Adequate moisture levels are required for nematode survival and subsequent efficacy as entomopathogens. Formulation of nematodes aimed at aboveground applications may assist in maintaining such moisture levels. In this study, we report the effects of a superabsorbent polymer formulation, Zeba® on the performance of an entomopathogenic nematode, Heterorhabditis zealandica Poinar, for controlling diapausing codling moth, Cydia pomonella (L.) larvae in cryptic habitats on trees. Water activity (aw-value) on bark was considered to be an indication of moisture levels on trees in cryptic habitats where codling moth larvae are known to occur, thereby influencing nematode efficacy. H. zealandica was only able to infect codling moth larvae at aw≥0.92, with aw50=0.94 and aw90=0.96. Laboratory experiments in which nematode concentration was investigated indicated a positive linear relationship between the concentration of nematodes applied and the level of control obtained, with the highest level of mortality recorded at 80 IJs/larva, requiring at least 4 h of conditions conducive to nematode activity to ensure infectivity and subsequent efficacy. Further experimentation showed that the use of the Zeba formulation, together with the nematodes, improved the level of control obtained at 60% and 80% RH in the laboratory and that it also enhanced the survival and infection-ability of the nematodes in the field. The study conclusively illustrates that the tested formulation assisted in maintaining adequate moisture levels on the application substratum, as required for nematode survival and subsequent efficacy.  相似文献

The non-insecticidal control strategies currently being implemented in South African orchards for the control of codling moth, Cydia pomonella (L.) may be hampered by wooden fruit bins being infested with diapausing codling moth larvae, acting as a potential source of re-infestation. Key factors contributing to the success or failure of an entomopathogenic nematode application were investigated using the SF 41 isolate of Heterorhabditis zealandica in laboratory bioassays with wooden minibins. Under operational conditions, an application rate of 100 IJs/mL (LD90=102 IJs/mL) effectively controlled codling moth larvae in these bins, and for further laboratory bioassays, the LD50 value of 18 IJs/mL (?25 IJs/mL) was identified as the discriminating dosage. Maximum mortality was attained when bins were pre-wet for at least 1 min (>90% RH) and maintained at maximum humidity (>95% RH) post-treatment for at least 3 days (LT90=73 h), to ensure nematode survival and subsequent satisfactory infection of diapausing codling moth larvae. Tarping bins achieved the desired high level of humidity required. Furthermore, adjuvants (specifically Reverseal 10?) also improved an application. The study conclusively illustrated that if all the above-mentioned conditions are met, H. zealandica has the potential to successfully disinfest wooden fruit bins of codling moth.  相似文献

The biocontrol potential of South African isolates of Heterorhabditis zealandica, Steinernema citrae, S. khoisanae, S. yirgalemense, and Steinernema sp., was evaluated against codling moth, Cydia pomonella. Codling moth was susceptible to all six nematode isolates at a concentration of 50 infective juveniles/insect (78–100% mortality). Low temperatures (10 h at 17°C; 14 h at 12°C) negatively affected larvicidal activity (≤3%) for all isolates. All tested isolates were most effective at higher levels of water activity (a w=1). The average a w50-values for all isolates tested was 0.94 (0.93–0.95), except S. khoisanae 0.97 (0.97–0.98). Regarding host-seeking ability, no positive attraction to host cues could be detected amongst isolates, except for H. zealandica. Three of the isolates, H. zealandica, S. khoisanae, and the undescribed Steinernema sp., were selected for field-testing and proven to be effective (mortality >50%). Insect containment methods used during field experimentation was shown to influence larvacidal activity, as different levels of mortality were obtained using various containment methods (wooden planks vs. pear tree logs vs. mesh cages). Pear tree logs were impractical. Predictive equations were subsequently developed, enabling future trials to be conducted using either planks or cages, enabling the prediction of the expected level of control on tree logs. All tested isolates therefore showed a certain degree of biological control potential, however, none of the experiments showed clear efficacy-differences amongst isolates. The study highlighted the importance of environmental factors to ensure the successful application of these nematodes for the control of diapausing codling moth larvae in temperate regions.  相似文献

Stacked wooden fruit bins are frequent overwintering sites for overwintering diapausing codling moth larvae. Control strategies against the codling moth (Cydia pomonella) (Lepidoptera: Tortricidae) in South Africa have been hampered by the reinfestation of orchards from nearby stacked infested fruit bins and by the movement of infested bins between orchards. Worldwide, wooden fruit bins are systematically being replaced with plastic bins, however in South Africa this will not be accomplished in the near future. The objective of this study was to evaluate the potential of two recycled commercially available entomopathogenic nematode (EPN) species, Heterorhabditis bacteriophora and Steinernema feltiae, as well as of a local species, Steinernema yirgalemense, to disinfest miniature wooden fruit bins under controlled conditions in the laboratory. After dipping miniature bins loaded with codling moth larvae in a suspension of 25?IJs/mL of each of the three EPN species, under optimum conditions of temperature and humidity, the highest percentage of control was obtained using S. feltiae (75%). The addition of adjuvants significantly increased S. feltiae infectivity to >95%, whereas it did not result in a significant increase in H. bacteriophora or S. yirgalemense infectivity.  相似文献

Cydia pomonella L. is an important insect pest of pome fruits worldwide. We have isolated and characterized 17 microsatellite loci from the enriched genomic libraries constructed using a biotin/streptavidin capture protocol. Among these loci, 11 were scored polymorphic when 50 individuals from a laboratory population were screened. Their alleles numbered two to four, with the observed heterozygosity (HO) ranging from 0.114 to 0.404. Successful amplification was obtained for all these loci when the designed primers were tested, showing the promise of use in genetic mapping and population studies.  相似文献

Codling moth (CM), Cydia pomonella (L.) is the most serious pest of apple and other pome fruit worldwide. In temperate climates, diapausing cocooned larvae make up 100% of the population. Control of this stage would reduce or eliminate damage by first generation CM in late spring and early summer. Entomopathogenic nematodes (EPNs) are good candidates for control of CM in the cryptic habitats where the larvae overwinter. The two predominant limiting factors for EPNs are adequate moisture and temperatures below 15°C. Formulation that maintains moisture and enables survival of EPN infective juveniles (IJs) until they can infect overwintering larvae would significantly improve their utility for protection of apple, pear and walnut. In laboratory studies conducted in moist mulch (consisting of apple and conifer wood), Galleria mellonella (L.) larvae infected with Steinernema carpocapsae (Weiser), S. feltiae (Filipjev), or Heterorhabditis bacteriophora Poinar and coated with starch and clay, produced mean mortalities of 42, 88, and 24%, respectively in CM larvae. Mulched field plots treated with formulated S. carpocapsae- or S. feltiae-infected G. mellonella larvae, then followed by an application of wood flour foam as an anti-desiccant, resulted in 56 and 86% mortality, respectively. Comparative tests of aqueous suspensions of S. carpocapsae IJs applied to cardboard bands on apple tree trunks followed by water, fire retardant gel or foam resulted in 11, 35, and 85% respective mortalities. Identical tests with S. feltiae resulted in 20, 19, and 97% respective mortalities. Our research with cadaver formulations of EPNs in mulch and aqueous suspensions on tree trunks combined with anti-desiccant agents, demonstrated significant improvement in larvicidal activity for diapausing cocooned CM larvae.  相似文献

Spread pachytene nuclei of codling moth Cydia pomonella (Linnaeus) (Lep., Tortricidae) females of a Syrian strain (SY) were used to investigate chromomere patterns of chromosome bivalents and determine their length. The karyotype of female codling moths consists of 28 chromosome bivalents, of which seven are clearly distinguishable using chromosome length and the number and size of the chromomeres in the pachytene stage. One autosome bivalent has two nucleolar organizing regions (NORs) that are located at the opposite ends of the chromosome and appear as distinct structural landmarks. In female codling moths, the WZ sex chromosome bivalent was easily identified in pachytene oocytes according to the heterochromatic thread of the W chromosome. This study contributed to the knowledge and identification of pachytene chromosomes of female codling moths.  相似文献

The possibility of controlling the codling moth Cydia pomonella (Linnaeus) using an attract and kill approach as an alternative to chemical sprays with contact insecticides was investigated in widely separated orchards. The results of a 4‐year study have shown that, using an attract and kill approach, three applications/season kept infestation rates in treated orchards below the economic injury level except in one with a too high codling moth population density. The mean number of male codling moths/trap/week in attract and kill‐treated orchards was much lower in comparison with control orchards which were treated with the usual cover sprays of insecticides. The results also showed that the efficacy of attract and kill under orchard conditions decreased with time and the relationship between time effect and codling moth death rate was very strong. These data indicate that the attract and kill technique applied at a rate of three application per season resulted in good control of codling moth in well managed orchards in Syria.  相似文献

Seasonal changes in the fatty acid composition of the total lipid extracted from the whole body of Cydia pomonella L. larvae were determined by gas chromatography. The six most abundant fatty acids in both non-diapause and diapause larvae of codling moth were oleic (35%-39%), palmitic (23%-33%), linoleic (16%-30%), palmitoleic (5%-10%), stearic (1.5%-3.0%) and linolenic acids (1.0%-2.5%). This represents a typical complement of Lepidopteran fatty acids. The fatty acid composition of total lipid of C. pomonella larvae was related to diapause. In similarity to most other reports, the proportion of unsaturated fatty acids increased in diapause initiation state. The total lipid of diapause larvae contained more linoleic acid (25.8% vs. 16.1%) and less palmitic acid (24.7% vs. 33.4%), than that of non-diapause larvae. The weight percentage of linoleic acid (C 18:2) increased from 16% to 26% from early-August through early-September during transition to diapause, while palmitic acid (C16:0) decreased from 33% to 25% at the same time. These changes resulted in an increase in the ratio of unsaturated to saturated fatty acids (UFA/SFA) from 1.72 in non-diapause larvae to 2.63 in diapause larvae.  相似文献

The resistance to deltamethrin was evaluated in diapausing larvae of 14 field populations of the codling moth (Cydia pomonella L.) From the main French orchard areas using biological assays. Resistance to deltamethrin was compared to mixed-function-oxidase (mfo) activity measured at the individual level through ethoxycoumarin-O-deethylase (ECOD) activity using a fluorescence microplate reader. The larvae were collected in corrugated paper band traps in the autumn of 1995. Analysis was previously performed on two laboratory strains, one susceptible and one resistant to deltamethrin, in order to characterize the changes in resistance during diapause development. Resistance to deltamethrin as well as the ECOD activity were stable during the diapause, and ECOD activity was always significantly lower in the susceptible strain than in the resistant one. The ECOD activity was significantly correlated to the frequency of resistant moths in the field populations. This strongly suggested the involvement of the mfo system in the resistance to deltamethrin in these populations. The intra-strain variabilities in ECOD activity of both laboratory and field resistant insects indicated that other resistance mechanisms might also be involved. Further investigations on these mechanisms are needed in order to develop complete diagnostic methods and to define suitable control strategies against each resistant population. Arch. Abstract:  Resistance of the codling moth Cydia pomonella (L.) (Lep., Tortricidae) to the organophosphorus compound (OP) azinphosmethyl was observed in apple orchards in Israel. The level of resistance varied with the pest control strategy. Compared with a sensitive laboratory population, the resistance level was highest in insects from the preventative pest control strategy, intermediate in integrated pest management (IPM) orchards, and relatively low in the organic orchards. The level of azinphosmethyl resistance in larvae (but not in adults) exposed for 17 generations in the laboratory to a pesticide-free diet was reduced by 50%. Codling moth larvae resistant to azinphosmethyl were also resistant to various insect growth regulators (IGRs). The IGRs include three chitin synthesis inhibitors (diflubenzuron, novaluron and teflubenzuron), two juvenile hormone mimics (pyriproxyfen and fenoxycarb) and one ecdysone agonist (methoxyfenozide). Codling moth resistant to azinphosmethyl was tolerant to methoxyfenozide and novaluron without previous history of application in apple orchards, indicating the possibility of cross-resistance. According to this study, managing resistance programs in apple orchards should be based on IPM principles with minimum use of conventional neuroactive pesticides.  相似文献

The morphology of Nosema carpocapsae and its development in experimentally infected codling moth larvae are described. Spherical uninucleate meronts were the first stages. Nuclear division produced binucleate meronts which were the most abundant vegetative stage, although additional uninucleate and a few tetranucleate meronts were also observed at this time. All meronts were spherical and ranged from 2.8 to 5.8 μm in diameter. Uninucleate and binucleate fusiform sporonts then appeared followed by some tetranucleate and dividing forms. Oval sporoblasts developed after these and did not divide before maturing into spores. Sporonts were approximately 5.0 to 7.9 × 2.4 to 3.0 μm. Spores developed in all host tissues except the nervous tissue. The binucleate spores showed considerable variation in spore size, 2.4 to 3.9 × 1.3 to 3.1 μm (alcohol fixed, Giemsa stained). The polar filament was usually coiled 11 times (range 9 to 13) at an angle of 53° to the long axis of the spore. Its maximum observed length was 75 μm.  相似文献

Predation of cocooned larvae of codling moth Cydia pomonella by silvereyes Zosterops lateralis was studied in an apple orchard in Nelson, New Zealand. Apple logs with known larval densities were made available to the birds for known periods of time, either in cages or exposed in the apple orchard. The numbers of silvereyes and the natural predation of codling moth were recorded in the same orchard. Predation was density dependent. On caged logs with an initial high density of 32 larvae, 1.1 larvae were consumed per bird‐hour; in contrast, one larva was consumed per 34.5 bird‐hours at three larvae per log. A curvilinear relationship was demonstrated between larval density and the bird‐hours required for predation; this relationship was consistent with the known density dependence of silvereye predation of codling moth. A regression of the total annual winter bird predation of larvae in the orchard on bird numbers was significant. However, the density dependence of predation resulted in declining rates of predation over the winter as larval density declined; the first birds to arrive in the orchard benefitted from particularly high predation rates. As a consequence, fluctuations in bird numbers during the winter had only a secondary influence on predation rates. The numbers of silvereyes in the orchard showed no relationship to the density of the codling moth population present. This study confirmed the importance of silvereyes in the predation of codling moth and a functional, not numerical, rseponse of these birds to codling moth density.  相似文献

在甘肃省高台县通过对三角胶粘式和水盆式这2种类型苹果蠹蛾Cydia pomonella(L.)诱捕器的田间比较研究。结果显示,二者在诱捕效率上存在的明显差异。在苹果蠹蛾密度较低的条件下,三角胶粘式诱捕器平均日诱捕量为2.50只,比水盆式诱捕器高出近2.84倍,最早监测到成虫的时间也比水盆式诱捕器提前3~4d,因此,三角胶粘式诱捕器具有更高的监测效率。结果还显示地面植被的遮盖极大地降低诱捕器的诱捕效果,严重时会使诱捕器对密度较低的苹果蠹蛾种群的监测功能丧失。  相似文献
