Thirty-day-old plants of mustard (Brassica juncea L.) were sprayed with 10−10, 10−8, or 10−6 M aqueous solution of 28-homobrassinolide (HBR). The HBR-treated plants were healthier than those treated with water and yielded more. Maximum increase over control was found in 60-d-old, 10−8 M-HBR-treated plants in fresh and dry mass per plant, carbonic anhydrase (CA, E.C. 4.2.1.1) activity, and net photosynthetic rate (P N), at harvest in number of pods per plant and seed yield per plant (the respective values were 25, 30, 34, 69, 24, and 29 %). A further increase in the concentration of HBR (10−6 M) did not make any additional impact on the growth and yield. Increased CA activity and P N were correlated with growth and seed yield. The leaves of 30-d-old plants of Brassica juncea Czern & Coss cv. Varuna were sprayed with 10–6 M aqueous solutions of indole-3-yl-acetic acid (IAA), gibberellic acid (GA3), kinetin (KIN), and abscisic acid (ABA) or 10–8 M of 28-homobrassinolide (HBR). All the phytohormones, except ABA, improved the vegetative growth and seed yield at harvest, compared with those sprayed with deionised water (control). HBR was most prominent in its effect, generating 32, 30, 36, 70, 25, and 29 % higher values for dry mass, chlorophyll content, carbonic anhydrase (E.C. 4.2.1.1) activity, and net photosynthetic rate in 60-d-old plants, pods per plant, and seed yield at harvest, over the control, respectively. A pot experiment was conducted to find out whether the foliar spray of salicylic acid (SA) could successfully ameliorate the adverse effects of salinity stress on periwinkle. Thirty-day-old plants were supplied with Control; 0 mM NaCl + 10−5 M SA (T1); 50 mM NaCl + 0 SA (T2); 100 mM NaCl + 0 SA (T3); 150 mM NaCl + 0 SA (T4); 50 mM NaCl + 10−5 M SA (T5); 100 mM NaCl + 10−5 M SA (T6); 150 mM NaCl + 10−5 M SA (T7). The plants were sampled 90 days after sowing to assess the effect of SA on stressed and unstressed plants. Salt stress significantly reduced the growth attributes including plant height, leaf-area index, shoot and root fresh weights, shoot and root dry weights. Increasing NaCl concentrations led to a gradual decrease in photosynthetic parameters and activities of nitrate reductase and carbonic anhydrase. Ascorbic acid, total alkaloids and antioxidants enzymes superoxide dismutase, catalase and peroxidase also declined in NaCl-treated plants. The plants, undergoing NaCl stress, exhibited a significant increase in electrolyte leakage and proline content. Foliar application of SA (10−5 M) reduced the damaging effect of salinity on plant growth and accelerated the restoration of growth processes. It not only improved the growth parameters but also reversed the effects of salinity. Total alkaloid content was improved by SA application both in unstressed and stressed plants. The highest level of total alkaloid content recorded in leaves of SA-treated stressed plants was 11.1%. Foliar spray of SA overcame the adverse effect of salinity by improving the content of vincristine (14.0%) and vinblastine (14.6%) in plants treated with 100 M NaCl.  相似文献

The leaves of 29-d-old plants of Brassica juncea Czern & Coss cv. Varuna were sprayed with 10–6 or 10–8 M aqueous solutions of indole-3-yl-acetic acid (IAA) or its substituted derivatives 4-Cl-IAA, 7-Cl-IAA, and 4,7-Cl2-IAA. All the auxins improved the vegetative growth and seed yield at harvest compared with those sprayed with de-ionised water (control). 4-Cl-IAA was most prominent in its effect, generating 21.6, 39.7, 61.0, 35.0, 65.5, and 56.2% higher values for dry mass, leaf chlorophyll content, carbonic anhydrase (E.C. 4.2.1.1) and nitrate reductase (E.C. 1.6.6.1) activities, net photosynthetic rate, and carboxylation efficiency, respectively, in 60-d-old plants. It also enhanced the seed yield by 31.1% over the control. The order of response of the plants to various auxins was 4-Cl IAA 7-Cl IAA > 4,7-Cl2 IAA = IAA > control.  相似文献

In the present study, it was aimed to investigate the influence of exogenous mammalian sex hormones (MSH) (progesterone, β-estradiol and androsterone) on the morphological (root and shoot growth) and biochemical parameters (protein and sugar content, antioxidant enzyme activities, and lipid peroxidation and H2O2 levels) of chickpea (Cicer arietinum L.) plants growing under control conditions. The solutions of hormones prepared at different concentrations (10−4, 10−6, 10−9, 10−12 and 10−15 M) were sprayed once on the leaves of 7-day plants. The plants were harvested on 18 days after the hormone treatment. Although all of the hormones at the tested concentrations significantly increased plant growth, soluble protein and sugar contents, and antioxidant enzyme activities [superoxide dismutase (SOD), peroxidase (POX) and catalase (CAT)], they decreased H2O2 content and lipid peroxidation level when compared with control plants. The activities of SOD, POX and CAT reached to the highest levels at 10−6 M for progesterone, and 10−9 M for β-estradiol and androsterone, which maximum growth, protein and sugar contents were determined. The same concentrations also resulted in the lowest levels for H2O2 content and lipid peroxidation. It can be interpreted that the MSH improve plant growth and development by affecting some biochemical parameters including antioxidative system.  相似文献

The effects of salicylic acid (SA) and jasmonic acid (JA) on plant defense responses were studied with aseptic potato (Solanum tuberosum L.) plantlets infected with Phytophthora infestans (Mont.) de Bary. Plant treatment with 10−6 M SA or 10−7 M JA induced plant resistance; the mixture of these acids was most efficient. After treatment with these compounds, phenolic compounds were accumulated and peroxidase was activated in the sites of pathogen localization, and this might be the reason of resistance enhancement. In addition, more H2O2 was accumulated in infected plants treated with JA or its mixture with SA but not in plants treated with SA alone. It might occur because of observed inhibition of catalase and activation of isoperoxidase with the isoelectric point (pI) of ∼9.3, which manifests an affinity for the pathogen cell wall. The data obtained allow us to recommend the application of these compounds for potato plant protection against late blight.  相似文献

This study was conducted to investigate the influence of salicylic acid (SA) on the growth and changes of nucleic acids, protein, photosynthetic pigments, sugar content and photosynthesis levels in the green alga Chlorella vulgaris Beijerinck (Chlorophyceae). The most significant changes in the content of nucleic acids and proteins was observed at the concentration 10−4 M SA between 8 and 12 day of cultivation. This concentration of SA increased the number of cells (about 40 %) and content of proteins (about 60 %) and its secretion to the medium. The slight stimulation of protein secretion occurred on the 12th day of cultivation at concentration 10−4 M, while in the range of 10−5 M to 10−6 M the protein secretion was inhibited. SA also stimulated the content of nucleic acids, especially RNA by 20–60 %, compared with the control. The most stimulating influence upon the contents of chlorophylls a and b (50–70 %), total carotenoids (25–57 %), sugar (27–41 %) and intensity of net photosynthesis (18–33 %) was found at 10−4 M of SA. At the concentration of 10−6 M SA the slight inhibition of growth and biochemical activity of the algae was recorded at the first days of cultivation.  相似文献

In three-day-old maize (Zea mays L.) seedlings, we removed the endosperm, coleoptile with leaflets, and adventitious roots. Primary roots were exposed to 0–10−3 M salicylic acid (SA) for 1–5 h; scutellum, to 10−2 M 2-desoxy-D-glucose (2dG). 2dG-sucrose synthesized from 2dG was transported from scutella to the roots along the phloem. Its accumulation in 5-mm-long root tips was the measure of phloem unloading. At the concentrations higher than 10−4 M, SA suppressed unloading. Simultaneously, the uptake of 14C-5,5-dimethyloxazolidinedione (DMO) by root segments was inhibited, indicating cytoplasm acidification. 10−3 M SA also inhibited root respiration and growth. The lower SA concentrations (10−5 and 10−6 M) activated unloading under conditions of weak sucrose phloem transport to the root. They did not affect DMO uptake, respiration, and growth. 10−4 M SA stimulated unloading during 1- or 2-h exposure but did not affect it at longer treatments. A dependence of SA action on its concentration and exposure duration implies its involvement in the control of phloem unloading in the root tip.  相似文献

Mustard (Brassica juncea L.) cultivars Alankar (salt-tolerant) and PBM16 (salt-sensitive) plants were grown with 50 mM NaCl and were sprayed with 0.1, 0.5, and 1.0 mM salicylic acid (SA) to study the physiological processes determining salt tolerance and to observe the influence of SA application on the alleviation of NaCl-induced adverse effects. The content of leaf Na+, Cl, H2O2, TBARS, and electrolyte leakage and the activity of SOD were higher in PBM16 than Alankar. In contrast, nutrients content, activity of APX and GR, glutathione content, photosynthetic and growth characteristics were higher in Alankar. Treatment of 50 mM NaCl resulted in increase of Na+ and Cl, oxidative stress, activity of antioxidant enzymes and glutathione content, while nutrients content, photosynthetic, and growth characteristics decreased in both the cultivars. Application of 0.5 mM SA alleviated the negative effects of 50 mM NaCl maximally, but 1.0 mM SA proved inhibitory. The effect of SA was more conspicuous in Alankar than PBM16. It is concluded that the higher tolerance of Alankar was due to its lower leaf Na+ and Cl content, higher nutrients content, and efficient antioxidant metabolism. The application of 0.5 mM SA substantially alleviated salt-induced adverse effects in Alankar.  相似文献

In this paper, we describe a study concerning the determination of phenylalanine ammonia lyase (PAL) activities leading to decline in disease progress caused by Rhizoctonia solani after application of salicylic acid (SA). Two applications of 1.4 mM SA (pH 6.5) followed by inoculation with Rhizoctonia solani resulted in a quantitative change in polyphenol oxidase (PPO), peroxidase (PRX) isoforms and increase in PAL activities from 4.38 to 19.48 unit g−1 (FM) h−1 in Bundel-1, UPC-4200 and IFC-902 cowpea genotypes. Increase in PAL activities was further observed specifically in UPC-4200 when plants were exposed with Rhizoctonia solani spores. Total soluble proteins did not change after SA treatment, however they were significantly increased in SA sprayed-inoculated UPC-4200 genotype. Of the ten detected isoforms of polyphenol oxidase, isoforms 7 and 10, and isoform 4 of peroxidase showed increased activities by SA application. The disease symptoms measured as areas under progress curve (AUDPC) indicated less A value in SA sprayed Bundel-1 and UPC-4200 genotypes over their controls.  相似文献

Foliar sprays of water or 1, 10 and 100 μM aqueous solutions of gibberellic acid (GA3) or kinetin (KIN) were applied to 40-d-old plants of Nigella sativa (L.) to study their effects on net photosynthetic rate, nitrogen metabolism, and the seed yield. 10 μM solutions of both the hormones, especially GA3, appreciably increased the activities of nitrate reductase and carbonic anhydrase, chlorophyll and total protein contents and net photosynthetic rate in the leaves, along with capsule number and seed yield plant−1, at harvest.  相似文献

Pepper (Capsicum annuum L.) plants were sprayed with salicylic acid (SA) and treated with ultraviolet radiation UV-A (320–390 nm), UV-B (312 nm), and UV-C (254 nm) of 6.1, 5.8, and 5.7 W m−2, respectively. UV significantly reduced contents of chlorophyll (Chl) a and b, and carotenoids (Car). SA treatment moderated Chl and Car reduction in plants treated with UV-B and UV-C. The quantity of antocyanins, flavonoids, rutin, and UV-absorbing compounds in plants that were treated with UV-B, UV-C, and SA were significantly increased. Foliar spray of SA counteracted the UV effects on pepper.  相似文献

Callus cultures initiated from shoot base explants of Curcuma aromatica Salisb. were maintained on Murashige and Skoog (MS) media supplemented with 2 mg dm−3 2,4-dichlorophenoxyacetic acid alone or with 0.5 mg dm−3 kinetin. Plantlets were regenerated from 60 and 180-d-old callus on MS media supplemented with 3 mg dm−3 benzyladenine and 0.5 mg dm−3 α-naphthalene acetic acid. Approximately 8–10 plantlets were produced after 30–40 d of culture per 50 mg of callus inoculated. Out of 113 regenerants analyzed 85 plants were exclusively diploid and 28 were predominantly diploid revealing presence of polyploid nuclei. Frequency of polyploid cells were more in regenerants obtained from 180-d-old callus then from 6-d-old callus which might be attributed to the ageing of callus.  相似文献

The present study was performed to determine the changes in inorganic element content in barley leaves of mammalian sex hormones (MSH). Barley leaves were sprayed with 10−4, 10−6, 10−9, 10−12, 10−15 M concentrations of progesterone, β-estradiol, and androsterone at 7th day after sowing. The plants were harvested at the end of 18 days after treatment with MSH solutions. The inorganic element concentrations were determined using wavelength dispersive X-ray fluorescence spectroscopy technique. Although the all MSH concentrations significantly (p < 0.05) increased the concentrations of calcium, magnesium, phosphorus, sulfur, copper, manganese, aluminum, zinc, iron, potassium, and chlorine, it decreased those of sodium concentration in barley leaves. The maximum changes in the element concentrations were obtained at 10−9 M for plant leaves treated with progesterone, 10−6 M for plant leaves treated with β-estradiol and androsterone. The present study elucidated that MSH significantly (p < 0.05) affected the inorganic element concentrations in barley leaves.  相似文献

The present study was undertaken to reveal the changes in inorganic constituents of plants exposed to mammalian sex hormones (MSH). Chickpea leaves were sprayed with 10−4, 10−6, 10−9, 10−12, and 10−15 M concentrations of progesterone, β-estradiol, and androsterone at 7th day after sowing. The plants were harvested at the end of 18 days after treatment of MSH solutions and the inorganic components determined using a wavelength-dispersive X-ray fluorescence spectroscopy technique. At all of the concentrations tested, MSH significantly increased the contents of K, S, Na, Ca, Mg, Zn, Fe, P, Cu, and Ni. Interestingly, only Mn and Cl contents decreased. The maximum changes in the inorganic composition were recorded at 10−6 M for plants treated with progesterone and 10−9 M for plants treated with β-estradiol and androsterone.  相似文献

The role of gibberellins (GAs) in the regulation of shoot elongation is well established but the phytohormonal control of dry-matter production is poorly understood. In the present study, shoot elongation and dry-matter production were resolved by growing Brassica napus L. seedlings under five light intensities (photon flux densities) ranging from 25 to 500 μmol m−2 s−1. Under low light, plants were tall but produced little dry weight; as light intensity was increased, plants were progressively shorter but had increasing dry weights. Endogenous GAs in stems of 16- and 17-d-old plants were analyzed by gas chromatography-selected ion monitoring with [2H2] internal standards. The contents of GAs increased dramatically with decreasing light intensity: GA1, GA3, GA8 and GA20 were 62, 15, 16 and 32 times higher, respectively, under the lowest versus highest light intensities. Gibberellin A19 was not measured at 25 μmol m−2 s−1 but was 9␣times greater in the 75 compared to 500 μmol m−2 s−1 treatment. Shoot and hypocotyl lengths were closely positively correlated with (log) GA concentration (for example: r 2 = 0.93 for GA1 and hypocotyl length) but shoot dry matter was negatively correlated with GA concentration. The application of gibberellic acid (GA3) produced elongation of plants grown under high light, indication that their low level of endogenous GA was limiting shoot elongation. Although endogenous GA20 showed the greatest influence of light treatment, metabolism of [3H]GA20 and of [3H]GA1 was only slightly influenced by light intensity, suggesting that neither 2β- nor 3β-hydroxylation were points of metabolic regulation. The results of this study indicate that GAs control shoot elongation but are not directly involved in the regulation of shoot dry weight in Brassica. The study also suggests a role of GAs in photomorphogenesis, serving as an intermediate between light condition and shoot elongation response. The present study describes a system for efficient plant regeneration via organogenesis and somatic embryogenesis of safflower (Carthamus tinctorius L.) cv. NARI-6 in fungal culture filtrates (FCF)-treated cultures. FCF was prepared by culturing Alternaria carthami fungal mycelia in selection medium for host-specific toxin production. Cotyledon explants cultured on callus induction medium with different levels of FCF (10–50%) produced embryogenic callus. In organogenesis, 42.2% microshoots formed directly from embryogenic callus tissues in plant regeneration medium with 40% FCF. Isolated embryogenic callus cultured on embryo induction medium containing 40% FCF induced 50.2% somatic embryogenesis. Embryo germination percentage was decreased from 64.5 to 28 in embryo maturation medium containing 40% FCF. However, nine plantlets from organogenesis and 24 plantlets from somatic embryogenesis were selected as FCF-tolerant. Alternaria carthami fungal spores (5 × 105 spores/ml) sprayed on the leaves of FCF-tolerant plants showed enhanced survival rate over control plants, which plants were more susceptible to fungal attack. The number of leaf spot lesions per leaf was decreased from 3.4 to 0.9 and their lesion length was also reduced from 2.9 to 0.7 mm in organogenic derived FCF-tolerant plants over control. In somatic embryo derived FCF-tolerant plants, the number of lesions was decreased from 3.1 to 0.4 and the lesion size was also reduced to 2.7–0.5 mm when compared to the control. This study also examined antioxidant enzyme activity in FCF-tolerant plants. Catalase (CAT) activity was slightly decreased whereas peroxidase (POD) activity was increased to a maximum of 42% (0.19 μmol min−1 mg−1 protein) from organogenesis and 47% (0.23 μmol min−1 mg−1 protein) from embryogenesis in FCF-tolerant plants. Superoxide dismutase (SOD) activity was also increased to 17% (149 U mg−1 protein) and 19.5% (145 U mg−1 protein) in FCF-tolerant plants derived from organogenesis and somatic embryogenesis when compared with control plants.  相似文献

Supplementary UV-B (12.2 kJ m−2 d−1 UV-BBE) provided to Vigna radiata for 2 h d−1 suppressed the length of root, shoot and whole plants, number of leaves, total leaf area, leaf area index, specific leaf mass, fresh and dry mass of leaves and shoot, relative growth rate and net productivity. In unstressed green gram plants (10 kJ m−2 d−1 UV-BBE), triadimefon (TRIAD) (20 mg dm−3) enhanced growth in all parameters over control. The growth promoting effect of TRIAD enabled the UV-B impacted plants to overcome the growth inhibitions to varying degrees indicating its protective potential against UV-B stress. Induction of high-frequency shoot regeneration using nodal segments containing axillary buds from a 1-yr-old mother plants of Cannabis sativa was achieved on Murashige and Skoog (MS) medium containing 0.05–5.0 μM thidiazuron. The quality and quantity of regenerants were better with thidiazuron (0.5 μM thidiazuron) than with benzyladenine or kinetin. Adding 7.0 μM of gibberellic acid into a medium containing 0.5 μM thidiazuron slightly increased shoot growth. Elongated shoots when transferred to half-strength MS medium supplemented with 500 mg l−1 activated charcoal and 2.5 μM indole-3-butyric acid resulted in 95% rooting. The rooted plants were successfully acclimatized in soil. Following acclimatization, growth performance of 4-mo-old in vitro propagated plants was compared with ex vitro vegetatively grown plants of the same age. The photosynthesis and transpiration characteristics were studied under different light levels (0, 500, 1,000, 1,500, or 2,000 μmol m−2 s−1). An increase in photosynthesis was observed with increase in the light intensity up to 1,500 μmol m−2 s−1 and then decreased subsequently at higher light levels in both types of plants. However, the increase was more pronounced at lower light intensities below 500 μmol m−2 s−1. Stomatal conductance and transpiration increased with light intensity up to highest level (2000 μmol m−2 s−1) tested. Intercellular CO2 concentration (C i) and the ratio of intercellular CO2 concentration to ambient CO2 (C i/C a) decreased with the increase in light intensity in both in vitro as well as ex vitro raised plants. The results show that in vitro propagated and hardened plants were functionally comparable to ex vitro plants of same age in terms of gas and water vapor exchange characteristics, within the limits of this study.  相似文献

Metrosideros polymorpha, a dominant tree species in Hawaiian ecosystems, occupies a wide range of habitats. Complementary field and common-garden studies of M. polymorpha populations were conducted across an altitudinal gradient at two different substrate ages to ascertain if the large phenotypic variation of this species is determined by genetic differences or by phenotypic modifications resulting from environmental conditions. Several characteristics, including ecophysiological behavior and anatomical features, were largely induced by the environment. However, other characteristics, particularly leaf morphology, appeared to be mainly determined by genetic background. Common garden plants exhibited higher average rates of net assimilation (5.8 μmol CO2 m−2 s−1) and higher average stomatal conductance (0.18 mol H2O m−2 s−1) than their field counterparts (3.0 μmol CO2 m−2 s−1, and 0.13 mol H2O m−2 s−1 respectively). Foliar δ13C of most common-garden plants was similar among sites of origin with an average value of −26.9‰. In contrast, mean values of foliar δ13C in field plants increased substantially from −29.5‰ at low elevation to −24.8‰ at high elevation. Leaf mass per unit area increased significantly as a function of elevation in both field and common garden plants; however, the range of values was much narrower in common garden plants (211–308 g m−2 for common garden versus 107–407 g m−2 for field plants). Nitrogen content measured on a leaf area basis in common garden plants ranged from 1.4 g m−2 to 2.4 g m−2 and from 0.8 g m−2 to 2.5 g m−2 in field plants. Photosynthetic nitrogen use efficiency (PNUE) decreased 50% with increasing elevation in field plants and only 20% in plants from young substrates in the common garden. This was a result of higher rates of net CO2 assimilation in the common garden plants. Leaf tissue and cell layer thickness, and degree of leaf pubescence increased significantly with elevation in field plants, whereas in common garden plants, variation with elevation of origin was much narrower, or was entirely absent. Morphological characteristics such as leaf size, petiole length, and internode length decreased with increasing elevation in the field and were retained when grown in the common garden, suggesting a potential genetic basis for these traits. The combination of environmentally induced variability in physiological and anatomical characteristics and genetically determined variation in morphological traits allows Hawaiian M. polymorpha to attain and dominate an extremely wide ecological distribution not observed in other tree species.
