In this study, we investigated whether disruption of Na+ and Ca2+ homeostasis via activation of Na+-K+-Cl cotransporter isoform 1 (NKCC1) and reversal of Na+/Ca2+ exchange (NCXrev) affects protein aggregation and degradation following oxygen–glucose deprivation (OGD). Cultured cortical neurons were subjected to 2 h OGD and 1–24 h reoxygenation (REOX). Redistribution of ubiquitin and formation of ubiquitin-conjugated protein aggregates occurred in neurons as early as 2 h REOX. The protein aggregation progressed further by 8 h REOX. There was no significant recovery at 24 h REOX. Moreover, the proteasome activity in neurons was inhibited by 80–90% during 2–8 h REOX and recovered partially at 24 h REOX. Interestingly, pharmacological inhibition or genetic ablation of NKCC1 activity significantly decreased accumulation of ubiquitin-conjugated protein aggregates and improved proteasome activity. A similar protective effect was obtained by blocking NCXrev activity. Inhibition of NKCC1 activity also preserved intracellular ATP and Na+ homeostasis during 0–24 h REOX. In a positive control study, disruption of endoplasmic reticulum Ca2+ with thapsigargin triggered redistribution of free ubiquitin and protein aggregation. We conclude that overstimulation of NKCC1 and NCXrev following OGD/REOX partially contributes to protein aggregation and proteasome dysfunction as a result of ionic dysregulation.  相似文献

The Indian meal moth (IMM), Plodia interpunctella (Hübner) (Lepidoptera: Pyralidae), engages in long-distance or foraging flights in the twilight hours of the scotophase when blue light dominates the irradiance spectrum of the sky. We tested the hypothesis that IMM uses wavelengths of visible blue/violet light as orientation cues that trigger phototactic responses. In four-choice laboratory experiments, blue light (400–475 nm) was significantly more effective than green (475–600 nm), orange (575–700 nm), or red (590–800 nm) light in attracting males and mated females. In subsequent experiments that tested light emitting diodes (LEDs) emitting peak wavelengths in the blue/violet-light range, the 405-nm 'violet' LED was significantly more effective than the 435-, 450-, or 470-nm 'blue' LED in attracting males as well as virgin and mated females. In electroretinogram recordings, the 405-nm wavelength elicited significantly stronger receptor potentials from female and male eyes than the 350-nm (UV) wavelength, and in a behavioral experiment it significantly enhanced the known attractiveness of UV light. Equal attraction of IMMs to 405-nm LEDs at 600–700 µW/cm2 with or without UV light, and significantly stronger attraction to a 405-nm LED than to a 350-nm LED at maximum light intensities, suggest that the deployment of violet instead of UV light could become one of several management tactics for control of IMMs.  相似文献

1. Lake Titicaca is a large, high altitude (3810 m a.s.l.) tropical lake (16°S, 68°W) that lies on the border of Bolivia and Perú, receiving high fluxes of ultraviolet radiation (UVR) throughout the year. Our studies were conducted during September of 1997 with the main objective of studying the impact of solar UVR upon phytoplankton photosynthesis.
2. Water samples were taken daily and incubated in situ (down to 14 m depth) under three radiation treatments to study the relative responses to PAR (Photosynthetic Available Radiation, 400–700 nm), UV-A (320–400 nm), and UV-B (280–320 nm) radiation.
3. Photosynthetic inhibition by UVR in surface waters was about 80%, with UV-A accounting for 60% and UV-B for 20%; the inhibition by high levels of PAR was less than 20%. The inhibition due to UVR decreased with depth so that there were no significant differences between treatments at 8.5 m depth.
4. The amount of inhibition per unit energy received by phytoplankton indicates that even though there was a significant inhibition of photosynthesis due to UVR, species in Lake Titicaca seem to be better adapted than species in high latitude environments.
5. The cellular concentration of UV-absorbing compounds, a possible mechanism of photoadaptation, was low in phytoplanktonic species. However, they were abundant in zooplankton, suggesting a high rate of bioaccumulation through the diet.  相似文献

Thylakoid dismantling is one of the most relevant processes occurring when chloroplasts are converted to non-photosynthetically active plastids. The process is well characterised in senescing leaves, but other systems could present different features. In this study, thylakoid dismantling has been analysed in dividing cells of the unicellular alga, Euglena gracilis , cultured in darkness. Changes in photosynthetic pigments and in the abundance of LHC and PSII core proteins (D2 and CP43) showed that: (i) during the 0–24 h interval, the decline in LHCII was faster than that in the PSII core; (ii) during the 24–48 h interval, PSII and LHCII were strongly degraded to nearly the same extent; (iii) in the 48–72 h interval, the PSII core proteins declined markedly, while LHCII was maintained. These changes were accompanied by variations in room temperature fluorescence emission spectra recorded from single living cells with a microspectrofluorimeter (excitation, 436 nm; range 620–780 nm). Emission in the 700–715 nm range was proposed to derive from LHCI-II assemblages; changes in emission at 678 nm relative to PSII matched PSII core degradation phases. Overall, the results suggest that, in degreening E. gracilis , thylakoid dismantling is somewhat different from that associated with senescence, because of the early loss of LHCII. Moreover, it is proposed that, in this alga, disruption of the correct LHCI-II stoichiometry alters the energy transfer to photosystems and destabilises membrane appression leading to the thylakoid destacking observed using transmission electron microscopy.  相似文献

In Antarctica ozone depletion is highest during spring, coinciding with the reproduction of many seaweed species. Propagules are the life-stage of an alga most susceptible to environmental perturbations. Therefore, fertile thalli of Iridaea cordata (Turner) Bory (Rhodophyta) were collected in the eulittoral of King George Island (Antarctica) to examine spore susceptibility to ultraviolet radiation (UVR). In the laboratory, freshly released tetraspores were exposed to photosynthetically active radiation (PAR) (400–700 nm), PAR+UV-A (320–700 nm) or PAR+UV-A+UV-B (280–700 nm). Photosynthetic efficiency was measured during 1–8 h of exposure and after 48 h of recovery. Additionally, mycosporine-like amino acids (MAAs) and DNA damage were determined. Saturating irradiance of photosynthesis of freshly released tetraspores was 57 µmol photons m−2 s−1. Exposure to increasing fluence of PAR reduced photosynthetic efficiency. UVR further decreased the photosynthetic efficiencies of the tetraspores but spores were able to recover completely after UVR exposure and 2 days post-cultivation under low PAR. DNA damage was minimal and lesions were effectively repaired under photoreactivating light. Concentrations of the MAAs shinorine and palythine were higher in tetraspores treated with UVR than in spores only exposed to PAR. Generally, the tetraspores show a good UV tolerance. This flexible response of the tetraspores of this species to changing radiation conditions enables the alga to grow along a considerable depth gradient from the sublittoral to the eulittoral where they can be exposed to enhanced UVBR under conditions of stratospheric ozone depletion.  相似文献

The presence of pheromone in cotton foliage after removal of pheromone dispensers was assessed by measuring the airborne pheromone concentration with an electroantennogram device. Plots of 0.4 ha in isolated cotton fields were treated with Shin-Etsu PBW-Rope® pheromone dispensers for mating disruption of Pectinophora gossypiella (Saunders) (Lepidoptera: Gelechiidae). The dispensers contained ( Z , Z )- and ( Z , E )-7,11-hexadecadienyl acetates (gossyplure) in a 49:51 ratio and were applied at a density of 1 000/ha. The 400 pheromone dispensers were removed 1–12 days later. In four experiments involving canopy heights from 30–150 cm, the decay of the pheromone concentration was recorded repeatedly in short intervals for up to 7 h. Decay to undetectable concentrations generally occurred within 1–10 h, depending on plant size and wind conditions. In all four experiments, pheromone concentration 24 h after removal was found to be near or below detection threshold of the electroantennogram. The presence of pheromone within and above the cotton after dispenser removal would be due to re-entrainment of pheromone that had been adsorbed on cotton foliage or possibly some residual airborne pheromone.  相似文献

Serial transmission electron microscopy and 3D reconstruction were used to document cell morphology and position of the motoneurones innervating somites 1 and 2 of a 12.5-day amphioxus larva, of Branchiostoma floridae , and also those innervating the dorsal compartment of somites 3 through 6 of an 8-day larva. Motoneurones supplying the ventral and dorsal compartments can be distinguished from one another on a number of morphological criteria. The ventral compartment motoneurones are neither symmetrical nor particularly ordered in arrangement. Their cilia are short and point forward or obliquely across the central canal; their axons run along the basal lamina adjacent to processes from muscle fibres, with which they make extended linear series of synapses containing 45–60 nm synaptic vesicles. The dorsal compartment motoneurones are paired and tend to be positioned at or near the junctions between somites. Their cilia are longer and project caudally; their axons are large, filled with mitochondria and 30–45 nm synaptic vesicles, and make synapses only at specific, segmentally repeated sites.
  An unusual feature of both cell types is that synaptic input occurs all along the axon, either by direct axo-axonal synapses or via slender dendritic processes. This allows for redundancy and multiple inputs, and is possible only because amphioxus somatic motor axons lie entirely within the nerve cord, which is itself an unusual feature among chordates. The possible significance of dual somatic innervation is discussed in relation to the dual innervation of the head in vertebrates, which has separate sets of somatic and visceral/branchiomotor nerves.  相似文献

Alteration of 'normal' levels of ultraviolet-B light (UV-B, 280–320 nm) can affect plant chemical composition as well as growth; however, little is known about how plants perceive UV-B light. We have carried out fluence response curves, and demonstrated that the growth inhibition of etiolated Arabidopsis thaliana seedlings by low fluence UV light is specific to UV-B and not UV-A (320–390 nm). The response shows reciprocity between duration and intensity, at least over a limited range, and thus depends only on photon fluence and not on photon flux. The action spectrum for this response indicates a peak of maximum effectiveness at 290 nm, and response spectra at different fluences indicate that the most effective wavelength at 30 000  µ mol m–2 is 290 nm, whereas 300 nm light was the most effective at 100 000  µ mol m–2. This response occurs in mutant seedlings deficient in cryptochrome, phytochrome or phototropin, suggesting that none of the known photoreceptors is the major UV-B photoreceptor. Some null mutants in DNA repair enzymes show hypersensitivity to UV-B, suggesting that even at low fluence rates, direct damage to DNA may be one component of the response to UV-B.  相似文献

Ring Ouzel Turdus torquatus breeding site occupancy in the Moorfoot Hills, southeast Scotland, was determined during the periods 1952–85 and 1998–2000, and sites were classed as either occupied or deserted during the latter period. Site occupancy during 1998–2000 was related to habitat data derived from a supervised classification of a Landsat 7 satellite image. Breeding sites were more likely to have remained occupied if they were at higher altitudes and had more heather cover within radii of both 200 and 450 m, although these effects became non-significant when spatial autocorrelation was accounted for. Using only topographical variables, potential breeding sites were predicted and the habitat surrounding them was compared with actual breeding sites. Actual breeding sites were more likely to have heather or grass–heather mosaic within 100 m than potential breeding sites, but there were no further habitat differences between actual and potential breeding sites within radii of 200 or 450 m. These results indicate that breeding sites at higher altitudes and with a higher cover of heather were less likely to have been deserted, and that lack of heather may prevent otherwise suitable breeding sites from becoming occupied by Ring Ouzels. More detailed field studies are needed to understand the mechanisms underlying these associations.  相似文献

The disruption of the trail-based communication system of eastern tent caterpillars (ETC) ( Malacosoma americanum ) and forest tent caterpillars (FTC) ( M. disstria ) by spraying host trees with a trail pheromone mimic caused the disintegration and collapse of young colonies. A 1-ppm formulation of 5β-cholestan-3-one or a control formulation was sprayed on trees either before or after eclosion of the caterpillars in the spring. For pre-eclosion trials, an average of 92% of the FTC colonies on control trees, but only 17% of colonies on treated trees consisted of viable, cohesive aggregates when survival was assessed 5–23 days post-spray. The remaining colonies had either completely disappeared or were reduced to small, disintegrated fragments. For the ETC, 100% of colonies on control trees and an average of 12% of colonies on treated trees consisted of cohesive aggregates inhabiting tents when survival was assessed 3–41 days post-spray. The remaining colonies were completely destroyed, reduced to scattered individuals, or to small, disintegrated, shelter-less fragments. For post-eclosion trials with the FTC in which colonies were sprayed during their first or second larval stadium, an average of 63% of individuals in control groups but only 7% in treatment groups were still on the sprayed trees 5–18 days post-spray. For the ETC, colonies were largely unaffected when trees were not sprayed until after the caterpillars had eclosed and established tents. For all trials, the loss of individuals on treated trees was largely attributable to the inability of dispersed and isolated first or second instar caterpillars to maintain secure purchase, causing them to fall from trees and perish. To our knowledge, this is the first study to investigate the potential of manipulating populations of pest species by interfering with a trail-based, chemical communication system.  相似文献

Because of high tendency of the prion protein (PrP) to aggregate, the exact PrP isoform responsible for prion diseases as well as the pathological mechanism that it activates remains still controversial. In this study, we show that a pre-fibrillar, monomeric or small oligomeric conformation of the human PrP fragment 90–231 (hPrP90–231), rather than soluble or fibrillar large aggregates, represents the neurotoxic species. In particular, we demonstrate that monomeric mild-denatured hPrP90–231 (incubated for 1 h at 53°C) induces SH-SY5Y neuroblastoma cell death, while, when structured in large aggregates, it is ineffective. Using spectroscopic and cellular techniques we demonstrate that this toxic conformer is characterized by a high exposure of hydrophobic regions that favors the intracellular accumulation of the protein. Inside the cells hPrP90–231 is mainly compartmentalized into the lysosomes where it may trigger pro-apoptotic 'cell death' signals. The PrP toxic conformation, which we have obtained inducing a controlled in vitro conformational change of the protein, might mimic mild-unfolding events occurring in vivo, in the presence of specific mutations, oxidative reactions or proteolysis. Thus, in light of this model, we propose that novel therapeutic strategies, designed to inhibit the interaction of the toxic PrP with the plasmamembrane, could be beneficial to prevent the formation of intracellular neurotoxic aggregates and ultimately the neuronal death.  相似文献

Some physicochemical properties and the microstructure of heat-induced aggregates of globulin from common buckwheat (Fagopyrum esculentum Moench) (BWG) formed at 100 °C in 0.01 M phosphate buffer containing 1.0 M NaCl, pH 7.4 were studied. Differential scanning calorimetric (DSC) analysis shows a re-distribution of native and extensively denatured proteins in the heat-induced aggregates of BWG, particularly in the ISA fraction. Sodium dodecyl sulfate polyacrylamide gel electrophoretic (SDS-PAGE) analysis suggests the occurrence of both dissociation and association of molecules and the involvement of intermolecular disulfide linkages during thermal aggregation. Transmission electron microscopy (TEM) reveals that native BWG appeared as uniform compact globules with diameters ranging between 11.7 and 12.5 nm. TEM examination of the buffer-soluble aggregates, fractionated by sucrose density gradient ultracentrifugation, demonstrates the formation of strand-like small aggregates and large compact globular soluble macroaggregates.  相似文献

This study has utilized the tools of lipid biomarker chemistry and molecular phylogenetic analyses to assess the archaeal contribution to diversity and abundance within a microbial mat and underlying sediment from a hypersaline lagoon in Baja California. Based on abundance of ether-linked isoprenoids, archaea made up from 1 to 4% of the cell numbers throughout the upper 100 mm of mat and sediment core. Below this depth archaeal lipid was two times more abundant than bacterial. Archaeol was the primary archaeal lipid in all layers. Relatively small amounts of caldarchaeol (dibiphytanyl glyceroltetraether) were present at most depths with phytanyl to biphytanyl molar ratios lowest (~10 : 1) in the 4–17 mm and 100–130 mm horizons, and highest (132 : 1) in the surface 0–2 mm. Lipids with cyclic biphytanyl cores were only detected below 100 mm. A novel polar lipid containing a C30 isoprenoid (squalane) moiety was isolated from the upper anoxic portion of the core and partially characterized. Hydrocarbon biomarker lipids included pentamethylicosane (2–10 mm) and crocetane (primarily below 10 mm). Archaeal molecular diversity varied somewhat with depth. With the exception of samples at 0–2 mm and 35–65 mm, Thermoplasmatales of marine benthic group D dominated clone libraries. A significant number of phylotypes representing the Crenarchaeota from marine benthic group B were generally present below 17 mm and dominated the 35–65 mm sample. Halobacteriaceae family made up 80% of the clone library of the surface 2 mm, and consisted primarily of sequences affiliated with the haloalkaliphilic Natronomonas pharaonis .  相似文献

Complex life strategies are common among plant pathogens belonging to rust fungi ( Uredinales ). The heteroecious willow rust Melampsora larici-epitea produces five spore stages and alternates on larch ( Larix ). To shed light on the epidemiology of this pathogen, amplified fragment length polymorphisms (AFLPs) were used to determine the genetic diversity and genetic structure of rust samples collected from coppice willow ( Salix ) plantations at three UK sites (LA, CA and MC) over three sampling dates (September 2000, July 2001 and September 2001). Of the total of 819 isolates, 465 were unique AFLP phenotypes and there was a shift in genotype diversity between the two seasons (0.67 in 2000 and 0.87–0.89 in 2001). No phenotypes were common between the two seasons within a site, suggesting that the rust did not overwinter as an asexual stage within plantations. A temporal analysis detected large amounts of genetic drift ( F S = 0.15–0.26) between the two seasons and very small effective population sizes ( N e  =  2–3) within sites. These results all point to a new colonization of the plantations by the rust in the second season (2001). The F ST-analogue values were ΦCT = 0.121, Weir and Cockerham's θ = 0.086 and the Bayesian estimate θB = 0.087–0.096. The results suggest that the sources of inoculum were somewhat localized and the same sources were mainly responsible for disease epidemics in LA and CA over the two seasons. The relatively low F ST-values among sites (0.055–0.13) suggest the existence of significant gene flow among the three sampled sites.  相似文献

The population genetic profiles of 60 specimens of thecommon intertidal sea anemone Actinia equina taken from foursampling sites (Peroj, Barbariga and the north and south shoresof the Limski Canal), along a 25 km strip of the coastof the Istra Peninsula, Croatia, were evaluated by AFLP markers(144 loci). All populations were characterized by aggregates ofspecimens confined within a few square meters. One of the populations(on the north shore of the canal) contained both large and small-sizedindividuals. Results revealed high within-aggregate genetic heterogeneity(36.1–55.6%). With respect to interpopulationprofiles, geographically remote populations from Peroj, Barbarigaand the south shore of the canal were more closely related geneticallythan those on the north and south shores of the canal. Abstract:  Data were obtained from mark recapture trials pertaining to the dispersal of medfly, Ceratitis capitata (Dipt., Tephritidae), over both short (10–160 m) and very long distances (0.5–9.5 km) within the surveillance trapping array in Adelaide, Australia. They could be related to previously reported data sets by expressing the capture rates of each set in common terms that corrected for differences in recapture rate resulting from type of trap, season or climate. The mean capture rate at each distance from the point of release in each data set was expressed as a percentage of the real or inferred rate of that set at a distance of 100 m. The resulting distribution of dispersal distances conformed to both an inverse power model and a modified Cauchy model regardless of whether the present and previous data were combined or not. The modified Cauchy model inferred that the median distance flown was extremely short and 90% of flies displaced only 400–700 m despite the fact that a consistent trend in declining catch rates was obtained up to 9.5 km. The spread of invading propagules in quarantined zones in the first generation is likely to be limited by a decline to non-viable density within 1 km or less of the incursion point and the spread of larger infestations could be limited by the longevity of the dispersers. The results also have significance to the ability of surveillance trapping arrays to detect infestations and also to methods of distributing insects for the 'sterile insect technique'.  相似文献

As part of a larger comparative phylogeographical study of Philippine fruit bats, I used fragments of the mitochondrial genes cytochrome  b and ND2 to investigate phylogeography and diversification in Haplonycteris fischeri , a pteropodid bat endemic to the Philippines but widespread within the archipelago. Genetic diversity in H. fischeri was extremely high in these commonly studied genes, with 101 unique haplotypes in 123 sequenced individuals, although small, continuously isolated islands had less diversity than had large island complexes. Seven monophyletic groups and one paraphyletic group were restricted to individual islands, groups of islands, or parts of islands. Each Pleistocene island complex had a single resident monophyletic lineage; these five groups were separated by approximately 6–8% sequence divergence and apparently have been diverging for 4–6 Myr. Within island groups, monophyletic lineages on some individual islands suggest that current ocean channels have also been barriers to gene flow; in some cases, multiple allopatric clades were present on single islands. Basal divergence dates were estimated to be in the early Pliocene, and most diversification was apparently connected to the ongoing geological evolution of the Philippines. Abstract. Measurements of the photosynthetic photon flux density (400–700 nm) and of the spectral distribution of photon flux density across the 370–800 nm waveband, were made under both clear and overcast sky conditions above and at various positions within two Pinus radiata canopies of different stocking but similar leaf area indices. The spectra obtained for the daylight conditions (i.e. above forest canopy) were generally similar to those published previously. The spectra for shadelight within the forest canopy showed no blue peak which was characteristic of previously reported measurements which were restricted to the diffuse radiation component. There was almost neutral absorption within the 400–700 nm waveband, and typical lower attenuation in the 700–800 nm waveband. The blue: red ratio was largely unchanged by either canopy type or sky conditions and varied between 0.57 and 0.81. The red: far-red ratio in shadelight was between 0.22 and 0.41 under clear sky and between 0.68 and 0.95 under overcast sky conditions. Values for daylight were between 1.16–1.22. Calculated phytochrome photoequilibrium values in shadelight were approximately 0.35 under clear sky and 0.46 under overcast sky conditions. In each case there appeared to be no differences between the two canopies with respect to these minimum values.  相似文献
