Agroinfiltration and PVX agroinfection are two efficient transient expression assays for functional analysis of candidate genes in plants. The most commonly used agent for agroinfiltration is Agrobacterium tumefaciens, a pathogen of many dicot plant species. This implies that agroinfiltration can be applied to many plant species. Here, we present our protocols and expected results when applying these methods to the potato (Solanum tuberosum), its related wild tuber-bearing Solanum species (Solanum section Petota) and the model plant Nicotiana benthamiana. In addition to functional analysis of single genes, such as resistance (R) or avirulence (Avr) genes, the agroinfiltration assay is very suitable for recapitulating the R-AVR interactions associated with specific host pathogen interactions by simply delivering R and Avr transgenes into the same cell. However, some plant genotypes can raise nonspecific defense responses to Agrobacterium, as we observed for example for several potato genotypes. Compared to agroinfiltration, detection of AVR activity with PVX agroinfection is more sensitive, more high-throughput in functional screens and less sensitive to nonspecific defense responses to Agrobacterium. However, nonspecific defense to PVX can occur and there is a risk to miss responses due to virus-induced extreme resistance. Despite such limitations, in our experience, agroinfiltration and PVX agroinfection are both suitable and complementary assays that can be used simultaneously to confirm each other''s results.  相似文献

The genus Mystrium is revised for the Malagasy region. Six species, Mystrium barrybressleri sp. n., Mystrium labyrinth sp. n., Mystrium eques sp. n., Mystrium mirror sp. n., Mystrium shadow sp. n., and Mystrium janovitzi sp. n. are described as new. Two existing names, Mystrium fallax Forel and Mystrium stadelmanni Forel, are synonymized with Mystrium voeltzkowi Forel and Mystrium mysticum Roger, respectively. All recognized species, including species outside of the Malagasy region, are assigned to one of the three newly proposed species groups. The associations between existing names and males are reexamined, and males of eight of the ten Malagasy species are described or redescribed. The taxonomic history of Mystrium highlights the importance of using unique identifiers when designating type specimens and the use of deposited vouchers in phylogenetic and ecological studies. Keys to species for workers, queens, and males are provided. Furthermore, a neotype for Mystrium mysticum is designated, as well as lectotypes for Mystrium camillae Emery, Mystrium rogeri Forel, Mystrium fallax Forel, Mystrium oberthueri Forel, Mystrium stadelmanni Forel, and Mystrium voeltzkowi Forel. Stigmatomma gingivale (Brown) is reassigned to Amblyopone as comb. rev. and Amblyopone awa Xu & Chu, Amblyopone kangba Xu & Chu, Amblyopone meiliana Xu & Chu, and Amblyopone zomae Xu & Chu are transferred to the genus Stigmatomma as comb. n.  相似文献

Six species of Cochylis Treitschke, 1829 are recorded for China. Among them, Cochylis triangula sp. n. is described as new; Cochylis atricapitana (Stephens, 1852), Cochylis discerta Razowski, 1970, Cochylis dubitana (Hübner, [1799]), Cochylis faustana (Kennel, 1919) and Cochylis posterana hyrcana (Toll, 1948) are recorded for the first time for China. The female of Cochylis discerta Razowski, 1970 is described for the first time. Adults and genitaliaare illustrated, a key is given for the identification of Chinese species based on male genitalia.  相似文献

The Andean bellflowers comprise an explosive radiation correlated with shifts to specialized pollination. One diverse clade has evolved with extremely curved floral tubes and is predicted to be pollinated exclusively by one of two parapatric species of sicklebill hummingbirds (Eutoxeres). In this study, we focused on the floral biology of Centropogon granulosus, a bellflower thought to be specialized for pollination by Eutoxeres condamini, in a montane cloud forest site in southeastern Peru. Using camera traps and a pollination exclusion experiment, we documented E. condamini as the sole pollinator of C. granulosus. Visitation by E. condamini was necessary for fruit development. Flowering rates were unequivocally linear and conformed to the “steady‐state” phenological type. Over the course of >1800 h of monitoring, we recorded 12 E. condamini visits totaling 42 s, indicating traplining behavior. As predicted by its curved flowers, C. granulosus is exclusively pollinated by buff‐tailed sicklebill within our study area. We present evidence for the congruence of phenology and visitation as a driver of specialization in this highly diverse clade of Andean bellflowers.  相似文献

A detailed examination of specimens of Cryptopleurum sichuanicum Ryndevich, 2005 from high altitudes of Sichuan Province, China, revealed that the species belongs in the genus Nipponocercyon Satô, 1963 previously endemic to Japan. The species is here transferred in Nipponocercyon, and Nipponocercyon sichuanicus (Ryndevich, 2005), comb. n. is redescribed and compared with Nipponocercyon shibatai Satô, 1963. The male genitalia of Nipponocercyon sichuanicus is described for the first time. An adapted diagnosis of Nipponocercyon is provided, and reasons for the inclusion of Nipponocercyon sichuanicus into Nipponocercyon and the general distribution of the genus are discussed.  相似文献

Targeted genetic manipulation using homologous recombination is the method of choice for functional genomic analysis to obtain a detailed view of gene function and phenotype(s). The development of mutant strains with targeted gene deletions, targeted mutations, complemented gene function, and/or tagged genes provides powerful strategies to address gene function, particularly if these genetic manipulations can be efficiently targeted to the gene locus of interest using integration mediated by double cross over homologous recombination.Due to very high rates of nonhomologous recombination, functional genomic analysis of Toxoplasma gondii has been previously limited by the absence of efficient methods for targeting gene deletions and gene replacements to specific genetic loci. Recently, we abolished the major pathway of nonhomologous recombination in type I and type II strains of T. gondii by deleting the gene encoding the KU80 protein1,2. The Δku80 strains behave normally during tachyzoite (acute) and bradyzoite (chronic) stages in vitro and in vivo and exhibit essentially a 100% frequency of homologous recombination. The Δku80 strains make functional genomic studies feasible on the single gene as well as on the genome scale1-4.Here, we report methods for using type I and type II Δku80Δhxgprt strains to advance gene targeting approaches in T. gondii. We outline efficient methods for generating gene deletions, gene replacements, and tagged genes by targeted insertion or deletion of the hypoxanthine-xanthine-guanine phosphoribosyltransferase (HXGPRT) selectable marker. The described gene targeting protocol can be used in a variety of ways in Δku80 strains to advance functional analysis of the parasite genome and to develop single strains that carry multiple targeted genetic manipulations. The application of this genetic method and subsequent phenotypic assays will reveal fundamental and unique aspects of the biology of T. gondii and related significant human pathogens that cause malaria (Plasmodium sp.) and cryptosporidiosis (Cryptosporidium).  相似文献

A review of the subgenera of the South American genus Praocis Eschscholtz (Pimeliinae: Praociini) is presented. Praocis comprises 77 species and 8 subspecies arranged in nine subgenera distributed in arid lands from Central Peru and Bolivia to the Southern part of Patagonia in Chile and Argentina. For each subgenus of Praocis: Praocis Eschscholtz, Mesopraocis Flores & Pizarro-Araya, subgen. n., Anthrasomus Guérin-Méneville, Filotarsus Gay & Solier, Postpraocis Flores & Pizarro-Araya, subgen. n., Hemipraocis Flores & Pizarro-Araya, subgen. n., Orthogonoderes Gay & Solier, Praonoda Flores & Pizarro-Araya, subgen. n., and Praocida Flores & Pizarro-Araya, subgen. n., we present a diagnosis using new and constant characters of adult morphology such as clypeal configuration, length and proportion of antennomeres 9, 10 and 11, arrangement of apical tomentose sensory patches on antennomeres 10 and 11, anterior margin of prosternum, lateral margin of elytron, ventral surface of profemora, and shape of protibiae. An identification key for the nine subgenera of Praocis is presented. Type species are designated for the five new subgenera; for Mesopraocis: Praocis calderana Kulzer, for Postpraocis: Praocis pentachorda Burmeister, for Hemipraocis: Praocis sellata Berg, for Praonoda: Praocis bicarinata Burmeister, for Praocida: Praocis zischkai Kulzer, and for the previously described subgenus Orthogonoderes: Praocis subreticulata Gay & Solier. The current number of species and the estimated number of species to be described are presented. The distribution ranges of the subgenera, including new records from collections and recent expeditions, are given. Habitat preferences and a discussion of the biogeography of the genus are also presented.  相似文献

Twenty-one isolates of 18 fungal species were tested on water agar for their pathogenicity to eggs of Heterodera glycines. An egg-parasitic index (EPI) for each of these fungi was recorded on a scale from 0 to 10, and hatch of nematode eggs was determined after exposure to the fungi on water agar for 3 weeks at 24 C. The EPI for Verticillium chlamydosporium was 7.6, and the fungus reduced hatch 74%. Pyrenochaeta terrestris and two sterile fungi also showed a high EPI and reduced hatch 42-73%. Arthrobotrys dactyloides, Fusarium oxysporum, Paecilomyces lilacinus, Stagonospora heteroderae, Neocosmospora vasinfecta, Fusarium solani, and Exophiala pisciphila were moderately pathogenic to eggs (EPI was 2.0-4.5, and hatch was reduced 21-56%). Beauveria bassiana, Hirsutella rhossiliensis, Hirsutella thompsonii, Dictyochaeta heteroderae, Dictyochaeta coffeae, Gliocladium catenulatum, and Cladosporium sp. showed little parasitism of nematode eggs but reduced hatch. A negative correlation was observed between hatch and fungal parasitism of eggs. Fusarium oxysporum, H. rhossiliensis, P. lilacinus, S. heteroderae, V. chlamydosporium, and sterile fungus 1 also were tested in soil in a greenhouse test. After 3 months, the nematode densities were lower in soil treated with H. rhossiliensis and V. chlamydosporium than in untreated soil. The nematode population densities were correlated negatively with the EPI, but not with the percentage of cysts colonized by the fungi. Plant weights and heights generally increased in the soil treated with the fungi.  相似文献

A PCR-based diagnostic assay was developed for early detection and identification of Aphelenchoides fragariae directly in host plant tissues using the species-specific primers AFragFl and AFragRl that amplify a 169-bp fragment in the internal transcribed spacer (ITS1) region of ribosomal DNA. These species-specific primers did not amplify DNA from Aphelenchoides besseyi or Aphelenchoides ritzemabosi. The PCR assay was sensitive, detecting a single nematode in a background of plant tissue extract. The assay accurately detected A. fragariae in more than 100 naturally infected, ornamental plant samples collected in North Carolina nurseries, garden centers and landscapes, including 50 plant species not previously reported as hosts of Aphelenchoides spp. The detection sensitivity of the PCR-based assay was higher for infected yet asymptomatic plants when compared to the traditional, water extraction method for Aphelenchoides spp. detection. The utility of using NaOH extraction for rapid preparation of total DNA from plant samples infected with A. fragariae was demonstrated.  相似文献

Namalycastis jaya sp. n. (Polychaeta: Nereididae: Namanereidinae)is described from the southern coast of Kerala in southwest India. One important characteristic feature of the species is the lack of notochaetae in all parapodia, a characteristic that it shares with at least two other species, Namalycastis elobeyensis Glasby, 1999and Namalycastis hawaiiensis Johnson, 1903. It differs from Namalycastis elobeyensis by virtue of its smaller antennae, unequal eye size, bilobed acicular neuropodial ligule and multi-incised pygidium rim. Moreover, it differs from Namalycastis hawaiiensis by having fewer teeth on the serrated blades of the sub-neuroacicular falciger in chaetiger 10, and by possessing finely serrated falcigers in posterior segments. Beyond morphological analyses, molecular phylogenetics was used for the first time for Namalycastis to support population monophyly and recognition of the new species.The analysis, using both mitochondrial and nuclear data, corroborated the morphological analysis in suggesting that our specimens represent an as yet undescribed species, Namalycastis jaya sp. n., which forms a monophyletic group among the sampled nereidid taxa. Finally, a taxonomic key for Namalycastis species recorded from the Indian region is provided.  相似文献

The distribution of Acomopterella species in the Palaearctic region has been re-examined in this study, using recently collected material. The European species was found to be distributed in the eastern Palaearctic as well. A second Palaearctic species from Honshu (Japan) is herein described. The morphology of adult specimens was studied by light microscopy and scanning electron microscopy. The shape of functional specialized setae on mid tibiae in Acomopterella and seven further fungus gnat genera is described and the suitability of this character for systematic studies is discussed. Details of a “hind tibial organ” are described.The position of Acomopterella in the tribe Gnoristini is briefly discussed. Acomopterella is found to be more closely related to Speolepta Edwards, 1925, than to any other recent genus.  相似文献

The Chinese fauna of the family Gasteruptiidae is revised, keyed and fully illustrated for the first time. Only one genus of this family, Gasteruption Latreille, 1796, is recorded from China. In total 28 valid species of the genus Gasteruption are recognized. Six species are new to science (Gasteruption angulatum sp. n., Gasteruption assectoides sp. n., Gasteruption coloratum sp. n., Gasteruption latitibia sp. n., Gasteruption sinepunctatum sp. n. and Gasteruption strigosum sp. n.) and eight species are reported new for China (Gasteruption bimaculatum Pasteels, 1958, Gasteruption birmanense Pasteels, 1958, Gasteruption dimidiatum Semenov, 1892, Gasteruption formilis Alekseev, 1995, Gasteruption subhamatum Pasteels, 1958, Gasteruption tonkinense Pasteels, 1958, Gasteruption tournieri Schletterer, 1885, Gasteruption transversiceps Pasteels, 1958). Three new synonyms are proposed: Gasteruption curiosum Pasteels, 1958, of Gasteruption amoyense Pasteels, 1958; Gasteruption sinense var. minus Kieffer, 1924, with Gasteruption japonicum Cameron, 1888,and Gasteruption sinense Kieffer, 1924, of Gasteruption sinarum Kieffer, 1911. Lectotypes are designated for Gasteruption corniculigerum Enderlein, 1913, Gasteruption sinense Kieffer, 1924, and Gasteruption transversiceps Pasteels, 1958. Gasteruption bihamatum Kieffer, 1911, previously reported from South China, is a South American species.  相似文献

We describe three new species of springsnails (genus Pyrgulopsis) from the Amargosa River basin, California and Nevada (P. licina sp. n., P. perforata sp. n., P. sanchezi sp. n.), each of which was previously considered to be part of P. micrococcus. We also restrict P. micrococcus to its type locality area (Oasis Valley) and redefine a regional congener, P. turbatrix, to include populations from the central Death Valley region and San Bernardino Mountains that had been previously identified as P. micrococcus. The five species treated herein form genetically distinct lineages that differ from each other by 4.2–12.6% for mtCOI and 5.2–13.6% for mtNDI (based on previously published and newly obtained data), and are diagnosable by shell and/or penial characters. The new molecular data presented herein confirm sympatry of P. licina and P. sanchezi in Ash Meadows (consistent with morphological evidence) and delineate an additional lineage of P. micrococcus (in the broad sense) that we do not treat taxonomically owing to the paucity of morphological material. Conservation measures are needed to ensure the long term persistence of populations of P. micrococcus and a genetically differentiated lineage of P. sanchezi which live in disturbed habitats on private lands.  相似文献

The species of Megachile subgenus Litomegachile are revised with a review of the species morphology, biology, and plant associations. A new species, Megachile pankus, is described and illustrated. Megachile mendica snowi Mitchell is elevated to species. Megachile var. nupta Cresson and Megachile texana var. cleomis Cockerell are synonymized with Megachile brevis and Megachile texana, respectively. One new genus and eight new species of anilline carabids are described from southern Mexico. The new genus, Zapotecanillus gen. n., is established for Z. oaxacanus (type species) sp. n., Z. nanus sp. n., Z. iviei sp. n., Z. ixtlanus sp. n., Z. montanus sp. n., and Z. kavanaughi sp. n. from the Sierra Madre de Oaxaca, Z. pecki sp. n. from the Sierra Madre del Sur, and Z. longinoi sp. n. from the Sierra Madre de Chiapas. A taxonomic key for all described species of Zapotecanillus and a cladistic analysis, based on morphological data, are provided. Morphological, behavioral and biogeographical aspects of the speciation in the genus obtained from the resulting cladogram are discussed.  相似文献

The praying mantis genus Liturgusa Saussure, 1869 occurs only in Central and South America and represents the most diverse genus of Neotropical Liturgusini (Ehrmann 2002). The genus includes bark dwelling species, which live entirely on the trunks and branches of trees and run extremely fast. All species included within the genus Liturgusa are comprehensively revised with a distribution stretching from central Mexico, the island of Dominica to the southeastern regions of Brazil and southern Bolivia. All known species are redescribed to meet the standards of the new treatment of the genus (11 species). Three new genera are described including Fuga gen. n., Velox gen. n., and Corticomantis gen. n. for species previously included in Liturgusa as well as Hagiomantis. Liturgusa mesopoda Westwood, 1889 is moved to within the previously described genus Hagiomantis Audinet Serville, 1838. A total of 19 species are newly described within Liturgusa, Fuga, and Velox including L. algorei sp. n., L. bororum sp. n., L. cameroni sp. n., L. cura sp. n., L. dominica sp. n., L. fossetti sp. n., L. kirtlandi sp. n., L. krattorum sp. n., L. manausensis sp. n., L. maroni sp. n., L. milleri sp. n., L. neblina sp. n., L. purus sp. n., L. stiewei sp. n., L. tessae sp. n., L. trinidadensis sp. n., L. zoae sp. n., F. grimaldii sp. n., and V. wielandi sp. n. Four species names are synonymized: Liturgusa peruviana Giglio-Tos, 1914, syn. n. = Liturgusa nubeculosa Gerstaecker, 1889 and Hagiomantis parva Piza, 1966, syn. n., Liturgusa sinvalnetoi Piza, 1982, syn. n., and Liturgusa parva Giglio-Tos, 1914, syn. n. = Mantis annulipes Audinet Serville, 1838. Lectotypes are designated for the following two species: Liturgusa maya Saussure & Zehntner, 1894 and Fuga annulipes (Audinet Serville, 1838). A male neotype is designated for Liturgusa guyanensis La Greca, 1939. Males for eight species are described for the first time including Liturgusa cayennensis Saussure, 1869, Liturgusa lichenalis Gerstaecker, 1889, Liturgusa guyanensis La Greca, 1939, Liturgusa maya Saussure & Zehntner, 1894, Liturgusa nubeculosa Gerstaecker, 1889, Fuga annulipes (Audinet Serville, 1838), Corticomantis atricoxata (Beier, 1931), and Hagiomantis mesopoda (Westwood, 1889). The female of Fuga fluminensis (Piza, 1965) is described for the first time. Complete bibliographic histories are provided for previously described species. The spelling confusion surrounding Liturgusa/Liturgousa is resolved. Full habitus images for males and females are provided for nearly all species. Habitus and label images of type specimens are provided when possible. Diagnostic illustrations of the head and pronotum for males and females are provided for all species when possible. Illustrations of male genital structures are provided for all species for which males are known. Measurement data, including ranges and averages, are provided for males and females of all species. Combined male and female genus and species level dichotomous keys are provided with a Spanish translation. A complete table of all examined specimens lists label data, museum codes, repositories, and other specimen specific information. A KML file with all georeferenced locality records is downloadable from mantodearesearch.com for viewing in Google Earth. The East Mediterranean species of Orthomus Chaudoir, 1838 are revised. The type series of Feronia longula Reiche & Saulcy, 1855, F. berytensis Reiche & Saulcy, 1855, F. proelonga Reiche & Saulcy, 1855, Orthomus longior Chaudoir, 1873, O. sidonicus Chaudoir, 1873, and O. berytensis akbensis Mateu, 1955 were studied and lectotypes for the first four are designated. Also, the following nomenclatural acts are proposed: Feronia proelonga Reiche & Saulcy, 1855, syn. n. of Orthomus berytensis (Reiche & Saulcy, 1855); Feronia elongata Chaudoir, 1859, syn. n. of Orthomus berytensis (Reiche & Saulcy, 1855); Orthomus sidonicus Chaudoir, 1873, syn. n. of Orthomus longior Chaudoir, 1873; Orthomus velocissimus andalusiacus Mateu, 1957, syn. n. of Orthomus velocissimus akbensis Mateu, 1955, new assignment for Orthomus berytensis akbensis Mateu, 1955. As a result, three species of the genus inhabit the East Mediterranean biogeographical region: O. berytensis, O. longior, and O. longulus. A key to these three species is given. O. longior is recorded for Turkey and Syria for the first time. In addition, a new synonymy of two West Mediterranean taxa is proposed: O. szekessyi (Jedlička, 1956), syn. n. of O. balearicus (Piochard de la Brûlerie, 1868), and a new genus and a species are described: Parorthomus gen. n. socotranus sp. n. (type locality: Republic of Yemen, Socotra Archipelago, Socotra Island, Fimihin env., 530 m.a.s.l.). Illustrations of the species dealt with here are provided including external characters, habitus, mentum and submentum, and genitalia are provided.Nine genera of the “African Series” of subtribe Euchroina Chaudoir, 1874 are keyed for the first time. Checklists of the species of Orthomus and of the Old World euchroine genera are given.  相似文献

Enzymes from females of five Pratylenchus brachyurus populations and one P. scribneri population were analyzed by isoelectric focusing electrophoresis. Of the 18 enzyme systems investigated, only malate dehydrogenase (MDH), phosphoglucomutase (PGM), and phosphoglucose isomerase (PGI) were detected from all five P. brachyurus populations and P. scribneri. Faint bands were detected for isocitrate dehydrogenase and phosphogluconate dehydrogenase from one P. brachyurus population. Three distinct phenotypic groups were found in the MDH and PGM systems for P. brachyurus populations, but only a single electromorph was detected for PGI. Multiple electromorphs for MDH, PGM, and PGI were detected for P. scribneri; there was no similarity among these patterns with those from P. brachyurus. No phenotypic differences in PGI were observed between females and mixed juveniles of one population of P. brachyurus.  相似文献

The parasitoid complex associated with Monochamus galloprovincialis (Olivier), vector of the pine wood nematode, is discussed. Four species of the family Braconidae and one Ichneumonidae were found associated with Monochamus galloprovincialis in Portugal, namely Atanycolus denigrator (Linnaeus), Atanycolus ivanowi (Kokujev), Cyanopterus flavator (Fabricius), Doryctes striatellus (Nees) (Braconidae), and Xorides depressus (Holmgren) (Ichneumonidae). Atanycolus ivanowi, Atanycolus denigrator, Doryctes striatellus and Xorides depressus are new species for Portugal fauna, and Monochamus galloprovincialis is recorded as a new host of Xorides depressus. A key for determination of the ichneumonoid parasitoids of the pine sawyer is provided for the Palaearctic fauna.  相似文献
