McVicar A. H. 1977. The bothridial hooks of Acanthobothrium quadripartitum Williams, 1968 (Cestoda: Tetraphyllidea): their growth and use in taxonomy. International Journal for Parasitology7: 439–442. Bothridial hooks are absent from juvenile Acanthobothrium quadripartitum in Raja naevus but develop to full size before there is much growth of the bothridia and strobila. There is a linear relationship between the lengths of the different components of the hooks during their development and it is suggested that the ratios of these lengths to the total hook length may be useful parameters in dstinguishing between species of Acanthobothrium. Principal component analysis of published hook dimensions gave good separation of most species of Acanthobothrium.  相似文献

The aim of this study was to search for entomopathogenic fungi that infect wild cockroaches in forest ecosystems in two protected natural areas of Argentina. Two isolates of Metarhizium argentinense were obtained and identified from wild cockroaches (Blaberidae: Epilamprinae) through the use of morphological characteristics and molecular phylogenetic analyses. This novel species was found in Argentina and is a member of the Metarhizium flavoviride species complex. Phylogenetic analyses, based on sequence similarity analysis using internal transcribed spacer (ITS) and a set of four protein-coding marker sequences (EF1A, RPB1, RPB2 and BTUB), supported the status of this fungus as a new species. In addition, we tested the biological activity of the new species through assays against Blattella germanica nymphs and found that the two evaluated isolates were pathogenic. However, isolate CEP424 was more virulent and caused a confirmed mortality of 76 % with a median lethal time of 7.2 d. This study reports the southernmost worldwide location of a Metarhizium species that infects cockroaches and will help expand the knowledge of the biodiversity of pathogenic fungi of Argentine cockroaches.  相似文献

Between 1993 and 1996, 26 individuals of two species of horn sharks were collected from the upper Gulf of California and their spiral intestines examined for onchobothriid tapeworms. Heterodontus francisci was found to host Acanthobothrium puertecitense n. sp. Based on the criteria of Ghoshroy & Caira (2001), this is a Category 4 species (with >15mm total length, >50 proglottids, >80 testes and a symmetrical ovary). It differs from the four Category 4 species previously reported from either the western Atlantic or eastern Pacific Oceans, including A. bajaense (emend.) which was described from H. francisci, in hook-shape, total length, number of proglottids, number of testes and number of columns of vitelline follicles. H. mexicanus was found to host Acanthobothrium santarosaliense n. sp., which, based on the criteria of Ghoshroy & Caira (2001) is a Category 3 species (with >15mm total length, >50 proglottids, >80 testes and an asymmetrical ovary). A. santarosaliense differs from the five Category 3 species previously reported from either the western Atlantic or eastern Pacific Oceans in its size, euapolytic nature, lack of dark matrix from the bases of the hooks, hook-shape, and distribution and number of testes. In addition to the difference in ovarian symmetry, A. puertecitense possesses fewer testes, fewer proglottids, is a shorter worm, possesses a more anterior genital pore and vitelline follicles which extend further forward anteriorly than in A. santarosaliense. This represents the first report of onchobothriids from H. mexicanus and the first report of onchobothriids from H. francisci in the Gulf of California. The apparent host-specificity of both new species should be viewed with caution until a greater sample of host individuals of both species can be examined. The fact that both new tapeworm species are known only from the Gulf is also considered to be a preliminary result, given the small sample of host individuals examined from these regions to date. This brings the total number of species of Heterodontus known to host Acanthobothrium to four and the total number of Acanthobothrium species described from heterodontiform sharks to five.  相似文献

Despite the large number of species described to date for the onchoprotepcephalid genus Acanthobothrium (207), only 16 named species have a genetic sequence. With this background, specimens of adult cestodes of the stingray Hypanus longus were collected off San Blas, Nayarit, and onchoproteocephalid larvae in the carangid fish Trachinotus rhodopus from Puerto Ángel, Oaxaca, both located on the Pacific coast of Mexico. The objective of this work is to investigate the phylogenetic position of these adults and larvae using nuclear ribosomal markers (18S rDNA and 28S rDNA). Morphologically, adult specimens were identified as Acanthobothrium cleofanus; larvae were identified only to family level. The phylogenetic position of both taxa was investigated based on the information of two nuclear molecular markers analyzed under Parsimony (PA) and Bayesian Inference (BI) methods. The newly generated sequences of A. cleofanus from Nayarit are identical to the sequences of several samples of Acanthobothrium sp. collected in the Mexican Pacific, which sequence are available in GenBank; DNA sequences obtained from onchoproteocephalid larva clearly place this taxon within Acanthobothrium but representing an independent lineage. In the resulting phylogenetic trees, Uncibilocularis okei was found nested within Acanthobothrium with an unstable position depending on the optimality criteria, indicating the need for more molecular analyzes with a greater number of species of both genera prior to define its phylogenetic relationships.  相似文献

Osteoarticular brucellosis is the most frequent complication of active disease. A large amount of cells in bone are osteocytes. Since bone remodeling process is regulated by hormones we sought to study the effect of cortisol and DHEA in Brucella abortus-infected osteocytes. Cortisol treatment inhibited the expression of IL-6, TNF-α, MMP-2 and RANKL in B. abortus-infected osteocytes. DHEA could reverse the inhibitory effect of cortisol on MMP-2 production. B. abortus infection inhibited connexin 43 (Cx43) expression in osteocytes. This expression was increased when cortisol was incorporated during the infection and DHEA treatment partially reversed the effect of cortisol. Osteocytes-infected with B. abortus induced osteoclast's differentiation. Yet, the presence of cortisol, but not DHEA, during osteocyte infection inhibited osteoclastogenesis. Glucocorticoid receptor (GR) is implicated in the signaling of cortisol. Infection with B. abortus was able to increase GRα/β ratio. Levels of intracellular cortisol are not only dependent on GR expression but also a result of the activity of the isoenzymes 11β-hydroxysteroid dehydrogenase (11β-HSD)-1 (cortisone to cortisol conversion), 11β-HSD2 (cortisol to cortisone conversion). B. abortus infection increased 11β-HSD 1/2 ratio and cortisone mimicked the effect of cortisol. Our results indicated that cortisol and DHEA could modulate osteocyte responses during B. abortus infection.  相似文献

Addition of nifurtimox (a nitrofuran derivative) to Trypanosoma cruzi culture (epimastigote) forms induced an increase in the respiratory rate and the release of H2O2 from the whole cells to the suspending medium. Growth-inhibiting concentrations of nifurtimox were able to stimulate O2? production by the T. cruzi mitochondrial fraction supplemented with NADH (or succinate), and also to enhance the generation of O2? by the microsomal fraction with NADPH as reductant.  相似文献

The Somuncurá Plateau is a Protected Natural Area located in the middle of the northern extra-Andean arid Patagonia. Inhabited by at least 20 small mammal species, is the place with the uppermost species richness in Patagonia. The aim of this study was to examine the parasite remains from micromammal coprolites collected in association with a bone sequence recovered at the east of the Somuncurá Plateau (site “Alero Las Lechuzas”). Coprolites came from the four temporal units previously defined: unit I (4790?±?100?yrs. 14C B.P.), unit II, unit III (7840?±?120?yrs. 14C B.P.) and unit IV. Each coprolite was processed, rehydrated, homogenized, processed by spontaneous sedimentation and examined using a light microscope. Coprolites and eggs were described, measured and photographed. Samples were positive for two nematode species: Helminthoxys caudatus Freitas, Lent & Almeida, 1937 (Oxyurida, Oxyuridae) and Trichuris spp. (Trichinellida: Trichuridae). This is the first paleoparasitological study developed for the Somuncurá Plateau Protected Area. Moreover, this is the first time that the genus Helminthoxys is reported from ancient times worldwide. Coprolites were attributed to the mountain cavy Microcavia australis (Rodentia, Caviidae).The presence of H. caudatus for the Middle Holocene of northern Patagonia contributes to the study of the history of the histricomorphs and pinworms relationships.  相似文献

Anurans are extremely diverse amphibians with a unique Bauplan, whose origin, early ecomorphological diversification, and adaptive significance remain elusive. Their early fossil record is improving at an accelerated pace worldwide, but its contribution to these issues is still wanting. Here we explore ecomorphological diversity among Early Cretaceous (Barremian) frogs that inhabited a large subtropical wetland from Iberia by inferring locomotor abilities with a phylogenetic flexible discriminant analysis on data from limb proportions, which strongly correlate with locomotion. The results show a remarkable diversity among these frogs when compared with the extant diversity from this region and from tropical and subtropical wetland assemblages worldwide, encompassing miniature to medium-size jumpers and dedicated swimmers with more extreme proportions than extant ‘archeobatrachians,’ but also more generalized jumping and/or swimming forms. This agrees with the inferred wetland paleoenvironment and the hypothesis regarding early frogs as small poor jumpers or swimmers that evolved their peculiar Bauplan in aquatic environments.  相似文献

A method for the isolation of oligodendroglial cells from young and adult whole rat brains, using a Percoll density gradient is presented. The minced tissue, incubated in a balanced salt solution containing 0.1% trypsin is further dissociated by forcing it through nylon screens to 145 and 74 μm pore size. The crude suspension is then mixed with an isosmotic Percoll solution and centrifuged for 15 min. An in situ generated density gradient allows the separation of five bands, only one of which (Band C) lying between δ1.050 and δ1.062 contains cellular elements. The isolated cells show the typical morphological characteristics of oligodendroglia.A detailed morphological study of the cells isolated from whole brains of 10-, 30- and 120-day old rats is presented for the first time in the literature and immunocytochemical characterization is carried out using specific (antigalactocerebroside) and non specific (anti-glial fibrillary acidic protein) anti-sera.The method is simple and rapid and isosmotic conditions are maintained throughout, resulting in a better preservation of cell integrity. It represents an improvement over the two previous methods described in the literature and will be useful for studying different developmental events (biochemical and morphological) occurring in oligodendroglial cells at early stages of myelin formation.  相似文献

Background

Candida auris and Candida haemulonii are emerging and multiresistant pathogens. C. auris has produced hospital outbreaks and is misidentified by phenotypic-based methods. The only reliable identification methods are DNA sequencing and MALDI-TOF.

Aims

To develop a classical-PCR method capable of rapidly and accurately identify C. auris and C. haemulonii.

Methods

A multiplex PCR was carried out in one tube that included an internal control and oligonucleotides that specifically hybridize to the ITS2 region of C. auris and C. haemulonii. The usefulness of the new method was verified by testing a collection of 50 strains of 20 different species (previously identified by ITS sequencing). The selection of species was made in order to emulate the C. auris panel used by the CDC to validate diagnostic tools. In addition, other yeast species not included in the aforementioned panel were incorporated based on reported identification errors.

Results

The results obtained with the proposed protocol were in total agreement with those obtained by ITS sequencing.

Conclusions

We present a PCR method able to unequivocally identify C. auris and differentiate it from C. haemulonii. It is inexpensive, fast and it could be a useful tool to reduce the chances of a C. auris outbreak.  相似文献

Five new species of Acanthobothrium (Tetraphyllidea: Onchobothriidae) are described from the spiral intestine of the Freshwater whipray, Himantura chaophraya, in the Kinabatangan River in Malaysian Borneo. Based on criteria set forth in a previous categorization scheme for species of Acanthobothrium, these consist of 3 Category 1 species, Acanthobothrium asnihae n. sp., Acanthobothrium saliki n. sp., and Acanthobothrium zainali n. sp.; a Category 8 species, Acanthobothrium etini n. sp.; and a Category 2 species, Acanthobothrium masnihae n. sp.. Acanthobothrium asnihae n. sp. differs from all Category 1 species in its possession of a horizontal band of weak musculature that divides the posterior loculus in half. Among Category 1 species, A. saliki n. sp. differs from all but Acanthobothrium southwelli in its possession of postovarian testes. It differs from A. southwelli in its possession of fewer testes and a greater number of proglottids. Acanthobothrium zainali n. sp. differs from the 25 other Category 1 species in a combination of overall size, muscular pad and hook shape, arrangement and number of testes, ovary configuration in cross section, position of ovarian isthmus, and genital pore position. Acanthobothrium etini n. sp. is distinguished from all 5 other Category 8 species in its lack of testes from the proglottid antiporal and postporal regions and in testis number. Acanthobothrium masnihae n. sp. differs from the 35 other Category 2 species in its possession of fewer testes, postporal testes, or a greater number of proglottids. A key to Acanthobothrium species parasitizing H. chayophraya is presented. This represents the first report of Acanthobothrium from freshwater stingrays belonging to a family other than the Potamotrygonidae.  相似文献

ABSTRACT. Described herein are the morphology and certain morphogenetic stages of a new freshwater ciliate species, Deviata polycirrata n. sp., and of Deviata bacilliformis recorded in the soil of a dried temporary pond from Argentina. Ciliates were studied alive and after silver impregnation with Protargol. Deviata polycirrata n. sp. measures 130–180 × 45–70 μm in vivo. The species possesses 8–9 long cirral rows on the right and 9–13 on the left of the oral zone, and 3 dorsal rows of dikinetids. The adoral zone is composed of 39–48 membranelles. There are four macronuclear nodules and usually two micronuclei. A single contractile vacuole is located equatorially on the left body margin. This new species mainly differs from its congeners in having a higher number of cirral rows, the three long dorsal rows of dikinetids (vs. usually one to two dorsal rows of dikinetids), and a higher number of adoral membranelles. The other species reported here, D. bacilliformis, is recorded for the first time in Argentina. Unlike previous observations on this species, on the dorsal surface there are cirral rows that are preceded by cilia (combined cirral rows), and stomatogenesis begins with the proliferation of non‐ciliferous basal bodies some distance posterior to the buccal vertex.  相似文献

A new Category 1 species of Acanthobothrium van Beneden, 1850 is described from the cowtail stingray Pastinachus atrus (Macleay) collected from the Gulf of Carpentaria near Weipa, Queensland, Australia. This species is unique among Acanthobothrium Category 1 species in that it retains gravid proglottids on its strobila. It differs further from the 34 other Category 1 species in total length, proglottid number and testis number. The host identities of other Acanthobothrium species reported from Pastinachus are revised based on recent taxonomic work on rays of this genus. Given the revised host taxonomy, according to which P. atrus is the only member of its genus occurring off Australia, this should be considered to be the fifth species of Acanthobothrium reported from this Australian endemic species.  相似文献

Abstract

Meristies and morphometries are reported for 81 specimens of Alepisaurus ferox and 31 of A. brevirostris, mostly from the Pacific Ocean. The holotype and topotype of A. ferox and two paratypes of A. brevirostris, all from the Atlantic Ocean, were also examined. For specimens longer than aJoout 500 mm SL, ferox is distinguishable from brevirostris by its relatively longer head and snout. In ferox the dorsal fin origin is a1bove or behind the hind margin of the operculum; in brevirostris it is in front. Paoific Ocean ferox have significantly fewer dorsal fin rays (mean 36.6) than Atlantic (40.5) and Indian Ocean (40.8) examples. In the Pacific, ferox can be distinguished from brevirostris by its fewer dorsal rays, but in the Atlantic and the Indian Ocean there is considerable overlap in dorsal ray counts. A. ferox is found world-wide, whereas brevirostris is not known from the North Pacific. The northernmost record of brevirostris in the Pacific is from 14°47′S.  相似文献

Nosema ceranae is the most prevalent endoparasite of Apis mellifera iberiensis and it is a major health problem for bees worldwide. The infective capacity of N. ceranae has been demonstrated experimentally in honey bee brood, however no data are available about its prevalence in brood under natural conditions. Thus, brood combs from 10 different hives were analyzed over two consecutive years, taking samples before and after winter. A total of 1433 larvae/pupae were analyzed individually and N. ceranae (3.53%) was the microsporidian most frequently detected, as opposed to Nosema apis (0.42%) which was more frequently detected in conjunction with N. ceranae (0.71%). The active multiplication of both microsporidians was confirmed by the expression (real-time-PCR) of the N. ceranae polar tube protein 3 gene and/or the N. apis RNA polymerase II gene in 24% of the brood samples positive for Nosema spp. Both genes are related to microsporidian multiplication. As such, N. ceranae multiplication was confirmed in 1.06% of the samples, while N. apis multiplication was only observed in co-infections with N. ceranae (0.07%). Brood cells were analyzed for the presence of Nosema spp., as those are the immediate environment where the brood stages develop. The brood samples infected by Nosema spp. were in brood cells in which that microsporidians were not detected, while brood cells positive for N. ceranae hosted brood stages that were not apparently infected, indicating that this is unlikely to be the main pathway of infection. Finally, the colonies with brood infected by N. ceranae showed higher levels (numbers) of infected adult bees, although the differences were not significant before (P = 0.260), during (P = 0.055) or after (P = 0.056) brood sampling. These results show that N. ceranae is a bee parasite ubiquitous to all members of the colony, irrespective of the age of the bee. It is also of veterinary interest and should be considered when studying the epidemiology of the disease.  相似文献

Chalicotherium brevirostris was named by Colbert based on a skull lacking mandibles from the late Middle Miocene Tunggur Formation, Tunggur, Inner Mongolia, China. Here we describe new mandibular materials collected from the same area. In contrast to previous expectations, the new mandibular materials show a long snout, long diastema, a three lower incisors and a canine. Cbrevirostris shows some sexual dimorphism and intraspecific variation in morphologic characters. The new materials differ from previously described C. cf. brevirostris from Cixian County (Hebei Province) and the Tsaidam Basin, which may represent a different, new species close to Cbrevirostris. The diagnosis of Cbrevirostris is revised.  相似文献

Preference for low salinity inland water may be an important factor in the immigration of postlarval penaeid shrimp from the sea to their nursery grounds, such as estuaries, bays, and coastal lagoons. In the present work, it was shown that four species from west Mexico - Penaeus vannamei Boone, P stylirostris Stimpson, P. californiesis (Holmes), and P. brevirostris Kingsley - all prefer salinities less than that of normal sea water. Older or larger postlarvae preferred lower salinities than early postlarvae. Of the four species, P. vannamei has the lowest preference and this species predominates in the coastal lagoon fisheries of the area. Postlarvae of P. californiensis and P. brevirostris were also shown to prefer lagoon water to sea water. The importance of these preferences is discussed in relation to the normal postlarval migrations.  相似文献

A method of affinity chromatography developed for the purification of species-specific antigens from Toxocara canis adult worms is described. Immunochemical analyses by polyacrylamide gel electrophoresis, immunoelectrophoresis and immunodiffusion showed that ‘pure’ antigen contained fewer but more specific proteins than ‘crude’ antigen. Purified antigens and parasite sections from four parasite species (Toxocara canis, Dirofilaria immitis, Angiostrongylus cantonensis and Ascaris lumbricoides) were used in immunofluorescence tests to measure serum antibody levels in animals with natural or experimental T. canis infections and people with zoonotic toxocariasis. ‘Pure’ antigen showed higher specificity and sensitivity than ‘crude’ antigen in serological testing.  相似文献
