A retrospective study was conducted to evaluate the outcome of 223 prostaglandin F(2)alpha (PGF) treatments given to lactating Holstein and Jersey cows over a 44-mo period. The cows were part of a commercial dairy herd. Treated cows were given PGF (25 mg i.m.) because they had not been observed in estrus or because they had been diagnosed as having a luteal cyst or pyometra. Outcome of treatments was assessed for frequency of response to treatment (the detection of estrus), time interval to detection and pregnancy outcome of breedings at PGF induced estrus. Of the 223 administrations of PGF, 70% (156 223 ) were given for unobserved estrus, 12.1% (27 223 ) were given for cystic ovarian disease and 15.2% (34 223 ) were given for pyometra. Estrus was detected in 55.3% (120 217 ) of cows over a period of 36 to 130 hours after PGF administration. Interval to estrus and response rates were not influenced by the reason for treatment. Mean time interval to response was 79.1 h. Time interval for response was affected by season (P < 0.01) housing location (P < 0.01) and lactation number (P < 0.05). Insemination was performed following 157 of the treatments. For cows treated for unobserved estrus, insemination was performed in response to estrus or at 80 h if no estrus had been previously seen. Of this group, 39.7% (58 146 ) failed to show estrus and were inseminated at 80 h. Reinsemination was required for 22.7% (20 88 ) of the cows that manifested estrus following insemination at 80 h. Overall conception rate was 41.4% (65 157 ). Cows inseminated in response to estrus had a 53.4% (47 88 ) conception rate, and appointment bred (80 h) cows had a 20.7% (12 58 ) conception rate. Factors significantly influencing conception were response to PGF administration (P < 0.05), season (P < 0.01) and lactation number (P < 0.01). It was concluded that estrus detection efficiency affects the outcome of PGF administration and that conception rates resulting from 80 h breeding when no estrus is observed may be unacceptably low.  相似文献

Data were collected on reproductive, health and production events in 45 dairy herds in southwestern and eastern Ontario, Canada, from July 1990 to July 1993. Mean and median days in milk at first estrus, at first service, and at conception were 85.1 and 83.9, 94.0 and 88.9, and 126.3 and 115.5 d, respectively. The mean estrus detection rate was 48.3%, with a minimum of 28.4% and a maximum of 64.2%. The mean overall conception rate was 46.7%, with a minimum of 29.8% and a maximum of 70.7%. The most commonly used used therapeutic agents were GnRH (11.5% lactational incidence risk) and PG (13.1% lactational incidence risk). Estrus management accounted for the largest portion of PG use in the herds studied (69.4%). Results of a two sample t-test indicated no significant difference in reproductive performance among herds housed in free-stall compared with those housed in tie-stall barns. Multivariate linear regression models of 3 reproductive performance measures indicated that cows with ovarian cysts and/or increased peak milk production had significantly longer calving-to-first estrus and calving-to-conception intervals as well as lower conception rates than average producing cows without cysts. Cows with metritis were found to have significantly shorter calving-to-conception intervals but no detectable difference in conception rate or calving-to-first estrus interval than their herdmates. As lactation number increased reproductive performance improved. Cows that were observed in estrus earlier conceived earlier. Animals receiving uterine infusions had significantly longer calving-to-conception intervals and lower conception rates than their herdmates. There was no detectable difference in reproductive performance for cows treated with GnRH or PG compared with the nontreated cows. An increased estrus detection rate was associated with shorter calving-to-conception intervals and lower conception rates. Based on the range of performance in the study herds, the factors with the greatest potential influence on calving-to-conception interval in the average herd were the estrus detection rate and conception rate.  相似文献

This study compared the calving to conception intervals for cows in AI pens with cows exposed to natural service sires, controlling for milk production, mastitis occurrence, parity and calving month effects. Records from 10 western United States dairy herds (mean herd size = 2058 cows) were evaluated retrospectively over an 18-month period. Eight bull breeding analysis cohorts were created (the first cohort 0-50 days in milk and the remaining cohorts at 25 days in milk intervals through 226 days). The cohorts contained non-pregnant cows that were first moved into bullpens during the described cohort period. Equal numbers of non-pregnant cows only exposed to AI during the cohort period were randomly selected from the pool of eligible non-pregnant cows. An AI cow was used only once in the data analysis, but was included in a bull breeding cohort at a later date if she remained non-pregnant and was transferred to a bullpen. Univariate and multivariate survival analysis was used to compare the calving to conception intervals. Cows in AI groups had higher hazard rates for pregnancy across all cohorts. Parity and milk production were significantly associated with risk for pregnancy. In herds that practice a mixture of AI and bull breeding, overall herd reproductive performance might be improved by allowing cows more opportunities at AI prior to moving them into clean-up bullpens.  相似文献

Two field trials were designed to evaluate use of the prostaglandin F analog cloprostenol (CP) in dairy cattle with unobserved estrus and a palpably mature corpus luteum (CL). In Trial 1, 98 cows were treated with CP (500 μg IM) and 83 were given saline. Cows in both groups were assigned to be inseminated based on observed signs of estrus. Cows treated with CP were inseminated sooner than controls although heat detection still presented some difficulty. Conception rates were the same for both groups. In Trial 2, 154 cows were treated with CP for unobserved estrus and were assigned to be inseminated at estrus (78 cows) or were inseminated twice by appointment at 72 and 96 hours after treatment (76 cows). Appointment breeding eliminated problems associated with detection of estrus so nearly all cows were inseminated soon after treatment. In addition, cows bred twice by appointment had a higher conception rate than those bred based on signs of estrus. Induction of luteolysis with the prostaglandin F analog CP in cows with unobserved estrus and a CL should prove to be a very effective addition to reproductive herd health programs. Successful use of CP for this purpose is dependent upon accurate diagnosis of the presence of a mature CL. In these trials 83 (92.4%) of 90 cows sampled had blood serum progesterone concentrations indicative (≥ 2.0 ng/ml) of significant luteal tissue activity.  相似文献

The objective of this study was to compare the conception rate for fixed-timed artificial insemination (FTAI) and observed heat artificial insemination (HAI) prior to the scheduled FTAI in Ovsynch and Heatsynch synchronization protocols. In Experiment 1, lactating dairy cows (n=535) received two set-up injections of 25mg prostaglandin F(2alpha) (PGF(2alpha)) i.m., 14 days apart starting at 36+/-3 days in milk (DIM). Cows were blocked by parity and were randomly allocated to either Ovsynch or Heatsynch groups. All cows received 100 microg of GnRH i.m. 14 days after the second set-up injection of PGF(2alpha), followed by a third injection of 25mg PGF(2alpha) i.m., 7 days later. In the Ovsynch group, HAI cows (n=29) were bred on standing estrus after the third PGF(2alpha) before the scheduled second GnRH, whereas FTAI cows (n=218) that were not observed in estrus, received a second injection of 100 microg of GnRH i.m., 48 h after the third PGF(2alpha) and received TAI 8 h after the second GnRH. In the Heatsynch group, all cows (n=288) received 0.5 mg of estradiol cypionate (ECP) 24 h after third PGF(2alpha) and HAI cows (n=172) were bred on standing estrus and FTAI cows (n=116) that were not observed in estrus, received TAI 72 h after the third PGF(2alpha). In Experiment 2, repeat breeder cows (n=186) were randomly assigned to either Ovsynch or Heatsynch groups. The FTAI and HAI cows were inseminated similar to Experiment 1. All cows were observed for estrus three times daily. The associations with the conception rate were modeled with logistic regression separately for Experiments 1 and 2. Of all the variables included in the model in Experiment 1, type of AI (HAI versus FTAI, P=0.0003) and parity (primiparous versus multiparous, P=0.05) influenced the first service conception rate. Over-all conception rate and first service conception rate for HAI cows were higher compared to FTAI cows (33.8% versus 21.3%, and 35.3% versus 21.0%; P=0.001). In the Heatsynch group, cows that received HAI had significantly higher over-all conception rate and first service conception rate compared to FTAI (35.2% versus 17.3% and 36.0% versus 15.5%; P=0.0001). The conception rates in repeat breeder cows for HAI and FTAI (30.1% versus 22.3%) were not different (P>0.1). In conclusion, it was recommended to include AI at observed estrus and fixed-time AI for cows not observed in estrus in order to improve the conception rate in synchronization protocols.  相似文献

Trials were carried out on 1184 dairy cows calved at least six weeks before treatment and 255 heifers to determine effectiveness of the prostaglandin analogue, cloprostenol to control estrus. In trial 1, following two injections of cloprostenol given 12 days apart, there was no difference in calving rate following AI either at 72 and 96 hr after treatment (71 163 ) or at a detected estrus (53 118 ) compared to control cows bred at estrus (54 110 ). In trial 2, treated cows were injected once after 5 to 7 days of estrous detection and AI. The calving rate following AI either at 72 and 96 hr after cloprostenol (46 100 ) or at a detected estrus (39 71 ) was similar to that in control cows bred at estrus (45 86 ). In trial 3, cows were bred at a detected estrus after the first cloprostenol injection. Twelve days after this injection, cows not bred were given a second injection and bred 72 and 96 hr later. The calving rate in treated cows bred at estrus after the first injection (66 138 ) was similar to calving rate in controls (55 95 ). However, calving rate in cows given a second injection and bred 72 and 96 hr later was significantly (P angle 0.05) lower (30 98 ). Similar results were obtained in heifers, except calving rate in trial 3 after the second cloprostenol injection was not reduced.  相似文献

This study was designed to determine conception rates in dairy cows after timed-insemination and simultaneous treatment with gonadotrophin releasing hormone (GnRH) and/or prostaglandin F2 alpha (PGF2alpha). A total of 2352 cows was randomly assigned to six groups. Cows in Groups 1 to 5 were palpated per rectum to determine the presence of a corpus luteum (CL) on the ovary, and blood samples were obtained for the determination of plasma progesterone (P4) concentrations. Cows with a CL and P4 concentrations >1 ng/ml were treated (Day 0) with PGF2alpha (25 mg, i.m.) and were observed for estrus. Cows in estrus prior to 72 hours after treatment (Group 5, n = 106) were bred, but were not treated. Cows not observed in estrus by 72 hours were divided into four remaining groups, were bred between 72 and 80 hours and were assigned as follows: Cows in Group 1 (n = 203) were not treated; Cows in Group 2 (n = 200) were treated with GnRH (100 ug, i.m.); Cows in Group 3 (n = 201) were treated with PGF2alpha (25 mg, i.m.); and cows in Group 4 (n = 202) were treated with both GnRH and PGF2alpha. Cows in Group 6 (n = 1440) were not treated with PGF2alpha on Day 0 and were estrual cows that were bred on days when cows in Groups 1 to 5 were time-inseminated. The percentage of cows in all groups pregnant at 45 to 50 days after one insemination was compared using analysis of variance (P<0.05). The conception rate of cows in Group 2 was significantly higher than that of cows in Groups 1 to 4. There was a significant group-by-season interaction. Cows treated with GnRH during the spring had a higher conception rate than at other times of the year. Conception rates of cows in Groups 1 to 4 that were inseminated during the summer were low and not significantly different from each other. Conception rates of cows in Groups 5 and 6 inseminated during the summer were not significantly different from each other, but were significantly higher than that of cows in Groups 1 to 4 that were inseminated during the summer.  相似文献

Dairy cows (n = 199) were assigned randomly at Day 14 post partum to a control group or a programmed reproductive treatment group (PRT). Cows in the PRT group received 8 ug of GnRH on Days 14 and 50 post partum and 25 mg of PGF2alpha injected on Days 21, 34 and 57 post partum. Cows in the PRT group had a greater frequency of progesterone (P4) concentrations > 1 ng/ml (50% vs 30%; P < 0.01). Frequency of cows having P4 > 1 ng/ml at both Days 21 and 34 post partum was greater in the PRT group than in the control group (39% vs 20%; P < 0.01). Accumulation of days with a palpable cystic ovary was lower in the PRT than the control group (11.4% vs 6.5%; P < 0.05). The frequency of anestrous cows through Day 57 post partum was not different between the 2 groups. Cows that were anestrous had a lower body condition score throughout the postpartum period than the cyclic cows (2.7 vs <3.0; P < 0.01). Conception rate to first service, conception rate to all services, services per conception and days open were not different between the groups. Intensity of estrus detection was low for the study based on a low estrus detection rate (42%) and a low palpation pregnancy rate index (below 68%) throughout the study period. Accuracy of estrus detection was low based on the distribution of normal interestrus intervals for the study (19 to 23 d; 13.5%). It is concluded that programmed reproductive treatments during the postpartum period are functionally effective relative to altered ovarian activity; however, potential advantages to such a system are not evident without good estrus detection practices in a large commercial herd.  相似文献

The aim of this study was to evaluate the effect of presynchronization with or without the detection of estrus on first service pregnancy per artificial insemination (P/AI) and on Ovsynch outcome in lactating dairy cows. A total of 511 cows were divided randomly but unevenly into 3 treatment groups at 44 to 50 days in milk (DIM). Ovsynch was started at the same time (69 to 75 DIM) in all three groups. Cows in the Ovsynch group (CON, N = 126) received no presynchronization before Ovsynch, and all cows were bred by timed AI (TAI). Cows in the presynchronization with estrus detection (PED) and the presynchronization with only TAI (PTAI) groups received two doses of prostaglandin F (PGF) 14 days apart, starting at 44 to 50 DIM. Ovsynch was initiated 11 days after the second PGF treatment. Cows in the PED group (N = 267) received AI if estrus was detected after either PGF injection. Cows that were not determined to be in estrus after PGF injection received Ovsynch and TAI. Cows in the PTAI group (N = 118) were not inseminated to estrus, with all cows receiving TAI after Ovsynch. The ovulatory response to the first GnRH injection administered as part of Ovsynch differed (P = 0.002) among treatment groups (83.1% in PTAI, 72.6% in PED, and 62.7% in CON). However, the ovulatory response to the second injection of GnRH during Ovsynch did not differ among treatment groups. Of the 267 PED cows, a total of 132 (49.4%) exhibited estrus and were inseminated. The P/AI at the 31-day pregnancy diagnosis was similar between the cows in the PED group with AI after estrus detection (37.9%; 50/132) and those bred with TAI (34.1%; 46/135). The P/AI in the CON group (46.8%; 59/126) was greater (P < 0.05) than that in the PED group (36.0%; 96/267). In addition, the P/AI in the CON group was greater (P = 0.04) than that in the PED cows receiving TAI (34.1%; 46/135) but less than that in the PED cows bred to estrus (37.9%; 50/132) (P = 0.16). At the 31-day pregnancy diagnosis, the cows in the PTAI group had greater P/AI (55.9%; 66/118) than both those in the PED group (P < 0.01; either estrus or TAI) and those in the CON group (P = 0.08). Thus, presynchronization with PGF (PTAI) increased the ovulatory response to Ovsynch and improved P/AI in dairy cows. Interestingly, the breeding of cows to estrus during presynchronization reduced fertility to the TAI and overall fertility, including cows bred to estrus and TAI. These results indicate that maximal fertility is obtained when all cows receive TAI after the presynchronization protocol.  相似文献

Fixed-time AI (TAI) after GnRH-PGF(2alpha)-GnRH treatment is a method to achieve pregnancies in dairy herds without estrous detection. However, cows that fail to respond to the initial GnRH may have compromised TAI conception rates due to asynchronous ovarian response. This study documented the percentage of GnRH-treated Holstein cows (n=345) in two herds that displayed estrus at an inopportune time for optimum TAI conception rate (< or =48h post-PGF(2alpha); premature estrus (PE)) and compared conception rates of two TAI protocols in cows that did not display PE. At biweekly herd health exams, cows diagnosed as not pregnant to a previous AI and cows >80 days postpartum with no AI were treated with 100 microg GnRH (day -7) and 25mg PGF(2alpha) (day 0). Cows detected in PE by twice-daily visual observation from day -7 to day 2 were bred by AI 8-12h later. Cows not detected in PE were randomly assigned by parity, body condition score, and postpartum interval to receive either: (1) 100microg GnRH at 48h after PGF(2alpha) and TAI 16 to 18h later (Ovsynch); or (2) TAI at 72h post-PGF(2alpha) and a concurrent 100 microg GnRH injection to those cows not detected in estrus between 48 and 72h post-PGF(2alpha) (modified Ovsynch (MOV)). All hormone injections were im. Twenty percent (68/345) of the cows were detected in estrus before 48 after PGF(2alpha), of which 5% (17/345) were detected in estrus before PGF(2alpha) (< or =day 0). Herd influenced the percentage of cows in the PE group (herd A versus herd B; 25% versus 14%; P<0.05). Conception rates were not affected by treatment (PE versus Ovsynch versus MOV; 32% (21/65) versus 30% (37/125) versus 32% (47/145); P>0.10). However, within MOV-treated cows, conception rates were greater (P<0.05) in cows detected in estrus (46% (23/50)) compared with cows not detected in estrus (25% (24/95)). In conclusion, 20% of GnRH-treated cows displayed PE and necessitates estrous detection during this period if maximal pregnancy rates are to be achieved. Although additional estrous detection is required compared to Ovsynch, reduced cow handling and hormone usage, efficient use of expensive semen through greater conception rates in cows detected in estrus, and comparable TAI conception rates, suggests the MOV protocol may be a cost effective alternative to Ovsynch in many dairy herd reproductive management programs.  相似文献

Single injection of estrogen and progesterone before prostaglandin (steroid priming) was used to shorten the prostaglandin-based estrus synchronization program. Sixty-five cyclic Sistani cattle, with parity ranging from 1 to 4 and postpartum period of >80 days were selected at unknown stages of the estrous cycle and assigned to 2 groups according to their age, weight and parity. Females in the control group (n=33; 58.4 +/- 4.3 months; 277 +/- 8 kg LW) received two consecutive injections of prostaglandin F2alpha analogue (500 microg; Cloprostenol, PG) 14 days apart (Day 0 = First PG injection). On Day 7, treated females (n=32; 60 +/- 4.8 months; 292 +/- 9 kg LW) were given an intramuscular injection of 100 mg progesterone and 2 mg estradiol benzoate followed by prostaglandin 7 days later, concurrent with the second PG injection of the control group. Estrus detection was carried out every 6 hours for 7 days, commencing from 24 hours after the last PG injection. Females that allowed to be mounted were identified (standing estrus) and inseminated with frozen semen 12 hours later. Pregnancy was diagnosed on Day 50 after AI through palpation per rectum. Data were analyzed using Chi-squared and t-test. The tightness of estrus synchrony (%), the interval from the end of treatment to estrus (h) and conception rates (%) were similar (P > 0.05) between control (69.6%, 77.7 +/- 5.96 h and 56.5%) and treatment (68.2%, 82.6 +/- 7.64 h and 54.5%) groups. In conclusion, steroid priming is an efficient way to shorten the prostaglandin-based estrus synchronization program from 14 to 7 days without compromising estrous response and fertility.  相似文献

In Argentina, most dairy cows with endometritis are treated with prostaglandin (PGF(2alpha) or its analogs) and insemination is withheld until there are no signs of endometritis. The objective of the present study was to evaluate if this method of managing endometritis enhances reproductive performance. Three experiments were conducted over 4 years in a large farm in the west of Buenos Aires province. In Experiment 1, half of the cows diagnosed with endometritis (>1.5-fold difference in diameter of uterine horns, as determined by rectal palpation) received standard endometritis management (treatment with tiaprost, a PGF(2alpha) analog, rectal palpation every 20 days, and withholding of AI until endometritis apparently resolved) and the other half was untreated, with AI at the first estrus after the voluntary waiting period. Untreated cows were inseminated and conceived 20 days earlier than treated cows, and the pregnancy rate by Day 90 postpartum was higher in the untreated group. In Experiment 2, cows with endometritis were divided into four groups according to the severity of symptoms; within each group, cows were allocated to treatment or control, as in Experiment 1. Although first service conception rate decreased as endometritis severity increased, reproductive performance in treated versus control cows was similar to that of Experiment 1 (with no interaction due to degree of endometritis). Re-evaluation of the treated cow (to confirm uterine "normality") may have been responsible for the delay in conception in both experiments. The objective of Experiment 3 was to determine the effects of tiaprost treatment on clinically normal postpartum cows (no evidence of endometritis). Tiaprost treatment reduced the interval from calving to conception in multiparous cows, but it delayed conception and reduced the conception rate in primiparous cows. In conclusion, treatment with tiaprost impaired reproductive performance in primiparous cows (in the absence of endometritis). Furthermore, the standard treatment for endometritis (treatment with a prostaglandin analog and withholding insemination until clinical signs abated) impaired reproductive performance and increased costs.  相似文献

The objective of the study was to analyze the effect of a spontaneous estrus cycle after synchronization of estrus with prostaglandin F2alpha (PGF2alpha) in dairy cows on the degree of synchronization and reproductive performance. We assigned 557 Holstein cows to two treatment groups. In Group 1 estrus was synchronized by two treatments with 25 mg of Dinoprost-Trometamol in 14-day intervals. Cows were treated 27 to 33 days postpartum (dpp) and 41 to 47 dpp, respectively. Cows in Group 2 were treated with 25 mg of Dinoprost-Trometamol three times in 14-day intervals, starting at 34 to 40 dpp. The second and third injections were administered at 48 to 54 dpp and 62 to 68 dpp, respectively. All cows were inseminated on observed estrus after a voluntary waiting period of 65 days post partum. Thus cows in Group 1 were inseminated on spontaneous estrus and cows in Group 2 on induced estrus. Cows not inseminated at 80 days post partum were palpated per rectum and treated according to a predefined protocol. Herd reproductive performance measures did not differ significantly between groups. The proportion of cows with low serum progesterone levels was significantly higher 3 days after synchronization than 24 days after synchronization (97% vs 39%). The first-service conception rate was 34.8% in Group 1 and 30.7% in Group 2 (P > 0.05). Days open were 113.5 in Group 1 and 110.9 in Group 2 (P > 0.05). It is concluded that postponing artificial insemination for one spontaneous estrus cycle after synchronization decreased the degree of synchronization. This procedure, however, had no effect on herd reproductive performance compared to insemination on first observed estrus after synchronization.  相似文献

The objective of this study was to determine the reproductive performance of lactating dairy cows treated with GnRH and/or PGF2a for synchronization of estrus and ovulation. Between Days 43 and 57 post partum, a total of 374 dairy cows was divided into 4 groups. Cows in Group 1 (n = 62) were treated with 25 mg, i.m. PGF2a on Days 43 and 57; cows in Group 2 (n = 65) were not treated at this time; cows in Group 3 (n = 118) were treated with 100 ug, i.m. GnRH on Day 50, 25 mg, i.m. PGF2a on Day 57, 100 ug, i.m. GnRH on Day 59, and time-inseminated 16 h later; cows in Group 4 (n = 129) were treated with 25 mg, i.m. PGF2a once on Day 57. Cows in Groups 1 and 4 were inseminated at an induced estrus within 7 d after the last PGF2a treatment, and cows in Group 2 were inseminated at a noninduced estrus within a corresponding period of time. Conception rate, estrus detection rate and pregnancy rate were analyzed using logistic regression, and controlled for lactation number, body condition score and time of year. Days from calving to conception were analyzed using the GLM procedures of SAS, and the model included group, body condition score, lactation number, time of year, and their interactions. Cows in Group 3 had a significantly higher pregnancy rate than cows in Groups 1, 2 and 4. Orthogonal contrasts of mean days from calving to conception showed that cows in Group 3 had significantly (P < 0.01) less days from calving to conception than cows in Group 1 and Group 4. There was a significant effect of time of year on pregnancy rate and days from calving to conception, but there was no interaction between time of year and these reproductive characteristics. There was no effect of body condition score and lactation number on the reproductive characteristics evaluated. From the results of this study, it was concluded that better reproductive performance was observed in cows inseminated at a synchronized ovulation than in those inseminated at a synchronized estrous period.  相似文献

The objective was to evaluate if uterine sampling by low-volume uterine lavage, done shortly before the end of the voluntary waiting period, affected reproductive performance, milk production, or culling. Approximately 20 cows (40 and 60 d postpartum) were sampled in each of 38 herds (total, 752 cows), whereas all remaining cows concurrently between 40 and 60 d were a non-sampled reference cohort (n = 2,252 cows). There was an interaction between parity and sampling for first-service conception, calving to conception interval, and milk production. When stratified by parity, there was a tendency for impaired reproductive performance in sampled primiparous cows compared with non-sampled primiparous cows (Odds Ratio for pregnancy to first-service = 0.78 (95% C.I. 0.58-1.04). The Hazard Ratio for pregnancy in sampled primiparous cows was 0.89 (95% C.I. 0.77-1.04) compared to non-sampled primiparous cows. Sampling did not affect first-service conception rate (Odds Ratio for pregnancy Odds ratio (OR) for pregnancy = 1.03; 95% C.I. 0.80-1.33) or calving-to conception interval in multiparous cows (Hazard Ratio = 1.04; 95% C.I. 0.91-1.18). Sampling did not affect culling risk (Hazard Ratio HR = 0.92; 95% C.I. 0.77-1.11) after accounting for covariates. After stratification by parity, milk production was not affected by sampling except in fourth- and fifth-parity cows where sampled cows produced more milk than non-sampled cows after controlling for first test-day milk production and days-postpartum at first test-day. In conclusion, sampling by low-volume uterine lavage did not have significant detrimental effects on reproduction, culling, or milk production. However, there was a tendency for lower first-service conception in sampled primiparous cows, but the procedure appeared to be benign in multiparous cows.  相似文献

Effect of stage of the estrous cycle at the time of prostaglandin F(2alpha) (PGF(2alpha)) injection on subsequent reproductive events in beef females was studied in four trials involving 194 animals. Cycling animals were given two injections of 25 mg PGF(2alpha) 11 days apart or, in some cases, the interval was altered to allow the second injection to fall on a specific day of the cycle. Day of estrous cycle at time of the second injection was determined by estrous detection. Interval from the second PGF(2alpha) injection to the onset of estrus (interval to estrus) was shorter (P<.01) in heifers than in cows. Both cows and heifers injected on days 5 to 9 (early cycle) had a shorter (P<.01) interval to estrus (estrus = day 0) than did those injected on days 10 to 15 (late cycle). Conception rate was lower (P<.05) for early-cycle heifers than for late-cycle heifers inseminated by appointment at 80 hours. There was no significant difference in conception rate of early-or late-cycle heifers or cows inseminated according to estrous detection or early- or late-cycle cows inseminated at 80 hours. Progesterone concentrations in blood samples collected in heifers at 4-hour intervals after the second PGF(2alpha) injection on either day 7 or day 14 declined linearly (P<.05) through 36 hours. Day of the estrous cycle at PGF(2alpha) injection had no effect on rate of progesterone decline, even though heifers injected on day 7 had a shorter (P<.05) interval to estrus. All animals whose cycle length was not affected by the second PGF(2alpha) injection were treated on days 5 through 8 of the cycle, indicating that PGF(2alpha) was less effective in regressing the corpus luteum between days 4 and 9 of the cycle than later in the cycle.  相似文献

The objective of this study was to evaluate the effects of early postpartum PGF two alpha treatment on reproductive performance in dairy cows with calving and puerperal traits. A total of 363 Holstein cows (128 primiparous and 235 multiparous) were selected based on the presence of at least one of calving and puerperal traits (dystocia, retained placenta, twin, abortion, and postpartum uterine infections) and were assigned to two groups (treatment and control) irrespective of presence or absence of luteal tissue. Cows in the treatment group were treated twice with 25 mg dinoprost 8 h apart on day 20 postpartum, and for the control group saline placebo was administered. As it was speculated that the timing of a second dose would mimic the release of endogenous PGF2α from the uterus, our hypothesis was that two doses of PGF2α 8 h apart may increase the duration of elevated plasma prostaglandin F2α metabolite concentration in these cows. Recorded reproductive variables included days to first estrus, days to first AI, first service conception rate, pregnancy by 150 days in milk, service per conception, open days, and the percentage of repeat breeder animals. The data were analyzed using SPSS (Version 15) (IBM North America, New York, NY, USA) and Minitab (Version 14) (Minitab, State College, PA, USA). Although early postpartum PGF2α treatment had no effect on days to first estrus (36.7 days vs. 34.9 days, P = 0.056) and days to first AI (70.5 days vs. 72.2 days, P = 0.537), it increased first service conception rate (47.1% vs. 27.6%, P < 0.001); and this was more remarkable in primiparous cows (64.7% vs. 25%, P < 0.001). PGF2α treatment reduced the mean service per conception (1.92 vs. 2.72, P < 0.001) and the mean open days (112 days vs. 144 days, P < 0.001), and increased pregnancy by 150 days in milk (DIM) (80% vs. 66%, P = 0.004). The prevalence of repeat breeder syndrome in cows with calving and puerperal traits was reduced by PGF2α treatment (10% vs. 29.8%, P < 0.001). In conclusion, treatment of cows with calving and puerperal traits twice with a luteolytic dose of PGF2α 8 h apart on Day 20 postpartum improved reproductive performance and reduced the prevalence of repeat breeder syndrome.  相似文献

Lactating Holstein cows (n=288) were grouped as pairs at parturition and randomly assigned to two treatments (control, C vs intervenient treatment, T). The reproductive management of the Group C cows (n=130) consisted of the intramuscular administration of 500 microg PGF2alpha analogue (PG) on Days 28 and 63 postpartum and breeding on the basis of estrus signs with the a.m.-p.m. rule after Day 63. Cows that were not bred by 77 d postpartum received another injection of PG and were bred at estrus or 84 h after PG treatment. Pregnancy diagnoses were perfomed by palpation of the uterus per rectum 42 to 48 d after AI. Cows in the T group (n=139) received intramuscular injections of 100 microg GnRH 14 d and PG 28 d after calving. On Day 56 postpartum, cows were given a second dose of GnRH followed by PG on Day 63 postpartum and a third GnRH injection 48 h after PG (OvSynch). Cows were inseminated at a fixed time (22+/-1 h) after GnRH. Five days after the fixed-time insemination cows were given 1500 IU hCG i.m.. Group C and T cows that returned to service or were diagnosed as non-pregnant continued to receive PG at intervals of 14 d with breeding at estrus or 84 h after the second PGF2alpha dose. A sustained increase in milk progesterone concentration was observed in 59.0% of T cows after GnRH administration on Day 14. A similar rise in milk progesterone concentrations was observed in 53.8% of C cows. The PG on Day 28 induced luteolysis more in Group T cows (53.2%) than in Group C cows (36.9%). The PG on Day 63 reduced milk progesterone concentrations to basal levels in 50.7% of T and 49.2% of Group C animals. The first service pregnancy rates (T, 40.3% vs C, 36.2%) and the overall pregnancy rates (all services, T, 83.5% vs C, 86.9%) were not different between the two groups. The two treatments did not differ in the interval from first service to pregnancy, calving to pregnancy or in calving interval, number of services per pregnancy or culling rates.  相似文献

Ineffective estrus detection is the foremost limiting factor in the fertility of farmed cattle worldwide. Failure to detect estrus or erroneous diagnosis of estrus results in great economic losses in Korea each year. This study was carried out in order to comprehensively describe the estrus behaviors and conception rates of different estrus synchronization protocols applied to 40 cycling native Korean cattle (Hanwoo). The cows were grouped into four (n = 10) and treated with the following protocols: (1) Day -15: controlled intravaginal drug-releasing device (CIDR) for 12 days; Day -5: prostaglandin F (PGF), (2) ovulation synchronization (OVS): Day -15: GnRH; Day -6: PGF; Day -4: GnRH, (3) Day -15: progesterone-releasing intravaginal device for 12 days; Day -5: PGF; and (4) Day -15: PGF; Day -4: PGF. Artificial insemination was performed 12 hours after the detection of estrus using frozen-thawed semen. Estrus signs were compared using a charge-coupled device camera (CCDC) and a control method (direct visual observation). The pregnancy of the cows was determined by transrectal ultrasonography at Days 25 to 30 postinsemination. The results indicated that the day of estrus return was significantly earlier using the CCDC method compared with direct visualization (P < 0.05). Mounting of other cows was the most predominant sign of estrus among the flock (P < 0.05), as analyzed using the CCDC. In the OVS group, a lower rate of mounting was observed than in the other three groups. Moreover, significantly fewer estrus behaviors were noticed in the OVS protocol group (P < 0.05). Both first service conception and overall conception rates were significantly higher (P < 0.05) in the CIDR and OVS treatment groups. In conclusion, the CIDR and OVS protocols appear to be the best practice for the synchronization of estrus for reproductive competence through the CCDC in Hanwoo cows. However, CIDR has a practical advantage over OVS with respect to estrus detection.  相似文献
