Internal tandem duplication (ITD) mutations within the FMS-like tyrosine kinase-3 (FLT3) render the receptor constitutively active driving proliferation and survival in leukemic blasts. Expression of FLT3-ITD from the endogenous promoter in a murine knockin model results in progenitor expansion and a myeloproliferative neoplasm. In this study, we show that this expansion begins with overproliferation within a compartment of normally quiescent long-term hematopoietic stem cells (LT-HSCs), which become rapidly depleted. This depletion is reversible upon treatment with the small molecule inhibitor Sorafenib, which also ablates the disease. Although the normal LT-HSC has been defined as FLT3(-) by flow cytometric detection, we demonstrate that FLT3 is capable of playing a role within this compartment by examining the effects of constitutively activated FLT3-ITD. This indicates an important link between stem cell quiescence/homeostasis and myeloproliferative disease while also giving novel insight into the emergence of FLT3-ITD mutations in the evolution of leukemic transformation.  相似文献

Intracellular membrane trafficking depends on the ordered formation and consumption of transport intermediates and requires that membranes fuse with each other in a tightly regulated and highly specific manner. Membrane anchored SNAREs assemble into SNARE complexes that bring membranes together to promote fusion. SNAP29 is a ubiquitous synaptosomal-associated SNARE protein. It interacts with several syntaxins and with the EH domain containing protein EHD1. Loss of functional SNAP29 results in CEDNIK syndrome (Cerebral Dysgenesis, Neuropathy, Ichthyosis and Keratoderma). Using fibroblast cell lines derived from CEDNIK patients, we show that SNAP29 mediates endocytic recycling of transferrin and β1-integrin. Impaired β1-integrin recycling affected cell motility, as reflected by changes in cell spreading and wound healing. No major changes were detected in exocytosis of VSVG protein from the Golgi apparatus, although the Golgi system acquired a dispersed morphology in SNAP29 deficient cells. Our results emphasize the importance of SNAP29 mediated membrane fusion in endocytic recycling and consequently, in cell motility.  相似文献

The mitochondrial kinase PINK1 and the ubiquitin ligase Parkin are participating in quality control after CCCP- or ROS-induced mitochondrial damage, and their dysfunction is associated with the development and progression of Parkinson''s disease. Furthermore, PINK1 expression is also induced by starvation indicating an additional role for PINK1 in stress response. Therefore, the effects of PINK1 deficiency on the autophago-lysosomal pathway during stress were investigated. Under trophic deprivation SH-SY5Y cells with stable PINK1 knockdown showed downregulation of key autophagic genes, including Beclin, LC3 and LAMP-2. In good agreement, protein levels of LC3-II and LAMP-2 but not of LAMP-1 were reduced in different cell model systems with PINK1 knockdown or knockout after addition of different stressors. This downregulation of autophagic factors caused increased apoptosis, which could be rescued by overexpression of LC3 or PINK1. Taken together, the PINK1-mediated reduction of autophagic key factors during stress resulted in increased cell death, thus defining an additional pathway that could contribute to the progression of Parkinson''s disease in patients with PINK1 mutations.  相似文献

造血干细胞是目前研究最为深入的成体干细胞,是极富应用前景的研究领域,然而其维持自我更新以及多向分化潜能的分子机制尚不明.MicroRNA (miRNA)是一类崭新的调控性非编码小分子RNA,在监控生物体个体发育和细胞增殖、分化进程中起着重要作用.miRNA参与包括胚胎干细胞和多种成体干细胞的发育进程,人类造血干细胞及其发育过程中也存在特征性miRNA表达谱,参与调控造血干细胞发育进程,以miRNA为分子靶点的防治造血功能低下疾患的研究具有广阔的应用前景.  相似文献

通过同种基因型小鼠构建造血干细胞移植模型,将预处理的全骨髓单个核细胞或c-Kit+造血干细胞移植至致死剂量照射的受体小鼠体内,动态监测移植2~16周后受体小鼠体内供体来源细胞造血重建以及嵌合情况,以期揭示不同群体的供体细胞以及预处理等因素对小鼠造血干细胞移植后造血重建的影响。实验结果显示,移植后早期(2周)全骨髓单个核细胞组髓系比例要高于c-Kit+细胞移植组,但全骨髓移植组受体小鼠呈现出较大的移植后不良反应,出现脱毛、食欲不振以及体重减轻的症状。c-Kit+细胞移植组在淋系重建上要早于全骨髓移植组,供体细胞的嵌合植入也早于全骨髓移植组,但两组实验组最终均能完成造血重建过程。实验结果表明c-Kit+细胞移植组在移植后能够较快地实现供体细胞植入,进而开始造血重建,且c-Kit+细胞移植组的不良反应要低于全骨髓移植组。结果说明在整体造血重建效果上c-Kit+细胞移植组要优于全骨髓移植组。  相似文献

目的研究Cramp基因敲除在衰老过程中对小鼠造血干细胞的作用。方法应用流式细胞仪分析3月龄及12月龄Cramp基因敲除小鼠及同窝野生型小鼠的骨髓造血干细胞的比例及不同发育阶段B淋巴细胞的比例。结果与野生型小鼠相比,12月龄Cramp基因敲除小鼠的骨髓长期造血干细胞增多,多潜能造血祖细胞减少;前体B淋巴细胞和未成熟B淋巴细胞减少,成熟B淋巴细胞增多。结论在衰老过程中,Cramp基因敲除对骨髓造血干细胞及B淋巴细胞发育有重要影响。  相似文献
