The aim of this study was to determine the influence of heat processing on denaturation and digestibility properties of protein isolates obtained from sweet quinoa (Chenopodium quinoa Willd) at various extraction pH values (8, 9, 10 and 11). Pretreatment of suspensions of protein isolates at 60, 90 and 120 °C for 30 min led to protein denaturation and aggregation, which was enhanced at higher treatment temperatures. The in vitro gastric digestibility measured during 6 h was lower for protein extracts pre-treated at 90 and 120 °C compared to 60 °C. The digestibility decreased with increasing extraction pH, which could be ascribed to protein aggregation. Protein digestibility of the quinoa protein isolates was higher compared to wholemeal quinoa flour. We conclude that an interactive effect of processing temperature and extraction pH on in vitro gastric digestibility of quinoa protein isolates obtained at various extraction pH is observed. This gives a first indication of how the nutritional value of quinoa protein could be influenced by heat processing, protein extraction conditions and other grain components.  相似文献

This study aimed at assessing the impact of four barley forms on total tract apparent digestibility of dietary fibre in horses fed a large amount of starch in the morning meal (0.27% BW). Processed barley forms had a greater pre-caecal starch digestibility than the whole form. Based on this result, we hypothesised that using barley-processing methods would limit the potential dumping of undegraded starch in the hindgut of horses and, consequently, the potential negative effect on fibre degradation in the hindgut. In a 4×4 latin square design, four mature geldings fitted with a right ventral colon-fistula were fed a meadow hay : concentrate (62 : 38; dry matter (DM) basis) diet at 1.7% BW. The concentrate was made of 80% barley distributed either as whole grain or as processed forms: 2.5 mm ground, pelleted or steam-flaked. For each period, total tract apparent digestibilities of DM, NDF and ADF were determined over 3 consecutive days by total faecal collection, whereas pH, volatile fatty acids (VFA) concentrations and cultural functional bacteria counts (total anaerobic, cellulolytic bacteria, lactic acid producers, amylolytic bacteria and lactic acid utilisers) in colonic content were evaluated on 1 day 4 h after the morning meal. Total tract apparent digestibility of DM and dietary fibre was influenced (P<0.05) by barley form. Diets including thermo-mechanically treated barley forms led to a higher (P<0.05) total tract apparent digestibility of NDF than those constituted of ground barley and also led to a greater (P<0.05) total tract apparent digestibility of ADF than those made of whole or ground barley forms. However, no significant difference was observed in colonic pH, VFA concentrations and cultural bacteria concentrations. Owing to a high starch supply in the morning meal, the concentration of the functional bacteria in the colonic content averaged 7.8 log CFU/ml, 5.9 NPM/ml, 6.9 and 7.3 CFU/ml for total anaerobic, cellulolytic, amylolytic and lactic acid-utilising bacteria, respectively. Consequently, providing horses with pelleted or steam-flaked instead of ground barley forms may limit the negative impact of starch on fibre digestibility in horses fed a high level of starch in the morning meal (0.27% BW). Moreover, the fibre-to-starch ratio fed in this experiment did not cause any digestive upset.  相似文献

The present study investigates the effect of guar gum on the digestibility of a waxy maize starch in vitro under simulated gastric and intestinal conditions. A detailed rheology and confocal scanning laser microscopy of the digesta were performed in order to study the possible mechanisms of interactions involved during in vitro hydrolysis of starch. No starch hydrolysis was observed under simulated gastric conditions, whereas more than 90% hydrolysis was observed at the end of digestion under simulated intestinal conditions. In the presence of guar gum, the starch hydrolysis was reduced by nearly 25% during the first 10 min and by 15% at the end of in vitro intestinal digestion. The rheological behavior of the digesta was significantly affected in the presence of the gum. The viscosity of digesta decreased during intestinal digestion; however, the extent of decrease was quite low in the presence of guar gum. The consistency index increased and flow behavior index of digesta decreased in the presence of gum after 1 min of intestinal digestion. All the samples (digested or undigested) displayed a pseudoplastic behavior as their apparent viscosity (η a) decreased with an increase in shear rate. A negative correlation between the starch hydrolysis (%) and storage modulus for the starch sample without gum and a positive correlation for the starch sample with gum were found. Large granule remnants observed through confocal micrographs have shown that the solubilization of starch granule remnants during in vitro digestion is significantly reduced in the presence of gum.  相似文献

A leaf protein concentrate (LPC) extracted from shelled plants of pea (Pisum sativum L.) yielded 108 kg DM ha?1. Dry matter (DM) was analysed for ether extract (13.59%), neutral detergent fibre (NDF, 1.79%), acid detergent fibre (ADF, 0.81%) and total ash (10.57%). The main nitrogen fraction was represented by protein-N (92% of total N). Mineral composition showed an adequate ratio of Ca to P (3). Lysine was present in nutritionally suitable amounts (7.42 g per 100 g recovered amino acids), but methionine was not (1.78). Apparent digestibility (AD; 75.5), true digestibility (TD; 87.6), biological value (BV; 68.8) and net protein utilisation (NPU; 59.7) were determined with Wistar albino rats. Methionine addition improved AD (81.3), BV (80.6) and NPU (70.5), although not TD (87.5) which was already very high.  相似文献

The aim of this study was to investigate the effect of different dietary levels of concentrate on feed intake, digestibility, ruminal fermentation and microbial population in steers. Eight Nellore steers fitted with ruminal cannulas were used in a double 4 × 4 Latin square design experiment. The dietary treatments consist of four different proportions of concentrate to roughage: 30:70, 40:60, 60:40 and 80:20% in the dry matter, resulting in Diets 30, 40, 60 and 80, respectively. The roughage was corn silage, and the concentrate was composed of corn, soybean meal and urea. Apparent digestibility of organic matter and crude protein showed a linear association with concentrate proportion (= 0.01), but the increased concentrate levels did not affect the digestibility of fibre. The lowest ruminal pH-values were observed in animals fed with Diet 80, remaining below pH 6.0 from 6 h after feeding, while in the other diets, the ruminal pH was below 6.0 not before 12 h after feeding. After feeding Diet 80, the ammonia concentration in the rumen was significantly the highest. Higher dietary concentrate levels resulted in a linear increase of propionic acid concentrations, a linear reduction of the ratio acetic acid to propionic acid (p < 0.01) and a linear increased synthesis of microbial nitrogen (p < 0.001). The predicted production of methane was lower in diets with greater amounts of concentrate (p = 0.032). The population of methanogens, R. flavefaciens and R. albus decreased with higher concentrate levels, while the population of S. ruminantium increased (p < 0.05). The results indicate that greater amounts of concentrate do not decrease ruminal pH-values as much as expected and inhibit some cellulolytic bacteria without impairing the dry matter intake and fibre digestibility in Nellore steers.  相似文献

Abstract

The experiment was conducted to study the effects of fermentation of barley, using two different strains of lactic acid bacteria, a Lactobacillus plantarum/pentosus strain isolated from spontaneously fermented rye sourdough (AD2) and a starch-degrading Lactobacillus plantarum (AM4), on contents of mixed-linked (1 → 3) (1 → 4)-β-glucans, α-amylase inhibitor activity, inositol phosphates, and apparent digestibility of macronutrients in mink. Effects of fermentation were compared with effects of gamma irradiation (γ-irradiation: 60Co γ-rays at 25 kGy). The diets were fed to mink with and without a supplementary enzyme preparation. Both lactic acid fermentation and γ-irradiation followed by soaking and incubation, reduced concentrations of soluble β-glucans, phytate and α-amylase inhibitor activity. Dietary enzyme supplementation increased significantly digestibility of crude protein, fat, starch and crude carbohydrate (CHO). Fermentation of the barley increased digestibility of starch and CHO. Fermentation with lactic acid bacteria AD2 resulted in higher starch and CHO digestibility than strain AM4, and had greater effect than γ-irradiation, soaking and incubation. The highest digestibility of starch and CHO was obtained after AD2 fermentation followed by enzyme supplementation. It is concluded that both lactic acid fermentation of barley and enzyme supplementation have positive nutritional implications in the mink by limiting the effects of antinutrients and improving digestibility and energy utilization.  相似文献

Pasta is popular for its ease of cooking and its low glycaemic index (GI). This interesting nutritional property can be attributed to its specific compact structure generally described as a protein network entrapping starch granules. Despite this low GI, pasta is poor in fibres and lack some essential amino acids. To enhance its nutritional composition, pasta can be fortified with non-traditional ingredients such as legume flours. The objective of this study was to investigate the impact of legume flour addition on pasta structure and the inherent consequences on the in vitro digestibility of starch. The addition of a high level (35%, w/w) of legume flour, especially split pea flour, induced some minor structural changes in pasta. The inclusion of fibres, the dilution of gluten proteins by albumins and globulins, and the larger amount of thin protein films (in split pea pasta) may have favoured higher susceptibility of starch to digestive enzymes. At the opposite, the presence of some partially gelatinised starch granules in the core of fortified pasta may have favoured the decrease in the in vitro starch digestibility. As a consequence, a high level of legume flour addition in pasta did not have any significant impact on its in vitro starch digestibility. A high level of split pea and faba bean flours can thus be added to pasta to increase its nutritional composition while keeping its low glycaemic index.  相似文献

A 2 × 2 factorial feeding experiment was conducted to examine the effects of varying the maturity level of the grass used to prepare silage and the nature of concentrate starch source and their interactions on dry matter intake (DMI), diet digestibility, energy corrected milk (ECM) production and milk composition in dairy cows. Twenty-eight multiparous Swedish Red dairy cows, 133 ± 45 days in milk (DIM), with an average milk yield of 30 ± 4 kg/day and a live weight of 624 ± 69 kg were blocked by DIM and randomly assigned to seven replicated balanced 4 × 4 Latin squares with four 21-day experimental periods. The experimental diets consisted of four total mixed rations (TMR) consisting of early-cut grass silage (EGS) supplemented with either barley- or maize-based concentrate and late-cut grass silage (LGS) supplemented with either barley- or maize-based concentrate. All TMR contained identical proportions of forage (51%) and concentrate (49%). Total tract digestibility was estimated by determining indigestible NDF (iNDF) concentrations in feeds and faeces and using iNDF as an internal marker. The feeds’ ruminal degradation parameters were determined using both in situ (nylon bag) and in vitro (gas production (GP)) techniques. Cows offered diets containing EGS had greater (P < 0.001) daily dry matter (DM) intakes, ECM yields and total tract digestibilities for DM and organic matter (OM), but these were not affected by the nature of the concentrate starch source. No interaction between the maturity of the silage and the nature of the concentrate starch source was observed for DMI, diet digestibility or ECM yield. Both grass silages and concentrates had similar rates of ruminal degradation of NDF when measured in situ. The in situ DM (P < 0.001) and starch (P = 0.001) degradation rates of barley-based concentrate were greater than those for maize-based concentrate. In vitro OM GP rates and extents were similar for both concentrate feeds. The results showed that diets containing EGS offered better animal performance and diet digestibility than diets containing LGS. The concentrate starch source did not affect animal performance, but total NDF digestibility was better with diet containing barley- than maize-based concentrate.  相似文献

The objective of this study was to analyse the impact of the gel structure obtained by different heat-induced temperatures on the in vitro gastric digestibility at pH 2. To achieve this, gels were prepared from soy protein, pea protein, albumin from chicken egg white and whey protein isolate at varying temperatures (90, 120 and 140 °C) for 30 min. Gels were characterised prior to digestion via microstructure and SDS-PAGE analysis. Subsequently, the gastric digestion process was followed via the protein hydrolysis and HPSEC analysis up to 180 min. Peptides of different sizes (<5 kDa) were gradually formed during the digestion. Our results showed that gels induced at 140 °C were digested faster. The protein source and gelation temperature had great influence on the in vitro gastric protein digestibility.  相似文献

The present study aimed to determine the effects of combined usage of β-cyclodextrin with whey protein isolate and sodium caseinate on the microencapsulation of Bifidobacterium bifidum-BB12 by spray drying.

From the results, the highest count of B. bifidum was provided by whey protein isolate as 8.62 log CFU/g. The increasing concentration of β-cyclodextrin considerably increases gastric and intestinal resistance to B. bifidum cells. In the gastric and intestinal test, the highest protection was determined in whey protein isolate substituted with 10% β-cyclodextrin with reduction rates of 0.98 and 3.30%, respectively. Moreover, free cells did not survive in the same gastric conditions. The lowest hygroscopicity was determined in whey protein isolate as 8.57%. It must be noted that increasing β-cyclodextrin concentration in carrier material combination led to an increase in hygroscopicity of microcapsules. In general, substitution with β-cyclodextrin increased the particle size of microparticles, and microcapsules produced with whey protein isolate had a smaller size than that of sodium caseinate.

An isolate of arabis mosaic virus (AMV) from a hop plant with symptoms of nettlehead disease induced unusually severe symptoms when transmitted to Chenopodium quinoa. This isolate, called AMV-Ta, yielded particle preparations in which up to 80% of the nucleic acids consisted of a species of low molecular weight (SNA), estimated to be about 75 000 daltons by polyacrylamide gel (PAG) electrophoresis. An isolate free of detectable S-NA (AMV-To) was derived from AMV-Ta by inoculating plants with the two high molecular weight genomic RNA species of AMV (2–8 times 106 and 1–3 times 106 daltons; Murant, 1981) separated from S-NA by PAG electrophoresis. This isolate induced much milder symptoms in C. quinoa. Hop seedlings inoculated with AMV-Ta, either mechanically or by nematodes, developed characteristic nettlehead symptoms. Hop seedlings similarly inoculated with AMV-To remained free of nettlehead symptoms. Two species of S-NA associated with hop nettlehead isolates of AMV were detected at two sites in Kent, and two West Midlands sites. At both sites in Kent and at one of the West Midlands sites, the occurrence of the S-NA species was closely correlated with the incidence of nettlehead symptoms. At the other site in the West Midlands, the occurrence of nettlehead symptoms was too erratic to allow positive correlation of SNA with symptom development. Our results show that S-NA plays an active part in symptom production in experimentally inoculated plants of both hops and C. quinoa. In addition, the close correlation between the occurrence of AMV with additional nucleic acid species, and the incidence of nettlehead symptoms in commercially grown hops, suggests a role for S-NA in the aetiology of this disease.  相似文献

Apart from being an energy source, dietary fibre is also discussed to act as anti-nutritional factor reducing apparent precaecal protein and amino acid (AA) digestibility due to reduced absorption or increased endogenous secretion or both. However, the amounts of protein and AA of endogenous origin determined at the terminal ileum in cannulated animals do not represent the total losses associated with endogenous secretion. A high proportion of secreted protein is reabsorbed and does not reach the terminal ileum, and losses occur during synthesis of endogenous protein. Therefore, the present study used an alternative indirect approach, taking the reduction of nitrogen (N) retention in a threonine (Thr) limited diet as a sensitive indicator for fibre-associated Thr losses. Two experiments were conducted with 12 castrated male pigs each between 37 and 75 kg body weight to measure the effect of the intake of Thr and 150 and 300 g/d fibre from wheat bran (Exp. 1), or 150 g/d fibre from rape seed, cassava leaves, and cassava root peels, respectively (Exp. 2), on N retention. During two (Exp. 1) and three (Exp. 2) balance periods the animals were subjected to the dietary treatments according to a cross-over design. All animals received 1350 g/d of a wheat–soybean-based diet supplemented with free AA to ensure Thr being the first-limiting AA. To determine the effect of Thr on N retention, intake of the basal diet was reduced to 1150 g/d and supplemented with corn starch to reach equal energy intake and an unchanged AA pattern. With increasing BW additional starch was added to all diets to ensure a constant energy intake of 1.25 MJ ME/kg BW0.75. Since the fibre sources contained small amounts of Thr, N retentions were corrected for precaecal digestible Thr intake originating from the fibre sources according to the Thr effect on N retention as determined in experiment 1. Corrected N retentions were affected by fibre level (p = 0.007) and source (p < 0.001). Fibre-associated Thr losses amounted to 3.3, 3.2, 1.2, and 1.1 g/kg fibre from wheat bran, rapeseed, cassava leaf, and cassava root peel, respectively. It is concluded that Thr losses per gram of dietary fibre depend on the fibre source and that fibre concentration and source in pig diets should be considered as a factor affecting Thr requirement.  相似文献

The effect of supplementing diet with yeast culture (Yea-sacc1026) on dairy cattle was studied by rumen in vitro studies and a feeding trial. Using rumen inoculum from a cow, yeast adapted (YA) and yeast unadapted (YU), incubations were carried out with finger millet (Eleusine coracana) straw (FMS) and a commercial cattle feed (CCF). The 24 h cumulative gas production and digestibility of neutral detergent fibre and acid detergent fibre were not different with YU and YA rumen inoculum for both FMS and CCF. Microbial nitrogen synthesis (mg N ml−1 gas produced) with rumen inoculum from YA feeding regime was higher (P < 0.01) than YU with starch (0.1068 vs. 0.1008) and cellulose (0.0900 vs. 0.0859).A feeding trial was conducted using twelve multiparous cows in midlactation in a switch over design. The cows were divided into two groups of six cows in each group. The duration of the trial was fourteen weeks, each period lasting for 7 weeks. There were no differences in dry matter intake, body weight gain and milk yield. Milk composition for the two groups were also similar.  相似文献

The working hypothesis was that a minor postprandial caecal pH decline would affect apparent total tract digestibility (ATTD) of the fibre fraction in horses and, hence, that soluble fibre would amplify fermentation and consequently increase ATTD of fibre. This study was a 4 × 4 Latin Square design with a sequence of 17 days adaptation to the ration followed by 8 sampling days. The feed rations consisted of only timothy hay (Group H), hay plus molassed sugar beet pulp combined with either whole oats (Group OB) or barley (Group BB) and hay plus loose chaff based concentrate (Group M). Four horses fitted with permanent caecal cannulas and collection harnesses were used. A pH electrode with logger was inserted through the cannula and caecal pH was recorded at 1 min intervals for 8 h. The mobile nylon bag technique was used to quantify pre-caecal loss (PCL) of individual feedstuffs. Fibre was analysed as dietary fibre (DF), non-starch polysaccharides, soluble non-cellulosic polysaccharides (S-NCP), insoluble non-cellulosic polysaccharide (I-NCP) and neutral detergent fibre. The ATTD of the S-NCP fraction was above 0.8, which was 60% higher than for the I-NCP fraction. The PCL of starch were 0.98 (oats) and 0.75 (barley). The BB diet lowered (p < 0.001) postprandial caecal pH more than the other diets and a significant correlation was found between the lowest pH and ATTD of I-NCP (r = ?0.66; p = 0.005). In conclusion, this study successfully measured the in vivo digestibility of individual fibre fractions and found that S-NCP was more digestible than the I-NCP, and that a single meal of unprocessed barley was sufficient to decrease caecal pH to such an extent that the fibre digestibility of the whole diet was negatively affected.  相似文献

Food intervention is a financially sensible way for prevention and treatment of diabetes. Extruded snack foods are considered high glycaemic products. Our previous research illustrated that postprandial glycaemic responses to snacks are manipulated by altering dietary fibre and starch contents. The current research assessed the effect of psyllium and oat bran on postprandial glycaemia and in?vitro digestibility. Addition of psyllium fibre to extruded snack products significantly reduced both the in?vitro and in?vivo glycaemic responses of products compared to a control snack product recipe. Oat bran inclusion reduced in?vitro starch digestibility but not in?vivo glycaemic response. The inclusion of oat bran into the snack products appeared to extend the glycaemic response of individuals compared to the control snack, suggesting a possibility of prolonging glucose release and potentially affecting satiety responses. The positive effect in attenuating glucose response means that psyllium fibre could be a target for inclusion by the snack food industry to effectively manipulate postprandial glucose response of individuals.  相似文献

In order to study the main effects of particle size, three ruminally fistulated cows (550 to 580 kg BW) were fed a constant low concentrate level (3.56 kg DM/d, 20% of total DMI) and a fibre‐rich hay (approximately 60% NDF in DM) in long (28.7 mm), chopped (9.2 mm) and fine ground (2.9 mm) form in a 3 x 3 Latin square design. In another three factorial experiment with 8 wethers (4 animals were ruminally fistulated, mean BW = 68 kg) the main effects and interactions of the above mentioned hay particle sizes at two concentrate levels (10.4 to 13.3% and 29.5 to 40.1% of DMI, resp.) and two intake levels (restricted and ad libitum) were investigated. In comparison to long hay (28.7 mm), feeding of chopped hay (9.2 mm) at low concentrate levels, increased not only the hay intake (7% in dairy cows and 13% in sheep) but also the intake of digestible organic matter (12% in dairy cows and 32% in sheep), due to an increase in the apparent digestibility of OM by 3.8% in dairy cows and 8.2% in sheep. Ad libitum feeding of fine ground hay in combination with low concentrate amount in the ration increased the passage rate in the hindgut and consequently the hay intake, but not the intake of DOM, due to a significant depression of digestibility, especially of fibre fractions (4 to 7% in dairy cows and 4.5 to 14% in sheep), in comparison to 28.7 and 9.2 mm hay particle sizes. The digestibility decreased significantly with restricted feeding of fine ground hay in sheep only in comparison to 9.2 mm particle size. A threefold increase of concentrate amount levelled out all effects of the particle size reduction. The effect of particle size was more pronounced in sheep than in dairy cows.  相似文献

Cytochrome P-450 substrate interactions were studied with cytochrome P-450 partially purified from livers of untreated, phenobarbital-treated, benzo[a]pyrene-treated and caffeine-treated rats. Partial inhibition of aminopyrine N-demethylase in presence of in vitro caffeine observed with intact microsomes was further investigated in a reconstituted system composed of partially purified cytochrome c reductase. Caffeine addition (in vitro) to partially purified cytochrome P-450 altered the hexobarbital, aniline and ethylisocyanide induced spectral change, and decreased NADPH oxidation in presence of substrates aminopyrine and acetanilide. NADPH oxidation was found to be increased in presence of aminopyrine and unaltered in presence of acetanilide in reconstituted system having partially purified cytochrome P-450 from caffeine-treated rats. Our studies suggest that caffeine acts as a true modifier of cytochrome P-450 and is possibly responsible for the formation of abortive complexes with aminopyrine.  相似文献

Aquatic ferns (AFs) such as Azolla filiculoides and Salvinia molesta are grown on swine lagoons in the tropics and used in diets for pigs. The present work is aimed at evaluating their potential as feed ingredients for sows. When presented with ad libitum AFs, gilts weighing 110 ± 14 kg (mean ± SD), were able to ingest 9.1–9.7 kg fresh AF per day (from 597 to 630 g dry matter (DM) per day) and from 1240 to 1428 g DM per day when presented in a dry, ground form. A digestibility study was conducted, using sows weighing 213 ± 9 kg (mean ± SD), which were fed diets containing maize, soybean meal and 0, 150 or 300 g AF kg−1 diet. The presence of AFs had a negative impact on the faecal digestibility of the crude protein, NDF and energy content of the whole diet (P<0.001) and on the ileal protein digestibility, especially with 300 g AFs kg−1 diet. The level of AFs in the diet had no effect on stomach weight (P>0.05) but increased the weight of the rest of the gastrointestinal tract (P<0.001). The rate of AF fibre fermentation in the pig large intestine was measured using an in vitro gas test. The rates were much lower than tropical tree foliage, which can also be used in pig diets in the tropics. This could partly explain the low apparent digestibility of AFs in pigs. In conclusion, the inclusion level of AFs in rations for sows should be limited to 150 g AFs kg−1 diet due to the low digestibility and energy density, as well as the negative impact on the digestibility of the whole diet.  相似文献

Livestock performance can be improved by increasing the digestibility of feeds, one of the key elements of nutritional quality. Digestibility of feeds can be improved by breeding forage crops with modified cell-wall structure, increasing the potential availability of energy from the cell wall to rumen microbes and livestock. The objectives of this research were to identify interrelationships among lignin and phenolic components of the fibre fraction of three perennial grasses and to determine their influence on in vitro fibre digestibility. Differences in etherified and esterified ferulate and esterified p-coumarate among clones of three perennial grasses were generally repeatable across harvests. The concentration of neutral detergent fibre (NDF) and Klason lignin within the NDF fraction were the factors most limiting to 24-h in vitro digestibility, with NDF being the most important. Klason lignin and etherified ferulate were the factors most limiting to 96-h in vitro digestibility for all three species. Due to its positive correlation with NDF, selection for low etherified ferulate should be avoided in smooth bromegrass (Bromus inermis Leyss) and cockfsoot (Dactylis glomerata L.). It should be possible to select and breed for low concentrations of lignin to improve digestibility without decreasing NDF in these two species. However, in reed canarygrass (Phalaris arundinacea L.), both lignin and etherified ferulate were positively correlated with NDF, indicating that selection for increased digestibility should be based directly on some measure of in vitro digestibility to avoid the fitness problems associated with reduced NDF.  相似文献
