Roles of abscisic acid (ABA) in water stress-induced oxidative stress were investigated in leaves of maize ( Zea mays L.) seedlings exposed to water stress induced by polyethylene glycol (PEG 6000). Treatment with PEG at -0.7 MPa for 12 and 24 h led to a reduction in leaf relative water content (RWC) by 7.8 and 14.1%, respectively. Duration of the osmotic treatments is considered as mild and moderate water stress. The mild water stress caused significant increases in the generation of superoxide radical ( O 2 - ) and hydrogen peroxide (H 2 O 2 ), the activities of superoxide dismutase (SOD), catalase (CAT), ascorbate peroxidase (APX) and glutathione reductase (GR) and the contents of ascorbate (ASC), reduced glutathione (GSH). The moderate water stress failed to further enhance the capacity of antioxidant defense systems, as compared to the mild water stress. The contents of catalytic Fe, which is critical for H 2 O 2 -dependent hydroxyl radical ( •OH) production, and the oxidized forms of ascorbate and glutathione pools, dehydroascorbate (DHA) and oxidized glutathione (GSSG), markedly increased, a significant oxidative damage to lipids and proteins took place under the moderate water stress. Pretreatment with ABA caused an obvious reduction in the content of catalytic Fe and significant increases in the activities of antioxidant enzymes and the contents of non-enzymatic antioxidants, and then significantly reduced the contents of DHA and GSSG and the degrees of oxidative damage in leaves exposed to the moderate water stress. Pretreatment with an ABA biosynthesis inhibitor, tungstate, significantly suppressed the accumulation of ABA induced by water stress, reduced the enhancement in the capacity of antioxidant defense systems, and resulted in an increase in catalytic Fe, DHA and GSSG, and oxidative damage in the water-stressed leaves. These effects were completely prevented by addition of ABA, which raised the internal ABA content. Our data indicate that ABA plays an important role in water stress-induced antioxidant defense against oxidative stress.  相似文献

This study investigated the effects of exogenous hydrogen sulfide on the ascorbate and glutathione metabolism in wheat seedlings leaves under water stress. The results showed that pretreatment with sodium hydrosulfide (NaHS), hydrogen sulfide donor, increased the activities of ascorbate peroxidase, glutathione reductase, dehydroascorbate reductase and gamma-glutamylcysteine synthetase, and the contents of reduced ascorbic acid, reduced glutathione, total ascorbate and total glutathione under water stress, compared to control and water stress without NaHS. Meanwhile, pretreatment with NaHS decreased the malondialdehyde content and electrolyte leakage induced by water stress in plants, compared to control and water stress without NaHS. Our results suggested that exogenous hydrogen sulfide alleviated oxidative damage by regulating the ascorbate and glutathione metabolism in wheat seedlings under water stress.  相似文献

通过盆栽实验, 对干旱胁迫下黄土高原地区冰草(Agropyron cristatum)叶片的抗坏血酸和谷胱甘肽合成及循环代谢相关酶及物质含量进行了研究。结果表明: 冰草可以通过增强叶片的抗坏血酸和谷胱甘肽合成及循环代谢酶: 抗坏血酸过氧化物酶、谷胱甘肽还原酶、脱氢抗坏血酸还原酶、单脱氢抗坏血酸还原酶、L-半乳糖酸-1, 4-内酯脱氢酶和γ-谷氨酰半胱氨酸合成酶活性, 维持植物体内抗坏血酸和谷胱甘肽水平及氧化还原状态, 从而抵御干旱造成的氧化胁迫。但叶片抗坏血酸和谷胱甘肽合成及循环代谢对不同水平干旱胁迫的响应, 随胁迫时间的延长而不同。在胁迫24天以前, 严重干旱下叶片的抗坏血酸和谷胱甘肽合成及循环代谢增强较显著; 在胁迫24天后, 由于该胁迫下植物所遭受的氧化胁迫较为严重, 叶片中上述6种酶的活性均呈降低趋势。而在中度干旱下叶片抗坏血酸和谷胱甘肽合成及循环代谢相关的6种酶在整个胁迫过程中均保持较高的活性。这说明, 冰草能够长时间有效地抵御中度干旱所造成的氧化胁迫, 但只能在一定时间范围内有效地抵御严重干旱所造成的氧化胁迫, 胁迫时间延长则会降低其抵御严重干旱的能力。  相似文献

To investigate the possible mechanisms of glutathione reductase (GR) in protecting against oxidative stress, we obtained transgenic tobacco (Nicotiana tabacum) plants with 30–70% decreased GR activity by using a gene encoding tobacco chloroplastic GR for the RNAi construct. We investigated the responses of wild type and transgenic plants to oxidative stress induced by application of methyl viologen in vivo. Analyses of CO2 assimilation, maximal efficiency of photosystem II photochemistry, leaf bleaching, and oxidative damage to lipids demonstrated that transgenic plants exhibited enhanced sensitivity to oxidative stress. Under oxidative stress, there was a greater decrease in reduced to oxidized glutathione ratio but a greater increase in reduced glutathione in transgenic plants than in wild type plants. In addition, transgenic plants showed a greater decrease in reduced ascorbate and reduced to oxidized ascorbate ratio than wild type plants. However, there were neither differences in the levels of NADP and NADPH and in the total foliar activities of monodehydroascorbate reductase and dehydroascorbate reductase between wild type and transgenic plant. MV treatment induced an increase in the activities of GR, ascorbate peroxidase, superoxide dismutase, and catalase. Furthermore, accumulation of H2O2 in chloroplasts was observed in transgenic plants but not in wild type plants. Our results suggest that capacity for regeneration of glutathione by GR plays an important role in protecting against oxidative stress by maintaining ascorbate pool and ascorbate redox state.  相似文献

In excess, iron can induce the production and accumulation of reactive oxygen species (ROS), causing oxidative stress. The objective of this work was to evaluate the impact of toxic concentrations of iron (Fe) on the antioxidative metabolism of young Eugenia uniflora plants. Forty-five-day-old plants grown in Hoagland nutrient solution, pH 5.0, were treated with three Fe concentrations, in the form of FeEDTA, during three periods of time. At the end of the treatment, the plants were harvested and relative growth rate, iron content, lipid peroxidation and enzymes and metabolites of the antioxidative metabolism were determined. Iron-treated plants showed higher iron contents, reduced relative growth rates and iron toxicity symptoms in both leaves and roots. There was an increase in lipid peroxidation with increasing Fe, only in the leaves. The enzymatic activities of superoxide dismutase (SOD) and glutathione reductase (GR) increased with increasing Fe concentration and treatment exposure time. The activities of catalase (CAT), peroxidase (POX) and ascorbate peroxidase (APX) also increased with increasing Fe concentration but decreased with increasing treatment exposure time. Glutathione peroxidase activity (GPX) decreased with increasing Fe concentration and exposure time. The ascorbate (AA) and reduced glutathione (GSH) contents and the AA/DHA and GSH/GSSG ratios, in general, increased with increasing Fe concentration and treatment exposure time. The results indicate that under toxic levels of Fe, young E. uniflora plants suffer increased oxidative stress, which is ameliorated through changes in the activities of antioxidative enzymes and in the contents of the antioxidants AA and GSH.  相似文献

The purpose of this work was to evaluate the effect of 2,4-dichlorophenol (2,4-DCP) and pentachlorophenol (PCP) on the activity of antioxidative system and lipid peroxidation in the leaves of reed canary grass (Phalaris arudinacea). The activity of catalase (CAT), ascorbate peroxidase (APX), guaiacol peroxidase (GPX), glutathione reductase (GR) and glutathione S-transferase (GST) as well as the content of glutathione, ascorbate and phenolic compounds were determined. An induced-increase in the APX, CAT, GPX and GR activities was stronger for PCP, while a significant increase in the GST activity was noted only for 2,4-DCP. Both compounds increased the content of phenolic compounds, oxidized and reduced glutathione as well as the content of ascorbic acid. PCP induced stronger increase in lipid peroxidation than 2,4-DCP. The observed changes revealed that chlorophenols induce oxidative stress and oxidative damage in the leaves of reed canary grass.  相似文献

Bean (Phaseolus vulgaris L.) seedlings were subjected to varying selenium levels (1, 2, 4, and 6 ppm) in a hydroponic culture. The germination reached 100% in 48 h in all Se levels except 6 ppm, where it took 72 h. The root and shoot growth was stimulated at 1 and 2 ppm Se levels that was commensurate with increase in chlorophyll content, leaf water content, and cellular respiration. At 4 and 6 ppm Se levels, the growth was inhibited appreciably, which was associated with increase in stress injury measured as damage to membranes and decrease in cellular respiration, chlorophyll, and leaf water content. The oxidative injury as elevation of lipid peroxidation was larger compared to hydrogen peroxide accompanied by reduced levels of enzymatic (superoxide dismutase, catalase, ascorbate peroxidase, and glutathione reductase) and non-enzymatic (ascorbic acid and glutathione) antioxidants. Proline content was significantly higher at 1 and 2 ppm Se but diminished considerably at 4 and 6 ppm levels concomitant with the reduced growth. Exogenous application of proline (50 μM) resulted in substantiation of its endogenous levels that antagonised the toxic effects of Se by improving the growth of seedlings. The stress injury was reduced significantly with simultaneous increase in enzymatic and non-enzymatic antioxidants. Especially the components of ascorbate–glutathione cycle showed larger stimulation with proline application. The role of proline in mitigating the toxic effects of Se is discussed.  相似文献

The effects of overwatering (flooding) on the oxidative potential, the level of low-molecular-weight antioxidants, the content of stress proteins, and activities of antioxidant enzymes in green barley (Hordeum vulgare L.) seedlings were studied. Overwatering retarded barley seedling growth and induced hydrogen peroxide accumulation, a decrease in the total ascorbate content and an increase in the content of reduced glutathione (GSH), but it did not affect the content of oxidized glutathione (GSSG). After the cessation of stress factor action (post-stress period), the content of hydrogen peroxide declined to the initial level, the content of ascorbate reduced still stronger, whereas the content of GSH continued to rise. Under flooding conditions, activities of glutathione reductase (GR) and superoxide dismutase (SOD) increased. After the cessation of stress factor action, activities of these enzymes decreased but remained at rather high levels as compared with control. Activity of catalase (CAT) reduced during stress, whereas activity of ascorbate peroxidase (APX) was not essentially changed. In the post-stress period, CAT activity remained to be low; in contrast, APX activity increased. Barley seedling flooding induced the synthesis of stress proteins, HSP70 and dehydrins (DH). In the post-stress period, the content of stress proteins decreased; however, the content of DH in experimental leaves remained rather high. The results obtained indicate that barley defense system manifested a complex response to overwatering, which may be related to the oxygen shortage under stress conditions and sharp metabolism activation at re-aeration in the post-stress period.  相似文献

The effect of light intensity on antioxidants, antioxidant enzymes, and chlorophyll content was studied in common bean (Phaseolus vulgaris L.) exposed to excess Mn. Leaves of bean genotypes contrasting in Mn tolerance were exposed to two different light intensities and to excess Mn; light was controlled by shading a leaflet with filter paper. After 5 d of Mn treatment ascorbate was depleted by 45% in leaves of the Mn-sensitive genotype ZPV-292 and by 20% in the Mn-tolerant genotype CALIMA. Nonprotein sulfhydryl groups and glutathione reductase were not affected by Mn or light treatment. Ten days of Mn-toxicity stress increased leaf ascorbate peroxidase activity of cv ZPV-292 by 78% in low light and by 235% in high light, and superoxide dismutase activity followed a similar trend. Increases of ascorbate peroxidase and superoxide dismutase activity observed in cv CALIMA were lower than those observed in the susceptible cv ZPV-292. The cv CALIMA had less ascorbate oxidation under excess Mn-toxicity stress. Depletion of ascorbate occurred before the onset of chlorosis in Mn-stressed plants, especially in cv ZPV-292. Lipid peroxidation was not detected in floating leaf discs of mature leaves exposed to excess Mn. Our results suggest that Mn toxicity may be mediated by oxidative stress, and that the tolerant genotype may maintain higher ascorbate levels under stress than the sensitive genotype.  相似文献

Exposure to ultraviolet-B (UV-B) radiation can lead to oxidative damage in plants. However, plants possess a number of UV-protection mechanisms including screening of potentially damaging UV-B and increased production or activities of antioxidants. The balance or trade-off between these two mechanisms has rarely been studied and is poorly understood. Two isolines of soybean (Glycine max [L.] Merr.) Clark cultivar, the normal line with moderate levels of flavonoids and the magenta line with reduced flavonoids levels, were grown in the field with or without natural levels of UV-B. Leaflet blades of the first trifoliate leaf were harvested after 4–12 days of exposure to the experimental conditions for analysis of active oxygen species (AOS) and antioxidant levels. Solar UV-B radiation caused oxidative stress in both lines and altered AOS metabolism primarily by decreasing superoxide dismutase activity and increasing the activities of ascorbate peroxidase, catalase and glutathione reductase. This resulted in decreased ascorbic acid content and increased dehydroascorbate content. The magenta line had greater oxidative stress than the normal line in spite of its enhanced oxidative defense capacity as compared to the normal line, even under UV-B exclusion. These results indicate enhanced sensitivity in the magenta line, especially under UV-B exclusion that was likely due to the absence of flavonoid epidermal screening compounds and subsequent increased penetration of solar ultraviolet radiation into the leaf.  相似文献

The physiological effects of lanthanum(III) ions on the ferritin-regulated antioxidant process were studied in wheat (Triticum aestivum L.) seedlings under polyethylene glycol (PEG) stress. Treatment with 0.1 mM La3+ resulted in increased levels of chlorophyll, carotenoid, proline, ascorbate, and reduced glutathione. The activities of superoxide dismutase, catalase, ascorbate peroxidase, monodehydroascorbate reductase, dehydroascorbate reductase, glutathione reductase, and peroxidase were also increased after La3+ treatment. Treatment with La3+ seems to enhance the capacity of the reactive oxygen species scavenging system, affect the Fe2+ and Fe3+ electron-transfer process in ferritin, and restrain the formation of hydroxyl radical (OH.), alleviating the oxidative damage induced by PEG stress.  相似文献

The in vitro response of sweet cherry (Prunus cerasus × P. canescens) rootstock Gisela 5 to increasing water deficit in the culture medium was studied. Water stress induced by the incorporation of 1, 2 and 4 % polyethylene glycol (PEG-8000) into the Murashige and Skoog medium was applied for 6 weeks. PEG-induced water stress reduced shoot dry mass, length, water content and relative chlorophyll content. Water stress also induced leaf necrosis without causing loss of viability in the explants. The increase in malondialdehyde content indicated oxidative stress. The activities of antioxidant enzymes, such as superoxide dismutase (SOD), catalase (CAT), ascorbate peroxidase (APX), peroxidase (POX) and glutathione reductase (GR) were also significantly elevated. The concentrations of K, Ca, Fe and Mn of shoots were decreased.  相似文献

The rising temperatures (>35°C) are proving detrimental to summer-sown mungbean genotypes that experience inhibition of vegetative and reproductive growth. In the present study, the mungbean plants growing hydroponically at varying temperatures of 30/20°C (control), 35/25, 40/30, and 45/35°C (as day/night 12 h/12 h) with (50 μM) or without ascorbic acid (ASC) were investigated for effects on growth, membrane damage, chlorophyll loss, leaf water status, components of oxidative stress, and antioxidants. The ASC-treated plants showed significant improvement in germination and seedling growth especially at 40/30 and 45/35°C. The damage to membranes, loss of water, decrease in cellular respiration, and chlorophyll were significantly prevented by ASC treatment to plants growing at these temperatures. The oxidative stress measured as malondialdehyde and hydrogen peroxide content was observed to be significantly lower at high temperatures with ASC application. The activities of superoxide dismutase, catalase, ascorbate peroxidase, and glutathione reductase increased at 40/30°C but decreased at 45/35°C in the absence of ASC while with its application, the activities of these enzymes were appreciably resorted. Among all the antioxidants, the endogenous ASC content decreased to the greatest extent at 45/35°C grown plants indicating its vital role in affecting the response of mungbean to heat stress. Exogenously applied ASC raised its endogenous content along with that of glutathione and proline at 45/35°C. The findings indicated that heat stress-induced inhibition in growth and chlorosis was associated with decrease in leaf water status and elevation of oxidative stress, which could partly be prevented by exogenous application of ASC. Its role in imparting protection against heat stress is discussed.  相似文献

Growth promoting potential of Bacillus subtilis (BS) in drought stressed Abelmoschus esculentus (L.) Moench (okra) was assessed by measuring the chlorophyll stability index (CSI), chlorophyll a (Chl-a) fluorescence, leaf osmotic potential and lipid peroxidation by malondialdehyde content, emission of reactive oxygen species (ROS), osmolyte content and the activity of non-enzyme and enzyme antioxidants. BS treatment significantly increased the leaf osmotic potential, osmolyte production and the activity of non-enzyme and enzyme antioxidants under drought stress. BS treatment mitigated the drought-induced reduction in Chl a fluorescence and CSI. Concomitant increase in total sugar, proline, non-enzyme antioxidants [glutathione and ascorbate] and enzyme antioxidants like superoxide dismutase, catalase, ascorbate peroxidase, glutathione reductase, monodehydroascorbate reductase and dehydroascorbate reductase modulate the intracellular ROS concentration in okra to resist the stress induced oxidative damage in BS treated plants led to fast recovery and less photodamage.Supplementary InformationThe online version contains supplementary material available at 10.1007/s12298-021-00982-8.  相似文献

Manganese (Mn) is an essential element for plant growth but in excess, specially in acidic soils, it can become phytotoxic. In order to investigate whether oxidative stress is associated with the expression of Mn toxicity during early seedling establishment of rice plants, we examined the changes in the level of reactive oxygen species (ROS), oxidative stress induced an alteration in the level of non-enzymic antioxidants and activities of antioxidative enzymes in rice seedlings grown in sand cultures containing 3 and 6 mM MnCl2. Mn treatment inhibited growth of rice seedlings, the metal increasingly accumulated in roots and shoots and caused damage to membranes. Mn treated plants showed increased generation of superoxide anion (O2 .−), elevated levels of H2O2 and thiobarbituric acid reactive substances (TBARS) and decline in protein thiol. The level of nonprotein thiol, however, increased due to Mn treatment. A decline in contents of reduced ascorbate (AsA) and glutathione (GSH) as well as decline in ratios of their reduced to oxidize forms was observed in Mn-treated seedlings. The activities of antioxidative enzymes superoxide dismutase (SOD) and its isoforms Mn SOD, Cu/Zn SOD, Fe SOD as well as guaiacol peroxidase (GPX) increased in the seedlings due to Mn treatment however, catalase (CAT) activity increased in 10 days old seedlings but it declined by 20 days under Mn treatment. The enzymes of Halliwell-Asada cycle, ascorbate peroxidase (APX) monodehydoascorbate reductase (MDHAR), dehyroascorbate reductase (DHAR) and glutathione reductase (GR) increased significantly in Mn treated seedlings over controls. Results suggest that in rice seedlings excess Mn induces oxidative stress, imbalances the levels of antioxidants and the antioxidative enzymes SOD, GPX, APX and GR appear to play an important role in scavenging ROS and withstanding oxidative stress induced by Mn.  相似文献

We investigated the relationship between H2O2 metabolism and the senescence process using soluble fractions, mitochondria, and peroxisomes from senescent pea (Pisum sativum L.) leaves. After 11 d of senescence the activities of Mn-superoxide dismutase, dehydroascorbate reductase (DHAR), and glutathione reductase (GR) present in the matrix, and ascorbate peroxidase (APX) and monodehydroascorbate reductase (MDHAR) activities localized in the mitochondrial membrane, were all substantially decreased in mitochondria. The mitochondrial ascorbate and dehydroascorbate pools were reduced, whereas the oxidized glutathione levels were maintained. In senescent leaves the H2O2 content in isolated mitochondria and the NADH- and succinate-dependent production of superoxide (O2·−) radicals by submitochondrial particles increased significantly. However, in peroxisomes from senescent leaves both membrane-bound APX and MDHAR activities were reduced. In the matrix the DHAR activity was enhanced and the GR activity remained unchanged. As a result of senescence, the reduced and the oxidized glutathione pools were considerably increased in peroxisomes. A large increase in the glutathione pool and DHAR activity were also found in soluble fractions of senescent pea leaves, together with a decrease in GR, APX, and MDHAR activities. The differential response to senescence of the mitochondrial and peroxisomal ascorbate-glutathione cycle suggests that mitochondria could be affected by oxidative damage earlier than peroxisomes, which may participate in the cellular oxidative mechanism of leaf senescence longer than mitochondria.  相似文献

The present study examines the salinity-induced oxidative damage and differential response of enzymatic and non-enzymatic antioxidants of Nostoc muscorum. As compared to carotenoid content which showed induction the chlorophyll and phycocyanin contents were inhibited after salt stress. Acceleration of lipid peroxidation and peroxide production suggested onset of oxidative damage. The activities of all studied enzymatic antioxidants were significantly increased by salt stress with maximum induction of superoxide dismutase (154.8% at 200 mM NaCl treatment). Interestingly under severe stress condition (250 mM NaCl) ascorbate peroxidase seems to be more crucial than catalase for peroxide scavenging. Among the studied non-enzymatic antioxidants alpha-tocopherol was induced maximally (56.0%), however, ascorbate and reduced glutathione were increased by only 8.9% after 250 mM NaCl treatment as compared to control cells. Therefore, salinity was found to induce antioxidative defense system of N. muscorum.  相似文献

Salinity is an important abiotic factor that adversely affects major agricultural soils of the world and hence limits crop productivity. An optimum mineral-nutrient status of plants plays critical role in determining plant tolerance to various stresses. A pot experiment was conducted on mustard (Brassica campestris L.) to study the protective role of added potassium (K, 40 mg kg−1 soil) against salinity-stress (0, 40 and 80 mM NaCl)-induced changes in plant growth, photosynthetic traits, ion accumulation, oxidative stress, enzymatic antioxidants and non-enzymatic antioxidants at 30 days after sowing. Increasing NaCl levels decreased the growth, photosynthetic traits and the leaf ascorbate and glutathione content but increased the leaf ion accumulation and oxidative stress, and the activity of antioxidant enzymes. In contrast, K-nutrition improved plant growth, photosynthetic traits, activity of antioxidant enzymes and the ascorbate and glutathione content, and reduced ion accumulation and oxidative stress traits in the leaves, more appreciably at 40 mM than at 80 mM NaCl. The study illustrates the physiological and biochemical basis of K-nutrition-induced NaCl tolerance in mustard as a means to achieving increased crop productivity in a sustainable way.  相似文献
