Field trials were conducted to evaluate the efficacy of wheat bran bait formulations of Paranosema locustae and Metarhizium anisopliae for controlling grasshoppers in southeast Niger. Treatments consisted of wheat bran baits mixed with M. anisopliae, P. locustae + M. anisopliae or with P. locustae spores and P. locustae + sugar. Oedaleus senegalensis, Pyrgomorpha cognata and Acrotylus blondeli were the predominant species at the time of application representing ca. 94% of the total population. Bran application was done when O. senegalensis (ca. 75% of the population) was at its early developmental stages, with first, second and third instars accounting for 64–85%. Grasshopper population reduction, P. locustae prevalence and level of infections in the predominant species were monitored. Manual application of P. locustae and M. anisopliae formulated in wheat bran has proven to induce consistent pathogen infection in grasshopper populations. Population density over the three weeks monitoring, typically decreased by 44.7 ± 6.9%, 52.8 ± 8.4%, 73.7 ± 5.5% and 89.1 ± 1.8% in P. locustae, P. locustae + sugar, M. anisopliae and P. locustae + M. anisopliae treated plots respectively. Paranosema locustae prevalence in surviving adult grasshoppers at 28 after application was 48.1 ± 2.3%, 28.9 ± 4.8% and 27.4 ± 3.7%, with infection level of 6.2 ± 0.8 × 106, 2.3 ± 0.3 × 104 and 2.1 ± 0.3 × 103 spores mg−1 host weight in O. senegalensis, A blondeli and P. cognate respectively. Other species that each accounted for <2% of the community, namely Aiolopus thalassinus, A. simulatrix, Acorypha glaucopsis, Acrotylus patruelis, Anacridium melanorhodon, Diabolocatantops axillaris, Kraussaria angulifera and Schistocerca gregaria were found to show sign of infection. The results from this study suggest that wheat bran application of M. anisopliae and P. locustae alone or in combination, targeting early instars grasshopper could be a valuable option in grasshopper control programs.  相似文献

Paranosema (Antonospora) locustae (Canning) is a microsporidium with an extensive host range including >100 reported insect hosts from the order Orthoptera. The susceptibility of two species of locusts (Orthoptera: Acrididae) – the migratory locust, Locusta migratoria subsp. migratorioides (Fairmaire & Reiche), and the desert locust, Schistocerca gregaria Forsskål – to P. locustae was compared under laboratory conditions at a decreased temperature of 27 °C to enhance susceptibility to infection. Locusta migratoria was found highly vulnerable as infection percentages exceeded 70% at 104, 105, and 106 spores per insect, and mortality increased with increasing dosage. Conversely, P. locustae spores were not found in S. gregaria throughout the experiment. Only at 107 spores per insect, as many as 20% of S. gregaria were infected. This suggests that the desert locust is resistant to P. locustae infection, as opposed to the migratory locust. The laboratory models of these parasite–host systems may be useful for elucidating mechanisms of insect immunity to microsporidia.  相似文献

Second instar nymphs of African migratory locust, Locusta migratoria migratorioides, and desert locust, Schistocerca gregaria, were tested for their susceptibility to the microsporidium pathogen Johenrea locustae (Lange et al. 1996 Lange, C.E., Becnel, J.J., Razafindratiana, E., Przybyszewski, J. and Razafindrafara, H. 1996. Johenrea locustae n.g., n.sp. (Microspora: Glugeidae): A Pathogen of Migratory Locusts (Ortrhoptera: Acrididae: Oedipodinae) from Madagascar. Spores of J. locustae were produced from live L.m. migratorioides, a conspecific to L. migratoria capito. Locusta m. migratorioides and S. gregaria were exposed to wheat seedlings sprayed with 20 mL of three concentrations (106, 107 and 108 spores mL?1). Both second-instar nymphs of L.m. migratorioides and S. gregaria were susceptible to J. locustae infection at the three concentrations. There was no effect of concentration of the microsporidium on mortalities of S. gregaria (92–98%) 19 days postinfection. Adjusted mortality in L.m. migratorioides at 19 days post-exposure was 24, 43 and 80% at the corresponding treatment concentrations. The effect of infection on fecundity was tested on both L.m. migratorioides and S. gregaria. Spinach leaf discs were treated with different concentrations (0, 104, 105 and 106 spores mL?1) of J. locustae and presented to female insects for 24 h. Female L.m. migratorioides surviving infection as nymphs laid significantly fewer egg pods than untreated controls at all levels of exposure. The number of eggs per female was also significantly lower in treated lots than in the controls. Higher spore concentrations also adversely affected egg hatching rate. The effect of J. locustae infection on feeding was tested on S. gregaria. There was a significant decrease in food intake among S. gregaria nymphs treated only at the high concentration (106 spores mL?1).  相似文献

Aenasius arizonensis (Girault) is an important solitary endoparasitoid of Phenacoccus solenopsis Tinsley. To optimise the mass production of high-quality females, it is important to assess the influence of mating regimes on the progeny fitness and sex allocation. We, therefore, hypothesise that mating combinations in A. arizonensis adults emerged from different host instars may influence parasitism and sex allocation in the subsequent generation. Therefore, we compared three nymphal instars (1st, 2nd and 3rd) and adults host stages of Psolenopsis for parasitism and sex allocation by A. arizonensis. Further, F1 female progeny of the parasitoid emerged from different host instars was henceforth evaluated for its fitness in six mating combinations. A. arizonensis females parasitised all the host stages except the 1st instar nymphs. The parasitised 2nd instar nymphs yielded only males, while the sex ratio in the later host instars was strongly female-biased. The parasitoid females preferred 3rd instar nymphs with respect to higher parasitism (74.0–84.0%) and produced more females in the F1 progeny as compared to other host stages. F1 females that emerged from 3rd instar nymphs produced significantly higher parasitism (74.0–79.0%). These mating combinations also yielded more female progeny in the F2 generation. However, parasitism by F1 females was significantly lower (9.0–12.0%) when mated with males that emerged from 2nd instar P. solenopsis nymphs. Moreover, latter combinations yielded only male progeny in F2 generation. These findings can be used in laboratory mass rearing of this parasitoid vis-à-vis biological control of P. solenopsis.  相似文献

The ability of parasites to modify the behaviour of their hosts is a wide spread phenomenon, but the effects of microsporidian parasites on locust behaviour remain unexplored. Here the frequencies of directional changes (ND) and jumping (NJ) per minute of gregarious locusts infected with 2000 spores of the microsporidian parasite Paranosema locustae were significantly different from those of untreated locusts 10 and 16 days after infection, being similar to values for solitary nymphs. In contrast, the behaviour of locusts inoculated with the lower doses of 200 spores/locust was sometimes like that of solitary nymphs. At other times, behaviour was intermediate between solitary and gregarious, i.e. transitional. The rearing density did not affect the turning and jumping behaviour of infected locusts, and their behaviours were similar to those of solitary locusts at 10–16 days after infection. Our study demonstrates that infection with P. locustae may lead gregarious locusts to change some of their behaviour to that typical of solitary locusts.  相似文献

The larvae of the pine processionary moth (PPM), Thaumetopoea pityocampa, feed on the needles of pine and cedar. The urticating hairs of older instars pose a threat to human and animal health. Strains of the entomopathogenic fungi, Metarhizium brunneum (V275, ARSEF 4556) and Beauveria bassiana (KTU-24), were assayed against first to fourth instar T. pityocampa using doses ranging from 1?×?105 to 1?×?108 conidia mL?1. The three strains differed slightly in their virulence but caused 100% mortality of all instars at the highest dose. The newly emerged or first instar larvae were extremely susceptible with 100% mortality being achieved 2–4 days post inoculation with V275 at all but the lowest dose. The fourth instar larvae appeared to be less susceptible than earlier instars. There was good horizontal transmission of conidia from treated to un-inoculated larvae. However, mortality was higher in third and fourth instars and where the ratio of inoculated versus untreated larvae was high. This we presume is due to spores being more readily trapped by the urticating hairs found on third and older instar larvae. Injection of the nests offers a simple and environmentally friendly way of controlling the pest with reduced risk to operators.  相似文献

Abstract

This study was carried out to determine the lethality of the entomopathogenic fungus, Lecanicillium longisporum on eggs, young and old nymphs of the greenhouse whitefly, Trialeurodes vaporariorum. Mortality percentage was significantly differed based on stage of T. vaporariorum and conidial concentrations of L. longisporum. Average of the infection level to insect was very low particularly in egg with only 9.81%, even with higher conidial concentrations (1×107 conidia mL-1). Whereas, it was higher in 1st and 2nd instar (46.56%) and 3rd and 4th instars (37.21%). Three parameters were assessed with T. vaporariorum eggs, namely; egg infection, egg hatchability and crawlers emergence. Egg mortality percentages averaged 3.56, 7.14, 9.64, 16.42 and 20.35% with fungal concentrations of 1×103, 1×104, 1×105, 1×106 and 1×107 conidia mL-1, respectively. Daily infection percentages were varied depend upon the conidia concentration where the highest infection rate of eggs was occurred with 1×107. Egg hatch was very high and the mortality among the emerged crawlers was neglectable compared with the control. Efficiency of L. longisporum on whitefly nymphs also was varied based on the insect instar and fungal concentration. Mortality percentages were obviously higher in young nymphs (1st and 2nd instars) than in older ones (3rd and 4th instars). The results indicated that nymphs were highly susceptible to fungal treatment compared with eggs.  相似文献

Studies were conducted to compare preference among Bemisia tabaci Gennadius, biotype B instars for parasitization by Eretmocerus mundus Mercet and Encarsia pergandiella Howard when provided one instar only, two different instars, and four different instars simultaneously. In the single‐instar no choice treatment, Er. mundus was more successful in parasitizing the younger host instars, while En. pergandiella parasitized a greater proportion of the older instars. Similar results were observed when parasitoids were provided a choice of two instars in six different pair combinations. When all four instars were provided simultaneously, the numbers of first, second, and third instars parasitized by Er. mundus were not significantly different from each other (range 10.3–16.4%), but all were significantly higher than parasitism of fourth instar nymphs (2.1%). The highest percentage parasitization by En. pergandiella was in third instar (17.2%), and the lowest in first instar (2.8%).  相似文献

The effects of crowding and isolation on body color and behavior were observed for the mid‐instar nymphs of the desert locust, Schistocerca gregaria. Some of the solitarious (isolation‐reared) nymphs that were crowded for 1 or 4 h during the third instar developed black patterns at the fourth instar, but most individuals remained unaffected. Black patterns appeared in all individuals that were crowded for 1 day or longer, but even after 4 days of crowding the black patterning for some individuals was not as intense as that for the gregarious (crowd‐reared) controls. Isolation of gregarious nymphs caused the black patterns to recede or disappear at the last (fifth) nymphal instar, but it was necessary to isolate the nymphs from the beginning of the first instar to obtain body coloration looking like solitarious nymphs in most individuals. Solitarious nymphs that were allowed to see gregarious nymphs developed different intensities of black patterns depending on the body size and number of nymphs shown. The behavioral phase shift from one phase to another was observed when the nymphs were crowded or isolated for 2 days or longer, as previously reported for the last nymphal instars of the same strain. Behavioral gregarization was induced for isolated nymphs that were allowed to see a group of nymphs through a transparent double wall. These results suggested that body‐color phase shift occurred more rapidly for mid‐instar nymphs than for late instar nymphs but the rate of behavioral phase shift was similar for the two instars.  相似文献

室内测定了球孢白僵菌Beauveria bassiana BBSG8702菌株对桃蚜Myzus persicae 若蚜的毒力,并分析了若蚜脱皮与白僵菌有效侵染之间的关系。在21℃、1×106/mL孢子浓度下, 1~4龄若蚜感菌后的累积死亡率分别为10.1%、2.1%、3.1%和40.2%,明显低于同样条件下成蚜的感菌死亡率(98.4%)。在若蚜各龄感菌死亡的个体中,1~4龄中分别有8.1%、50.0%、44.4%和98.0%的若蚜在死亡前发育到成蚜,且都能产下正常发育的后代。若蚜的感菌死亡率与接菌后到第一次脱皮的时间密切相关,脱皮早则感菌死亡率低,反之亦然。但试验中也发现有部分若蚜特别是低龄若蚜感菌后未完成第一次脱皮便死亡。观察了桃蚜在1、4龄早期感染球孢白僵菌后的发育、存活和生殖情况,结果表明:1、4龄早期若蚜接菌后第一次脱皮的时间早于未接菌若蚜,但提前幅度少于5%,在羽化为成蚜之前被致死的概率分别只有4.1%和0,在羽化为成蚜后8天内的生殖率与未接菌蚜虫的无明显差异。这些结果表明,若蚜的脱皮,尤其当蚜虫在低龄若蚜时受感染后的多次脱皮,可有效地摆脱球孢白僵菌分生孢子的侵染,降低该菌的毒力。  相似文献

The activity of entomopathogens on insect pests has been investigated for many species but the influence of entomopathogenic fungi on factors other than mortality relating to population increase has not been frequently studied. The influence of Lecanicillium attenuatum CS625 (=Verticillium lecanii CS625) on development and reproduction of cotton aphid (Aphis gossypii) was investigated. A conidia suspension of the isolate was applied onto first instar nymphs. Increased spore concentration did not significantly affect each nymphal stage, total nymphal period, pre-reproductive period and the age of first larviposition. A significant dose effect on reduction of life span, reproductive period and fecundity was observed in 1st and 3rd instars after spore application. When conidia were applied to 1st instars, life span was significantly reduced to 10.8 and 8.4 days at 1 × 104 and 1 × 108 conidia/ml, respectively from 12.2 days in the control. During the life span, total fecundity was 41 ± 7.3, 26 ± 0.8 and 22 ± 5.7 nymphs per female at 1 × 104, 1 × 106 and 1 × 108 conidia/ml, respectively compared with 51 ± 2.0 nymphs per untreated female. Reproduction period was also significantly shortened with increasing spore concentration. Application of spores to 3rd instars showed a similar trend. However, daily fecundity of individual aphids was not affected by spore dose. It was concluded that the isolate of L. attenuatum is able to affect populations of cotton aphid by reducing life span and total fecundity as well as by killing the aphids directly.  相似文献

Eretmocerus mundus Mercet is a parasitoidof Bemisia tabaci (Genn.) indigenous tothe Mediterranean and is used commercially foraugmentative biological control in Spain andelsewhere. A better understanding of thesuitability of different host instars wouldhelp optimize production and field application.Incidence of parasitism, development time,survivorship and sex ratio were evaluated whendifferent nymphal instars of the sweetpotatowhitefly Bemisia tabaci biotype `Q' wereoffered for parasitization. Experiments wereconducted on sweet pepper at 25 °C, 75%RH and 16:8 (L:D) photoperiod. E. mundusoviposited in all nymphal instars of B.tabaci except the mature 4th instar orpharate adult (previously designated, `pupa').Incidence of parasitism was greatest (33.8± 5.1 parasitized nymphs) and developmenttime shortest (14.1 ± 0.1 d) whenoviposition occurred under 2nd and3rd instar nymphs compared to 1st or4th instars. Survivorship (85%) andoffspring sex ratio (39.8% female) did notdiffer statistically for parasitoids developingin whiteflies that were parasitized asdifferent instars. Although 2nd and3rd instars were clearly the mostfavorable host stage for E. mundus, itscapacity to parasitize and develop on a widerange of host stages is a favorablecharacteristic for both rearing and fieldapplication.  相似文献

Even though Paranosema locustae is widely used in China as a biological agent for controlling grasshoppers,the mortality rate is initially quite low.This study sought to determine whether the simultaneous use of P.locustae and Beauveria bassiana would be a more effective control strategy.Additionally,changes in the intestinal microbial communities of migratory locusts infected with the two pathogens were analyzed to investigate the roles of gut microbes in pathogen-host interactions.The mortality rate of locusts inoculated with B.bassiana and P.locustae simultaneously was not significantly higher than expected but the mortality rates of locusts inoculated with B.bassiana 3,6,and 9 days after inoculation with P.locustae were significantly higher than if their effects were additive,indicating synergism.A MiSeq analysis found that Weissella was the most common bacterium,representing 41.48%and 51.62%of the total bacteria in the mid-and hindguts,respectively,and the bacterial declines were greatest during dual infections with B.bassiana and P.locustae.The appropriately timed combined application of P.locustae and B.bassiana was more effective against locusts than either treatment alone.Moreover,the combined inoculation of the two pathogens changed the gut microflora of locusts,indicating the potential relevancy of their synergistic effects on locust control.  相似文献

Abstract

The susceptibility of immature Choristoneura rosaceana (Harris) to Beauveria bassiana-GHA (BotaniGard® 22WP) was evaluated under laboratory and field conditions. Although egg masses ≤24 h old were susceptible to infection by topically sprayed B. bassiana spores in the laboratory and resulting mycosis significantly reduced the percent of neonates able to emerge, no significant egg mass infections resulted from orchard treatment of ≤24–48 h eggs. Exposure to high levels of the B. bassiana product on apple leaf surfaces in the laboratory caused significant dose-related mortality in first to fourth instar C. rosaceana. First instar C. rosaceana were the most susceptible of the larval stages assayed in the laboratory although only 36% of neonates introduced into the orchard prior to Beauveria treatment and 27% of the neonates emerging from orchard treated egg masses became infected. Fourth instar mortalities were similar but moderate when treated with 1×108 spores mL?1 in the orchard and on leaf surfaces in the laboratory. Beauveria bassiana-induced larval mortalities were significantly higher when the spores were applied directly to the larval integument as opposed to the leaf surface where the insect would encounter the pathogen in a treated orchard. Feeding of C. rosaceana larvae on antibiotic containing meridic diet prior to their use in trials did not impact the susceptibility of the larvae to B. bassiana. Antibiotic containing meridic diet significantly reduced larval C. rosaceana mortality when treated larvae or the spores were placed directly on the diet as opposed to leaf tissue.  相似文献

Antonospora locustae is a microsporidian parasite of grasshopper insects that is used as a biological control agent. We report on laboratory selection of isolates from different regions with increased virulence. Bioassays were conducted against third instar nymphs of Locusta migratoria manilensis. AL2008L01 was originally imported from the USA in 1986, AL2008M01 was isolated from Melanoplus differentialis in USA and AL2008F01 was isolated from infected Fruhstorferiola tonkinensi collected in Guangdong, China. The results showed that all three isolates can infect the locust and that pathogenicity increased gradually with increased dose. The LD50 values of the original isolates at the highest dose (5×106 spores/nymph) were 19, 23 and 22 days and LD50 values were 3.2×105, 3.4×106 and 0.7×106 spores/g, respectively. After selecting for three generations, the virulence of all isolates increased significantly. LT50s were reduced to 17, 20 and 21 days at the highest dose (5×106 spores/nymph) and LD50s were reduced to 1.4×105, 2.5×105 and 1.7×105 spores/g.  相似文献

Encarsia formosa Gahan (Hymenoptera: Aphelinidae), a thelytokous parasitoid, is an important biological control agent of whiteflies because of its outstanding reproduction and host‐feeding ability. In this study, we evaluated the parasitism, host feeding and developmental time of E. formosa populations reared on Trialeurodes vaporariorum (Westwood) (EFT) or on Bemisia tabaci (Gennadius) (EFB) when different nymphal instars of the castor whitefly, Trialeurodes ricini (Misra), were offered as hosts, with an aim to understand the preference of the parasitoid on nymphal instars of T. ricini. Experiments were conducted on castor bean plants at 26 ± 2°C, 50–60% RH and 16 : 8 (L : D) photoperiod. The results showed that E. formosa successfully oviposited and fed on all nymphal instars of T. ricini. However, numbers of the first instars fed on by the E. formosa populations reared on T. vaporariorum (EFT) and B. tabaci (EFB) were significantly greater (45.9 and 31.3, respectively) than those of the second (EFT: 30.4 and EFB: 15.8), the third (EFT: 22.4 and EFB: 13.2) and the fourth nymphal instars (EFT: 6.0 and EFB: 3.8). The number of T. ricini nymphs parasitized by E. formosa varied significantly among different instars, and the parasitism rates on the first instar (EFT: 15.2; EFB: 7.7) and fourth instar (EFT: 19.3; EFB: 4.9) were greater than those on the second and third instars. Encarsia formosa reared on T. vaporariorum had a significantly higher host feeding and ovipositing potential on T. ricini than EFB. When parasitizing the fourth instar nymphs, E. formosa completed development in a significantly shorter time (12.9 day) than when ovipositing in other instars (17.8–19.1 day). These results showed that EFT had a better host adaption than EFB. The information from this study should be useful for us to better understand the performance and nymphal preference of E. formosa from T. vaporariorum and B. tabaci when they parasitized and fed on T. ricini, and the interactions of parasitoids with different host whitefly species.  相似文献

Nabis roseipennis Reuter nymphs demonstrated a preference for nuclear polyhedrosis virus (NPV) — infected over healthyAnticarsia gemmatalis Hübner larvae when offered a choice of larval prey in Petri dishes and on soybean. In Petri dishes, small (second-third instar) and large (fifth-sixth instar) nymphs attacked a significantly greater number of diseased than healthy larvae at all larval instars tested (first-fifth instars) and exposure periods (2, 5 and 24 h), except that at 2 h the number of 1st and 3rd instar larvae attacked by large nymphs did not differ significantly (P≤0.05). Nabis roseipennis caged with larvae on individual soybean plants in the greenhouse resulted in a generally low percentage of attack by small and large nymphs after 2 days, ranging from 5.6 to 36.7%. As in the Petri dishes, the nabids showed a significant preference for diseased larvae over healthy larvae attacked for all nabid and larval sizes on soybean, with the percentage of diseased larvae attacked ranging from 28.0 to 65.4% (P≤0.05). This preference for diseased larvae on soybean as well as in Petri dishes demonstrates that the preference was not due to the close proximity in which the host and prey were found in the Petri dishes. The preference for diseased larvae may be due to a reduction in a defensive response in late stages of disease. Three behaviors of epidemiological importance, namely feeding latency, feeding duration and defecation latency, for six populations of Meccus phyllosomus longipennis (Usinger) from areas of central, western and north-central Mexico with high (HP) and low (LP) prevalence of Trypanosoma cruzi (Chagas) human infection were evaluated in this study. The median feeding latency (the time taken to begin feeding) was highly variable between instars. Within–instar comparisons showed that at least 65% of the LP populations (N3 to adult) started to feed significantly (P < 0.05) later than the HP population, with N1 showing no difference, and N2 from LP populations feeding sooner than those from HP populations. The six populations had similar median feeding durations within instars. A higher (P < 0.05) percentage of the instars from HP populations defecated faster than the respective instars from the three LP populations. Approximately 25% of the young nymphs (N1 to N3) and females in the HP populations defecated < 2 min postfeeding, compared with 4%–6% of the young nymphs and 1.3%–3% of females in the LP populations. Moreover, 17.7%–38.8% of the older nymphs (N4 to N5) in the HP populations and 6.8%–13.4% in the LP populations defecated during or immediately after feeding. Our results indicate that the HP populations have a greater potential than the LP populations to transmit T. cruzi infections, which may underlie the differences in the prevalence of T. cruzi infection in some areas where M. p. longipennis is currently distributed.  相似文献

With the aim of developing better procedures for rearing the microtype tachinid fly Pales pavida (Meigen), we performed ecological studies in the laboratory using the natural host Mythimna separata (Walker), investigating larval development, mating behaviour, individual oviposition patterns and relationships between parasitisation and number of eggs ingested (NEI) per host. The host instar at the time of parasitoid egg ingestion significantly affected the development time of the immature parasitoid: development took longer when the hosts ingested eggs when at the fifth instar than at the sixth (last) instar. There was no difficulty obtaining mated females in the laboratory when day 0–1 female flies were kept with day 2–4 males. Mean lifetime fecundity was 5805?±?568 eggs per female. Daily rates of oviposition by individual females varied widely; the greatest number of eggs laid in a day was exactly 1700. When the NEI by day 1 fifth instars or day 0 or 3 last instars was 1, 3, 6 or 10, the parasitisation percentage tended to increase with increasing NEI, although it did not differ significantly between NEI 6 and 10.
