We report the isolation and characterization of polymorphic microsatellite loci for the Siberian flying squirrel Pteromys volans. The seven most useful loci had between six and 11 alleles and expected heterozygosities ranging from 0.477 to 0.866. We also tested the utility of these loci in other squirrel species, northern flying squirrels (Glaucomys sabrinus and G. volans) and the common red squirrel (Sciurus vulgaris). Three of the Siberian flying squirrel loci were polymorphic in other squirrel species, suggesting a limited potential for cross‐species use.  相似文献

Climate is an important factor limiting species distributions. Historic climate‐change related range movements have modified the genetic diversity of species by the merging and splitting of gene pools and by the effects associated with recurrent founder events. These effects are often inferred, either from retrospective analyses of current genetic patterns or from simulations. Rarely has it been possible for the population genetic effects of range expansion to be examined with contemporaneous demographic data. We characterized the genetic signature of rapid range expansion by southern flying squirrels (Glaucomys volans) and compared these results to a stationary population of the closely related northern flying squirrel (Glaucomys sabrinus) in Ontario, Canada. Samples were taken during an approximately 200 km range expansion by G. volans (1994–2003) and genotyped at 6 (G. sabrinus) and 8 (G. volans) microsatellite loci. For G. volans, but not G. sabrinus, we found evidence of a latitudinal gradient in allele frequencies and a decrease in allelic richness along the axis of expansion. We found no evidence of isolation‐by‐distance in either species or of genetic bottlenecks in the area of G. volans expansion. These results suggest that serial founder events can cause an immediate reduction in genetic diversity following rapid range expansion with high levels of gene flow giving rise to heterogeneity within what would classically be termed panmixia. Given the pace of anthropogenic climate change, and the increasing incidence of range movements in response, this may be an important, immediate consequence of climate change.  相似文献

We tested whether species-specific behavioral traits could explain patterns of habitat patch occupancy by five different squirrel species in Ontario, Canada: the northern and southern flying squirrel (Glaucomys sabrinus and Glaucomys volans), the North American red squirrel (Tamiasciurus hudsonicus), the eastern chipmunk (Tamias striatus), and the eastern gray squirrel (Sciurus carolinensis). Flying squirrel species exhibit group nesting in winter, which may put them at risk of extirpation in small patches with few individuals to contribute to group nests. Flying squirrels are also volant, potentially making non-treed matrix a barrier. Our surveys revealed that G. sabrinus was most likely to occur in large patches that were embedded in landscapes with low connectivity, and least likely to occur in small patches in highly connected landscapes. Conversely, G. volans was most likely to occur in large, well-connected patches and least likely to occur in small, unconnected patches. Patch occupancy by the cursorial squirrels was not strongly influenced by patch area or isolation. These findings reinforce previous studies suggesting that an understanding of species-specific traits such as behavior is an important consideration when interpreting habitat fragmentation effects.  相似文献

The PCR primers were tested on 21 terrapins from Cape Romain, SC, USA. We isolated 11 polymorphic microsatellite loci for wood stork (Mycteria americana). Polymerase chain reaction (PCR) primers and conditions are described for the amplification of five dinucleotide, one trinucleotide and five tetranucleotide microsatellite loci. The PCR primers were tested on two wood stork populations, Fazenda Ipiranga, Mato Grosso, Brazil (n = 11) and Tamiami West, Everglades, Florida, USA (n = 20). The primers yielded two to four alleles per locus, an observed heterozygosity of 0.0–0.727 and a polymorphic information content of 0.048–0.604. The low level of polymorphism for these markers is consistent with previous studies of this species.  相似文献

We describe polymerase chain reaction (PCR) primers and conditions to amplify 11 microsatellite DNA loci isolated from the oldfield mouse (Peromyscus polionotus subgriseus). These were tested for amplification using nine species and subspecies maintained at the Peromyscus Genetic Stock Center, with an average success rate of 65% and two loci amplifying in all species. Polymorphism was tested within the P. polionotus subgriseus and the recently obtained P. maniculatus sonorensis colonies. P. p. subgriseus had modest numbers of alleles per locus (1–4), whereas P. m. sonorensis had many alleles per locus (5–10) and high expected heterozygosities (0.625–0.878).  相似文献

Anecdotal reports of ultrasound use by flying squirrels have existed for decades, yet there has been little detailed analysis of their vocalizations. Here we demonstrate that two species of flying squirrel emit ultrasonic vocalizations. We recorded vocalizations from northern (Glaucomys sabrinus) and southern (G. volans) flying squirrels calling in both the laboratory and at a field site in central Ontario, Canada. We demonstrate that flying squirrels produce ultrasonic emissions through recorded bursts of broadband noise and time-frequency structured frequency modulated (FM) vocalizations, some of which were purely ultrasonic. Squirrels emitted three types of ultrasonic calls in laboratory recordings and one type in the field. The variety of signals that were recorded suggest that flying squirrels may use ultrasonic vocalizations to transfer information. Thus, vocalizations may be an important, although still poorly understood, aspect of flying squirrel social biology.  相似文献

We describe specific primers and conditions to amplify two dinucleotide and five trinucleotide microsatellite DNA loci isolated from the oomycete Phytophthora ramorum, the causal agent of sudden oak death. The primer sets were tested on 14–30 isolates from North America and Europe. Seven of 14 loci differentiated between A1 and A2 mating types. All seven loci successfully amplified DNA isolated from infected plant tissue. Four loci may be useful for the diagnosis of P. ramorum because they do not amplify closely related Phytophthora species.  相似文献

Siberian flying squirrel (Pteromys volans) has declined in Finland and it is considered an endangered species. We studied microsatellite variation in four flying squirrel populations in a fragmented landscape in Finland to determine the amount of gene flow and genetic diversity in the populations. Demographic data from these areas suggest that the populations are declining. All the populations are significantly differentiated (F ST = 0.23). The most notable result is the high degree of differentiation between two adjacent populations (F ST = 0.11) and low genetic variability (number of alleles 3.0) in one of the populations. These findings suggest problems in dispersal and possible fragmentation effects in the landscape where only 10–20% of habitat favorable for the flying squirrel is left. Conservation ensuring dispersal should be urgently considered. Future studies should concentrate on the modeling of the population viability and on the effects of inbreeding in these small populations. Summary The flying squirrel (Glaucomys volans) is a strongly nocturnal rodent. Previous anatomical observations suggested that the retina of this animal contains some cone-like receptors in addition to large numbers of rods. Evidence for duplicity of function in this visual system was obtained from an examination of three indices of visual activity: the electroretinogram (ERG), the isolated PIII retinal response, and the visually evoked cortical potential (VECP). The spectral sensitivity of the dark-adapted flying squirrel is similar to that of other mammals — it has a 500 nm peak (Figs. 3, 8). Responses of the ERG and isolated PIII to flickering light indicate the operation of two processes (Figs. 4, 7), one of which is unable to follow flickering light at repetition rates above 10–15 Hz. Spectral sensitivity measurements reveal that these two processes have different spectral sensitivities. The photopic mechanism in the flying squirrel visual system has peak sensitivity at about 520 nm (Figs. 5, 7, 9). The effects of steady light adaptation are much more obvious in the cortical potentials than they are in the retinal potentials.We thank David Birch for his advice and assistance. This paper reports the isolation and characterization of 24 polymorphic microsatellite markers in an important tropical timber species, Koompassia malaccensis (Leguminosae). The primers were designed from a genomic library enriched for dinucleotide (CT) repeats and screened on 24 samples from a natural population. The number of alleles detected per locus ranged from two to 13 while the observed heterozygosity ranged from 0.042 to 1.000. Significant departure from Hardy–Weinberg equilibrium (P < 0.05) was detected in two loci. These microsatellite markers were tested across 13 timber species of the same family. The amplification success appeared to be associated with taxonomy classification at the genus but not subfamily levels.  相似文献

The primers were tested on 22 individuals collected from a population located within southwestern Georgia (USA). Southern flying squirrels (Glaucomys volans) can have significant negative impacts on red-cockaded woodpecker (Picoides borealis) reproductive success and group size. Although direct control of southern flying squirrels may be necessary in small red-cockaded woodpecker populations (<30 groups), creation of high quality habitat through landscape management is the preferred method for managing larger woodpecker populations. Thus, we determined the habitat and landscape factors within 100 m, 400 m, and 800 m of cluster centers that were related to southern flying squirrel use of red-cockaded woodpecker cavities at the Carolina Sandhills National Wildlife Refuge, South Carolina. At all spatial scales, the number of cavities in the cluster was the most influential variable determining use by southern flying squirrels. At the 400-m and 800-m scales, the amount of stream length was also positively associated with the presence of flying squirrels. The proximity and amount of hardwoods surrounding clusters were not related to southern flying squirrel use at any spatial scale; thus, removal or conversion of hardwood stands surrounding red-cockaded woodpeckers may not be necessary for reducing cavity kleptoparasitism by flying squirrels. However, when establishing recruitment clusters, areas with streams should be avoided and addition of artificial cavities to existing clusters should be done judiciously to minimize the number of excess cavities. A total of 15 microsatellite primers pairs were developed for the Australian small‐eyed snake Rhinoplocephalus nigrescens. Five primers were used to screen 93 individuals of R. nigrescens and were also tested against eight species of the closely related genus Suta. Allelic diversity in R. nigrescens was high in three loci (12–27) and there was high heterozygosity (0.58–0.82). Observed heterozygosity did not deviate from Hardy–Weinberg expectations for the five loci tested. These primers will be useful in studies of population genetics and mating systems of small‐eyed snakes and related species.  相似文献

We report on the identification and characterization of two dinucleotide, two trinucleotide and eight tetranucleotide microsatellite DNA loci isolated from the European subterranean termite Reticulitermes santonensis. We tested the loci on 51–92 individuals from 46 colonies from different regions of France. Eleven loci were polymorphic with 2–8 alleles per locus and low observed heterozygosities (0.10–0.48). We also tested the loci on 17–20 individuals from 10 colonies in the closely related North American species R. flavipes and found significantly more alleles (2–9 alleles per locus) and higher observed heterozygosities (0.15–0.80) than in R. santonensis. The lower observed heterozygosities in R. santonensis are consistent with higher levels of inbreeding in these colonies due to the presence of numerous inbred replacement reproductives.  相似文献

We present 11 dinucleotide microsatellite DNA loci isolated from the narrow‐leaved cattail (Typha angustifolia) and describe conditions for their amplification. The PCR primers were tested on at least 20 individuals of Typha angustifolia and T. latifolia from two Ukrainian populations per species. The primers amplify loci with relatively high numbers of alleles (averaging 7.22 and 4.95 alleles per locus in T. angustifolia and T. latifolia, respectively), and polymorphic information content (averaging 0.61 and 0.46 in T. angustifolia and T. latifolia, respectively).  相似文献

We describe the cloning and characterization of five highly polymorphic microsatellite loci cloned from aduncus dolphins (Tursiops aduncus) from Western Australia. Five polymorphic microsatellite loci were isolated and tested on up to 350 animals, showing 7–23 alleles and expected heterozygosity values from 0.68 to 0.89. We also tested the loci on striped dolphins and franciscana dolphins, where we also found high levels of polymorphism (9–16 alleles in 102 striped, and 4–5 alleles in 13 franciscana dolphins). Considering that the cetacean genome is highly conserved, the characterized markers are likely to be useful in a number of cetacean population studies.  相似文献

We describe primers and polymerase chain reaction conditions to amplify 15 tetranucleotide microsatellite loci from the loggerhead sea turtle (Caretta caretta). The primers were tested on 30 individuals that nested along the Georgia, USA coast. The primer pairs developed in this study yielded an average of 13.9 alleles per locus (range of 10–21), an average observed heterozygosity of 0.91 (range 0.79–1.00), and an average polymorphic information content of 0.88 (range 0.84–0.92).  相似文献
