This paper introduces a dose-response model for teratological quantal response data where the probability of response for an offspring from a female at a given dose varies with the litter size. The maximum likelihood estimators for the parameters of the model are given as the solution of a nonlinear iterative algorithm. Two methods of low-dose extrapolation are presented, one based on the litter size distribution and the other a conservative method. The resulting procedures are then applied to a teratological data set from the literature. 相似文献

Prenatal inhalation toxicity of xylene (industrial mixture of isomers) was studied in experiments of white Wistar rats exposed daily (6 hours a day, 5 days in a week) to concentrations of 10, 50 (MAC for xylene in the air of work environment in Bulgaria) and 500 mg.m-3 throughout the period of gestation from the first to the 21st day. Both routine teratological indices and biochemical and physiological methods of observation were used to evaluate the integrity of the individual organs - liver, brain, lungs and myocardium of the generation in the postnatal period of development. In concentrations of 50 and 500 mg.m-3, xylene exhibits pronounced embryotoxic and teratogenic effects. Postimplantation embryonal mortality increases, the process of physical development of the fetus is delayed, the incidence of induced anomalies of internal organs (hydrocephalus, microphthalmia, intracerebral haematomas, haemorrhages in the liver) is enhanced, the processes of ossification of the sternum and the skull are impaired. In concentrations of 50 and 500 mg.m-3, xylene causes disturbances in postnatal development of F1 generation. The concentration of 50 mg.m-3 is the threshold of the embryotropic effect of the solvent. Measures for the protection of women at work are proposed to reduce industrial hazard of developing antenatal pathology in the newborn of workwomen in xylene works. 相似文献

Sensitivities of six avian species, black vulture (Coragyps atratus), American kestrel (Falco sparverius), Japanese quail (Coturnix japonica), domestic chicken (Gallus domesticus), eastern screech-owl (Otus asio), and European starling (Sturnus vulgaris), to acute poisoning by sodium cyanide (NaCN) were compared by single dose LD50's. Three species, domestic chickens, black vultures, and turkey vultures (Cathartes aura), were dosed with NaCN to determine cyanide residues in those that died and also in survivors, in addition to postmortem fate. Three flesh-eating species (black vulture, American kestrel, and eastern screech-owl; LD50's 4.0-8.6 mg/kg) were more sensitive to NaCN than three species (Japanese quail, domestic chicken, and European starling; LD50's 9.4-21 mg/kg) that fed predominantly on plant material. Elevated concentrations of cyanide were found in the blood of birds that died of cyanide poisoning; however, concentrations in birds that died overlapped those in survivors. Blood was superior to liver as the tissue of choice for detecting cyanide exposure. No gross pathological changes related to dosing were observed at necropsy. 相似文献

A model for cyanide species uptake by willow (Salix eriocephala L. var. Michaux) was developed to interpret data from hydroponic experiments quantitatively. While the potential for cyanide phytoremediation has been demonstrated modeling will aid in determining plant processes that contribute to cyanide transport and metabolism in willow and will target specific physiological parameters for field-scale phytoremediation design and optimization. The objective of the model development was to gain insight into the relative role of different processes with respect to dissolved free and iron-complexed cyanide transport and assimilation in plants and to determine rates at which these processes occur within the willow plant under the experimental conditions. A physiologically-based model describing plant uptake, transport, and metabolism of cyanide species was developed to reflect the processes that influence the movement of cyanide into and throughout the plant. Plant compartmentalization (root, stem, and leaf) corresponded to the level of detail in the data collected via hydroponic experiments. Inclusion of more detailed intra- and intercellular processes would create a model inconsistent with the macroscale nature of the data. Mass balances around each compartment were developed via kinetic representations for the mass transfer processes and were combined to form a model describing the fate of cyanide species within plant-water systems. 相似文献

In liver fractions from male Sprague-Dawley rats, the metabolism of methacrylonitrile (MeAN) to cyanide (CN-) was localized in microsomal fraction and required reduced nicotinamide adenine dinucleotide phosphate (NADPH) and oxygen for maximal activity. The biotransformation of MeAN to CN- was characterized with respect to time, microsomal protein concentration, pH, and temperature. Metabolism of MeAN was increased in microsomes obtained from phenobarbital-treated rats (310% of control) and decreased with CoCl2 and SKF 525 A treatments (55% and 61%, respectively). Addition of the epoxide hydratase inhibitor, 1,1,1-trichloropropane 2,3-oxide, decreased the formation of CN- from MeAN. Addition of glutathione, cysteine, D-penicillamine, and 2-mercaptoethanol enhanced the released of CN- from MeAN. These findings indicate that MeAN is metabolized to CN- via a cytochrome P-450-dependent mixed-function oxidase system. 相似文献

Weekly weighings of the laboratory rats are required to determine the correct dosage for mixing in the food. This creates problems in that the food mixing must be done immediately after the weighings and staff are often heavily taxed to perform the task. A discounted least squares growth prediction model allows for prediction of weights a week ahead of time, obviating the necessity for instantaneously processing the weight data. When dosages were prepared based on these predictions, for 10 treatment combinations 100% of the doses proved to be within 8-0% of the required dosage; 98-4% were within 5% of the required dosage; 78-7% were within 2% of the required dosage; and 51-6% were within 1% of the required dosage. The quadratic weight prediction model can also be incorporated into a model for predicting food consumption. 相似文献

An original, simple, and inexpensive diluter for flow-through toxicity studies is described. The diluter is run only by water gravity and has no moving parts. The main units of the device are made of very cheap disposable components and may be mounted without qualified technical assistance. It has neither metal nor any other kind of chemically active parts contacting experimental media. The diluter may be easily modified to study toxicants independently or in mixtures, in fact, as many toxicants as are necessary. When properly calibrated, it does not require further adjustments even during long-term experiments. The main original feature of the diluter described here is its dosing system, which is based on a vacuum air lock. This key system is very simple, reliable, includes neither mechanical nor electrical devices, and ensures adequate accuracy in dosing. 相似文献

Zamzam water is well documented for plenty of medicinal value for curing illness. In the present study, the effects of perinatal consumption of Zamzam and normal drinking water by the pregnant mice on their offspring’s physical parameters, early sensory motor reflexes, locomotor activities, acetylcholinesterase (AChE) activity in the homogenize brain tissue and blood parameters were compared. To achieve that; Zamzam water was given to female Swiss-Webster strain mice as the only source of drinking fluid and the control animals were administered plain tap water. Treatment started from the first day of pregnancy and continued until the postnatal day fifteen of delivery. All offspring were subjected to various tests. The rate of body weight gain remained relatively unaffected until the second week of weaning period, however; in the last week the offspring exposed to Zamzam water gained significant body weight as compared to their control offspring. Furthermore, the opening of eyes and appearance of body hairs in Zamzam exposed pups remained unaffected as compared to the controls. The sensory motor reflexes in Zamzam exposed pups after birth and during the first two weeks of weaning period were significantly increased. Locomotor Activity Test performed in the male and female offspring after weaning period showed a significant decrease in the male and increase in the female on most of the elements of this test due to Zamzam exposure. AChE activity in the homogenized brain tissue and blood parameters were unaffected as compared to the controls, the present Zamzam effects in the offspring are possibly via in utero action and/or via mother’s milk. 相似文献

The effect of lyophilised aqueous extract of Telfairia occidentalis (TO) on induced cyanide toxicity in rats was investigated. Twenty 3-week old male wistar albino rats were randomly distributed into one control and three treatment groups of five rats each: control group (group1), group treated with 3mg/kg body wt of cyanide only (group2), group treated with 3mg/kg body wt. each of cyanide and extract (group3), and a group treated with 3mg/kg Body wt of extract only (group4) were used for the investigation. Cyanide toxicity reduced both food and water intake, while the food intake was improved in group3, this effect of the extract on food was not observed on water intake. Cyanide reduced average body weight of rats significantly. The reduction effect of cyanide on body weight was countered by Telfairia occidentalis extract. The extract did not have an observable effect on rats' body weight. Ocular lesion was observed in 67% of rats in group2 . This ocular effect of cyanide was mitigated significantly by Telfairia occidentalis as only 17% of the rats in group3 had ocular lesion. Cyanide toxicity produced nasal discharge in 39% of the rat population in group2 while there was a partial but considerable reduction (21%) in the severity of nasal discharge in group 3. There was no significant difference in the organ/body wt.ratio between the treatments and the control groups for all the organs examined in the study. Biochemical analysis of liver enzymes showed that cyanide (group2) damaged the liver as there was significantly elevated presence of Aspartate aminotransferase (AST) and Alanine aminotransferase (ALP) above those of the control group. The damaging effect of cyanide on the liver was ameliorated by Telfairia occidentalis considerably.Histopathological effect of cyanide toxicity on the organs examined included multifocal degeneration and necrosis of the liver, mild kidney congestion and congestion of the brain. These effects were moderated mildly by Telfairia occidentalis. Group 4, treated with the vegetable alone had none of the observed histopathology in the organs examined. We concluded that lyophilised aqueous extracts of Telfairia occidentalis showed good potential as a safe antidote for cyanide poisoning when administered concomitantly or very shortly after ingestion of sub-lethal dose of cyanide. However, further bioassay guided fractionation and analytical studies are needed to identify the actual chemical compound or molecule in the vegetable responsible for or associated with the observed effects. 相似文献

Background
Rabies is a widespread disease in African domestic dogs and a serious public health problem in developing countries. Canine rabies became established in Africa during the 20th century, coinciding with ecologic changes that favored its emergence in canids. This paper reports the results of a cross-sectional study of dog ecology in the Antananarivo urban community in Madagascar. A questionnaire survey of 1541 households was conducted in Antananarivo from October 2007 to January 2008. The study addressed both owned and unowned dogs. Various aspects of dog ecology were determined, including size of dog population, relationship between dogs and humans, rabies vaccination.Results
Dog ownership was common, with 79.6 to 94.1% (mean 88.9%) of households in the six arrondissements owning dogs. The mean owned dog to person ratio was 1 dog per 4.5 persons and differed between arrondissements (administrative districts), with ratios of 1:6.0 in the first arrondissement, 1:3.2 persons in the 2nd, 1:4.8 in the 3rd, 1:5.2 in the 4th, 1:5.6 in the 5th and 1:4.4 in the 6th arrondissement. Overall, there were more male dogs (61.3%) and the male/female sex ratio was estimated to be 1.52; however, mature females were more likely than males to be unowned (OR: 1.93, CI 95%; 1.3919.
Electron spin echo envelope modulation studies are performed on human hemoglobin cyanide, hemoglobin nitroxide and hemoglobin nitroxide + inositol hexaphosphate at neutral pH. The modulation data are Fourier transformed and are analyzed in the frequency domain. The frequency components observed from hemoglobin cyanide indicate modulation from the coordinated nitrogen N-1 of the proximal imidazole (His-F8). In the case of hemoglobin nitroxide, the binding of inositol hexaphosphate causes the nitrogen N-3 of the proximal imidazole to be protonated in some of the subunits. From a comparison with other studies on these derivatives of hemoglobin, these subunits are identified as the alpha-subunits. 相似文献

Pre-exposure of rainbow trout, Salmo gairdneri to sublethal levels of arsenic or cyanide, resulted in significant changes in the incipient lethal levels of the respective toxicants. Fish gradually increased their tolerance to arsenic (as arsenite) by 47% during a three week preexposure to 0.22 of the incipient lethal level. Mean whole-body aresenic concentrations increased from 2 to 27 μg g−1 dry weight during the first week, then decreased to a stable level of 15 after two and three weeks. Fish decreased their tolerance to cyanide (as HCN) by 32 and 15% after one and two weeks of pre-exposure to 0–35 of the incipient lethal level. This initial sensitization disappeared after three weeks. Renal tubular parenchymatous oedema was observed after two and three weeks of cyanide pre-exposure. Neither arsenic nor cyanide affected growth at these levels of pre-exposure. 相似文献
