A survey on the occurrence of aflatoxin M1 was carried out in the province of Bari (Italy) on a total of 106 samples of raw milk from local farms, commercially heattreated milk, and dried milk. About 72% of the samples were contaminated with aflatoxin M1 within the concentration range 4–280ng/Kg. The incidence of aflatoxin M1 was much higher in commercial milk (91%) than in farm milk (26%), and all the dried milk samples contained aflatoxin Ml (10 to 280ng per Kg of powder milk).  相似文献

Contamination of milk and dairy products with aflatoxin M1 (AFM1) presents a risk for human health. The aim of this study was to investigate the presence of AFM1 in pasteurized milk samples in Fariman, located in the province of Khorasan Razavi, Iran, by enzyme-linked immunosorbent assay (ELISA). Forty-five samples of pasteurized milk from different supermarkets were collected during 3 months in summer (July to September, 2012). AFM1 contamination was detected in all of milk samples. The mean concentration of aflatoxin M1 was 27.2 ng/l. The range of AFM1 content was 8.8–64 ng/l. Thirteen (28.8 %) of the samples had AFM1 levels exceeding the maximum levels (50 ng/l) accepted by the European Union. Due to the fact that milk is used by all the age groups including infants and children in Fariman city, it is necessary to minimize the health risk from AFM1 contamination in milk. For this reason, the level of its precursor, aflatoxin B1 (AFB1), in dairy feeds must be reduced, requiring constant aflatoxin monitoring of relevant agricultural commodities.  相似文献

This study was performed to determine aflatoxin M1 (AFM1) in human breast milk samples collected in ?anl?urfa, located in Southeastern region of Turkey, and to investigate a possible correlation between AFM1 occurrence (frequency and levels) and sampling seasons. Human breast milk samples collected in December 2014 and in June 2015 from a total of 74 nursing women, both outpatient and inpatient volunteers in hospitals located in ?anl?urfa, Turkey, were analyzed using competitive enzyme-linked immunosorbent assay (ELISA) for the presence of AFM1. AFM1 was detected in 66 (89.2%) out of 74 samples at an average concentration of 19.0 ± 13.0 ng/l (min.-max., 9.6–80 ng/l). There was a statistically significant difference between December and June concerning AFM1 levels (p < 0.05). Further detailed studies will be needed to determine the main sources of aflatoxins in food, to establish protection strategies against maternal and infant exposure to these mycotoxins.  相似文献

Sequestering agents bind dietary aflatoxin B1 (AFB1) and reduce absorption from an animal's gastrointestinal tract. As a result, they protect an animal from the toxic effects of AFB1 and reduce transfer of the metabolite, aflatoxin M1 (AFM1), into milk. Three experiments, using late-lactation Holstein cows fed AFB1-contaminated feed, were conducted to evaluate several potential sequestering agents for their abilities to prevent or reduce the transmission of AFM1 into milk. Six agents previously tested in our laboratory for AFB1 binding in vitro were evaluated in these experiments. These were: SA-20, an activated carbon (AC-A); Astra-Ben-20, a sodium bentonite (AB-20); MTB-100, an esterified glucomannan (MTB-100); Red Crown, a calcium bentonite (RC); Flow Guard, a sodium bentonite (FG); and Mycrosorb, a sodium bentonite (MS). Five of the six sequestering agents significantly (P < 0.01) reduced AFM1 contamination of milk (AB-20, 61%; FG, 65%; MS, 50%; MTB-100, 59%; and RC, 31%); whereas, AC-A, activated carbon, had no effect on AFM1 transmission at 0.25% of feed. By the first milking (1 day after cows consumed contaminated feed), AFM1 appeared in milk, then reached maximum levels after three days, and was absent from milk within four days after AFB1 was removed from the feed. Sodium bentonites at 1.2% of feed showed good potential as AFB1 binders; MTB-100, a yeast cell wall product, was equally effective at 0.05% in feed. Potential AFB1 binding agents should be evaluated experimentally to demonstrate efficacy. Our data show that sequestering agents can reduce AFM1 in milk of cows fed AFB1-contaminated feed.  相似文献

BACKGROUND: The principal Aflatoxin B(1) (AFB(1)) hydroxylated metabolite excreted in milk is Aflatoxin M(1) (AFM(1)) classified in group 2B by the International Agency for Research on Cancer (IARC). Human exposure to AFM(1) is due to the consumption of contaminated dairy products and partly to endogenous production through AFB(1) liver metabolism. METHODS: Since no data are available on AFM(1) embryotoxicity, its lethal and teratogenic potential was investigated using the Frog Embryo Teratogenesis Assay-Xenopus (FETAX). Stage-8 blastulae were exposed to AFM(1) at 1, 4, 16, 64, and 256 microg/L concentrations until stage 47, free-swimming larva. RESULTS: A slight increase of mortality and malformed larva percents was found in AFM(1)-exposed groups but these differences were not statistically significant in comparison with the controls. CONCLUSIONS: Therefore, AFM(1) is a non-embryotoxic compound when evaluated with a FETAX model at concentrations under the conditions tested. However, AFM(1) merits further studies using mammals as experimental models to identify a possible risk during human pregnancy.  相似文献

IntroductionThe aim of this study was to investigate the fatty acid (FA) pattern in breast milk of obese mothers and their neonates' plasma compared to those of normal weight mothers.Patients and methodsThis was an observational study of 41 obese and 41 normal weight pregnant women. Twenty-nine obese women participating in a weight reduction program were investigated for comparison. FAs were analyzed in breast milk collected at 3 and 10 days and one and two months postnatally and in infant's plasma 3 days after birth.Results and conclusionsThe concentration of long-chain n-3 FA were lower and the ratio n-6/n-3 FA higher in neonates and in consecutive samples of breast milk of obese mothers compared to normal weight mothers. The obese mothers that participated in an intervention program with general dietary advice had FA concentrations approaching that of the normal-weight mothers. The study indicates importance of dietary advice in pregnancy.  相似文献

In liquid cultures growing and stationary phase cells ofFlavobacterium aurantiacum NRRL B-184 eliminated aflatoxin M1. Toxin concentrations of 15µg/ml and 37.5µg/ml interfered with bacterial growth, and at the higher level 4.4µg M1 was removed from the growth medium by a milligram (dry weight) of bacteria. Toxin was completely removed from the liquid medium by incubating 5 × 1010 resting cells per milliliter with 8µg/ml of aflatoxin M1 for 4 h. Attempted recovery of M1 from cells following incubation of the bacteria with the toxin demonstrated that the M1 was essentially nonextractable. Bacterial cells also removed aflatoxin M1 from toxin-contaminated milk.  相似文献

目的探讨大连地区分泌型和非分泌型母亲哺乳期间母乳菌群的主要差异。方法于大连市妇幼保健院纳入42名志愿者产妇,收集其产后第6天的母乳样本。提取母乳样本DNA,并对包含rs601338和rs1047781单核酸位点多态性(SNP)的片段用聚合酶链式反应(polymerase chain reaction,PCR)检测并加以测序,以检测受试母亲岩藻糖基转移酶2基因(FUT2)的类型。采用16S rRNA高通量测序法对不同母乳样本中微生物的多样性进行分析,并且对不同母乳样本中的微生物丰度及类型展开探讨。结果母亲分泌型分布情况分析:42名志愿者中,36名母亲是分泌型,6名母亲是非分泌型。母乳菌群多样性分析:分泌型与非分泌型组母亲母乳菌群比较,发现反映组内差异的Alpha多样性指数(包括Ace、Chao1以及Shannon等)在2组间差异无统计学意义;通过使用主坐标分析,发现分泌型组和非分泌型组的距离相对较远,母乳菌群组内菌群结构类似,组间差异性较显著,说明母乳中的蛋白核心岩藻糖基化水平可明显改变母乳中菌群结构。乳汁菌群物种构成分析:从门水平分析,变形菌门与厚壁菌门为主要优势菌;从属水平分析,2组母乳菌群中的双歧杆菌属丰度差异较显著。相关性分析:分泌型母乳的菌群中相关基因的表达更强,说明分泌型母乳对菌群基因的表达有促进作用。结论母乳菌群构成在一定程度上受到母体FUT2基因类型,即母亲分泌型和非分泌型的影响。  相似文献

Forty-seven samples of commercial ultra-high-temperature-treated milk from a dairy facility in the northwest part of Spain were analyzed for the presence of aflatoxin M1. A total of 14 samples (29.8%) were positive for aflatoxin M1 (4 in May, 3 in November, 3 in December, 1 in January, 1 in April, 1 in July, and 1 in August), 29 (61.7%) were negative, and 4 (8.5%) were doubtful, i.e., they showed trace quantities of aflatoxin M1. The range of aflatoxin M1 content was 0.02 to 0.1 ng/ml.  相似文献

Kuwaiti diet is exceptionally rich in fat, carbohydrates and proteins. In addition, subjects in Kuwait are exposed to extreme heat and sun light. Fatty acid profiles of human milk obtained from 19 full breast feeding Kuwaiti mothers were analyzed. Dietary patterns for individual mothers were determined by 24 h dietary recall and food frequency questionnaire. The fatty acid content of human milk was affected by the diet consumed by the lactating mother. The content of long chain polyunsaturated fatty acids (LCP) in human milk lipids did not correlate with their parent fatty acids like linoleic and alpha-linolenic acids. However the human milk LCP were related to the of content of LCP in the maternal diet. Mothers reporting a high fish consumption showed significant amounts of C22:6, omega 3 and C20:5, omega 3 fatty acids. As a general conclusion, breast milk produced by a well nourished mother is better suited to meet the lipid requirements of infants.  相似文献

Breast milk contributes towards optimal nutrition for infants. However, studies showed that it can also contain different toxins and heavy metals, which reduce its health benefits. The aim of this study is to determine the level of contaminants such as aflatoxin M1 (AFM1), Pb, Cd, As, and Hg in breast milk samples from Famagusta, Cyprus. Correlations between moldy food consumption, smoking habits of the mothers, and contaminant levels in breast milk were also investigated. Breast milk samples from 50 lactating mothers in rural and urban areas of Famagusta District were analyzed for AFM1 by ELISA. Eighty percent of them were found to be contaminated with AFM1 with the mean measurement of 7.84?±?1.72 ng/l. Socio-demographic status, moldy food consumption habits, and smoking status do not have any effect on the AFM1 levels observed in breast milk. Heavy metal levels in breast milk were examined by inductively coupled plasma mass spectrometry, and the mean measurements were1.19?±?1.53 ppm for Pb, 0.73?±?0.58 ppm for As, 0?±?0.20 ppm for Hg, and 0.45?±?0.23 ppm for Cd. This study indicates that the levels of these contaminants in breast milk samples obtained in Famagusta District are well within the acceptable levels. However, the presence of AFM1 and heavy metals still may pose risks for infant health.  相似文献

To achieve optimal development of a newborn, breastfeeding is extensively recommended, but little is known about the role of non-nutritive bioactive milk components. We aimed to characterize the fatty acid esters of hydroxy fatty acids (FAHFAs), namely palmitic acid hydroxystearic acids (PAHSAs)—endogenous lipids with anti-inflammatory and anti-diabetic properties, in human breast milk.Breast milk samples from 30 lean (BMI = 19–23) and 23 obese (BMI > 30) women were collected 72 h postpartum. Adipose tissue and milk samples were harvested from C57BL/6J mice. FAHFA lipid profiles were measured using reverse phase and chiral liquid chromatography-mass spectrometry method.PAHSA regioisomers as well as other FAHFAs were present in both human and murine milk. Unexpectedly, the levels of 5-PAHSA were higher relative to other regioisomers. The separation of both regioisomers and enantiomers of PAHSAs revealed that both R- and S-enantiomers were present in the biological samples, and that the majority of the 5-PAHSA signal is of R configuration. Total PAHSA levels were positively associated with weight gain during pregnancy, and 5-PAHSA as well as total PAHSA levels were significantly lower in the milk of the obese compared to the lean mothers.Our results document for the first time the presence of lipid mediators from the FAHFA family in breast milk, while giving an insight into the stereochemistry of PAHSAs. They also indicate the negative effect of obesity on 5-PAHSA levels. Future studies will be needed to explore the role and mechanism of action of FAHFAs in breast milk.  相似文献

During an initial survey, using thin layer chromatography, 10 of 64 samples of mothers’ breast milk, collected from donors at the Corniche Maternity Hospital and the Al-Nehyan Clinic for Maternity and Childhood, were found to contain Aflatoxin M1 at concentrations ranging from 0.3 to 1.3 ng mL?1. A second survey using HPLC showed Aflatoxin M1 at concentrations ranging from 7 to 23 pg mL?1 in all of the 15 samples collected. 6 of 20 samples of camel milk collected from several sources in Abu Dhabi were also found to contain Aflatoxin M1 at levels ranging from 0.25 to 0.8 ng mL?1.  相似文献

Aflatoxin G1 (AFG1) was transformed into aflatoxin B3 (AFB3) by the fungiRhodotorula sp,Sporobolomyces sp,Rhizopus oryzae NRRL395,Pythium ultimum, Aspergillus terreus, A clavatus and Penicillium frequentans grown in a medium containing AFG1, Difco potato dextrose broth, yeast extract, and peptone both in liquid shaken cultures and in solid static cultures at 25°C in the dark. A maximum rate of transformation of 10 % was obtained after 2 to 3 weeks of incubation. The transformation was correlated with an increase in the pH of the media from 5.7 –5.9 to 8.3 – 8.8.Saccharomyces cerevisiae also transformed AFG1 into AFB3, but at a slower rate; the pH of the media did not reach above 8.0 until 5 weeks after incubation. No transformation was observed whenA niger andP chrysogenum were tested; in both cases, no increase in pH was noticed. However, some transformation of AFG1 to AFB3 by both fungi was observed when the initial pH of the media was adjusted to 9.0. The rate of transformation increased to 15 – 20% in the static culture where the same medium was adsorbed onto vermiculite andRhizopus andAspergilli gave the highest increase in AFB3 yield.  相似文献
