本文根据GenBank中已登录的蜡蚧蚧霉Lecanicillium lecanii,粉棒束孢Isaria farinosa,球孢白僵菌Beauveria bassiana和金龟子绿僵菌Metarhizium anisopliae的几丁质酶基因序列的同源性比较, 以它们高度保守的核苷酸序列设计一对引物,采用RT-PCR和3′/5′-RACE相结合的方法,首次从玫烟色棒束孢Isaria fumosorosea中克隆出几丁质酶Ifuchi1全长cDNA基因。其全序列长为1 542 bp,编码阅读框由1 275个核苷酸组成, 5′非翻译区(5′-UTR)与3′非翻译区(3′-UTR)分别为33个核苷酸和234个核苷酸。基因编码424个氨基酸, 信号肽长度为24个氨基酸, 成熟蛋白理论分子量为47.6 kDa,理论等电点为4.89。该蛋白可归于几丁质酶18族V类。运用制备芽生孢子转化法将Ifuchi1基因导入球孢白僵菌。最适产酶时间36 h条件下, 挑选的转基因菌株的几丁质酶活性相对于野生型菌株提高了86.2%;在对马尾松毛虫的生物测定中,在孢子浓度为1×107个孢子/mL时,与野生型菌株Bb13相比,转基因菌株的致死中时间LT50平均缩短了29%,同时死亡率提高了52.9%;野生型菌株和转基因菌株致死中浓度LC50分别为4.71×106 个孢子/ mL和1.74×106 个孢子/ mL,降低了约1.7倍的剂量,故通过基因工程方法获得的转基因工程菌株显著地提高了球孢白僵菌的杀虫效率。  相似文献

The influence of six chemical insecticides on growth, sporulation, conidial germination and cuticle-degrading enzyme production by Isaria fumosorosea were investigated under laboratory conditions. Maximum reduction in vegetative growth, sporulation and conidial germination in relation to the control treatment was observed for Chloranthraniliprole whereas Indoxacarb proved to be the safest insecticide causing lowest reduction in these parameters. Chloranthraniliprole, Chlorpyrifos and Chlorfenapyr caused higher reduction in enzyme activities (chitinase, Pr1, Pr2 and lipase) at all three concentrations whereas very low reduction in enzyme activities was caused by Hubendamide+ Avermectin and Indoxacarb when used at 10 µg/ml. The data presented can be used for future recommendations of these insecticides in IPM programmes where I. fumosorosea is an important control agent.  相似文献

Large amounts of conidia must be recovered to use Isaria fumosorosea as a biological control agent. Aerial conidia are mass-produced in solid substrate culture. Conidia obtained from growth on different substrates must be similar in quantity and quality (viability, purity and virulence). In the present work, several solid substrates were evaluated: rice, sorghum, sugarcane, corncob, plantain and wheat (derived from the production of Trichogramma sp.) to find alternatives that are economical and available in different regions. The highest conidia production was obtained on rice, plantain and corncob substrates, which yielded 5.33 × 109 conidia/g, 5.24 × 109 conidia/g and 4.8 × 109 conidia/g substrate, respectively. At concentrations of 1 × 107 conidia/mL, the mortality rates obtained with conidia developed on rice, plantain and corncob substrates were 98%, 97% and 88%, respectively. The lethal concentration 50 (LC50) values of conidia obtained from rice and plantain were 7.2 × 104 conidia/mL and 5.1 × 104 conidia/mL, respectively.  相似文献

Catalases are the most important enzymatic systems used to degrade hydrogen peroxide (H2O2) into water and oxygen, thereby lowering intracellular hydrogen peroxide levels. Entomopathogenic fungi display increased catalase activity during germination and growth, which is necessary to counteract the hyperoxidant state produced by oxidative metabolism. We studied the influence of five different hydrocarbons on catalase production by Lecanicillium muscarium to determine the importance of catalase induction in fungal germination, stress tolerance and virulence. Conidia produced by colonies grown on different hydrocarbons showed higher rates of catalase activity compared to the control and the catalase activity of conidia produced on n-octacosane was three times higher than the activity of the control. This increase in catalase activity was accompanied by a higher level of resistance to exogenous hydrogen peroxide and a reduction in the germination time. Our study has helped to identify that increased catalase activity improves the germination and tolerance to different antioxidant stress response of L. muscarium.  相似文献

The imperfect fungus Metarhizium anisopliae infects and kills larvae of many insect species, including the mosquito Culex pipiens. Mutants of M. anisopliae selected for enhanced production of amylase have been found to have simultaneously acquired hypervirulence against C. pipiens larvae. In the present work, wild-type and some mutant strains of M. anisopliae were found to excrete an acid or acids which alter the pH of fungal cultures below that permissible for amylase activity. The amylase-enhanced hypervirulence mutants did not excrete acid. Detection of protease and lipase activities was complicated by the acid excretion. When this was taken into account, mutants having altered lipase or protease production were found to have no alteration in virulence against mosquito larvae. The link between acid excretion, amylase activity, and hypervirulence is discussed.  相似文献

Effects of Ca2+ ions on the intensity of lipid peroxidation, activities of guaiacol peroxidase, superoxide dismutase (SOD), and catalase, as well as on heat resistance of winter wheat (Triticum aestivium L.) coleoptiles were examined. A preliminary incubation of coleoptile segments in a 5 mM CaCl2 solution was shown to improve their survival rates after an injuring heat treatment (43.5°C). The effect of Ca2+ was suppressed by the inhibitor of Ca2+ channels (1 mM LaCl3). An incubation of coleoptiles in the presence of 5 mM CaCl2 prior to the stress treatment elevated the content of lipid peroxidation product, malondialdehyde (MDA) and stimulated the activities of guaiacol peroxidase, SOD, and catalase. After the heat exposure of untreated and Ca2+-treated seedlings, differential changes in MDA content and in activities of guaiacol peroxidase, SOD, and catalase were observed.
