Incubation of normal mouse spleen cells with homologous IgE resulted in the formation of soluble factors that inhibited rosette formation of mouse Fc epsilon R+ cells with IgE-coated ox erythrocytes. The soluble factors could be absorbed with mouse or rat IgE coupled to Sepharose and recovered from the beads by acid elution. However, the factors had no affinity for either human IgE or mouse IgG. The IgE-binding factors were derived from T cells. Production of the factors required Lyt1+ T cells and Fc gamma R+ cells, which suggests that the factors are derived from Fc gamma R+ Lyt 1+ T cells. The molecular size of IgE-binding factors was approximately 15,000 daltons. When IgE-binding factors were formed by BALB/c spleen cells, nearly one-half of the factors had affinity for lentil lectin, and the remaining half of the factors failed to bind to the lectin. The proportion of the two species of IgE-binding factors differed depending on mouse strains. The majority of the factors formed by B6D2F1 spleen cells had affinity for lentil lectin, but those formed by SJL spleen cells failed to bind to the lectin. The IgE-binding factors were also induced by incubation of normal spleen cells with polyinosinic-polycytidylic acid (pI:pC). The nucleotide stimulated splenic adherent cells to form "inducers" of IgE-binding factors, which in turn induced normal lymphocytes to form IgE-binding factors. The inducers of IgE-binding factors were inactivated (or neutralized) by antibodies specific for mouse Type I interferon. It was also found that purified mouse beta interferon could induce the formation of IgE-binding factors. IgE-binding factors induced by pI:pC consisted of two different molecules: one had a m.w. of 15,000 daltons, and another had a m.w. of between 40,000 and 60,000 daltons.  相似文献

Goal, Scope and Background Although LCA is traditionally a site-independent tool, there is currently a trend towards making LCA more site-dependent if not site-specific. For Europe, site-dependent impact factors have been calculated on a country basis for acidification, terrestrial eutrophication and toxicological impacts. It is, however, an open question whether this is the optimum level for site-dependent factors. The aim of this paper is to develop site-dependent characterisation factors for different parts of Sweden for air emissions of NOx, SOx and particulates regarding ecosystem and human health impacts. Based on experiences from a case-study, the usability of the site-dependent factors for LCA are discussed, as well as the appropriate level of site-dependency for ecosystem and human health impacts. Method logy. The Ecosense model is used for calculating site-dependent factors for some atmospheric pollutants. Characterisation factors are calculated for four different places in Sweden with two different stack heights. Results and Conclusions The characterisation factors for ecosystem impacts show fairly small differences between different parts of Sweden (within a factor of two). For health impacts, the differences between different parts of the country were larger and more significant (up to one order of magnitude). Also the difference between low and high stack heights may be relevant, especially in densely populated areas. These results suggest that for ecosystems, site-dependent characterisation factors for the considered atmospheric pollutants on a country level may be sufficient for most applications. However, for health impacts, site-dependent factors on a country level may be inappropriate. Beside LCA, the calculated factors and the methodology used should also be useful for other environmental system analysis tools, such as Strategic Environmental Assessment, Cost-Benefit Analysis and Environmental Management Systems.  相似文献

The principles of enzyme kinetic analysis were applied to quantitate the relationships among serum-derived growth factors, nutrients, and the rate of survival and multiplication of human fibroblasts in culture. The survival or multiplication rate of a population of cells plotted against an increasing concentration of a growth factor or nutrient in the medium exhibited a hyperbolic pattern that is characteristic of a dissociable, saturable interaction between cells and the ligands. Parameters equivalent to the Km and Vmax of enzyme kinetics were assigned to nutrients and growth factors. When all nutrient concentrations were optimized and in steady state, serum factors accelerated the rate of multiplication of a normal cell population. The same set of nutrients that supported a maximal rate of multiplication in the presence of serum factors supported the maintenance of non-proliferating cells in the absence of serum factors. Therefore, under this condition, serum factors are required for cell division and play a purely regulatory iole in multiplication of the cell population. The quantitative requirement for 18 nutrients of 29 that were examined was significantly higher (P < 0.001) for cell multiplication in the presence of serum factors than for cell maintenance in the absence of serum factors. This indicated specific nutrients that may be quantitatively important in cell division processes as well as in cell maintenance. The quantitative requirement for Ca2+, Mg2+, K+, Pi, and 2-oxocarboxylic acid for cell multiplication was modified by serum factors and other purified growth factors. The requirement for over 30 other nutrients could not clearly be related to the level of serum factors in the medium. Serum factors also determined the Ca2+, K+, and 2-oxocarboxylic acid requirement for maintenance of non-proliferating cells. Therefore, when either Ca2+, K+, or 2-oxocarboxylic acid concentration was limiting, factors in serum played a role as cell “survival or maintenance” factors in addition to their role in cell division as “growth regulatory” factors. However, with equivalent levels of serum factors in the medium, the requirement for Ca2+, K+, and 2-oxocarboxylic acids was still much higher for multiplication than for maintenance. Kinetic analysis revealed that the concentrations of individual nutrients modify the quantitative requirement for others for cell multiplication in a specific pattern. Thus, specific quantitative relationships among different nutrients in the medium are important in the control of the multiplication rate of the cell population. When all nutrient concentrations were optimal for multiplication of normal cells, the multiplication response of SV40-virus-transformed cells to serum factors was similar to that of normal cells. When serum factors were held constant, transformed cells required significantly less (P < 0.001) of 12 of the 26 nutrients examined. Therefore, the transformed cells only have a growth advantage when the external concentration of specific nutrients limits the multiplication rate of normal cells. Taken together, the results suggest that the control of cell multiplication is intimately related to external concentrations of nutrients. Specific growth regulatory factors may stimulate cell proliferation by modification of the response of normal cells to nutrients. Transforming agents may confer a selective growth advantage on cells by a constitutive alteration of their response to extracellular nutrients.  相似文献

The lack of spatial differentiation in current life-cycle impact assessment (LCIA) affects the relevance of the assessed impact. This article first describes a framework for constructing factors relating the region of emission to the acidifying impact on its deposition areas. Next, these factors are established for 44 European regions with the help of the RAINS model, an integrated assessment model that combines information on regional emission levels with information on long-range atmospheric transport to estimate patterns of deposition and concentration for comparison with critical loads and thresholds for acidification, eutrophication via air; and tropospheric ozone formation. The application of the acidification factors in LCIA is very straightforward. The only additional data required, the geographical site of the emission, is generally provided by current life-cycle inventory analysis. The acidification factors add resolving power of a factor of 1,000 difference between the highest and lowest ratings, while the combined uncertainties in the RAINS model are canceled out to a large extent in the acidification factors as a result of the large number of ecosystems they cover The framework presented is also suitable for establishing similar factors for eutrophication and tropospheric ozone formation for regions outside Europe as well.  相似文献

The epidemiologic approach to determining the etiology of disease involves identification of potential risk factors and then comparison of disease incidence among people with varying levels of exposure to the potential risk factors. This paper defines risk factors which correspond to different levels of genetic and environmental proximity to index cases of birth defects. Genetic proximity is estimated by the coefficient of relationship (R): 0.5 for siblings and dizygotic twins and 1.0 for monozygotic twins. Environmental proximity is measured by a combination of two variables: one variable for those potentially preventable risk factors common to siblings (S) and another for those common only to twins (T). Discordance in identical twins is attributed to a third type of environmental factors (U) that are unshared by twins and include random (stochastic) factors. The association between these risk factors and birth defects is estimated by using a linear model of the correlation of liability for different relatives. The coefficients derived from the model reflect the relative importance of genetic and different types of environmental risk factors as causes for the defects and can be used to identify birth defects most likely to be caused by measurable and possibly preventable risk factors. These defects could then be assigned high priority for future studies and preventive efforts.  相似文献

This study aims to determine the effect of climatic factors (temperature, relative humidity and rainfall) on mosquito abundance and to map mosquito larva breeding sites using GIS application in Eastern Province, Saudi Arabia. The data pertaining to larval and adult mosquito abundance/distribution and climatic factors were collected for the study period of 2014. Bi-variate and multivariate analyses were performed to determine the relationship between mosquito abundance and climatic factors (temperature, relative humidity and rainfall). The utilization of GIS with GPS facilitates to identify and map larva breeding sites in the study area. The result showed strong negative correlation between mosquito abundance and temperature while there appeared a strong positive correlation with relative humidity and moderate positive correlation with rainfall. Low mosquito abundance was observed at high temperatures whereas high and moderate mosquito abundance was observed at high humidity and during rainy months, respectively. In the adult mosquito, the regression model for three climatic factors (temperature, RH and rainfall) and other factors showed a variation of 84.5% of the variance accounted for the climatic factors while the remaining 15.5% were attributed to other factors. In larva, 64.3% of the variance accounted for climatic factors, and the remaining 35.7% attributed to other factors such as the presence of vegetation, waste materials and water reservoirs such as ditches. In comparison, the larva seems influenced by the presence of vegetation, waste material, water reservoirs and ditches and less influenced by climatic factors than the adult mosquito. Development of a risk map by considering the flying distance of the adult mosquito from the studied sites showed three major clusters where a recommendation for management control program was suggested.  相似文献

Many human cancers develop as a result of exposure to risk factors related to the environment and ways of life. The aim of this study was to estimate attributable fractions of 25 types of cancers resulting from exposure to modifiable risk factors in Brazil. The prevalence of exposure to selected risk factors among adults was obtained from population-based surveys conducted from 2000 to 2008. Risk estimates were based on data drawn from meta-analyses or large, high quality studies. Population-attributable fractions (PAF) for a combination of risk factors, as well as the number of preventable deaths and cancer cases, were calculated for 2020. The known preventable risk factors studied will account for 34% of cancer cases among men and 35% among women in 2020, and for 46% and 39% deaths, respectively. The highest attributable fractions were estimated for tobacco smoking, infections, low consumption of fruits and vegetables, excess weight, reproductive factors, and physical inactivity. This is the first study to systematically estimate the fraction of cancer attributable to potentially modifiable risk factors in Brazil. Strategies for primary prevention of tobacco smoking and control of infection and the promotion of a healthy diet and physical activity should be the main priorities in policies for cancer prevention in the country.  相似文献

Goal, Scope and Background Country-dependent characterisation factors for acidification have been derived for use in life cycle assessments to describe the effect on ecosystem protection of a change in national emissions. They have recently also been used in support of European air pollution abatement policies and related cost benefit analyses. We demonstrate that the characterisation factors as calculated to date are unstable due to being derived from the non-smooth and highly varying part of the underlying emission-impact functions. The purpose of this paper is to discuss the currently available characterisation factors and to propose a modification that makes use of the full range of the underlying functions. Method The characterisation factors derived in this paper are based on updates of data used to support European air pollution agreements under the UNECE Convention on Long-range Transboundary Air Pollution (LRTAP) and the European Commission. The focus in this paper is on the analysis of characterisation factors for acidification. The analysis of characterisation factors for terrestrial eutrophication from nitrogen compounds is a simple extension of the methods described here. The analysis is conducted for 25 European nations, i.e. for 23 EU countries plus Norway and Switzerland; Cyprus and Malta are excluded due to lack of data on critical loads. Results and Conclusions We show that a linear model which is calibrated to emission changes of –50% is generally more reliable than characterisation factors which are based on emission changes of plus or minus 10%. Application of these characterisation factors are justified for emission reductions up to 30% in total European emissions, compared to 2000. This is within the range of currently agreed upon emission reductions in 2010 relative to 2000. Therefore, characterisation factors can be used in LCA as well as for the support of the revision of existing European air pollution agreements.  相似文献

We have developed a solid-phase clotting assay which uses peroxidase-fibrinogen in solution and fibrinogen bound to microtiter plates as a substrate for the thrombin generated from the clotting cascade. We have developed this assay for measurement of the extrinsic pathway factors thromboplastin (tissue factor, factor III), VII and VIIa, X, and II. Using long incubation times (40-90 min), thromboplastin could be measured in extracts of human brain at very low concentrations. Specificity for thromboplastin was demonstrated by showing a requirement for factors II, V, X, and VII but not for VIII, IX, XI, or XII; both substrate plasmas monodeficient in single factors and mixtures of the pure factors were used in demonstrating this specificity. The assay was modified to measure factors II, VII, VIIa, and X using appropriate deficient plasmas. The limit of detection was 2-3 orders of magnitude lower than a one-stage clotting test for all factors assayed. This assay has the advantages of convenience, specificity comparable to standard clotting tests, and high sensitivity.  相似文献

Social epidemiology has increasingly looked to psychological factors as both risk factors for physical health and mechanisms behind disparities. Yet, there has been little resolution to the question of whether psychological factors explain disparities, and skepticism has begun to mount about whether psychological factors are causally linked to health. Furthermore, some have questioned the nature of the relationship: most research suggests that psychological factors mediate the relationship between socioeconomic status and health, but recent research suggests that they moderate the relationship. The present paper attempts to provide a more comprehensive appraisal of the current debate. It uses four popular psychological factors (i.e., self-esteem, mastery, neuroticism, and depressive symptoms), three health outcomes, and a nationally representative, three-panel longitudinal survey. The results illustrate the promise and limitations of psychological mechanisms. In the cross-section, the results provide evidence for substantial moderating effects, but these effects disappear entirely when estimated prospectively. The results also provide some evidence for mediating effects, but these effects are very weak and the prospective effects of psychological factors diminish over time and with controls for baseline health. Implications for theories of socioeconomic status and health are discussed and a more social psychologically sophisticated approach is encouraged.  相似文献

This study determines the major weather factors affecting hiking activity and builds a prediction model to estimate participation. An empirical assessment of hiking participation using weather factors was demonstrated for trails on Kuanyin Mountain, Taiwan. By adapting the concepts of the range of tolerance and the eclectic model, a nonlinear function was used to explain hiking participation with weather factors. Stepwise multiple-regression analysis was carried out to determine the major weather factors affecting hiking participation. The results indicate that not only did participation vary with the season but hiking behavior was affected by different weather factors in each season. The explanation rates for the seasons exceeded 90% except that for spring.  相似文献

Background, Aims and Scope Several authors have shown that spatially derived characterisation factors used in life cycle impact assessment (LCIA) can differ widely between different countries in the context of regional impact categories such as acidification or terrestrial eutrophication. Previous methodology studies in Europe have produced country-dependent characterisation factors for acidification and terrestrial eutrophication by using the results of the EMEP and RAINS models and critical loads for Europe. The unprotected ecosystem area (UA) is commonly used as a category indicator in the determination of characterisation factors in those studies. However, the UA indicator is only suitable for large emission changes and it does not result in environmental benefits in terms of characterisation factors if deposition after the emission reduction is still higher than the critical load. For this reason, there is a need to search for a new category indicator type for acidification and terrestrial eutrophying in order to calculate site-dependent characterisation factors. The aim of this study is to explore new site-dependent characterisation factors for European acidifying and eutrophying emissions based on accumulated exceedance (AE) as the category indicator, which integrates both the exceeded area and amount of exceedance. In addition, the results obtained for the AE and UA indicators are compared with each other. Methods The chosen category indicator, accumulated exceedance (AE), was computed according to the calculation methods developed in the work under the United Nations Economic Commission for Europe (UNECE) Convention on Long-range Transboundary Air Pollution (LRTAP). Sulphur and nitrogen depositions to 150x150 km2 grid cells over Europe were calculated by source-receptor matrices derived from the EMEP Lagrangian model of long-range transport of air pollution in Europe. Using the latest critical load data of Europe, the site-dependent characterisation factors for acidification and terrestrial eutrophication were calculated for 35 European countries and 5 sea areas for 2002 emissions and emissions predicted for 2010. In the determination of characterisation factors, the emissions of each country/area were reduced by various amounts in order to find stable characterisation factors. In addition, characterisation errors were calculated for the AE-based characterisation factors. For the comparison, the results based on the use of UA indicator were calculated by 10% and 50% reductions of emissions that corresponded to the common practice used in the previous studies. Results and Discussion The characterisation factors based on the AE indicator were shown to be largely independent of the reduction percentage used to calculate them.. Small changes in emissions (≤100 t) produced the most stable characterisation factors in the case of the AE indicator. The characterisation errors of those characterisation factors were practically zero. This means that the characterisation factors can describe the effects of small changes in national emissions that are mostly looked at in LCAs. The comparison between country-dependent characterisation factors calculated by the AE and UA indicators showed that these two approaches produce differences between characterisation factors for many countries/areas in Europe. The differences were mostly related to the Central and Northern European countries. They were greater for terrestrial eutrophication because the contribution of ammonia emission differ remarkably between the two approaches. The characterisation factors of the AE indicator calculated by the emissions of 2002 were greater than the factors calculated by the predicted emissions for 2010 in almost all countries/sea areas, due to the presumed decrease of acidifying and eutrophying emissions in Europe. Conclusions and Recommendations. In this study, accumulated exceedance was shown to be an appropriate category indicator in LCIA applications for the determination of site-dependent characterisation factors for acidification and terrestrial eutrophication in the context of integrated assessment modelling. In the future, it would be useful to calculate characterisation factors for emissions of separate parts of large countries and sea areas in Europe. In addition, it would also be useful to compare the approach based on the AE indicator with the method of the hazard index, as recommended in the latest CML guidebook.  相似文献

Two protein factors (A and B) have been partially purified from Escherichia coli supernatant which, in combination, are more effective than 0.5 M NH4Cl in stimulating ribosomes for AcPhe-tRNA and fMet-tRNA binding, for the puromycin reaction, and for incorporating acetylphenylalanine from AcPhe-tRNA into polypeptide. The factors appear to differ from the initiation factors, the elongation factor EF-T, and ribosomal proteins. Some uncertainty exists as to whether factor B is different from EF-G. To maximize the effect of the factors in initiator tRNA binding, we preincubated the ribosomes with the factors and carried out the binding assay for a short period at 15 degrees C. Maximal stimulation of binding occurred after about a 2-min preincubation at 37 degrees C. Longer preincubation times were required at 15 degrees C, and only slight stimulation was observed after preincubation at 0 degrees C. The extent of stimulation by the factors was not affected when the NH4Cl concentration was increased from 40 to 500 mM in the preincubation. The presence of both the 30S and 50S ribosomal subunits is required for the enhancement of AcPhe-tRNA binding. Polyphenylalanine synthesis carried out without AcPhe-tRNA is inhibited by the factors. It is suggested that the factors may act by inducing a structural rearrangement of the ribosomes.  相似文献

Determining the number of factors is a critical first step in exploratory factor analysis. Although various criteria and methods for determining the number of factors have been evaluated in the usual between-subjects R-technique factor analysis, there is still question of how these methods perform in within-subjects P-technique factor analysis. A novel feature of P-technique data is that observations are usually sequentially dependent in some way. The current study evaluates 10 criteria for determining the number of factors for data simulated to resemble intensive repeated-measures data for which P-technique is usually applied. The number of factors, loading size, observed-to-latent ratio, serial-dependency, and sample size were simulation design factors with conditions simulated to resemble P-technique data. The methods are compiled from various statistical techniques and have been demonstrated in previous R-technique simulations. Finally, a real data example demonstrates applying the acceleration factor method to several time series data sets.  相似文献

Factors Enhancing the Host-Cell Penetration of Toxoplasma gondii   总被引:7,自引:2,他引:5  
The penetration into HeLa cells of Toxoplasma gondii was studied with a cell culture technique. The influence on the rate of penetration and the number of penetrating Toxoplasma parasites was tested by use of preparations of disintegrated parasites mixed with test parasites. These preparations were found to contain factors enhancing the penetrating rate of the parasites. This effect was demonstrable by use of untreated parasites as well as parasites lacking active motility owing to a previous exposure to Formalin. The preparations of disintegrated parasites contained, in addition, components inhibitory to the penetration-enhancing factors. These inhibitory components were able to reduce the penetrating capacity of normal Toxoplasma parasites, suggesting that the studied enhancing factors may play a role in the natural process of penetration. The efficacy of various techniques for disintegration of Toxoplasma parasites was investigated for release of penetration-enhancing factors from Toxoplasma parasites. The methods used resemble those used for liberation of lysosomal enzymes. Reduced osmotic pressure was obviously not adequate for release of enhancing factors, whereas the freezing and thawing procedure, sonic treatment, and irradiation produced high yields. It was difficult to evaluate the effect of incubation at acid pH on release of enhancing activity, because the penetration-promoting factors seemed unstable on both the acid and the alkaline sides of pH 7.6.  相似文献

The effectiveness of management strategies for invasive species is often hampered by a lack of clear understanding of the factors that limit species distributions. The distribution of exotic species, especially those that are invasive, are often so dynamic that limiting factors are difficult to identify. Comparisons of exotic species between their native ranges, where they are presumably close to equilibrium with controlling factors, and their ranges in areas of introduction can circumvent this difficulty. Such studies would help identify (1) limiting factors for distributions in native ranges, (2) factors associated with a high degree of invasiveness, (3) changes in genetics and morphology since introduction, which also might contribute to invasiveness, and (4) future directions and rates of invasion as a basis for developing detection/warning systems. Findings from such comparative studies would be highly valuable for understanding the dynamics of biological invasions and for improving the effectiveness of management to prevent or control invasives.  相似文献
