Abstract

Water availability is one of the most critical factors that limits the growth and development of plants. Salicylic acid (SA) is an important signal molecule that modulates plant responses to abiotic stress. To elucidate the regulating mechanism of exogenous SA on Torreya grandis cv. Merrillii under different water stresses, a pot experiment was conducted in a greenhouse. Exposure of T. grandis seedlings to drought conditions resulted in reduced growth rate that was associated with a decline in water content and CO2 assimilation. Foliar application of SA effectively increased the water content, net CO2 assimilation rate, proline content and antioxidant enzymes activity in the plants, which helped T. grandis to acclimate to moderate drought stress and increase the shoot dry matter. However, when the plants were under severe drought stress, the relative water content and CO2 assimilation in the SA-treated plants were significantly lower than those in the control plants. Therefore, our results indicated that SA can effectively ameliorate the negative effect of moderate drought stress on T. grandis seedling growth.  相似文献

5-aminolevulinic acid (ALA) is a key precursor for the biosynthesis of porphyrins such as heme and chlorophyll. ALA alleviates salinity stress damage in germinating seeds and improves seedling growth. Exogenous application of ALA at low concentrations has been shown to enhance salt tolerance in a number of plants. In the present study, we studied the effect of exogenous application of ALA on enhancing salt stress tolerance in Isatis indigotica Fort. (Anhui population as S1, Shanxi population as S2). A foliar application of 0, 12.5, 16.7, 25.0, and 50.0 mg/L ALA was given to the leaves of I. indigotica plants treated with 100 mmol/L NaCl. The fresh weight of leaves and roots; chlorophyll relative content (SPAD value); photosynthetic parameters, such as net photosynthetic rate (Pn), stomatal conductance (Gs), intercellular carbon dioxide concentration (Ci) and water use efficiency of the treated plants were determined. The third leaf of each treated plant was used to determine the activities of antioxidant enzymes. Superoxide dismutase (SOD), peroxidase (POD), catalase (CAT), glutamate synthase (GOGAT), nitrate reductase (NR) activities and the malondialdehyde (MDA) content increased in response to 100 mmol/L NaCl in both S1 and S2 plants. However, the fresh weight of leaf and root, chlorophyll relative content, Pn, Gs, Ci decreased in response to salt stress in both S1 and S2 plants. In all foliar application of ALA in S1 plants, the MDA content, and the activities of SOD and POD were the highest in response to 50.0 mg/L foliar application of ALA. GOGAT and NR activities were the highest in response to 16.7 mg/L foliar ALA. Chlorophyll content and Pn were the highest in S1 plants treated with by 25.0 mg/L ALA. In S2 plants, plant fresh weight, chlorophyll relative content, SOD, CAT, NR activities and Pn treated with 16.7 mg/L ALA were higher than that of the control (CK0). POD, MDA, GOGAT activities in S2 plants treated with 25.0 mg/L ALA were the highest among all treatments. Thus, our results showed that the optimal concentration of ALA (16.7 ~ 25.0 mmol/L) increases the activity of antioxidant enzymes, which in turn helps to abate the damage caused by salt stress in I. indigotica seedlings. Furthermore, ALA also results in an increase in chlorophyll content, Pn and the activities of GOGAT and NR.  相似文献

In the present study, the physiological responses of Nitraria tangutorum Bobr. seedlings to NaCl stress and the regulatory function of exogenous application of salicylic acid (SA) were investigated. NaCl in low concentration (100 mM) increased while in higher concentrations (200–400 mM) decreased the individual plant dry weights (wt) of seedlings. Decreased relative water content (RWC) and chlorophyll content were observed in the leaves of seedlings subjected to salinity stress (100–400 mM NaCl). Furthermore, NaCl stress significantly increased electrolyte leakage and malondialdehyde (MDA) content. The levels of osmotic adjustment solutes including proline, soluble sugars, and soluble protein were enhanced under NaCl treatments as compared to the control. In contrast, exogenous application of SA (0.5–1.5 mM) to the roots of seedlings showed notable amelioration effects on the inhibition of individual plant dry wt, RWC, and chlorophyll content. The increases in electrolyte leakage and MDA content in the leaves of NaCl-treated seedlings were markedly inhibited by SA application. The SA application further increased the contents of proline, soluble sugars, and soluble protein. The activities of antioxidant enzymes including superoxide dismutase (SOD), peroxidase (POD), catalase (CAT), and ascorbate peroxidase (APX) were up-regulated by NaCl stress and the activities of SOD, POD, and CAT were further enhanced by SA treatments. Application of SA in low concentration (0.5 mM) enhanced while in higher concentrations (1.0 and 1.5 mM) inhibited APX activities in leaves of NaCl-treated seedlings. These results indicate that SA effectively alleviated the adverse effects of NaCl stress on N. tangutorum.  相似文献

随着融雪剂在国内外寒冷地区的广泛应用及其在城市使用量的逐年增加,融雪剂对城市生态环境的危害引起了广泛的重视。其中,融雪剂在城市道路土壤中的积累对植物生长的影响已日益凸现。以油松幼苗为材料,通过分析0.2%浓度融雪剂胁迫下外源钾(K+)和水杨酸(salicylic acid,SA)对油松幼苗各生长生理指标的影响,探讨外源K+和SA在缓解融雪剂对油松幼苗生长抑制中的机理与剂量效应关系。结果表明,0.2%浓度的融雪剂处理对油松生长有明显的抑制作用,而20 mmol/L KNO3和2 mmol/L SA能明显诱导过氧化物酶(peroxidase,POD)活性的增强,缓解膜脂过氧化作用,降低丙二醛(malondialdehyde,MDA)在叶片中的积累,维持细胞膜的稳定性。虽然外源K+和SA对油松幼苗叶片胞间CO2浓度(intracellular CO2concentrations,Ci)和气孔导度(stomatal conductance,Gs)的缓解作用并不显著,但其可通过提高叶绿素含量促进光合作用的进行,缓解融雪剂胁迫对油松幼苗生长的抑制,分别增加生物量24.9%和63.6%。可见,20 mmol/L KNO3和2 mmol/L SA处理能有效缓解融雪剂对油松幼苗的伤害,为城市化学融雪剂的污染防治提供科学依据。  相似文献

We investigated the effects of jasmonic acid (JA) and gamma irradiation on the growth and metabolic responses to salt stress in rice (Oryza sativa L.) plants. The relative growth rate (RGR), relative water content (RWC), and chlorophyll (Chl) content were lower in NaCI-treated plants than in the control, whereas the malondialdehyde content (MDA), electrolyte leakage (EL), and contents of proline and abscisic acid (ABA) were higher in the treated plants. When induced by the salt stress, those effects, however, were somewhat alleviated by the application of JA or gamma irradiation. The most significant response was manifested by the proline content, with relatively lower values for alleviation being recorded for the contents of RGR, RWC, Chl, and MDA, as well as EL. Moreover, although total Chl content was not significantly influenced by JA or gamma irradiation in plants under salt stress, an increase in the level of Chl a resulted in a markedly changed ratio of Chl a/b. The degree of alleviation, in terms of growth and metabolic responses, was more extensive for JA-treated plants than for those exposed to gamma irradiation.  相似文献

Aims

The role of salicylic acid (SA) in plant responses to salinity is still a matter of controversy. To address the effect of endogenous SA variation in level and signaling on plant responses to salinity, biochemical and physiological analyses were performed on SA-altering Arabidopsis mutants including snc1 with high level of SA, transgenic line nahG with low SA, npr1-1 with SA signaling blockage, snc1/nahG plants (expression of nahG in the snc1 background), as well as wild type plants.

Methods

Plants were cultured in 1?×?Hoagland solution under controlled conditions. For salt exposure, NaCl at final concentrations of 100?mM, 200?mM, and 300?mM, respectively, was added to the culture solution after 25?d of seed germination. Except where mentioned, plant leaves were harvested after 14?d of salt stress, and used for physiological and chemical analyses.

Results

Salt stress caused all plants growth retardation with a dose-effect relationship relative to control. However, compared to wild type plants, a greater growth inhibition occurred in snc1, while a less inhibition was observed in nahG and npr1-1 plants, and a comparable extent was detected in snc1/nahG plants in which the SA level was near to that in wild type plants. The snc1 plants had lower net photosynthetic rate, variable to maximum fluorescence ratio, quantum efficiency of photosystem 2, reduced glutathione/oxidized glutathione ratio, proline levels, and higher malondiadehyde levels and electrolyte leakage rates as compared to wild type plants under salt stress. These values were effectively reversed by the expression of nahG gene in snc1 plants. The nahG and npr1-1 plants always exhibited more tolerance to salinity in above-mentioned indices than wild type plants. However, higher activities of superoxide dismutase and peroxidase in snc1 plants did not contribute to salt tolerance.

Conclusions

These data showed that SA deficit or signaling blockage in Arabidopsis plants was favorable to salt adaptation, while a high accumulation of SA potentiated salt-induced damage to Arabidopsis plants.  相似文献

This study was aimed to assess physiological responses of melon (Cucumis melo L.) cultivars to salinity stress under field conditions. Seventeen melon cultivars including 16 widely distributed native and one exotic (‘Galia’) were subjected to 2-year (2014–2015) field salinity stress. Leaf relative water content (RWC), membrane stability index (MSI), pigments [chlorophyll a, b, total chlorophyll (TChl), carotenoid (Car) and their ratios], malondialdehyde (MDA), H2O2 content, proline content (Pro), total soluble sugar content (TSC), salinity tolerance and susceptibility indices as well as yield were evaluated. The results of combined analysis of variance showed significant genotypic variation for all the traits and significant effect of salinity stress on all the traits with the exception of Chla/Chlb and TChl/Car ratios. Overall, field salinity stress caused an increase in leaf MDA, H2O2, Chla, Chlb, TChl, Car, Pro and TSC and caused a reduction in leaf MSI and RWC as well as yield. The results of correlation coefficients showed that accumulation of osmolytes (proline and TSC) led to an increase in RWC and a decrease in MDA contents. In addition, the results of multiple regression analysis showed that leaf MDA, TSC, MSI and Chla contents were the most important predictors of yield justifying 72% total variation of yield under saline conditions. These results may highlight a dynamic interplay among biomarkers for lipid peroxidation (MDA), sugar osmolytes (TSC) and photosynthetic pigment (Chla) to maintain cell viability and cell wall integrity under salinity stress conditions in melon.  相似文献

Salinity and waterlogging are two stresses which in nature often occur simultaneously. In this work, effects of combined waterlogging and salinity stresses are studied on the anatomical alteration, changes of enzymatic antioxidant system and lipid peroxidation in Mentha aquatica L. plants. Seedlings were cultured in half-strength Hoagland medium 50 days after sowing, and were treated under combination of three waterlogging levels (well drained, moderately drained and waterlogging) and NaCl (0, 50, 100, 150 mM) for 30 days. Moderately drained and waterlogging conditions induced differently aerenchyma formation in roots of M. aquatica salt-treated and untreated plants. Moreover, stele diameter and endodermis layer were also affected by salt stress and waterlogging. Salt stress significantly decreased growth, relative water content (RWC), protein level, catalase (CAT) and polyphenol oxidase (PPO) activities, and increased proline content, MDA content, H2O2 level and activities of superoxide dismutase (SOD), peroxidase (POX), and ascorbate peroxidase (APX). Waterlogging in salt-untreated plants increased significantly growth parameters, RWC, protein content, antioxidant enzyme activity, and decreased proline content, H2O2 and MDA levels. In salt-treated plant, waterlogging caused strong induction of antioxidant enzymes activities especially at severe stress condition. These results suggest M. aquatica is a waterlogging tolerant plant due to significant increase of antioxidant activity, membrane stability and growth under water stress. High antioxidant capacity under waterlogging can be a protective strategy against oxidative damage, and help to salt stress alleviation.  相似文献

Salt stress is a critical factor that affects the growth and development of plants. Salicylic acid (SA) is an important signal molecule that mitigates the negative effects of salt stress on plants. To elucidate salt tolerance in large pink Dianthus superbus L. (Caryophyllaceae) and the regulatory mechanism of exogenous SA on D. superbus under different salt stresses, we conducted a pot experiment to evaluate leaf biomass, leaf anatomy, soluble protein and sugar content, and the relative expression of salt-induced genes in D. superbus under 0.3, 0.6, and 0.9% NaCl conditions with and without 0.5 mM SA. The result showed that exposure of D. superbus to salt stress lead to a decrease in leaf growth, soluble protein and sugar content, and mesophyll thickness, together with an increase in the expression of MYB and P5CS genes. Foliar application of SA effectively increased leaf biomass, soluble protein and sugar content, and upregulated the expression of MYB and P5CS in the D. superbus, which facilitated in the acclimation of D. superbus to moderate salt stress. However, when the plants were grown under severe salt stress (0.9% NaCl), no significant difference in plant physiological responses and relevant gene expression between plants with and without SA was observed. The findings of this study suggest that exogenous SA can effectively counteract the adverse effects of moderate salt stress on D. superbus growth and development.  相似文献

In some regions of the world, low annual precipitation necessitates irrigation of crop plants which usually leads to soil salinity. Due to climatic changes this effect is also expected in the countries of Central Europe, and so in Poland. The aim of the study was (1) to compare tolerance to salt stress of Polish Triticum aestivum cvs. ‘Bogatka’ and ‘Banderola’ with T. durum cv. ‘Komnata’ and breeding line 121, and (2) to indicate the physiological parameter/parameters most suitable for such comparison. The investigation was performed in two experiments. In the first one, the germination ability of caryopses and coleoptiles’ growth were estimated at 0–250 mM of NaCl. The second experiment was conducted on plants grown in a glasshouse in saline soil at 0–150 mM of NaCl for 6 weeks. Salt tolerance was evaluated on the basis of following parameters: chlorophyll fluorescence, net photosynthesis rate (P N), transpiration rate (E), stomatal conductance (g s), cell membrane permeability (EL), proline content, fresh weight (FW), dry weight (DW), and relative water content (RWC). Highest germination of caryopses of durum cultivars was recorded at all the salinity levels; however, their coleoptiles were shorter than coleoptiles of bread wheat cultivars. Analysis of chlorophyll fluorescence showed that applied salt doses did not disturb the light phase of photosynthesis in all cultivars under study. Plants of durum wheat showed the higher dissipation of energy excess at the level of the antenna chlorophyll (DIo/CSm) under salinity as compared to plants of bread wheat. Both ‘Komnata’ and line 121 showed stronger P N reduction as an effect of salinity. A decline of P N was closely connected with a decrease in g s. The P N correlated with a decrease in DW in all studied cultivars except ‘Bogatka’. Control plants of ‘Komnata’ and line 121 were characterized by higher EL and proline level than bread wheat cultivars. An increasing cell membrane permeability correlated with a decrease of RWC in ‘Banderola’ and ‘Komnata’. The content of proline under the increasing salinity correlated with changes of RWC in ‘Banderola’, ‘Komnata’ and line 121, which indicate protectoral role of proline against dehydration of tissue. Dry weight and RWC seem to be the parameters most useful in the salt-tolerance estimation of wheat plants. Taking into account the studied parameters ‘Banderola’ could be recognized as more salt tolerant, the degree of salinity tolerance of ‘Bogatka’ is the same as line 121, while ‘Komnata’ seems to be the most salt sensitive. The salt tolerance of T. aestivum and T. durum depends on the cultivar rather than the wheat species.  相似文献

In Arabidopsis, it has been clarified that AGO4 protein is implicated in a phenomenon termed RNA-directed DNA methylation (RdDM). Previously, four orthologs of AtAGO4 were cloned in tomato, designated as SlAGO4ASlAGO4D. Here, we studied the role of the SlAGO4A gene in regulating salt and drought tolerance in tomato. SlAGO4A-down-regulating (AS) transgenic tomato plants showed enhanced tolerance to salt and drought stress compared to wild-type (WT) and SlAGO4A-overexpressing (OE) transgenic plants, as assessed by physiological parameters such as seed germination rate, primary root length, chlorophyll/proline/MDA/soluble sugar/RWC content, and survival rate. Moreover, several genes involved in ROS scavenging and plant defense, including CAT, SOD, GST, POD, APX, LOX, and PR1, were up- or down-regulated consistently under salt and drought stress. Notably, expression levels of some DNA methyltransferase genes and RNAi pathway genes were significantly lower in AS plants than in WT. Taken together, our results suggest that SlAGO4A gene plays a negative role under salt and drought stress in tomato probably through the modulation of DNA methylation as well as the classical RNAi pathway. Hence, it may serve as a useful biotechnological tool for the genetic improvement of stress tolerance in crops.  相似文献

Water stress is one of the main environmental stresses that affect plant growth and development. Salicylic acid (SA) induces water stress tolerance in plants. In this study, the effect of exogenous SA on physiological and biochemical process in Red bayberry (Myric rubra) seedlings, of three different genotypes, that were grown under water stress (soil ranging from 20 to 50 % of field capacity) was evaluated. Results showed that water stress severely affected the relative water content (RWC), photosynthesis, stomatal conductance and enzymes activities. Genotypes differed in RWC, Chlorophyll content, gas exchange parameter, antioxidant enzymes activities and proline, and the genotype Biqi had the RWC, photosynthesis, stomatal conductance and enzymes activities greater than the other two genotypes Wangdao and Shenhong. SA treated plants showed, in general, a higher RWC, chlorophyll content, photosynthetic rate, stomatal conductance, superoxide dismutase activity and proline content, and a lower relative electrolyte conductivity, methane dicarboxylic aldehyde content and catalase activity compared to those of untreated seedlings. These results signified the role of SA in diminishing the negative effects of drought on Red bayberry plants and suggest that SA could be used as a potential growth regulator, for improving plant growth under water stress.  相似文献

Effects of isoflavones on plant salt tolerance were investigated in soybean (Glycine max L. Merr. cultivar N23674) and tobacco (Nicotiana tabacum L.). Leaf area, fresh weight, net photosynthetic rate (Pn), and transpiration rate (Tr) of soybean N23674 plants treated with 80 mM NaCl were significantly reduced, while a gene (GmIFS1) encoding for 2-hydroxyisoflavone synthase was highly induced, and isoflavone contents significantly increased in leaves and seeds. To test the impact of isoflavones to salt tolerance, transgenic soybean cotyledon hairy roots expressing GmIFS1 (hrGmIFS1) were produced. Salt stress slightly increased isoflavone content in hairy roots of the transgenic control harboring the empty vector but substantially reduced the maximum root length, root fresh weight, and relative water content (RWC). The isoflavone content in hrGmIFS1 roots, however, was significantly higher, and the above-mentioned root growth parameters decreased much less. The GmIFS1 gene was also transformed into tobacco plants; plant height and leaf fresh weight of transgenic GmIFS1 tobacco plants were much greater than control plants after being treated with 85 mM NaCl. Leaf antioxidant capacity of transgenic tobacco was significantly higher than the control plants. Our results suggest that salt stress-induced GmIFS1 expression increased isoflavone accumulation in soybean and improved salt tolerance in transgenic soybean hairy roots and tobacco plants.  相似文献

The present study was undertaken to find out individual and interactive effects of arsenic (As) and salicylic acid (SA) on an important medicinal plant, Artemisia annua. As uptake and its accumulation was detected and found to be maximum in roots at higher As concentration (150 μM). Under As treatments, H2O2 and MDA content were induced. Biomass and chlorophyll content were negatively affected under As treatments. Furthermore, enzymatic (SOD, CAT, APX, and GR) and non-enzymatic antioxidants were also enhanced under As treatments. Exogenous application of SA reduced the extent of H2O2 and O2 ? generation and lipid peroxidation, while reverted biomass and chlorophyll content to overcome oxidative stress. Simultaneous application of SA with As increased endogenous SA level, artemisinin, and dihydroartemisinic acid as compared with individual As treatment and pre-application of SA with As treatments. The expression of four key artemisinin biosynthetic pathway genes, i.e., ADS, CYP71AV1, DBR2, and ALDH1 were upregulated at a maximum in plants simultaneously treated with SA and As. Similar pattern of artemisinin accumulation and glandular trichome size was observed which attest that SA has a stimulatory impact on artemisinin biosynthesis under As stress. Our study suggests that exogenous application of SA and As together induced more tolerance in A. annua than a comparable dose of SA pre-treatment. The study may provide a platform with dual benefits by developing As-tolerant plants to be used for phytoremediation of arsenic from As-contaminated soil and obtaining high artemisinin-producing A. annua plants.  相似文献
