Genetic studies on Atlantic herring, Clupea harengus, have generally revealed a low level of genetic variation over large geographic areas. Genetically distinct herring populations in some of the Norwegian fjords are exceptions, and juvenile herring from the large oceanic herring, Norwegian Spring Spawners (NSS), are often found in mixture with local fjord populations as well as widely distributed in the Barents Sea. Research surveys in the eastern Barents Sea (Goose Bank) in 1993, 1994 and 2001 included collection of herring samples for allozyme analyses. As expected the results identified juveniles from NSS stock, but an additional unique group of herring (low vertebrae number), being almost fixed for alternative alleles at several allozyme loci, was detected. In some cases, the two groups of herring were taken in the same trawl catches as documented by highly significant departure from Hardy—Weinberg expectation with large excess of homozygotes providing evidence for population mixing. Large genetic differences (Nei's genetic distance = 1.53; FST = 0.754) were detected in pairwise comparisons based on five allozyme loci. The two herring groups were also compared with reference samples of Pacific herring, Clupea pallasi, including one sample from Japan Sea and three Alaskan samples. UPGMA dendrogram based on five allozyme loci revealed a close genetic relationship between the low vertebrae herring in the Barents Sea and the group of samples of Pacific herring. Although significant different in allele frequencies, one of the herring samples clustered together with the reference sample from Bering Sea with genetic distance of 0.008 and FST value of 0.032. The close genetic relationship found in this paper, suggest a re-evaluation of the taxonomic status of the Barents Sea herring populations investigated. 相似文献

Ripe unfertilized eggs of the Pacific herring, Clupea pallasii , release sperm-activating proteins into seawater at the time of fertilization. Five species of herring sperm-activating proteins (HSAP) with different pl values (4.8, 4.9, 5.0, 5.1 and 5.4) were purified from the egg-conditioned medium by gel filtration and isoelectric focusing. Molecular mass of the HSAP (pl = 5.1), the major species of the five HSAP, was determined to be 8.1 kDa by mass spectrometry. Molecular weights of all of the HSAP were estimated to be 7700 by sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE). The first 20 amino acid sequences from N-terminal ends of three HSAP (pl = 4.9, 5.0 and 5.1) were almost identical, suggesting that the HSAP have similar structures. 相似文献

Variation of microsatellite loci Cpa 10, Cpa 113, Cpa4, and Cpa7 was for the first time examined in Pacific-type herring Clupea pallasii from the White Sea (CI. pallasii marisalbi), the Kara Sea (CI. pallasii suworowi), the Sea of Okhotsk, and Lake Nerpich'e, Kamchatka Bay, northwestern Pacific (CI. pallasiipallasii). All loci exhibitedhigh genetic diversity. The estimates of expected heterozygosity varied from 41.5 to 95.6% (mean, 82%). The level of pairwise genetic differentiation Fst at all microsatellite loci varied from 0.005 to 0.076 (0.019, on average) and t was statistically significant (P < 0.05) in most of the pairs of herring samples. Estimates of genetic differentiation among the herring of one subspecies were lower than between the groups belonging to different subspecies. 相似文献

Production and characterization of monoclonal antibodies to IgM of Pacific herring (Clupea pallasii)

Pacific herring (Clupea pallasii) have a central role in the North Pacific ecosystem as a forage fish species and are natural reservoirs of several important finfish pathogens, including Viral hemorrhagic septicemia virus (VHSV). Here, we report the identification of the gene encoding the immunoglobulin mu (IgM) heavy chain, as well as the development and characterization of monoclonal antibodies (MAbs) that specifically react with Pacific herring IgM. Pacific herring immunoglobulin was purified and consisted of heavy and light chains of approximately 80 and 25?kDa. Three hybridoma clones were initially identified by ELISA as reactive with purified immunoglobulin but only one clone was able to detect an 80?kDa protein in Pacific and Atlantic herring (Clupea harengus) whole plasma by denaturing western blot. However, all three MAbs were able to precipitate an 80?kDa protein from Pacific herring and LCMS sequencing of peptide fragments derived from this protein matched the predicted amino acid sequence of the cloned, heavy chain gene. In addition, two of the MAbs stained cells within the putative lymphocyte gates for the spleen, anterior kidney and posterior kidney but were not reactive for myeloid/granulocyte gates, which is consistent with these MAbs reacting with surface IgM(+) B-cells. To our knowledge, this is the first report of IgM-related gene sequences and anti-IgM monoclonal antibodies from any member of the family Clupeidae. The antibodies produced in this study are critical for achieving our long-term goal of conducting serological surveillance to assess pathogen exposure in natural populations of Pacific herring. 相似文献

The genetic variations among spawning groups of herrings from different spawning grounds of the northwestern part of the Sea of Okhotsk was assessed using ten microsatellite loci. All loci were polymorphic with the expected heterozygosity estimates varying at different loci in the range of 0.7–95.0% (with a mean of 68.5%). The degree of genetic differentiation displayed by the herrings from the Sea of Okhotsk was not statistically significant (θ = 0.74%). The level of pairwise genetic differentiation F ST varied in the range of 0.002–0.014, nor was it statistically significant in all comparison pairs between the herring samples. 相似文献

Infectious hematopoietic necrosis (IHN) leads to periodic epidemics among certain wild and farmed fish species of the Northeast (NE) Pacific. The source of the IHN virus (IHNV) that initiates these outbreaks remains unknown; however, a leading hypothesis involves viral persistence in marine host species such as Pacific herring Clupea pallasii. Under laboratory conditions we exposed specific pathogen-free (SPF) larval and juvenile Pacific herring to 10(3) to 10(4) plaque-forming units (pfu) of IHNV ml(-1) by waterborne immersion. Cumulative mortalities among exposed groups were not significantly different from those of negative control groups. After waterborne exposure, IHNV was transiently recovered from the tissues of larvae but absent in tissues of juveniles. Additionally, no evidence of viral shedding was detected in the tank water containing exposed juveniles. After intraperitoneal (IP) injection of IHNV in juvenile herring with 10(3) pfu, IHNV was recovered from the tissues of sub-sampled individuals for only the first 5 d post-exposure. The lack of susceptibility to overt disease and transient levels of IHNV in the tissues of exposed fish indicate that Pacific herring do not likely serve a major epizootiological role in perpetuation of IHNV among free-ranging sockeye salmon Oncorhynchus nerka and farmed Atlantic salmon Salmo salar in the NE Pacific. 相似文献

We gathered mitochondrial DNA sequences (557 bp from the control region in 935 specimens and 668 bp of the cytochrome b gene in 139 specimens) of Pacific herring collected from 20 nearshore localities spanning the species' extensive range along the North Pacific coastlines of Asia and North America. Haplotype diversity and nucleotide diversity were high, and three major phylogeographic lineages (sequence divergences ca. 1.5%) were detected. Using a variety of phylogenetic methods, coalescent reasoning, and molecular dating interpreted in conjunction with paleoclimatic and physiographic evidence, we infer that the genetic make-up of extant populations of C. pallasii was shaped by Pleistocene environmental impacts on the historical demography of this species. A deep genealogical split that cleanly distinguishes populations in the western vs. eastern North Pacific probably originated as a vicariant separation associated with a glacial cycle that drove the species southward and isolated two ancestral populations in Asia and North America. Another deep genealogical split may have involved either a vicariant isolation of a third herring lineage (perhaps originally in the Gulf of California) or it may have resulted simply from the long coalescent times that are possible in large populations. Coalescent analyses showed that all the three evolutionary lineages of C. pallasii experienced major expansions in their most recent histories after having remained more stable in the preceding periods. Independent of the molecular calibration chosen, populations of C. pallasii appear to have remained stable or grown throughout the periods that covered at least two major glaciations, and probably more. 相似文献

Eleven samples of Pacific herring from the four seas of Eurasia (Sea of Japan, Sea of Okhotsk, Bering Sea, and White Sea), and one sample of Atlantic herring were analyzed. Complete and partial sequences of the mtDNA control region with the sizes up to 1071 bp were used. To verify the haplogroups identified, additional sequencing of the cytochrome oxidase I gene was performed. It was demonstrated that Pacific herring from the seas of Eurasia belonged to one mitochondrial haplogroup. The gene flow between the localities from different parts of the Far Eastern sea basins was about 11% per locality, per generation, which led to constant leveling of herring intraspecific differentiation. The data presented gave no reasons for subdivision of the herring populations in accordance to ecological characters (lacustrine and marine). Analysis of global molecular variance (global AMOVA) demonstrated that in Asian water basins, more than 98% of molecular polymorphism was found within the samples at the low level of significance (P < 0.05). 相似文献

A spawning pheromone in the milt (semen) and testes of the Pacific herring,Clupea harengus pallasi,is thought to facilitate school spawning of this species. We found that responsiveness to the spawning pheromone was variable among ripe fish (milt-producing or ovulated). Measurement of five principle reproductive steroids in the free form and five steroids in conjugated forms in the plasma of male fish early in the spawning season (newly ripe fish) showed that elevated plasma levels of 3α,17α-dihydroxy-5β-pregnan-20-one and 17α-hydroxyprogesterone coincided with responsiveness to the spawning pheromone in these fish; levels of other steroids did not differ. In contrast, responsiveness to the pheromone by female fish later in the spawning season (ripe-and-holding fish) coincided with lower levels of glucuronated 17α,20β-dihydroxyprogesterone and a lower gonadosomatic index. We suggest that these differences indicate a more advanced mature reproductive state in the responsive individuals among both the newly ripe male and the ripe-and-holding female fish. We found no differences in the level of cortisol in the blood of the herring that could be correlated with differences in pheromonal responsiveness. We conclude that differences in responsiveness to the spawning pheromone coincide to some extent with levels of reproductive maturation but probably not with recent stress. 相似文献

We examined the spatial and temporal variability of juvenile Pacific herring, Clupea pallasi, growth within Prince William Sound, Alaska. Pacific herring, ranging from post-larval to mature fish, were collected from four spatially segregated bays between October 1995 and March 1998. Linear growth equations from each bay were similar. However, growth rates and wet weight-at-length, reflecting condition, of juvenile Pacific herring cohorts varied seasonally and annually. The short term spatial variability in juvenile Pacific herring growth suggested that each bay was a unique nursery area. The physical and biological conditions within each bay appeared to dictate Pacific herring growth rate. 相似文献

The genetic diversity among spawning groups of herring from different parts of the White Sea was assessed using ten microsatellite loci. All loci were polymorphic with the expected heterozygosity estimates varying in the range of 12.7–94.1% (mean was 59.5%). The degree of genetic differentiation displayed by White Sea herring was statistically significant (θ = 2.03%). The level of pairwise genetic differentiation F ST was 0–0.085, and it was statistically significant in most of the comparison pairs between the herring samples. A hierarchical analysis of molecular variance (AMOVA) revealed the statistically significant differentiation of White Sea herring. 96.59% genetic variation was found within the samples and 3.41% variation was found among the populations. The main component of interpopulation diversity (1.85%) falls at the differences between two ecological forms of herring, spring- and summer-spawning. Within the spring-spawning form, the presence of local stocks in Kandalaksha Bay, Onega Bay, and Dvina Bay was demonstrated. 相似文献

This study investigated effects of microalgae (Rhodomonas baltica) and heterotrophic protists (Oxyrrhis marina) on the daily growth, activity, condition and feeding success of Atlantic herring (Clupea harengus) larvae from hatch, through the end of the endogenous (yolk sac) period. Yolk sac larvae were reared in the presence and absence of microplankton and, each day, groups of larvae were provided access to copepods. Larvae reared with microalgae and protists exhibited precocious (2 days earlier) and ≥ 60% increased feeding incidence on copepods compared to larvae reared in only seawater (SW). In the absence and presence of microalgae and protists, life span and growth trajectories of yolk sac larvae were similar and digestive enzyme activity (trypsin) and nutritional condition (RNA-DNA ratio) markedly declined in all larvae directly after yolk sac depletion. Thus, microplankton promoted early feeding but was not sufficient to alter life span and growth during the yolk sac phase. Given the importance of early feeding, field programs should place greater emphasis on the protozooplankton-ichthyoplankton link to better understand match-mismatch dynamics and bottom-up drivers of year class success in marine fish. 相似文献

Synopsis This study compares simple fish communities of ten oligotrophic lakes in south-central Ontario. Species densities and population
size structure vary significantly among these lake communities depending on fish species present beyond the littoral zone.
Lake whitefish are fewer and larger in the presence of lake herring than in their absence. Diet analysis indicates that lake
whitefish shift from feeding on both plankton and benthic prey when lake herring are absent to a primarily benthic feeding
niche in the presence of lake herring. When benthic round whitefish are present, lake whitefish size and density decline and
they move lower in the lake compared to round whitefish. Burbot are also fewer and larger in lakes with lake herring than
in lakes without herring. Burbot, in turn, appear to influence the population structure of benthic coregonine species. Lower
densities of benthic lake whitefish and round whitefish are found in lakes containing large benthic burbot than in lakes with
either small burbot or where burbot are absent. Predation on the pelagic larvae of burbot and lake whitefish by planktivorous
lake herring alters the size and age structure of these populations. As life history theory predicts, those species with poor
larval survival appear to adopt a bet-hedging life history strategy of long-lived individuals as a reproductive reserve. 相似文献
