Two trials were conducted to evaluate treatments combining progesterone pretreatment and prostaglandin F(2alpha) (PGF(2alpha)) on estrus response, pregnancy and calving rate in heifers. Treatments in Trial 1 were 1) control (T(1); n=59), 2) 25 mg PGF(2alpha) on Day 0 (T(2); n=58), 3) 150 mg progesterone (P(4), i.m.) in corn oil on Day -24 plus PGF(2alpha) (T(3); n=61), and 4) 150 mg P(4) on Day -5 plus PGF(2alpha) (T(4); n=59). Trial 2 had T(2) and T(4) only. Heifers were artificially inseminated 8 to 16 h after detection of estrus for 10 and 5 d in Trials 1 and 2, respectively. In Trial 1 more heifers in T(3) and T(4) showed estrus by 72 h compared to T(1) and T(2). In T(3), percentages were greater at 84 and 96 h than in T(1) and T(2). There were no differences between T(3) and T(4) or T(1) and T(2) over time. Cumulative distributions of responses showed that more heifers in T(3) and T(4) were in estrus by 84 h after PGF(2alpha) than after other treatments, while T(3) showed the greatest total number of heifers in estrus by 84 h; this difference persisted for 180 h. In Trial 2, percentages of heifers observed in estrus for T(1) and T(4) were not different. Average interval from PGF(2alpha) to estrus was shorter in Trial 1 for T(3) heifers compared to other treatments. No difference was observed in interval to estrus for T(2) and T(4) in Trial 2; this interval averaged 58 h. Artificial insemination pregnancy rates were not different among treatments in either trial and averaged 67.4%. In Trial 1, a greater proportion of heifers in T(2), T(3) and T(4) calved by 35 days into the calving season compared to T(1), but in Trial 2 calving rates for T(2) and T(4) were not different. Progesterone pretreatment combined with PGF(2alpha) appeared to enhance estrus synchronization without influencing either pregnancy or calving rates.  相似文献

The prostaglandin F(2alpha) analog (PGFA) alfaprostol was used in 277 cyclic dairy heifers for the purpose of estrous cycle synchronization. A dose of 5 mg was used in all the trials during spring 1979 and 1.5 mg 100 kg body weight were used during winter 1979/80. Animals were treated according to 3 schedules: two doses 11 days apart without prior examination (Schedule I), one dose without prior observation and a second dose 11 days later only for those animals that failed to show estrus after the first treatment (Schedule II) and one dose for animals not showing estrus after a 5-day observation period (Schedule III). Estrus synchronization was achieved with peak estrus activity occurring form 32 to 72 h after treatment in 95% of the responding animals. Of the animals treated according to schedules I, II and III, 93%, 100% and 100% showed synchronized estrus activities respectively. Conception rates in all trials, from insemination at observed heat (Schedules I, II and III) or from fixed time insemination at 48 and 72 h after the first or second treatments in schedules III and I, respectively, compared well with that of untreated contemporary controls with a range of 34.2 to 66.7% for animals in these trials, and an overall conception rate of 49.48% for treated and of 48.30% for control animals. This observation, together with the pregnancy rate at 60 days after breeding (78.30% for all treated animals and 73.50% for all untreated controls), indicates that alfaprostol had no adverse effects on fertility.  相似文献

Over a two year period, 276 dairy heifers (German Black Pied x Holstein Friesian) received one or two injections i.m. of 1.5 mg/100 kg body weight of the PGF analog alfaprostol. Only animals not showing heat after the first injection were treated twice, with the same dose, 11 days apart. Before treatment, 71% of the heifers were cycling regularly, 12% were cycling irregularly and 17% were not observed in heat prior to treatment. Animals in heat were inseminated once or twice, 18 to 24 h apart. In response to the first injection, 60.5% of all treated animals came into heat; 38% showed heat after the 2nd treatment. Three of the six non-responding animals were later diagnosed as freemartins. Heat was well synchronized. Twenty seven percent, 60% and 9% of all first inseminations occurred on days two, three and four after treatment (s), respectively; 65% of all heifers conceived from the first insemination. Pregnancy rates after one or two inseminations were 66.0% and 63.8%, respectively. At the second a.i., 45% of all animals had already ovulated. Pregnancy rates from first a.i. after one or two treatments with alfaprostol were 60.1% and 72.2%, respectively. Years had no influence on any of the parameters observed. Stage of cycle in which the first treatment occurred had a strong influence on the occurrence of heat and on conceptions as well. Of the animals treated between days 6 and 18% of their cycles, 92.6% responded with heat to the first treatment and 79% conceived. When animals were treated for the first time between days 19 and 5 of their cycles, only 28.8% came into heat and only 23.8% conceived. After the 2nd treatment, 71.2% of the latter group came into heat and 73.1% conceived.  相似文献

This study was conducted to determine the efficacy of feeding melengestrol acetate (MGA) for 14 days and administering prostaglandin F(2)alpha (PGF) 17 days after MGA to synchronize or induce estrus in yearling beef heifers. The study involved 56 Angus (n = 19), Hereford (n = 15) and Simmental (n = 22) heifers that were assigned by breed and pubertal status to either MGA+PGF or to control groups. Heifers in the synchronized group were fed 0.5 mg MGA per head per day for 14 days from a grain carrier and were injected with 25 mg, i.m. PGF 17 days after the last daily feeding of MGA. Control heifers were fed from a grain carrier without MGA and were not treated with PGF. Heifers were classified as pubertal when concentrations of progesterono in the serum exceeded 1 ng/ml in 1 of 2 samples collected prior to the initiation of treatments. Blood samples were collected 7 days before and on the day that treatment with MGA or carrier began and 7 days before and on the day that PGF was administered. Progesterone concentrations in the serum were elevated ( > 1 ng/ml) in 61% (17 28 ) of the MGA+PGF-treated heifers and in 61% (17 28 ) of the control heifers prior to feeding MGA. However, concentrations of progesterone in the serum at the time PGF was administered differed (P<0.05) between MGA+PGF and control groups. Concentrations of progesterone in the serum exceeded 1 ng/ml in 100% (28 28 ) of the MGA+PGF-treated heifers and in 71% (20 28 ) of control heifers at the time PGF was administered (P<0.05). All heifers were inseminated 12 hours after the first detected estrus. Twenty-two of 28 (79%) of the MGA+PGF-treated heifers exhibited estrus within 6 days after PGF compared with 9 of 28 (32%) of control heifers (P<0.05). The conception rate at first service did not differ between MGA+PGF and control groups (64% and 67%, respectively). Synchronized pregnancy rates were higher (P<0.05) for MGA+PGF-treated heifers than for control heifers (14 28 , 50% vs 6 28 , 21%). Increased concentrations of progesterone in serum at the time PGF was administered and higher pregnancy rates during the synchronized period among MGA+PGF-treated heifers demonstrate the efficacy of this treatment for use in estrus synchronization. Moreover, this treatment may have a potential effect on inducing puberty in breeding age heifers.  相似文献

The effect of pregnancy on the release of prostaglandin F2 alpha (PGF2 alpha) in response to oxytocin (OT) has been examined. Fourteen cyclic heifers received one intravenous injection of 1 IU OT (n = 6) or 100 IU OT (n = 8) 17, 18, or 19 days (Day 17-19) after the onset of estrus (Day 0). Five of these animals also received 100 IU OT at Days 6 and 13 to determine the effect of OT at different times of the cycle. Frequent blood samples were taken for 60 min before and for 90 min after OT injection for the measurement of 15-keto-13,14-dihydro-PGF2 alpha (PGFM) by radioimmunoassay. The experiment was then repeated using the same animals at Day 17-19 of pregnancy (confirmed by the recovery of an embryo the day after OT injection). Following the injection of 1 IU OT, plasma PGFM reached its peak within 30 min with the increase significantly lower (P less than 0.05) in pregnant (1.13 +/- 0.10-fold) than in nonpregnant animals (2.07 +/- 0.27-fold). However, because only 3 of the 6 cyclic animals showed a response to 1 IU OT, the dose was increased to 100 IU in subsequent experiments. The animals that received 100 IU at Days 6 and 13 had no significant increase in PGFM concentrations (1.18 +/- 0.05-fold and 1.01 +/- 0.04-fold, respectively). At Day 17-19 the increase in plasma PGFM reached its peak 5-15 min after 100 IU OT and the increase was significantly greater in nonpregnant (3.23 +/- 0.17-fold) than in pregnant (1.21 +/- 0.02-fold; P = 0.003) heifers. Six of 11 animals injected at Day 17-19 of the cycle showed a decrease in progesterone (P4) the day after OT administration. These data show that the release of PGF2 alpha in response to OT is suppressed in pregnant animals in vivo, suggesting an antiluteolytic role for the embryo in luteostasis.  相似文献

In a 5-year study (1973-1977), 281 cycling beef heifers were treated with a 7-day norgestomet (SC21009) ear implant and an intramuscular injection of prostaglandin F(2alpha) (PGF(2alpha)) at the time of implant removal or 24 hr before implant removal. Percentages of heifers in estrus by 36, 48, 60, 72, and 120 hr after implant removal were 32.4, 52.7, 71.6, 80.1, and 93.2, respectively. Onset of estrus occurred an average of 49.8 +/- 4.7 hr after treatment. Percentages of heifers in estrus 36 hr after treatment were 5.7 and 51.7 for those with a corpus luteum and those without a corpus luteum (or determined regressing by palpation) at implant removal, respectively. When PGF(2alpha) was injected 24 hr before implant removal, 55% of the heifers were in estrus by 36 hr after implant removal compared to 30% when PGF(2alpha) was injected at the time of implant removal; however, by 60 hr after implant removal the difference was 76% vs. 71%. First-service conception rates for synchronized and nonsynchronized heifers were 62.2% and 59.6%, respectively. During 1976 and 1977 heifers were checked for estrus every 4 hr and inseminated 2, 6, 10, 14, 18, 22, 26, or 30 hr after first detected to be in standing estrus. Conception rate was not significantly affected by time of insemination but tended to be higher for heifers bred 26 and 30 hr after first being detected in standing estrus (78.9% and 70.0% vs. average 59.2%). Treatment with a 7-day norgestomet implant plus a single injection of PGF(2alpha) 24 hr before or at implant removal appears to be a practical technique for synchronizing estrus in cycling heifers without affecting conception.  相似文献

Two mature heifers were slaughtered on days 3, 6-7, 10-11, 16, 18-19 or on day 21 of the oesterus cycle. Endometrium was incubated in quadruplicates with medium-199 at 37 C and a water saturated gas phase of 95% O2 + 5% CO2. Half ml medium samples were taken after 6, 12 and 24 h of incubation for determination of PGF2 alpha and PGFM. PGF2 alpha was secreted by endometrium at each stage of the oestrous cycle. Maximal secretion was measured around oestrus (p less than 0.01) compared with days 6-16 of the cycle. Concentration of PGFM in medium had a similar trend. Highest ability of endometrium for PGF2 alpha metabolism (indicated by the ratio PGF2 alpha:PGFM) was on days 6-16 of the cycle. Data suggest that PGF2 alpha metabolism by the endometrium may depend on ovarian steroids and that this metabolism may also protect the corpus luteum from the luteolytic action of PGF2 alpha besides reduced production of this prostaglandin during the luteal phase.  相似文献

Cloprostenol was previously believed to be unable to release endogenous prostaglandin F2alpha (PGF2alpha) when administered during early bovine diestrus. A prostaglandin release is, however, seen in late diestrus. The aim of this study is to find out whether dexcloprostoenol (containing the only biologically active isomer, d-isomer, of cloprostenol) induces endogenous PGF2alpha release during early and late diestrus. Twelve heifers of the Finnish Ayrshire breed were allocated into two equal groups. Their estrous cycles were synchronized with dexcloprostenol. A further luteolysis was induced with 0.15 mg of dexcloprostenol either on Day 7 (group D7 or early diestrus) or on Day 14 (group D14 or late diestrus) after ovulation. Blood for progesterone and the PGF2alpha metabolite 15-ketodihydro-PGF2alpha determinations was collected immediately before dexcloprostenol treatment and thereafter every second hour for 48 h. Five of the six heifers in both groups showed significantly increased blood levels of 15-ketodihydro-PGF2alpha at some time during the 48-h experimental period. The intervals from treatment to the first significant increases of the PGF2alpha metabolite were 32.8+/-2.3 h (min. 30 h, max. 36 h) and 20.0+/-4.2 h (min. 14 h, max. 24 h) in groups D7 and D14, respectively (P < 0.01). We have concluded that dexcloprostenol induced endogenous PGF2alpha release in most cases, regardless the time of its administration (early or late diestrus). This release, however, differs from that observed during spontaneous luteolysis.  相似文献

Fifteen does received intravaginal pessaries impregnated with 20 mg fluorogestone acetate (Cronolone) at various times after the end of estrus for a period of fourteen days. The does were then treated on days 2, 4, 5, 12, or 16 after onset of estrus following the progestin treatment, with 15 mg of prostaglandin F2α (Lutalyse) injected intramuscularly. Seventy three percent of the does showed estrus within 2 days after withdrawal of progestin treatment. Ten out of 11 does treated with prostaglandins 4 to 16 days after the end of estrus exhibited estrus within 44 to 72 hours after treatment. Breeding following the use of prostaglandin in these does resulted in a high pregnancy rate. It is concluded that a sequential use of progestin and prostaglandin may be a useful method for achieving estrus synchronization associated with high fertility in does.  相似文献

This study was designed to test the efficacy of melengestrol acetate (MGA) in combination with prostaglandin F(2alpha) (PGF(2alpha)) in synchronizing estrus in cyclic and noncyclic heifers. One hundred thirty-one cyclic and prepubertal crossbred heifers were randomly assigned to three treatment groups: Controls (n = 43); MGA (0.5 mg/d for 7 d) and PGF(2alpha) (25 mg i.m. on Day 7; n = 44); and PGF(2alpha) (25 mg i.m. on Day 7; n = 44). Observations for estrus were made at 6-n intervals throughout the 7-d treatment period followed by a 34-d artificial insemination breeding season. A greater percentage (P < 0.05) of MGA-PGF(2alpha) noncyclic heifers showed behavioral estrus (91%) than did Control (67%) or PGF(2alpha) heifers (61%) during the 34-d artificial insemination period. There was no difference (P > 0.05) between synchronization rates of the MGA-PGF(2alpha) heifers and PGF(2alpha) heifers 7 d after PGF(2alpha) administration. The percentage of control animals in estrus during the first 25 d of the breeding season did non differ from the synchronized rates of MGA-PGF(2alpha) and PGF(2alpha) heifers (P > 0.05). Conception rates (heifers pregnant/heifers inseminated) did not differ (P > 0.05) for cyclic or prepubertal heifers among Control, MGA-PGF(2alpha) or PGF(2alpha) heifers. Though conception rates did not differ, there was a trend toward lowered conception rates in MGA-PGF(2alpha) heifers.  相似文献

Clenbuterol, as other sympathomimetic drugs, relaxes the myometrium, thus causing a short-term inhibition of labor and the delay of parturition. This study has examined the influence of clenbuterol on the release of prostaglandin F2 alpha (PGF2 alpha) induced by oxytocin alone or with estradiol-17 beta. Five bilaterally ovariectomized heifers, primed with progesterone for 14 days, were used in two experiments. In the first they received two i.v. injections of oxytocin 6h apart, with and without an i.v. injection of clenbuterol before the second oxytocin injection; the second experiment was similar to the first except that the animals were given estradiol-17 beta 30 min after the first oxytocin injection. Frequent blood samples were taken for the measurement of 13,14-dihydro-15-keto-PGF2 alpha by radioimmunoassay. The data show that clenbuterol does not influence PGF2 alpha release in response to oxytocin alone or with estradiol-17 beta, and it does not inhibit the basal release of PGF2 alpha. This suggests that clenbuterol does not act on the endometrium to alter the secretion of PGF2 alpha in the non-pregnant cow.  相似文献

Ninety-five normal cyclic crossbred beef heifers were used to determine if the proportions of heifers showing estrus, intervals to estrus and corpus luteum (CL) function were influenced by PGF(2alpha) dosage and (or) the stage of luteal phase when PGF(2alpha) was administered. Heifers were assigned randomly to treatments in a 4 x 3 factorial arrangement. Treatments were 5, 10, 25 or 30 mg PGF(2alpha) injected either in early (5 to 9 d), mid (10 to 14 d) or late (15 to 19 d) stages of the luteal phase. Jugular samples were taken at 0 h and at 8 h-intervals for 48 h and again at 60 h after PGF(2alpha) treatment for progesterone assay. Heifers were observed for estrus continuously for 120 h PGF(2alpha) treatment. The proportion of heifers showing estrus was dependent upon (P<0.05) both dosage of PGF(2alpha) and stage of luteal phase. Heifers given 5 mg of PGF(2alpha) showed estrus only if treated during the late stage, while those given 10 mg of PGF(2alpha) showed a progressive increase of heifers in estrus as stage of luteal phase advanced. The proportion of heifers showing estrus after 25 and 30 mg of PGF(2alpha) increased from 56% for the early stage to 100% for the mid and late stages. Interval to estrus in heifers showing estrus within 120 h after PGF(2alpha) treatment did not differ (P>0.05) among dosages but tended (P=0.10) to be longer in heifers treated during the mid luteal stage (67 h) than in heifers treated in the two other stages (56 h). A greater proportion of heifers (P<0.05) showed estrus by 60 h after PGF(2alpha) when treated during the early and late luteal stages (75.5%) than for heifers treated during the mid luteal stage (30.4%). Patterns of progesterone concentrations were influenced (P=0.08) by the three way interaction of dosage, stage and time. In heifers that showed estrus, rate of decline in progesterone tended (P=0.07) to be slower during the mid luteal stage than during the early and late stages. Progesterone did not drop below 1 ng/ml until 32 h in heifers treated during the mid luteal stage; whereas progesterone dropped below 1 ng/ml by 24 h in heifers treated during the early and late stages. These data may be useful in designing more efficient systems for using PGF(2alpha) or its analogues in estrus synchronization of beef cattle.  相似文献

Using radioimmunoassay procedures, the levels of plasma, uterine and ovarian prostaglandin (PG) F2alpha, and those of plasma estradiol and progesterone were measured in intact, hysterectomized or ovariectomized immature female rats pretreated with PMS and subsequent HCG. Occurrence of ovulation was confirmed at 8 hours after the HCG administration not only in the intact rats but also in the hysterectomzied rats. The levels of plasma estradiol and progesterone, and of uterine and ovarian PGF2alpha rose with the PMS injection alone, but they did not reach the peaks before the HCG administration. Both plasma estradiol and uterine PGF2alpha showed a peak at 2 hours after the HCG injection. These peaks were antecedent 2 or 6 hours before the peaks of ovarian and plasma PGF2alpha, respectively. However, such increase of uterine PGF2alpha does not seem to be indispensable for ovulation, because ovulation could occur in the hysterectomized rats. The levels of ovarian PGF2alpha showed a high plateau from 4 to 8 hours after the HCG injection, and then rapidly decreased after ovulation. The levels of plasma PGF2alpha peaked not only in the intact rats but also in the hysterectomized rats at 8 hours after the HCG treatment. But in the ovariectomized rats, this plasma PGF2alpha peak at 8 hours disappeared and there was no statistical change of plasma PGF2alpha throughout the PMS-HCG treatment. Plasma progesterone gradually increased and reached the maximum at 10 hours after the HCG injection. These results conclude that the main source of increased plasma PGF2alpha during the ovulatory process induced with the PMS-HCG treatment is the ovary, and it is strongly suggested that a rapid increase of PGF2alpha in the ovary may play some important role(s) in the ovulatory process.  相似文献
