This study describes the binding of [3H]corticosterone and [3H] dexamethasone to soluble macromolecules in the pituitaries of adrenalectomized rats perfused at sacrifice to remove blood contamination. (1) Unlabeled dexamethasone competes only slightly for [3H] corticosterone binding when the steroids are added to cytosol as well as when the steroids are present from the time of disruption of the tissue. In the latter case, corticosterone is as good a competitor as dexamethasone for [3H]dexamethasone binding. (2) Gel permeation chrmatography with BioRad A-5M reveals the presence of a [3H] corticosterone-macromolecular complex with an elution volume comparable to transcortin as well as a very large hormone-binding complex eluting in the void volume of the column...  相似文献

Analysis of rat uterine cytosol for Tamoxifen binding reveals that the saturable binding sites are only partially inhibited by estradiol-17β. Partial thermal denaturation of the cytosol at 30° C for 2 h 30 allows the characterization of a high affinity (Kd = 3.3 × 10?9M) saturable Tamoxifen class of binding sites insensitive to estradiol-17β while remaining sensitive to the antiestrogens CI628 and Nafoxidine. The uterine concentration of these binding sites is lower in the uterus of immature or castrated animals, increases from metestrus to proestrus and reaches a peak on the day of estrus.  相似文献

Reuber H-35 hepatoma (H4-II-E-C3) and HTC cells are known to retain differentiated corticosteroid induced functions in cell culture and to bind corticosteroids to macromolecules in cytosol which subsequently enter the cell nuclear fraction. Using both cell types we have demonstrated the major macromolecular fraction in cytosol to have properties (elution position from DEAE columns, pI, 3H-dexamethasone binding), very similar to those of rat liver corticosteroid Binder II which may be the hormone receptor.  相似文献

A macromolecular binder of folic acid and folic acid derivatives has been identified in the particulate fraction of homogenates of rabbit choroid plexus. Within the choroid plexus, there are 2.3 nmol of folate-binding activity (binder) per g of tissue. The molecular weight of the folate binder complex, separated from the particulate fraction after solubilization with Triton X-100, was 340,000 to 400,000 by Sephadex gel filtration. The partially purified binder, when freed of endogenous folates, bound equivalent amounts of both [3H]folic acid and [methyl-14C]methyltetrahydrofolic acid per mg of protein. Folic acid, homofolic acid, 5-methyltetrahydrofolic acid, and to a lesser degree, methotrexate, inhibited the binding of both [3H]folic acid and [14C]methyltetrahydrofolic acid. Binding activity, which decreased below pH = 7.0, was unaffected by pretreatment with ribonuclease but was eliminated completely by papain and a protease (Streptomyces griseus). Although dihydrofolate reductase was present in choroid plexus, the binder was distinct from dihydrofolate reductase as judged by gel filtration and methotrexate sensitivity. This high affinity binder of folates may be responsible, in part, for the rapid, saturable uptake of folic acid and methyltetrahydrofolic acid by rabbit choroid plexus in vitro.  相似文献

The intestine of ricketic rats, a recognized target tissue of vitamin D, contains s soluble macromolecule capable of specific in vitro binding of both 25-hydroxycholecalciferol and 1,25-dihydroxycholecalciferol. Its sedimentation behavior on linear 5–20% sucrose gradients suggests as a molecular weight of approximately 100 000. The binding is specific for sterols possessing both an open B ring and 25-hydroxyl group, and is destroyed by pre-incubation with trypsin. The binding affinity for 25-hydroxycholecalciferol (KA = 2 · 109 1/mole) was 2.8 times that for 1,25-dihydroxycholecalciferol.  相似文献

1. The effect of free fatty acids [FFAs (saturated (S) and unsaturated (U))] on dexamethasone binding in vitro using liver cytosol from rainbow trout was examined. 2. All UFFAs but none of the SFFAs tested suppressed binding. This suppression is dose-dependent and correlated roughly with the degree of unsaturation of the FFAs. 3. Scatchard analysis indicated that the addition of linoleic C18:2 (150 microM) increased the dissociation constant (Kd = 5.1 +/- 0.4 x 10(-8) M vs control of 1.7 +/- 0.3 x 10(-8) M) but minimally affected the binding capacity (Bmax = 68 +/- 6.2 vs control of 88 +/- 15.2 fmol/mg protein) suggesting C18:2 caused a conformational change of the receptor. 4. Lineweaver-Burk plot revealed a mixed non-competitive type of inhibition by C18:2. 5. Free acid appeared to be required for inhibition as esterification or derivatization of the acid greatly diminished its potency. 6.C18:2 also promotes the dissociation of bound [3H]-dexamethasone from the steroid-receptor complex but slower in rate and lesser in magnitude compared to that caused by dexamethasone or the glucocorticoid antagonist RU 38486. 7. UFFAs and some of their derivatives can thus modulate glucocorticoid receptor function in vitro and might play essential roles in regulating glucocorticoid action in fish as well. 8. These fatty acids presumably acts at a site different from that of the glucocorticoid binding site.  相似文献

Analysis of the interactions of two synthetic estrogen antagonists, tamoxifen and CI 628, with rat uterine and chick oviduct cytosol revealed significant differences in the antiestrogen binding properties of these tissues. In the rat uterus CI 628, tamoxifen and estradiol were bound to a similar number of saturable binding sites and estradiol could completely inhibit the binding of tritiated antiestrogens to these sites. In contrast, high affinity, saturable antiestrogen binding sites in chick oviduct were present at three times the concentration of estradiol binding sites and estradiol could only partially inhibit the binding of tritiated antiestrogens to these sites. It is concluded that antiestrogens bind to the estrogen receptor in both tissues and that chick oviduct has an additional saturable antiestrogen binding site distinct from the classical estrogen receptor site.  相似文献

The relationship between the glucocorticoid binding capacity of rat liver cytosol and the activity of tyrosine aminotransferase has been studied in adrenalectomized male rats. Bilateral adrenalectomy of male rats caused an increase within 3 days in the level of specific dexamethasone binding of liver cytosol accompanied by a rapid decrease in tyrosine aminotransferase activity. Known inducers of tyrosine aminotransferase were administered in vivo to test their effect on dexamethasone binding capacity, in order to determine whether the induction was by an indirect mechanism involving an increase in glucocorticoid binding capacity. Insulin, adrenalin, glucagon, dibutyryl cyclic AMP and oestradiol caused a significant increase in the activity of the enzyme, with no change in the specific dexamethasone binding. Tetracosactrin, a synthetic analogue of ACTH, had no effect on either parameter. It was concluded that the induction of tyrosine aminotransferase by the compounds tested was not mediated by an increase in glucocorticoid receptor activity.  相似文献

The incubation of rat thymocytes with 3H-cortisol in the presence of cortisol immobilized on polyvinylpyrrolidone (PVP-GC) produces a decrease in 3H-cortisol uptake by these cells. PVP-GC is shown to compete with cortisol for specific binding sites on thymocytes, without penetrating into cells. It is therefore, concluded that plasma membranes of thymocytes contain specific glucocorticoid binding sites.  相似文献

In the presence of 0.3 M potassium chloride and 0.5 mM dithiothreitol, rat intestinal cytosol contains two binding proteins for 1,25-dihydroxyvitamin D3 (1,25-(OH)2D3)1 having sedimentation coefficients of 3.2S and 5–6S. The 3.2S protein is specific for 1,25-(OH)2D3 as determined by competition analysis, whereas the 5–6S protein binds 25-hydroxyvitamin D3 (25-OH-D3) exclusively.  相似文献

Studies of the temperature sensitivity of estradiol receptor binding in rabbit uterine cytosol have revealed the existence of an enzyme which catalyzes the covalent binding of estradiol to cytosol proteins. A fraction, prepared by chromatography on Biogel P-200 and incubated at 37 degrees C in the presence of Mn++, exhibited a time-dependent, temperature-sensitive, oxidative binding of estradiol not seen in the crude cytosol preparation. Although the activity of this enzyme was shown to be independent of estradiol binding by the high affinity estrogen receptor, its presence may complicate studies of estrogen receptor action which involve the use of elevated temperatures.  相似文献

An alpha-tocopherol-binding macromolecule was isolated from the heart cytosol of rabbits fed for 1 month with an alpha-tocopherol-deficient diet. The amount of [3H]-tocopherol bound to nuclear chromatin was increased when the alpha-tocopherol-deficient heart nuclei were incubated in the presence of [3H]tocopherol-cytosol complex. In this condition, large amounts of [3H]tocopherol were associated with a subnuclear fraction that contained non-histone acidic proteins.  相似文献
