为探讨解脲脲原体(Uu)的脂质相关膜蛋白(LAMPs)诱导小鼠巨噬细胞表达诱导性一氧化氮合酶(iNOS)的分子机制,从解脲脲原体提取的脂质相关膜蛋白,刺激小鼠巨噬细胞,以RT_PCR、Western blot等方法分析iNOS的表达及NO的产生;用细胞免疫化学、间接免疫荧光及Western blot等方法检测核因子κB(NF_κB)的激活,另外检测了NF_κB的特异性抑制剂二硫代氨基甲酸吡咯烷(PDTC)和蛋白酶抑制剂放线菌酮(CHX)对iNOS的表达及NF_κB激活的影响。结果表明,解脲脲原体的LAMPs通过激活NF_κB诱导小鼠巨噬细胞表达iNOS的mRNA和蛋白,且能以时间和剂量依赖方式刺激小鼠巨噬细胞产生NO,NF_κB的抑制剂PDTC或蛋白酶抑制剂放线菌酮(CHX),可抑制NF_κB的激活及iNOS的表达。由于解脲脲原体的脂质相关膜蛋白通过激活NF_κB诱导小鼠巨噬细胞表达iNOS和产生NO,因而可能是一个重要的致病因素。  相似文献

目的:研究大鼠肢体缺血/再灌注后急性肺损伤时,内皮型一氧化氮合酶(eNOS)和诱导型一氧化氮合酶(i-NOS)的表达及其在急性肺损伤发生中的作用。方法:雄性Wistar大鼠于后肢根部阻断血流后松解(4h/4h),分别给予L-Arg和氨基胍(AG)预先干预,分为control、IR、L-Arg和AG组,免疫组织化学方法检测肺组织中iNOS和eNOS的表达,同时检测肺组织中MDA、MPO、W/D和NO2^-/NO3^-值,肺组织形态学观察以评价肺损伤的程度。结果:与control组比较,I/R组eNOS表达降低,iNOS表达增强,MDA、MPO、W/D和NO2^-/NO3^-值增加。肺组织充血、炎细胞浸润,肺泡腔渗液;与I/R组比较,L-Arg组eNOS、iNOS表达无明显变化,NO2^-/NO3^-增加。MDA、MPO、W/D降低,肺组织损伤有减轻趋势,AG组eNOS表达无明显变化,iNOS活性降低,NO2^-/NO3^-减少,MDA、MPO、W/D增加,肺组织损伤有加重趋势。结论:肢体缺血/再灌注急性肺损伤过程中,iNOS表达增加,NO生成增多,在肺损伤发生中有一定的保护作用。  相似文献

摘要 目的:探讨阿司匹林通过调节Hippo途径抑制大鼠颅内动脉瘤形成的机制。方法:选择40只健康SD大鼠分为对照组(不做任何处理)、假手术组(暴露双侧肾动脉后支和左颈总动脉,但不结扎)、动脉瘤组(结扎双侧肾动脉后支和左颈总动脉后生理盐水灌胃)和阿司匹林组(动脉结扎后阿司匹林灌胃),各10只。灌胃12周后检测血清炎性因子[肿瘤坏死因子-α(TNF-α)、单核细胞趋化蛋白-1(MCP-1)、白细胞介素-6(IL-6)、IL-10]、血管内皮损伤标志物[一氧化氮(NO)、内皮素-1(ET-1)、血管内皮生长因子(VEGF)]。处死大鼠后,检测动脉瘤大小、壁厚比、内腔面积和中膜变薄长度,检测动脉瘤血管组织中Yes相关蛋白(YAP)表达情况。结果:(1)动脉瘤组和阿司匹林组大鼠Willis环上有明显凸起,且阿司匹林组大鼠凸起明显小于动脉瘤组。阿司匹林组大鼠动脉瘤大小、内腔面积和中膜变薄长度均显著小于动脉瘤组,壁厚比显著大于动脉瘤组(P<0.05)。(2)动脉瘤组和阿司匹林组大鼠血清TNF-α、MCP-1、IL-6水平显著高于对照组和假手术组,IL-10水平显著低于对照组和假手术组(P<0.05);阿司匹林组大鼠血清TNF-α、MCP-1、IL-6水平显著低于动脉瘤组,IL-10水平显著高于动脉瘤组(P<0.05)。(3)动脉瘤组和阿司匹林组大鼠血清NO水平显著低于对照组和假手术组,ET-1和VEGF水平显著高于对照组和假手术组(P<0.05);阿司匹林组大鼠血清NO水平显著高于动脉瘤组,ET-1和VEGF水平显著低于动脉瘤组(P<0.05)。(4)动脉瘤组和阿司匹林组大鼠YAP蛋白表达相对吸光值显著高于对照组和假手术组(P<0.05);阿司匹林组大鼠YAP蛋白表达相对吸光值显著低于动脉瘤组(P<0.05)。结论:阿司匹林能够显著减轻颅内动脉瘤大鼠炎性反应,改善血管内皮功能,抑制颅内动脉瘤形成,这可能与阿司匹林调控Hippo信号通路有关。  相似文献

目的:探讨脊髓水平诱导型一氧化氮合酶在吗啡依赖大鼠戒断反应中的作用。方法:健康雄性SD大鼠72只,体重200~250 g,吗啡剂量每次10 mg/kg,每日2次,隔日每次增加10 mg/kg,至第6天末次注射50 mg/kg,大鼠腹腔注射纳洛酮4 mg/kg建立吗啡依赖及戒断模型,在纳洛酮激发戒断前30 min鞘内注射iNOS特异性抑制剂氨基胍(AG)150μg。分为正常对照组、吗啡依赖组、吗啡戒断组、AG组。采用行为学(n=8)、免疫组织化学(n=6)和Western blot(n=4)方法观察鞘内应用iNOS特异性抑制剂氨基胍对吗啡依赖大鼠纳洛酮催促戒断反应和脊髓神经元iNOS表达的影响。结果:AG组戒断症状评分和戒断组促诱发痛评分均低于戒断组(P<0.05)。免疫组织化学和Western blot显示戒断组大鼠脊髓iNOS阳性神经元的数目和蛋白的表达增高,而AG组大鼠脊髓iNOS阳性神经元的数目和iNOS蛋白的表达低于戒断组(P<0.05)。结论:脊髓水平iNOS表达上调可能参与介导吗啡戒断反应。  相似文献

目的:观察鞘内注射选择性一氧化氮合酶(nNOS)和诱导型一氧化氮合酶(iNOS)抑制剂对吗啡依赖大鼠纳洛酮催促戒断反应、脊髓Fos蛋白表达和脊髓神经元nNOS和iNOS表达的影响,以探讨nNOS和iNOS在吗啡依赖和戒断反应中的作用。方法:在大鼠吗啡依赖和戒断模型上,采用行为学、免疫组织化学和Western blot方法观察鞘内应用nNOS抑制剂7-硝基吲哚(7-Ni)和iNOS抑制剂氨基胍(AG)对吗啡依赖大鼠纳洛酮催促戒断反应、脊髓Fos蛋白表达和脊髓神经元nNOS和iNOS表达的影响。结果:①鞘内注射7-Ni、AG可明显减轻吗啡依赖大鼠戒断症状,戒断组戒断症状评分为28.6±4.89,7-Ni组为16.2±3.99(P<0.01),AG组为22.94±4.0(P<0.05);戒断组TEA评分为13.5±2.55,7-Ni、AG组分别为7.5±2.56、10.5±2.71(P<0.05);②鞘内注射7-Ni、AG可减少脊髓背角Fos阳性神经元的数目,7-Ni、AG组为228.2±49.5、296.8±50.6,低于戒断组(380±71,P<0.05);③7-Ni、AG组nNOS和iNOS阳性神经元的数目分别为169±32、10.2±2.85,均低于戒断组(239±45,16.8±5.1,P<0.05),两给药组脊髓NOS蛋白的表达也显著减少。结论:nNOS和iNOS抑制剂能减轻吗啡依赖及戒断大鼠的戒断症状和在脊髓水平抑制nNOS和iNOS的表达,nNOS起主要作用而iNOS可能起辅助作用。  相似文献

目的:探讨肢体缺血/再灌注(I/R)致肝损伤时肝组织iNOS表达的变化及其意义.方法:夹闭大鼠双侧股动脉根部4 h、开放2~24 h,制备肢体I/R模型.RT-PCR检测肝组织iNOSmRNA表达的改变,免疫组化染色法观察iNOS蛋白及过氧亚硝基阴离子(ONOO-)的生成与分布,比色法测定肝组织MDA含量及SOD活性;对肢体I-R大鼠用氨基胍抑制iNOS活性后,观察其肝组织的病理学变化.结果:肢体I-R后,肝组织iNOS mRNA的表达水平较对照组显著上调(P<0.05),肝组织内出现大量iNOS及ONOO-阳性肝细胞,肝组织MDA含量升高及SOD活性降低均与对照组有显著性差异(P<0.01).应用氨基胍抑制iNOS活性,使肢体I-R所致肝组织病变减轻.结论:肢体I/R后,肝组织iNOSmRNA及蛋白表达显著上调,所诱生的高浓度NO参与介导了肢体I/R引发的肝脏损伤.  相似文献

目的探讨大鼠局灶性脑缺血再灌注后海马神经细胞一氧化氮合酶(NOS)的表达与神经细胞凋亡的关系及中药复方丹参的保护作用。方法采用大脑中动脉内栓线阻断法(MCAO)造成局灶性脑缺血再灌注模型。用原位细胞凋亡检测方法观察海马神经细胞凋亡;用免疫组织化学方法检测大鼠海马神经细胞(nNOS、iNOS)的表达并做图像分析。结果与假手术对照组比较,脑缺血再灌注2h后缺血侧海马CA1、CA3区神经细胞nNOS、iNOS表达升高,并出现神经细胞凋亡,随着再灌注时间的延长,神经细胞iNOS的表达明显增强,凋亡神经细胞数逐渐增多,至24h达高峰,但神经细胞nNOS的表达并未见明显增强。复方丹参保护组神经细胞nNOS、iNOS的表达和凋亡神经细胞数明显低于缺血再灌组(P<0.01)。结论脑缺血再灌注后缺血侧海马CA1、CA3区神经细胞nNOS的表达增强,iNOS的表达显著升高,使NO的形成增加,这可能是介导脑缺血再灌注后神经细胞凋亡的机制之一。复方丹参具有下调神经细胞nNOS、iNOS的表达,减少NO的生成,抑制细胞凋亡,减轻缺血再灌注对大鼠海马损伤的作用。  相似文献

目的: 研究一氧化氮(NO)和内皮素-1(ET-1)在大鼠肢体缺血/再灌注(LI/R)后脑损伤中的作用,探讨NO/ET-1平衡关系的变化对脑损伤的影响.方法: 在大鼠LI/R损伤模型上,应用NO合成前体物质L-精氨酸(L-Arg)、一氧化氮合酶(NOS)抑制剂氨基胍(AG)、ETA受体阻断剂BQl23进行干预,观察血浆 NO、ET-1、MDA、XOD、SOD、LDH及脑组织tNOS、iNOS、cNOS、NO、ET-1、MDA、XOD、MPO、 SOD的变化.结果: 与对照组比较,I/R组血浆MDA、XOD、LDH及脑组织MDA、XOD、MPO升高,SOD活性降低(P<0.01),脑组织tNOS和iNOS明显升高,而cNOS明显降低(P<0.01),I/R组血浆及脑组织NO、ET-1增加,NO/ET-1比值降低,脑损伤加重.应用L-Arg及BQ123后,血浆及脑组织NO/ET-1比值较I/R组升高,脑损伤减轻,应用AG后,NO/ET-1比值降低,脑损伤进一步加重.结论: 肢体缺血/再灌注后,一氧化氮与内皮素-l的比值降低时脑损伤加重.  相似文献

采用Spprague-Dawley大鼠胸主动脉中膜、外膜和培养的血管平滑肌细胞(VSMCs)作材料,鉴定不同类型的血管组织经炎性介质刺激后其一氧化氮(NO)的产生来源,闻明蛋白激酶C(PKC)和蛋白酪氨酸激酶(PTK)介导大鼠VSMCs生成NO的调控机制。大鼠VSMCs经脂多糖(LPG)和细胞因子(TNF-α,IL-1β)处理后,以剂量依赖方式促进NO释放。采用Western Blot证实经刺激的VSMCs伴有iNOS表达上调。进一步实验表明PKC和PTK参与LPS和细胞因子诱导NO生成的胞内信号转导。用PKC抑制剂H7与VSMCs共培育,H7能明显减少LPS、TNF-α和IL-1β诱导细胞NO的形成。白屈菜赤碱亦可抑制NO的生成,但HAl004对VSMCs的NO生成无抑制作用,提示PKC参与NO的生成与调控。PTK抑制剂genistein和tyrphostin AG18均能抑制由LPS、TNF-α和IL-1β引发VSMCs释放NO,同时伴iNOS蛋白表达下调,而PKC抑制剂不能阻断iNOS的表达。上述观察结果提示,PKC介导LPS和细胞因子诱导细胞合成NO可能是通过iNOS翻译后加工;而PTK则以上调iNOS表达而促增NO生成。  相似文献

空间微重力导致的心肌收缩功能下降是航天医学的重要问题, 其发生机制尚不清楚. 采用电子自旋共振(ESR)、免疫细胞化学和核酸原位杂交等技术研究了模拟微重力效应对心肌细胞一氧化氮(NO)水平、诱导型一氧化氮合成酶(iNOS)表达的影响及其调控的信号转导途径, 以探讨模拟微重力影响心肌细胞收缩功能的可能机制. 结果表明, 模拟微重力导致心肌细胞NO水平增高, iNOS蛋白及其mRNA表达上调; 非选择性的蛋白激酶抑制剂staurosporine和选择性的蛋白激酶C ( PKC )抑制剂calphostin C均可显著抑制模拟微重力下心肌细胞NO水平增高, 说明模拟微重力对iNOS表达的调节至少是部分地依赖于PKC途径. 结果提示, 心肌细胞NO途径对模拟微重力条件敏感, 该途径可能在模拟微重力影响心肌细胞收缩功能的机制中发挥重要作用.  相似文献

本实验旨在探讨诱导型一氧化氮合酶(iNOS)的激活与血压之间的关系,三组SD大鼠分别静脉输注不同浓度(0.3%,4%及8%)NaCl溶液以使其处于不同的血压水平,运用同位素标记的L-精氨酸转换成L-Citrulline 的转换率变化及Greiss反应,分别测定不同血压时iNOS的活性及NO的生成量,另四组大鼠包括正常Wistar,正常SD,高盐诱导的高血压(NaHR)及自发性高血压大鼠(SHR),经测定血压后,取主动脉血管并以Western印迹印交法测定其iNOS蛋白水平,结果表明,血压较低时,SD大鼠iNOS活基本没有改变,而在输入4%和8%NaCl并处于较高血压水平的SD大鼠,其iNOS活性及NO生存均明显升高,。此外Western 印迹表明,两种高血压大鼠主动脉组织iNOS蛋白水平均较正常Wistar及正常SD大鼠高,密度扫描表明,NaHR及SHR主动脉组织iNOS蛋白分别较正常SD大鼠及正常Wistar大鼠升高149%及261%,这一结果提示,诱导型一氧化氮合酶是血液动力学调控的重要组成部分,尤其是在血压处于较高水平时,iNOS具有重要的代偿调节作用,除细胞因子,细菌产物等之外,血压也是调节iNOS表达及活性的重要因素之一。  相似文献

目的通过观察糖皮质激素对机械通气大鼠肺组织诱导型一氧化氮合酶(iNOS)及一氧化氮(NO)表达的影响,探讨糖皮质激素对呼吸机所致肺损伤(ventilator induced lung injury,VILI)的干预作用。方法 24只雄性Wistar大鼠随机分为对照组、机械通气组、地塞米松(DXM)干预组。用逆转录-聚合酶链反应(RT-PCR)法检测肺组织iNOS mRNA表达,用免疫组织化学染色法检测肺组织iNOS蛋白表达,用硝酸还原酶法测定肺组织和血浆NO含量。结果机械通气组和DXM干预组大鼠肺组织iNOS mRNA及其蛋白表达水平,以及血浆和肺组织NO含量均明显高于对照组(P〈0.01);DXM干预组上述指标与机械通气组比较均明显降低(P〈0.01)。结论糖皮质激素可通过抑制肺组织iNOS的表达,减少NO的生成,对机械通气大鼠肺组织具有保护作用。  相似文献

本文通过慢性血液动力学实验,观察了肾髓质局部输入诱导型一氧化酶(iNOS)抑制剂AG(aminoguanidine)对Dahl盐敏感大鼠(DS)、Dahl盐抵抗大鼠(DR)及SD(Sprague Dawley)大鼠动脉血压的影响,并测定了一氧化氮(NO)代谢终产物NO2及NO3含量(UNOX)、iNOS活性、肾功能以及血浆肾素活性(PRA)。结果表明:AG能明显放大高盐(8%)引起的DS及SD大鼠  相似文献

目的:观察儿茶酚抑素(CST)在两肾一夹(2K1C)肾性高血压大鼠中的表达改变,并初步探讨其对肾性高血压的影响及作用机制。方法:36只SD大鼠随机分为假手术组(Sham)(n=15)和肾性高血压模型组(Model组)(n=21)。Model组采用两肾一夹(2K1C)手术法建立肾性高血压模型,Sham组手术操作同Model组,但不结扎左肾动脉,每周动态监测大鼠尾动脉血压。6周后各组大鼠行颈总动脉插管测定动脉压,Model组再随机分为2K1C组(n=15)与2K1C+CST组(n=6)。2K1C+CST组经颈外静脉一次性给予CST (80 μg/100 g·BW),Sham组与2K1C组给予等容积的生理盐水。各组动物经测血压、采集血标本后被处死,称取左心室加室间隔(LV+S)重量,计算(左心室+室间隔)/体重;高效液相色谱-电化学方法测定血浆中去甲肾上腺素(NE)含量,ELISA法测定血浆CST含量,硝酸还原酶法测定血浆及心室肌一氧化氮(NO)浓度;Western blot法检测延髓、肾上腺髓质、左心室和肾脏的嗜铬蛋白A (Chga)及左心室内皮型一氧化氮合酶(eNOS)、诱导型一氧化氮合酶(iNOS)蛋白表达量。结果:①与Sham组相比,2K1C组大鼠尾动脉压显著升高,左心室明显肥厚(P<0.01);血浆NE含量增高246%(P<0.01),CST水平降低56%(P<0.05);延髓Chga含量增高108%,左心室和肾脏分别降低60%和30%(P<0.05);左心室NO含量增高46%,血浆NO含量增高24%(P<0.05);左心室eNOS、iNOS蛋白表达分别增高66%和40%(P<0.05);②外源性CST显著降低2K1C大鼠颈总动脉压(P<0.05);③与2K1C组相比,2K1C+CST组左心室和血浆NO含量分别增高35%和19%(P<0.05);左心室eNOS蛋白表达高50%(P<0.05),而iNOS表达无显著统计学差异。结论:两肾肾性高血压时大鼠CST表达下调,外源性CST可能通过NO/NOS系统降低肾性高血压的作用,推测CST可能与肾性高血压的发生发展有关。  相似文献

实验采用生后14天Wistar大鼠缺血缺氧(HI)动物模型。用免疫组织化学方法观察HI复苏(HI/R0后前脑一氧化氮合酶动态表达。结果显示;神经元一氧化氮合酶(nNOS)阳性神经元主要分布于新生大鼠大脑皮层的Ⅲ-Ⅳ层。尾状核,隔核及嗅结节,HI/R早期其表达水平无明显变化;复苏48小时及5天后,可分别在右侧大脑顶皮层或右侧大脑顶皮层和尾状核区出现梗塞灶,该区nNOS阳性神经元明显减少,而诱导型一氧化氮合酶(iNOS)阳性细胞在HI/R后12小时始现于损伤侧的侧脑室;随时间的推移在损伤侧缰核,皮层,尾状核以及丘脑背外侧核,丘脑腹侧核可见iNOS阳性细胞逐渐增多并染色加深,用识别单核巨噬细胞的克隆ED1单克隆抗体检测可见ED1阳性细胞出现的时间和在脑区的分布与iNOS阳性细胞相似,本实验提示,在局灶性脑缺血缺氧早期,脑内NO的释放不依赖于nNOS阳性神经元或iNOS阳性细胞,而在局灶性脑缺血缺氧晚期,iNOS阳性细胞产生的NO可能参与了脑损伤的过程。  相似文献

The mechanism of nitrate tolerance is poorly defined. We studied the rat P450 (CYP)-catalyzed conversion of organic nitrate to nitric oxide (NO) by purified CYP isoforms and the relationship between P450 expression and nitrate tolerance following continuous infusion of organic nitrates in rats. CYP1A2 effectively formed NO from isosorbide dinitrate and nitroglycerine (NTG). The hypotensive effect of an NTG bolus injection was abolished in rats which had been previously given a continuous 48 h infusion of NTG. Nitrate tolerance was reversible to control levels 2 days after cessation of the continuous infusion. At 48 h after infusion, NTG-induced NO generation of the vessels increased in acetone (a P450 inducer)-pretreated rats, and nitrite and nitrate levels were markedly greater than in normal rats. The appearance and disappearance of P450 isoforms paralleled the conversion of organic nitrates to NO as assessed by immunohistochemistry and Western blotting. Our observations indicate that nitrate tolerance is in large part the result of decreased P450 expression and activity. Interventions that maintain or increase P450 activity may be a useful strategy to provide sustained relief from ischemic conditions in humans.  相似文献

Portal hypertension, a major complication of cirrhosis, is caused by both increased portal blood flow due to arterial vasodilation and augmented intrahepatic vascular resistance due to sinusoidal constriction. In this study, we examined the possible involvement of resident macrophages in the tone regulation of splanchnic blood vessels using bile duct ligated (BDL) portal hypertensive rats and an in vitro organ culture method. In BDL cirrhosis, the number of ED2-positive resident macrophages increased by two- to fourfold in the vascular walls of the mesenteric artery and extrahepatic portal vein compared with those in sham-operated rats. Many ED1-positive monocytes were also recruited into this area. The expression of inducible nitric oxide (NO) synthase (iNOS) mRNA was increased in the vascular tissues isolated from BDL rats, and accordingly, nitrate/nitrite production was increased. Immunohistochemistry revealed that iNOS was largely expressed in ED1-positive and ED2-positive cells. We further analyzed the effect of iNOS expression on vascular smooth muscle contraction using an in vitro organ culture system. iNOS mRNA expression and nitrate production significantly increased in vascular tissues (without endothelium) incubated with 1 μg/ml lipopolysaccharide (LPS) for 6 h. Immunohistochemistry indicated that iNOS was largely expressed in ED2-positive resident macrophages. α-Adrenergic-stimulated contractility of the mesenteric artery was greatly suppressed by LPS treatment and was restored by N(G)-nitro-L-arginine methyl ester (NO synthase inhibitor); in contrast, portal vein contractility was largely unaffected by LPS. Sodium nitroprusside (NO donor) and 8-bromo-cGMP showed greater contractile inhibition in the mesenteric artery than in the portal vein with decreasing myosin light chain phosphorylation. In the presence of an α-adrenergic agonist, the mesenteric artery cytosolic Ca(2+) level was greatly reduced by sodium nitroprusside; however, the portal vein Ca(2+) level was largely unaffected. These results suggest that the induction of iNOS in monocytes/macrophages contributes to a hypercirculatory state in the cirrhosis model rat in which the imbalance of the responsiveness of visceral vascular walls to NO (mesenteric artery > portal vein) may account for the increased portal venous flow in portal hypertension.  相似文献

目的:观察硫化氢(H2S)对1型糖尿病大鼠膈肌一氧化氮(NO)含量和诱导型一氧化氮合酶(iNOS)活性的影响。方法:将32只雄性SD大鼠随机分为4组:正常组(NC组)、糖尿病组(DM组)、糖尿病治疗组(DM + NaHS组)和NaHS对照组(NaHS组)(n=8)。采用一次性腹腔注射链脲佐菌素55 mg/kg制备1型糖尿病大鼠模型,造模成功后第4周起,DM + NaHS组和NaHS组大鼠腹腔注射NaHS溶液14μmol/kg干预治疗。连续注射5周后,测大鼠空腹血糖值(FBG)和膈肌重量/体重量比(DW/BW);HE染色观察膈肌显微结构变化;利用NOS分型测试盒测膈肌组织iNOS活性;硝酸还原法测定膈肌组织NO含量;利用RT-PCR和Western blot分别检测膈肌组织iNOS mRNA和蛋白表达。结果:与NC组比较,DM组大鼠FBG显著升高,膈肌显微结构损伤明显,DW/BW下降,膈肌组织iNOS活性和NO含量显著增加,iNOS mRNA和蛋白表达明显增高,NaHS组各项指标差异无统计学意义。与DM组比较,DM + NaHS组膈肌显微结构明显改善,DW/BW增高,膈肌组织iNOS活性和NO含量明显下降,iNOS mRNA和蛋白表达显著降低。结论:外源性补充H2S可能通过下调膈肌组织iNOS活性和蛋白表达,降低NO含量,进而保护糖尿病大鼠膈肌的功能。  相似文献

Hemorrhagic shock (HS) has been implicated in the induction of inducible nitric oxide synthase (iNOS) that leads to increase production of nitric oxide (NO). Recently, NO has been implicated to cause hyporesponsiveness of blood vessel in vitro towards vasoconstrictors in refractory (decompensated) HS. In our in vivo model, we examined the effects of aminoguanidine (AG), a known iNOS inhibitor, with angiotensin II (ANG II), a vasoconstrictor, following hemorrhagic shock decompensatory phase (HSDP) on percentage survival, vascular responsiveness, mean arterial blood pressure (MABP), heart rate and mean nitrate/nitrite levels in anaesthetized rats. HSDP (3 h) was achieved via constant pressure method (40-45 mmHg). MABP and heart rate was measured via the left carotid artery. Plasma collected from HSDP rats was used to measure nitrate/nitrite levels. Vascular hyporeactivity to ANG II was carried out using HSDP aortic strips, precontracted with KCl and noradrenaline. Sham-operated rats served as controls. HSDP rats decreased percentage survival, vascular contractility to ANG II and noradrenaline, MABP, heart rate while showing increased levels of nitrate/nitrite. Infusion of AG with ANG II, increased percentage survival and had reversed these cardiovascular effects of HSDP rats. This study indicates that excessive NO formation from iNOS activity induces vascular hyporeactivity and decompensation in HSDP. This might suggest that selective NOS inhibitor, AG, when coupled with ANG II, show reduction in NO's effect in HSDP.  相似文献

The aim of the present work was to investigate the alterations in nitric oxide synthase (NOS) expression and nitrate and nitrite (NOx) content of different arteries from simulated microgravity rats. Male Wistar rats were randomly assigned to either a control group or simulated microgravity group. For simulating microgravity, animals were subjected to hindlimb unweighting (HU) for 20 days. Different arterial tissues were removed for determination of NOS expression and NOx. Western blotting was used to measure endothelial NOS (eNOS) and inducible NOS (iNOS) protein content. Total concentrations of NOx, stable metabolites of nitric oxide, were determined by the chemiluminescence method. Compared with controls, isolated vessels from simulated microgravity rats showed a significant increase in both eNOS and iNOS expression in carotid arteries and thoracic aorta and a significant decrease in eNOS and iNOS expression of mesenteric arteries. The eNOS and iNOS content of cerebral arteries, as well as that of femoral arteries, showed no differences between the two groups. Concerning NOx, vessels from HU rats showed an increase in cerebral arteries, a decrease in mesenteric arteries, and no change in carotid artery, femoral artery and thoracic aorta. These data indicated that there were differential alterations in NOS expression and NOx of different arteries after hindlimb unweighting. We suggest that these changes might represent both localized adaptations to differential body fluid redistribution and other factors independent of hemodynamic shifts during simulated microgravity.  相似文献
