From a (GT)13-enriched genomic library of Oplegnathus fasciatus, 14 polymorphic microsatellite were isolated and characterized in a test population with alleles ranging from two to nine, the observed and expected heterozygosities from 0.0000 to 1.0000, and from 0.1726 to 0.8507, respectively. Five loci deviated from the Hardy–Weinberg equilibrium, and linkage disequilibrium between two loci was significant. Two loci were also polymorphic in Pagrosomus major assessed for cross-species amplification. These polymorphic microsatellites will be useful for genetic diversity analysis and molecule-assisted breeding for O. fasciatus. Changwei Shao contributed equally.  相似文献

Lymphocytotoxic reagents allowed us to define 11 lymphocyte factors in sheep. After a genetic control of monospecificity had been made, a genetic study based on 400 crossings indicated that nine factors are transmitted in two haplotypes containing 0.1 or 2 factors. The linked factors of the bifactorial haplotypes are the products of two genes at two closely linked loci,OLA- A andOLA- B, having a recombination rate of about 0.6%. The nine factors are the products of fiveOLA- A and fourOLA- B alleles. Allelic frequencies at the two loci, ranging from 0.05 to 0.23 were obtained by two methods. A linkage disequilibrium between the loci is described. Among the possible causes of this, inbreeding is excluded, whereas selective pressure or more probably old fusions between sheep populations must be retained. A tenth factor (frequency 0.33 to 0.35) has a 26% recombination with OLA factors; its locus (OL-X) would be on the same chromosome but very far from theOLA complex. An 11th factor (frequency 0.30) is probably independent ofOLA; its locus (OL-Z) may be on another chromosome. The two linkedOLA loci give evidence of a major histocompatibility complex in sheep; the observed linkage disequilibrium allows one to foretell certain applications in phylogeny and perhaps in selection.  相似文献

We have isolated 16 microsatellite loci in the field cricket, Gryllus bimaculatus. Nine loci were found to be polymorphic in G. bimaculatus and the number of alleles varied from seven to 14. All 16 loci were tested for amplification in nine other species. In the five species tested belonging to the same subfamily (Gryllinae), a minimum of nine loci amplified. These loci will be used to determine paternity as part of a study to investigate the genetic benefits of polyandry.  相似文献

We have identified 15 polymorphic microsatellite loci for the barn owl (Tyto alba), five from testing published owl loci and 10 from testing non‐owl loci, including loci known to be of high utility in passerines and shorebirds. All 15 loci were sequenced in barn owl, and new primer sets were designed for eight loci. The 15 polymorphic loci displayed two to 26 alleles in 56–58 barn owls. When tested in 10 other owl species (n = 1–6 individuals), between four and nine loci were polymorphic per species. These loci are suitable for studies of population structure and parentage in owls.  相似文献

Eurytoma brunniventris is a parasitic wasp with a wide range of host species. We have developed primers for nine polymorphic microsatellite loci to allow examination of intraspecific population subdivision associated with two aspects of the biology of their hosts: host species (oak gallwasps) and the tree on which the gallwasp develops (different oak species). All nine loci amplified well across individuals collected from a range of gallwasp species and across two oak taxa. These microsatellite loci are potentially of value in the study of closely related economic pests such as seed predators of almonds (E. amygdali) and pistachios (E. plotnikovi).  相似文献

Kearney's bluestar (Amsonia kearneyana) is a highly endangered herbaceous perennial in the family Apocynaceae. The species is found only in the Baboquivari Mountains of southern Arizona. We report the isolation and development of 12 microsatellite loci for Kearney's bluestar. Numbers of alleles ranged from two to four and observed heterozygosities ranged from 0.20 to 0.80 in the nine loci found to be polymorphic in the test population. All loci were also tested for cross‐amplification in five other Amsonia species representing two subgenera from the southwestern United States. Some loci that were not polymorphic in the Kearney's bluestar were polymorphic in other species.  相似文献

We characterize 15 microsatellite loci from two microsatellite libraries developed from the Australian agamid lizards Amphibolurus muricatus and Ctenophorus pictus. All loci were tested for amplification in four other agamids: Ctenophorus fordi, Ctenophorus decresii, Chlamydosaurus kingii, and Physignathus lesueurii. These loci were highly polymorphic within and across species, with nine to 12 loci amplifying for each species tested.  相似文献

Primers have been developed for nine polymorphic tetranucleotide microsatellite loci isolated from the swift fox (Vulpes velox). These can be amplified in three multiplex reactions and all loci can be scored easily and without ambiguity. The number of alleles per locus ranged from three to 14 and the overall heterozygosities ranged from 0.54 to 1.00. All loci were in Hardy‐Weinberg equilibrium, but two loci were in linkage disequilibrium. These loci will be used to monitor endangered swift fox populations on the Canadian prairies.  相似文献

A total of nine polymorphic microsatellite loci were obtained from a genomic library of Pangasius pangasius (order Siluriformes, family Pangasiidae). Samples from rivers Bhagirathi (n = 22) and Mahanadi (n = 20) were genotyped for each of the nine microsatellite loci to determine genetic variation. The mean number of alleles per locus was 5.22 in Bhagirathi and 5.78 in Mahanadi; and expected heterozygosity ranged from 0.567 (Bhagirathi) to 0.578 (Bhagirathi). Significant deviation (P < 0.003) from Hardy–Weinberg expectations was evident at three loci, Ppa2 (Bhagirathi), Ppa14 (Mahanadi) and Ppa28 (Bhagirathi and Mahanadi). The identified microsatellite loci were found to be promising for population genetics studies of P. pangasius.  相似文献

Microsatellites represent an important tool for characterising population structure, for attributing individuals to stocks, and for revealing ecological processes taking place on population and meta-population levels. A sound knowledge of population structure is essential for sustainable management of exploited fish stocks, and helps to understand population connectivity and speciation. We developed for the first time primers for microsatellite loci in the carapine grenadier, Coryphaenoides carapinus, inhabiting the abyssal Atlantic. Eleven microsatellites were obtained from partial genomic DNA libraries enriched for tetranucleotide repeats. The loci were characterised in three unrelated individuals and nine loci were found to be polymorphic. Cross-amplification in two commercially exploited deep-sea macrourid species (Coryphaenoides rupestris and Macrourus berglax) resolved two polymorphic loci in each species.  相似文献

Microsatellite loci were developed from hoary marmots (Marmota caligata) to aid in the investigation of the social structure and mating system of this species. Seven of the microsatellite loci developed were found to be moderately polymorphic with between two and seven alleles per locus. In addition to the microsatellites developed in hoary marmots we also tested markers developed for other scuirids, namely European alpine marmots (M. marmota), Columbian ground squirrels (Spermophilus columbianus) and European ground squirrels (S. citellus). Of these markers, 13 were polymorphic when amplified in hoary marmots with between two and nine alleles per locus.  相似文献

Primers for five polymorphic microsatellite loci were developed for the midget faded rattlesnake (Crotalus viridis concolor), a rare subspecies of western rattlesnake (Crotalus viridus) found only in parts of Wyoming, Colorado, and Utah. Five polymorphic microsatellites were isolated, four of which had relatively high levels of diversity (eight to nine alleles). We found only two departures from Hardy–Weinberg equilibrium and they occurred in different loci, so null alleles are likely not a problem. Moreover, we found that no two loci were linked. These loci will be applicable for population genetic analysis and perhaps analysis of paternity and mating systems.  相似文献

We isolated and characterized 16 microsatellite loci from the blacktip shark, Carcharhinus limbatus, and tested cross‐species amplification in 11 Carcharhinus species and five additional shark genera. Thirty‐six (1.6%) and 180 (48%) colonies were positive for dinucleotide repeat motifs from unenriched and enriched libraries, respectively. Heterozygosities of polymorphic loci ranged from 0.04 to 0.96 with two to 22 alleles per locus. Amplification products were observed at nine to 13 loci (five to 11 of which where polymorphic) in 10 Carcharhinus species. Several loci were also polymorphic in each of the additional genera examined.  相似文献

Nine microsatellite loci were isolated from the insect Dolichogenidea homoeosomae (Hymenoptera: Braconidae), an important parasitoid of the sunflower moth Homosoeosoma electellum (Lepidoptera: Pyralidae), and assayed for polymorphism. All nine loci were polymorphic within the five populations tested, with two to 14 alleles per locus. Expected and observed heterozygosities ranged from 0.39 to 0.90 and 0.25 to 0.72 respectively. These are the first microsatellite primers developed for D. homeosomae and will be useful for studies of population dynamics and connectivity.  相似文献

Sea urchin is an important model organism for evolutionary biology, embryology, and gene regulation study. We developed and evaluated simple sequence repeat (SSR) markers from expressed sequence tags (ESTs) of Strongylocentrotus purpuratus and Hemicentrotus pulcherrimus. Characteristics of nine EST-SSR loci were investigated using 41 Hemicentrotus pulcherrimus individuals. The number of alleles per locus ranged from two to five. The observed heterozygosity ranged from 0.122 to 0.7805, while the expected heterozygosity ranged from 0.4472 to 0.7696. These loci and markers will be useful for population genetics and systemic evolution among species of sea urchin.  相似文献

Fourteen microsatellite loci were isolated from capybaras (Hydrochoerus hydrochaeris), which are large, diurnal social rodents that occur in the wet savannas of South America. Five of these loci were monomorphic. The remaining nine loci were polymorphic (two to seven alleles per locus), with observed levels of heterozygosity ranging from 0.063 to 0.800 (n = 17 animals). The latter loci provide a valuable tool for assessing patterns of parentage and kinship within capybara social groups.  相似文献

Six polymorphic simple sequence repeat (SSR) markers were developed for the ectomycorrhizal fungus Pisolithus microcarpus. A polymerase chain reaction (PCR)‐based technique was used in which random amplified polymorphic DNA (RAPD) fingerprints were probed with labelled SSR oligonucleotides by southern hybridization. The number of alleles per locus ranged from two to nine with expected heterozygosity values from 0.33 to 0.76. These loci will be potentially useful for genetic structure and gene flow studies of P. microcarpus populations. Cross‐species amplification with Pisolithus albus isolates at all loci was also observed.  相似文献

This study reports the isolation and characterization of seven polymorphic microsatellite loci in Citrus. The loci were isolated from two libraries constructed from genomic DNA nonenriched for TC and AC repeats and enriched for AC repeats. These markers yielded four to nine alleles per locus (mean 6.14) in a survey of 32 Citrus cultivars. Average observed heterozygosity ranged from 0.43 to 0.72. The levels of polymorphism found in this study suggest that these microsatellite loci can become an important tool for genetic studies in Citrus.  相似文献

Thirty‐nine polymorphic microsatellite DNA markers were isolated from the guppy (Poecilia reticulata) genomic library. All of the loci showed moderate allelic variation ranging from two to seven alleles, with observed heterozygosities from 0.000 to 0.938. The microsatellite DNA markers isolated will be available for use in analysis of quantitative trait loci in breeding programmes and for population genetic studies on experimental fish.  相似文献

We describe nine new polymorphic tetranucleotide microsatellite loci isolated from the fire‐bellied toad (Bombina bombina). The relative yield of new loci was higher than described in previous studies in amphibians: out of 12 loci initially evaluated, nine were polymorphic and amplifying reliably. Number of alleles ranged from four to 10 and observed heterozygosities from 0.47 to 0.91. Seven loci were polymorphic also in Bombina variegata and five in Bombina orientalis. Enrichment protocols yielding long flanking regions potentially overcome difficulties (i.e, low yield of reliable loci relative to number of clones screened) which have been reported in microsatellite development in anurans.  相似文献
