SUMMARY. From 16 air-lift samplers described in the literature, three were selected for operation from a small boat: Mackey, Pearson et al. and Verollet & Tachet samplers. Random samples (number of sampling units n= 10) were taken in a large tank with a known number of dried peas, representing invertebrates, amongst stones of uniform size. Separate experiments were performed with three sizes of stones (modal sizes 2–4, 16–20 and 32–36 mm). Stratified-random samples (usually n= 10) were taken in rivers at lour sites with modal particle sizes of < 0.1 2–4. 32–128 and 64–128 mm. The samplers were compared with a Ponar grab in the tank experiments and the field, and with a Naturalist's dredge in the field. Rates of pumping at different air-flows were established for each sampler. Water-How increased with increasing air-How and increasing submergence of the riser-pipe. The Mackey sampler achieved the highest water-flow and lifted more substratum and larger stones (64–128 mm) than the other samplers. The Verollet & Tachet sampler had the lowest pumping rate and failed to lift stones > 8 mm long, whilst the Pearson et al. sampler, with an intermediate performance, lifted mud and stones < 32 mm long. In the tank experiments, the area from which the Mackey and Pearson et al. samplers collected peas varied with air-flow, operating time and type of substratum. The Maekey sampler over-sampled and the Pearson et al. sampler usually under-sampled their respective sampling areas, except the latter sampler at high air-flows gave approximately quantitative results comparable to those of the Ponar grab for peas on the surface of stones 16–20 mm. No sampler collected much at a depth of 3 cm on larger stones (32–36 mm), and the Verollet & Tachet sampler failed completely on all substrata. In field trials, the Mackey and Pearson et al. samplers provided good qualitative samples except on large stones (32–128mm). The Verollet & Tachet sampler lifted little material. The relative abundance of taxa often differed between samplers. Estimates of the number of invertebrates per m2 differed widely between samplers except for the Pearson et al. and Ponar grab which gave similar quantitative results at three sites. Values were very low for the Verollet & Tachet sampler and Naturalist's dredge and always very high for the Mackey sampler which often grossly over-sampled its sampling area. Therefore, these three samplers cannot be considered as quantitative. The relationship between the variances and the means of samples of peas in the tank and most invertebrates in the field followed a power law with values of the exponent b in the range 1.13–2.51. The present study completes the evaluation of 14 samplers used to sample benthic macro-invertebrates in deep rivers, and the more important conclusions from the comparative studies of seven grabs, four dredges and three air-lift samplers are summarized in a table.  相似文献

Using two different surface microlayer sampling devices (the Harvey and Burzell glass plate and the Garrett screen), two microlayer fractions could be distinguished. The first was from 0 to 50 µm and the second was from 51 to 320 µm. Significantly different (p < 0.05) concentrations of dissolved nutrients between the two microlayer fractions strongly suggests stratification within the surface microlayer. The performance of a micro soil solution sampling device was tested in a laboratory examination and in a field experiment. The instrument allows detection of temporal and spatial changes in soil solution chemistry at a spatially high resolution. The flexible tube of the suction cell is made of a porous polymer with a diameter of 2.3 mm. To achieve more stability and to minimize disturbance of the instrument during field installation, the original device was modified by embedding the suction cell in a stainless steel and pressure absorbing corpus. During a laboratory test the new sampling system was compared to ceramic P-80 suction cells. Solution samples taken with the new device adapted more quickly to the given concentrations compared to the ceramic suction cells. In a field test, micro samplers were implanted in an existing soil solution monitoring plot, equipped with standard ceramic samplers. Bi-weekly sampling using the micro cells indicated high temporal and spatial variation, and in June 1995 it was possible, to identify a distinct nitrification. However, in a statistical comparison of the entire sampling period and respective sub-sampling areas the two sampler types indicated identical concentration ranges for nitrate. It is concluded that the new micro samplers can help to identify processes in soils which may cause short-term changes in the soil solution chemistry, whereas the standard sampling technique with ceramic cells seems to be still a suitable tool if long-term mean soil solution concentrations are to be measured.  相似文献

A series of experiments was conducted to explore the utility of composite-based collection of surface samples for the detection of a Bacillus anthracis surrogate using cellulose sponge samplers on a nonporous stainless steel surface. Two composite-based collection approaches were evaluated over a surface area of 3716 cm2 (four separate 929 cm2 areas), larger than the 645 cm2 prescribed by the standard Centers for Disease Control (CDC) and Prevention cellulose sponge sampling protocol for use on nonporous surfaces. The CDC method was also compared to a modified protocol where only one surface of the sponge sampler was used for each of the four areas composited. Differences in collection efficiency compared to positive controls and the potential for contaminant transfer for each protocol were assessed. The impact of the loss of wetting buffer from the sponge sampler onto additional surface areas sampled was evaluated. Statistical tests of the results using ANOVA indicate that the collection of composite samples using the modified sampling protocol is comparable to the collection of composite samples using the standard CDC protocol (p  =  0.261). Most of the surface-bound spores are collected on the first sampling pass, suggesting that multiple passes with the sponge sampler over the same surface may be unnecessary. The effect of moisture loss from the sponge sampler on collection efficiency was not significant (p  =  0.720) for both methods. Contaminant transfer occurs with both sampling protocols, but the magnitude of transfer is significantly greater when using the standard protocol than when the modified protocol is used (p<0.001). The results of this study suggest that composite surface sampling, by either method presented here, could successfully be used to increase the surface area sampled per sponge sampler, resulting in reduced sampling times in the field and decreased laboratory processing cost and turn-around times.  相似文献

  • 1 The chemical characteristics of floodwaters in ephemeral rivers are little known, particularly with regard to their organic loads. These rivers typically exhibit a pronounced downstream hydrological decay but few studies have documented its effect on chemical characteristics and material transport. To develop a better understanding of the dynamics of floods and associated material transport in large ephemeral rivers, floods of the ephemeral Kuiseb River in south‐western Africa were tracked and repeatedly sampled at multiple points along the river's lower 220 km.
  • 2 We quantified the composition and transport of solute and sediment loads in relation to longitudinal hydrological patterns associated with downstream hydrological decay. Source and sink areas for transported materials were identified, and the composition and transport dynamics of the organic matter load were compared to those described from more mesic systems.
  • 3 Concentrations of sediments and solutes transported by floods in the Kuiseb River tended to increase downstream in association with pronounced hydrological decay. The contribution of particulate organic matter to total organic load is among the highest recorded, despite our observation of unusually high levels of dissolved organic matter. Hydrological decay resulted in deposition of all transported material within the lower Kuiseb River, with no discharge of water or materials to the Atlantic Ocean.
  • 4 Our results suggest that longitudinal variation in surface flow and associated patterns of material transport renders the lower Kuiseb River a sink for materials transported from upstream. The downstream transport and deposition of large amounts of labile organic matter provides an important carbon supplement to heterotrophic communities within the river's lower reaches.

SUMMARY.
  • 1 The transport of particulate material in a multiply-impounded southern African river was studied from source to mouth over a period of 2 years during base flow conditions.
  • 2 More than 95% of the dissolved and particulate material in transport was ultra fine (<80 μm). We conclude that too much emphasis has been placed on coarse material (>1 mm) in theories of river ecosystem functioning.
  • 3 The ratio of CPOM:FPOM (1000–4000:250–1000 μm) in transport decreased with increasing stream order as a result of inputs of FPOM from pollution sources and from plankton blooms discharged from reservoirs.
  • 4 The downstream effects of impoundment depended largely on the type of release and the quality of the inflowing water: surface-releasing reservoirs in the clean, upper reaches of the river had the least effect on transported material. A polluted surface-release impoundment in the mid-reaches of the river converted the particle size spectrum of the river from small (<80 μm) to large (250–1000 μm). Bottom-released water carried high concentrations (36–190 g m3) of small (<5 μm) and largely inorganic (86–93%) material.
  • 5 The distance required for zooplankton densities to reduce by 95% was between 4 and 8 km downstream of an impoundment, except below a polluted impoundment where zooplankton densities took 32–35 km to reduce by 95%.
  • 6 Most of the downstream changes in transported organic matter in the Buffalo River are due to inflows of agricultural and urban effluent. These disturbances to the river cause greater perturbations than do the impoundments.

Only few of the numerous samplers for collecting invertebrate fauna associated with vegetation permit sampling at depths of more than approximately 0.5 m. This paper describes a sampler allowing collection of epifauna from submerged plants at a depth of up to approximately 2 m, growing in varied densities. The sampler is composed of two panels made of duralumin, connected on one side by means of piano-hinges. One of the panels has an opening covered with mesh, and the other is equipped with a cone-shaped net and detachable sample concentrator at its end. The sampler is coupled with a manipulator with a several meter extension for lowering the sampler under water, as well as for its opening, and closing. The initial assessment of the sampler efficiency, verified in beds of Potamogeton perfoliatus, provided similar results as two other, older and commonly used samplers. In comparison with the older devices, the sampler has the advantage of permitting collecting samples from greater depths. It also enables studying both the vertical and horizontal distribution of invertebrates within a plant patch.  相似文献

Evidence from spore samples collected amongst infected straw spread on fallow ground supported the conclusion that spores of Pseudocercosporella herpotrichoides are dispersed mostly by rainsplash. Most spores travelled a short distance in the larger ballistic splash droplets, although some may have travelled further in smaller airborne droplets. Weekly spore counts from microscope slides under rainshields, a funnel and an impinger, evaluated as samplers for spores of P. herpotrichoides, showed a similar seasonal pattern. The funnel, as the largest sampler, generally collected most spores, but the impinger collected more spores per unit area of sampling surface. Slides sometimes collected spores when none was recovered from other samplers. Young wheat plants, exposed with the samplers and changed weekly, subsequently developed eyespot symptoms for most of the season.  相似文献

Previous field and laboratory studies investigating airborne Burkholderia pseudomallei have used a variety of different aerosol samplers to detect and quantify concentrations of the bacteria in aerosols. However, the performance of aerosol samplers can vary in their ability to preserve the viability of collected microorganisms, depending on the resistance of the organisms to impaction, desiccation, or other stresses associated with the sampling process. Consequently, sampler selection is critical to maximizing the probability of detecting viable microorganisms in collected air samples in field studies and for accurate determination of aerosol concentrations in laboratory studies. To inform such decisions, the present study assessed the performance of four laboratory aerosol samplers, specifically the all-glass impinger (AGI), gelatin filter, midget impinger, and Mercer cascade impactor, for collecting aerosols containing B. pseudomallei generated from suspensions in two types of culture media. The results suggest that the relative performance of the sampling devices is dependent on the suspension medium utilized for aerosolization. Performance across the four samplers was similar for aerosols generated from suspensions supplemented with 4% glycerol. However, for aerosols generated from suspensions without glycerol, use of the filter sampler or an impactor resulted in significantly lower estimates of the viable aerosol concentration than those obtained with either the AGI or midget impinger. These results demonstrate that sampler selection has the potential to affect estimation of doses in inhalational animal models of melioidosis, as well as the likelihood of detection of viable B. pseudomallei in the environment, and will be useful to inform design of future laboratory and field studies.  相似文献

  • 1 A simple periphyton sampler, which is cheap and easy to construct, is described. The key component is a disposable scouring disc which also serves to adsorb detached material.
  • 2 Field trials show that the scouring sampler is significantly more efficient at removing and retaining periphyton from stones in streams for biomass estimation than other commonly used techniques.
  • 3 The scouring sampler requires only one operator. Its small size and speed of use facilitate replication of samples. Samples can be collected from stones as small as 30mm diameter.

Results from the field testing of some innovative sampling methods developed to evaluate risk management strategies for polychlorinated biphenyl (PCB) contaminated sediments are presented. Semipermeable membrane devices (SPMDs) were combined with novel deployment methods to quantify the availability of PCBs at the Sangamo-Weston Lake Hartwell Superfund Site in SC. Three locations in Lake Hartwell were examined: a background site (BKG) with little detectable contamination and two contaminated sites (T-M/N and T-O). PCB availability was quantified using sediment surface samplers designed to hold SPMDs in contact with surface sediments, benthic dome samplers designed to enclose and suspend SPMDs at the sediment-water interface, and commercially obtained SPMD cages suspended in the water column. A two-way analysis of variance showed significant effects by sampler type (P < 0.0001) and site (P < 0.0001) for mean time-weighted average (TWA) total PCBs (t-PCBs). Regardless of the SPMD sampler used, mean TWA t-PCBs were directly proportional to the level of contamination present (BKG < T-M/N < T-O). Water column and surface sediment sampler t-PCB uptake patterns were described by a significant linear fit of the data (r2 = 0.9625, p < 0.0001, and r2 = 0.8188, p < 0.0001, respectively). Dome and sediment surface SPMD samples had a higher percentage of higher chlorinated PCBs compared to water column samples.  相似文献

The Biotest RCS Plus air sampler was compared with the well-established Casella slit sampler for the quantitative analysis of the microbiological content of air. The results of the tests reported here show that the RCS Plus sampling efficiency is similar to that of the Casella sampler over the range of particles likely to be encountered in the environment. For particles less than 4 μm down to the sub-micronic sizes the efficiency of sampling falls off only gradually so that the efficiency of sampling for 1·0 μm particles is only reduced to about 50%. The ability to pre-select 10 different volumes (from 1 to 1000 1) enables the samplers to be used for measuring a wide range of concentrations of airborne micro-organisms in a variety of locations.  相似文献

A suction sampler designed to minimize sampling error, and quantitatively collect benthic macroinvertebrates from coarse substrata under various flow regimes is described. This sampler uses a 12 V bilge pump (capacity 4400 1 h–1) to suck animals into a collecting net from substrata disturbed by a single operator. Macroinvertebrates were collected more rapidly by this sampler than by a Surber sampler or a Boulton suction sampler. Sources of sampling error are discussed for all three samplers. The electric pump sampler described here is suitable for sampling habitats where depth and velocity vary spatially and with time.  相似文献

Monitoring the number of bacterial colony-forming units is an important step in assuring compliance with the recommendation that water from dental units contain <200 CFU mL–1. Media that have been used for this purpose include R2A, a standard plate counting medium for water samples, and the Millipore HPC Sampler device, designed to facilitate sampling in dental offices. Discrepancies between the two media have been observed. This study tested the hypothesis that differences in counts on the two media were due to the failure of some bacteria to grow on the HPC sampler or to grow at less efficiency than on R2A. Of four different bacterial colony phenotypes tested in three independent experimental trials, one phenotype did not grow on the HPC device, and another grew inconsistently and at lower efficiency. These results confirmed the hypothesis. From these findings, users of the HPC sampler should be aware that microbial undercounts may occur.  相似文献

SUMMARY.
  • 1 The development of stone surface organic layers was investigated in dark and light experimental channels at two field sites. Layer formation was monitored by measuring organic carbon, chlorophyll-a, ATP and rates of oxygen consumption, and using scanning electron microscopy.
  • 2 In the darkened forest stream channel an organic layer consisting of slime, fine particles, bacteria and fungi developed and attained maximum biomass (=0.08 mg cm-2) in about 2 months. At the second site, channels were fed by spring water low in dissolved and particulate organic matter (DOC < 0.5 g m-3) and no organic layer developed on stones in the dark. Organic layers grown in channels subject to natural light intensities and photoperiods were dominated by diatoms and/or filamentous algae at both sites.
  • 3 Laboratory experiments carried out in enclosed, recirculating stream channels demonstrated the importance of dissolved organic matter (DOM) as a prerequisite for layer formation. Also. DOM additions in the form of leaf leachates stimulated oxygen consumption by preformed layers. Uptake by microorganisms accounted for most of the reduction in water-column DOM.
  • 4 Radiotracer experiments (14C and 144Ce) showed that several common stream invertebrates could feed on ‘heterotrophic’ layers. Calculated assimilation efficiencies ranged from 18% to 74% and imply that nonautotrophic components of stone surface organic layers are likely to play a significant role in carbon transfer to the benthos, particularly in small, shaded streams.

SUMMARY.
  • 1 Use of a multi-column thin-layer pneumatic sampler and modified analytic procedures has enabled resolutions of chemical and biological strata at 2.5 or 5.0 cm depth intervals. Examination of meromictic Lake Fidler, Tasmania, indicates the presence of a thin, intense stratum of bacteriochlorohyll d in the upper monimolimnion, associated with a discrete stratum of Chlorobium cf. limicola at the microaerobic interface between the oxygenated and sulphide-rich zones.
  • 2 Algae included small populations of Chlorophyceae, Chrysophyceae, Bacillariophyceae and Cryptophyceae. Bacteria included microaerophils and obligate anaerobes, pigmented and colourless, in well-defined strata in the upper monimolimnion. A population of the microcrustacean Calamoecia tasmanica tasmanica was present in the mixolimnion. Chaoborus larvae were concentrated within the Chlorobium layer.
  • 3 The chemical profile of Lake Fidler was stable, with a chemocline constant in position relative to the lake bottom. The surface water levels rose and fell through a distance of 1m in conjunction with heavy rainfall in the rainforest, and with river level variation, but had no measurable effect on the absolute position of the chemocline. Marked heterogeneity of dissolved substances at depths in the vicinity of bacteria suggested endogenous influence on pH and gelbstoff (‘gilvin’ in Australia).
  • 4 Downwelling light attenuation was influenced primarily by gelbstoff (‘gilvin’) in the mixolimnion, with only red light (peak at 700 nm) measurable below 2 m. Light was absorbed mainly by Chlorobium in the monimolimnion, and was unmeasurable deeper than 3 m.
  • 5 The absorption spectrum of the bacteriochlorophyll d in vivo, with a maximum absorbance at 721 nm, corresponds with the available downwelling light penetrating the mixolimnion to the Chlorobium layer.

Sampling, storage, and preparation of biological material for analysis are some of the most important factors in the study of trace elements in biomedical fields. If due care is not given to these factors, the resulting data may often be meaningless, regardless of the analytical technique chosen for analysis. A survey of the data in the literature would support this view, in which wide differences in the trace element composition of biological tissues have been observed, and not all these differences are attributed to variations in the biological systems. In this paper, the following points are discussed in detail:
  1. Sampling and Storage Sampling methods and program. Contamination of the sample during sampling and storage. Changes in the composition during storage. Various storage methods for solid and liquid samples.
  2. Sample Preparation Drying and ashing. Wet digestion. Different methods for preparing samples of hard, semihard, and soft tissues, and of biological fluids.

SUMMARY. A study of the vertical distribution of benthic macro-invertebrates in the upper Wye, Wales, using basket samplers, indicated no difference between the number of organisms collected, at one sampling time, after 28, 61 and 93 days. However, densities in samples collected after periods greater than 370 days were generally lower than might have been expected. Overall, 59.4%, 22.6% and 18.0% of the benthic invertebrates were recorded in the top (0–11 cm), middle (12–22 cm) and bottom (23–33 cm) levels of the basket samplers. Some organisms were generally confined to the top level (e.g. Ephemeroptera, Simuliidae, Mollusca), others were more evenly distributed (e.g. Oligochaeta, Chironomidae) and some increased with depth at certain times ( Sericostoma personatum ). Except for organisms confined to the upper layer it was not possible to predict with any certainty the vertical distribution of most organisms within the substratum. The community structure and abundance of invertebrates in the basket samplers differed from collections obtained with a conventional surface sampler.  相似文献
