteins. In this paper, we investigate the application of infrared spectroscopy to the quantification of protein structural stability by measuring the extent of amide hydrogen/deuterium exchange in buffers containing D2O for proteins in solution and interacting with ligands and lipid membranes. We report the thermodynamic stability of several protein preparations, including chick egg-white lysozyme, trypsin bound by benzamidine inhibitors, and cytochrome c interacting with lipid membranes of varying net-negative surface charge density. The results demonstrate that infrared spectroscopy can be used to compare protein stability as determined by amide hydrogen/deuterium exchange for a variety of cases.  相似文献

Peak area matching and linear regression were used to quantify eight chlorinated pesticides and polychlorinated biphenyls (as Aroclor 1260) in human serum. There are no statistically significant differences in data obtained by these two quantifying techniques which were indicated by the paired t-test. For chlorinated pesticides, p = 0.053-0.62, and for polychlorinated biphenyls (as Aroclor 1260), p = 0.64. Analyte residues for the chlorinated pesticides ranged from 0.5 ppb for hexachlorobenzene (HCB) to 186 ppb for dichlorodiphenyldichloroethylene (DDE). Analyte residues for the polychlorinated biphenyls (as Aroclor 1260) ranged from 5-114 ppb. The absolute mean percent difference between the two quantifying techniques ranged from 0.06% for DDE to 8.06% for dieldrin (HEOD) among the chlorinated pesticides. The absolute mean percent difference between the two quantifying techniques for the polychlorinated biphenyls (as Aroclor 1260) was 3.4%. Peak area matching and linear regression were found to be comparable for quantifying these environmental residues in serum when the following conditions apply: 1) the concentration of the chlorinated pesticides is greater than or equal to 0.5 ppb (e.g., HCB, hexachlorocyclohexane (HCCH), oxychlordane (OC), heptachlor epoxide (HE), transnonachlor (TN), HEOD, and dichlorodiphenyltrichloroethane (DDT); 2) the concentration of the chlorinated pesticide is greater than or equal to 3 ppb (e.g., DDE); and 3) the total concentration of polychlorinated biphenyls (e.g., as Aroclor 1260) is greater than or equal to 5 ppb.  相似文献

Animals emit visual signals that involve simultaneous, sequential movements of appendages that unfold with varying dynamics in time and space. Algorithms have been recently reported (e.g. Peters et al. in Anim Behav 64:131–146, 2002) that enable quantitative characterization of movements as optical flow patterns. For decades, acoustical signals have been rendered by techniques that decompose sound into amplitude, time, and spectral components. Using an optic-flow algorithm we examined visual courtship behaviours of jumping spiders and depict their complex visual signals as “speed waveform”, “speed surface”, and “speed waterfall” plots analogous to acoustic waveforms, spectrograms, and waterfall plots, respectively. In addition, these “speed profiles” are compatible with analytical techniques developed for auditory analysis. Using examples from the jumping spider Habronattus pugillis we show that we can statistically differentiate displays of different “sky island” populations supporting previous work on diversification. We also examined visual displays from the jumping spider Habronattus dossenus and show that distinct seismic components of vibratory displays are produced concurrently with statistically distinct motion signals. Given that dynamic visual signals are common, from insects to birds to mammals, we propose that optical-flow algorithms and the analyses described here will be useful for many researchers.Damian O. A challenging topic in undergraduate physiology courses is the complex interaction between the vertebrate endocrine system and the immune system. There are relatively few established and accessible laboratory exercises available to instructors to help their students gain a working understanding of these interactions. The present laboratory module was developed to show students how glucocorticoid receptor activity can be pharmacologically modulated in Xenopus laevis tadpoles and the resulting effects on thymus gland size visualized and quantified in vivo. After treating young tadpoles with a cortisol receptor agonist (dexamethasone) for 1 wk, students can easily visualize the suppressive effects of glucocorticoids on the intact thymus gland, which shrinks dramatically in size in response to this steroid hormone analog. However, the suppressive effect of dexamethasone is nullified in the presence of the glucocorticoid receptor antagonist RU-486, which powerfully illustrates the specific effects of glucocorticoid receptor inhibition on the immune system. Image analysis and statistics software are used to quantify the effects of glucocorticoid modulation on thymus size.  相似文献

Conservation efforts to better understand how wildlife populations respond to environmental change and anthropogenic disturbance has led to a proliferation of research examining physiological indicators of stress response in wildlife. Glucocorticoid stress hormones (GCs), typically cortisol and corticosterone, are among the most frequently measured indicators of the vertebrate stress response. To review the current state of research on stress physiology of free-ranging animals and its application to conservation, we canvassed more than 1000 articles on GC measures in wildlife published since 1969. For 454 studies published since 1990, we assessed the most commonly analysed correlates and disturbances and conducted a meta-analysis on commonly studied species. We noted a prominent divide in the legacies of fish-related analyses and those of higher order vertebrates and the need and opportunity to transfer knowledge between fields. Fish studies most frequently measured physiological indicators, condition, and the relationship between stress and mortality, whereas other vertebrate studies most frequently measured reproduction, condition, and environmental correlates. Correlates that significantly influenced GC levels across all vertebrate groups and are thus important to control for in study design and analyses include density and dispersal of conspecifics, season, reproductive status, and social status. Consistent trends across commonly studied species included positive GC response to capture and handling, reduced GC response related to acclimation, and a lack of correlation between condition and baseline GC levels. Our synthesis within and across diverse taxonomic orders reveals substantial research coverage but a lack of depth in multivariate analyses and a disparity in how correlates are controlled. This paper provides a comprehensive assessment of correlates and disturbances that influence GC measures and, as such, has useful applications to assist conservation physiologists in study design, analysis, and interpretation.  相似文献

Interest in the field of neuroimmunoendocrinology is in full expansion. With regard to this, steroid influence on the immune system, in particular sex steroids and glucocorticoids, has been known for a long time. Sex steroids are part of the mechanism underlying the immune sexual dimorphism, as particularly emphasized in autoimmune diseases. Immunosuppressive and anti-inflammatory effects of glucocorticoids are now considered a physiological negative feedback loop to cytokines produced during an immune and/or inflammatory response. Psychosocial factors may play a role in the development of immunologically-mediated diseases, e.g. autoimmune diseases. The nonobese diabetic (NOD) mouse, that develops an immunologically-mediated insulin-dependent diabetes mellitus (IDDM) is an interesting model to study the role of endogenous steroids. Insulitis is present in both sexes, but diabetes has a strong preponderance in females. Hormonal alteration, such as castration, modulates the incidence of diabetes, whereas environmental factors, such as stress, accelerate the disease. In the present paper, we have reviewed the role of gender, sex steroid hormones, stress and glucocorticoids in autoimmunity as well as analyzed their different levels of actions and interrelationships, focusing particular attention on the immunologically-mediated IDDM of the NOD mouse.  相似文献

Proteins damaged by stressors such as heat, oxidizing conditions or toxic agents are deleterious to cells and must be properly taken care of. Accordingly, misfolded proteins trigger a cellular stress response that aims to either repair defective polypeptides or eliminate faulty elements when salvage is not possible. This stress response provides time for additional stressor-specific pathways that adapt the cell to the changed environment if necessary. Recent studies have investigated how proteins that frustrate the folding machinery are recognized and cleared from the cell. Surprisingly, these clearance mechanisms are not restricted to the protein level. The stress response can also eliminate the mRNA of polypeptides that are refractory to folding.  相似文献

The intravenous injection of cholera toxin in rats 17 h prior to experimentation results in increased levels of insulin and corticosterone in the blood. This is accompanied by a rise in lipoprotein lipase activity in muscle and a decrease in adipose tissue. Pre- and postheparin blood levels of the enzyme are increased, representing the higher overall muscle activity. Hepatic lipase is decreased by cholera toxin treatment. These enzyme changes are accompanied by increased levels of non-esterified fatty acids, ketone bodies and unesterified cholesterol in the blood, whereas triacylglycerol levels are lowered. The lipoprotein triacylglycerol secretion is not affected by cholera toxin, suggesting increased triacylglycerol removal from the blood. On the other hand the unesterified cholesterol removal may be decreased due to the decreased hepatic lipase activity. Administration of excess glucocorticoid 2 days prior to blood and tissue sampling also resulted in a rise in lipoprotein lipase, a decrease in hepatic lipase activity and an increase of non-esterified fatty acids. In contrast to the effect of cholera toxin, the triacylglycerol levels were increased. Adrenalectomy, whether by inhibition of 11-beta-steroid hydroxylase or by surgical intervention, did not abolish the choleratoxin effects. It is concluded that corticosterone increase is not essential to the cholera toxin effects. Corticosterone itself probably causes an increase of cyclic AMP and/or Ca2+ levels, as is discussed.  相似文献

Animal movements in the landscape are influenced by linear features such as rivers, roads and power lines. Prior studies have investigated how linear features, particularly roads, affect movement rates by comparing animal's movement rate measured as step lengths (i.e., the distance between consecutive observations such as GPS locations) before, during and after crossing of a linear feature. The null hypothesis has been that the length of crossing steps should not differ from other steps, and a deviation from this, mainly that steps are longer during crossing, has been taken as support for a disturbance effect of the linear feature. However, based on the simple relationship between the length of a step and its probability to cross a linear feature, we claim that this assumption is inappropriate to test for behavioural responses to linear features. The probability is related to the proportion of the total length of the trajectory (i.e., the path of movement) a step constitutes. Consequently, care should be taken when formulating hypotheses about how animal moves in relation to linear features in the landscape. Statistical tests should be set up with respect to the expected length based on the distribution of step lengths in the trajectory. We propose two methods that accounts for the bias in crossing frequency that is caused by step lengths, and illustrates their applications by using simulated animal trajectories as well as empirical data on reindeer in an area with a power line.  相似文献

Objective and reliable assessment of animal colouration is of great value to workers in the biological sciences. We present the results of a comparison between two colour measurement techniques: Munsell colour standard matching and spectroradiometry. As part of ongoing research on sexual selection in red jungle fowl ( Gallus gallus ), eather and comb colour of 49 roosters was measured using both techniques. Previous research showed that hens use variation in feather and comb colour in mate choice, and we allowed hens to choose between paired roosters. Colour matching and spectroradiometry scores were generally correlated, but spectroradiometry was more sensitive in detecting variation and also provided a better estimate of the role of secondary sexual characters in male mating success.  相似文献

Introduction

Studies in the spontaneous ankylosis model in aging male DBA/1 mice and in patients with ankylosing spondylitis provide evidence that inflammation and new tissue formation leading to joint or spine ankylosis are likely linked but largely uncoupled processes. We previously proposed the ''entheseal stress'' hypothesis that defines microdamage or cell stress in the enthesis as a trigger for these disease processes. Here, we further investigated the relationship between inflammation and ankylosis by focusing on the early phase of the spontaneous arthritis model.

Methods

Aging male DBA/1 mice from different litters were caged together at the age of ten weeks and studied for signs of arthritis. A group of DBA/1 mice were treated daily with dexamethasone (0.5 μg/g body weight). Severity of disease was assessed by histomorphology and by positron emission tomography (PET) using 2-[18F]fluoro-2-deoxy-D-glucose (18F-FDG) as a tracer. Bone loss in dexamethasone-treated or control mice was determined by in vivo dual-energy X-ray absorptiometry. Chemokine gene expression was studied ex vivo in dissected paws and in vitro in mesenchymal cells (periosteal and bone marrow stromal cells) by quantitative real-time PCR in the presence or absence of bone morphogenetic protein 2 (BMP2) and dexamethasone.

Results

Dexamethasone treatment did not affect incidence or severity of ankylosis, but led to an expected reduction in inflammation in the paws at week 15 as measured by PET tracer uptake. Treatment with dexamethasone negatively affected bone mineral density. Chemokines attracting neutrophils and lymphocytes were expressed in affected paws. In vitro, BMP2 stimulation upregulated chemokines in different mesenchymal joint-associated cell types, an effect that was inhibited by dexamethasone.

Conclusions

BMP signaling may be a trigger for both inflammation and ankylosis in the spontaneous model of ankylosing enthesitis. The lack of inhibition by glucocorticoids on new bone formation while causing systemic bone loss highlights the paradoxical simultaneous loss and gain of bone in patients with ankylosing spondylitis.  相似文献

Mycoplasma, the smallest self-replicating life-forms, are responsible for a variety of diseases in humans, domestic animals, insects, and plants. Although the exact role of these organisms in wildlife is not available, now it is increasingly recognized that mycoplasmosis is an emerging disease in many wildlife populations. As only scattered data regarding these are available, an attempt has been made to assemble the available literature. To our knowledge, this is the first review paper regarding mycoplasmosis in wildlife. The outbreaks of mycoplasmoses such as contagious pleuropneumonia (CCPP), infectious keratoconjunctivitis (IKC), Mycoplasma ovipneumoniae and Mycoplasma capricolum subspp. capricolum infections, and contagious agalactia in wild ruminants; mycoplasmosis in raccoons; upper respiratory tract disease (URTD) and Mycoplasma crocodyli and Mycoplasma alligatoris infections in wild reptiles; and house finch conjunctivitis are showing that the prevalence of mycoplasmosis in wildlife has reached an alarming level, thus raising concerns for the conservation of many endangered species in captivity and free-ranging populations. These also emphasize the urgent need for increased surveillance and control of mycoplasma in wildlife. Some control approaches that can be followed are prevention of the spillover from domestic livestock and birds, proper quarantine of newly received animals along with mycoplasma screening, isolation of affected and suspected animals, screening of mycoplasmoses before the release of captive animals into free-ranging population, etc. At the same time, the previous dogma that mycoplasma have strict host specificity should be reconsidered in the light of new findings.  相似文献

Prolonged high secretion of glucocorticoids normally reflects a state of chronic stress, which has been associated with an increase in disease susceptibility and reduction in Darwinian fitness. Here, we hypothesize that an increase in oxidative stress accounts for the detrimental effects of prolonged high secretion of glucocorticoids. We performed a meta-analysis on studies where physiological stress was induced by administration of glucocorticoids to evaluate the magnitude of their effects on oxidative stress. Glucocorticoids have a significant effect on oxidative stress (Pearson r = 0.552), although this effect depends on the duration of treatment, and is larger in long-term experiments. Importantly, there was a significant effect on tissue, with brain and heart being the most and the least susceptible to GC-induced oxidative stress, respectively. Furthermore, effect size was larger (1) in studies using both sexes compared to males only, (2) when corticosterone rather than dexamethasone was administered and (3) in juveniles than in adults. These effects were not confounded by species, biochemical biomarker, or whether wild or laboratory animals were studied. In conclusion, our meta-analysis suggests that GC-induced oxidative stress could be a further mechanism underlying increases in disease susceptibility and decreases in Darwinian fitness observed under chronic stress.  相似文献

Mounting evidence suggests that acute and chronic stress, especially the stress-induced release of glucocorticoids, induces changes in glutamate neurotransmission in the prefrontal cortex and the hippocampus, thereby influencing some aspects of cognitive processing. In addition, dysfunction of glutamatergic neurotransmission is increasingly considered to be a core feature of stress-related mental illnesses. Recent studies have shed light on the mechanisms by which stress and glucocorticoids affect glutamate transmission, including effects on glutamate release, glutamate receptors and glutamate clearance and metabolism. This new understanding provides insights into normal brain functioning, as well as the pathophysiology and potential new treatments of stress-related neuropsychiatric disorders.  相似文献

Stress relaxation (or equivalently creep) allows a large range of the relaxation (retardation) spectrum of materials to be examined, particularly at lower frequencies. However, higher frequency components of the relaxation curves (typically of the order of Hertz) are attenuated due to the finite time taken to strain the specimen. This higher frequency information can be recovered by deconvolution of the stress and strain during the loading period. This paper examines the use of three separate deconvolution techniques: numerical (Fourier) deconvolution, semi-analytical deconvolution using a theoretical form of the strain, and deconvolution by a linear approximation method. Both theoretical data (where the exact form of the relaxation function is known) and experimental data were used to assess the accuracy and applicability of the deconvolution methods. All of the deconvolution techniques produced a consistent improvement in the higher frequency data up to the frequencies of the order of Hertz, with the linear approximation method showing better resolution in high-frequency analysis of the theoretical data. When the different deconvolution techniques were applied to experimental data, similar results were found for all three deconvolution techniques. Deconvolution of the stress and strain during loading is a simple and practical method for the recovery of higher frequency data from stress-relaxation experiments.  相似文献

It is well known that the release of glucocorticoids from the adrenal gland is increased in response to many types of stressors and plays a principal role in stress responses. We have shown that the synthesis of prostaglandins (PGs) in the brain is increased under several stress conditions including immobilization (IMO), and that endogenous glucocorticoids counteract this stress-induced PG synthesis. It was also recently reported that IMO damages dopaminergic (DA) neurons in the substantia nigra (SN), which is known to cause symptoms similar to Parkinson's disease (PD). The present study was therefore undertaken to determine the role of glucocorticoids in modulating the signs of PD induced by IMO. The pole test, in which each mouse was placed head upward at the top of a pole and the time taken to turn downward and to arrive on the floor was recorded, and immunohistochemistry for tyrosine hydroxylase (TH) in the SN were performed to evaluate bradykinesia and injury of DA neurons, respectively. Intact and adrenalectomized (ADX) mice were immobilized for 2 h twice, 1 day apart. Both bradykinesia and a decrease in the number of TH-immunoreactive cells in the SN were observed in ADX mice, but not in intact mice, following IMO. These effects of IMO on ADX mice were restored by treatment with corticosterone or indomethacin, a PG synthesis inhibitor. These results suggest that glucocorticoids play a role in preventing the detrimental effect of IMO on nigral DA neurons and resulting bradykinesia, and that this effect of IMO involves PG-mediated mechanisms.  相似文献
