Phenylalanin ammonia-lyase (PAL) plays a pivotal role in the production of phenolic compounds, which are responsible for the success of the defense strategies in harsh environments in response to different stimuli. Measurements of the PAL activity, total phenolics, total flavonoids and anthocyanin contents were performed in flowers, leaves and fruits of three pistachio cultivars “Ahmadaghaii”, “Ohadi” and “Kallehghuchi”. The results showed that PAL activity was different in cultivars and in plant organs of pistachio trees (flowers, leaves and fruits). The highest activity rate of their compounds was observed in Ahmadaghaii cultivar. A positive correlation was observed between PAL activity, total phenolics and total flavonoids in leaves, and a negative correlation between PAL activity and anthocyanin contents in leaves and flowers of Ahmadaghaii cultivar. PAL activity and total phenolics in fruits of pistachio suffered a decrease when the maturation processes began. It is suggested that the hulls of the pistachio fruits, containing high level of phenolic compounds (especially in Ahmadaghaii cultivar), may function as a protective layer of defense chemicals against ultraviolet radiation and pathogens. The final concentration of phenolic compounds, flavonoids and antocyanins in the kernel depend on PAL activity in the kernel’s cultivar. The results led to the conclusion that increase in PAL activity, phenolic compounds and flavonoids in Ahmadaghaii can help the plant to cope with the stresses better than the other cultivars. Since phenolic compounds are antioxidant and scavenge free oxygen, it is postulated that Ahmadaghaii is the most resistant cultivar to the environmental stresses.  相似文献

Anatomical, histochemical and phytochemical methods were used to investigate the structure, localization and dynamic changes of total saponin and senegenin of vegetative organs in Polygala tenuifolia Willd. Histochemical localization results showed that saponin accumulated mainly in parenchyma cells of vegetative organs. The phytochemical results also showed that the saponin accumulated in the vegetative organs of P. tenuifolia, with higher content in roots and lower content in the aerial parts that included stems and leaves. The saponin content and dry weight of the vegetative organs of P. tenuifolia had dynamic variance at the developmental stages and all reached the highest level in the post-fruit period. Hence, the roots and aerial parts should be gathered in August to make full use of the plant. As the root is the main medicinal organ of P. tenuifolia, the content of total saponin and senegenin of different aged and different parts of the root were determined. The content of total saponin and senegenin exhibited a sustained decreasing trend with increasing root age; therefore, the annual roots had high quality. The content of total saponin and senegenin in different parts of the root showed obvious variation. The content in the "skin areas" was much higher than that of xylem. The results offer a theoretical basis for determining the appropriate harvesting stage and a reasonable harvest of P. tenuifolia.  相似文献

Contents of rutin and catechin in leaves and under-ground part of Potentilla fragarioides L.in spring and autumn were determined by HPLC technology.The results show that in spring,rutin content in leaves and under-ground part of P.fragarioides is 1.670 3 and 0.054 4 mg·g-1,and catechin content is 0.263 5 and 0.723 5 mg·g-1,respectively.In autumn,rutin content is 0.586 9 and 0.010 7 mg·g-1,and catechin content is 0.784 3 and 0.366 4 mg·g-1,respectively.The rutin content in leaves is the highest in spring,and catechin content in leaves is the highest in autumn.Therefore,collecting leaves in spring is suitable for taking rutin as a purpose,while collecting leaves in autumn is suitable for taking catechin as a purpose.  相似文献

The effects of ultradry storage on the starch mobilization in maize (Zea mays L.) seed after aging were investigated. The results indicated that there were no significant differences in the content of ATP, starch, and soluble sugar, as well as the activity of amylase, between ultradried seeds and seeds stored at -20℃ during germination. These results were consistent with the higher level of vigor of the ultradried seed. Sieve tube introduction of a fluorescence dye (carboxyl fluoresceindiacetate) and laser confocal microscopy were used to study the development of plasmodesmata in the ultradried seeds. The results indicated that plasmodesmata developed well in ultradried seeds. Fluorescence analysis also showed that the fluorescence intensity in the radicle of ultradried seeds was stronger than that in seeds with a higher moisture content. This suggests that ultradry treatment has no adverse effects on the seeds. After seed imbibition, cell orgaelles could be resumed. It is concluded that ultradry seed storage is beneficial for maintaining seed vigor and that starchy mobilization proceeds regularly during germination.  相似文献

In the leaf of rice (Oryza sativa L.) cultivar Yunnan purple rice,the anthocyanins with an obvious absorption peak at 530nm were distributed in the cells of upper and lower epidermis,bulliform tissue and bristle. The maximal photosynthetic oxygen evolution rate and chlorophyll content in flag leaves were 28% and 23%,respectively,more than the common green leaf rice cultivar Chijiaoru-anzhan. Higher chlorophyll content is probably one of the physiological adaptations for enhancing light harvesting capacity of the antenna in photosystems in this cyanic leaves species. Upon the photooxidation of leaf segments mediated by methyl viologen in weak light for 3 days,the distinct bleaching of anthocyanins in purple rice was associated with the reduction of scavenging ability to DPPH· free radical ability and the increase in membrane leakage rate. But almost no changes in contents of flavonoids and total phenolics were observed. Chlorophyll fluorescence parameters Fv/Fo,qP and φPSⅡ decreased with the increase in NPQ and DES of xanthophylls cycle after photooxidation treatment. Green rice leaves showed more decrease in DPPH· scavenging rate and more increase in cell membrane leakage rate but showed a trace of anthocyanins during photooxidation. It is sug-gested that anthocyanin may be a beneficial and primary antioxidant in sun cyanic rice leaves against oxidative stress induced by environmental adversity. And photooxidation could induce different changing patterns of anthocyanins between the tested purple and green rice leaves.  相似文献

Four-leaf rice seedlings (Oryza sativa L.), which had been cultivated in Kimura B complete nutrient solution, were treated with two nitrogen forms by replacing the nitrogen element in the complete solution with sole nitrate or ammonium (2.86 mmol/L). Nitrate-N nutrition tended to increase oxalate content in all parts of the plant, including the leaves, stems, roots, and root exudates, whereas ammonium had the opposite effect. Consequently, marked differences in oxalate content were observed between the two treatments throughout the time tested (0-12 d), with maximal differences of approximately 12-fold at 6d after treatment. Photosynthetic/respiratory parameters were examined over time simultaneously with changes in oxalate content. Net photosynthetic rate, chlorophyll fluorescence parameters (i.e. maximal photochemical efficiency (Fv/Fm) and photochemical quantum yields of photosystem (PS)II (ΦPSⅡ)), and respiratory rate were not significantly different between plants treated with the two nitrogen forms, although ammonium-fed plants had apparently higher leaf chlorophyll content than nitrate-fed plants. Leaf glucose content was altered little, but the content of fructose, sucrose, and total soluble sugar was significantly higher in the leaves of ammonium-fed plants than nitrate-fed plants. The results indicate that nitrate/ammonium may serve as efficient regulators of oxalate accumulation owing to regulation of metabolism in rice leaves rather than oxalate downward transfer and root excretion, and that photosynthetic metabolism is not directly correlated with the regulation of oxalate accumulation in rice plants.  相似文献

The influence of abscisic acid (ABA) on the processes of formation of different polysomal populations, their structures and stability in embryonal tissue during pea seeds germination was studied. The contents of total ribosomal fraction increased in all samples up to 72 h of germination and then decreased. The contents of polysomal population (FP, MBP, CBP and CMBP) extracted from the embryonal tissue after 72 hrs of germination of pea seeds were then quantified. It turned out that in examined tissue of control sample, fraction of free polysomes (FP) was the most abounded. This population of polysomes in sprouts decreased after ABA treatment. FP content decreased even more when the higher ABA concentration was applied during germination. Similar changes were observed in the fraction of membrane-bound polysomes (MBP). Quite different tendencies were found, however, in forming population of the cytoskeleton-membrane-bound polysomes (CMBP). The CMBP population content in embryonal tissue increased in a dosage dependent manner with increasing concentration of ABA applied during seed germination. This indicates the important role of CMBP fraction in synthesis of specific proteins in embryos in the time when processes of seeds germination are retarded by ABA. In the final part we examined the stability of polysomes isolated from sprouts of germinating seeds in water and sprouts isolated from seeds treated with ABA (100 μM) during germination. Total polysomes isolated from embryonal tissue of germinating seeds treated with ABA showed much higher resistance to exogenous ribonuclease digestion than total polysomes of control sample. The obtained results suggest that ABA influence on different polysomal population formation also controls their stability.  相似文献

The concentration of chlorophyll and a carotenoids in the bark of stems of different age and in the leaves of lilac (Syringa vulgaris L.) was determined. The thickness of bark changes with the age of the stems, ranging from 0.73 mm in the current-year stems to 1.22 mm in 3-year-old ones. Chlorophyll and carotenoids were present through the whole thickness of the bark, except the cork. It was found that chlorophyll and carotenoids are located mainly in the outer layer of the bark, immediately under the cork, to a depth of 400 μm. In this layer the chlorophyll a/b ratio is the highest and the content of chlorophyll is four times larger than that of carotenoids. When penetrating deeper into the bark, the content of chlorophyll and carotenoids as well as the chlorophyll a/b ratio diminishes. Investigations of the leaves showed that most of the chlorophyll is found in the palisade parenchyma, the chlorophyll a/b ratio is the highest in the upper layer. The highest concentration of chlorophyll in the bark is 0.44 mg·dm−2 and in leaves −1.2 mg−2·dm−2. The highest value of the chlorophyll a/b ratio in the bark is 3.8, and the lowest 0.5, while in the leaves it varies from 4.5 to 3.8 Low values of the chlorophyll a/b ratio are due to the shade conditions existing in the bark and they are evidence of very great differentiation of light conditions within it.  相似文献

With three different methods, the total flavonoids from the roots of cultivated Gtycyrrhiza pallidiflora Maxim. were determined and compared by ultraviolet-visible spectrophotometry. The results showed that the extract yield of total flavonoids in crude extracts was 4.54% by direct determination method, 4.27% by AI( NO3)3 colorimetry method and 2.09% by AlCl3 colorimetry method. The content of total flavonoids in roots of cultivated G. pallidiflora was only 0.05%. The cultivated G. pallidiflora in Lanzhou is not unfit for the medicine source of flavonoids.  相似文献

Paramyxovirus-mediated membrane fusion usually requires an interaction between the viral-attachment and -fusion proteins. The mechanism by which this interaction regulates fusion differs between paramyxoviruses that bind to sialic acid-containing receptors and those that recognize specific proteins. The recently solved structure of the globular head of the measles virus hemagglutinin suggests that this difference might be related to the location of the receptor-binding sites on the attachment proteins of the two classes of paramyxoviruses.  相似文献

Papillomaviruses (PVs) infect stratified squamous epithelia in vertebrates. Some PVs are associated with different types of cancer and with certain benign lesions. It has been assumed that PVs coevolved with their hosts. However, recently it has been shown that different regions of the genome have different evolutionary histories. The PV genome has a modular nature and appeared after the addition of pre-existent blocks. This order of appearance in the PV genome is evident today in the different evolutionary rates of the different genes, with new genes--E5, E6 and E7--diverging faster than old genes--E1, E2, L2 and L1. Here, we propose an evolutionary framework aiming to integrate genome evolution, PV biology and epidemiology of PV infections.  相似文献

Abstract Artificial structures, such as seawalls, pilings and pontoons, are common features of urban estuaries. They replace natural structures or add to the amount of hard substratum in an area and provide habitats for many fish and invertebrates. Previous work has concentrated on fish or on the invertebrates that occupy the primary substratum of artificial structures. Mussels often grow on different types of structures (pontoons, pilings, seawalls and natural reefs) and provide a secondary substratum for other organisms to inhabit. Counting and identifying organisms associated with mussel beds is traditionally done to species level, which is very time‐consuming. To save time, organisms in this study were identified to coarse levels of taxonomic resolution (a mix of taxa, such as class, order, family and genus), which showed similar patterns to those when particularly speciose and abundant groups were identified to species. This study tests hypotheses that the distribution and abundance of mobile and sessile organisms that inhabit mussel beds will differ among natural and various types of artificial structures. When the associated assemblages of mussel beds from different types of structures and from different locations were examined, assemblages varied according to the type of structure they inhabited and its location. Assemblages associated with mussels on pontoons differed consistently from those on other types of structures. Patterns in the assemblages were also consistent through time. These data show that the types and amounts of artificial structures added to an environment can affect the types, distribution and abundances of organisms living in biogenic habitats.  相似文献

This study evaluates the antimicrobial effects of ethanolic extract of five herbal plants; Guava (Psidium guajava), Sage (Salvia officinalis), Rhamnus (Ziziphusspina Christi), Mulberry (Morusalba L.), and Olive (Oleaeuropaea L) leaves against several microbial population representing Gram positive, Gram negative and Mollicutes; S. aureus, E. coli, Pasteurella multocida, B. cereus, Salmonella Enteritidis and M. gallisepticum using standard agar disc diffusion technique and minimal inhibitory concentration (MIC). Different extracts reveal variable results against the microorganism under study. All extracts have no antibacterial potency for Mycoplasma gallisepticum except Psidium guajava. The results of minimal inhibitory concentration (MIC) and Minimum bactericidal concentration (MBC) of the extracts against the six bacteria ranged from 625 to 5000 μg/ml. The used herbal extract could inhibit the selected microorganism under study with variable minimal inhibitory concentration (MIC) and minimum bactericidal concentration (MBC).  相似文献

Ras signalling has classically been thought to occur exclusively at the inner surface of a relatively uniform plasma membrane. Recent studies have shown that Ras proteins interact dynamically with specific microdomains of the plasma membrane as well as with other internal cell membranes. Energy balance is maintained via a homeostatic system involving both the brain and the periphery. A key component of this system is the hypothalamus. Over the past two decades, major advances have been made in identifying an increasing number of peptides within the hypothalamus that contribute to the process of energy homeostasis. Under stable conditions, equilibrium exists between anabolic peptides that stimulate feeding behavior, as well as decrease energy expenditure and lipid utilization in favor of fat storage, and catabolic peptides that attenuate food intake, while stimulating sympathetic nervous system (SNS) activity and restricting fat deposition by increasing lipid metabolism. The equilibrium between these neuropeptides is dynamic in nature. It shifts across the day-night cycle and from day to day and also in response to dietary challenges as well as peripheral energy stores. These shifts occur in close relation to circulating levels of the hormones, leptin, insulin, ghrelin and corticosterone, and also the nutrients, glucose and lipids. These circulating factors together with neural processes are primary signals relaying information regarding the availability of fuels needed for current cellular demand, in addition to the level of stored fuels needed for long-term use. Together, these signals have profound impact on the expression and production of neuropeptides that, in turn, initiate the appropriate anabolic or catabolic responses for restoring equilibrium. In this review, we summarize the evidence obtained on nine peptides in the hypothalamus that have emerged as key players in this process. Data from behavioral, physiological, pharmacological and genetic studies are described and consolidated in an attempt to formulate a clear statement on the underlying function of each of these peptides and also on how they work together to create and maintain energy homeostasis.  相似文献
